Despite its massive potential, Raman imaging represents just a modest fraction of all research and clinical microscopy to date. This is due to the ultralow Raman scattering cross-sections of most biomolecules that impose low-light or photon-sparse conditions. Bioimaging under such conditions is suboptimal, as it either results in ultralow frame rates or requires increased levels of irradiance. Here, we overcome this tradeoff by introducing Raman imaging that operates at both video rates and 1,000-fold lower irradiance than state-of-the-art methods. To accomplish this, we deployed a judicially designed Airy light-sheet microscope to efficiently image large specimen regions. Further, we implemented subphoton per pixel image acquisition and reconstruction to confront issues arising from photon sparsity at just millisecond integrations. We demonstrate the versatility of our approach by imaging a variety of samples, including the three-dimensional (3D) metabolic activity of single microbial cells and the underlying cell-to-cell variability. To image such small-scale targets, we again harnessed photon sparsity to increase magnification without a field-of-view penalty, thus, overcoming another key limitation in modern light-sheet microscopy.

Burkitt and Burkitt like lymphoma (BL/BLL) are highly proliferative germinal or post-germinal B cell tumors. Few studies have evaluated the impact of autologous stem cell transplantation (ASCT) on disease outcomes.We performed a systematic review to analyze the efficacy of ASCT as frontline consolidation and for treatment of relapsed/refractory cases in adult BL/BLL.Eligible studies with clear outcome measures on the efficacy of ASCT in adult patients with BL/BLL were identified through systematic search. The overall survival (OS), progression-free survival (PFS), complete response (CR), partial response (PR), and progression/relapse were used to assess the efficacy.For patients who underwent ASCT in first CR, 5-year PFS and OS ranged between 70-78% and 70-83% respectively. For relapsed/refractory disease, 5-year PFS and OS were 27% and 31%, respectively. Patients undergoing ASCT for chemoresistant disease fared poorly with 3-year OS of 7% vs 37% for chemosensitive disease (p ≤ 0.00001). The overall response rate to ASCT for patients transplanted in first CR ranged between 71% and 93% and was 37% for patients who were transplanted in disease status other than first CR. Disease progression/relapse was observed in 16-29% of the patients transplanted in first CR, and 55% to 60% in relapsed disease.We found insufficient evidence to support ASCT over chemotherapy alone in the first remission for adult BL/BLL. Evidence supports guidelines recommending ASCT for chemosensitive disease but suggests there is no benefit to ASCT for chemoresistant disease.

Two types of cobalt porphyrins, viz., meso-tetrakis(tolylporphyrinato)cobalt(II), (TTP)Co (1), and meso-tetrakis(triphenylamino porphyrinato)cobalt(II), [(TPA)4P]Co, (2) were self-assembled via metal-ligand axial coordination of phenyl imidazole functionalized fulleropyrrolidine, ImC[Formula: see text] to form a new series of donor–acceptor constructs. A 1:2 complex formation with ImC[Formula: see text] was established in the case of (TTP)Co while for [(TPA)4P]Co only a 1:1 complex was possible to positively identify. The binding constants [Formula: see text] and [Formula: see text] for step-wise addition of ImC[Formula: see text] to (TTP)Co were found to be 1.07 × 105 and 3.20 × 104 M[Formula: see text], respectively. For [(TPA)4P]Co:ImC[Formula: see text], the measured [Formula: see text] values was found to be 6.48 × 104 M[Formula: see text], slightly smaller than that observed for (TTP)Co. Although both cobalt porphyrins were non-fluorescent, they were able to quench the fluorescence of ImC[Formula: see text] indicating occurrence of excited state events in the supramolecular donor-acceptor complexes. Electrochemistry coupled with spectroelectrochemistry, revealed the formation of cobalt(III) porphyrin cation instead of a cobalt(II) porphyrin radical cation, as the main product, during oxidation of phenyl imidazole coordinated cobalt porphyrin. With the help of computational and electrochemical results, an energy level diagram was constructed to witness excited state photo-events. Competitive energy and electron transfer from excited CoP to coordinated ImC[Formula: see text], and electron transfer from Im1C[Formula: see text]* to cobalt(II) porphyrin resulting into the formation of PCo[Formula: see text]:ImC[Formula: see text] charge separated state was possible to envision from the energy diagram. Finally, using femtosecond transient absorption spectroscopy and data analysis by Glotaran, it was possible to establish sequential occurrence of energy transfer and charge separation processes. The lifetime of the final charge separated state was [Formula: see text] 2 ns. A slightly better charge stabilization was observed in the case of [(TPA)4P]Co:ImC[Formula: see text] due to the presence of electron rich, peripheral triphenylamine substituents on the cobalt porphyrin.

