The COVID-19 pandemic drastically affected higher education and higher education students around the world, but few studies of college students' experiences during the COVID-19 pandemic have been conducted in Latin America. This study describes the COVID-19-related experiences and perspectives of Peruvian college students. We surveyed 3,427 full-time college students (average age: 23 years) attending a multi-campus Peruvian university in fall 2020. Participants were recruited through the digital platform of the learning management system at their university, email, and social media. We asked participants how they were managing risks related to COVID-19; the continuity of social, educational, and work activities; and the psychological and economic impacts of the pandemic on their lives. Since March 2020, 73.0% of participants reported COVID-19-related symptoms, but only 33.9% were tested for COVID-19. During the national quarantine imposed by the Peruvian government (March 15-June 30, 2020), 64.3% of participants remained in their house. Furthermore, while 44.0% of participants were working in February 2020 (95% CI: [41.7%, 46.4%]), only 23.6% (95% CI: [21.7%, 25.7%]) were working immediately after the pandemic began (i.e., at the end of April 2020). Participants were more stressed about the health and educational implications of COVID-19 for Peruvian society and their families than about themselves. The public health, economic, and educational implications of COVID-19 on college students are continuing to unfold. This study informed Peruvian higher education institutions' continued response to the COVID-19 pandemic, the progressive return to postpandemic activities, as well as other future pandemics and other crises.

Abstract During pregnancy the immune system needs to maintain immune tolerance of the foetus while also responding to infection, which can cause premature activation of the inflammatory pathways leading to the onset of labour and preterm birth. The vaginal microbiome is an important modifier of preterm birth risk, with Lactobacillus dominance during pregnancy associated with term delivery while high microbial diversity is associated with an increased risk of preterm birth. Glycans on glycoproteins along the lower female reproductive tract are fundamental to microbiota-host interactions and the mediation of inflammatory responses. However, the specific glycan epitopes involved in these processes are not well understood. To address this, we conducted glycomic analyses of cervicovaginal fluid (CVF) from 36 pregnant women at high risk of preterm birth and 4 non-pregnant women. Our analysis of N- and O-glycans revealed a rich CVF glycome. While O-glycans were shown to be the main carriers of ABO blood group epitopes, the main features of N-glycans were the presence of abundant paucimannose and high mannose glycans, and a remarkable diversity of complex bi-, tri-, and tetra-antennary glycans decorated with fucose and sialic acid. We identified immuno-regulatory epitopes, such as Lewis antigens, and found that fucosylation was negatively correlated to pro-inflammatory factors, such as IL-1β, MMP-8, C3a and C5a, while glycans with only sialylated antennae were mainly positively correlated to those. Similarly, paucimannose glycans showed a positive correlation to pro-inflammatory factors. We revealed a high abundance of glycans which have previously been identified as hallmarks of cancer and viral glycosylation, such as Man8 and Man9 high mannose glycans. Although each pregnant woman had a unique glycomic profile, longitudinal studies showed that the main glycosylation features were consistent throughout pregnancy in women who delivered at term, whereas women who experienced extreme preterm birth exhibited sharp changes in the CVF glycome shortly before delivery. These findings shed light on the processes underlying the role of glycosylation in maintaining a healthy vaginal microbiome and associated host immune responses. In addition, these discoveries facilitate our understanding of the lower female reproductive tract which has broad implications for women’s health.

Purpose: End-user participation is often encouraged to promote the uptake of Digital Livestock Technologies (DLTs). However, managing participation during DLT development can be challenging. We explore how participation decisions can impact end-users' engagement and attitudes towards the process, before suggesting strategies for improved management of the participation process. Methodology: We explored the experiences of end-users (e.g. farmers and farm assessors) and other stakeholders (e.g. developers, researchers, industry) involved in the development and testing of DLTs on UK farms, using semi-structured, in-depth interviews (N = 31). Findings: Participation can help develop technologies that better align with users' needs, promote learning, and encourage feelings of ownership. However, participation can be a double-edged sword. Inadequate levels of involvement, management of stakeholder relationships and expectations, and available support can negatively impact end-users' engagement and attitudes. Practical implications: Our study highlights the importance of understanding how management decisions during the participatory development of DLTs can influence the engagement and attitudes of end-users towards the process. Theoretical implications: The study contributes to the participation literature in agriculture and demonstrates the importance of using a critical lens to avoid making normative assumptions that participation necessarily promotes uptake in a linear, uncomplicated fashion. Originality/Value: Participation is seen as key for technology adoption. However, the potential downsides of participation have received less attention in relation to the engagement of end-users in the process.

