1. Antiproliferative effect of mifepristone (RU486) on human neuroblastoma cells (SK-N-SH): in vitro and in vivo studies

Abstract

RU486 (mifepristone), a glucocorticoid and progesterone receptor antagonist, has been reported to exert antiproliferative effects on tumor cells. Experiments were performed to analyze the effects of RU486 on the proliferation of the human neuroblastoma, both in vitro and in vivo, using the human neuroblastoma SK-N-SH cell line. The exposure in vitro of SK-N-SH cells to RU486 revealed a dose-dependent inhibition of 3H-thymidine incorporation due to a rapid but persistent inhibition of MAPKinase activity and ERK phosphorylation. A significant decrease of SK-N-SH cell number was evident after 3, 6, and 9 days of treatment (up to 40% inhibition), without evident cell death. The inhibitory effect exerted by RU486 was not reversed by the treatment of the cells with dexamethasone or progesterone. Moreover, RU486 induced a shift in SK-N-SH cell phenotypes, with an almost complete disappearance of the neuronal-like and a prevalence of the epithelial-like cell subtypes. Finally, the treatment with RU486 of nude mice carrying a SK-N-SH cell xenograft induced a strong inhibition (up to 80%) of tumor growth. These results indicated a clear effect of RU486 on the growth of SK-N-SH neuroblastoma cells that does not seem to be mediated through the classical steroid receptors. RU486 acted mainly on the more aggressive component of the SK-N-SH cell line and its effect in vivo was achieved at a concentration already used to inhibit oocyte implantation.

Abstract

Tana del Barletta is an upland cave used from the Late Neolithic to the Middle Bronze Age, located in the vicinity of the coast in Liguria (NW Italy). The excavation revealed the presence of a faunal assemblage dominated by caprine and cattle remains. In order to gain new data on late prehistoric farming strategies (e.g. seasonal mobility, coastal grazing, animal diet), intra-tooth series of stable oxygen and carbon isotopes have been obtained from cattle and sheep/goat tooth enamel, along with intra-tooth series of nitrogen and carbon isotopes from cattle dentine collagen. Due to the prevalence of maxillary teeth, a modern calf has also been analysed to assess intra-individual isotopic differences between the maxillary and mandibular dentition. Modern data on oxygen isotope values of meteoric water from different altitudes around the area of the site were used as a reference for interpretation. The results indicate that the water ingested by the herd was mostly characterised by particularly low δ18O values, highlighting the importance of the uplands for the late prehistoric farmers of the region. However, the input of water sourced from lower elevations, especially during the winter months, cannot be dismissed. In addition, the nitrogen isotopic composition of cattle collagen rules out the ingestion of salt-tolerant vegetation or seaweed, suggesting that grazing did not occur directly on the coastal plain.

5. Dexamethasone blocks the migration of the human neuroblastoma cell line SK-N-SH

Abstract

Glucocorticoids (Gc) influence the differentiation of neural crest-derived cells such as those composing sympathoadrenal tumors like pheochromocytomas, as well as neuroblastomas and gangliomas. In order to obtain further information on the effects of Gc on cells evolving from the neural crest, we have used the human neuroblastoma cell line SK-N-SH to analyze: 1) the presence and the binding characteristics of Gc receptors in these cells, 2) the effect of dexamethasone (Dex) on the migration of SK-N-SH cells, and 3) the effect of Dex on the organization of the cytoskeleton of SK-N-SH cells. We show that: 1) receptors that bind [³H]-Dex with high affinity and high capacity (Kd of 9.6 nM, Bmax of 47 fmol/mg cytosolic protein, corresponding to 28,303 sites/cell) are present in cytosolic preparations of SK-N-SH cells, and 2) treatment with Dex (in the range of 10 nM to 1 µM) has an inhibitory effect (from 100% to 74 and 43%, respectively) on the chemotaxis of SK-N-SH cells elicited by fetal bovine serum. This inhibition is completely reversed by the Gc receptor antagonist RU486 (1 µM), and 3) as demonstrated by fluorescent phalloidin-actin detection, the effect of Dex (100 nM) on SK-N-SH cell migration is accompanied by modifications of the cytoskeleton organization that appear with stress fibers. These modifications did not take place in the presence of 1 µM RU486. The present data demonstrate for the first time that Dex affects the migration of neuroblastoma cells as well as their cytoskeleton organization by interacting with specific receptors. These findings provide new insights on the mechanism(s) of action of Gc on cells originating in the neural crest.

