Abstract: A global heat flux model based on a fractional derivative of plasma pressure is proposed for the heat transport in fusion plasmas. The degree of the fractional derivative of the heat flux, α, is defined through the power balance analysis of the steady state. The model was used to obtain the experimental values of α for a large database of the Joint European Torus (JET) carbon-wall as well as ITER like-wall plasmas. Abstract: Most Chemical Companies create and maintain a set of Standards, Requirements and Recommended practices for Process and Occupational Safety. These augment the legal and industry consensus standards which exist. The process of creating, updating and maintaining these is a necessary commitment to Process Safety Management. The process of making sure that these are properly applied throughout the corporation is a very important task. Without this, all the other efforts fail. When accidents occur, it is occasionally found by the investigation that a requirement of the company has not been applied. In these cases, the root cause is found to be a failure in the implementation process. This part often turns out to be the most laborious of all. It is not enough to simply create a requirement or a recommendation and tell the organisation that it exists and they must apply it. When the topic has been researched within EPSC this has been cited as a problem. There have been particular challenges in the Mergers and Acquisitions field where an acquired facility has either continued to operate in ignorance or has not implemented necessary changes quickly enough. By breaking down the challenge into discrete steps with the issues elaborated for each, the EPSC study group has identified good practice and tools for creating, maintaining, implementing and compliance checking for each. This paper reviews these tools with a view to wider use and better performance.

Abstract: The anticipated heat flux limit of the European DEMO first wall is ∼1−2 MW/m2. During transient and off normal events, the heat load deposited on the wall would be much larger than the steady state heat load and exceed the first wall limit, therefore the breeding blanket first wall needs to be protected. This involves dedicated discrete limiters in certain regions of the machine that would take the brunt of the heat load as well as adequate shaping of the first wall. The current concept envisages limiters at a few (3–4) equatorial ports to cope with the ramp-up of the plasma; upper limiters (in ∼8 upper ports) are considered for upward vertical displacement events. Two design options have been considered for these limiters: a modular design where the limiter plasma facing components are attached to individual plates that are assembled together so that transient electromagnetic loads can be reduced, and in case of damage the plates can be replaced/repaired individually; and a divertor-like design where the plasma facing components are attached to a single Eurofer cassette. Other limiters considered include inner wall limiters in case of plasma contraction and lower limiters may be needed for downward vertical displacement events. The thermal hydraulic finite element analysis results show that the integrity of the cooling pipes can be maintained during the anticipated transient events. The limiters are considered to be sacrificial and designed to be replaceable independently from the breeding blanket system. The design has to allow that installation, removal or replacement of the limiters can be performed remotely. Strategy to tackle outstanding issues and required R&D is also discussed.

Abstract: The 2014–2016 JET results are reviewed in the light of their significance for optimising the ITER research plan for the active and non-active operation. More than 60 h of plasma operation with ITER first wall materials successfully took place since its installation in 2011. New multi-machine scaling of the type I-ELM divertor energy flux density to ITER is supported by first principle modelling. ITER relevant disruption experiments and first principle modelling are reported with a set of three disruption mitigation valves mimicking the ITER setup. Insights of the L–H power threshold in Deuterium and Hydrogen are given, stressing the importance of the magnetic configurations and the recent measurements of fine-scale structures in the edge radial electric. Dimensionless scans of the core and pedestal confinement provide new information to elucidate the importance of the first wall material on the fusion performance. H-mode plasmas at ITER triangularity (H = 1 at ßN ~ 1.8 and n/nGW ~ 0.6) have been sustained at 2 MA during 5 s. The ITER neutronics codes have been validated on high performance experiments. Prospects for the coming D–T campaign and 14 MeV neutron calibration strategy are reviewed. Abstract: Objective To assess the anal function of women who have suffered a third degree perineal tear during parturition. Patients Fifty-three consecutive women who sustained a third degree tear, between January 1998 and March 2000, at the Princess Anne Maternity Unit, Royal Bolton Hospital were assessed. Methods Women were assessed at 3 months post partum using the Cleveland Clinic Incontinence Score, digital assessment of anal sphincter and endo-anal ultrasound scan. Results At 3 months post partum 75% of the participants had no symptoms of anal incontinence, 18% had mild symptoms and 7% had more severe symptoms. Anal endosonography demonstrated normal anal sphincters in 66% of participants, an abnormality in the external sphincter in 29% and a defect in both sphincters in 2%. There was poor correlation between symptoms and scan defects. Conclusion The incidence of anal incontinence following repair of a third degree tear was not high and it is unlikely that we are missing a hidden pool of symptomatic women. No major change in management policy is required. The routine assessment of anal function in women who had sustained a third degree tear was appreciated by the women and enabled us to identify the small portion of women with significant symptoms.

