Many Coupled Model Intercomparison Project phase 6 (CMIP6) models have exhibited a substantial cold bias in the global mean surface temperature (GMST) in the latter part of the 20th century. An overly strong negative aerosol forcing has been suggested as a leading contributor to this bias. An updated configuration of UK Earth System Model (UKESM) version 1, UKESM1.1, has been developed with the aim of reducing the historical cold bias in this model. Changes implemented include an improved representation of SO2 dry deposition, along with several other smaller modifications to the aerosol scheme and a retuning of some uncertain parameters of the fully coupled Earth system model. The Diagnostic, Evaluation and Characterization of Klima (DECK) experiments, a six-member historical ensemble and a subset of future scenario simulations are completed. In addition, the total anthropogenic effective radiative forcing (ERF), its components and the effective and transient climate sensitivities are also computed. The UKESM1.1 preindustrial climate is warmer than UKESM1 by up to 0.75 K, and a significant improvement in the historical GMST record is simulated, with the magnitude of the cold bias reduced by over 50 %. The warmer climate increases ocean heat uptake in the Northern Hemisphere oceans and reduces Arctic sea ice, which is in better agreement with observations. Changes to the aerosol and related cloud properties are a driver of the improved GMST simulation despite only a modest reduction in the magnitude of the negative aerosol ERF (which increases by +0.08 W m−2). The total anthropogenic ERF increases from 1.76 W m−2 in UKESM1 to 1.84 W m−2 in UKESM1.1. The effective climate sensitivity (5.27 K) and transient climate response (2.64 K) remain largely unchanged from UKESM1 (5.36 and 2.76 K respectively).

Neuroblastoma (NB) is a cancer that develops in the neuroblasts. It is the most common cancer in children under the age of 1 year, accounting for approximately 6% of all cancers. The prognosis of NB is linked to both age and degree of cell differentiation. This results in a range of survival rates for patients, with outcomes ranging from recurrence and mortality to high survival rates and tumor regression. Our previous work indicated that PKC-ι promotes cell proliferation in NB cells through the PKC-ι/Cdk7/Cdk2 cascade. We report on two atypical protein kinase inhibitors as potential therapeutic candidates against BE(2)-C and BE(2)-M17 cells: a PKC-ι-specific 5-amino-1-2,3-dihydroxy-4-(methylcyclopentyl)-1H-imidazole-4-carboxamide and a PKC-ζ specific 8-hydroxy-1,3,6-naphthalenetrisulfonic acid. Both compounds induced apoptosis and retarded the epithelial-mesenchymal transition (EMT) of NB cells. Proteins 14-3-3 and Smad2/3 acted as central regulators of aPKC-driven progression in BE(2)-C and BE(2)-M17 cells in relation to the Akt1/NF-κB and TGF-β pathways. Data indicates that aPKCs upregulate Akt1/NF-κB and TGF-β pathways in NB cells through an association with 14-3-3 and Smad2/3 that can be diminished by aPKC inhibitors. In summary, both inhibitors appear to be promising potential neuroblastoma therapeutics and merit further research.

We document and evaluate the aerosol schemes as implemented in the physical and Earth system models, the Global Coupled 3.1 configuration of the Hadley Centre Global Environment Model version 3 (HadGEM3-GC3.1) and the United Kingdom Earth System Model (UKESM1), which are contributing to the sixth Coupled Model Intercomparison Project (CMIP6). The simulation of aerosols in the present-day period of the historical ensemble of these models is evaluated against a range of observations. Updates to the aerosol microphysics scheme are documented as well as differences in the aerosol representation between the physical and Earth system configurations. The additional Earth system interactions included in UKESM1 lead to differences in the emissions of natural aerosol sources such as dimethyl sulfide, mineral dust and organic aerosol and subsequent evolution of these species in the model. UKESM1 also includes a stratospheric–tropospheric chemistry scheme which is fully coupled to the aerosol scheme, while GC3.1 employs a simplified aerosol chemistry mechanism driven by prescribed monthly climatologies of the relevant oxidants. Overall, the simulated speciated aerosol mass concentrations compare reasonably well with observations. Both models capture the negative trend in sulfate aerosol concentrations over Europe and the eastern United States of America (US) although the models tend to underestimate sulfate concentrations in both regions. Interactive emissions of biogenic volatile organic compounds in UKESM1 lead to an improved agreement of organic aerosol over the US. Simulated dust burdens are similar in both models despite a 2-fold difference in dust emissions. Aerosol optical depth is biased low in dust source and outflow regions but performs well in other regions compared to a number of satellite and ground-based retrievals of aerosol optical depth. Simulated aerosol number concentrations are generally within a factor of 2 of the observations, with both models tending to overestimate number concentrations over remote ocean regions, apart from at high latitudes, and underestimate over Northern Hemisphere continents. Finally, a new primary marine organic aerosol source is implemented in UKESM1 for the first time. The impact of this new aerosol source is evaluated. Over the pristine Southern Ocean, it is found to improve the seasonal cycle of organic aerosol mass and cloud droplet number concentrations relative to GC3.1 although underestimations in cloud droplet number concentrations remain. This paper provides a useful characterisation of the aerosol climatology in both models and will facilitate understanding in the numerous aerosol–climate interaction studies that will be conducted as part of CMIP6 and beyond.