The main objective of the study was to see the association of dyslipidemia in preeclampsia (PET) as compare to normotensive (NT) patients. It was a descriptive study done in Nepal Medical College Teaching Hospital Department of Obstetrics and Gynecology from April 2019 from April 2020. A total of 75 PET (test group) and 75 NT (control group) in their third trimester were included. Their fasting lipid profile was studied. It was found that triglyceride (TG) and very low density lipoprotein (VLDL) was significantly high among PET group as compared to that of NT patients. The mean TG of PET group was 266.11± 98.61 and the NT group was 187±58.56 (p=0.00) and the mean VLDL of PET group was 46.00±15.31 and NT group was 37.88±12.85 (p=0.001). On the other hand, the total cholesterol (TC), low density lipoprotein (LDL) and high density lipoprotein (HDL) were not significantly different in both the groups. It was also found that with the severity of the disease (Preeclampsia), the level of TG and VLDL increased significantly. Therefore, among the investigation done for PET, we should include lipid profile, especially the TG and VLDL. Furthermore, TG and VLDL should be measured serially (at least once a week) so as to predict the progress of PET and manage the patient accordingly.

Garden pea is one of the economically and nutritionally most important winter vegetable crops in Nepal. Garden pea production is an emerging farm enterprise of Nepalese farmers of Eastern hill and mountain. The study was conducted to determine profitability, to investigate whether factors of production are being used in economic way, and to find out extension strategies for increasing income of farmers in Dhankuta district of Eastern Nepal. A total of 120 garden pea producing farmers, (60 from Chhathar-Jorpati Rural Municipality and 60 from Mahalaxmi Municipality of Dhankuta), were randomly selected and interviewed using semistructured interview schedule. Descriptive statistics and Cobb-Douglas production function were used to analyze the collected data. The benefit cost ratio (3.54) indicates that the garden pea production in study site was profitable with productivity of 6.59 Mt ha-1 per hectare. The production function revealed that cost on seed and labor have statistically significant effect on gross returns. In contrast, the effect of cost cost on plant nutrients and pesticides were not statistically significant. The sum of regression coefficients was 1.04 which indicates that production function exhibited an increasing return to scale. Thus, extension strategies should be primarily focused on introducing high yielding varieties, use of high quality seeds, providing trainings on improved farming practices, increasing value addition practices and strengthening market infrastructures. Intestinal parasitic infections are among the major public and socioeconomic concerns that have adversely affected the well-being of the poor in developing countries. This research work aimed to inspect the prevalence of gastro-intestinal (GI) among Sarki ethnic group of Pala Rural Municipality, Baglung, Nepal. Total 498 stool samples collected in dry, screw capped clean plastic vails and studied for the presence of parasites using direct smear method as well as by concentration method. Overall parasite positive rate was found to be 31.32% (156/498). The prevalence of single infection was higher than double infection among Sarki ethnic group. There were no significant differences in prevalence of parasites among different age-groups, sexes and villages. Positive rate was considerably higher in females (47.5%; 38/80) compared with males (32.5%; 26/80) (P0.05). Altogether six genera of parasites were encountered. Of them, Trichuris trichura (53.20%; 264/498) was the most prevalent parasite followed by Ascaris lumbricoides (33.97%; 169/498), Entamoeba coli (4.49%; 22/498), Taenia sp. (3.21%; 16/498), Strongyloides stercoralis (2.56%; 13/498) and Hymenolepis nana (2.56%; 13/498). Remarkable prevalence of intestinal parasitic infection was indicated by the present study carried out among Sarki ethnic group of Pala Rural Municipality, Baglung district of Nepal.

The formation of a high-energy, long-lived radical ion-pair by electron transfer exclusively from the triplet excited state, is demonstrated in a newly synthesized platinum porphyrin-fullerene dyad, in which the porphyrin ring is modified with three electron rich triphenylamine entities. The spin selectivity of the electron transfer leading to the formation of the radical ion-pair is demonstrated using time-resolved optical and EPR spectroscopic techniques.

