Tensile drawing experiments on poly(ethylene terephthalate) (PET) have shown that there is no significant strain-induced crystallisation until draw ratios around 2.3. The actual onset of strain-induced crystallisation depends on the deformation rate, and it occurs at lower draw ratios for higher draw rates. The development of strain-induced crystallisation has a significant impact on the relaxation behaviour of PET. Online birefringence measurements, during the relaxation of PET drawn to different draw ratios showed that: (a) at low draw ratios, the orientation relaxes over long periods of time and Fourier transform infrared (FTIR) spectroscopy showed that the overall orientation falls after relaxation; (b) at higher draw ratios, when significant strain-induced crystallisation has occurred, the orientation decreases over short times, of the order of 10 s and remains constant thereafter. This behaviour is probably due to the crystallites that lock in the extension of the chains in the amorphous material. Post relaxation FTIR measurements showed that the orientation increases due to annealing effect. Differential scanning calorimetry (DSC) showed that there is an increase in the crystalline fraction after relaxation for all draw ratios, which is probably due to the conversion of oriented amorphous material (trans conformers) into the crystalline phase.

The effect of processing conditions on the structure and properties of roll drawn poly(ethylene terephthalate) (PET) was examined. It was found that, when roll drawing amorphous PET at temperatures just above the glass transition, only very low draw ratios were obtained. This is probably because there were no crystallites to lock in the applied extension. Roll drawing at high temperatures, above 130°C, where there was significant thermal crystallization, produced film of high strength. At temperatures between 130°C and 190°C, the properties were almost independent of processing temperature. Mechanical tests performed on roll drawn samples, processed in this temperature range, showed that the initial modulus and the yield stress increased linearly with draw ratio. The yield strain decreased with draw ratio up to λ = 4.0, and then became almost constant. The processing temperature that produced samples with the greatest strength was 170°C. This was because the highest draw ratios were obtained at this temperature while maintaining constant width deformation. At low draw ratios, the crystallinity increased with the processing, whereas at higher draw ratios, it was independent of temperature. This constant level of crystalline fraction may have produced the constant failure strain that was observed at high draw ratios. The orientation functions were similarly unaffected by the processing temperature, although birefringence measurements did suggest that lower processing temperatures may have produced higher levels of orientation. The orientation of the trans conformers was independent of the temperature, but the overall content was increased at higher processing temperatures.

In this paper, we discuss the development and use of a birefringence technique for on-line or off-line quantitative measurement of orientation in transparent films, sheets, bottles, etc. Absolute values of birefringence are measured in two directions using a technique based on an incident multi-wavelength double beam and a photodiode array assembly, combined with in-house developed software. Both machine and transverse direction birefringence (relative to the normal direction) are measured quasi-simultaneously. Film and sheet thicknesses from 10 microns to 10 mm were tested and birefringence values from 0.0005 to 0.25 were measured. The technique was tested on different materials and under different conditions and several applications will be discussed.

The mechanical relaxation behaviour of high and linear low density polyethylenes has been examined using tensile dynamic mechanical measurements and compared to the deformation behaviour observed in tensile drawing. In this way the relationship between the mechanical relaxations and the tensile deformation has been investigated. It is shown that the brittle-ductile transition relates to the γ-relaxation while the yield behaviour is related to the interlamellar shear process and the c-shear process.

Dynamic mechanical measurements are reported on two polyethylene copolymers with different types of comonomers and well-characterized comonomer distributions. Measurements on both isotropic and oriented samples were undertaken over a wide frequency range. The mechanical anisotropy and activation energies of the α and β relaxations are consistent with the former relating to c-shear in the crystals, and the latter to interlamellar shear.

In Escherichia coli K-12, expression of the lysU gene is regulated by the lrp gene product, as indicated by an increase in the level of lysyl-tRNA synthetase activity and LysU protein in an lrp mutant. Comparison of the patterns of protein expression visualized by two-dimensional gel electrophoresis indicated that LysU is present at higher levels in an lrp strain than in its isogenic lrp+ parent. The purified lrp gene product was shown to bind to sites upstream of the lysU gene and to protect several sites against DNase I digestion. A region extending over 100 nucleotides, between 60 and 160 nucleotides upstream from the start of the lysU coding sequence, showed altered sensitivity to DNase I digestion in the presence of the Lrp protein. The extent of protected DNA suggests a complex interaction of Lrp protein and upstream lysU DNA.