We present the average rest-frame spectrum of the final catalog of dusty star-forming galaxies (DSFGs) selected from the South Pole Telescope's SPT-SZ survey and measured with Band 3 of the Atacama Large Millimeter/submillimeter Array. This work builds on the previous average rest-frame spectrum, given in Spilker et al. (2014) for the first 22 sources, and is comprised of a total of 78 sources, normalized by their respective apparent dust masses. The spectrum spans 1.9 < z < 6.9 and covers rest-frame frequencies of 240–800 GHz. Combining this data with low- J CO observations from the Australia Telescope Compact Array, we detect multiple bright line features from ^12 CO, [C i ], and H _2 O, as well as fainter molecular transitions from ^13 CO, HCN, HCO ^+ , HNC, CN, H _2 O ^+ , and CH. We use these detections, along with limits from other molecules, to characterize the typical properties of the interstellar medium (ISM) for these high-redshift DSFGs. We are able to divide the large sample into subsets in order to explore how the average spectrum changes with various galaxy properties, such as effective dust temperature. We find that systems with hotter dust temperatures exhibit differences in the bright ^12 CO emission lines, and contain either warmer and more excited dense gas tracers or larger dense gas reservoirs. These observations will serve as a reference point to studies of the ISM in distant luminous DSFGs ( L _IR > 10 ^12 L _⊙ ), and will inform studies of chemical evolution before the peak epoch of star formation at z = 2–3.

Abstract The Global Coupled 3 (GC3) configuration of the Met Office Unified Model is presented. Among other applications, GC3 is the basis of the United Kingdom's submission to the Coupled Model Intercomparison Project 6 (CMIP6). This paper documents the model components that make up the configuration (although the scientific descriptions of these components are in companion papers) and details the coupling between them. The performance of GC3 is assessed in terms of mean biases and variability in long climate simulations using present‐day forcing. The suitability of the configuration for predictability on shorter time scales (weather and seasonal forecasting) is also briefly discussed. The performance of GC3 is compared against GC2, the previous Met Office coupled model configuration, and against an older configuration (HadGEM2‐AO) which was the submission to CMIP5. In many respects, the performance of GC3 is comparable with GC2, however, there is a notable improvement in the Southern Ocean warm sea surface temperature bias which has been reduced by 75%, and there are improvements in cloud amount and some aspects of tropical variability. Relative to HadGEM2‐AO, many aspects of the present‐day climate are improved in GC3 including tropospheric and stratospheric temperature structure, most aspects of tropical and extratropical variability and top‐of‐atmosphere and surface fluxes. A number of outstanding errors are identified including a residual asymmetric sea surface temperature bias (cool northern hemisphere, warm Southern Ocean), an overly strong global hydrological cycle and insufficient European blocking.

There is mounting evidence that resolving mesoscale eddies and western boundary currents as well as topographically controlled flows can play an important role in air–sea interaction associated with vertical and lateral transports of heat and salt. Here we describe the development of the Met Office Global Coupled Model version 2 (GC2) with increased resolution relative to the standard model: the ocean resolution is increased from 1/4 to 1/12° (28 to 9 km at the Equator), the atmosphere resolution increased from 60 km (N216) to 25 km (N512) and the coupling period reduced from 3 hourly to hourly. The technical developments that were required to build a version of the model at higher resolution are described as well as results from a 20-year simulation. The results demonstrate the key role played by the enhanced resolution of the ocean model: reduced sea surface temperature (SST) biases, improved ocean heat transports, deeper and stronger overturning circulation and a stronger Antarctic Circumpolar Current. Our results suggest that the improvements seen here require high resolution in both atmosphere and ocean components as well as high-frequency coupling. These results add to the body of evidence suggesting that ocean resolution is an important consideration when developing coupled models for weather and climate applications.