Abstract Background Endometrial cancer is a multifactorial disease with inflammatory, metabolic and potentially microbial cues involved in disease pathogenesis. The endometrial cancer microbiome has been poorly characterised so far and studies have often overestimated bacterial biomass due to lack of integration of appropriate contamination controls. There is also a scarcity of evidence on the functionality of microbial microenvironments in endometrial cancer. This work addresses that knowledge gap by interrogating the genuine, contamination-free microbial signatures in the female genital tract and rectum of women with endometrial cancer and the mechanistic role of microbiome on carcinogenic processes. Results Here we sampled different regions of the reproductive tract (vagina, cervix, endometrium, fallopian tubes and ovaries) and rectum of 61 patients (37 endometrial cancer; 24 benign controls). We performed 16S rRNA gene sequencing of the V1–V2 hypervariable regions and qPCR of the 16S rRNA gene to qualitatively and quantitatively assess microbial communities and used 3D benign and endometrial cancer organoids to evaluate the effect of microbial products of L. crispatus, which was found depleted in endometrial cancer patients following primary analysis, on endometrial cell proliferation and inflammation. We found that the upper genital tract of a subset of women with and without endometrial cancer harbour microbiota quantitatively and compositionally distinguishable from background contaminants. Endometrial cancer was associated with reduced cervicovaginal and rectal bacterial load together with depletion of Lactobacillus species relative abundance, including L. crispatus, increased bacterial diversity and enrichment of Porphyromonas, Prevotella, Peptoniphilus and Anaerococcus in the lower genital tract and endometrium. Treatment of benign and malignant endometrial organoids with L. crispatus conditioned media exerted an anti-proliferative effect at high concentrations but had minimal impact on cytokine and chemokine profiles. Conclusions Our findings provide evidence that the upper female reproductive tract of some women contains detectable levels of bacteria, the composition of which is associated with endometrial cancer. Whether this is a cause or consequence of cancer pathophysiology and what is the functional significance of this finding remain to be elucidated to guide future screening tools and microbiome-based therapeutics . Fault slip is a complex natural phenomenon involving multiple spatiotemporal scales from seconds to days to weeks. To understand the physical and chemical processes responsible for the full fault slip spectrum, a multidisciplinary approach is highly recommended. The Near Fault Observatories (NFOs) aim at providing high-precision and spatiotemporally dense multidisciplinary near-fault data, enabling the generation of new original observations and innovative scientific products. The Alto Tiberina Near Fault Observatory is a permanent monitoring infrastructure established around the Alto Tiberina fault (ATF), a 60 km long low-angle normal fault (mean dip 20°), located along a sector of the Northern Apennines (central Italy) undergoing an extension at a rate of about 3 mm yr−1. The presence of repeating earthquakes on the ATF and a steep gradient in crustal velocities measured across the ATF by GNSS stations suggest large and deep (5–12 km) portions of the ATF undergoing aseismic creep. Both laboratory and theoretical studies indicate that any given patch of a fault can creep, nucleate slow earthquakes, and host large earthquakes, as also documented in nature for certain ruptures (e.g., Iquique in 2014, Tōhoku in 2011, and Parkfield in 2004). Nonetheless, how a fault patch switches from one mode of slip to another, as well as the interaction between creep, slow slip, and regular earthquakes, is still poorly documented by near-field observation. With the strainmeter array along the Alto Tiberina fault system (STAR) project, we build a series of six geophysical observatory sites consisting of 80–160 m deep vertical boreholes instrumented with strainmeters and seismometers as well as meteorological and GNSS antennas and additional seismometers at the surface. By covering the portions of the ATF that exhibits repeated earthquakes at shallow depth (above 4 km) with these new observatory sites, we aim to collect unique open-access data to answer fundamental questions about the relationship between creep, slow slip, dynamic earthquake rupture, and tectonic faulting.

