Aspen and poplar trees are important horticultural plants grown in Canada for aesthetic, commercial woodlot and windbreak applications. Bronze leaf is a destructive disease in Populus spp. and is caused by the fungal pathogen Apioplagiostoma populi Barr. This pathogen is often difficult to isolate and confirm from infected plant tissues and has been mainly identified by disease symptoms and morphological characteristics of A. populi when fruiting bodies form on infected leaves or branches. Affected leaves and branches typically become necrotic and bronze in colour. Air-borne spores and nursery shipments containing infected plants play an important role in the efficient movement of the pathogen. In this study, bronze leaf disease samples from symptomatic trees in Canada were examined microscopically for A. populi perithecia and asci. Pathogen-specific genomic sequences were identified for the development of sensitive stringent diagnostics that indicated branches and petioles were the most effective tissues for detecting A. populi. Leaf samples from symptomatic trees were collected in Canada and examined for perithecia to microscopically characterize A. populi asci and ascospores. Disease associated DNA sequences of the internal transcribed spacer (ITS) 5.8S region of the nuclear ribosomal, β-tubulin 2, and translation elongation factor-1-γ chain, were isolated from perithecia and symptomatic tree samples. Morphological and molecular biological data from this study characterized the relationship and epidemiology of A. populi and enabled the development of rapid diagnostic methods that restrict the extent of further losses in amenity and commercial plantings of aspen and poplar.

Research suggests that children develop best in families. However, millions of children live in residential care centres worldwide. Many residential centres desire to transition their programs from a residential to a family-based model of care, but face barriers surrounding funding and donor support. Little research exists on how organisations address these concerns. The current article investigated the financial impact of transitioning to a family-based model of care, donor engagement practices used in this process, and changes in donor support resulting from the transition. Twenty-six organisations that had fully or partially transitioned their model completed a brief survey. Data revealed an initial increase in cost per child, but long-term the cost of services decreased. Further, findings suggested that involving donors early and using multiple methods and types of communication led to better long-term donor support. Based on these findings, five recommendations were made for organisations planning to transition their care model.

Objective To test the hypothesis that infants born Study design Randomized trial (3/2017-01/2019) at 5 NICUs. The primary outcome was CPAP failure; subgroup analyses (gestational age, receipt antenatal corticosteroids) were performed. Results A total of 232 infants were randomized. Infants in the Seattle-PAP and FP-CPAP groups had mean gestational ages of 27.0 and 27.2 weeks, respectively. Clinical Relevance Using the deep margin elevation technique in preparations extending beyond the cemento-enamel junction appears to be beneficial in maintaining structural integrity of CAD/CAM-fabricated feldspathic ceramic inlays. SUMMARY Objective: To evaluate the effect of deep margin elevation on structural and marginal integrity of ceramic inlays. Methods and Materials: Forty extracted human third molars were collected and randomly separated into four groups (n=10/group). In group 1 (enamel margin group), the gingival margin was placed 1 mm supragingival to the cemento-enamel junction (CEJ). In group 2 (cementum margin group), the gingival margin was placed 2 mm below the CEJ. In group 3 (glass ionomer [GI] margin group), the gingival margin was placed 2 mm below the CEJ, and then the margin elevated with GI to the CEJ. In group 4 (resin-modified glass ionomer [RMGI] margin group), the gingival margin was placed 2 mm below the CEJ, and then the margin elevated with RMGI to the CEJ. Standardized ceramic class II inlays were fabricated with computer-aided design/computer-aided manufacturing and bonded to all teeth, and ceramic proximal box heights were measured. All teeth were subjected to 10,000 cycles of thermocycling (5°C/55°C) and then underwent 1,200,000 cycles of vertical chewing simulation at 50 N of force. Ceramic restorations and marginal integrity were assessed with a Hirox digital microscope. The Fisher exact test (two-tailed) with adjusted p-values (α=0.05) and logistic regression were used for statistical analysis. Results: The cementum margin group had a significantly higher ceramic fracture rate (90%) compared to other groups (10% in enamel margin and GI margin groups, p=0.007; 0% in RMGI group, p Conclusion: Deep marginal elevation resulted in decreased ceramic fracture when preparation margins were located below the CEJ. There was no difference found between margin elevation with GI or RMGI. Increased heights of ceramic proximal box may lead to an increased probability of ceramic fracture.

