The Mission Support System (MSS) is an open source software package that has been used for planning flight tracks of scientific aircraft in multiple measurement campaigns during the last decade. It consists of three major components: a web map server located close to the model data storage site that is capable of producing a variety of 2-D figures from 4-D meteorological data; a client application capable of displaying the figures in combination with the planned flight track and an assortment of additional information; and a new collaboration server component that enables real-time collaboration of multiple remote parties. During the last decade, these components were constantly improved towards being simple to set up and use and being standard compliant. Here, we describe the use of MSS during the Southern Hemisphere Transport, Dynamics, and Chemistry–Gravity Waves (SouthTRAC-GW) measurement campaign in 2019. This campaign, based in Rio Grande, Argentina, used the German research aircraft HALO to investigate several scientific objectives related to the Southern Hemisphere chemistry and dynamics. We present the diverse data products offered by the MSS web map server dedicated to the campaign, which were derived from the European Centre for Medium-Range Weather Forecasts (ECMWF) forecast data, Chemical Lagrangian Model of the Stratosphere (CLaMS) simulations, and Atmospheric Infrared Sounder (AIRS) near-real time brightness temperature measurements. As an example for how the MSS software is used in conjunction with the different data sets, we describe the planning of a single flight, which eventually took place on 12 September 2019, probing orographic gravity waves propagating up into the lower mesosphere.

Intro: Performance of SARS-CoV-2 antigen tests is described relative to RT-PCR cycle threshold (Ct) values. However, Ct values may not be consistent between tests from different manufacturers, or even between runs. Here we correlate the Roche Elecsys SARS-CoV-2 Antigen assay results to a quantitative RT-PCR readout as a more reproducible measure of viral load. Additionally, we look at the relationship between the antigen test results, viral load and infectious titer. Methods: Longitudinal nasopharyngeal swab samples from patients (N=452) with severe Covid-19 pneumonia collected between 03 April and 28 May 2020 in a randomized, double-blind, placebo controlled, multicenter study to evaluate the safety and efficacy of Tocilizumab (COVACTA), were assessed for SARS-CoV-2 viral load (RNA copies/ mL) and a qualitative and semi-quantitative readout of Elecsys SARS-CoV-2 Antigen assay. Viral culture experiments were performed to determine the infectious titer (TCID50/ mL) in a subset of samples. Agreement analysis was performed to compare the results of the assays. Please note that the current intended use of Elecsys SARS-CoV-2 Antigen assay is the qualitative detection of SARS-CoV-2 antigen. Findings: We observed high negative percent agreement between the Elecsys SARS-CoV-2 Antigen assay results and the RT-PCR results, while the positive percent agreement was only high in samples exceeding a certain viral load and at earlier time points from symptom onset. Infectious titer values and both the antigen assay semi-quantitative readout and the quantitative RT-PCR results correlated well. Positive percent agreement of RT-PCR and antigen results in relation to infectious titer was very high in both cases, while negative percent agreement was moderate to low. Conclusion: These data show that in patients with high viral load the Elecsys SARS-CoV-2 Antigen assay correlates well qualitatively and quantitatively with the presence of SARS-CoV-2 RNA and infectious virus as determined by RT-PCR and viral culture, respectively.

This study presents a framework for regional smart energy planning for the optimal location and sizing of small hybrid systems. By using an optimization model – in combination with weather data – various local energy systems are simulated using the Calliope and PyPSA energy system simulation tools. The optimization and simulation models are fed with GIS data from different volunteered geographic information projects, including OpenStreetMap. These allow automatic allocation of specific demand profiles to diverse OpenStreetMap building categories. Moreover, based on the characteristics of the OpenStreetMap data, a set of possible distributed energy resources, including renewables and fossil-fueled generators, is defined for each building category. The optimization model can be applied for a set of scenarios based on different assumptions on electricity prices and technologies. Moreover, to assess the impact of the scenarios on the current distribution infrastructure, a simulation model of the low- and medium-voltage network is conducted. Finally, to facilitate their dissemination, the results of the simulation are stored in a PostgreSQL database, before they are delivered by a RESTful Laravel Server and displayed in an angular web application.

