Abstract

International audience; This paper presents a microwave sensor composed of one millifluidic channel that operates over a frequency range from 1 GHz to 4 GHz and is used for microwave spectroscopy of liquids with millilitre volumes. The accuracy of the device is first evaluated with BSA solutions of different concentrations and then with complex biological liquids such as hen egg yolk and albumen samples. Low standard deviations of the measured values of modulus and phase contrasts of the transmission coefficient S21 are obtained. These values correspond to 0.02 and 0,18 respectively for BSA solutions, as well as 0.05 and 0.03 for 10 samples of hen egg yolk and white respectively over the selected frequency range. Such accuracies enable the use of such a sensor for various applicative domains, including agronomy for which the investigation of the dielectric properties of eggs may be of interest for quality, freshness or status discrimination purpose.

Abstract

International audience; The PPILOW project aims to co-construct innovations to improve Poultry and Pig Welfare in Low-input outdoor and Organic farming systems through a multi-actor approach. PPILOW implements a participatory approach for proposing and studying welfare-improvement levers. It will provide a combination of practical solutions that can be applied at a pan-European level with specific adjustments depending on citizen’s expectations and the target market. The multi-actor approach consists in involving end-users including farmers, breeding companies, feed producers, consumer associations, retailers, advisers, processors, and scientists in National Practitioner Groups (NPG) in six participating countries. PPILOW partners facilitate the groups by connecting NPG at European level, transferring scientific information, interacting with partners engaged in animal experiments, and co-creating innovations rising from NPG-specific demands. They co-build with PPILOW partners welfare self-assessment tools (development of the PIGLOW app for pigs and refinement of the EBENE® app for poultry), and innovative breeding, feeding, and rearing strategies and techniques to improve the welfare of animals. They co-design protocols, test innovations on farm, and disseminate the results. In turn, they receive insights on methods and scientific results, and inputs from other NPG reinforcing the value of the expected outcomes. Approaches focus on avoiding physical damage and the elimination of layer male chicks, on reducing boar taint of intact male pigs, promoting positive behaviours, animal health, and robustness through field studies with pigs and poultry. Multicriteria analyses of the most effective levers of welfare improvement will be performed to evaluate their economic, social, and environmental impacts based on the ‘One Welfare’ concept; economic and business models will also be developed. To ensure the rapid uptake of the project results by end-users, the close involvement of PPILOW’s NPG throughout the EU will ensure disseminationactivities and the facilitation of change. Abstract

International audience; The remarkable reproductive success of avian species relies upon the integrated defensive features of the eggshell, acellular membranes, physicochemical characteristics and antimicrobial molecules. These egg structures are dramatically modified during embryonic development: solubilization of internal eggshell components, disintegration of acellular membranes that serve as substratum for the growth of extraembryonic structures, and assimilation/transfer of egg white and yolk antimicrobials to other compartments. The apparent degradation in basic egg defenses is counter balanced by the establishment of other protective and functional structures (namely the amniotic, the allantoic and the yolk sacs) that support several vital functions until hatching. Their cellular complexity, as well as the functional and molecular specificity of these structures, add an additional defensive layer. This chapter reviews the different levels of these interconnected egg defenses that assist the embryo throughout its development, beginning with the egg defenses from maternal origin, to the development of extra-embryonic protective structures.

Abstract

International audience; In oviparous species, the egg contains protective systems and provides energy and nutrients that support the development of the embryo until hatching. During embryonic development, extraembryonic structures are vital to assimilate egg nutrients, to protect the embryo, to ensure gaseous exchange and to accumulate metabolic waste. The chorioallantoic membrane (CAM), which develops in close contact with the inner eggshell from day 5 of incubation, is a highly vascularized structure. The CAM is located between the eggshell membranes (ESM, outer side) and the allantoic fluid that collects embryonic waste (inner side). The CAM was reported to be involved in decalcification of the inner eggshell and mineral transport from the eggshell to the embryo (for mineralization of its skeleton), in acid-base homeostasis, and in ion and H2O reabsorption from the allantoic fluid. To decipher the kinetics of its structural maturation, we compared the structure of the inner and outer surfaces of the CAM by electron microscopy. In parallel, ESM and the eggshell were analyzed to better appreciate the impact of CAM development on its surrounding structures. Eggs from Ross 308 broiler hens were incubated for 11 or 15 days (ED11 vs ED15, early and mature stage of CAM development, respectively). At each time point, eggs were broken, and the eggshell, ESM and the CAM were gently separated for analysis by scanning electron microscopy. Images corresponding to the cross-section and the inner side of the eggshell, eggshell membranes (ESM, outer and inner sides) and CAM (outer and inner sides) were obtained. At ED15, results showed that the inner side of the eggshell is degraded (decalcification) during embryonic development while the mammillary knobs of the eggshell remain tightly attached to the external membrane of the ESM. The CAM also undergoes major structural changes with some villous cells that emerge from its outer surface to reach the eggshell membrane and the eggshell. The molecular components associated with eggshell decalcification and mineral transport from the eggshell to the embryo via CAM’s vasculature remain essentially unknown. This study has been recently complemented by a transcriptomic analysis of the CAM (RNA sequencing) at these two developmental stages. Altogether, we believe that the combined findings of this work will lead to better understanding of CAM functions that are vital for embryonic development and skeletal mineralization during incubation.

