Quantification of land surface–atmosphere fluxes of carbon dioxide (CO2) and their trends and uncertainties is essential for monitoring progress of the EU27+UK bloc as it strives to meet ambitious targets determined by both international agreements and internal regulation. This study provides a consolidated synthesis of fossil sources (CO2 fossil) and natural (including formally managed ecosystems) sources and sinks over land (CO2 land) using bottom-up (BU) and top-down (TD) approaches for the European Union and United Kingdom (EU27+UK), updating earlier syntheses (Petrescu et al., 2020, 2021). Given the wide scope of the work and the variety of approaches involved, this study aims to answer essential questions identified in the previous syntheses and understand the differences between datasets, particularly for poorly characterized fluxes from managed and unmanaged ecosystems. The work integrates updated emission inventory data, process-based model results, data-driven categorical model results, and inverse modeling estimates, extending the previous period 1990–2018 to the year 2020 to the extent possible. BU and TD products are compared with the European national greenhouse gas inventory (NGHGI) reported by parties including the year 2019 under the United Nations Framework Convention on Climate Change (UNFCCC). The uncertainties of the EU27+UK NGHGI were evaluated using the standard deviation reported by the EU member states following the guidelines of the Intergovernmental Panel on Climate Change (IPCC) and harmonized by gap-filling procedures. Variation in estimates produced with other methods, such as atmospheric inversion models (TD) or spatially disaggregated inventory datasets (BU), originate from within-model uncertainty related to parameterization as well as structural differences between models. By comparing the NGHGI with other approaches, key sources of differences between estimates arise primarily in activities. System boundaries and emission categories create differences in CO2 fossil datasets, while different land use definitions for reporting emissions from land use, land use change, and forestry (LULUCF) activities result in differences for CO2 land. The latter has important consequences for atmospheric inversions, leading to inversions reporting stronger sinks in vegetation and soils than are reported by the NGHGI. For CO2 fossil emissions, after harmonizing estimates based on common activities and selecting the most recent year available for all datasets, the UNFCCC NGHGI for the EU27+UK accounts for 926 ± 13 Tg C yr−1, while eight other BU sources report a mean value of 948 [937,961] Tg C yr−1 (25th, 75th percentiles). The sole top-down inversion of fossil emissions currently available accounts for 875 Tg C in this same year, a value outside the uncertainty of both the NGHGI and bottom-up ensemble estimates and for which uncertainty estimates are not currently available. For the net CO2 land fluxes, during the most recent 5-year period including the NGHGI estimates, the NGHGI accounted for −91 ± 32 Tg C yr−1, while six other BU approaches reported a mean sink of −62 [-117,-49] Tg C yr−1, and a 15-member ensemble of dynamic global vegetation models (DGVMs) reported −69 [-152,-5] Tg C yr−1. The 5-year mean of three TD regional ensembles combined with one non-ensemble inversion of −73 Tg C yr−1 has a slightly smaller spread (0th–100th percentiles of [-135,+45] Tg C yr−1), and it was calculated after removing net land–atmosphere CO2 fluxes caused by lateral transport of carbon (crop trade, wood trade, river transport, and net uptake from inland water bodies), resulting in increased agreement with the NGHGI and bottom-up approaches. Results at the category level (Forest Land, Cropland, Grassland) generally show good agreement between the NGHGI and category-specific models, but results for DGVMs are mixed. Overall, for both CO2 fossil and net CO2 land fluxes, we find that current independent approaches are consistent with the NGHGI at the scale of the EU27+UK. We conclude that CO2 emissions from fossil sources have decreased over the past 30 years in the EU27+UK, while land fluxes are relatively stable: positive or negative trends larger (smaller) than 0.07 (−0.61) Tg C yr−2 can be ruled out for the NGHGI. In addition, a gap on the order of 1000 Tg C yr−1 between CO2 fossil emissions and net CO2 uptake by the land exists regardless of the type of approach (NGHGI, TD, BU), falling well outside all available estimates of uncertainties. However, uncertainties in top-down approaches to estimate CO2 fossil emissions remain uncharacterized and are likely substantial, in addition to known uncertainties in top-down estimates of the land fluxes. To become carbon neutral by 2050, the European Union (EU27) net carbon sink from forests should increase from the current level of about −360 to −450 Mt CO2eq yr−1 by 2050. Reaching this target requires additional efforts, which should be informed by the expected interactions between current age-class distributions, the effect of forest management practices and the expected impacts of future climate change. However, modelling the combined effect of these drivers is challenging, since it requires a mechanistic assessment of climate impacts on primary productivity and heterotrophic respiration and a detailed representation of the forest age structure and of the management practices across the entire EU. To achieve this goal, we combined the output provided by four land–climate models – run under two different Representative Concentration Pathway scenarios (RCP2.6 and RCP6.0) – to parameterize the input data used in an empirical forest growth model. This hybrid modelling approach aims to quantify the impact of climate change and forest management on the long-term (i.e. to 2100) evolution of the EU27 + UK forest carbon budget. This approach was tested using a business-as-usual (BAU) scenario, based on the continuation of the management practices applied by EU member states within the historical period 2000–2015. We emphasize that our study does not explore a specific policy scenario but describes a methodological framework. Our results highlight that, under our BAU case, the EU27 + UK forest C sink would decrease to about −250 Mt CO2eq yr−1 in 2050 and −80 Mt CO2eq yr−1 by 2100. The main driver of the long-term evolution of the forest C sink is the ongoing ageing process of the European forests, mostly determined by past and ongoing management. In addition, climate change may further amplify or mitigate this trend. Due to the large uncertainty in climate projections, in 2050 the net C sink may range from −100 to −400 Mt CO2eq yr−1 under RCP2.6 and from −100 to −300 Mt CO2eq yr−1 under RCP6.0. These results suggest that while a change in management practices would be needed to reverse an otherwise declining trend in the sink, climate change adds a considerable uncertainty, potentially nearly doubling or halving the sink associated with management.

