Abstract: We empirically test the relation between the SFR(LIR) derived from the infrared luminosity, LIR, and the SFR(Ha) derived from the Ha emission line luminosity using simple conversion relations. We use a sample of 474 galaxies at z = 0.06 - 0.46 with both Ha detection (from 20k zCOSMOS survey) and new far-IR Herschel data (100 and 160 {\mu}m). We derive SFR(Ha) from the Ha extinction corrected emission line luminosity. We find a very clear trend between E(B - V) and LIR that allows to estimate extinction values for each galaxy even if the Ha emission line measurement is not reliable. We calculate the LIR by integrating from 8 up to 1000 {\mu}m the SED that is best fitting our data. We compare SFR(Ha) with the SFR(LIR). We find a very good agreement between the two SFR estimates, with a slope of m = 1.01 \pm 0.03 in the SFR(LIR) vs SFR(Ha) diagram, a normalization constant of a = -0.08 \pm 0.03 and a dispersion of sigma = 0.28 dex.We study the effect of some intrinsic properties of the galaxies in the SFR(LIR)-SFR(Ha) relation, such as the redshift, the mass, the SSFR or the metallicity. The metallicity is the parameter that affects most the SFR comparison. The mean ratio of the two SFR estimators log[SFR(LIR)/SFR(Ha)] varies by approx. 0.6 dex from metal-poor to metal-rich galaxies (8.1 < log(O/H) + 12 < 9.2). This effect is consistent with the prediction of a theoretical model for the dust evolution in spiral galaxies. Considering different morphological types, we find a very good agreement between the two SFR indicators for the Sa, Sb and Sc morphologically classified galaxies, both in slope and normalization. For the Sd, irregular sample (Sd/Irr), the formal best-fit slope becomes much steeper (m = 1.62 \pm 0.43), but it is still consistent with 1 at the 1.5 sigma level, because of the reduced statistics of this sub-sample.

Abstract: We set out to determine the ratio, q(IR), of rest-frame 8-1000um flux, S(IR), to monochromatic radio flux, S(1.4GHz), for galaxies selected at far-IR and radio wavelengths, to search for signs that the ratio evolves with redshift, luminosity or dust temperature, and to identify any far-IR-bright outliers - useful laboratories for exploring why the far-IR/radio correlation is generally so tight when the prevailing theory suggests variations are almost inevitable. We use flux-limited 250-um and 1.4-GHz samples, obtained in GOODS-N using Herschel (HerMES; PEP) and the VLA. We determine bolometric IR output using ten bands spanning 24-1250um, exploiting data from PACS and SPIRE, as well as Spitzer, SCUBA, AzTEC and MAMBO. We also explore the properties of an L(IR)-matched sample, designed to reveal evolution of q(IR) with z, spanning log L(IR) = 11-12 L(sun) and z=0-2, by stacking into the radio and far-IR images. For 1.4-GHz-selected galaxies, we see tentative evidence of a break in the flux ratio, q(IR), at L(1.4GHz) ~ 10^22.7 W/Hz, where AGN are starting to dominate the radio power density, and of weaker correlations with z and T(d). From our 250-um-selected sample we identify a small number of far-IR-bright outliers, and see trends of q(IR) with L(1.4GHz), L(IR), T(d) and z, noting that some of these are inter-related. For our L(IR)-matched sample, there is no evidence that q(IR) changes significantly as we move back into the epoch of galaxy formation: we find q(IR) goes as (1+z)^gamma, where gamma = -0.04 +/- 0.03 at z=0-2; however, discounting the least reliable data at z1., A&A Herschel Special Issue, in press as a Letter. 5 pages

Abstract: The influence of stellar metallicity on stellar coronal properties is still unclear, since most of the stars with well‐studied coronal spectra have photospheric abundances similar to (or assumed to be) the solar photospheric values. We have used XMM‐Newton to observe 11 LMi, a known X‐ray source, which has supersolar (2.0–2.5 times solar) photospheric abundances, in order to study the effect of the high underlying intrinsic metallicity on the temperature structure and abundances of its coronal plasma. We present and discuss the EPIC spectra of 11 LMi that we have obtained, and compare the properties of its coronal emission with those of similar stars of solar metallicity.