Abstract: Objective: This study employs a Developmental Origins of Health and Disease (DOHaD) approach to assess the effect of vitamin D deficiency (VDD) in childhood on the risk of osteoporosis in adulthood in an archaeological sample of skeletons dating from the 18th to 19th centuries., Materials: Femora and lumbar vertebrae of 65 adults aged 18+ years (26 diagnosed with residual rickets and 39 without) from an 18th-19th century Quaker burial ground at Coach Lane, North Shields, England., Methods: Bone mineral density (BMD) was measured for the femoral neck and first four lumbar vertebrae of each individual using a dual energy X-ray absorptiometry (DXA) scanner as a proxy for assessing osteoporotic fracture risk., Results: 3-way ANOVA revealed no statistically significant differences in BMD between individuals with and without residual rickets across age and sex., Conclusions: A combination of lifestyle and environmental factors likely influenced the BMD of people buried at Coach Lane across the life course. The impact of childhood VDD on BMD later in life can be mitigated through other factors such as physical activity and diet., Significance: This is one of the first bioarchaeological studies to take a DOHaD approach to understand osteoporosis risk in 18th-19th century England. It highlights the complexity of aetiological factors for osteoporosis and that VDD in early life does not necessarily predispose a person to osteoporosis in adulthood., Limitations: BMD is not the only indicator of osteoporosis. Abstract: The Spherical Tokamak for Energy Production (STEP) requires high-field magnet designs and has therefore adopted the REBCO-based high-temperature superconductor (HTS) as its current carrier. The HTS enables the toroidal field (TF) coils to be remountable, which unlocks STEP's vertical maintenance approach; however, remountable joints, approximately 18 GJ of stored energy and limited space down the centre of a spherical tokamak, make the TF coils the most challenging. STEP has pursued a passive approach to TF coil quench protection in order to limit coil terminal voltage. Initial results suggest that a solution may rely on tuning internal coil resistance coupled with actively powered heaters. The pre-conceptual inter-coil structure demonstrates acceptable stresses and deflections under steady-state operating conditions and preliminary fault scenarios, and loads are distributed to limit the tensile force on the TF centre rod. Finally, the HTS must operate reliably in a high radiation environment and endure high neutron fluences, ensuring commercially relevant magnet lifetimes. Initial experiments indicate that instantaneous gamma irradiation of HTS has no negative impact on current carrying capacity. Experimental programmes are underway to cold irradiate HTS to fusion-relevant fluences and to develop a method of assuring tape irradiation tolerance using oxygen ions as an analogue for neutrons.This article is part of the theme issue 'Delivering Fusion Energy - The Spherical Tokamak for Energy Production (STEP)'.

13. Vitamin D status in post-medieval Northern England: Insights from dental histology and enamel peptide analysis at Coach Lane, North Shields (AD 1711-1857).