We report measurements of the parity-conserving beam-normal single-spin elastic scattering asymmetries $B_n$ on $^{12}$C and $^{27}$Al, obtained with an electron beam polarized transverse to its momentum direction. These measurements add an additional kinematic point to a series of previous measurements of $B_n$ on $^{12}$C and provide a first measurement on $^{27}$Al. The experiment utilized the Qweak apparatus at Jefferson Lab with a beam energy of 1.158 GeV. The average lab scattering angle for both targets was 7.7 degrees, and the average $Q^2$ for both targets was 0.02437 GeV$^2$ (Q=0.1561 GeV). The asymmetries are $B_n$ = -10.68 $\pm$ 0.90 stat) $\pm$ 0.57 (syst) ppm for $^{12}$C and $B_n$ = -12.16 $\pm$ 0.58 (stat) $\pm$ 0.62 (syst) ppm for $^{27}$Al. The results are consistent with theoretical predictions, and are compared to existing data. When scaled by Z/A, the Q-dependence of all the far-forward angle (theta < 10 degrees) data from $^{1}$H to $^{27}$Al can be described by the same slope out to $Q \approx 0.35$ GeV. Larger-angle data from other experiments in the same Q range are consistent with a slope about twice as steep.

We report the integration of quantitative-phase imaging (QPI) with light-sheet (LS) fluorescent microscopy on to a standard inverted microscope that retains compatibility with microfluidics. QPI enables label-free imaging and number-density quantification of single cells and their organelles. Conversely, LS yields considerable speed and phototoxicity gains in quantifying the 4D dynamics of gene-encoded fluorescent biomarkers. We will detail the system design that relied on spatial light interferometry for QPI and an accelerating Airy-beam light-sheet for fluorescence, its performance, as well as results of a representative multivariate imaging analysis of single-cell metabolism.

Understanding the nucleon spin structure in the regime where the strong interaction becomes truly strong poses a challenge to both experiment and theory. At energy scales below the nucleon mass of about 1 GeV, the intense interaction among the quarks and gluons inside the nucleon makes them highly correlated. Their coherent behaviour causes the emergence of effective degrees of freedom, requiring the application of non-perturbative techniques, such as chiral effective field theory. Here, we present measurements of the neutron's generalized spin-polarizabilities that quantify the neutron's spin precession under electromagnetic fields at very low energy-momentum transfer squared down to 0.035 GeV$^2$. In this regime, chiral effective field theory calculations are expected to be applicable. Light-sheet microscopy enables considerable speed and phototoxicity gains, while quantitative-phase imaging confers label-free recognition of cells and organelles, and quantifies their number-density that, thermodynamically, is more representative of metabolism than size. Here, we report the fusion of these two imaging modalities onto a standard inverted microscope that retains compatibility with microfluidics and open-source software for image acquisition and processing. An accelerating Airy-beam light-sheet critically enabled imaging areas that were greater by more than one order of magnitude than a Gaussian beam illumination and matched exactly those of quantitative-phase imaging. Using this integrative imaging system, we performed a demonstrative multivariate investigation of live-cells in microfluidics that unmasked that cellular noise can affect the compartmental localization of metabolic reactions. We detail the design, assembly, and performance of the integrative imaging system, and discuss potential applications in biotechnology and evolutionary biology.

Background: Plants harbor many trophic groups of nematodes in them. The plant production is also determined by the occurrence of nematodes adjacent to the rhizosphere of plants, such as parasitic, free-living etc. Methods: Altogether 60 samples (30 from 20 cm depth and 30 from 30 cm depth) were examined from the Kiwi plants to detect the nematode distribution in Kiwi plants in ICIMOD, Godawari, Lalitpur, Nepal. Overall, 1510 individuals of 10 nematodes genera (both free living/beneficial and parasitic) belonging to four orders were found to be associated with Kiwi plants, among them the highest report was of order Mononchida (44.04%) which was followed by Rhabditida (33.44%), Dorylaimida (17.88%) and Tylenchida (4.64%). The number of nematodes were higher at 20 cm depth than at 30 cm depth. Iotonchus spp. was highest (32.68%) and the number of Helichotylenchus spp. (3.93%) and Mesorhabditis spp. (3.92) were lowest at 20 cm depth. Similarly, Iotonchus spp. was highest (20.13%) and the number of Discolaimus spp. (4.03%) was lowest at 30 cm depth.