Reliable quantification of the sources and sinks of atmospheric carbon dioxide (CO2), including that of their trends and uncertainties, is essential to monitoring the progress in mitigating anthropogenic emissions under the Kyoto Protocol and the Paris Agreement. This study provides a consolidated synthesis of estimates for all anthropogenic and natural sources and sinks of CO2 for the European Union and UK (EU27 + UK), derived from a combination of state-of-the-art bottom-up (BU) and top-down (TD) data sources and models. Given the wide scope of the work and the variety of datasets involved, this study focuses on identifying essential questions which need to be answered to properly understand the differences between various datasets, in particular with regards to the less-well-characterized fluxes from managed ecosystems. In comparing NGHGIs with other approaches, a key source of uncertainty is that related to different system boundaries and emission categories (CO2 fossil) and the use of different land use definitions for reporting emissions from land use, land use change and forestry (LULUCF) activities (CO2 land). At the EU27 + UK level, the NGHGI (2019) fossil CO2 emissions (including cement production) account for 2624 Tg CO2 in 2014 while all the other seven bottom-up sources are consistent with the NGHGIs and report a mean of 2588 (± 463 Tg CO2). The inversion reports 2700 Tg CO2 (± 480 Tg CO2), which is well in line with the national inventories. Over 2011–2015, the CO2 land sources and sinks from NGHGI estimates report −90 Tg C yr−1 ± 30 Tg C yr−1 while all other BU approaches report a mean sink of −98 Tg C yr−1 (± 362 Tg of C from dynamic global vegetation models only). For the TD model ensemble results, we observe a much larger spread for regional inversions (i.e., mean of 253 Tg C yr−1 ± 400 Tg C yr−1). This concludes that (a) current independent approaches are consistent with NGHGIs and (b) their uncertainty is too large to allow a verification because of model differences and probably also because of the definition of “CO2 flux” obtained from different approaches. Emission of greenhouse gases (GHGs) and removals from land, including both anthropogenic and natural fluxes, require reliable quantification, including estimates of uncertainties, to support credible mitigation action under the Paris Agreement. This study provides a state-of-the-art scientific overview of bottom-up anthropogenic emissions data from agriculture, forestry and other land use (AFOLU) in the European Union (EU281). The data integrate recent AFOLU emission inventories with ecosystem data and land carbon models and summarize GHG emissions and removals over the period 1990–2016. This compilation of bottom-up estimates of the AFOLU GHG emissions of European national greenhouse gas inventories (NGHGIs), with those of land carbon models and observation-based estimates of large-scale GHG fluxes, aims at improving the overall estimates of the GHG balance in Europe with respect to land GHG emissions and removals. Whenever available, we present uncertainties, its propagation and role in the comparison of different estimates. While NGHGI data for the EU28 provide consistent quantification of uncertainty following the established IPCC Guidelines, uncertainty in the estimates produced with other methods needs to account for both within model uncertainty and the spread from different model results. The largest inconsistencies between EU28 estimates are mainly due to different sources of data related to human activity, referred to here as activity data (AD) and methodologies (tiers) used for calculating emissions and removals from AFOLU sectors. The comprehensive analysis of carbon stocks and fluxes of managed European forests is a prerequisite to quantify their role in biomass production and climate change mitigation. We applied the Carbon Budget Model (CBM) to 26 European countries, parameterized with country information on the historical forest age structure, management practices, harvest regimes and the main natural disturbances. We modeled the C stocks for the five forest pools plus harvested wood products (HWPs) and the fluxes among these pools from 2000 to 2030. The aim is to quantify, using a consistent modeling framework for all 26 countries, the main C fluxes as affected by land-use changes, natural disturbances and forest management and to assess the impact of specific harvest and afforestation scenarios after 2012 on the mitigation potential of the EU forest sector. Substitution effects and the possible impacts of climate are not included in this analysis. Results show that for the historical period from 2000 to 2012 the net primary productivity (NPP) of the forest pools at the EU level is on average equal to 639 Tg C yr−1. The losses are dominated by heterotrophic respiration (409 Tg C yr−1) and removals (110 Tg C yr−1), with direct fire emissions being only 1 Tg C yr−1, leading to a net carbon stock change (i.e., sink) of 110 Tg C yr−1. Fellings also transferred 28 Tg C yr−1 of harvest residues from biomass to dead organic matter pools. The average annual net sector exchange (NSE) of the forest system, i.e., the carbon stock changes in the forest pools including HWP, equals a sink of 122 Tg C yr−1 (i.e., about 19 % of the NPP) for the historical period, and in 2030 it reaches 126, 101 and 151 Tg C yr−1, assuming constant, increasing (+20 %) and decreasing (−20 %) scenarios, respectively, of both harvest and afforestation rates compared to the historical period. Under the constant harvest rate scenario, our findings show an incipient aging process of the forests existing in 1990: although NPP increases (+7 %), heterotrophic respiration increases at a greater rate (+13 %) and this leads to a decrease in the sink in the forest pools (−6 %) in 2030 compared to the historical period. By comparing the evolution of the biomass as a function of the NPP (i.e., the turnover time) for each country, we highlighted at least three groups of countries and turnover times. This means that, contrary to the assumptions proposed by other authors, this relationship cannot be assumed as a constant for all the EU countries, but specific conditions, such as the harvest rate, the current age structure and the forest composition, may contribute to the country-specific evolution of biomass stocks. The detailed picture of the C fluxes condensed in this study, and their evolution under different harvest scenarios, may represent both a benchmark for similar studies and a basis for broader analyses (e.g., including substitution effects of wood) on the mitigation potential of the EU forest sector.