The annual variation of δD in the tropical lower stratosphere is a critical indicator for the relative importance of different processes contributing to the transport of water vapour through the cold tropical tropopause region into the stratosphere. Distinct observational discrepancies of the δD annual variation were visible in the works of Steinwagner et al. (2010) and Randel et al. (2012). Steinwagner et al. (2010) analysed MIPAS (Michelson Interferometer for Passive Atmospheric Sounding) observations retrieved with the IMK/IAA (Institut für Meteorologie und Klimaforschung in Karlsruhe, Germany, in collaboration with the Instituto de Astrofísica de Andalucía in Granada, Spain) processor, while Randel et al. (2012) focused on ACE-FTS (Atmospheric Chemistry Experiment Fourier Transform Spectrometer) observations. Here we reassess the discrepancies based on newer MIPAS (IMK/IAA) and ACE-FTS data sets, also showing for completeness results from SMR (Sub-Millimetre Radiometer) observations and a ECHAM/MESSy (European Centre for Medium-Range Weather Forecasts Hamburg and Modular Earth Submodel System) Atmospheric Chemistry (EMAC) simulation (Eichinger et al., 2015b). Similar to the old analyses, the MIPAS data set yields a pronounced annual variation (maximum about 75 ‰), while that derived from the ACE-FTS data set is rather weak (maximum about 25 ‰). While all data sets exhibit the phase progression typical for the tape recorder, the annual maximum in the ACE-FTS data set precedes that in the MIPAS data set by 2 to 3 months. We critically consider several possible reasons for the observed discrepancies, focusing primarily on the MIPAS data set. We show that the δD annual variation in the MIPAS data up to an altitude of 40 hPa is substantially impacted by a “start altitude effect”, i.e. dependency between the lowermost altitude where MIPAS retrievals are possible and retrieved data at higher altitudes. In itself this effect does not explain the differences with the ACE-FTS data. In addition, there is a mismatch in the vertical resolution of the MIPAS HDO and H2O data (being consistently better for HDO), which actually results in an artificial tape-recorder-like signal in δD. Considering these MIPAS characteristics largely removes any discrepancies between the MIPAS and ACE-FTS data sets and shows that the MIPAS data are consistent with a δD tape recorder signal with an amplitude of about 25 ‰ in the lowermost stratosphere.

Abstract Background Nonadherence with mood stabilizers is a major problem that negatively impacts the course of bipolar disorder. Medication adherence is a complex individual behavior, and adherence rates often change over time. This study asked if distinct classes of adherence trajectories with mood stabilizers over time could be found, and if so, which patient characteristics were associated with the classes. Methods This analysis was based on 12 weeks of daily self-reported data from 273 patients with bipolar 1 or II disorder using ChronoRecord computer software. All patients were taking at least one mood stabilizer. The latent class mixed model was used to detect trajectories of adherence based on 12 weekly calculated adherence datapoints per patient. Results Two distinct trajectory classes were found: an adherent class (210 patients; 77%) and a less adherent class (63 patients; 23%). Within the framework of the second SPARC (Stratosphere-troposphere Processes And their Role in Climate) water vapour assessment (WAVAS-II), we evaluated five data sets of δD(H2O) obtained from observations by Odin/SMR (Sub-Millimetre Radiometer), Envisat/MIPAS (Environmental Satellite/Michelson Interferometer for Passive Atmospheric Sounding), and SCISAT/ACE-FTS (Science Satellite/Atmospheric Chemistry Experiment – Fourier Transform Spectrometer) using profile-to-profile and climatological comparisons. These comparisons aimed to provide a comprehensive overview of typical uncertainties in the observational database that could be considered in the future in observational and modelling studies. Our primary focus is on stratospheric altitudes, but results for the upper troposphere and lower mesosphere are also shown. There are clear quantitative differences in the measurements of the isotopic ratio, mainly with regard to comparisons between the SMR data set and both the MIPAS and ACE-FTS data sets. In the lower stratosphere, the SMR data set shows a higher depletion in δD than the MIPAS and ACE-FTS data sets. The differences maximise close to 50 hPa and exceed 200 ‰. With increasing altitude, the biases decrease. Above 4 hPa, the SMR data set shows a lower δD depletion than the MIPAS data sets, occasionally exceeding 100 ‰. Overall, the δD biases of the SMR data set are driven by HDO biases in the lower stratosphere and by H2O biases in the upper stratosphere and lower mesosphere. In between, in the middle stratosphere, the biases in δD are the result of deviations in both HDO and H2O. These biases are attributed to issues with the calibration, in particular in terms of the sideband filtering, and uncertainties in spectroscopic parameters. The MIPAS and ACE-FTS data sets agree rather well between about 100 and 10 hPa. The MIPAS data sets show less depletion below approximately 15 hPa (up to about 30 ‰), due to differences in both HDO and H2O. Higher up this behaviour is reversed, and towards the upper stratosphere the biases increase. This is driven by increasing biases in H2O, and on occasion the differences in δD exceed 80 ‰. Below 100 hPa, the differences between the MIPAS and ACE-FTS data sets are even larger. In the climatological comparisons, the MIPAS data sets continue to show less depletion in δD than the ACE-FTS data sets below 15 hPa during all seasons, with some variations in magnitude. The differences between the MIPAS and ACE-FTS data have multiple causes, such as differences in the temporal and spatial sampling (except for the profile-to-profile comparisons), cloud influence, vertical resolution, and the microwindows and spectroscopic database chosen. Differences between data sets from the same instrument are typically small in the stratosphere. Overall, if the data sets are considered together, the differences in δD among them in key areas of scientific interest (e.g. tropical and polar lower stratosphere, lower mesosphere, and upper troposphere) are too large to draw robust conclusions on atmospheric processes affecting the water vapour budget and distribution, e.g. the relative importance of different mechanisms transporting water vapour into the stratosphere.