Background: Antibiotics are commonly used in animal husbandry for medicinal, prophylactic and growth promotion purposes. Method: The prevalence of three groups of antibiotics; namely Gentamycin, Streptomycin and sulfonamides was studied in fresh milk available in five different study areas; namely Budhanilkantha, Dharmasthali, Tokha, Nepaltar and Gaushala from Kathmandu Valley of Nepal. The milk samples (n=100) were collected from local dairy collection centers of Kathmandu valley. Qualitative and semi-quantitative analysis with rapid screening kits revealed that 6% (n=6) samples were positive for antibiotic residues in the fresh milk for gentamycin, streptomycin and sulfonamide groups. Total 5% milk samples contained unidentified antibiotics residues. Sulfonamides residues were found to be highest 3% (n=3) followed by gentamycin 2% (n=2) and streptomycin 1% (n=1) with no any milk sample with multiple residues. Total 2 samples were positive in Dharmasthali, 2 in Gaushala, 1 in Nepaltar, 1 in Tokha and no positive samples from Budhanilkantha area. There was no statistical significant difference between study areas and commonly used antibiotics residues in milk samples (P is greater than 0.05). Conclusion: The antibiotic residues were found above the maximum residue limits that presented serious threat to consumer health and raised a serious concern regarding the implementation and monitoring of international regulations.

A beam-normal single-spin asymmetry generated in the scattering of transversely polarized electrons from unpolarized nucleons is an observable related to the imaginary part of the two-photon exchange process. We report a 2% precision measurement of the beam-normal single-spin asymmetry in elastic electron-proton scattering with a mean scattering angle of theta_lab = 7.9 degrees and a mean energy of 1.149 GeV. The asymmetry result is B_n = -5.194 +- 0.067 (stat) +- 0.082 (syst) ppm. The spin-structure functions $g_1$ and $g_2$, and the spin-dependent partial cross-section $\sigma_\mathrm{TT}$ have been extracted from the polarized cross-sections differences, $\Delta \sigma_{\parallel}\hspace{-0.06cm}\left(\nu,Q^{2}\right)$ and $\Delta \sigma_{\perp}\hspace{-0.06cm}\left(\nu,Q^{2}\right)$ measured for the $\vec{^\textrm{3}\textrm{He}}(\vec{\textrm{e}},\textrm{e}')\textrm{X}$ reaction, in the E97-110 experiment at Jefferson Lab. Polarized electrons with energies from 1.147 to 4.404 GeV were scattered at angles of 6$^{\circ}$ and 9$^{\circ}$ from a longitudinally or transversely polarized $^{3}$He target. The data cover the kinematic regions of the quasi-elastic, resonance production and beyond. From the extracted spin-structure functions, the first moments $\overline{\Gamma_1}\hspace{-0.06cm}\left(Q^{2}\right)$, $\Gamma_2\hspace{-0.06cm}\left(Q^{2}\right)$ and $I_{\mathrm{TT}}\hspace{-0.06cm}\left(Q^{2}\right)$ are evaluated with high precision for the neutron in the $Q^2$ range from 0.035 to 0.24~GeV$^{2}$. These measurements were carried out to improve knowledge of the proton electromagnetic form factor ratio R=μpGEp∕GMp at large values of Q2 and to search for effects beyond the Born approximation in polarization transfer observables at Q2=2.5GeV2. The final results of both experiments were reported in a recent archival publication. A full reanalysis of the data from both experiments was carried out in order to reduce the systematic and, for the GEp-2γ experiment, statistical uncertainties. This technical note provides additional details of the final analysis omitted from the main publication, including the final evaluation of the systematic uncertainties.