Clubroot caused by Plasmodiophora brassicae is an important disease of brassica crops. The use of vital stains to determine the viability of P. brassicae resting spores can provide useful information regarding spore longevity, inoculum potential, or the efficacy of antimicrobial treatments. Evans blue is one example of a vital stain that has been reported to differentially stain viable and nonviable resting spores. Some previously published protocols using Evans blue to stain P. brassicae resting spores have not provided accurate or consistent results. In this study, we modified the Evans blue method by increasing the staining time to 8 h or more and evaluated P. brassicae resting spores after heat treatment at various combinations of temperature and time. Extending staining times significantly increased the numbers of stained resting spores up to 7 h, after which the numbers of stained spores did not change significantly (R2 = 96.88; P ≤ 0.001). The accuracy of the modified method to discriminate viable and nonviable spores was evaluated in repeated experiments and by comparing the staining data with those derived from inoculation assays and propidium monoazide quantitative PCR (qPCR). The results demonstrated that the modified Evans blue staining method improved the accuracy and consistency of measurement of P. brassicae resting spore viability. Additionally, it was equivalent to the qPCR method for differentiating viable and nonviable spores (R2 = 99.84; P ≤ 0.001) and confirmed in canola infection bioassays.

Background: At birth, the majority of neonates born at Methods: We designed a multicenter, non-blinded, randomized controlled trial that will enroll 230 premature infants (220/7 to 296/7 weeks of gestation). Infants will be randomized to receive Seattle-PAP or FP-CPAP. The primary outcome is respiratory failure requiring intubation and mechanical ventilation. Secondary outcomes include measures of short and long-term respiratory morbidity and cost effectiveness. Discussion: This trial will assess whether Seattle-PAP is more efficacious and cost effective than FP-CPAP in real-world practice among premature neonates. Trial Registration: This trial has been registered with the United States National Library of Medicine (www.clinicaltrials.gov, Trial Identifier #NCT03085329). Registered on 21 March 2017.

The goal of this project was to validate the functional relevance and utility of mucus produced by an in vitro intestinal cell culture model. This is facilitated by the need to physiologically replicate both healthy and abnormal mucus conditions from native intestinal tissue, where mucus properties have been connected to intestinal disease models. Mucus harvested from colonic cell cultures derived from healthy donors was compared to mucus collected from surgically resected, noninflamed transverse colon tissue. The rheological and biochemical properties of these mucus samples were compared using oscillational rheometry, particle-tracking microrheology, multiangle laser light scattering, refractometry, and immunohistochemical imaging. An air-liquid interface culture of primary human colonic epithelial cells generated a continuous monolayer with an attached mucus layer that displayed increasing weight percent (wt%) of solids over 1 week (1.3 ± 0.5% at 2 days vs. 2.4 ± 0.3% at 7 days). The full range of mucus concentrations (0.9–3.3%) observed during culture was comparable to that displayed by ex vivo mucus (1.3–1.9%). Bulk rheological measurements displayed similar wt%-based complex viscosities between in vitro and ex vivo mucus, with the complex viscosity of both systems increasing with wt% of solids. Particle-tracking microrheology showed higher complex viscosities for ex vivo mucus samples than in vitro mucus which was explained by a greater fraction of water present in in vitro mucus than ex vivo, i.e., in vitro mucus is more heterogeneous than ex vivo. Refractometry, multiangle laser light scattering, and immunostaining showed increased mucus complex size in ex vivo mucus compared with in vitro mucus, which may have been due to the admixture of mucus and cellular debris during ex vivo mucus collection. The air-liquid interface culture system produced intestinal mucus with similar composition and rheology to native human gut mucus, providing a platform to analyze pathological differences in intestinal mucus.

