Combining hydroacoustics and underwater video is an effective tool for generating fish population estimates. However, hydroacoustics cannot be used to differentiate fish from the seafloor within an area known as the acoustic dead zone. A common way to address this is to exclude data near the bottom. The effect of this exclusion zone on population estimates of nearshore semi-pelagic rockfish is unknown. This study explores the effect of a near bottom (0–1 m) exclusion zone by comparing ROV video data to data from a combined hydroacoustic and video method. Higher densities of semi-pelagic species (Black and Blue/Deacon Rockfish) were observed in the combined acoustic and video method, suggesting that most of the population resides above the exclusion zone. Demersal rockfish observed by the ROV did not contaminate acoustic data of semi-pelagic species, since they remained within the exclusion zone. Results demonstrate that extrapolation of school data into the exclusion zone provided a realistic correction to the acoustic data for Black Rockfish. Our work demonstrates that excluding the data within 1 m of the bottom does not negatively affect the ability of the combined video hydroacoustic method to sample semi-pelagic rockfish.

Despite decades of climate science research, existing climate actions have had limited impacts on mitigating climate change. Efforts to reduce emissions, for example, have yet to spur sufficient action to reduce the most severe effects of climate change. We draw from our experiences as Ojibwe knowledge holders and community members, scientists, and scholars to demonstrate how the lack of recognition of traditional knowledges (TK) within climate science constrains effective climate action and exacerbates climate injustice. Often unrecognized in science and policy arenas, TK generates insights into how justice-driven climate action, rooted in relational interdependencies between humans and older-than-human relatives, can provide new avenues for effectively addressing climate change. We conclude by arguing for a shift toward meaningful and respectful inclusion of plural knowledge systems in climate governance.

Although tornadoes produced by quasi-linear convective systems (QLCSs) generally are weak and short lived, they have high societal impact due to their proclivity to develop over short time scales, within the cool season, and during nighttime hours. Precisely why they are weak and short lived is not well understood, although recent work suggests that QLCS updraft width may act as a limitation to tornado intensity. Herein, idealized simulations of tornadic QLCSs are performed with variations in hodograph shape and length as well as initiation mechanism to determine the controls of tornado intensity. Generally, the addition of hodograph curvature in these experiments results in stronger, longer-lived tornadic-like vortices (TLVs). A strong correlation between low-level mesocyclone width and TLV intensity is identified (R2 = 0.61), with a weaker correlation in the low-level updraft intensity (R2 = 0.41). The tilt and depth of the updraft are found to have little correlation to tornado intensity. Comparing QLCS and isolated supercell updrafts within these simulations, the QLCS updrafts are less persistent, with the standard deviations of low-level vertical velocity and updraft helicity approximately 48% and 117% greater, respectively. A forcing decomposition reveals that the QLCS cold pool plays a direct role in the development of the low-level updraft, providing the benefit of additional forcing for ascent while also having potentially deleterious effects on both the low-level updraft and near-surface rotation. The negative impact of the cold pool ultimately serves to limit the persistence of rotating updraft cores within the QLCS.

Background: Heterozygote carriers of potentially pathogenic variants in the cystic fibrosis transmembrane conductance regulator ( CFTR) gene have increased asthma risk. However, the frequency and impact of CFTR variation among individuals with asthma is unknown. Objective: To determine whether potentially pathogenic CFTR variants associate with disease severity and whether individuals with two potentially pathogenic variants exist in a severe asthma-enriched cohort . Methods: We analyzed sequencing data spanning a 190.5Kb region of CFTR in participants from the Severe Asthma Research Program (SARP1-3). Potentially pathogenic, rare CFTR variants (frequencyCFTR genotypes (0-2 potentially pathogenic variants) and severity outcomes. Results: Of 1401 participants, 9.5% (134) had one potentially pathogenic variant, occurring more frequently in non-Hispanic white (NHW, 10.1% [84 of 831]) compared to African American individuals (AA, 5.2% [22 of 426]). We found ≥2 potentially pathogenic CFTR variants in 1.4% (19); 0.5% (4) of NHW and 2.8% (12) of AA. Potentially pathogenic CFTR variant genotypes (≥1 or ≥2 variants) were not cumulatively associated with lung function or exacerbations. In NHW, we found three F508del compound heterozygotes with F508del and a VVCC (two 5T;TG12[c.1210-11T>G] and one Arg1070Trp) and a homozygote for the VVCC, 5T;TG12. Conclusions: We found potentially pathogenic CFTR variants within a severe asthma-enriched cohort , including three compound heterozygote genotypes variably associated with CF in NHW individuals. These findings provide the rationale for CFTR sequencing and phenotyping of CF-related traits in individuals with severe asthma.