Pig liver methylenetetrahydrofolate reductase catalyzes the reduction of quinonoid dihydropterins in vitro. Either NADPH or methyltetrahydrofolate can serve as the electron donor. Methylenetetrahydrofolate reductase can also suppor phenylalanine hydroxylation in vitro by regeneration of the tetrahydropterin cofactor. These results lend support to the proposal that reduction of methylenetetrahydrofolate proceeds by tautomerization of the 5-iminium cation to form quinonoid 5-methyldihydrofolate, which is then reduced to methyltetrahydrofolate (Matthews, R. G., and Haywood, B. J. (1979) Biochemistry 18, 4845-4851). Under Vmax conditions, the turnover numbers for the NADPH-linked reductions of the quinonoid forms of 6,7-dimethyldihydropterin, dihydrobiopterin, and dihydrofolate are all about the same as that for the reduction of methylenetetrahydrofolate. The Km values for racemic mixtures of the same quinonoid acceptors are 40, 30, and 20 microM, respectively, while the Km for (6R,S)methylenetetrahydrofolate is 20 microM at pH 7.2 in phosphate buffer. The reduction of quinonoid dihydropterins is inhibited by adenosylmethionine and dihydropteroylhexaglutamate, which are known to modulate methylenetretrahydrofolate reductase activity.

The mode of inspiratory augmentation (1A) of the A2-P2 interval was investigated in seven normal volunteers (group 1) and six patients with pulmonary hypertension of diverse etiology (group 2) using catheter-tip micromanometers. In group 1 subjects, inspiratory widening of this interval was found to average 27.2 msec, of which 7.6 plus or minus 2.7 msec (1 sd) or 27 plus or minus 7% was due to a decrease in the Q-A2 interval. The major contribution of Q-P2 interval prolongation was divided into two components: a) Q-O was measured from the onset of the QRS to the onset of the rapid descent of the right ventricular (RV) negative dp/dt, which was felt to reflect the duration of RV electromechanical systole, b) O-P2 or Q-P2 - Q-O. Increase in the Q-O interval accounted for only 7.7 plus or minus 5.0 msec or 28 plus or minus 12% of the total IA. The major single component of IA was the increase in O-P2 which averaged 11.9 plus or minus 3.0 msec. Five of six group 2 patients demonstrated significant respiratory change in Q-P2 intervals. In contrast to group 1 subjects, however, this was accomplished primarily via increases in the duration of RV electromechanical systole. The O-P2 interval is felt to primarily reflect the impedance characteristics of the pulmonary vascular bed. It is concluded that physiologic splitting of the second heart sound in normal subjects is most probably due to an inspiratory decrease in impedance of the pulmonary bed rather than the traditional explanation of prolongation of RV systole secondary to an increase in venous return. When the normal impedance characteristics of this bed are lost, as in pulmonary hypertension, IA must occur primarily via increases in the duration of RV systole. The inspiratory delay from the conclusion of RV systole to the occurrence of P2 is attributed to the inertiance of the RV stroke mass.

Tetrahydrofolate (H4folate) derivatives are chemically analogous to tetrahydropterins and dihydroflavins, and like these compounds can be oxidized readily. The roles of pterin and flavin cofactors in biological oxidoreductions are well documented. However, with the exception of the reaction catalyzed by thymidylate synthase, explicit oxidoreductions of the H4folate cofactor do not occur in enzyme-catalyzed folate-dependent reactions. We have been studying methylenetetrahydrofolate reductase from pig liver. This enzyme catalyzes the reversible conversion of the exocyclic N5,N10-methylene substituent to an N5-methyl group. Our studies suggest that the detailed mechanism of this reduction of an exocyclic group involves the oxidoreduction of the tetrahydropterin ring system in a manner that is not apparent from the overall reaction stoichiometry. We suggest that such latent oxidoreductions of H4folate cofactors may also occur in other H4folate-dependent reactions, such as the reaction catalyzed by methionine synthase.