Abstract: Objectives: The post-medieval period in Europe saw a dramatic increase in metabolic bone disease related to vitamin D deficiency (VDD). Recent paleopathological work has utilized interglobular dentin (IGD) as a proxy for poor vitamin D status during development, while enamel peptide analysis allows the identification of chromosomal sex in non-adult remains. Here we explore the relationship between sex, the presence of IGD, and macroscopic markers of VDD in an industrial era assemblage from Northeast England., Materials and Methods: 25 individuals (9 females, 9 males, 9 unknown sex) from the cemetery site at Coach Lane, North Shields (1711-1857) were selected for paleopathological analysis, histological assessment of IGD, and enamel peptide determination of chromosomal sex., Results: Ground tooth sections from 21 individuals were of suitable quality for detection of IGD, and enamel peptide analysis confirmed the chromosomal sex of ten individuals. Sixteen individuals (76.1%) exhibited ≥1 episode of IGD. Nine of these (42.8%) exhibited >1 episode and four (19%) exhibited ≥4 episodes in regular intervals. Male sex was significantly associated with the presence of IGD (p = 0.0351; 100% males vs. 54.5% females). Females were more likely to exhibit macroscopic evidence of VDD (45.5% females vs 30% males) but this was not statistically significant., Discussion and Conclusions: Periods of poor mineral metabolism during childhood appear much more prevalent at Coach Lane than macroscopic evidence suggests. Evidence of seasonal IGD episodes indicates that northern latitude played a major role in poor VD status in the Northeast of England. The significant association of IGD with male sex may be due to sex-related differences in dentinal mineralization or a higher risk of poor VD status in males aged <5 years. Abstract: Nucleic acid aptamers are short sequences of single-stranded (ss) DNA or RNA that fold into a three-dimensional shape with useful binding properties. Traditionally, these properties have included specific recognition and binding of ions, small-molecules, proteins, and enzyme targets. Increasingly though, aptamers are being raised against complex subcellular or cellular targets. These broader-affinity aptamers can be usefully employed for detection, labeling, or therapeutic targeting of intact/living cells, whether prokaryotic or eukaryotic. Aptamers are usually developed from a random-sequence oligonucleotide library by repeated rounds of selection and amplification, a process named "systematic evolution of ligands by exponential enrichment" (SELEX). We describe here a widely applicable cell-SELEX method for raising aptamers against bacteria, using Escherichia coli strain HB101 as an example. Our cell-SELEX method uses a cycle of four stages: (1) incubation of a fluorescently labeled random-sequence ssDNA library with bacterial cells; (2) separation of cell-associated ssDNA from free ssDNA; (3) amplification of bound ssDNA by PCR, and (4) use of lambda-exonuclease to selectively regenerate ssDNA for further rounds of selection., (© 2023. Abstract: This article presents outcomes from a Workshop entitled "Bioarchaeology: Taking Stock and Moving Forward," which was held at Arizona State University (ASU) on March 6-8, 2020. Funded by the National Science Foundation (NSF), the School of Human Evolution and Social Change (ASU), and the Center for Bioarchaeological Research (CBR, ASU), the Workshop's overall goal was to explore reasons why research proposals submitted by bioarchaeologists, both graduate students and established scholars, fared disproportionately poorly within recent NSF Anthropology Program competitions and to offer advice for increasing success. Therefore, this Workshop comprised 43 international scholars and four advanced graduate students with a history of successful grant acquisition, primarily from the United States. Ultimately, we focused on two related aims: (1) best practices for improving research designs and training and (2) evaluating topics of contemporary significance that reverberate through history and beyond as promising trajectories for bioarchaeological research. Among the former were contextual grounding, research question/hypothesis generation, statistical procedures appropriate for small samples and mixed qualitative/quantitative data, the salience of Bayesian methods, and training program content. Topical foci included ethics, social inequality, identity (including intersectionality), climate change, migration, violence, epidemic disease, adaptability/plasticity, the osteological paradox, and the developmental origins of health and disease. Abstract: Objective: The present study explores growth and health in Roman (1st-3rd centuries CE) and Post-Roman (4th-7th centuries CE) Gaul, incorporating a life-course approach, to better understand the influence of Roman practices and lifestyles on health, and the impact of cultural change from the Roman to the Post-Roman period., Materials and Methods: The skeletal remains of 844 individuals were analyzed for non-specific signs of physiological stress, including growth disruption (diaphyseal and adult maximum femur length), dental enamel hypoplastic defects (DEH), cribra orbitalia (CO), and periosteal reaction of the tibiae (Tibia PR)., Results: The Gallo-Roman sample demonstrated shorter femoral lengths, and higher rates of DEH and Tibia PR. Post-Roman groups demonstrated longer femoral lengths and higher rates of CO., Conclusions: Gallo-Roman individuals may have been more regularly exposed to infectious pathogens throughout childhood, inhibiting opportunities for catch-up growth, resulting in high rates of DEH and shorter femoral lengths ('intermittent stress of low lethality'). This could be the result of overcrowding and insalubrious urban environments. Higher rates of CO in the Post-Roman samples may have been influenced by dietary changes between the periods., Significance: The intertwined and often synergistic relationships between early life environment, nutrition and settlement structure is highlighted, helping to further understandings of life experiences during the Roman and Post-Roman periods., Limitations: It was not possible to obtain sufficient data from northern regions during the Gallo-Roman period, limiting this analysis., Suggestions for Further Research: Further application of life course approaches can reveal subtle patterns in stress indicators., (Copyright © 2021 Elsevier Inc. 17. Health inequality in Britain before 1750.