Abstract Overexpression of the longevity gene Klotho prolongs lifespan, while its knockout shortens lifespan and impairs cognition via perturbation of myelination and synapse formation. However, comprehensive analysis of Klotho knockout effects on mammalian brain transcriptomics is lacking. Here, we report that Klotho knockout alters the levels of aging- and cognition related mRNAs, long non-coding RNAs, microRNAs and tRNA fragments. These include altered neuronal and glial regulators in murine models of aging and Alzheimer’s disease and in human Alzheimer’s disease post-mortem brains. We further demonstrate interaction of the knockout-elevated tRNA fragments with the spliceosome, possibly affecting RNA processing. Last, we present cell type-specific short RNA-seq datasets from FACS-sorted neurons and microglia of live human brain tissue demonstrating in-depth cell-type association of Klotho knockout-perturbed microRNAs. Together, our findings reveal multiple RNA transcripts in both neurons and glia from murine and human brain that are perturbed in Klotho deficiency and are aging- and neurodegeneration-related.

Abstract Cities concentrate problems that affect human well-being and biodiversity. Exploring the link between mental health and biodiversity can inform more holistic public health and urban planning. Here we examined associations between bird and tree species diversity estimates from eBird community science datasets and national forest inventories with self-rated mental health metrics from the Canadian Community Health Survey. We linked data across 36 Canadian Metropolitan Areas from 2007-2022 at a postal code level. After controlling for covariates, we found that bird and tree species diversity were significantly positively related to good self-reported mental health. Living in a postal code with bird diversity one standard deviation higher than the mean increased reporting of good mental health by 6.64%. Postal codes with tree species richness one standard deviation more than the mean increased reporting of good mental health by 5.36%. Our results suggest that supporting healthy urban ecosystems may also benefit human well-being.

Abstract Understanding the dynamics of intracellular signaling pathways, such as ERK1/2 (ERK) and Akt1/2 (Akt), in the context of cell fate decisions is important for advancing our knowledge of cellular processes and diseases, particularly cancer. While previous studies have established associations between ERK and Akt activities and proliferative cell fate, the heterogeneity of single-cell responses adds complexity to this understanding. This study employed a data-driven approach to address this challenge, developing machine learning models trained on a dataset of growth factor-induced ERK and Akt activity time courses in single cells, to predict cell division events. The most predictive models were developed by applying discrete wavelet transforms (DWTs) to extract low-frequency features from the time courses, followed by using Ensemble Integration, a data integration and predictive modeling framework. The results demonstrated that these models effectively predicted cell division events in MCF10A cells (F-measure=0.524, AUC=0.726). ERK dynamics were found to be more predictive than Akt, but the combination of both measurements further enhanced predictive performance. The ERK model`s performance also generalized to predicting division events in RPE cells, indicating the potential applicability of these models and our data-driven methodology for predicting cell division across different biological contexts. Interpretation of these models suggested that ERK dynamics throughout the cell cycle, rather than immediately after growth factor stimulation, were associated with the likelihood of cell division. Overall, this work contributes insights into the predictive power of intra-cellular signaling dynamics for cell fate decisions, and highlights the potential of machine learning approaches in unraveling complex cellular behaviors.

Arctic biodiversity is under threat from both climate induced environmental change and anthropogenic activity. However, the rapid rate of change and the challenging conditions for studying Arctic environments mean that many research questions must be answered before we can strategically allocate resources for management. Addressing threats to biodiversity in the Arctic is further complicated by the region's complex geopolitics, as eight countries claim jurisdiction over the area, with multiple local considerations such as Indigenous sovereignty and resource rights. Here, we identify research priorities to serve as a starting point for addressing the most pressing threats to Arctic biodiversity. We began by collecting pressing research questions about Arctic biodiversity, thematizing them as either threats or actions, and then categorizing them further into 18 groups. Then, drawing on cross-disciplinary and global expertise of professionals in Arctic science, management, and policy, we considered the barriers to answering these questions and proposed potential solutions that could be implemented if barriers were overcome. Overall, our horizon scan provides an expert assessment of threats (e.g., species’ responses to climate change) and actions (e.g., a lack of fundamental information regarding Arctic biodiversity) needing attention and is intended to guide future conservation action within the Arctic.

