1. Charge-density-wave control by adatom manipulation and its effect on magnetic nanostructures

Abstract

Charge-density waves (CDWs) are correlated states of matter, where the electronic density is modulated periodically as a consequence of electronic and phononic interactions. Often, CDW phases coexist with other correlated states, such as superconductivity, spin-density waves or Mott insulators. Controlling CDW phases may therefore enable the manipulation of the energy landscape of these interacting states. 2H-NbSe$_2$ is a prime example of a transition metal dichalcogenide (TMDC) hosting CDW order and superconductivity. The CDW is of incommensurate nature resulting in different CDW-to-lattice alignments at the atomic scale. Here, we use the tip of a scanning tunneling microscope (STM) to position adatoms on the surface and induce reversible switching of the CDW domains. We show that the domain structure critically affects other local interactions, namely the hybridization of Yu-Shiba-Rusinov (YSR) states, which arise from exchange interactions of magnetic Fe atoms with the superconductor. Our results suggest that CDW manipulation could also be used to introduce domain walls in coupled spin chains on superconductors, potentially also affecting topological superconductivity.

Abstract

Single-stranded RNA viruses co-assemble their capsid with the genome and variations in capsid structures can have significant functional relevance. In particular, viruses need to respond to a dehydrating environment to prevent genomic degradation and remain active upon rehydration. Theoretical work has predicted low-energy buckling transitions in icosahedral capsids which could protect the virus from further dehydration. However, there has been no direct experimental evidence, nor molecular mechanism, for such behaviour. Here we observe this transition using X-ray single particle imaging of MS2 bacteriophages after aerosolization. Using a combination of machine learning tools, we classify hundreds of thousands of single particle diffraction patterns to learn the structural landscape of the capsid morphology as a function of time spent in the aerosol phase. We found a previously unreported compact conformation as well as intermediate structures which suggest an incoherent buckling transition which does not preserve icosahedral symmetry. Finally, we propose a mechanism of this buckling, where a single 19-residue loop is destabilised, leading to the large observed morphology change. Our results provide experimental evidence for a mechanism by which viral capsids protect themselves from dehydration. Abstract

Magnetic adatoms on superconductors give rise to Yu-Shiba-Rusinov (YSR) states that hold considerable interest for the design of topological superconductivity. Here, we show that YSR states are also an ideal platform to engineer structures with intricate wave-function symmetries. We assemble structures of iron atoms on the quasi-two-dimensional superconductor $2H$-NbSe$_2$. The Yu-Shiba-Rusinov wave functions of individual atoms extend over several nanometers enabling hybridization even at large adatom spacing. We show that the substrate can be exploited to deliberately break symmetries of the adatom structure in ways unachievable in the gas phase. We highlight this potential by designing chiral wave functions of triangular adatom structures confined within a plane. Our results significantly expand the range of interesting quantum states that can be engineered using arrays of magnetic adatoms on superconductors.

Abstract

Abstract Microbubbles (MB) are widely used as contrast agents for ultrasound (US) imaging and US‐enhanced drug delivery. Polymeric MB are highly suitable for these applications because of their acoustic responsiveness, high drug loading capability, and ease of surface functionalization. While many studies have focused on using polymeric MB for diagnostic and therapeutic purposes, relatively little attention has thus far been paid to improving their inherent imaging and drug delivery features. This study here shows that manipulating the polymer chemistry of poly(butyl cyanoacrylate) (PBCA) MB via temporarily mixing the monomer with the monomer‐mimetic butyl cyanoacetate (BCC) during the polymerization process improves the drug loading capacity of PBCA MB by more than twofold, and the in vitro and in vivo acoustic responses of PBCA MB by more than tenfold. Computer simulations and physisorption experiments show that BCC manipulates the growth of PBCA polymer chains and creates nanocavities in the MB shell, endowing PBCA MB with greater drug entrapment capability and stronger acoustic properties. Notably, because BCC can be readily and completely removed during MB purification, the resulting formulation does not include any residual reagent beyond the ones already present in current PBCA‐based MB products, facilitating the potential translation of next‐generation PBCA MB.

