Abstract

Among the meson decays, K → π ν ν ‾ are the cleanest environment, from the theoretical point of view, where to search for new physics effects. The NA62 Experiment at CERN SPS aims to measure the BR ( K + → π + ν ν ‾ ) with a 10% precision by the end of 2018. It has been commissioned with technical runs in 2014 and 2015, and some preliminary results of the detector performances and quality of data are here reported.

Abstract

The $K^+\rightarrow \pi^+ \nu \bar \nu$ decay is one of the theoretically cleanest meson decay where to look for indirect effects of new physics complementary to LHC searches. The NA62 experiment at CERN is designed to measure the branching ratio (BR) of this decay with 10\% precision. NA62 has been successfully launched in October 2014, took data in pilot runs in 2014 and 2015 reaching the final designed beam intensity. The NA62 experimental setup is illustrated and quality of data acquired in view of the final measurement is reported. The $K^+\rightarrow \pi^+ \nu \bar \nu$ decay is theoretically one of the cleanest meson decays and so a good place to look for indirect effects of new physics complementary to LHC searches. The NA62 experiment at CERN SPS aims to collect about 100 of such events in two years of data taking, keeping the background at the level of 10%. Abstract

Pexastimogene devacirepvec (Pexa-Vec) is a vaccinia virus-based oncolytic immunotherapy designed to preferentially replicate in and destroy tumor cells while stimulating anti-tumor immunity by expressing GM-CSF. An earlier randomized Phase IIa trial in predominantly sorafenib-naïve hepatocellular carcinoma (HCC) demonstrated an overall survival (OS) benefit. This randomized, open-label Phase IIb trial investigated whether Pexa-Vec plus Best Supportive Care (BSC) improved OS over BSC alone in HCC patients who failed sorafenib therapy (TRAVERSE). 129 patients were randomly assigned 2:1 to Pexa-Vec plus BSC vs. BSC alone. Pexa-Vec was given as a single intravenous (IV) infusion followed by up to 5 IT injections. The primary endpoint was OS. Secondary endpoints included overall response rate (RR), time to progression (TTP) and safety. A high drop-out rate in the control arm (63%) confounded assessment of response-based endpoints. Median OS (ITT) for Pexa-Vec plus BSC vs. BSC alone was 4.2 and 4.4 months, respectively (HR, 1.19, 95% CI: 0.78–1.80; p = .428). There was no difference between the two treatment arms in RR or TTP. Pexa-Vec was generally well-tolerated. The most frequent Grade 3 included pyrexia (8%) and hypotension (8%). Induction of immune responses to vaccinia antigens and HCC associated antigens were observed. Despite a tolerable safety profile and induction of T cell responses, Pexa-Vec did not improve OS as second-line therapy after sorafenib failure. The true potential of oncolytic viruses may lie in the treatment of patients with earlier disease stages which should be addressed in future studies. ClinicalTrials.gov: NCT01387555

Abstract

Ten healthy subjects were submitted to alcohol loading (0.8 g/kg body weight; average alcohol blood level 0.97 g 0/00). The number of platelets, hematocrit and ADP-induced platelet aggregation curves did not show any variation from baseline. Significant changes (p less than 0.05) were observed as regards latent period (increased) and maximum amplitude (reduced) for collagen-induced platelet aggregation. In agreement with others, the authors conclude that alcohol definitely inhibits platelet aggregation.

Abstract

The NA62 experiment at SPS is a continuation of the long standing CERN kaon physics program. The high statistics and the unprecedent precision allow to probe the Standard Model and test the description of the strong interactions at low energy. The final results on the the lepton universality test by measuring the ratio $R_K = \Gamma(K^{+}\to e^{+}\nu) / \Gamma(K^{+}\to \mu^{+}\nu)$ and the study of the $K^\pm\to\pi^{\pm}\gamma\gamma $ decay are presented. The major goal of the NA62 experiment is to perform a measurement of the $Br(K^{+} \to \pi^{+} \nu \bar{\nu})$ with a precision of 10% in two years of data taking. Abstract

The main goal of the NA62 experiment at the CERN SPS accelerator is to measure the branching fraction of the ultra-rare decay K+→π+ν with 10% accuracy. Key aspects of the detector configuration are described, with emphasis on tagging and timestamping the minority kaons in the high intensity, unseparated, charged-particle beam using KTAG, an upgraded version of a CEDAR differential Cherenkov detector. Data are presented showing that KTAG was successfully commissioned at CERN in November 2012.