Alpha-1 antitrypsin deficiency (AATD) is one of the most common genetic diseases that is associated with severe complications and yet remains underdiagnosed. The pulmonary symptoms of both AATD and asthma include cough, excessive sputum production, dyspnea, and wheezing. These symptoms overlap significantly leading to difficulty distinguishing between these two conditions and suspicion that there may be an overlap syndrome. We aim to discuss the pathophysiology, clinical manifestations, and treatment of both alpha-1 antitrypsin and asthma and how they may overlap.Recent literature suggests that there is an association between asthma and AATD. This association has been hypothesized to be secondary to an imbalance of elastase and anti-elastase leading to a pro-inflammatory state in patients with AATD. This review serves to overview the pathophysiology, clinical manifestations, and treatment of alpha-1 antitrypsin, asthma, and the increasingly recognized intersection of the two, AATD-asthma overlap syndrome.

Feminist political ecologists have transformed mainstream political ecology since its inception. The foundational and current work of feminist political ecologists indicate that their field is attentive to the epistemological foundations of power, inequities, and inequalities that cut across intersectional identities and hierarchies of difference and at the sites of environmental conflict and governance. Feminist political ecologists have made important theoretical interventions in the interdisciplinary community of political ecologists, but the use of feminist methodologies and 'team-based environmental science' can be expanded. We argue that revisiting feminist methodological commitments is critical for furthering how feminist political ecology examines how, and in what way, power and privilege operate in the contexts where environmental knowledge is produced. We make our argument by drawing upon a multi-year, multi-sited project to describe how collaborative event ethnography (CEE) offers many possibilities to reassess feminist political ecology research designs. We show how the recognition of diverse and plural epistemologies are foundational preconditions to integrating feminist principles in feminist political ecology research. We find that integrating reflexivity, responsibility, and co-production in research designs create opportunities for, and challenges to, carrying out feminist political ecological practice. In so doing, the integration of feminist methodologies are critical to disrupting knowledge hegemonies and providing new modes of practicing feminist political ecologies. The politics of environmental justice increasingly feature in environmental governance across multiple levels. Environmental defenders risk their lives to protect land, water, and forests. Non-human actors like rivers are gaining rights. Frontline environmental justice communities now include nation-states like Fiji that faces existential threats from climate change. Indigenous Peoples’ fights for self-determination illuminate how deeply connected and inseparable are the politics of sovereignty, representation, and environment. This chapter explores these developments to chart and examine how a politics of environmental justice can inform environmental and social policies by treating environmental justice as a driver, rather than unintended consequence, of policy and politics. Through this critical, comparative review, the chapter illuminates how and why environmental justice concerns matter for environmental governance and for the study of comparative environmental politics.

Although Indigenous Peoples make significant contributions to global environmental governance and were prominent actors at the 2015 Paris Climate Summit, COP21, they remain largely invisible in conventional, mainstream, and academic accounts of COP21. In this article, we adopt feminist collaborative event ethnography to draw attention to often marginalized and unrecognized actors and help make visible processes that are often invisible in the study of power and influence at sites of global environmental governance. Specifically, we integrate current approaches to power from international relations and political ecology scholarship to investigate how Indigenous Peoples, critical actors for solving global environmental challenges, access, navigate, and cultivate power at COP21 to shape global environmental governance. Through conceptual and methodological innovations that illuminate how Indigenous Peoples overcome structural and spatial barriers to engagement, this article demonstrates how attention to the politics of representation through pluralistic approaches to power can help expand the repertoire of possibilities for advancing global environmental governance.