Abstract

Introduction Inadequate treatment of enlarged lateral lymph nodes (LLNs) in rectal cancer patients is associated with an increased lateral local recurrence (LLR) risk, despite neoadjuvant treatment and total mesorectal excision (TME) surgery. There is a promising role for LLN dissection (LLND) to lower this risk, but this challenging procedure requires appropriate training. This study protocol describes a prospective evaluation of oncological outcomes after standardised treatment based on multidisciplinary training, thereby aiming for a 50% reduction in LLR rate.Methods and analysis A prospective registration study will be opened in hospitals in which the involved multidisciplinary team members (radiologists, radiation oncologists, surgeons and pathologists) have received dedicated training to enhance knowledge and awareness of LLNs and in which standardised treatment including LLND has been implemented. Patients with rectal cancer and at least one enlarged LLN (short-axis ≥7.0 mm), or intermediate LLN (short-axis 5.0–6.9 mm) with at least one malignant feature on primary MRI, evaluated by a trained radiologist, are eligible. Patients will undergo neoadjuvant treatment by trained radiation oncologists, followed by TME surgery in combination with a minimally invasive, nerve-sparing LLND performed by trained surgeons. LLND specimens are evaluated by trained pathologists or grossing assistants. The primary outcome is LLR rate 3 years postoperatively. Secondary outcomes are morbidity, disease-free survival, overall survival and quality of life. To demonstrate a significant reduction in LLR rate from 13% (based on historical control data) to 6% after optimised treatment, 200 patients with enlarged LLNs are required.Ethics and dissemination The medical ethics board of the Vrije Universiteit Medical Centre (VUMC), the Netherlands, approved the study on 23 November 2022 (reference: 2021.0524). Participating centres must obtain local approval and participants are required to provide written informed consent. Background and purpose: This study assessed quality of life (QoL) and clinical outcomes in rectal cancer patients treated with magnetic resonance (MR) guided short-course radiation therapy (SCRT) on a 1.5 Tesla (T) MR-Linac during the first 12 months after treatment. Abstract

Magnetic adatom chains on surfaces constitute fascinating quantum spin systems. Superconducting substrates suppress interactions with bulk electronic excitations but couple the adatom spins to a chain of subgap Yu-Shiba-Rusinov (YSR) quasiparticles. Using a scanning tunneling microscope, we investigate such correlated spin-fermion systems by constructing Fe chains adatom by adatom on superconducting NbSe$_2$. The adatoms couple entirely via the substrate, retaining their quantum spin nature. In dimers, we observe that the deepest YSR state undergoes a quantum phase transition due to Ruderman-Kittel-Kasuya-Yosida interactions, a distinct signature of quantum spins. Chains exhibit coherent hybridization and band formation of the YSR excitations, indicating ferromagnetic coupling. Longer chains develop separate domains due to coexisting charge-density-wave order of NbSe$_2$. Despite the spin-orbit-coupled substrate, we find no signatures of Majoranas, possibly because quantum spins reduce the parameter range for topological superconductivity. We suggest that adatom chains are versatile systems for investigating correlated-electron physics and its interplay with topological superconductivity.

Abstract

A novel scaffold design has been created to enhance tissue engineering and regenerative medicine by optimizing the controlled, prolonged release of Hepatocyte Growth Factor (HGF), a powerful chemoattractant for endogenous mesenchymal stem cells. We present a new stacked scaffold that is made up of three different fibrin gel layers, each of which has HGF integrated into the matrix. The design attempts to preserve HGF’s regenerative properties for long periods of time, which is necessary for complex tissue regeneration. These multi-layered fibrin gels have been mechanically evaluated using rheometry, and their degradation behavior has been studied using D-Dimer ELISA. Understanding the kinetics of HGF release from this novel scaffold configuration is essential for understanding HGF’s long-term sustained bioactivity. A range of cell-based tests were carried out to verify the functionality of HGF following extended incorporation. These tests included 2-photon microscopy using phalloidin staining to examine cellular morphology, SEM analysis for scaffold–cell interactions, and scratch and scatter assays to assess migration and motility. The analyses show that the novel stacking scaffold promotes vital cellular processes for tissue regeneration in addition to supporting HGF’s bioactivity. This scaffold design was developed for in situ tissue engineering. Using the body as a bioreactor, the scaffold should recruit mesenchymal stem cells from their niche, thus combining the regenerative abilities of HGF and MSCs to promote tissue remodeling and wound repair.