Methylenetetrahydrofolate reductase commits tetrahydrofolate-bound one carbon units to use in the regeneration of the methyl group of adenosylmethionine (AdoMet) in eucaryotes and its activity is allosterically inhibited by AdoMet. Limited proteolysis and scanning transmission electron microscopy have been employed to show that the enzyme is a dimer of identical subunits and that each subunit is composed of spatially distinct domains with molecular masses of approximately 40 and 37 kDa (Matthews, R. G., Vanoni, M. A., Hainfeld, J. F., and Wall, J. (1984) J. Biol. Chem. 259, 11647-11650). We now report the use of the photoaffinity label 8-azido-S-adenosylmethionine (8-N3AdoMet) to locate the binding site for the allosteric inhibitor on the 37-kDa domain. In the absence of light, 8-N3AdoMet is itself an inhibitor of methylenetetrahydrofolate reductase activity, with a Ki value 4.8-fold higher than AdoMet, and like AdoMet it induces slow transitions between active and inactive forms. Photoaffinity labeling is dependent on irradiation with ultraviolet light and is prevented by AdoMet but not by ATP. Limited proteolysis of the photolabeled enzyme results in the formation of a labeled 37-kDa fragment which is further processed to a labeled 34-kDa fragment. On conversion of the 34-kDa fragment to a 31-kDa polypeptide, all label is lost, suggesting that the labeling is restricted to an approximately 3-kDa region near one end of the 37-kDa polypeptide. Limited proteolysis of the native enzyme, while completely desensitizing the enzyme to inhibition by AdoMet or 8-N3AdoMet, does not prevent subsequent photolabeling of the 37-kDa peptide fragment. This photolabeling does not occur in the presence of excess AdoMet. These latter experiments suggest that the desensitization of the enzyme eliminates the ability of allosteric effectors to stabilize an inactive form of the enzyme, but does not abolish specific binding of 8-N3AdoMet or AdoMet.

We have previously reported the isolation of old yellow enzyme complexed with a ligand of low molecular weight which imparts a distinctive charge-transfer absorption to the enzyme, making it green in color (Matthews, R. G., and Massey, V. (1969) J. Biol. Chem. 244, 1779-1789). This ligand has now been identified as p-hydroxybenzaldehyde by removal from the enzyme and characterization of its optical spectrum and subsequent mass spectral analysis. Similar compounds which are also bound to old yellow enzyme have been isolated from yeast extract and identified. These compounds give rise to complexes with old yellow enzyme which are characterized by broad long wavelength absorption bands, and they lead to inhibition of the NADPH-O2 oxidoreductase activity catalyzed by old yellow enzyme.

A gene encoding cobalamin-dependent methionine synthase (EC 2.1.1.13) has been isolated from a plasmid library of Escherichia coli K-12 DNA by complementation to methionine prototrophy in an E. coli strain lacking both cobalamin-dependent and -independent methionine synthase activities (RK4536:metE, metHH). Maxicell expression of a series of plasmids containing deletions in the metH structural gene was employed to map the position and orientation of the gene on the cloned DNA fragment. A 6.3-kilobase EcoRI-SalI fragment containing the gene was cloned into the sequencing vector pGEM3B for double-stranded DNA sequencing; the MetH coding region consists of 3372 nucleotides. The enzyme was purified from an overproducing strain of E. coli harboring the recombinant plasmid, in which the level of methionine synthase was elevated 30- to 40-fold over wild-type E. coli. Recombinant enzyme is a protein of 123,640 molecular weight and has a turnover number of 1,450 min-1 in the standard assay. These values are to be compared with previously reported values of 133,000 for the molecular weight and 1,240-1,560 min-1 for the turnover number of the homogenous enzyme purified from a wild-type strain of E. coli B (Frasca, V., Banerjee, R. V., Dunham, W. R., Sands, R. H., and Matthews, R. G. (1988) Biochemistry 27, 8458-8465). Limited proteolysis of the native enzyme with trypsin resulted in loss of enzyme activity but retention of bound cobalamin on a peptide fragment of 28,000 molecular weight. This fragment has been shown to extend from residue 643 to residue 900 of the 1124-residue deduced amino acid sequence.