Abstract

The control of egg quality is essential for the egg industry. Indeed, any alterations related to external factors, such as hen age or egg storage conditions, may have serious consequences on consumer’s health and/or on the economy of the egg industry. Some internal egg quality indicators are visible and are mostly measured after breaking the egg, while others (air chamber volume, yolk movement during egg storage) are less easily measurable. The objective of this work is to assess the use of tomographic imaging tools (magnetic resonance imaging or MRI for soft tissues, computed tomography or CT-scan for hard tissues) as non-invasive and non-destructive methods for the characterization of egg quality. Unfertilized eggs from Lohmann Tradition hens were collected on the day of lay, stored for 7, 14 and 28 days at two temperatures (4 and 20°C), analyzed by MRI (determination of yolk and albumen volumes, mapping of T2 relaxation times) and CT-scan (air chamber volumetry), then characterized with standard egg quality measurement tools, after egg breaking. The results of this study show that imaging can be used as a non-invasive tool for measuring internal egg quality while providing new possibilities for measuring criteria that are difficult to assess once the egg has been broken. In the future, the results of this study may allow the creation of a new reference for egg quality., La maitrise qualité de l’oeuf est essentielle pour la filière des oeufs de consommation. Certaines altérations, liées à des facteurs externes comme l’âge des poules ou les conditions de conservation par exemple, peuvent en effet avoir des impacts négatifs d’un point de vue sanitaire et économique, notamment. Certains indicateurs de qualité interne de l’oeuf sont visibles et sont pour la plupart mesurés après le cassage de l’oeuf. D’autres, comme le volume de la chambre à air ou le déplacement du jaune dans l’oeuf au cours du stockage sont moins facilement mesurables. L’objectif de ce travail est d’évaluer l’utilisation d’outils d’imagerie tomographique (l’imagerie par résonance magnétique ou IRM pour les tissus mous, la tomodensitométrie ou CT-scan pour les tissus durs) comme méthodes non invasives et non destructives pour caractériser la qualité des oeufs. Des oeufs de consommation de poules Lohmann Tradition ont été collectés le jour de la ponte, stockés pendant 7, 14 et 28 jours à deux températures (4 et 20°C), analysés par IRM (détermination des volumes du blanc et du jaune, cartographie des temps de relaxation T2) et CT-scan (volumétrie de la chambre à air), puis caractérisés avec les outils de mesures standards de la qualité de l’oeuf, après cassage. Les résultats obtenus révèlent que l’imagerie peut être utilisée comme outil de mesure non invasive de la qualité interne de l’oeuf tout en apportant des nouvelles possibilités de mesure de critères difficilement appréciables une fois l’oeuf cassé. Les résultats de cette étude pourront permettre à terme d’envisager la création d’un nouveau référentiel de la qualité de l’oeuf.

Abstract

Le stockage des œufs fécondés avant incubation est une pratique couramment utilisée dans les couvoirs pour synchroniser les éclosions. Cependant, certaines conditions choisies pour stocker les œufs peuvent avoir un impact négatif sur la qualité des œufs et la viabilité des embryons. Alors que la température de stockage est relativement comparable d’un couvoir à l’autre (16 à 18°C), la durée de stockage peut varier de trois à plus de dix jours. L’objectif principal de ce projet était donc d’évaluer l’impact de la durée de stockage sur la qualité de l’œuf à couver et sur le développement embryonnaire. Nous avons, dans un premier temps, exploré l'effet de la durée de stockage (0, 3 ou 10 jours) sur la qualité des œufs (Poulet de chair, Ross 308), en utilisant la tomographie assistée par ordinateur (CT-scan, mesure non invasive) et des mesures classiques des paramètres de qualité des œufs (après cassage : poids des œufs, poids et résistance de la coquille, pH du blanc d'œuf, unités de Haugh, indice et couleur du jaune). Les résultats ont révélé que la durée de stockage jusqu'à 10 jours affecte négativement certains paramètres de qualité tels que l’indice et volume du jaune, le volume de la chambre à air et le pH du blanc d'œuf. Les lots d'œufs conservés pendant trois ou dix jours ont été ensuite incubés pendant 11, 13 ou 15 jours (38,5°C, 55% d’humidité relative). Les œufs ont ensuite été analysés par imagerie par résonance magnétique (IRM) et CT-scan pour suivre le développement de l'embryon et les changements/mouvements des différents constituants de l’œuf pendant l'incubation. Les résultats ont révélé que le stockage des œufs pendant dix jours est associé à une diminution de la croissance de l'embryon, visible dès le onzième jour d'incubation (diminution des volumes du cerveau et de l'embryon). Par ailleurs, nous avons vérifié que la durée de stockage dans les conditions du couvoir n’affectait ni la viabilité embryonnaire, ni le sex-ratio. Nos analyses montrent donc que la durée de conservation des œufs a un impact négatif sur la croissance embryonnaire. Elles corroborent le fait que ce paramètre est à prendre en compte pour synchroniser les stades embryonnaires et potentiellement réduire la fenêtre d'éclosion. En outre, nos résultats révèlent que certaines technologies d'imagerie (CT, IRM) sont des outils pertinents pour évaluer la qualité des œufs avant l'incubation de manière non invasive et pour étudier l'impact des conditions de stockage (voire d'incubation) sur le développement et la croissance de l'embryon.

