Subtropical marine stratus clouds regulate coastal and global climate, but future trends in these clouds are uncertain. In coastal Southern California (CSCA), interannual variations in summer stratus cloud occurrence are spatially coherent across 24 airfields and dictated by positive relationships with stability above the marine boundary layer (MBL) and MBL height. Trends, however, have been spatially variable since records began in the mid-1900s due to differences in nighttime warming. Among CSCA airfields, differences in nighttime warming, but not daytime warming, are strongly and positively related to fraction of nearby urban cover, consistent with an urban heat island effect. Nighttime warming raises the near-surface dew point depression, which lifts the altitude of condensation and cloud base height, thereby reducing fog frequency. Continued urban warming, rising cloud base heights, and associated effects on energy and water balance would profoundly impact ecological and human systems in highly populated and ecologically diverse CSCA.

Six decades of observations at 20 coastal airports, from Alaska to southern California, reveal coherent interannual to interdecadal variation of coastal low cloudiness (CLC) from summer to summer over this broad region. The leading mode of CLC variability represents coherent variation, accounting for nearly 40% of the total CLC variance spanning 1950–2012. This leading mode and the majority of individual airports exhibit decreased low cloudiness from the earlier to the later part of the record. Exploring climatic controls on CLC, we identify North Pacific Sea Surface Temperature anomalies, largely in the form of the Pacific Decadal Oscillation (PDO) as well correlated with, and evidently helping to organize, the coherent patterns of summer coastal cloud variability. Links from the PDO to summer CLC appear a few months in advance of the summer. These associations hold up consistently in interannual and interdecadal frequencies.

Observations from four periods (three late springs and one early summer) at temperate forest sites in western and eastern Canada offer the first estimation of how the concentrations of submicron forest organic aerosol mass (SFOM) from the oxidation of biogenic volatile organic compounds (BVOC) vary over the ambient temperature range of 7 °C to 34 °C. For the measurement conditions of clear skies, low oxides of nitrogen and within approximately one day of emissions, 50 estimates of SFOM concentrations show the concentrations increase exponentially with temperature. The model that is commonly used to define terpene emissions as a function of temperature is able to constrain the range of the SFOM values across the temperature range. The agreement of the observations and model is improved through the application of an increased yield of SFOM as the organic mass concentration increases with temperature that is based on results from chamber studies. The large range of SFOM concentrations at higher temperatures leaves open a number of questions, including the relative contributions of changing yield and of isoprene, that may be addressed by more ambient observations at higher temperatures.

Ambient particles collected on teflon filters at the Peak of Whistler Mountain, British Columbia (2182 m a.s.l.) during spring and summer 2009 were measured by Fourier transform infrared (FTIR) spectroscopy for organic functional groups (OFG). The project mean and standard deviation of organic aerosol mass concentrations (OM) for all samples was 3.2±3.3 (μg m−3). Measurements of aerosol mass fragments, size, and number concentrations were used to separate fossil-fuel combustion and burning and non-burning forest sources of the measured organic aerosol. The OM was composed of the same anthropogenic and non-burning forest components observed at Whistler mid-valley in the spring of 2008; during the 2009 campaign, biomass burning aerosol was additionally observed from fire episodes occurring between June and September. On average, organic hydroxyl, alkane, carboxylic acid, ketone, and primary amine groups represented 31 %±11 %, 34 %±9 %, 23 %±6 %, 6 %±7 %, and 6 %±3 % of OM, respectively. Ketones in aerosols were associated with burning and non-burning forest origins, and represented up to 27 % of the OM. The organic aerosol fraction resided almost entirely in the submicron fraction without significant diurnal variations. OM/OC mass ratios ranged mostly between 2.0 and 2.2 and O/C atomic ratios between 0.57 and 0.76, indicating that the organic aerosol reaching the site was highly aged and possibly formed through secondary formation processes.

Submicron particles collected at Whistler, British Columbia, at 1020 m a.s.l. during May and June 2008 on Teflon filters were analyzed by Fourier transform infrared (FTIR) and X-ray fluorescence (XRF) techniques for organic functional groups (OFG) and elemental composition. Organic mass (OM) concentrations ranged from less than 0.5 to 3.1 μg m−3, with a project mean and standard deviation of 1.3±1.0 μg m−3 and 0.21±0.16 μg m−3 for OM and sulfate, respectively. On average, organic hydroxyl, alkane, and carboxylic acid groups represented 34%, 33%, and 23% of OM, respectively. Ketone, amine and organosulfate groups constituted 6%, 5%, and