Abstract: Background: This study examines the claim that social inequality in health in European populations was absent prior to 1750. This claim is primarily based on comparisons of life expectancy at birth in England between general and ducal (elite aristocrat) social classes from the 1550s to the 1870s., Methods: We examined historic childhood mortality trends among the English ducal class and the general population, based on previously published data. We compared mid-childhood to adolescent mortality (age 5-14) and early-childhood mortality (age 0-4) between the ducal class and the general population from the 17 th to 19 th centuries., Results: Prior to 1750, ducal early-childhood mortality was higher than the general population. However, mid-childhood to adolescent mortality was lower among the ducal class than the general population in all observed periods for boys, and almost all periods for girls. Among the ducal class, but not the general population, there was a sharp decline in early-childhood mortality around the 1750s which may partly explain the divergent trends in overall life expectancy at birth., Conclusion: Health inequality between the ducal class and general population was present in England from the 16 th to mid-18 th centuries, with disadvantages in mortality for ducal children in infancy and early childhood, but survival advantages in mid-childhood and adolescence. These opposing effects are obscured in life expectancy at birth data. Abstract: Present in all organisms, DNA ligases catalyse the formation of a phosphodiester bond between a 3' hydroxyl and a 5' phosphate, a reaction that is essential for maintaining genome integrity during replication and repair. Eubacterial DNA ligases use NAD + as a cofactor and possess low sequence and structural homology relative to eukaryotic DNA ligases which use ATP as a cofactor. These key differences enable specific targeting of bacterial DNA ligases as an antibacterial strategy. In this study, four small molecule accessible sites within functionally important regions of Escherichia coli ligase (EC-LigA) were identified using in silico methods. Molecular docking was then used to screen for small molecules predicted to bind to these sites. Eight candidate inhibitors were then screened for inhibitory activity in an in vitro ligase assay. Five of these (geneticin, chlorhexidine, glutathione (reduced), imidazolidinyl urea and 2-(aminomethyl)imidazole) showed dose-dependent inhibition of EC-LigA with half maximal inhibitory concentrations (IC 50 ) in the micromolar to millimolar range (11-2600 µM). Two (geneticin and chlorhexidine) were predicted to bind to a region of EC-LigA that has not been directly investigated previously, raising the possibility that there may be amino acids within this region that are important for EC-LigA activity or that the function of essential residues proximal to this region are impacted by inhibitor interactions with this region. We anticipate that the identified small molecule binding sites and inhibitors could be pursued as part of an antibacterial strategy targeting bacterial DNA ligases.