Data on the potential generation of energy from wind, solar and biomass is crucial for analysing their development, as it sets the limits on how much additional capacity it is feasible to install. This paper presents the methodologies used for the development of ENSPRESO, ENergy System Potentials for Renewable Energy SOurces, an EU-28 wide, open dataset for energy models on renewable energy potentials, at national and regional levels for the 2010–2050 period. In ENSPRESO, coherent GIS-based land-restriction scenarios are developed. For wind, resource evaluation also considers setback distances, as well as high resolution geo-spatial wind speed data. For solar, potentials are derived from irradiation data and available area for solar applications. Both wind and solar have separately a potential electricity production which is equivalent to three times the EU's 2016 electricity demand, with wind onshore and solar requiring 16% and 1.4% of total land, respectively. For biomass, agriculture, forestry and waste sectors are considered. Their respective sustainable potentials are equivalent to a minimum 10%, 1.5% and 1% of the total EU primary energy use. ENSPRESO can enrich the results of any energy model (e.g. JRC-EU-TIMES) by improving its analyses of the competition and complementarity of energy technologies. Traditional pulp-capping materials like mineral trioxide aggregate (MTA) offer excellent biocompatibility and sealing, but limitations such as prolonged setting time, low bioactivity, and high costs persist. Metformin, with its potential in craniofacial regeneration, could enhance dentin synthesis by targeting pulp cells. This study aimed to: (1) develop a calcium phosphate cement with chitosan (CPCC) with improved physio-mechanical properties; (2) incorporate metformin (CPCC-Met) to assess release; and (3) evaluate human dental pulp stem cells (hDPSCs) response. CPCC was mixed at different powder-to-liquid ratios to evaluate physio-mechanical properties compared to MTA. The optimized CPCC formulation was loaded with 0, 50, 100, and 150 µg of metformin to measure release and assess hDPSCs attachment and proliferation (1, 4, and 7 d) via live/dead imaging and SEM. One-way ANOVA was used for statistical analysis. Results showed CPCC at a 3.25:1 ratio significantly reduced setting time to 41.5 min versus 123 min for MTA (p < 0.05). Metformin release correlated with concentration, and SEM confirmed the presence of a porous, hydroxyapatite-rich surface. Cell viability was consistently high across groups (>93% at 1 d, >95% at 4 d, ≈98% at 7 d), with no significant differences (p > 0.05). These findings suggest that the novel CPCC-Met demonstrates promise as a fast-setting, cost-effective pulp-capping material, offering metformin delivery to enhance dentin repair.

Abstract Following a decision to require label warnings for concurrent use of opioids and benzodiazepines and increased risk of respiratory depression and death, the US Food and Drug Administratioin (FDA) recognized that other sedative psychotropic drugs may be substituted for benzodiazepines and be used concurrently with opioids. In some cases, data on the ability of these alternatives to depress respiration alone or in conjunction with an opioid are lacking. A nonclinical in vivo model was developed that could detect worsening respiratory depression when a benzodiazepine (diazepam) was used in combination with an opioid (oxycodone) compared to the opioid alone based on an increased arterial partial pressure of carbon dioxide (pCO2). The current study used that model to assess the impact on respiration of non‐benzodiazepine sedative psychotropic drugs representative of different drug classes (clozapine, quetiapine, risperidone, zolpidem, trazodone, carisoprodol, cyclobenzaprine, mirtazapine, topiramate, paroxetine, duloxetine, ramelteon, and suvorexant) administered alone and with oxycodone. At clinically relevant exposures, paroxetine, trazodone, and quetiapine given with oxycodone significantly increased pCO2 above the oxycodone effect. Analyses indicated that most pCO2 interaction effects were due to pharmacokinetic interactions resulting in increased oxycodone exposure. Increased pCO2 recorded with oxycodone‐paroxetine co‐administration exceeded expected effects from only drug exposure suggesting another mechanism for the increased pharmacodynamic response. This study identified drug‐drug interaction effects depressing respiration in an animal model when quetiapine or paroxetine were co‐administered with oxycodone. Clinical pharmacodynamic drug interaction studies are being conducted with these drugs to assess translatability of these findings.