The latest coupled configuration of the Met Office Unified Model (Global Coupled configuration 2, GC2) is presented. This paper documents the model components which make up the configuration (although the scientific description of these components is detailed elsewhere) and provides a description of the coupling between the components. The performance of GC2 in terms of its systematic errors is assessed using a variety of diagnostic techniques. The configuration is intended to be used by the Met Office and collaborating institutes across a range of timescales, with the seasonal forecast system (GloSea5) and climate projection system (HadGEM) being the initial users. In this paper GC2 is compared against the model currently used operationally in those two systems. Overall GC2 is shown to be an improvement on the configurations used currently, particularly in terms of modes of variability (e.g. mid-latitude and tropical cyclone intensities, the Madden–Julian Oscillation and El Niño Southern Oscillation). A number of outstanding errors are identified with the most significant being a considerable warm bias over the Southern Ocean and a dry precipitation bias in the Indian and West African summer monsoons. Research to address these is ongoing.

Photosynthesis by marine diatoms contributes substantially to global biogeochemical cycling and ecosystem productivity. It is widely accepted that diatoms are extremely sensitive to changes in Fe availability, with numerous in situ experiments demonstrating rapid growth and increased export of elements (e.g. C, Si and Fe) from surface waters as a result of Fe addition. Less is known about the effects of Fe enrichment on the phenotypes of diatoms, such as associated changes in nutritional value – furthermore, data on taxon-specific responses are almost non-existent. Enhanced supply of nutrient-rich waters along the coast of the subantarctic Kerguelen Island provide a valuable opportunity to examine the responses of phytoplankton to natural Fe enrichment. Here we demonstrate the use of synchrotron radiation Fourier Transform Infrared (SR-FTIR) microspectroscopy to analyse changes in the macromolecular composition of diatoms collected along the coast and plateau of Kerguelen Island, Southern Ocean. SR-FTIR microspectroscopy enabled the analysis of individual diatom cells from mixed communities of field-collected samples, thereby providing insight into in situ taxon-specific responses in relation to changes in Fe availability. Phenotypic responses were taxon-specific in terms of intraspecific variability and changes in proteins, amino acids, phosphorylated molecules, silicate/silicic acid and carbohydrates. In contrast to some previous studies, silicate/silicic acid levels increased under Fe enrichment, in conjunction with increases in carbohydrate stores. The highly abundant taxon Fragilariopsis kerguelensis displayed a higher level of phenotypic plasticity than Pseudo-nitzschia spp., while analysis of the data pooled across all measured taxa showed different patterns in macromolecular composition compared to those for individual taxon. This study demonstrates that taxon-specific responses to Fe enrichment may not always be accurately reflected by bulk community measurements, highlighting the need for further research into taxon-specific phenotypic responses of phytoplankton to environmental change.

Confirmatory factor analysis (CFA) is a commonly used statistical technique. Recommendations for evaluating CFA highlight scholars should outline the expected model, conduct data screening, report model estimation and evaluation, and report key information about results to provide evidence for latent variables. The purpose of the current study was to review and evaluate the use of CFA in research published from 2013 to 2022 in the journal of "Measurement in Physical Education and Exercise Science" (MPEES), the most measurement focused journal in kinesiology, in our view. Results were cross-checked by examining research published in Research Quarterly for Exercise and Sport during the same time period. Strengths included providing information about the expected model, evaluating the final model, and reporting parameter estimates. Areas for improvement included conducting data screening and evaluating factor quality. Finally, recommendations for scholars to improve reporting of CFA in kinesiology are provided.

The time of onset of changes in liver metabolism of the 14-hour-insulin-deprived, alloxan-diabetic rat following continuous portal vein infusion of insulin was studied. The earliest effect of insulin—on glycogen content of the liver and on the capacity of the liver slice to synthesize cholesterol from acetate—occurred between 10 and 30 minutes after the start of the infusion. The liver's capacity for incorporating acetate carbon into cholesterol rose dramatically between 30 and 60 minutes after the start of the hormone infusion, and returned to normal in 90 minutes. The significance of this biphasic response in hepatic cholesterogenesis to portal vein insulin infusion is discussed. Fatty acid synthesis was slowest to respond to insulin. An effect on hepatic lipogenesis was first detected between 60 and 90 minutes after injection, and the liver's capacity for incorporation of acetate-l-C14 into fatty acids continued to increase with duration of insulin administration up to 180 minutes.

The time relationships in the changes in hepatic lipogenesis and cholesterogenesis induced by the feeding of corn oil and lard have been studied. A pronounced decrease in the capacity of the liver to convert acetate carbon to fatty acids was observed as early as 1 hour after fat administration. The increase in the liver's capacity to incorporate acetate carbon into cholesterol developed slowly after fat feeding; the earliest change was observed in 12 hours. The restoration of lipogenesis to normal, following the withdrawal of fat, was more rapid than that of cholesterogenesis. The time changes in hepatic lipogenesis and cholesterogenesis induced by fat feeding were correlated with the time of appearance of exogenous fat in the liver.