Abstract Maintaining and restoring ecological connectivity will be key in helping to prevent and reverse the loss of biodiversity. Fortunately, a growing body of research conducted over the last few decades has advanced our understanding of connectivity science, which will help inform evidence‐based connectivity conservation actions. Increases in data availability and computing capacity have helped to dramatically increase our ability to model functional connectivity using more sophisticated models. Keeping track of these advances can be difficult, even for connectivity scientists and practitioners. In this article, we highlight some key advances from the past decade and outline many of the remaining challenges. We describe the efforts to increase the biological realism of connectivity models by, for example, isolating movement behaviors, population parameters, directional movements, and the effects of climate change. We also discuss considerations of when to model connectivity for focal or multiple species. Finally, we reflect on how to account for uncertainty and increase the transparency and reproducibility of connectivity research and discuss situations where decisions may require forgoing sophistication for more simple approaches.

Two species of the genus Agriotypus Curtis, 1832, A. jilinensis Chao, 1981 and A. wangpiensis sp. nov., are added to the South Korean fauna. In total, four species, A. gracilis, A. jilinensis, A. silvestris, and A. wangpiensis sp. nov., have now been found in South Korea. The female of A. jilinensis is described for the first time and the species is redescribed based on the first fully eclosed specimens. Photographs of collecting sites, the larva and the adult of A. wangpiensis sp. nov., and a key to the South Korean species of Agriotypus are provided.

Much attention in recent years has been focused on making biodiversity data open and accessible to researchers. Yet ensuring the availability of these data is only the first step in preventing data waste. Here, we argue that researchers need to do a better job of using available datasets. We recommend that researchers search for existing data sources to serve their needs first, that they work to integrate multiple data sources when one alone will not suffice, and that they aim to explore research topics that will directly inform conservation action. We provide a roadmap with resources and examples to help guide conservation researchers towards better data-use practices. The vast quantities of biodiversity data, coupled with advanced techniques for using and integrating datasets, will play a key role in determining how to halt biodiversity declines. Making data open and accessible is only the start; we must be sure that we are using that existing data to conduct further research and inform decisions.

Abstract Mucin protein glycosylation is important in determining biological properties of mucus gels, which form protective barriers at mucosal surfaces of the body such as the intestine. Ecological factors including: age, sex, and diet can change mucus barrier properties by modulating mucin glycosylation. However, as our understanding stems from controlled laboratory studies in house mice, the combined influence of ecological factors on mucin glycosylation in real-world contexts remains limited. In this study, we used histological staining with ‘Alcian Blue, Periodic Acid, Schiff’s’ and ‘High-Iron diamine’ to assess the acidic nature of mucins stored within goblet cells of the intestine, in a wild mouse population (Mus musculus). Using statistical models, we identified sex as among the most influential ecological factors determining the acidity of intestinal mucin glycans in wild mice. Our data from wild mice and experiments using laboratory mice suggest estrogen signalling associates with an increase in the relative abundance of sialylated mucins. Thus, estrogen signalling may underpin sex differences observed in the colonic mucus of wild and laboratory mice. These findings highlight the significant influence of ecological parameters on mucosal barrier sites and the complementary role of wild populations in augmenting standard laboratory studies in the advancement of mucus biology.

Large dam removal is being used to restore river systems, but questions remain regarding their outcomes. We examine how the removal of two large dams in the Elwha River, coupled with hatchery production and fishing closures, affected population attributes of Chinook salmon (Oncorhynchus tshawytscha) and steelhead (O. mykiss). Initial responses by returning adult Chinook salmon and steelhead was an increase in the number and spatial extent of natural and hatchery origin fish. Although few naturally produced juvenile Chinook salmon and steelhead outmigrants were observed prior to and during dam removal, abundances increased three years after adult fish passage was restored, suggesting that impacts due to downstream sedimentation after dam removal were reduced. The Chinook salmon population demographics remain dominated by hatchery production, while increases in winter steelhead abundance included both hatchery and natural-origin spawners. The spatial expansion of winter steelhead upstream of former dam sites was predominantly by natural-origin spawners. We also observed a natural “reawakening” of summer steelhead that were in part derived from an up-river resident population that returned to the Upper Elwha. Our results showed that a combination of habitat, hatchery, and harvest actions can result in positive responses for salmonid populations.