Abstract

Patients with high-risk endometrial carcinoma (stage IcG3, IIG3 with myometrial invasion >50%, and III) receive adjuvant therapy after surgery but it is not clear whether radiotherapy (RT) or chemotherapy (CT) is better. We randomly assigned 345 patients with high-risk endometrial carcinoma to adjuvant CT (cisplatin (50 mg m(-2)), doxorubicin (45 mg m(-2)), cyclophosphamide (600 mg m(-2)) every 28 days for five cycles, or external RT (45-50 Gy on a 5 days week(-1) schedule). The primary end points were overall and progression-free survival. After a median follow-up of 95.5 months women in the CT group as compared with the RT group, had a no significant hazard ratio (HR) for death of 0.95 (95% confidence interval (CI), 0.66-1.36; P = 0.77) and a nonsignificant HR for event of 0.88 (95% CI, 0.63-1.23; P = 0.45). The 3, 5 and 7-year overall survivals were 78, 69 and 62% in the RT group and 76, 66 and 62% in the CT group. The 3, 5 and 7-year progression-free survivals were, respectively, 69, 63 and 56 and 68, 63 and 60%. Radiotherapy delayed local relapses and CT delayed metastases but these trends did not achieve statistical significance. Overall, both treatments were well tolerated. This trial failed to show any improvement in survival of patients treated with CT or the standard adjuvant radiation therapy. Randomised trials of pelvic RT combined with adjuvant cytotoxic therapy compared with RT alone are eagerly awaited.

Abstract

Study aim: the Ebf gene family has been implicated in several developmental processes, ranging from B-cell development to neuronal differentiation. As the murine Ebf2 gene is expressed in numerous sites of nervous system, Ebf2-null mice develop hypogonadotropic hypogonadism, due to defective migration of gonadotropin releasing hormone-synthesizing neurons, and a peripheral neuropathy as well. Therefore, we aimed to study whether electrophysiological tests would be able to detect abnormalities of peripheral nerve function. Methods: 2 groups of mice were studied, which consisted of 8 Ebf2-/- mice and 9 age-matched controls. The sciatic nerve was stimulated at the ankle and at the ischiatic notch; the compound motor action potential (cMAP) was recorded from the paw muscles with a pair of needle electrodes to measure the motor conduction velocity (MCV). Abstract

Purpose: Longitudinal tenectomy of the reflected tendon of the superior oblique muscle resulted in full recovery in a series of cases of Brown's syndrome after surgery by one of the authors. Methods: A survey of the records of cases of Brown's syndrome was done to assess the criteria for patients requiring surgery and to evaluate the methods used. Included in the study were 9 eyes of 7 patients who underwent surgery over the course of 1 year. Criteria identified in cases in this study include: preoperative compulsory head tilt in primary position of gaze, severe restriction of Hess chart in the affected eye, and an enlarged field in the healthy eye; intraoperative thickening of the tendon preventing free slippage through the trochlea in the forced duction test. The surgical method consisted of trimming a swelling that was consistently found on the reflected tendon of the superior oblique muscle in all patients undergoing surgery. Enough of the swelling was removed to allow free slippage of the tendon through the trochlea, as judged by the forced duction test. Sulphamide powder soaked in delayed action steroid compound (methy !prednisolone acetate) was applied to prevent postoperative adhesions. Results: A normal head posture and a satisfactory upward vergence were achieved in all patients from the first days after surgery, and were maintained throughout the follow up, as shown in the pre- and postoperative photographs and Hess charts. Conclusions: A preliminary exploration of the reflected tendon of the superior oblique muscle is warranted in cases of Brown's syndrome; when a thickening of the tendon is found, its surgical trimming is advised. Our findings suggest that delayed development of the trochlea may be a possible cause of Brown's syndrome. Journal of Pediatric Ophthalmology and Strabismus 2002;39:33-38.