The Solar Orbiter spacecraft will be equipped with a wide range of remote-sensing (RS) and in situ (IS) instruments to record novel and unprecedented measurements of the solar atmosphere and the inner heliosphere. To take full advantage of these new datasets, tools and techniques must be developed to ease multi-instrument and multi-spacecraft studies. In particular the currently inaccessible low solar corona below two solar radii can only be observed remotely. Furthermore techniques must be used to retrieve coronal plasma properties in time and in three dimensional (3D) space. Solar Orbiter will run complex observation campaigns that provide interesting opportunities to maximise the likelihood of linking IS data to their source region near the Sun. Several RS instruments can be directed to specific targets situated on the solar disk just days before data acquisition. To compare IS and RS, data we must improve our understanding of how heliospheric probes magnetically connect to the solar disk. Aims. The aim of the present paper is to briefly review how the current modelling of the Sun and its atmosphere can support Solar Orbiter science. We describe the results of a community-led effort by European Space Agency's Modelling and Data Analysis Working Group (MADAWG) to develop different models, tools, and techniques deemed necessary to test different theories for the physical processes that may occur in the solar plasma. The focus here is on the large scales and little is described with regards to kinetic processes. To exploit future IS and RS data fully, many techniques have been adapted to model the evolving 3D solar magneto-plasma from the solar interior to the solar wind. A particular focus in the paper is placed on techniques that can estimate how Solar Orbiter will connect magnetically through the complex coronal magnetic fields to various photospheric and coronal features in support of spacecraft operations and future scientific studies. Methods. Recent missions such as STEREO, provided great opportunities for RS, IS, and multi-spacecraft studies. We summarise the achievements and highlight the challenges faced during these investigations, many of which motivated the Solar Orbiter mission. We present the new tools and techniques developed by the MADAWG to support the science operations and the analysis of the data from the many instruments on Solar Orbiter. Results. This article reviews current modelling and tool developments that ease the comparison of model results with RS and IS data made available by current and upcoming missions. It also describes the modelling strategy to support the science operations and subsequent exploitation of Solar Orbiter data in order to maximise the scientific output of the mission. Conclusions. The on-going community effort presented in this paper has provided new models and tools necessary to support mission operations as well as the science exploitation of the Solar Orbiter data. The tools and techniques will no doubt evolve significantly as we refine our procedure and methodology during the first year of operations of this highly promising mission. © 2020 A. P. The ultimate goal is to understand how the Sun produces and controls the heliosphere, filling the Solar System and driving the planetary environments. With six remote-sensing and four in-situ instrument suites, the coordination and planning of the operations are essential to address the following four top-level science questions: (1) What drives the solar wind and where does the coronal magnetic field originate?; (2) How do solar transients drive heliospheric variability?; (3) How do solar eruptions produce energetic particle radiation that fills the heliosphere?; (4) How does the solar dynamo work and drive connections between the Sun and the heliosphere? Maximising the mission's science return requires considering the characteristics of each orbit, including the relative position of the spacecraft to Earth (affecting downlink rates), trajectory events (such as gravitational assist manoeuvres), and the phase of the solar activity cycle. Furthermore, since each orbit's science telemetry will be downloaded over the course of the following orbit, science operations must be planned at mission level, rather than at the level of individual orbits. It is important to explore the way in which those science questions are translated into an actual plan of observations that fits into the mission, thus ensuring that no opportunities are missed. First, the overarching goals are broken down into specific, answerable questions along with the required observations and the so-called Science Activity Plan (SAP) is developed to achieve this. The SAP groups objectives that require similar observations into Solar Orbiter Observing Plans, resulting in a strategic, top-level view of the optimal opportunities for science observations during the mission lifetime. This allows for all four mission goals to be addressed. To meet the scientific objectives of the mission, the Solar Orbiter spacecraft carries a suite of in-situ (IS) and remote sensing (RS) instruments designed for joint operations with inter-instrument communication capabilities. Indeed, previous missions have shown that the Sun (imaged by the RS instruments) and the heliosphere (mainly sampled by the IS instruments) should be considered as an integrated system rather than separate entities. Many of the advances expected from Solar Orbiter rely on this synergistic approach between IS and RS measurements. Aims. Many aspects of hardware development, integration, testing, and operations are common to two or more RS instruments. In this paper, we describe the coordination effort initiated from the early mission phases by the Remote Sensing Working Group. We review the scientific goals and challenges, and give an overview of the technical solutions devised to successfully operate these instruments together. Methods. A major constraint for the RS instruments is the limited telemetry (TM) bandwidth of the Solar Orbiter deep-space mission compared to missions in Earth orbit. Hence, many of the strategies developed to maximise the scientific return from these instruments revolve around the optimisation of TM usage, relying for example on onboard autonomy for data processing, compression, and selection for downlink. The planning process itself has been optimised to alleviate the dynamic nature of the targets, and an inter-instrument communication scheme has been implemented which can be used to autonomously alter the observing modes. We also outline the plans for in-flight cross-calibration, which will be essential to the joint data reduction and analysis. Results. The RS instrument package on Solar Orbiter will carry out comprehensive measurements from the solar interior to the inner heliosphere. Thanks to the close coordination between the instrument teams and the European Space Agency, several challenges specific to the RS suite were identified and addressed in a timely manner. © 2020 F. Aims. We present the design and pre-launch performance of the Solar Orbiter Heliospheric Imager (SoloHI) which is an instrument prepared for inclusion in the ESA/NASA Solar Orbiter mission, currently scheduled for launch in 2020.Methods. The goal of this paper is to provide details of the SoloHI instrument concept, design, and pre-flight performance to give the potential user of the data a better understanding of how the observations are collected and the sources that contribute to the signal.Results. The paper discusses the science objectives, including the SoloHI-specific aspects, before presenting the design concepts, which include the optics, mechanical, thermal, electrical, and ground processing. Finally, a list of planned data products is also presented.Conclusions. The performance measurements of the various instrument parameters meet or exceed the requirements derived from the mission science objectives. SoloHI is poised to take its place as a vital contributor to the science success of the Solar Orbiter mission.