Extreme events can be defined in many ways. Typical definitions of weather and climate extremes consider either the maximum value during a specified time interval (such as season or year) or exceedance of a threshold (the “peaks-over-threshold” [POT] approach), in which universal rather than local thresholds are frequently applied. For example, temperatures above 95°F (35°C) are often considered extreme in most locations across the United States, except in areas such as the low-lying deserts of Arizona and California, where such temperatures are typical in the summer. Temperatures at these levels are obviously extreme for living organisms from a non-adapted, physiological perspective, and technological adaptation for humans is required for day-to-day functioning in such temperatures. But such temperatures are not necessarily extreme from the statistical or local climate perspectives. In statistics, extremes are considered low-probability events that differ greatly from typical occurrences. The IPCC defines extremes as 1% to 10% of the largest or smallest values of a distribution (Trenberth et al. 2007). Studies over large or complex regions marked by significant climatic variation require definitions that are relevant to local climate. Across the Southwest, location-specific definitions of extreme temperature, precipitation, humidity, and wind are required if a meaningful region-wide perspective is desired.

[1] The winters of 2009–2010 and 2010–2011 brought frigid temperatures to parts of Europe, Russia, and the U.S. We analyzed regional and Northern Hemispheric (NH) daily temperature extremes for these two consecutive winters in the historical context of the past 63 years. While some parts clearly experienced very cold temperatures, the NH was not anomalously cold. Extreme warm events were much more prevalent in both magnitude and spatial extent. Importantly, the persistent negative state of the North Atlantic Oscillation (NAO) explained the bulk of the observed cold anomalies, however the warm extremes were anomalous even accounting for the NAO and also considering the states of the Pacific Decadal Oscillation (PDO) and El Nino Southern Oscillation (ENSO). These winters' widespread and intense warm extremes together with a continuing hemispheric decline in cold snap activity was a pattern fully consistent with a continuation of the warming trend observed in recent decades.

Biologically active extracts of the Caribbean sponge Agelas conifera have yielded, in exhaustive studies, the diacetate salts of seven new bromopyrroles (1,3-8), as well as that of the known debromooroidin dimer sceptrin (2). These compounds were found to be antiviral and antibacterial and were active in barnacle settlement and biochemical prophage induction assays. The structures assigned were based on spectroscopic comparisons to sceptrin and two-dimensional NMR data. Synthetic bromopyrroles were used to verify bromine substitution patterns. The oxysceptrins (4,5) are characterized by their aminoimidazolinone group, the ageliferins (6-8) by a unique cyclohexene-based skeleton

Morning Glory syndrome is an uncommon congenital optic disc anomaly with occasional systemic associations. A case of unsuspected difficult intubation in a three-year old patient is described in this case report.

Intravenous cannulation is obtained in almost all patients scheduled for operative intervention under anaesthesia. In our practice, inhalational induction precedes cannulation in children in order to avoid pain and discomfort, and cannulation is delayed until the child is adequately anaesthetized in fear of precipitating laryngospasm due to painful stimulus of venepuncture in the light stage of anaesthesia. This study was performed on 150 patients between two to eight years of age to determine if there is a difference in the incidence of untoward incidents, if cannulation is performed when children are lightly anaesthetized (Early, Group E), as compared to when they are deeply anaesthetized (Late, Group L). In patients randomized to early cannulation, the results showed that there was a significantly shorter time from induction to venous cannulation, the halothane concentration was lower at the time of cannulation, there was a greater incidence of movement on cannulation and a greater incidence of changes in heart rate, blood pressure, and respiratory rate. There was no significant differences in the incidence of laryngospasm or in the success rate of intravenous cannulation between the two groups. We conclude that venous cannulation can be safely performed during the light stages of anaesthesia.

The antimitotic depsipeptide cryptophycin 1 (CP1) was compared to the antimitotic peptide dolastatin 10 (D10) as an antiproliferative agent and in its interactions with purified tubulin. The potent activity of CP1 as an inhibitor of cell growth was confirmed. The agent had an IC50 of 20 pM against L1210 murine leukemia cells versus 0.5 nM for D10. Both drugs were comparable as inhibitors of the glutamate-induced assembly of purified tubulin, with D10 being slightly more potent. CP1, like D10, was a noncompetitive inhibitor of the binding of [3H]vinblastine to tubulin (apparent Ki, 3.9 microM); and the depsipeptide was a competitive inhibitor of the binding of [3H]D10 to tubulin (apparent Ki, 2.1 microM). CP1 was less potent than D10 as an inhibitor of nucleotide exchange on tubulin, but the two drugs were equivalent in stabilizing the colchicine binding activity of tubulin. CP1, like D10, caused the formation of extensive structured aggregates of tubulin when present in stoichiometric amounts relative to the protein. Whereas at lower concentrations the drugs were equivalent in causing formation of small oligomers detected by gel permeation high-performance liquid chromatography, there were notable differences in the aggregation reactions induced by the two drugs. The electron micrographic appearance of the D10-induced aggregate differed substantially from that of the CP1-induced aggregate. With D10, but not CP1, aggregate morphology was greatly altered in the presence of microtubule-associated proteins. Finally, although CP1 caused the formation of massive aggregates, as did D10, there was little turbidity change with the depsipeptide as opposed to the peptide.