Abstract

Limb girdle muscular dystrophy (LGMD) constitutes a family of rare genetic disease characterized by progressive weakness of pelvic or shoulder girdle musculature due to impairment of the dystrophin-associated protein complex (DPC) components. The sarcoglycanopathies are a common cause of LGMDs, accounting for 3–18%, with a high percentage of severe cases. In particular, LGMD2E is a recessive autosomal disease caused by mutation in the gene, located on chromosome 4q12, encoding the beta-sarcoglycan, a major component of the DPC. Age of onset is between 2 years and the mid-teenage years. The clinical presentation includes progressive limb weakness (mainly of proximal muscles). Cardiac involvement occurs in 20% of the cases. LGMD2E is classified as a “neglected” disease by EU, underlying a substantial absence of dedicated scientific research; no specific treatments are known. Of interest, sarcoglycanopathies should be cured by gene therapy since the sarcoglycans genes are relatively short and with few exons, making them suitable for adenovirus-based therapy. Actually, a phase II clinical trial for gene therapy of alpha-sarcoglycanopathy is ongoing in USA. Another possible approach relies on autologous transplantation of stem cells, such as muscle progenitor cells. In 2013 the volunteer organization named Family Group of Beta-sarcoglycanopathy ONLUS (GFB ONLUS www.lgmd2e.org) was established to: a) contact the highest number of patients affected by LGMD and their families b) collect all data and information available on LGMD2E c) stimulate both basic and clinical research. Moreover, we would like to promote scientific research specific on LGMD2E, create collaborations with scientists to organize both informative and scientific meetings and, eventually, support researchers interested to study this disease. On April 19th 2013 the GFB ONLUS held its first scientific Meeting in Milan that allowed a fruitful confrontation among national and international researches.

Abstract

Glucocorticoids (Gc) influence the differentiation of neural crest-derived cells such as those composing sympathoadrenal tumors like pheochromocytomas, as well as neuroblastomas and gangliomas. In order to obtain further information on the effects of Gc on cells evolving from the neural crest, we have used the human neuroblastoma cell line SK-N-SH to analyze: 1) the presence and the binding characteristics of Gc receptors in these cells, 2) the effect of dexamethasone (Dex) on the migration of SK-N-SH cells, and 3) the effect of Dex on the organization of the cytoskeleton of SK-N-SH cells. We show that: 1) receptors that bind [³H]-Dex with high affinity and high capacity (Kd of 9.6 nM, Bmax of 47 fmol/mg cytosolic protein, corresponding to 28,303 sites/cell) are present in cytosolic preparations of SK-N-SH cells, and 2) treatment with Dex (in the range of 10 nM to 1 µM) has an inhibitory effect (from 100% to 74 and 43%, respectively) on the chemotaxis of SK-N-SH cells elicited by fetal bovine serum. This inhibition is completely reversed by the Gc receptor antagonist RU486 (1 µM), and 3) as demonstrated by fluorescent phalloidin-actin detection, the effect of Dex (100 nM) on SK-N-SH cell migration is accompanied by modifications of the cytoskeleton organization that appear with stress fibers. These modifications did not take place in the presence of 1 µM RU486. The present data demonstrate for the first time that Dex affects the migration of neuroblastoma cells as well as their cytoskeleton organization by interacting with specific receptors. These findings provide new insights on the mechanism(s) of action of Gc on cells originating in the neural crest.

Abstract

The efficacy and tolerability of granisetron in the management of acute and delayed emesis was compared with that of a multiple antiemetic drug combination regimen, including metoclopramide, dexamethasone, lorazepam and orphenadrine. The trial was a randomized, cross-over study involving 111 patients with gynecological cancers undergoing chemotherapy with cisplatin. Granisetron was significantly more effective than the combination regimen during the first 24 h after chemotherapy; complete response, rates were 67 and 48%, respectively (p = 0.002). There was a significant reduction in the effectiveness of the combination during the second treatment cycle, compared with the first. In contrast, the efficacy of granisetron did not differ between the two cycles. The response rate during the 6 days after chemotherapy was 40.8% in both groups. At the end of the study, 55% of patients preferred granisetron and 23% preferred the combination (p < 0.001). Granisetron was well tolerated. The principal adverse event was headache, which was reported in 7% of patients. The results of this study confirm that granisetron is effective in the treatment of cisplatin-induced nausea and vomiting during the 24 h after chemotherapy.