Abstract: Objectives: This study tests, for the first time, the applicability of a new method of sex estimation utilizing enamel peptides on a sample of deciduous and permanent teeth at different stages of mineralization, from nonadults of unknown sex, including perinates., Materials and Methods: A total of 43 teeth from 29 nonadult individuals aged from 40 gestational weeks to 19 years old were analyzed. The sample included pairs of fully mineralized and just developing teeth from the same individual. The individuals were from four archaeological sites in England: Piddington (1st-2nd centuries AD), Coach Lane, Victoria Gate, and Fewston (all 18th-19th centuries). A method that identifies sex chromosome-linked isoforms of the peptide amelogenin from human tooth enamel was applied. The method utilizes a minimally destructive acid etching procedure and subsequent nano liquid chromatography tandem mass spectrometry., Results: It was possible to determine the sex of 28 of the nonadult individuals sampled (males = 20, females = 8, undetermined = 1). Only one sample failed (CL9), due to insufficient mineralization of the sampled tooth enamel. Data are available via ProteomeXchange with identifier PXD021683., Discussion: Sufficient peptide material to determine sex can be recovered even from the crowns of developing perinatal teeth that are not fully mineralized. The minimally destructive and inexpensive (compared to ancient DNA) nature of this procedure has significant implications for bioarchaeological studies of infancy and childhood., (© 2021 The Authors. Abstract: Objectives: Cortisol is a glucocorticoid hormone produced through activation of the hypothalamic pituitary adrenal axis. It is known as the "stress hormone" for its primary role in the body's stress response and has been the focus of much modern clinical research. Within archaeology, only a few studies have analyzed cortisol in human remains and these have been restricted to hair (Webb et al., 2010; Webb, White, van Uum, & Longstaffe, 2015a; Webb, White, van Uum, & Longstaffe, 2015b). This study examines the utility of dentine and enamel, which survive well archaeologically, as possible reservoirs for detectable levels of cortisol., Materials and Methods: Then, 69 teeth from 65 individuals from five Roman and Post-Roman sites in France were tested via competitive enzyme-linked immunosorbent assay (ELISA) to assess and quantify the cortisol concentrations present within tooth dentine and enamel., Results: In both tooth dentine and enamel, detectable concentrations of cortisol were identified in multiple teeth. However, concentrations were low and not all teeth yielded results that were measurable through cortisol ELISA. Differences in cortisol values between dentine and enamel could suggest different uptake mechanisms or timing., Discussion: These results suggest that cortisol is incorporated within tooth structures and merits further investigation in both modern and archaeological contexts. Analysis of the results through liquid chromatographic-mass spectrometry would verify current results and might yield values that could be better integrated with published cortisol studies. Future studies of cortisol in tooth structures would greatly expand the research potential of cortisol in the past and could have implications for studies of human stress across deep time., (© 2020 The Authors. 21. 'Til Poison Phosphorous Brought them Death': A potentially occupationally-related disease in a post-medieval skeleton from north-east England.

Abstract: This paper describes the pathological changes observed on the skeleton of a c.12-14 year old person buried in a north-east England Quaker cemetery dated to AD 1711-1857. Bone formation (woven and lamellar) and destruction are present mainly on the mandible, clavicles, sternum and scapulae, long bones of the right arm, left ribs, spine, ilia, and the femora and tibiae. Differential diagnoses of tuberculosis and other pulmonary diseases, smallpox, actinomycosis, neoplastic disease, and "phossy jaw" are considered. While the pathological changes could represent all previously described diseases and thus be associated with the insalubrious conditions in which this person lived, it is also possible that this person worked in the matchmaking industry known to be present in the region at the time. Attention is drawn to the previously overlooked condition "phossy jaw" caused by phosphorus poisoning, which was strongly associated with this industry. While matchstick making was an industry often associated with women and girls, DNA analysis of a bone sample from the skeleton did not successfully identify biological sex. Two dental calculus samples from this person were analysed for phosphorus, and comparisons were made with samples from the same and a different site; the levels did not indicate the person was more exposed to phosphorus than any of the other people at Coach Lane. However, the pathological lesions described also have relevance in a clinical context, because "phossy jaw" has been observed in living populations, arising as a consequence of ingesting phosphorous contained within some pharmaceuticals used for treating neoplastic disease and osteoporosis., (Copyright © 2015 Elsevier Inc. 22. Gendered Differences in Accidental Trauma to Upper and Lower Limb Bones at Aquincum, Roman Hungary.