Resistance towards VEGF-centered anti-angiogenic therapy is an important clinical challenge. Here, the authors show that mast cells mediate resistance to anti-angiogenetic inhibitors by altering the proliferative and organizational state of endothelial cells through mobilization of FGF-1 and GM-CSF from the tumor matrix and secretion of FGF-2.

The AquaVIT-1 intercomparison of atmospheric water vapor measurement techniques was conducted at the aerosol and cloud simulation chamber AIDA (Aerosol Interaction and Dynamics in the Atmosphere) at the Karlsruhe Institute of Technology, Germany, in October 2007. The overall objective was to intercompare state-of-the-art and prototype atmospheric hygrometers with each other and with independent humidity standards under controlled conditions. This activity was conducted as a blind intercomparison with coordination by selected referees. The effort was motivated by persistent discrepancies found in atmospheric measurements involving multiple instruments operating on research aircraft and balloon platforms, particularly in the upper troposphere and lower stratosphere, where water vapor reaches its lowest atmospheric values (less than 10 ppm). With the AIDA chamber volume of 84 m3, multiple instruments analyzed air with a common water vapor mixing ratio, by extracting air into instrument flow systems, by locating instruments inside the chamber, or by sampling the chamber volume optically. The intercomparison was successfully conducted over 10 days during which pressure, temperature, and mixing ratio were systematically varied (50 to 500 hPa, 185 to 243 K, and 0.3 to 152 ppm). In the absence of an accepted reference instrument, the absolute accuracy of the instruments was not established. To evaluate the intercomparison, the reference value was taken to be the ensemble mean of a core subset of the measurements. For these core instruments, the agreement between 10 and 150 ppm of water vapor is considered good with variation about the reference value of about ±10% (±1σ). In the region of most interest between 1 and 10 ppm, the core subset agreement is fair with variation about the reference value of ±20% (±1σ). The upper limit of precision was also derived for each instrument from the reported data. The implication for atmospheric measurements is that the substantially larger differences observed during in-flight intercomparisons stem from other factors associated with the moving platforms or the non-laboratory environment. The success of AquaVIT-1 provides a template for future intercomparison efforts with water vapor or other species that are focused on improving the analytical quality of atmospheric measurements on moving platforms.

Record breaking loss of ozone (O3) in the Arctic stratosphere has been reported in winter–spring 2010/2011. We examine in detail the composition and transformations occurring in the Arctic polar vortex using total column and vertical profile data products for O3, bromine oxide (BrO), nitrogen dioxide (NO2), chlorine dioxide (OClO), and polar stratospheric clouds (PSC) retrieved from measurements made by SCIAMACHY (Scanning Imaging Absorption SpectroMeter for Atmospheric CHartography) on-board Envisat (Environmental Satellite), as well as total column ozone amount, retrieved from the measurements of GOME-2 (Global Ozone Monitoring Experiment) on MetOp-A (Meteorological Experimental Satellite). Similarly we use the retrieved data from DOAS (Differential Optical Absorption Spectroscopy) measurements made in Ny-Ålesund (78.55° N, 11.55° E). A chemical transport model (CTM) has been used to relate and compare Arctic winter–spring conditions in 2011 with those in the previous year. In late winter–spring 2010/2011 the chemical ozone loss in the polar vortex derived from SCIAMACHY observations confirms findings reported elsewhere. More than 70% of O3 was depleted by halogen catalytic cycles between the 425 and 525 K isentropic surfaces, i.e. in the altitude range ~16–20 km. In contrast, during the same period in the previous winter 2009/2010, a typical warm Arctic winter, only slightly more than 20% depletion occurred below 20 km, while 40% of O3 was removed above the 575 K isentrope (~23 km). This loss above 575 K is explained by the catalytic destruction by NOx descending from the mesosphere. In both Arctic winters 2009/2010 and 2010/2011, calculated O3 losses from the CTM are in good agreement to our observations and other model studies. The mid-winter 2011 conditions, prior to the catalytic cycles being fully effective, are also investigated. Surprisingly, a significant loss of O3 around 60%, previously not discussed in detail, is observed in mid-January 2011 below 500 K (~19 km) and sustained for approximately 1 week. The low O3 region had an exceptionally large spatial extent. The situation was caused by two independently evolving tropopause elevations over the Asian continent. Induced adiabatic cooling of the stratosphere favoured the formation of PSC, increased the amount of active chlorine for a short time, and potentially contributed to higher polar ozone loss later in spring.