Abstract

Osteosarcopenia, the concurrent presence of sarcopenia and osteopenia/osteoporosis, poses a significant health risk to older adults, yet its impact on clinical outcomes is not fully understood. The aim of this prospective, longitudinal multicentre study was to examine the impact of osteosarcopenia on 3-year mortality and unplanned hospitalizations among 572 older hospitalized patients (mean age 75.1 ± 10.8 years, 78% female). Sarcopenia and low bone mineral density (BMD) were evaluated using Dual Energy X-ray Absorptiometry and the European Working Group on Sarcopenia in Older People (EWGSOP2) and WHO criteria, respectively. Among participants, 76% had low BMD, 9% were sarcopenic, and 8% had osteosarcopenia. Individuals with osteosarcopenia experienced a significantly higher rate of mortality (46%, p < 001) and unplanned hospitalization (86%, p < 001) compared to those without this condition. Moreover, “healthy” subjects—those without sarcopenia or low BMD—showed markedly lower 3-year mortality (9%, p < 001) and less unplanned hospitalization (53%, p < 001). The presence of osteosarcopenia (p = 0.009) increased the 3-year mortality risk by 30% over sarcopenia alone and by 8% over low BMD alone, underscoring the severe health implications of concurrent muscle and bone deterioration. This study highlights the substantial impact of osteosarcopenia on mortality among older adults, emphasizing the need for targeted diagnostic and therapeutic strategies.

Abstract

This study focuses on enhancing controllable fibrin-based hydrogels for tissue engineering, addressing existing weaknesses. By integrating a novel copolymer, we improved the foundation for cell-based angiogenesis with adaptable structural features. Tissue engineering often faces challenges like waste disposal and nutrient supply beyond the 200 µm diffusion limit. Angiogenesis breaks through this limitation, allowing the construction of larger constructs. Our innovative scaffold combination significantly boosts angiogenesis, resulting in longer branches and more capillary network junctions. The copolymer attached to fibrin fibers enables precise adjustment of hydrogel mechanical dynamic properties for specific applications. Our material proves effective for angiogenesis, even under suppression factors like suramin. In our study, we prepared fibrin-based hydrogels with and without the copolymer PVP12400-co-GMA10mol%. Using a co-culture system of human umbilical vein endothelial cells (HUVEC) and mesenchymal stem cells (MSC), we analyzed angiogenetic behavior on and within the modified hydrogels. Capillary-like structures were reproducibly formed on different surfaces, demonstrating the general feasibility of three-dimensional endothelial cell networks in fibrin-based hydrogels. This highlights the biomaterial’s suitability for in vitro pre-vascularization of biohybrid implants.