Abstract: It was hypothesized that men and women living in the border provinces of the Roman Empire may have encountered different risks associated with their different occupations and activities. Limb bone trauma data were used to assess sex-based differences in physical hazards and evidence for fracture healing and treatment. Two hundred and ten skeletons were examined from a late 1st to early 4th century AD cemetery at Aquincum (Budapest, Hungary). Upper and lower limb bone fracture types, frequencies, distributions, and associated complications were recorded, and gendered patterns in injury risks were explored. Of the 23 fractures identified, both sexes had injuries indicative of falls; males exhibited the only injuries suggestive of higher-energy and more direct forces. Most fractures were well-healed with few complications. The extremity trauma at Aquincum suggests that people buried here experienced less hazardous physical activities than at other Roman provincial sites. The patterns of trauma indicate the occurrence of "traditional" gender roles, whereby male civilians participated in more physically dangerous activities than females. Additionally, treatment may have been equally accessible to men and women, but certain fracture types proved more challenging to reduce using the techniques available., (Copyright © 2015 Elsevier Inc. 23. An unusual exostotic lesion of the maxillary sinus from Roman Lincoln.

Abstract: This report provides a differential diagnosis of an exostotic bony lesion within the left maxillary sinus of a Romano-British (3rd to 4th century AD) adult male from Newport, Lincoln. Macroscopic, radiographic, and cone beam computed tomography (CBCT) analyses suggest that the lesion is likely of odontogenic origin. The overall size of the lesion and areas of sclerosis and radiolucency, together with its hypothesised odontogenic origin, suggest that the lesion represents a chronic exostotic osteomyelitic reaction to the presence of odontogenic bacteria. While modern case studies of odontogenic maxillary sinus osteomyelitis are noteworthy, published cases of this condition are extremely rare in an archaeological context and may be underreported due to the enclosed nature of the sinuses. Such infections may have serious implications for individual and population health, and non-destructive investigation should be considered in cases where significant maxillary caries are present., (Copyright © 2015 Elsevier Inc. 24. The proximal ulna as an additional diagnostic feature of advanced rheumatoid arthritis.

Abstract: Rheumatoid arthritis (RA) is considered to be an uncommon condition in paleopathology, although several case studies have recently been published. These studies tend to focus on changes in the small joints of the hands and feet, which are the most diagnostic, though these skeletal elements are often poorly preserved in archaeological contexts. This study aims to highlight another common trait that has been observed in multiple cases of RA in the clinical and paleopathological record: erosive lesions on the proximal ulna. RA frequently affects the elbow in clinical cases, with erosions observable in the radial head, the proximal ulna, and the distal humerus, in the later stages of the disease. Lesions produced by RA in the proximal ulna demonstrate a remarkable consistency in appearance between paleopathological cases from different periods and places. Although overlooked, erosive lesions on the proximal ulna provide an important diagnostic indicator of RA and yields additional information concerning disease progression., (Copyright © 2015 Elsevier Inc. Abstract: This study compares the infant mortality profiles of 128 infants from two urban and two rural cemetery sites in medieval England. The aim of this paper is to assess the impact of urbanization and industrialization in terms of endogenous or exogenous causes of death. In order to undertake this analysis, two different methods of estimating gestational age from long bone lengths were used: a traditional regression method and a Bayesian method. The regression method tended to produce more marked peaks at 38 weeks, while the Bayesian method produced a broader range of ages and were more comparable with the expected "natural" mortality profiles.At all the sites, neonatal mortality (28-40 weeks) outweighed post-neonatal mortality (41-48 weeks) with rural Raunds Furnells in Northamptonshire, showing the highest number of neonatal deaths and post-medieval Spitalfields, London, showing a greater proportion of deaths due to exogenous or environmental factors. Of the four sites under study, Wharram Percy in Yorkshire showed the most convincing "natural" infant mortality profile, suggesting the inclusion of all births at the site (i.e., stillbirths and unbaptised infants)., ((c) 2007 Wiley-Liss, Inc.)

Abstract: In the last ten years, layer of protection analysis (LOPA) emerged as a simplified form of quantitative risk assessment (QRA). The European Commission funded project Accidental Risk Assessment Methodology for Industries in the context of the Seveso 2 Directive (ARAMIS) has recently been completed. ARAMIS has several modules which give a consistent simplified approach to risk assessment which does not approach the complexity or expense of full QRA. LOPA is potentially a means of carrying out the assessment of barriers required in ARAMIS. This paper attempts to explain the principles of LOPA and the means by which it can be used within ARAMIS.