Abstract

The use of digital pathology for the histomorphologic profiling of pathological specimens is expanding the precision and specificity of quantitative tissue analysis at an unprecedented scale; thus, enabling the discovery of new and functionally relevant histological features of both predictive and prognostic significance. In this study, we apply quantitative automated image processing and computational methods to profile the subcellular distribution of the multi-functional transcriptional regulator, Kaiso (ZBTB33), in the tumors of a large racially diverse breast cancer cohort from a designated health disparities region in the United States. Multiplex multivariate analysis of the association of Kaiso's subcellular distribution with other breast cancer biomarkers reveals novel functional and predictive linkages between Kaiso and the autophagy-related proteins, LC3A/B, that are associated with features of the tumor immune microenvironment, survival, and race. These findings identify effective modalities of Kaiso biomarker assessment and uncover unanticipated insights into Kaiso's role in breast cancer progression.

Abstract

Enhancer profiling is a powerful approach for discovering cis-regulatory elements that define the core transcriptional regulatory circuits of normal and malignant cells. Gene control through enhancer activity is often dominated by a subset of lineage-specific transcription factors. By integrating measures of chromatin accessibility and enrichment for H3K27 acetylation, we have generated regulatory landscapes of chronic lymphocytic leukemia (CLL) samples and representative cell lines. With super enhancer-based modeling of regulatory circuits and assessments of transcription factor dependencies, we discover that the essential super enhancer factor PAX5 dominates CLL regulatory nodes and is essential for CLL cell survival. Targeting enhancer signaling via BET bromodomain inhibition disrupts super enhancer-dependent gene expression with selective effects on CLL core regulatory circuitry, conferring potent anti-tumor activity., (Copyright © 2018 Elsevier Inc. Abstract

Master transcription factors interact with DNA to establish cell type identity and to regulate gene expression in mammalian cells. The genome-wide map of these transcription factor binding sites has been termed the cistrome. Here we show that the androgen receptor (AR) cistrome undergoes extensive reprogramming during prostate epithelial transformation in man. Using human prostate tissue, we observed a core set of AR binding sites that are consistently reprogrammed in tumors. FOXA1 and HOXB13 colocalized at the reprogrammed AR binding sites in human tumor tissue. Introduction of FOXA1 and HOXB13 into an immortalized prostate cell line reprogrammed the AR cistrome to resemble that of a prostate tumor, functionally linking these specific factors to AR cistrome reprogramming. These findings offer mechanistic insights into a key set of events that drive normal prostate epithelium toward transformation and establish the centrality of epigenetic reprogramming in human prostate tumorigenesis.

Abstract

The vast majority of disease-associated single-nucleotide polymorphisms (SNPs) mapped by genome-wide association studies (GWASs) are located in the non-protein-coding genome, but establishing the functional and mechanistic roles of these sequence variants has proven challenging. Here we describe a general pipeline in which candidate functional SNPs are first evaluated by fine mapping, epigenomic profiling, and epigenome editing, and then interrogated for causal function by using genome editing to create isogenic cell lines followed by phenotypic characterization. To validate this approach, we analyzed the 6q22.1 prostate cancer risk locus and identified rs339331 as the top-scoring SNP. Epigenome editing confirmed that the rs339331 region possessed regulatory potential. By using transcription activator-like effector nuclease (TALEN)-mediated genome editing, we created a panel of isogenic 22Rv1 prostate cancer cell lines representing all three genotypes (TT, TC, CC) at rs339331. Introduction of the 'T' risk allele increased transcription of the regulatory factor 6 (RFX6) gene, increased homeobox B13 (HOXB13) binding at the rs339331 region, and increased deposition of the enhancer-associated H3K4me2 histone mark at the rs339331 region compared to lines homozygous for the 'C' protective allele. The cell lines also differed in cellular morphology and adhesion, and pathway analysis of differentially expressed genes suggested an influence of androgens. In summary, we have developed and validated a widely accessible approach that can be used to establish functional causality for noncoding sequence variants identified by GWASs.

Abstract

Ten healthy subjects were submitted to alcohol loading (0.8 g/kg body weight; average alcohol blood level 0.97 g 0/00). The number of platelets, hematocrit and ADP-induced platelet aggregation curves did not show any variation from baseline. Significant changes (p less than 0.05) were observed as regards latent period (increased) and maximum amplitude (reduced) for collagen-induced platelet aggregation. In agreement with others, the authors conclude that alcohol definitely inhibits platelet aggregation.