Abstract

Adrenal glucocorticoids (Gc) are among the most significant hormones in the mammalian organisms; these steroids may reach and penetrate all tissues where they interact with cytoplasmic/nuclear receptors, through which they exert multiple and very multifaceted actions. The effects of physiological concentrations of Gc on brain functions have not been completely clarified, even though Gc are recognized to influence behavioral responses, emotions, cognitive processes and to take part in the neuroendocrine control of body homeostasis. Developmental programming effects of Gc in animal models and humans have been proposed. Actually, pre-natal stress, or exposure to high Gc levels, would somehow affect neuronal developmental events in some structure and this can lead to central nervous system altered functions, as the impairment of neuroendocrine activities, cognitive processes, sleep and mood disorders. Interestingly, it has been observed that these abnormalities may not be limited to the first directly exposed individuals but transmissible across generations. The establishment of animal models with localized pre-natal glucocorticoid receptors deficiency led to the accumulation of data on the possible roles of these hormones on development of the central and peripheral nervous system. The most recent findings on the effects of Gc on neuroblast development, with particular attention to neuronal migration, will be presented.

Abstract

This paper describes essential aspects of the hydrotreatment of pyrolytic oils in the light of results obtained until now at the Universite Catholique de Louvain. Stability of pyrolysis oils necessitates a two-step processing. A low temperature hydrotreatment enables stabilization through reactions like olefin, carbonyl and carboxylic groups reduction. Further hydrotreatment aims at hydrodeoxygenation of phenols and hydrocracking of larger molecules. Results about catalysts, reaction conditions and parameters enabling or influencing the control of the reaction are summarized. Based on these laboratory data, a preliminary techno-economical evaluation is made. 50 wt.-% yields in hydrocarbons for deep hydrorefining of pyrolysis oils can be expected. Nevertheless, a moderate hydroconversion with partial elimination of oxygen would be, economically, more advantageous.

Cobalt(II) phthalocyanine was synthesized in situ within the cavities of a Y-zeolite and was characterized using different physicochemical techniques. Catalytic tests of hydrocarbon oxidation were carried out using iodosylbenzene and t-butylhydroperoxide as oxygen donors and chloroform as solvent. The formation of epoxides, ketones and alcohols was detected. It was observed that the encapsulated phthalocyanine is still active after 20 h of reaction while the free Co phthalocyanine loses its activity in a few minutes.

This paper presents the results obtained in the characterization of different pyrolysis oils, five oils produced by carbonization and one by flash pyrolysis. Chemical and physical properties such as density, viscosity, elemental composition, char content, water content, solubility and heating value were first determined, then an in-depth chemical characterization was carried out by liquid-liquid fractionation. A diagram indicating the separation procedure, providing four fractions (acids, bases, polars and hydrocarbons), is presented. Polar molecules which were retained in the aqueous layer ('aqueous' fraction) are recuperated. Each fraction was subsequently analysed by gas chromatography-mass spectrometry (g.c.-m.s.) and Fourier transform infrared spectroscopy (FT-i.r.). The acidic fraction was the most abundant and contained essentially phenolic structures. However, phenols were differently substituted in the two types of oils: alkyl and methoxy groups in the carbonization oils; methoxy, acidic, aldehydic and ketonic functions in the flash pyrolysis oil. The other fractions present a similar composition for all oils. The basic fraction was always very small; some aromatic N-containing compounds were identified. The 'polar' neutral fraction was also small and its characterization was very difficult. The hydrocarbon fraction, especially, was constituted of aromatics and cyclics, and some aliphatics were also identified. The 'aqueous' fraction contained mainly carboxylic esters, alcohols and ethers.

Flash pyrolysis oils have a high oxygen content. The presence of numerous and various oxygenated functions give these oils unwanted characteristics that may be improved by the elimination of oxygen by catalytic hydrotreating. This paper deals with the chemical characterization of RTP oils by liquid-liquid fractionation followed by mass spectrometric determination of the individual compounds. In a second part, the hydrodeoxygenation reactions were approached using model compounds selected on the basis of the characterization. The CH2Cl2 acidic fractions of the oils are mainly composed of phenolic structures. Nevertheless, the phenolic molecules are differently substituted depending on the pyrolysis conditions: carboxyl and carbonyl groups for short residence times and methoxy groups for long residence times. All the main chemical groups present in pyrolysis oils can be eliminated using nickel-molybdenum and cobalt-molybdenum traditional hydrotreating catalysts. The reactivity, the activation energies and the products of the hydrodeoxygenation conversion of the various oxygenated groups were determined.