Abstract: Introduction After pressure from patient and clinical groups NICE have reversed their position on biologics 1,2 in subacute ulcerative colitis (UC) and it will be available to patients in 2015. Method A retrospective review of the use of infliximab for the treatment of UC in a single centre DGH between 2011 and 2014. Results Ten patients received infliximab for acute severe UC and ten for subacute UC. In the 12 months pre-infliximab these patients received 40 courses of prednisolone between them (Table 1). All subacute patients had received immunomodulators. All subacute patients had a partial or complete clinical (MAYO score) and biochemical response following infliximab induction, compared to 50% of the acute severe group. Two patients remain on long-term infliximab and in remission. Colectomy was required within 12 months for 55.6% of patients with acute severe disease and 44.4% of those with subacute activity. 60% of the remaining subacute patients (n = 3) have received steroids since infliximab. Conclusion NICE change of position 2 echos current practice in our centre and with the change in guidelines we may hope to see a reduction in steroid use prior to the use of biological treatment. However the long term success of infliximab in UC is limited. Subacute patients showed better immediate outcomes in terms of biochemical and clinical response, but at 12 months, outcomes were similar in both groups. Work is needed to identify the likely responders to make its use cost effective and evidence based guidance is needed on the clinical benefits of switching to alternative biological agents if infliximab fails. Abstract: We use deep 70, 100 and 160 μm observations taken with PACS, the Photodetector Array Camera and Spectrometer on board of Herschel, as part of the PACS Evolutionary Probe (PEP) guaranteed time, to study the relation between star formation rate and environment at redshift ~ 1 in the GOODS-S and GOODS-N fields. We use the SDSS spectroscopic catalog to build the local analog and study the evolution of the star formation activity dependence on the environment. At z ∼ 1 we observe a reversal of the relation between star formation rate and local density, confirming the results based on Spitzer 24 μm data. However, due to the high accuracy provided by PACS in measuring the star formation rate also for AGN hosts, we identify in this class of objects the cause for the reversal of the density-SFR relation. Indeed, AGN hosts favor high stellar masses, dense regions and high star formation rates. Without the AGN contribution the relation flattens consistently with respect to the local analog in the same range of star formation rates. As in the local universe, the specific star formation rate anti-correlates with the density. This is due to mass segregation both at high and low redshift. The contribution of AGN hosts does not affect this anti-correlation, since AGN hosts exhibit the same specific star formation rate as star forming galaxies at the same mass. The same global trends and AGN contribution is observed once the relations are studied per morphological type. We study the specific star formation rate vs. stellar mass relation in three density regimes. Our data provides an indication that at M/M⊙ > 1011 the mean specific star formation rate tends to be higher at higher density, while the opposite trend is observed in the local SDSS star forming sample. © 2011 ESO.