Surveys of alfalfa (Medicago sativa L.) seed production fields on the Canadian Prairies from 1993 to 2017 demonstrated that epidemics of blossom blight caused by Botrytis cinerea Pers. and Sclerotinia sclerotiorum (Lib.) de Bary developed almost every year at some location(s). Blossom blight symptoms were prominent when epidemics were severe, but more difficult to identify when present at low levels, which likely contributes to the infrequent reporting of blossom blight in alfalfa seed production. Epidemics were observed across the entire region, from the Peace River region of northern Alberta to southern Manitoba, consistently associated with relatively cool, wet weather conditions. The widespread nature of blossom blight on alfalfa in the years following its initial diagnosis in Canada in 1993, together with the ubiquitous distribution of the pathogens involved on other crops, indicated that blossom blight was almost certainly present on alfalfa seed crops on the Canadian Prairies for many years before the initial identification. It appears likely that blossom blight continues to cause yield loss at some locations in most years.

Objective. The role of the crypt microarchitecture and surrounding tissue curvature on intestinal stem/proliferative cell physiology is unknown. The utility of liquid lithography in creating polydimethylsiloxane (PDMS) micropillar stamps with controlled tip curvature was assessed. Using these stamps, the impact of microcurvature at the crypt base on intestinal cell and cytoskeletal behavior was studied. Approach. An SU-8 master mold as a support, polyols of varying surface energies as sacrificial liquids, and liquid PDMS as the solidifiable material were combined using liquid lithography to form PDMS micropillar arrays. Vapor phase deposition of organosilane onto the master mold was used to modify the surface energy of the master mold to shape the micropillar tips. Collagen was molded using the micropillar arrays forming a scaffold for culture of human primary colonic epithelial cells. Cell proliferation and cytoskeletal properties were assessed using fluorescent stains. Main results. Liquid lithography using low surface energy polyols (−1) generated convex-tipped PDMS micropillars, while polyols with higher surface energies (>55 dynes cm−1) yielded concave-tipped PDMS micropillars. Gradients of octyltrichlorosilane deposition across a master mold with an array of microwells yielded a PDMS micropillar array with a range of tip curvatures. Human primary colonic epithelial cells cultured on micropillar-molded collagen scaffolds demonstrated a stem/proliferative cell compartment at the crypt base. Crypts with a convex base demonstrated significantly lower cell proliferation at the crypt base than that of cells in crypts with either flat or concave bases. Crypts with a convex base also displayed higher levels of G-actin activity compared to that of crypts with flat or concave bases. Significance. Liquid lithography enabled creation of arrays of in vitro colonic crypts with programmable curvature. Primary cells at the crypt base sensed and responded to surface curvature by altering their proliferation and cytoskeletal properties.