Plastics are extensively used in agriculture, but their weathering and degradation generates microplastics (MPs) that can be carried by runoff into water bodies where they can accumulate and impact wildlife. Due to its physicochemical properties, biochar has shown promise in mitigating contaminants in agricultural runoff. However, few studies have examined its effectiveness at removing MPs. In this study, we assessed MP pollution (>30 μm) in runoff from a farm in the Mississippi Delta and examined the effectiveness of biochar (pinewood and sugarcane) to remove MPs from aqueous solutions. Using micro-Fourier Transform Infrared spectroscopy (µ-FTIR), we observed an average of 237 MPs/L (range 27–609) in the runoff, with most particles identified as polyethylene, polyamide, polyvinyl chloride, polyurethane, acrylonitrile butadiene styrene, and polyarylamide. Biochar columns effectively removed MPs from runoff samples with reductions ranging from 86.6% to 92.6%. MPs of different sizes, shapes, and types were stained with Nile red dye (to facilitate observation by fluorescence) and quantified their downward progress with multiple column volumes of water and wet/dry cycles. Smaller MPs penetrated the columns further, but ≥90% of MPs were retained in the ∼20 cm columns regardless of their shape, size, and type. We attribute these results to physical entrapment, hydrophobic behaviors, and electrostatic interactions. Overall, this proof-of-concept work suggests biochar may serve as a cost-effective approach to remove MPs from runoff, and that subsequent field studies are warranted.

Canada’s provinces and territories govern species at risk across most of Canada, with the federal Species at Risk Act generally covering only aquatic species, migratory birds, and species living on federal land. More than a decade after a 2012 report by the environmental law charity Ecojustice on species at risk protection in Canada, we use the same criteria to evaluate the current state of provincial and territorial species at risk legislation, and we provide updates on changes in each jurisdiction since 2012. These criteria are as follows: whether at-risk species are being identified, whether these species are being protected, whether their habitat is being protected, and whether species recovery plans are being created and implemented. We find that there is considerable variation across jurisdictions, with shortcomings that result in inadequate protections for at-risk species, as well as strong components that should be adopted by all jurisdictions. We recommend seven key areas for improvement: dedicated and harmonized legislation, limited discretionary power, increased embrace of scientific and Indigenous knowledge, appropriate timelines for actions, reasonable exemptions to protections, habitat protection across land ownership types, and transparency throughout the process. We urge policymakers to address current shortcomings as they work toward meeting Canada’s biodiversity conservation commitments.

Abstract Counter‐wildlife crime (CWC) interventions—those that directly protect target wildlife from illegal harvest/persecution, detect and sanction rule‐breakers, and interdict and control illegal wildlife commodities—are widely applied to address biodiversity loss. This systematic map provides an overview of the literature on the effectiveness of CWC interventions for conserving African, Asian and Latin American wildlife directly threatened by exploitation, including human–wildlife conflicts that trigger poaching. Following our systematic map protocol (Rytwinski, Öckerman, et al., 2021), we compiled peer‐reviewed and grey literature and screened articles using pre‐defined inclusion criteria. Included studies were coded for key variables of interest, from which we produced a searchable database, interactive map and structured heatmaps. A total of 530 studies from 477 articles were included in the systematic map. Most studies were from Africa and Asia (81% of studies) and focused on African and Asian elephants (16%), felids (14%) and turtles and tortoises (11%). Most evaluations of CWC interventions targeted wildlife products (rather than species) and the transfer of those products along the wildlife crime continuum (40% of cases). Population/species outcomes were most commonly measured via indicators of threat reduction (65% of cases) and intermediate outcomes (25%). We identified knowledge clusters where studies investigated the links between (1) patrols and other preventative actions to increase detection and population abundance and (2) information analysis and sharing and wildlife crime/trade levels. However, the effectiveness of most interventions was not rigorously evaluated. Most investigations used post‐implementation monitoring only (e.g. lacking a comparator), and no experimental designs were found. We identified several key knowledge gaps including a paucity of studies by geography (Latin America), taxonomy (plants, birds and reptiles), interventions (non‐patrol‐based CWC interventions) and outcomes (biological and the combination of biological and human well‐being outcomes). Our map reveals an opportunity to improve the rigour and documentation of CWC intervention evaluations, which would enable the evidence‐based selection of effective approaches to improve wildlife conservation and national security.