Abstract: The Herschel Space Observatory enables us to accurately measure the bolometric output of starburst galaxies and active galactic nuclei (AGN) by directly sampling the peak of their far-infrared (IR) emission. Here we examine whether the spectral energy distribution (SED) and dust temperature of galaxies have strongly evolved since z~2.5. We use Herschel deep extragalactic surveys from 100 to 500um to compute total IR luminosities in galaxies down to the faintest levels, using PACS and SPIRE in the GOODS-North field (PEP and HerMES key programs). We show that measurements in the SPIRE bands can be used below the statistical confusion limit if information at higher spatial resolution is used to identify isolated galaxies whose flux is not boosted by bright neighbors. Below z~1.5, mid-IR extrapolations are correct for star-forming galaxies with a dispersion of only 40% (0.15dex), therefore similar to z~0 galaxies. This narrow distribution is puzzling when considering the range of physical processes that could have affected the SED of these galaxies. Extrapolations from only one of the 160um, 250um or 350um bands alone tend to overestimate the total IR luminosity. This may be explained by the lack of far-IR constraints around and above ~150um (rest-frame) on local templates. We also note that the dust temperature of luminous IR galaxies around z~1 is mildly colder by 10-15% than their local analogs and up to 20% for ULIRGs at z~1.6. Above z=1.5, distant galaxies are found to exhibit a substantially larger mid- over far-IR ratio, which could either result from stronger broad emission lines or warm dust continuum heated by a hidden AGN. Two thirds of the AGNs identified in the field with a measured redshift exhibit the same behavior as purely star-forming galaxies. Abstract: We exploit the deep and extended far infrared data sets (at 70, 100 and 160 um) of the Herschel GTO PACS Evolutionary Probe (PEP) Survey, in combination with the HERschel Multi tiered Extragalactic Survey (HerMES) data at 250, 350 and 500 um, to derive the evolution of the restframe 35 um, 60 um, 90 um, and total infrared (IR) luminosity functions (LFs) up to z~4. We detect very strong luminosity evolution for the total IR LF combined with a density evolution. In agreement with previous findings, the IR luminosity density increases steeply to z~1, then flattens between z~1 and z~3 to decrease at z greater than 3. Galaxies with different SEDs, masses and sSFRs evolve in very different ways and this large and deep statistical sample is the first one allowing us to separately study the different evolutionary behaviours of the individual IR populations contributing to the IR luminosity density. Galaxies occupying the well established SFR/stellar mass main sequence (MS) are found to dominate both the total IR LF and luminosity density at all redshifts, with the contribution from off MS sources (0.6 dex above MS) being nearly constant (~20% of the total IR luminosity density) and showing no significant signs of increase with increasing z over the whole 0.8, Comment: 32 pages, 20 figures, 9 tables. Abstract: By making use of Herschel-PEP observations of the COSMOS and Extended Groth Strip fields, we have estimated the dependence of the clustering properties of FIR-selected sources on their 100um fluxes. Our analysis shows a tendency for the clustering strength to decrease with limiting fluxes: r0(S100um >8 mJy)~4.3 Mpc and r0(S100um >5 mJy)~5.8 Mpc. These values convert into minimum halo masses Mmin~10^{11.6} Msun for sources brighter than 8 mJy and Mmin~10^{12.4} Msun for S100um > 5 mJy galaxies. We show such an increase of the clustering strength to be due to an intervening population of z~2 sources, which are very strongly clustered and whose relative contribution, equal to about 10% of the total counts at S100um > 2 mJy, rapidly decreases for brighter flux cuts. By removing such a contribution, we find that z ~100 Msun/yr). Our analysis shows that the same amount of (intense) star forming activity takes place in extremely different environments at the different cosmological epochs. Abstract: Gravitational lensing by massive galaxy clusters allows study of the population of intrinsically faint infrared galaxies that lie below the sensitivity and confusion limits of current infrared and submillimeter telescopes. We present ultra-deep PACS 100 and 160 microns observations toward the cluster lens Abell 2218, to penetrate the Herschel confusion limit. We derive source counts down to a flux density of 1 mJy at 100 microns and 2 mJy at 160 microns, aided by strong gravitational lensing. At these levels, source densities are 20 and 10 beams/source in the two bands, approaching source density confusion at 160 microns. The slope of the counts below the turnover of the Euclidean-normalized differential curve is constrained in both bands and is consistent with most of the recent backwards evolutionary models. By integrating number counts over the flux range accessed by Abell 2218 lensing (0.94-35 mJy at 100 microns and 1.47-35 mJy at 160 microns, we retrieve a cosmic infrared background (CIB) surface brightness of ~8.0 and ~9.9 nW m^-2 sr^-1, in the respective bands. These values correspond to 55% (+/- 24%) and 77% (+/- 31%) of DIRBE direct measurements. Combining Abell 2218 results with wider/shallower fields, these figures increase to 62% (+/- 25%) and 88% (+/- 32%) CIB total fractions, resolved at 100 and 160 microns, disregarding the high uncertainties of DIRBE absolute values., Accepted for publication on the A&A Herschel Special Issue, final version