Several Bartonella species (spp.) have been identified in dogs diagnosed with infectious endocarditis (IE) or myocarditis.To interrogate cardiac tissues of dogs with suspected IE for the presence of Bartonella spp. DNA of dogs in the Rocky Mountain states.Nine dogs with a clinical diagnosis of endocarditis from January 1990 to June 2008 were included.In this retrospective study, medical records at the Veterinary Teaching Hospital were searched. Animals were excluded if there was no diagnosis of IE in the original necropsy report. Paraffin embedded tissue blocks and medical records were available from 9 dogs. Total DNA was extracted from the cardiac tissues and assessed for Bartonella spp. DNA by 3 polymerase chain reaction (PCR) methods. For positive samples, the Bartonella spp. were determined by genetic sequencing or fluorogenic real-time PCR.Bartonella henselae DNA was amplified from the tissues of 7 dogs; Bartonella vinsonii subsp berkhoffii DNA was amplified concurrently from 3 dogs. Six dogs were from Colorado and 1 was from Wyoming. Flea or tick infestations were reported in 2 dogs.Bartonella spp. should be on the differential list for dogs in the Rocky Mountain states. The results emphasize the need for routine use of external parasite control products even in regions perceived to have low risk for flea and tick infestations.

PIXE analysis of 10 commercial powdered milk has been performed for 16 elements: P, Cl, K, Ca, Cr, Fe, Co, Ni, Cu, Zn, Ga, Br, Rb, Sr, Mo and Cd. None of the analyzed samples was found to contain any of these elements at concentrations exceeding toxicity limits. A qualitative analysis of the powdered milk containers was also done. Analysis of a standard reference milk sample, A-11 from IAEA, for the 16 elements listed above gave a good agreement with the certified values, except for Cl, K, Ca, Cr and Rb.

Abstract

Community-acquired pneumonia (CAP) is a leading cause of childhood death. There are few published reports of radiographic findings among children with severe CAP.To describe chest X-ray (CXR) findings and assess association between these radiographic findings and pneumococcal isolation in children with severe CAP.A prospective, multicenter, observational study was conducted in 12 centers in Argentina, Brazil, and the Dominican Republic. Children aged 3-59 months, hospitalized with severe pneumonia, were included. On admission, blood and pleural effusion cultures were performed. Streptococcus pneumoniae was identified according to standard procedures in the respective national reference laboratory. Chest X-rays were taken on admission and read before the culture results were reported.Out of 2,536 enrolled patients, 283 (11.2%) had S. pneumoniae isolated, in 181 cases (7.1%) from blood. The follow radiographic patterns were observed: alveolar infiltrate (75.2%), pleural effusion (15.6%), and interstitial infiltrate (9.2%). Overall, pleural effusion was associated with pneumococcal isolation and pneumococcal bacteremia (P0.001). Infiltrates were unilateral (78.7%) or bilateral (21.3%), right-sided (76%) or left-sided (24%), in the lower lobe (53.6%) or the upper lobe (46.4%). Multivariate analysis including patients with affection of only one lobe showed that upper lobe affection and pleural effusion were associated with pneumococcal isolation (OR 1.8, 95% CI, 1.3-2.7; OR 11.0, 95% CI, 4.6-26.8, respectively) and with pneumococcal bacteremia (OR 1.7, 95% CI, 1.2-2.6; OR 3.1, 95% CI, 1.2-8.0, respectively).Three-quarters of the patients studied had alveolar infiltrates. Upper lobe compromising and pleural effusion were associated with pneumococcal invasive disease.

An IOL with direct attachment to the sclera has been implanted in the absence of the posterior capsule in a total of 114 cases: as primary implant during ICCE in 93 eyes, and as secondary implant in 21 eyes. Vitrectomy was not necessary in any patient. No part of the haptics or non-absorbable sutures were left on the surface of the sclera. The haptics were completely embedded in the scleral thickness without sutures and were anchored by their terminal knob.

Lipid extracts from two acidic hydromorphic soils contained simple lipids and a slightly soluble fraction (named polar fraction or complex lipids). Ether- and ester-bond cleavage and KMnO4 oxidation of the polar fraction was followed by GC-MS analyses of the released compounds. In contrast to n-alkanols and α,ω-diacids, normal monoacids were found to be the same as in simple lipids. It is concluded that the polar fraction of the soil corresponds to highly aliphatic macromolecules with a polyethylene-like structure, in which some simple lipids may be incorporated to form a protokerogen.