Abstract

International audience; Avian eggs possess very efficient and orchestrated systems to protect the embryo during incubation, until hatch. Although the internal components of the egg are assumed to be sterile, the surface of the eggshell is covered by microbes (essentially bacteria) that may contribute to prevent eggshell colonization by pathogenic bacteria, through direct inhibition and/or competitive exclusion. The composition of the eggshell microbiome is a heritage from both maternal microbiota (caeca/faeces) where the egg meets caecal secretions in the cloacal segment during oviposition, and from the nesting environment (contaminated litter/feathers and air environment including dust). At laying time, the egg surface is still moisturized, but will progressively dehydrate during incubation. The surface characteristics of the egg, the loss of the moisture layer and the presence of antimicrobial molecules composing the cuticle are likely to dictate the bacterial communities that will survive on the surface of the eggshell. Although the literature on the composition of the eggshell microbiome of eggs originating from current commercial chicken hens is quite sporadic, this talk aims to provide an overview of the bacterial communities that colonize the chicken eggshell surface, and will discuss the role of the eggshell microbiota as the first barrier against pathogenic bacteria.

Abstract

International audience; Biomineralized structures can function as a barrier to the external environment, and as such are conceptually entwined with innate immune processes. Disentangling immune and biomineralization mechanisms represents a significant challenge for researchers trying to understand how organisms could integrate biomineral formation and plasticity with maintenance of critical innate immune protection. In fact, there is increasing evidence that immune proteins can serve genuine dual-functional roles, both in regulation of biomineralization, as well as resisting pathogens. This awareness is growing in models as diverse as the dual-functionning haemocytes of marine bivalves, and in mineralization / demineralization of the avian eggshell. Moreover, reef corals, in which calcification is coupled to the photosynthetic activity of their mutualistic symbiots, must adapt their innate immune system to achieve this tolerance. Cnidarian immune-related processes in response to abiotic stresses are increasingly implicated in loss of symbiosis and coral bleaching.This conference aims to bring together scientists working with diverse models of biomineralization, for an exchange of perspectives on the innate immune function of the calcified barrier and the dual role played by specific genes / proteins in these two critical functions.

Abstract

National audience; Le projet H2020 PPILOW - "Poultry and PIg Low-input and Organic production systems’ Welfare" a pour objectif la co-conception d'innovations pour améliorer le bien-être des volailles et des porcs élevés dans des systèmes de production biologiques et plein air, grâce à une approche multi-acteurs mise en œuvre par 23 partenaires dans 9 pays européens. Une approche participative permettra d’impliquer les acteurs des chaînes de production, citoyens et scientifiques pour proposer et étudier des leviers d’amélioration du bien-être des animaux. Les objectifs du projet sont en particulier de : Mettre au point des méthodes d’auto-évaluation par l’éleveur du bien-être de ses animaux ; Réduire le risque d’odeur sexuelle de la viande et les problèmes comportementaux (agressivité, montes) des porcs mâles, afin de permettre l’arrêt de la castration des porcelets ; Réduire les risques de picage chez les poules afin de permettre l’arrêt de l’épointage du bec ; Permettre l’arrêt de l’élimination juste après l’éclosion des poussins mâles de souche ponte, en explorant deux méthodes : a) utiliser des croisements génétiques qui permettent d’élever les mâles pour la viande et les femelles pour la ponte b) sexer les œufs (ovosexage) pour repérer le plus tôt possible les œufs mâles et stopper leur incubation ; Favoriser l’expression du comportement exploratoire des poulets sur les parcours plein-air; Améliorer la survie des porcelets par sélection génétique ou amélioration du logement des truies ; Améliorer la santé et le bien-être des volailles en agissant sur l’environnement dès l’incubation ou peu après l’éclosion des œufs ; Réduire voire éviter l’usage de médicaments chez le porc et la poule pondeuse en utilisant des aliments ou extraits végétaux favorisant la résistance aux maladies.Les innovations les plus prometteuses seront testées dans des élevages commerciaux en interaction avec les groupes participatifs nationaux. Des évaluations multicritères seront également réalisées en mobilisant le concept «One Welfare»1, afin d’englober les objectifs de durabilité, de bien-être animal mais aussi de bien-être humain. Des modèles économiques seront créés pour évaluer les bénéfices des stratégies élaborées dans le projet PPILOW. Enfin, des actions de diffusion seront développées en impliquant les groupes multi-acteurs tout au long du projet pour faciliter les changements de pratiques.