Natural and anthropogenic hazards have the potential to impact all aspects of society including its economy and the environment. Diagnostic data to inform decision-making are critical for hazard management whether for emergency response, routine monitoring or assessments of potential risks. Imaging spectroscopy (IS) has unique contributions to make via the ability to provide some key quantitative diagnostic information. In this paper, we examine a selection of key case histories representing the state of the art to gain an insight into the achievements and perspectives in the use of visible to shortwave infrared IS for the detection, assessment and monitoring of a selection of significant natural and anthropogenic hazards. The selected key case studies examined provide compelling evidence for the use of the IS technology and its ability to contribute diagnostic information currently unattainable from operational spaceborne Earth observation systems. User requirements for the applications were also evaluated. The evaluation showed that the projected launch of spaceborne IS sensors in the near-, mid and long term future, together with the increasing availability, quality and moderate cost of off the shelf sensors, the possibilities to couple unmanned autonomous systems with miniaturized sensors, should be able to meet these requirements. The challenges and opportunities for the scientific community in the future when such data become available will then be ensuring consistency between data from different sensors, developing techniques to efficiently handle, process, integrate and deliver the large volumes of data, and most importantly translating the data to information that meets specific needs of the user community in a form that can be digested/understood by them. The latter is especially important to transforming the technology from a scientific to an operational tool. Additionally, the information must be independently validated using current trusted practices and uncertainties quantified before IS derived measurement can be integrated into operational monitoring services.

How and why does participation matter for procedural justice in environmental governance? Drawing insights from multiple empirical examples that include local, national, and international decision-making processes, this chapter explores the synergies and tensions of the participatory landscape for procedural justice in global environmental governance. It takes the reader through the evolution of approaches to participation and procedural justice to demonstrate how considerations of power—where it is located, how it is sourced and exercised, and how it operates—are central for advancing procedural justice. By directing attention to how accessibility, representation, and capabilities can shape power and influence in decision-making processes, this chapter provides readers with analytical tools for approaching environmental justice through a procedural lens.

Chitinases and chitinase-like proteins are an evolutionary conserved group of proteins. In the absence of chitin synthesis in mammals, the conserved presence of chitinases suggests their roles in physiology and immunity, but experimental evidence to prove these roles is scarce. Chitotriosidase (chit1) is one of the two true chitinases present in mammals and the most prevalent chitinase in humans. In this study, we investigated the regulation and the role of chit1 in a mouse model of Klebsiella pneumoniae lung infection. We show that chitinase activity in bronchoalveolar lavage fluid is significantly reduced during K. pneumoniae lung infection. This reduced activity is inversely correlated with the number of neutrophils. Further, instilling neutrophil lysates in lungs decreased chitinase activity. We observed degradation of chit1 by neutrophil proteases. In a mouse model, chit1 deficiency provided a significant advantage to the host during K. pneumoniae lung infection by limiting bacterial dissemination. This phenotype was independent of inflammatory changes in chit1−/− mice as they exerted a similar inflammatory response. The decreased dissemination resulted in improved survival in chit1−/− mice infected with K. pneumoniae in the presence or absence of antibiotic therapy. The beneficial effects of chit1 deficiency were associated with altered Akt activation in the lungs. Chit1−/− mice induced a more robust Akt activation postinfection. The role of the Akt pathway in K. pneumoniae lung infection was confirmed by using an Akt inhibitor, which impaired health and survival. These data suggest a detrimental role of chit1 in K. pneumoniae lung infections.

Strong to violent tornadoes cause a disproportionate amount of damage, in part because the width and length of a tornado damage track are correlated to tornado intensity (as now estimated through enhanced Fujita scale ratings). The tendency expressed in the observational record is that the most intense tornadoes are often the widest. Herein the authors explore the simple hypothesis that wide intense tornadoes should form more readily out of wide rotating updrafts. This hypothesis is based on an application of Kelvin’s circulation theorem, which is used to argue that the large circulation associated with a wide intense tornado is more plausibly associated with a wide mesocyclone. Because a mesocyclone is, strictly speaking, a rotating updraft, the mesocyclone width should increase with increasing updraft width. A simple mathematical model that is quantified using observations of mesocyclones supports this hypothesis, as do idealized numerical simulations of supercellular thunderstorms.