Functionalization of C(sp 3 )-H bonds represents the most straightforward and atom-economical transformation in organic synthesis. An innovative approach integrating photocatalytic hydrogen atom transfer (HAT) and transition metal catalysis has made significant progress in the coupling of α-heterosubstituted C-H bonds with alkyl halides. However, unactivated alkanes were ineffective as a result of the preponderance of byproduct formation. Herein, we demonstrate direct HAT and nickel catalysis in the coupling of cycloalkanes and benzyl bromides/primary alkyl iodides. Additionally, tetrabutylammonium decatungstate (TBADT) was recovered and recycled.

Forest biomass is an essential resource in relation to the green transition and its assessment is key for the sustainable management of forest resources. Here, we present a forest biomass dataset for Europe based on the best available inventory and satellite data, with a higher level of harmonisation and spatial resolution than other existing data. This database provides statistics and maps of the forest area, biomass stock and their share available for wood supply in the year 2020, and statistics on gross and net volume increment in 2010-2020, for 38 European countries. The statistics of most countries are available at a sub-national scale and are derived from National Forest Inventory data, harmonised using common reference definitions and estimation methodology, and updated to a common year using a modelling approach. For those counties without harmonised statistics, data were derived from the State of Europe's Forest 2020 Report at the national scale. Background: The European Union (EU) has committed to achieve climate neutrality by 2050. This requires a rapid reduction of greenhouse gas (GHG) emissions and ensuring that any remaining emissions are balanced through CO 2 removals. Forests play a crucial role in this plan: they are currently the main option for removing CO 2 from the atmosphere and additionally, wood use can store carbon durably and help reduce fossil emissions. To stop and reverse the decline of the forest carbon sink, the EU has recently revised the regulation on land use, land-use change and forestry (LULUCF), and set a target of - 310 Mt CO 2 e net removals for the LULUCF sector in 2030., Results: In this study, we clarify the role of common concepts in forest management - net annual increment, harvest and mortality - in determining the forest sink. We then evaluate to what extent the forest sink is on track to meet the climate goals of the EU. For this assessment we use data from the latest national GHG inventories and a forest model (Carbon Budget Model). Our findings indicate that on the EU level, the recent decrease in increment and the increase in harvest and mortality are causing a rapid drop in the forest sink. Furthermore, continuing the past forest management practices is projected to further decrease the sink. Finally, we discuss options for enhancing the sinks through forest management while taking into account adaptation and resilience., Conclusions: Our findings show that the EU forest sink is quickly developing away from the EU climate targets. Stopping and reversing this trend requires rapid implementation of climate-smart forest management, with improved and more timely monitoring of GHG fluxes. Alzheimer's disease (AD) is one of the most known causes of dementia which can be characterized by continuous deterioration in the cognitive skills of elderly people. It is a non-reversible disorder that can only be cured if detected early, which is known as mild cognitive impairment (MCI). The most common biomarkers to diagnose AD are structural atrophy and accumulation of plaques and tangles, which can be detected using magnetic resonance imaging (MRI) and positron emission tomography (PET) scans. Therefore, the present paper proposes wavelet transform-based multimodality fusion of MRI and PET scans to incorporate structural and metabolic information for the early detection of this life-taking neurodegenerative disease. Further, the deep learning model, ResNet-50, extracts the fused images' features. The random vector functional link (RVFL) with only one hidden layer is used to classify the extracted features. The weights and biases of the original RVFL network are being optimized by using an evolutionary algorithm to get optimum accuracy. All the experiments and comparisons are performed over the publicly available Alzheimer's Disease Neuroimaging Initiative (ADNI) dataset to demonstrate the suggested algorithm's efficacy.

Schizophrenia (SCZ) is a devastating mental condition with significant negative consequences for patients, making correct and prompt diagnosis crucial. The purpose of this study is to use structural magnetic resonance image (MRI) to better classify individuals with SCZ from control normals (CN) and to locate a region of the brain that represents abnormalities associated with SCZ. Deep learning (DL), which is based on the nervous system, could be a very useful tool for doctors to accurately predict, diagnose, and treat SCZ. Gray Matter (GM), Cerebrospinal Fluid (CSF), and White Matter (WM) brain regions are extracted from 99 MRI images obtained from the open-source OpenNeuro database to demonstrate SCZ's regional relationship. In this paper, we use a pretrained ResNet-50 deep network to extract features from MRI images and an ensemble deep random vector functional link (edRVFL) network to classify those features. By examining the results obtained, the edRVFL deep model provides the highest classification accuracy of 96.5% with WM and is identified as the best-performing algorithm compared to the traditional algorithms. Furthermore, we examined the GM, WM, and CSF tissue volumes in CN subjects and SCZ patients using voxel-based morphometry (VBM), and the results show 1363 significant voxels, 6.90 T-value, and 6.21 Z-value in the WM region of SCZ patients. In SCZ patients, WM is most closely linked to structural alterations, as evidenced by VBM analysis and the DL model.