Previous work has found that child welfare professionals have both higher self-report prevalence of adverse childhood experiences (ACEs) (Howard et al., 2015) and higher rates of insecure attachment (Howard et al., 2013) than normative samples. Although adult attachment and ACEs are both prolific areas of research in the developmental literature, little is known about how attachment representations may affect an individual's report of attachment-related ACEs. The current study investigated the role of attachment classification on disclosure of adverse childhood experiences on a self-report questionnaire and a semi-structured clinical interview in a sample of 228 child welfare professionals. Participants completed the Adult Attachment Interview (AAI) and ACE Questionnaire. Data revealed significant differences in self-report and rated disclosure of adverse childhood experiences by attachment classification. No significant differences were found for participants classified as secure-autonomous. Participants classified as dismissing reported fewer ACEs in their self-report questionnaire than independent scorers rated in their AAI. Participants classified as preoccupied reported more ACEs in their self-report questionnaire than independent scorers rated in their AAI. These differences raise questions and may be concerning for both the child welfare professionals and the families they serve. Specifically, future research should focus on possible consequences of having an insecure internal working model and ways to facilitate secure status for professionals if attachment classification is impacting their decisions regarding families. The current study provides a first step in investigating such relations.

Youth with a history of adverse childhood experiences (ACEs) are more likely to develop risky health behaviors. With the increase of media use in the general population, it is likely that these high-risk youth are developing maladaptive behaviors associated with media use (i.e., problematic media use). The goals of this article are (1) to describe symptoms of problematic media use in high-risk youth and (2) to determine whether ACEs are related to problematic media use in this population. Data were collected through online questionnaires from 348 parents or legal guardians of children ages 5 to 18 years, the majority of whom had been adopted. Parents and guardians reported on the child’s history of ACEs and completed the Problematic Media Use Measure-Short Form (PMUM-SF). Almost half of the participants reported that their child had a history of four or more ACEs (48.9%). Caregivers of foster or adopted children reported more symptoms of problematic media use than those reporting on their biological children. After adjusting for covariates, the number of ACEs predicted problematic media use above and beyond variance explained by demographic factors or screen time amount. Children with a history of ACEs had higher problematic media use compared to children without ACEs.

Context. In 1929, Russell predicted that dust particles cannot survive in a region close to any star, hence giving justification for a dust free zone to exist inside a certain distance from the star. This theoretical prediction has not been confirmed, even with our Sun. Aims. We use the unique vantage points and new perspectives of the Parker Solar Probe (PSP) mission to study the dust environment close to the Sun with imaging observations from the Wide Field Imager for Solar Probe (WISPR) as PSP orbits, progressively closer to the Sun (PSP will ultimately reach a perihelion distance of 9.86 R⊙). Methods. We analyze the radial brightness profile of the axis of symmetry of the F-corona in the WISPR images obtained from heliocentric distances between about 0.350 AU (75 R⊙) and 0.129 AU (28 R⊙) to detect any change from earlier observations. Historically, at observer locations between 1 and 0.3 AU, the brightness of the axis of symmetry has been shown to fall off as a power law of solar distance, r−n, with an exponent of n = 2.3. Results. We show that as PSP approaches its perihelion distance of 28 R⊙ (orbits 4 and 5), the radial gradient of the brightness profile of the axis of symmetry of the F-corona gradually becomes less steep, starting at about 19 R⊙ down to the shortest elongations reached with current WISPR observations at about 7.65 R⊙. This observational signature is modeled with an ad hoc homogeneous dust density model (i.e., it is not based on any physical model) along the symmetry axis of the zodiacal dust cloud, which (1) varies as the historical density profile, r−1.3, down to 19 R⊙, then (2) stays approximately constant down to 10 R⊙, and finally (3) decreases exponentially to become zero at 3 R⊙. The density profile below 19 R⊙ is accomplished by using a multiplier on the historical density profile that decreases linearly down to 3 R⊙. The distance dependence and range of the multiplier were chosen to best match the brightness observations below 30 R⊙. Conclusions. The observed brightness decrease in the axis of symmetry is interpreted as the signature of the existence of a dust density depletion zone between about 19 R⊙ and 3 R⊙, which at the inner limit of WISPR’s field of view of 7.65 R⊙ has a dust density that is ~5% lower than the density at 19 R⊙, instead of the expected density which is three times if no depletion zone exists. No noticeable variations in the brightness of the F-corona axis of symmetry were observed from 2018 to 2020.