Abstract: Quasi-stellar objects (QSOs) occur in galaxies in which supermassive black holes (SMBHs) are growing substantially through rapid accretion of gas. Many popular models of the co-evolutionary growth of galaxies and SMBHs predict that QSOs are also sites of substantial recent star formation, mediated by important processes, such as major mergers, which rapidly transform the nature of galaxies. A detailed study of the star-forming properties of QSOs is a critical test of such models. We present a far-infrared Herschel/PACS study of the mean star formation rate (SFR) of a sample of spectroscopically observed QSOs to z~2 from the COSMOS extragalactic survey. This is the largest sample to date of moderately luminous AGNs studied using uniform, deep far-infrared photometry. We study trends of the mean SFR with redshift, black hole mass, nuclear bolometric luminosity and specific accretion rate (Eddington ratio). To minimize systematics, we have undertaken a uniform determination of SMBH properties, as well as an analysis of important selection effects within spectroscopic QSO samples that influence the interpretation of SFR trends. We find that the mean SFRs of these QSOs are consistent with those of normal massive star-forming galaxies with a fixed scaling between SMBH and galaxy mass at all redshifts. No strong enhancement in SFR is found even among the most rapidly accreting systems, at odds with several co-evolutionary models. Finally, we consider the qualitative effects on mean SFR trends from different assumptions about the star-forming properties of QSO hosts and redshift evolution of the SMBH-galaxy relationship. Abstract: In the local Universe, galaxy properties show a strong dependence on environment. In cluster cores, early type galaxies dominate, whereas star-forming galaxies are more and more common in the outskirts. At higher redshifts and in somewhat less dense environments (e.g. galaxy groups), the situation is less clear. One open issue is that of whether and how the star formation rate (SFR) of galaxies in groups depends on the distance from the centre of mass. To shed light on this topic, we have built a sample of X-ray selected galaxy groups at 010^10.3 M_sun in order to have a high spectroscopic completeness. As we use only spectroscopic redshifts, our results are not affected by uncertainties due to projection effects. We use several SFR indicators to link the star formation (SF) activity to the galaxy environment. Taking advantage of the extremely deep mid-infrared Spitzer MIPS and far-infrared Herschel PACS observations, we have an accurate, broad-band measure of the SFR for the bulk of the star-forming galaxies. We use multi-wavelength SED fitting techniques to estimate the stellar masses of all objects and the SFR of the MIPS and PACS undetected galaxies. We analyse the dependence of the SF activity, stellar mass and specific SFR on the group-centric distance, up to z~1.6, for the first time. We do not find any correlation between the mean SFR and group-centric distance at any redshift. We do not observe any strong mass segregation either, in agreement with predictions from simulations. Our results suggest that either groups have a much smaller spread in accretion times with respect to the clusters and that the relaxation time is longer than the group crossing time., Comment: Accepted for publication in MNRAS

Abstract: This is the first paper of a series aiming at investigating galaxy formation and evolution in the giant-void class of the Lemaitre-Tolman-Bondi (LTB) models that best fits current cosmological observations. Here we investigate the Luminosity Function (LF) methodology, and how its estimates would be affected by a change on the cosmological model assumed in its computation. Are the current observational constraints on the allowed Cosmology enough to yield robust LF results? We use the far-infrared source catalogues built on the observations performed with the Herschel/PACS instrument, and selected as part of the PACS evolutionary probe (PEP) survey. Schechter profiles are obtained in redshift bins up to z approximately 4, assuming comoving volumes in both the standard model, that is, Friedmann-Lemaitre-Robertson-Walker metric with a perfect fluid energy-momentum tensor, and non-homogeneous LTB dust models, parametrized to fit the current combination of results stemming from the observations of supernovae Ia, the cosmic microwave background, and baryonic acoustic oscillations. We find that the luminosity functions computed assuming both the standard model and LTB void models show in general good agreement. However, the faint-end slope in the void models shows a significant departure from the standard model up to redshift 0.4. We demonstrate that this result is not artificially caused by the used LF estimator which turns out to be robust under the differences in matter-energy density profiles of the models. The differences found in the LF slopes at the faint end are due to variation in the luminosities of the sources, which depend on the geometrical part of the model. It follows that either the standard model is over-estimating the number density of faint sources or the void models are under-estimating it., 23 pages, 11 figures. Accepted for publication in A&A