Nepal has an appreciative history on conservation of rhino with remarkable proportions of Asian rhino population protected and managed within the protected areas. Poaching risk map promises early warning and a way to target preventive action, which can safeguard both human and ecosystem. This study was designed to identify and map risk areas of rhino (Rhinoceros unicornis) poaching within and around eastern sector of Chitwan National Park (CNP). A multi criterion GIS method was used to analyze and derive the risky zone. A binary logistic regression and expert consultation were done to finalize variables and risk rating, then weighted sum index model was performed in ArcGIS to derive risk zonation map. Presence/poaching and pseudo absence data were dependent variables and distance to guard post, settlement and road network from poaching events, land cover, slope and elevation were predictor variables for logistic regression model. Poaching events were observed to be spatially distributed around the park except in the south part. Harvesting of mature forest crop, regenerating the harvested area and tending of young crop are major silvicultural operations in forest management. Silvicultural system provides a framework for carrying out these silvicultural operations in a systematic manner. Yield is the annual amount of produce that can be realized sustainably. Similarly, thinning is the principal tending operation carried out in immature crop, is more often complicated in natural forest. This paper attempts to review silvicultural system applied in Terai region of Nepal under scientific forest management in order to regulate yield and to suggest new methods of thinning in natural forests. The study was conducted in Buddha Shanti Collaborative Forest in Nawalparasi district, western Terai Nepal. Data related to implementation status of CFM Plan were collected through field observation. Harvesting data were collected during harvesting operation. The collected harvesting data were analyzed for yield regulation. Irregular shelter-wood system was applied in Buddha Shanti CFM. The applied method of yield regulation is combination of area and stem control. Area control is applied for regeneration period. Annual harvest is controlled by number of stems. Tree harvesting area is fixed for ten years and annual harvesting amount is fixed by number of tree. In this method annual numbers of trees for harvesting are fixed. Total enumeration method was applied for thinning to find out the most frequent size of pole in selected sub compartment. During the enumeration process; distance between the every stem and fore bearing was also recorded with the help of laser distance meter and compass. A stem map was prepared with the help of Arc-GIS and the grid distance was fixed based on the most frequent size of stem. Then, one stem from each grid was selected for retaining after harvesting and remaining stems were cut down. This method provides appropriate guideline for implementing it objectively by reducing subjective judgment and provide more systematic method for thinning. The field practices of application of silvicultural system and yield regulation were found in accordance to the scientific forest management Procedures 2014 in Buddha Shanti CFM. Light-sheet fluorescence microscopy has greatly improved the speed and overall photostability of optically sectioning cellular and multi-cellular specimens. Similar gains have also been conferred by light-sheet Raman imaging; these schemes, however, rely on diffraction limited Gaussian beams that hinder the uniformity and size of the imaging field-of-view, and, as such, the resulting throughput rates. Here, we demonstrate that a digitally scanned Airy beam increases the Raman imaging throughput rates by more than an order of magnitude than conventional diffraction-limited beams. Overall, this, spectrometer-less, approach enabled 3D imaging of microparticles with high contrast and 1 µm axial resolution at 300 msec integration times per plane and orders of magnitude lower irradiation density than coherent Raman imaging schemes. We detail the apparatus and its performance, as well as its compatibility with fluorescence light-sheet and quantitative-phase imaging towards rapid and low phototoxicity multimodal imaging.

Measurements of elastic electron scattering data within the past decade have highlighted two-photon exchange contributions as a necessary ingredient in theoretical calculations to precisely evaluate hydrogen elastic scattering cross sections. This correction can modify the cross section at the few percent level. In contrast, dispersive effects can cause significantly larger changes from the Born approximation. The purpose of this experiment is to extract the carbon-12 elastic cross section around the first diffraction minimum, where the Born term contributions to the cross section are small to maximize the sensitivity to dispersive effects. The analysis uses the LEDEX data from the high resolution Jefferson Lab Hall A spectrometers to extract the cross sections near the first diffraction minimum of $$^{12}$$ C at beam energies of 362 MeV and 685 MeV. The results are in very good agreement with previous world data, although with less precision. The average deviation from a static nuclear charge distribution expected from linear and quadratic fits indicate a 30.6% contribution of dispersive effects to the cross section at 1 GeV. The magnitude of the dispersive effects near the first diffraction minimum of $$^{12}$$ C has been confirmed to be large with a strong energy dependence and could account for a large fraction of the magnitude for the observed quenching of the longitudinal nuclear response. These effects could also be important for nuclei radii extracted from parity-violating asymmetries measured near a diffraction minimum.