Background: The contribution of EU forests to climate change mitigation in 2021-2025 is assessed through the Forest Reference Levels (FRLs). The FRL is a projected country-level benchmark of net greenhouse gas emissions against which the future net emissions will be compared. The FRL models the hypothetical development of EU forest carbon sink if the historical management practices were continued, taking into account age dynamics. The Member States' FRLs have been recently adopted by the European Commission with the delegated Regulation (EU) 2021/268 amending the Regulation (EU) 2018/841. Considering the complexity of interactions between forest growth, management and carbon fluxes, there is a need to understand uncertainties linked to the FRL determination., Results: We assessed the methodologies behind the modelled FRLs and evaluated the foreseen impact of continuation of management practices and age dynamics on the near-future EU27 + UK forest carbon sink. Most of the countries implemented robust modelling approaches for simulating management practices and age dynamics within the FRL framework, but faced several challenges in ensuring consistency with historical estimates. We discuss that the projected 16% increase in harvest in 2021-2025 compared to 2000-2009, mostly attributed to age dynamics, is associated to a decline of 18% of forest sink (26% for living biomass only)., Conclusions: We conclude that the FRL exercise was challenging but improved the modelling capacity and data availability at country scale. Forests provide a series of ecosystem services that are crucial to our society. In the European Union (EU), forests account for approximately 38% of the total land surface 1 . These forests are important carbon sinks, and their conservation efforts are vital for the EU's vision of achieving climate neutrality by 2050 2 . However, the increasing demand for forest services and products, driven by the bioeconomy, poses challenges for sustainable forest management. Here we use fine-scale satellite data to observe an increase in the harvested forest area (49 per cent) and an increase in biomass loss (69 per cent) over Europe for the period of 2016-2018 relative to 2011-2015, with large losses occurring on the Iberian Peninsula and in the Nordic and Baltic countries. Satellite imagery further reveals that the average patch size of harvested area increased by 34 per cent across Europe, with potential effects on biodiversity, soil erosion and water regulation. The increase in the rate of forest harvest is the result of the recent expansion of wood markets, as suggested by econometric indicators on forestry, wood-based bioenergy and international trade. If such a high rate of forest harvest continues, the post-2020 EU vision of forest-based climate mitigation may be hampered, and the additional carbon losses from forests would require extra emission reductions in other sectors in order to reach climate neutrality by 2050 3 .

The electrophile-electrophile cross-coupling of carboxylic acid derivatives and alkylpyridinium salts via C-N bond cleavage is developed. The method is distinguished by its simplicity and steers us through a variety of functionalized ketones in good to excellent yields. Besides acid chlorides, carboxylic acids were also employed as acylating agents, which enabled us to incorporate acid-sensitive functional groups such as MOM, BOC, and acetal. Control experiments with TEMPO revealed a radical pathway.

A simple and efficient iron-catalyzed protodehalogenation of alkyl and aryl halides using phenylhydrosilane is disclosed. The reaction utilizes FeCl3 without the requirement of ligands. Unactivated alkyl and aryl halides were successfully reduced in good yields; sterically hindered tertiary halides were also reduced including the less reactive chlorides. The scalability of this methodology was demonstrated by a gram-scale synthesis with a catalyst loading as low as 0.5 mol%. Notably, disproportionation of phenylsilane leads to diphenylsilane that further reduces the halides. Preliminary mechanistic studies revealed a non-radical pathway and the source of hydrogen is PhSiH3via deuterium labeling studies. Our methodology represents simplicity and provides a good alternative to typical tin, aluminum and boron hydride reagents.

Background: The credibility and effectiveness of country climate targets under the Paris Agreement requires that, in all greenhouse gas (GHG) sectors, the accounted mitigation outcomes reflect genuine deviations from the type and magnitude of activities generating emissions in the base year or baseline. This is challenging for the forestry sector, as the future net emissions can change irrespective of actual management activities, because of age-related stand dynamics resulting from past management and natural disturbances. The solution implemented under the Kyoto Protocol (2013-2020) was accounting mitigation as deviation from a projected (forward-looking) "forest reference level", which considered the age-related dynamics but also allowed including the assumed future implementation of approved policies. This caused controversies, as unverifiable counterfactual scenarios with inflated future harvest could lead to credits where no change in management has actually occurred, or conversely, failing to reflect in the accounts a policy-driven increase in net emissions. Instead, here we describe an approach to set reference levels based on the projected continuation of documented historical forest management practice, i.e. reflecting age-related dynamics but not the future impact of policies. We illustrate a possible method to implement this approach at the level of the European Union (EU) using the Carbon Budget Model., Results: Using EU country data, we show that forest sinks between 2013 and 2016 were greater than that assumed in the 2013-2020 EU reference level under the Kyoto Protocol, which would lead to credits of 110-120 Mt CO 2 /year (capped at 70-80 Mt CO 2 /year, equivalent to 1.3% of 1990 EU total emissions). By modelling the continuation of management practice documented historically (2000-2009), we show that these credits are mostly due to the inclusion in the reference levels of policy-assumed harvest increases that never materialized. With our proposed approach, harvest is expected to increase (12% in 2030 at EU-level, relative to 2000-2009), but more slowly than in current forest reference levels, and only because of age-related dynamics, i.e. increased growing stocks in maturing forests., Conclusions: Our science-based approach, compatible with the EU post-2020 climate legislation, helps to ensure that only genuine deviations from the continuation of historically documented forest management practices are accounted toward climate targets, therefore enhancing the consistency and comparability across GHG sectors. It provides flexibility for countries to increase harvest in future reference levels when justified by age-related dynamics. It offers a policy-neutral solution to the polarized debate on forest accounting (especially on bioenergy) and supports the credibility of forest sector mitigation under the Paris Agreement.