Abstract: We present results from the deepest Herschel-PACS (Photodetector Array Camera and Spectrometer) far-infrared blank field extragalactic survey, obtained by combining observations of the GOODS (Great Observatories Origins Deep Survey) fields from the PACS Evolutionary Probe (PEP) and GOODS-Herschel key programmes. We describe data reduction and the construction of images and catalogues. In the deepest parts of the GOODS-S field, the catalogues reach 3-sigma depths of 0.9, 0.6 and 1.3 mJy at 70, 100 and 160 um, respectively, and resolve ~75% of the cosmic infrared background at 100um and 160um into individually detected sources. We use these data to estimate the PACS confusion noise, to derive the PACS number counts down to unprecedented depths and to determine the infrared luminosity function of galaxies down to LIR=10^11 Lsun at z~1 and LIR=10^12 Lsun at z~2, respectively. For the infrared luminosity function of galaxies, our deep Herschel far-infrared observations are fundamental because they provide more accurate infrared luminosity estimates than those previously obtained from mid-infrared observations. Maps and source catalogues (>3-sigma) are now publicly released. Based on this rich dataset, we reproduce the restframe UV to FIR ten-colors distribution of galaxies using a superposition of multi-variate Gaussian modes. The median SED of each mode is then fitted with a modified version of the MAGPHYS code that combines stellar light, emission from dust heated by stars and a possible warm dust contribution heated by an AGN. The defined Gaussian grouping is also used to identify rare sources. The zoology of outliers includes Herschel-detected ellipticals, very blue z~1 Ly-break galaxies, quiescent spirals, and torus-dominated AGN with star formation. Out of these groups and outliers, a new template library is assembled, consisting of 32 SEDs describing the intrinsic scatter in the restframe UV-to-submm colors of infrared galaxies. This library is tested against L(IR) estimates with and without Herschel data included, and compared to eight other popular methods often adopted in the literature. When implementing Herschel photometry, these approaches produce L(IR) values consistent with each other within a median absolute deviation of 10-20%, the scatter being dominated more by fine tuning of the codes, rather than by the choice of SED templates. Finally, the library is used to classify 24 micron detected sources in PEP GOODS fields. AGN appear to be distributed in the stellar mass (M*) vs. star formation rate (SFR) space along with all other galaxies, regardless of the amount of infrared luminosity they are powering, with the tendency to lie on the high SFR side of the "main sequence". The incidence of warmer star-forming sources grows for objects with higher specific star formation rates (sSFR), and they tend to populate the "off-sequence" region of the M*-SFR-z space., Comment: Accepted for publication in A&A. Abstract: International audience; Using new homogeneous luminosity functions (LFs) in the far-ultraviolet (FUV) from VVDS and in the far-infrared (FIR) from Herschel/PEP and Herschel/HerMES, we studied the evolution of the dust attenuation with redshift. With this information, we were able to estimate the redshift evolution of the total (FUV + FIR) star formation rate density (SFRDTOT). By integrating SFRDTOT, we followed the mass building and analyzed the redshift evolution of the stellar mass density (SMD). This article aims at providing a complete view of star formation from the local Universe to z ~ 4 and, using assumptions on earlier star formation history, compares this evolution with previously published data in an attempt to draw a homogeneous picture of the global evolution of star formation in galaxies. Our main conclusions are that: 1) the dust attenuation AFUV is found to increase from z = 0 to z ~ 1.2 and then starts to decrease until our last data point at z = 3.6; 2) the estimated SFRD confirms published results to z ~ 2. At z > 2, we observe either a plateau or a small increase up to z ~ 3 and then a likely decrease up to z = 3.6; 3) the peak of AFUV is delayed with respect to the plateau of SFRDTOT and a probable origin might be found in the evolution of the bright ends of the FUV and FIR LFs; 4) using assumptions (exponential rise and linear rise with time) for the evolution of the star formation density from z = 3.6 to zform = 10, we integrated SFRDTOT and obtained a good agreement with the published SMDs.