The superior self-regulation and attention-regulation abilities of securely attached children have been repeatedly demonstrated. However, the mechanisms that allow securely attached children to exhibit higher levels of attention focus than insecurely attached (anxious-ambivalent and anxious-avoidant) children need to be explored. One possible mechanism that has been hypothesized to play a role in focusing attention is self-touch. Previous research has shown that 10-year-old children exhibit more bilateral self-touch (i.e., both hands are simultaneously moving onto each other or on the body, and both hands are in contact with each other or with the body), but not lateral self-touch (i.e., one hand is moving on the other hand or on the body, and the hand is in contact with the other hand or with the body), when they focus attention on a task. Because bilateral coordination is still developing during childhood, we expected that lateral self-touch, instead of bilateral self-touch, may be associated with attention focus for toddlers. The objectives of the present study were to examine whether securely attached toddlers exhibit more self-touch, particularly lateral self-touch, while they focus on a task than while they do not focus on a task. We expected to find that the association between lateral self-touch and attention focus is not as strong for insecurely attached toddlers. Data from forty-nine mother-child dyads were employed for analyses. The attachment classification of the children was determined using the Strange Situation. The duration of attention focus and self-touch behavior during a reading task were coded. An association between lateral self-touch and attention focus was found for children of all attachment classifications. This association was particularly strong for securely attached children. We discuss the possibility that securely attached toddlers may use lateral self-touch to regulate attention.

Maternal mental state language is thought to influence children's mental state language and sociocognitive understanding (e.g., theory of mind), but the mechanism is unclear. The current study examined the longitudinal development of mental state language in mother-child interactions. The methodology included assessments of the child and/or mother-child dyad at six time points between 12 to 52 months of the child's age. Measures determined child's attachment style and language abilities, and mental state language used by mother and child during a block-building task. Results showed that (a) mental state talk, including belief and desire language, increased over time; (b) there were differences between the type of mental state words used by the mother in insecure versus secure dyads; (c) there were differences in patterns of mental state words used in both mothers and children in insecure versus secure dyads; and (d) attachment appeared to exert a consistent influence over time.

Family drawings are thought to be a valuable tool for assessing attachment representations, but have not often been used with post-institutionalized adopted children thought to be at risk for attachment disturbances. The current study compared family drawings from a sample of post-institutionalized adopted children to a sample of children who entered their family by birth with similar levels of executive functioning. All families were residing in a major metropolitan area in the southwestern United States of America. Drawings were scored using Fury, Carlson, and Sroufe (1997) rating scales. The overall pattern of findings indicates family drawings of at risk adopted children and biological comparison children differ significantly in both objective scores and subjective ratings, suggesting that children in the adopted sample are at greater risk for attachment-related disturbances than children in the comparison sample. These findings corroborate other studies reporting attachment difficulties in post- institutionalized adopted children and indicate that family drawings are a sensitive and valid approach to assessing disturbances related to attachment representations during childhood.