Background: This study aimed to assess bond strength of the resin/bracket interface, under in-vitro shear stress, of metal brackets recycled by different clinical protocols., Methods: Sixty stainless steel orthodontic brackets were bonded on acrylic resin. The Transbond XT™ resin was applied at the base of the bracket aided by a matrix, obtaining 1 mm of thickness, and photoactivated with a LED device (40 s; 500 mW/cm2). Samples were randomly divided into four groups (N.=15) according to the reconditioning/recycling protocol: aluminum oxide (AO) 90 µm; hydrofluoric acid 60 s (HA60); hydrofluoric acid 120 s (HA120); hydrofluoric acid 60 s + silane (HA60S). After recycling, the resin was applied at the base of the bracket for shear testing in a universal testing machine (0.5 mm/min). After reconditioning/recycling, the surfaces were analyzed by Scanning Electron Microscopy. Data obtained after the shear test were subjected to ANOVA and Tukey's test (P<0.05)., Results: The AO group presented higher values of shear bond strength compared to the other reconditioning/recycling protocols (P<0.05). The HA120 and HA60S groups presented statistically similar results, but HA120 presented strength below the recommended limit., Conclusions: The recycling technique by aluminum oxide sandblasting was more effective for reconditioning orthodontic brackets when compared to the other protocols. The reconditioning technique with 10% hydrofluoric acid followed by the application of silane bonding agent may be used as an alternative protocol.

Background: Forests and the forest sector may play an important role in mitigating climate change. The Paris Agreement and the recent legislative proposal to include the land use sector in the EU 2030 climate targets reflect this expectation. However, greater confidence on estimates from national greenhouse gas inventories (GHGI) and more comprehensive analyses of mitigation options are needed to seize this mitigation potential. The aim of this paper is to provide a tool at EU level for verifying the EU GHGI and for simulating specific policy and forest management scenarios. Therefore, the Carbon Budget Model (CBM) was applied for an integrated assessment of the EU forest carbon (C) balance from 2000 to 2012, including: (i) estimates of the C stock and net CO 2 emissions for forest management (FM), afforestation/reforestation (AR) and deforestation (D), covering carbon in both the forest and the harvest wood product (HWP) pools; (ii) an overall analysis of the C dynamics associated with harvest and natural disturbances (mainly storms and fires); (iii) a comparison of our estimates with the data reported in the EU GHGI., Results: Overall, the average annual FM sink (-365 Mt CO 2 year -1 ) estimated by the CBM in the period 2000-2012 corresponds to about 7 % of total GHG emissions at the EU level for the same period (excluding land use, land-use change and forestry). The HWP pool sink (-44 Mt CO 2 year -1 ) contributes an additional 1 %. Emissions from D (about 33 Mt CO 2 year -1 ) are more than compensated by the sink in AR (about 43 Mt CO 2 year -1 over the period). For FM, the estimates from the CBM were about 8 % lower than the EU GHGI, a value well within the typical uncertainty range of the EU forest sink estimates. For AR and D the match with the EU GHGI was nearly perfect (difference <±2 % in the period 2008-2012). Our analysis on harvest and natural disturbances shows that: (i) the impact of harvest is much greater than natural disturbances but, because of salvage logging (often very relevant), the impact of natural disturbances is often not easily distinguishable from the impact of harvest, and (ii) the impact of storms on the biomass C stock is 5-10 times greater than fires, but while storms cause only indirect emissions (i.e., a transfer of C from living biomass to dead organic matter), fires cause both direct and indirect emissions., Conclusions: This study presents the application of a consistent methodological approach, based on an inventory-based model, adapted to the forest management conditions of EU countries. The approach captures, with satisfactory detail, the C sink reported in the EU GHGI and the country-specific variability due to harvest, natural disturbances and land-use changes. To our knowledge, this is the most comprehensive study of its kind at EU level, i.e., including all the forest pools, HWP and natural disturbances, and a comparison with the EU GHGI. The results provide the basis for possible future policy-relevant applications of this model, e.g., as a tool to support GHGIs (e.g., on accounting for natural disturbances) and to verify the EU GHGI, and for the simulation of specific scenarios at EU level.