Abstract: We study the dust properties of galaxies in the redshift range 0.10.5 with L_IR>5x10^{10} L_\odot, appears to be 2-5 K colder than that of AKARI-selected local galaxies with similar luminosities; and the dispersion in T_dust for high-z galaxies increases with L_IR due to the existence of cold galaxies that are not seen among local galaxies. Abstract: The star formation rate (SFR) is a key parameter in the study of galaxy evolution. The accuracy of SFR measurements at z ∼ 2 has been questioned following a disagreement between observations and theoretical models. The latter predict SFRs at this redshift that are typically a factor 4 or more lower than the measurements. We present star-formation rates based on calorimetric measurements of the far-infrared (FIR) luminosities for massive 1.5 < z < 2.5, normal star-forming galaxies (SFGs), which do not depend on extinction corrections and/or extrapolations of spectral energy distributions. The measurements are based on observations in GOODS-N with the Photodetector Array Camera and Spectrometer (PACS) onboard Herschel, as part of the PACS evolutionary probe (PEP) project, that resolve for the first time individual SFGs at these redshifts at FIR wavelengths. We compare FIR-based SFRs to the more commonly used 24 μm and UV SFRs. We find that SFRs from 24 μm alone are higher by a factor of ∼4-7.5 than the true SFRs. This overestimation depends on luminosity: gradually increasing for log L(24 μm) > 12.2 L ⊙. The SFGs and AGNs tend to exhibit the same 24 μm excess. The UV SFRs are in closer agreement with the FIR-based SFRs. Using a Calzetti UV extinction correction results in a mean excess of up to 0.3 dex and a scatter of 0.35 dex from the FIR SFRs. The previous UV SFRs are thus confirmed and the mean excess, while narrowing the gap, is insufficient to explain the discrepancy between the observed SFRs and simulation predictions. © 2010 ESO.

24. Donor T cells for CAR T cell therapy.

Abstract: Adoptive cell therapy using patient-derived chimeric receptor antigen (CAR) T cells redirected against tumor cells has shown remarkable success in treating hematologic cancers. However, wider accessibility of cellular therapies for all patients is needed. Manufacture of patient-derived CAR T cells is limited by prolonged lymphopenia in heavily pre-treated patients and risk of contamination with tumor cells when isolating T cells from patient blood rich in malignant blasts. Donor T cells provide a good source of immune cells for adoptive immunotherapy and can be used to generate universal off-the-shelf CAR T cells that are readily available for administration into patients as required. Genome editing tools such as TALENs and CRISPR-Cas9 and non-gene editing methods such as short hairpin RNA and blockade of protein expression are currently used to enhance CAR T cell safety and efficacy by abrogating non-specific toxicity in the form of graft versus host disease (GVHD) and preventing CAR T cell rejection by the host., (© 2022. Here, we describe a protocol for the robust ex vivo expansion of human CD34(+) HSC isolated from umbilical cord blood. The protocol described can efficiently generate large numbers of HSC. We also describe a flow cytometry-based method using high-resolution division tracking to characterize the kinetics of HSC growth and differentiation. Utilizing the guidelines discussed, it is possible for investigators to use this protocol as presented or to modify it for their specific needs.