The increasing amounts of reactive nitrogen in the stratosphere necessitate accurate global measurements of stratospheric nitrogen dioxide (NO2). Over the past decade, the SCIAMACHY (SCanning Imaging Absorption spectroMeter for Atmospheric CHartographY) instrument on ENVISAT (European Environmental Satellite) has been providing global coverage of stratospheric NO2 every 6 days. In this study, the vertical distributions of NO2 retrieved from SCIAMACHY limb measurements of the scattered solar light are validated by comparison with NO2 products from three different satellite instruments (SAGE II, HALOE and ACE-FTS). The retrieval algorithm based on the information operator approach is discussed, and the sensitivity of the SCIAMACHY NO2 limb retrievals is investigated. The photochemical corrections needed to make this validation feasible, and the chosen collocation criteria are described. For each instrument, a time period of two years is analyzed with several hundreds of collocation pairs for each year. As NO2 is highly variable, the comparisons are performed for five latitudinal bins and four seasons. In the 20 to 40 km altitude range, mean relative differences between SCIAMACHY and other instruments are found to be typically within 20 to 30%. The mean partial NO2 columns in this altitude range agree typically within 15% (both global monthly and zonal annual means). Larger differences are seen for SAGE II comparisons, which is consistent with the results presented by other authors. For SAGE II and ACE-FTS, the observed differences can be partially attributed to the diurnal effect error.

This study reports the first systematic measurements of nitric acid (HNO3) uptake in contrail ice particles at typical aircraft cruise altitudes. During the CIRRUS-III campaign cirrus clouds and almost 40 persistent contrails were probed with in situ instruments over Germany and Northern Europe in November 2006. Besides reactive nitrogen, water vapor, cloud ice water content, ice particle size distributions, and condensation nuclei were measured during 6 flights. Contrails with ages up to 12 h were detected at altitudes 10–11.5 km and temperatures 211–220 K. These contrails had a larger ice phase fraction of total nitric acid (HNO3ice/HNO3tot = 6%) than the ambient cirrus layers (3%). On average, the contrails contained twice as much HNO3ice as the cirrus clouds, 14 pmol/mol and 6 pmol/mol, respectively. Young contrails with ages below 1 h had a mean HNO3ice of 21 pmol/mol. The contrails had higher nitric acid to water molar ratios in ice and slightly higher ice water contents than the cirrus clouds under similar meteorological conditions. The differences in ice phase fractions and molar ratios between developing contrails and cirrus are likely caused by high plume concentrations of HNO3 prior to contrail formation. The location of the measurements in the upper region of frontal cirrus layers might account for slight differences in the ice water content between contrails and adjacent cirrus clouds. The observed dependence of molar ratios as a function of the mean ice particle diameter suggests that ice-bound HNO3 concentrations are controlled by uptake of exhaust HNO3 in the freezing plume aerosols in young contrails and subsequent trapping of ambient HNO3 in growing ice particles in older (age > 1 h) contrails.

Upper tropospheric observations outside and inside of cirrus clouds indicate water vapour mixing ratios sometimes exceeding water saturation. Relative humidities over ice (RHice) of up to and more than 200% have been reported from aircraft and balloon measurements in recent years. From these observations a lively discussion continues on whether there is a lack of understanding of ice cloud microphysics or whether the water measurements are tainted with large uncertainties or flaws. Here, RHice in clear air and in ice clouds is investigated. Strict quality-checked aircraft in situ observations of RHice were performed during 28 flights in tropical, mid-latitude and Arctic field experiments in the temperature range 183–240 K. In our field measurements, no supersaturations above water saturation are found. Nevertheless, super- or subsaturations inside of cirrus are frequently observed at low temperatures (ice deviating from saturation, we analysed the number densities of ice crystals recorded during 20 flights. From the combined analysis – using conventional microphysics – of supersaturations and ice crystal numbers, we show that the high, persistent supersaturations observed inside of cirrus can possibly be explained by unexpected, frequent very low ice crystal numbers that could scarcely be caused by homogeneous ice nucleation. Heterogeneous ice formation or the suppression of freezing might better explain the observed ice crystal numbers. Thus, our lack of understanding of the high supersaturations, with implications for the microphysical and radiative properties of cirrus, the vertical redistribution of water and climate, is traced back to the understanding of the freezing process at low temperatures.