Abstract

Summary: Background: For cancer patients to effectively engage in decision making, they require comprehensive and understandable information regarding treatment options and their associated outcomes. We developed an online prediction tool and supporting communication skills training to assist healthcare providers (HCPs) in this complex task. This study aims to assess the impact of this combined intervention (prediction tool and training) on the communication practices of HCPs when discussing treatment options. Methods: We conducted a multicenter intervention trial using a pragmatic stepped wedge design (NCT04232735). Standardized Patient Assessments (simulated consultations) using cases of esophageal and gastric cancer patients, were performed before and after the combined intervention (March 2020 to July 2022). Audio recordings were analyzed using an observational coding scale, rating all utterances of treatment outcome information on the primary outcome–precision of provided outcome information–and on secondary outcomes–such as: personalization, tailoring and use of visualizations. Pre vs. post measurements were compared in order to assess the effect of the intervention. Findings: 31 HCPs of 11 different centers in the Netherlands participated. The tool and training significantly affected the precision of the overall communicated treatment outcome information (p = 0.001, median difference 6.93, IQR (−0.32 to 12.44)). In the curative setting, survival information was significantly more precise after the intervention (p = 0.029). In the palliative setting, information about side effects was more precise (p < 0.001). Interpretation: A prediction tool and communication skills training for HCPs improves the precision of treatment information on outcomes in simulated consultations. The next step is to examine the effect of such interventions on communication in clinical practice and on patient-reported outcomes. Abstract Hintergrund Das Ausmaß in dem Menschen körperlich aktiv sind hängt vom sozialen Gradienten ab. Zahlreiche Studien haben gezeigt, dass insbesondere Menschen mit sozialer Benachteiligung die Empfehlungen für Bewegung nicht umsetzen. Bevölkerungsbezogene Maßnahmen sind geeignet, um alle Menschen zu erreichen, gleichzeitig gibt es wissenschaftliche Hinweise dafür, dass dadurch gesundheitliche Ungleichheiten verstärkt werden können. Ziel dieses Umbrella Reviews war es daher, wirksame bevölkerungsbezogene Ansätze zur Förderung der körperlichen Aktivität mit besonderem Fokus auf gesundheitliche Chancengleichheit zu identifizieren. Methoden Mittels sechs elektronischen Wissenschaftsdatenbanken wurde für den Zeitraum von 2015 bis 2021 nach systematischen Reviews zur bevölkerungsbezogenen Bewegungsförderung gesucht. Darüber hinaus wurde auch eine Handsuche in den Referenzlisten gefundener Artikel sowie eine Suche nach grauer Literatur durchgeführt. Zwei unabhängige Gutachter:innen überprüften anhand von Ein- und Ausschlusskriterien die Titel und Abstracts der potenziell relevanten Literatur und führten für jedes eingeschlossene Review eine Qualitätsbewertung durch. Die Ergebnisse der identifizieren Übersichtsarbeiten zu bevölkerungsbezogenen Ansätzen zur Bewegungsförderung mit Fokus auf Menschen mit sozialer Benachteiligung und/oder gesundheitliche Chancengleichheit wurden narrativ zusammengefasst. Ergebnisse Insgesamt wurden 4.411 Reviews identifiziert. Nach einer systematischen Überprüfung, erfüllten sechs Reviews die Einschlusskriterien und wurden eingeschlossen, nachdem sie alle als qualitativ hochwertig eingestuft worden waren. Massenmediale Kampagnen, motivationale Entscheidungshilfen, umweltbezogene Ansätze, politikbezogene Ansätze und gemeindebasierte Mehrkomponentenansätze können das Bewegungsverhalten in der Allgemeinbevölkerung fördern. Für Bevölkerungsgruppen mit sozialer Benachteiligung liegt Evidenz vor, dass massenmediale Kampagnen, motivationale Entscheidungshilfen sowie politikbezogene Ansätze wirksam sind, sofern diese bedarfsgerecht (tailored) angepasst werden. Über umweltbezogene Ansätze wurden widersprüchliche Ergebnisse berichtet. Zu gemeindebasierten Mehrkomponentenansätzen fehlen bislang Ergebnisse hinsichtlich der Verbesserung von gesundheitlicher Chancengleichheit. Schlussfolgerung Es wurden mehrere wirksame Ansätze zur Bewegungsförderung der Allgemeinbevölkerung identifiziert, jedoch sind Erkenntnisse über den Einfluss auf die gesundheitliche Chancengleichheit gering. Künftige Studien sollten daher diesem fehlenden Fokus mehr Aufmerksamkeit widmen. Des Weiteren mangelt es an Wissen über die Art der bedarfsgerechten Anpassung (Tailoring) sowie die langfristigen Auswirkungen bevölkerungsbezogener Ansätze zur Bewegungsförderung. Abstract

Damit Lehrkräfte ihre Schülerinnen und Schüler adaptiv unterstützen können, sind diagnostische Kompetenzen wichtig. Lehrkräfte müssen falsche oder ungewöhnliche Aufgabenbearbeitungen rasch einschätzen und den Lernenden geeignete Hinweise geben können. Das erfordert fachliches Wissen, in der Grundschule beispielsweise zum dezimalen Stellenwertsystem und zu den Grundoperationen. Präsentiert werden ausgewählte Ergebnisse einer Fragebogenuntersuchung. n = 224 amtierenden Regellehrkäften und n = 144 Förderlehrkräften (Grundschule) wurde eine Fallvignette vorgelegt, in der ein Kind Divisionsaufgaben wie folgt gelöst hatte: 35 : 5 = 7; 350 : 50 = 700; 350 : 5 = 70. Die Lehrkräfte beantworteten schriftlich folgende Frage: Warum macht das Kind vermutlich diesen Fehler? Begründen Sie. Die Antworten wurden nach einem vorgängig erarbeiteten Manual codiert. Der größte Teil der Lehrkräfte sah die Schwierigkeit des Kindes in einer Verwechslung der Division mit der Multiplikation. Nur ein kleiner Teil der Lehrkräfte nannte als Problem die fehlende Einsicht ins Stellenwertsystem als mögliche Fehlerursache. Zudem zeigte sich, dass viele Lehrkräfte Schwierigkeiten hatten, ihre Vermutungen fachsprachlich korrekt auszudrücken. Die Ergebnisse geben Hinweise, welche Themen und Aspekte in der Ausbildung besonders berücksichtigt werden müssen.