Summary: Background: AI models have shown promise in performing many medical imaging tasks. However, our ability to explain what signals these models have learned is severely lacking. Explanations are needed in order to increase the trust of doctors in AI-based models, especially in domains where AI prediction capabilities surpass those of humans. Moreover, such explanations could enable novel scientific discovery by uncovering signals in the data that aren’t yet known to experts. Methods: In this paper, we present a workflow for generating hypotheses to understand which visual signals in images are correlated with a classification model’s predictions for a given task. This approach leverages an automatic visual explanation algorithm followed by interdisciplinary expert review. We propose the following 4 steps: (i) Train a classifier to perform a given task to assess whether the imagery indeed contains signals relevant to the task; (ii) Train a StyleGAN-based image generator with an architecture that enables guidance by the classifier (“StylEx”); (iii) Automatically detect, extract, and visualize the top visual attributes that the classifier is sensitive towards. For visualization, we independently modify each of these attributes to generate counterfactual visualizations for a set of images (i.e., what the image would look like with the attribute increased or decreased); (iv) Formulate hypotheses for the underlying mechanisms, to stimulate future research. Specifically, present the discovered attributes and corresponding counterfactual visualizations to an interdisciplinary panel of experts so that hypotheses can account for social and structural determinants of health (e.g., whether the attributes correspond to known patho-physiological or socio-cultural phenomena, or could be novel discoveries). Findings: To demonstrate the broad applicability of our approach, we present results on eight prediction tasks across three medical imaging modalities—retinal fundus photographs, external eye photographs, and chest radiographs. We showcase examples where many of the automatically-learned attributes clearly capture clinically known features (e.g., types of cataract, enlarged heart), and demonstrate automatically-learned confounders that arise from factors beyond physiological mechanisms (e.g., chest X-ray underexposure is correlated with the classifier predicting abnormality, and eye makeup is correlated with the classifier predicting low hemoglobin levels). We further show that our method reveals a number of physiologically plausible, previously-unknown attributes based on the literature (e.g., differences in the fundus associated with self-reported sex, which were previously unknown). Interpretation: Our approach enables hypotheses generation via attribute visualizations and has the potential to enable researchers to better understand, improve their assessment, and extract new knowledge from AI-based models, as well as debug and design better datasets. Though not designed to infer causality, importantly, we highlight that attributes generated by our framework can capture phenomena beyond physiology or pathophysiology, reflecting the real world nature of healthcare delivery and socio-cultural factors, and hence interdisciplinary perspectives are critical in these investigations. Finally, we will release code to help researchers train their own StylEx models and analyze their predictive tasks of interest, and use the methodology presented in this paper for responsible interpretation of the revealed attributes. Abstract The invasive emerald ash borer (Agrilus planipennis) causes damage to street trees which is estimated to reach US$ 900 million over the next 30 years. Although millions of dollars are spent annually to control this species, spatiotemporal management plans are often based on rules of thumb that ignore future pest dispersal. Here, we reveal an optimal management strategy to protect urban trees in North America from A. planipennis. To achieve this, we embedded a pest dispersal model within a mixed integer programming framework. We discovered that optimized strategies consistently outperformed those based on rules of thumb, potentially resulting in the protection of an additional nearly 1 million street trees and savings of $ 629 million. Critically, the best management strategies always relied on quarantines and biological control (constituting 98–99% and 1–2% of the project budget, respectively), in contrast with current practices, where federal spending has been diverted to biological control. Our findings serve to inform future pest control efforts and can help protect many more trees from this invasive species.

Abstract A lack of composable and tunable gene regulators has hindered efforts to engineer non-model bacteria and consortia. Toward addressing this, we explore the broad-host potential of small transcription activating RNA (STAR) and propose a design strategy to achieve tunable gene control. First, we demonstrate that STARs optimized for E. coli function across different Gram-negative species and can actuate using phage RNA polymerase, suggesting that RNA systems acting at the level of transcription are portable. Second, we explore an RNA design strategy that uses arrays of tandem and transcriptionally fused RNA regulators to precisely alter regulator concentration from 1 to 8 copies. This provides a simple means to predictably tune output gain across species and does not require access to large regulatory part libraries. Finally, we show RNA arrays can be used to achieve tunable cascading and multiplexing circuits across species, analogous to the motifs used in artificial neural networks.