Methylenetetrahydrofolate reductase is a flavoprotein which has recently been purified to homogeneity from pig liver (Daubner, S. C., and Matthews, R. G. (1982) J. Biol. Chem. 257, 140-145). The enzyme catalyzes the transfer of reducing equivalents from NADPH to menadione or methylenetetrahydrofolate, or from methyltetrahydrofolate to menadione. We have now examined the individual reductive and oxidative half-reactions comprising these activities, measuring the rate of flavin reduction or oxidation in an anaerobic stopped flow apparatus. In all cases, the individual half-reactions occur at rates which are sufficiently fast to account for catalytic turnover, indicating that the enzyme is kinetically competent to catalyze these oxidoreductions by Ping Pong Bi Bi mechanisms. NADPH-linked reduction of the flavin appears to be rate-limiting for the NADPH-menadione oxidoreductase reaction, while reoxidation of the flavin is partially rate-limiting in the NADPH-methylenetetrahydrofolate oxidoreductase reaction. Reduction of the flavin by methyltetrahydrofolate is rate-limiting in the methyltetrahydrofolate-menadione oxidoreductase reaction. No intermediates are seen in any of these half-reactions. In agreement with our postulate of ping-pong mechanisms for the catalytic reactions of the enzyme is our observation of exchange of radiolabel between [methyl-14C]methyltetrahydrofolate and [methylene-14C]methylenetetrahydrofolate in the absence of either menadione or pyridine nucleotides. The rate of exchange when both substrates are saturating is in good agreement with the rate of the methyltetrahydrofolate-menadione oxidoreductase reaction measured under Vmax conditions.

The oxidation-reduction potential, E2, for the couple oxidized lipoamide dehydrogenase/2-electron reduced lipoamide dehydrogenase has been determined by measurement of equilibria of these enzyme species with lipoamide and dihydrolipoamide or with oxidized and reduced azine dyes. E2 is -0.280 V at pH 7, and deltaE2/deltapH is -0.06 V in the pH range 5.5 to 7.6. Values for E1, the oxidation-reduction potential for the couple 2-electron reduced enzyme/4-electron reduced enzyme, were obtained from measurements of the extent of dismutation of 2-electron reduced enzyme to form mixtures containing oxidized and 4-electron reduced enzyme. E1 is -0.346 V at pH 7, and deltaE1/deltapH is -0.06 V in the pH range 5.7 to 7.6. Spectra of oxidized enzyme and 4-electron reduced enzyme do not show variations with pH over this range, but the spectrum of the 2-electron reduced enzyme is pH-dependent, with the molar extinction at 530 nm changing from 3250 M-1 cm-1 at pH 8 to 2050 M-1 cm-1 at pH 5.2. The pH-dependent changes which are observed in the absorption properties of the 2-electron reduced enzyme are consistent with the disappearance of a charge transfer complex between an amino acid side chain and the oxidized flavin at the lower pH values, with the apparent pK of the side chain at pH 5. It has been suggested that the 530 nm absorbance of 2-electron reduced enzyme is due to a charge transfer complex between thiolate anion and oxidized flavin, and we propose that the thiolate anion is stabilized by interaction with a protonated base. The thermodynamic data predict that the amount of 4-electron reduced enzyme formed when the enzyme is reduced by excess NADH will be pH-dependent, with the greatest amounts seen at low pH values. These data support earlier evidence (Matthews, R.G., Wilkinson, K.D., Ballou, D,P., and Williams, C.H., Jr. (1976) in Flavins and Flavoproteins (Singer, T.P., ed) pp. 464-472; Elsevier Scientific Publishing Co., Amsterdam) that the role of NAD+ in the NADH-lipoamide reductase reaction catalyzed by lipoamide dehydrogenase is to prevent accumulation of inactive 4-electron reduced enzyme by simple reversal of the reduction of 2-electron reduced enzyme by NADH.