Background: According to the post-2012 rules under the Kyoto protocol, developed countries that are signatories to the protocol have to estimate and report the greenhouse gas (GHG) emissions and removals from forest management (FM), with the option to exclude the emissions associated to natural disturbances, following the Intergovernmental Panel on Climate Change (IPCC) guidelines. To increase confidence in GHG estimates, the IPCC recommends performing verification activities, i.e. comparing country data with independent estimates. However, countries currently conduct relatively few verification efforts. The aim of this study is to implement a consistent methodological approach using the Carbon Budget Model (CBM) to estimate the net CO 2 emissions from FM in 26 European Union (EU) countries for the period 2000-2012, including the impacts of natural disturbances. We validated our results against a totally independent case study and then we compared the CBM results with the data reported by countries in their 2014 Greenhouse Gas Inventories (GHGIs) submitted to the United Nations Framework Convention on Climate Change (UNFCCC)., Results: The match between the CBM results and the GHGIs was good in nine countries (i.e. the average of our results is within ±25 % compared to the GHGI and the correlation between CBM and GHGI is significant at P < 0.05) and partially good in ten countries. When the comparison was not satisfactory, in most cases we were able to identify possible reasons for these discrepancies, including: (1) a different representation of the interannual variability, e.g. where the GHGIs used the stock-change approach; (2) different assumptions for non-biomass pools, and for CO 2 emissions from fires and harvest residues. In few cases, further analysis will be needed to identify any possible inappropriate data used by the CBM or problems in the GHGI. Finally, the frequent updates to data and methods used by countries to prepare GHGI makes the implementation of a consistent modeling methodology challenging., Conclusions: This study indicates opportunities to use the CBM as tool to assist countries in estimating forest carbon dynamics, including the impact of natural disturbances, and to verify the country GHGIs at the EU level, consistent with the IPCC guidelines. A systematic comparison of the CBM with the GHGIs will certainly require additional efforts-including close cooperation between modelers and country experts. This approach should be seen as a necessary step in the process of continuous improvement of GHGIs, because it may help in identifying possible errors and ultimately in building confidence in the estimates reported by the countries.

Background: The new rules for the Land Use, Land Use Change and Forestry sector under the Kyoto Protocol recognized the importance of Harvested Wood Products (HWP) in climate change mitigation. We used the Tier 2 method proposed in the 2013 IPCC KP Supplement to estimate emissions and removals from HWP from 1990 to 2030 in EU-28 countries with three future harvest scenarios (constant historical average, and +/-20% in 2030)., Results: For the historical period (2000-2012) our results are consistent with other studies, indicating a HWP sink equal on average to -44.0 Mt CO 2 yr -1 (about 10% of the sink by forest pools). Assuming a constant historical harvest scenario and future distribution of the total harvest among each commodity, the HWP sink decreases to -22.9 Mt CO 2 yr -1 in 2030. The increasing and decreasing harvest scenarios produced a HWP sink of -43.2 and -9.0 Mt CO 2 yr -1 in 2030, respectively. Other factors may play an important role on HWP sink, including: (i) the relative share of different wood products, and (ii) the combined effect of production, import and export on the domestic production of each commodity., Conclusions: Maintaining a constant historical harvest, the HWP sink will slowly tend to saturate, i.e. to approach zero in the long term. The current HWP sink will be maintained only by further increasing the current harvest; however, this will tend to reduce the current sink in forest biomass, at least in the short term. Overall, our results suggest that: (i) there is limited potential for additional HWP sink in the EU; (ii) the HWP mitigation potential should be analyzed in conjunction with other mitigation components (e.g. sink in forest biomass, energy and material substitution by wood).

Background: Withania somnifera is an herbal medicine that has been known to possess memory-enhancing properties. The current study involved an assessment of cognitive and psychomotor effects of Withania somnifera extract in healthy human participants., Materials and Methods: In this prospective, double-blind, multi-dose, placebo-controlled, crossover study, 20 healthy male participants were randomized to receive 250 mg two capsules twice daily of an encapsulated dried aqueous extract of roots and leaves of Withania somnifera or a matching placebo for a period of 14 days. Cognitive and psychomotor performance was assessed pre-dose (day 1) and at 3 hrs post-dose on day 15 using a battery of computerized psychometric tests. After a washout period of 14 days, the subjects crossed-over to receive the other treatment for a further period of 14 days as per prior randomization schedule. Same battery of test procedures were performed to assess cognitive and psychomotor performance., Results: Significant improvements were observed in reaction times with simple reaction, choice discrimination, digit symbol substitution, digit vigilance, and card sorting tests with Withania somnifera extract compared to placebo. However, no effect can be seen with the finger tapping test., Conclusion: These results suggest that Withania somnifera extract can improve cognitive and psychomotor performance and may, therefore, be a valuable adjunct in the treatment of diseases associated with cognitive impairment.