Pneumocandin D0 (9), a new member of the echinocandin class of antifungal agents, has been isolated as a minor constituent from fermentation broths of the filamentous fungi Zalerion arboricola (ATCC 20957). The structure of 9 has been determined mainly on the basis of spectroscopic analysis and by comparison with published data for similar compounds. To date, pneumocandin D0 has been found to be the most potent inhibitor of Pneumocystis carinii development in vivo within the natural-occurring echinocandin family of antifungal agents.

In May 1988, the Centers for Disease Control's Model Performance Evaluation Program (Atlanta, Ga) surveyed 1092 laboratories that performed enzyme immunoassays and Western blot tests for human immunodeficiency virus type 1 antibody on mailed plasma samples of known human immunodeficiency virus type 1 antibody reactivity and that described their laboratory characteristics and testing practices. The study objective was to evaluate the quality of laboratory performance in testing for human immunodeficiency virus type 1 antibody. After identifying relevant variables in univariate analyses, multivariate analyses were performed using stepwise logistic models. Human immunodeficiency virus type 1 antibody test performance was independently associated with analytic variables such as commercial test kit used and with nonanalytic variables such as experience, training, and degree requirements of laboratory personnel. These results validate the importance of nonanalytic variables to the quality of outcomes in laboratory testing.

Retrieval analysis of two total knee replacements incorporating porous-coated components for biologic fixation was performed. The significant finding in a 63-year-old woman was fracture of the porous-coated metal backing of the tibial component, secondary to aseptic loosening. The significant findings at the time of revision surgery in an 82-year-old woman were nonuniform bone ingrowth (primarily around fixation pegs) and fracture of metal beads from the metal backing of the patellar component.

Most DNA intercalators and epipodophyllotoxins inhibit mammalian topoisomerase II by trapping the enzyme within DNA cleavage complexes that can be detected in cells as protein-associated DNA strand breaks. We have characterized previously a line of Chinese hamster cells (DC3F/9-OHE cells) the resistance of which to the cytotoxic effect of intercalators and etoposide is associated with a reduced formation of protein-associated DNA strand breaks. In the present study, topoisomerases of these cells were compared to those of the parental sensitive cells (DC3F). NaCl extracts (0.35 M) of isolated DC3F/9-OHE nuclei did not form 4'-(9-acridinylamino)methanesulfon-m-anisidide-induced DNA-protein linking, whereas DC3F nuclear extracts did. In addition, DC3F/9-OHE nuclear extract had an unusually high level of DNA linking activity in the absence of 4'-(9-acridinylamino)methanesulfon-m-anisidide. Topoisomerases II from DC3F/9-OHE and DC3F nuclei appeared similar qualitatively. DC3F/9-OHE nuclear extract had approximately twice less topoisomerase II molecules than did DC3F nuclear extract but similar topoisomerase II activity. Topoisomerase I activities appeared also similar in sensitive and resistant cells. However, part of DC3F/9-OHE topoisomerase I copurified with a DNA linking activity which was not present in DC3F nuclei. This unusual DNA linking activity was not sensitive to the stimulatory effect of 4'-(9-acridinylamino)methanesulfon-m-anisidide.

DNA intercalating drugs and the epipodophyllotoxins etoposide and teniposide interfere with the action of mammalian DNA topoisomerase II by trapping an intermediate complex of the enzyme covalently linked to the 5'-termini of DNA breaks. This effect can be observed in intact cells by alkaline elution measurement of protein-associated DNA strand breaks. To assess the cytotoxic role of this effect, we have studied a subline of DC3F Chinese hamster lung cells selected for resistance to the intercalating agent 9-hydroxyellipticine. This subline (DC3F/9-OHE) was cross-resistant to other intercalators as well as to etoposide. Resistance to Adriamycin was associated with reduced uptake. However, resistance to 4'-(9-acridinylamino)methanesulfon-m-aniside and 2-methyl-9-hydroxyellipticinium was observed in the absence of changes in drug uptake, suggesting a second mode of resistance. DC3F/9-OHE cells formed fewer protein-associated DNA strand breaks in response to 4'-(9-acridinylamino)methanesulfon-m-aniside, 2-methyl-9-hydroxyellipticinium, or etoposide than did the sensitive parental cells. The same was true for isolated nuclei from these cells, which is consistent with a mode of resistance unrelated to drug uptake through the plasma membrane. These data suggest that resistance to DNA topoisomerase II inhibitors exhibited by DC3F/9-OHE cells is due in part to a modification of topoisomerase II activity.