The three-dimensional structure of nucleons (protons and neutrons) is embedded in so-called generalized parton distributions, which are accessible from deeply virtual Compton scattering. In this process, a high energy electron is scattered off a nucleon by exchanging a virtual photon. Then, a highly-energetic real photon is emitted from one of the quarks inside the nucleon, which carries information on the quark's transverse position and longitudinal momentum. By measuring the cross-section of deeply virtual Compton scattering, Compton form factors related to the generalized parton distributions can be extracted. Here, we report the observation of unpolarized deeply virtual Compton scattering off a deuterium target. From the measured photon-electroproduction cross-sections, we have extracted the cross-section of a quasi-free neutron and a coherent deuteron. Due to the approximate isospin symmetry of quantum chromodynamics, we can determine the contributions from the different quark flavours to the helicity-conserved Compton form factors by combining our measurements with previous ones probing the proton's internal structure. These results advance our understanding of the description of the nucleon structure, which is important to solve the proton spin puzzle.

Short Range Correlations (SRCs) have been identified as being responsible for the high momentum tail of the nucleon momentum distribution, n(k). Hard, short-range interactions of nucleon pairs generate the high momentum tail and imprint a universal character on n(k) for all nuclei at large momentum. Triple coincidence experiments have shown a strong dominance of np pairs, but these measurements involve large final state interactions. This paper presents the results from Jefferson Lab experiment E08014 which measured inclusive electron scattering cross-section from Ca isotopes. The fields of particle and nuclear physics have undertaken extensive programs to search for evidence of physics beyond that explained by current theories. The observation of the Higgs boson at the Large Hadron Collider completed the set of particles predicted by the Standard Model (SM), currently the best description of fundamental particles and forces. However, the theory's limitations include a failure to predict fundamental parameters and the inability to account for dark matter/energy, gravity, and the matter-antimater asymmetry in the universe, among other phenomena. Given the lack of additional particles found so far through direct searches in the post-Higgs era, indirect searches utilizing precise measurements of well predicted SM observables allow highly targeted alternative tests for physics beyond the SM. Indirect searches have the potential to reach mass/energy scales beyond those directly accessible by today's high-energy accelerators. The value of the weak charge of the proton Q_W^p is an example of such an indirect search, as it sets the strength of the proton's interaction with particles via the well-predicted neutral electroweak force. Parity violation (invariance under spatial inversion (x,y,z) -> (-x,-y,-z)) is violated only in the weak interaction, thus providing a unique tool to isolate the weak interaction in order to measure the proton's weak charge. Here we report Q_W^p=0.0719+-0.0045, as extracted from our measured parity-violating (PV) polarized electron-proton scattering asymmetry, A_ep=-226.5+-9.3 ppb. Our value of Q_W^p is in excellent agreement with the SM, and sets multi-TeV-scale constraints on any semi-leptonic PV physics not described within the SM., Direct link to Nature Version "https://rdcu.be/954U"

International audience; We report on new p(e,e′p)π∘p(e,e′p)π∘ measurements at the Δ+(1232)Δ+(1232) resonance at the low momentum transfer region, where the mesonic cloud dynamics is predicted to be dominant and rapidly changing, offering a test bed for chiral effective field theory calculations. The new data explore the Q2Q2 dependence of the resonant quadrupole amplitudes and for the first time indicate that the Electric and the Coulomb quadrupole amplitudes converge as Q2→0Q2→0. The measurements of the Coulomb quadrupole amplitude have been extended to the lowest momentum transfer ever reached, and suggest that more than half of its magnitude is attributed to the mesonic cloud in this region. The new data disagree with predictions of constituent quark models and are in reasonable agreement with dynamical calculations that include pion cloud effects, chiral effective field theory and lattice calculations. The measurements indicate that improvement is required to the theoretical calculations and provide valuable input that will allow their refinements.

We report on a precise measurement of double-polarization asymmetries in electron-induced breakup of He3 proceeding to pd and ppn final states, performed in quasi-elastic kinematics at Q2=0.25(GeV/c)2 for missing momenta up to 250MeV/c. These observables represent highly sensitive tools to investigate the electromagnetic and spin structure of He3 and the relative importance of two- and three-body effects involved in the breakup reaction dynamics. The measured asymmetries cannot be satisfactorily reproduced by state-of-the-art calculations of He3 unless their three-body segment is adjusted, indicating that the spin-dependent part of the nuclear interaction governing the three-body breakup process is much smaller than previously thought.