Background: Acute and chronic stress is a risk factor for the development and progression of coronary artery disease. Increased arterial stiffness is an independent marker for cardiovascular disease. Cold pressor test (CPT) is known to be associated with substantial activation of the autonomic nervous system., Objective: The aim of this study was to evaluate the effect of Phyllanthus emblica extract on cold pressor stress test induced changes on cardiovascular parameters and aortic wave reflections in healthy human subjects., Materials and Methods: This was a double-blind, placebo-controlled, crossover study. Participants were randomized to receive either two capsules of P. emblica extract 250 mg (containing aqueous extract of P. emblica, highly standardized by high-performance liquid chromatography to contain low molecular weight hydrolysable tannins emblicanin-A, emblicanin-B, pedunculagin and punigluconin) or two capsules of placebo twice daily for 14 days. Pharmacodynamic parameters such as heart rate, augmentation pressure, augmentation index (AIx), subendocardial viability ratio (SEVR), radial and aortic blood pressure (BP) were recorded before and after CPT at baseline and end of treatment. After washout period of 14 days, subjects crossed over to the other treatment and the same test procedure was repeated again. Safety assessments were done at baseline and at the end of treatment., Results: A total of 12volunteers completed the study. Compared with baseline and placebo, P. emblica extract produced a significant decrease of mean percent change in the indices of arterial stiffness (AIx, radial and aortic BP) and increase in SEVR, an index of myocardial perfusion with CPT. Both treatments were well-tolerated and no serious adverse events were reported., Conclusion: Proprietary P. emblica extract, showed a significant decrease in cold pressor stress test induced changes on aortic wave reflections.

Objective: To evaluate the effects of centrally active drugs using a new indigenously developed automated psychometric test system and compare the results with that obtained using pencil- and paper-based techniques., Materials and Methods: The tests were standardized in 24 healthy participants. Reproducibility of the test procedure was evaluated by performing the tests by a single experimenter on two occasions (interday reproducibility). To evaluate the sensitivity of the tests, the effects of zolpidem (5 mg) and caffeine (500 mg) versus placebo were studied in 24 healthy participants in a randomized, double-blind three-way crossover design., Results: Psychometric tests were performed at baseline and at 1, 2, and 3 h after administration of study medication. The effects of zolpidem and caffeine on the psychomotor performance were most pronounced 1 h after administration. At this time, a significant impairment of performance in the simple reaction test (SRT), choice discrimination test (CDT), digit symbol substitution test (DSST), digit vigilance test (DVT), and card sorting test (CST) was observed with zolpidem. In contrast, caffeine showed a significant improvement in performance in CDT and DVT only., Conclusion: The results suggest that the tests of the computerized system are more sensitive and reliable then the pencil and paper tests in detecting the effects of central acting agents and are suitable for use in clinical areas to conduct studies with patients.

Background: The Subjective Well-Being Under Neuroleptic Treatment Scale short form (SWN-K) is a self-rating scale developed to measure mentally ill patients' well-being under the antipsychotic drug treatment. This paper reports on adaptation and psychometric properties of the instrument in an Estonian psychiatric sample., Methods: In a naturalistic study design, 124 inpatients or outpatients suffering from the first psychotic episode or chronic psychotic illness completed the translated SWN-K instrument. Item content analysis, internal consistency analysis, exploratory principal components analysis, and confirmatory factor analysis were used to construct the Estonian version of the SWN-K (SWN-K-E). Additionally, socio-demographic and clinical data, observer-rated psychopathology, medication side effects, daily antipsychotic drug dosages, and general functioning were assessed at two time points, at baseline and after a 29-week period; the associations of the SWN-K-E scores with these variables were explored., Results: After having selected 20 items for the Estonian adaptation, the internal consistency of the total SWN-K-E was 0.93 and the subscale consistencies ranged from 0.70 to 0.80. Good test-retest reliabilities were observed for the adapted scale scores, with the correlation of the total score over about 6 months being r = 0.70. Confirmatory factor analysis replicated the presence of a higher-order factor (general well-being) and five first-order factors (mental functioning, physical functioning, social integration, emotional regulation, and self-control); the model fitted the data well. The results indicated a moderate-high correlations r = 0.54 between the SWN-K-E total score and the evaluation how satisfied patients were with their lives in generally. No significant correlations were found between the overall subjective well-being score and age, severity of the psychopathology, drug adverse effects, or prescribed drug dosage., Conclusion: Taken together, the results demonstrated that the Estonian version of the SWN-K is a reliable and valid instrument with psychometric properties similar to the original English version. The potential uses of the scale in both research and clinical settings are considered.

Background: The Stroop paradigm evaluates susceptibility to interference and is sensitive to dysfunction in frontal lobes and drug effects. The aim of the present study was to establish a simple and reliable computerized version of Stroop color-word test, which can be used for screening of various psychotropic drugs., Materials and Methods: The standardized method was followed in all cases, by recording the reaction time (RT) in msec in 24 healthy participants using computerized version of Stroop color-word test. Reproducibility of the test procedure was evaluated by recording the RTs by a single experimenter on two sessions (interday reproducibility). Validity of the model was further tested by evaluating the psychotropic effect of Zolpidem 5 mg, Caffeine 500 mg, or Placebo on 24 healthy subjects in a randomized, double blind three-way crossover design., Results: The method was found to produce low variability with coefficient of variation less than 10%. Interday reproducibility was very good as shown by Bland-Altman plot with most of the values within ±2SD. There was a significant increase in RTs in Stroop performance with Zolpidem at 1 hr and 2 hrs; in contrast, caffeine significantly decreased RTs in Stroop performance at 1 hr only compared to placebo., Conclusion: The Stroop color-word recording and analysis system is simple, sensitive to centrally acting drug effects, and has potential for future experimental psychomotor assessment studies.