Abstract: Objectives: Cord blood (CB) has been established to be an alternative source of haematopoietic stem/progenitor cells (HPC) for transplantation. The number of HPC per CB unit is limited, which results in engraftment delay. Ex vivo expansion of HPC improvement must overcome this., Materials and Methods: Flow cytometry was used to extensively phenotype HPC pre- and post-expansion and CFDA-SE staining was used to track cell divisions. The NSG mouse model was employed in transplantation studies to determine long and short term repopulation in human cells. Gene array analysis was used to evaluate signalling pathways regulated following ex vivo expansion of HPC., Results: expansion of CD34(+) HPC impaired their regenerative function. In this xenograft transplantation model we showed that repopulating activity of CB cells declined following expansion. Expanded HPC had delayed engraftment at early and late stages post-transplant. High resolution division tracking revealed that the cultured HPC had reduced expansion and self-renewal probability and increased differentiation rate compared to non-expanded cells. Gene expression analysis exposed significant modulation of a complex network of genes and pathways that normally maintain HPC proliferation and limit their differentiation., Conclusions: The decline in short-term engraftment is consistent with the loss of rapid SCID repopulating ability r(SRA) by expanded CD34(+) CD38(+) cells recently reported. Abstract: Glycogen synthase kinase-3β (GSK-3β) has been identified as an important regulator of stem cell function acting through activation of the wingless (Wnt) pathway. Here, we report that treatment with an inhibitor of GSK-3β, 6-bromoindirubin 3'-oxime (BIO) delayed cell cycle progression by increasing cell cycle time. BIO treatment resulted in the accumulation of late dividing cells enriched with primitive progenitor cells retaining the ability for sustained proliferation. In vivo analysis using a Non-obese diabetic/severe combined immunodeficient (NOD/SCID) transplantation model has demonstrated that pretreatment with BIO promotes engraftment of ex vivo-expanded hematopoietic stem cells. BIO enhanced the engraftment of myeloid, lymphoid and primitive stem cell compartments. Limiting dilution analysis of SCID repopulating cells (SRC) revealed that BIO treatment increased human chimerism without increasing SRC frequency. Clonogenic analysis of human cells derived from the bone marrow of transplant recipient mice demonstrated that a higher level of human chimerism and cellularity was related to increased regeneration per SRC unit. Gene expression analysis showed that treatment with BIO did not modulate the expression of canonical Wnt target genes upregulated during cytokine-induced cell proliferation. BIO increased the expression of several genes regulating Notch and Tie2 signaling downregulated during ex vivo expansion, suggesting a role in improving stem cell engraftment. In addition, treatment with BIO upregulated CDK inhibitor p57 and downregulated cyclin D1, providing a possible mechanism for the delay seen in cell cycle progression. Abstract: We have developed a fusion protein (CBD-LG) incorporating a cellulose-binding domain and an antibody binding domain, protein LG, to provide an adaptor molecule for cell separation with regenerated cellulose hollow fiber arrays. A single hollow fiber cell adhesion assay utilizing a CD34+ cell line, KG1a, was used to investigate whether ligand affinity interactions were strong enough for cell attachment and separation. CBD-LG efficiently captured CD34+ cells labeled with the mouse IgG2a monoclonal antibody MHCD3400. However, it was not possible to bind CD34+ cells labeled with an IgG1 antibody (HPCA-2). The low affinity of HPCA-2 for LG was overcome by secondary antibodies: KG1a cells that were dual labeled with HPCA-2 followed by rat anti-mouse IgG1 adhered inside hollow fibers coated with CBD-LG. Alternatively, immobilized rabbit polyclonal anti-mouse IgG1 captured cells labeled with HPCA-2. The development of an adaptor molecule to display recombinant domains at the surface of hollow fibers will be an effective tool to investigate cellular ligand-receptor interactions, a necessary step in the development of hollow fiber bioreactors for manufacture of human cellular products.

Abstract: The cohesin-dockerin interaction, which is responsible for the formation of the cellulosome complex of cellulolytic bacteria, is a calcium-dependent, high affinity interaction. In this study, the cohesin (Cip7) and dockerin (Doc) domains of Clostridium thermocellum were fused to the cellulose-binding domain (CBD) of C. cellulovorans and the antibody-binding domain, protein LG, respectively, to form CBD-Cip7 and LG-Doc. Immobilised CBD-Cip7 was able to bind LG-Doc and subsequently antibody as determined using surface plasmon resonance. Binding was reversed by the removal of Ca2+ with EDTA. The dockerin containing fusion protein was affinity purified using an immobilised cohesin domain. Elution of the LG-Doc from the cohesin column was with EDTA. This affinity chromatography was repeated using an LG-dockerin column for the purification of cohesin fusion protein. The fusion proteins created in this report have shown that the properties of the cohesin and dockerin domains can be transferred to other protein domains and that the interaction between the cohesin and dockerin is specific, Ca2+ -dependent and reversible. We have shown that the cohesin-dockerin interaction has several properties making it suitable for use in recombinant fusion protein production and purification.

Abstract: Retroviral transduction efficiency is related to the multiplicity of infection and the physiological state of the target cells. It is generally not known what proportion of a cell population is susceptible to transduction. We used coinfection with two retroviral vectors containing the marker genes for green fluorescent protein and the truncated human nerve growth factor receptor. In the CD34+ cell line TF-1 or human primary CD34+ hematopoietic progenitor cells, it was found that cells transduced with one vector had a better than random chance of transduction by the other vector. A probability model was developed to estimate target cell susceptibility; susceptibility was calculated as the product of the proportions of transgene-positive cells divided by the proportion of double-positive cells. By using this relationship, it was found that susceptibility was related to the target cell type and culture conditions but not the retroviral titer or the retroviral packaging envelope protein used in this study. Cotransduction with two vectors is a relatively simple procedure that provides a means to assess the maximum transduction level possible in a given cell population.