Scanning transmission electron microscopy of individual unfixed molecules of methylenetetrahydrofolate reductase has been used to determine the molecular mass distribution of the protein. Methylenetetrahydrofolate reductase, which has a subunit molecular mass of 77 kilodaltons, was found to exist predominantly as a dimer with an apparent molecular mass of 136 +/- 29 kilodaltons. The mass distribution of the enzyme molecules was unchanged in the presence of the allosteric inhibitor S-adenosylmethionine. Examination of negatively stained protein molecules suggested that each subunit of the dimer consists of two globular domains of approximately equal size. Limited proteolysis of the enzyme by trypsin gave results which were entirely consistent with the presence of two domains per subunit. In the presence of 1% trypsin, the enzyme was cleaved into two fragments. The masses of these fragments were 39 and 36 kilodaltons as assessed by polyacrylamide gel electrophoresis in the presence of sodium dodecyl sulfate. Tryptic cleavage did not lead to loss of NADPH-menadione or NADPH-methylenetetrahydrofolate oxidoreductase activity, and the flavin prosthetic group remained bound to the protein. However, the cleaved protein was completely desensitized with respect to inhibition by S-adenosylmethionine. These results suggest that each subunit of methylenetetrahydrofolate reductase contains two domains and that allosteric inhibition requires specific interactions between these domains. The region between these two domains appears to be very sensitive to proteolysis, while the domains themselves are relatively resistant to further degradation.

Nitrous oxide (N2O) is commonly used as an anesthetic agent. Prolonged exposure to N2O leads to megaloblastic anemia in humans and to loss of methionine synthase activity in vertebrates. We now report that purified preparations of cobalamin-dependent methionine synthase (5-methyltetrahydrofolate-homocysteine methyltransferase, EC 2.1.1.13) from both Escherichia coli and pig liver are irreversibly inactivated during turnover in buffers saturated with N2O. Inactivation by N2O occurs only in the presence of all components required for turnover: homocysteine, methyltetrahydrofolate, adenosylmethionine, and a reducing system. Reisolation of the inactivated E. coli enzyme after turnover in the presence of N2O resulted in significant losses of bound cobalamin and of protein as compared to controls where the enzyme was subjected to turnover in N2-equilibrated buffers before reisolation. However, N2O inactivation was not associated with major changes in the visible absorbance spectrum of the remaining enzyme-bound cobalamin. We postulate that N2O acts by one-electron oxidation of the cob(I)alamin form of the enzyme which is generated transiently during turnover with the formation of cob(II)alamin, N2, and hydroxyl radical. Generation of hydroxyl radical at the active site of the enzyme could explain the observed irreversible loss of enzyme activity.

Methylenetetrahydrofolate reductase from pig liver has been purified to homogeneity, as judged by several criteria: (i) a single band with a subunit molecular weight of 77,300 following polyacrylamide gel electrophoresis in the presence of sodium dodecyl sulfate; (ii) a molecular weight determined by amino acid analysis of 74,500 per flavin, in agreement with the subunit molecular weight; and (iii) constant specific activities in the peak fractions during the final chromatography step. The purified enzyme exhibits a typical flavoprotein absorption spectrum. Methylenetetrahydrofolate reductase is a minor constituent of pig liver, and to obtain homogeneous enzyme, a 32,000-fold purification must be accomplished. The preparation described herein attains such purification in 5 steps and with a 14% yield. The enzyme isolated in this fashion is active and stable, and contains a stoichiometric complement of FAD. The enzyme is reducible under anaerobic conditions by 5-deazaflavin/EDTA/light or by NADPH. Reduction of 1 mol of enzyme-bound FAD requires 1.1 mol of NADPH. The reduced enzyme can be reoxidized by (6-R)-methylenetetrahydrofolate, again with nearly 1:1 stoichiometry. Steady state kinetic measurements of the NADPH-methylenetetrahydrofolate oxidoreductase activity give parallel line double reciprocal plots. The turnover number per mol of enzyme-bound flavin is 1600/min under Vmax conditions. The spectrum of the enzyme-bound flavin is significantly perturbed by the binding of S-adenosylmethionine, a metabolite known to be an allosteric modulator of the enzyme.