The NASA Aerosol Cloud meTeorology Interactions oVer the western ATlantic Experiment (ACTIVATE) produced a unique dataset for research into aerosol–cloud–meteorology interactions, with applications extending from process-based studies to multi-scale model intercomparison and improvement as well as to remote-sensing algorithm assessments and advancements. ACTIVATE used two NASA Langley Research Center aircraft, a HU-25 Falcon and King Air, to conduct systematic and spatially coordinated flights over the northwest Atlantic Ocean, resulting in 162 joint flights and 17 other single-aircraft flights between 2020 and 2022 across all seasons. Data cover 574 and 592 cumulative flights hours for the HU-25 Falcon and King Air, respectively. The HU-25 Falcon conducted profiling at different level legs below, in, and just above boundary layer clouds (< 3 km) and obtained in situ measurements of trace gases, aerosol particles, clouds, and atmospheric state parameters. Under cloud-free conditions, the HU-25 Falcon similarly conducted profiling at different level legs within and immediately above the boundary layer. The King Air (the high-flying aircraft) flew at approximately ∼ 9 km and conducted remote sensing with a lidar and polarimeter while also launching dropsondes (785 in total). Collectively, simultaneous data from both aircraft help to characterize the same vertical column of the atmosphere. In addition to individual instrument files, data from the HU-25 Falcon aircraft are combined into “merge files” on the publicly available data archive that are created at different time resolutions of interest (e.g., 1, 5, 10, 15, 30, 60 s, or matching an individual data product's start and stop times). This paper describes the ACTIVATE flight strategy, instrument and complementary dataset products, data access and usage details, and data application notes. Abstract Conservation planning requires extensive amounts of data, yet data collection is expensive, and there is often a trade‐off between the quantity and quality of data that can be collected. Researchers are increasingly turning to community science programs to meet their biodiversity data needs, yet the reliability of such data sources is still a common source of debate. Here, we argue that professionally collected data are subject to many of the limitations and biases present in community science datasets. We explore four common criticisms of community science data, and comparable issues that exist in data collected by experts: spatial biases, observer variability, taxonomic biases and the misapplication of data. We then outline solutions to these problems that have been developed to make better use of community science data, but can (and should) be equally applied to both kinds of data. We highlight four main solutions based on research using community science data that can be applied across all biodiversity data collection and research. Statistical techniques that have been developed for processing community science data can equally help account for spatial biases and observer variation in professional datasets. Benchmarking or vetting one dataset against another can strengthen evidence and uncover unknown sources of biases. Professional and community science datasets can be used together to fill knowledge gaps that are unique to each. Careful study design that accounts for the collection of relevant and important covariate data can help statistically account for sources of bias. Currently, a double standard exists in how researchers view data collected by professionals versus those collected by community scientists. Our aim is to ensure that valuable community science data are given the prominent place they deserve, and that data collected by experts are appropriately vetted and biases accounted for using all the tools at our disposal.

Recent studies have suggested that high levels of social support can encourage better health behaviours and result in improved cardiovascular health. In this study we evaluated the association between social support and ideal cardiovascular health among urban Jamaicans. We conducted a cross-sectional study among urban residents in Jamaica's south-east health region. Socio-demographic data and information on cigarette smoking, physical activity, dietary practices, blood pressure, body size, cholesterol, and glucose, were collected by trained personnel. The outcome variable, ideal cardiovascular health, was defined as having optimal levels of ≥5 of these characteristics (ICH-5) according to the American Heart Association definitions. Social support exposure variables included number of friends (network size), number of friends willing to provide loans (instrumental support) and number of friends providing advice (informational support). Principal component analysis was used to create a social support score using these three variables. Survey-weighted logistic regression models were used to evaluate the association between ICH-5 and social support score. Analyses included 841 participants (279 males, 562 females) with mean age of 47.6 ± 18.42 years. ICH-5 prevalence was 26.6% (95%CI 22.3, 31.0) with no significant sex difference (male 27.5%, female 25.7%). In sex-specific, multivariable logistic regression models, social support score, was inversely associated with ICH-5 among males (OR 0.67 [95%CI 0.51, 0.89], p = 0.006) but directly associated among females (OR 1.26 [95%CI 1.04, 1.53], p = 0.020) after adjusting for age and community SES. Living in poorer communities was also significantly associated with higher odds of ICH-5 among males, while living communities with high property value was associated with higher odds of ICH among females. In this study, higher level of social support was associated with better cardiovascular health among women, but poorer cardiovascular health among men in urban Jamaica. Further research should explore these associations and identify appropriate interventions to promote cardiovascular health.

Objective: Non-alcoholic fatty liver disease (NAFLD) involves hepatic accumulation of intracellular lipid droplets via incompletely understood processes. Here, we report distinct and cooperative NAFLD roles of LysTTT-5′tRF transfer RNA fragments and microRNA miR-194-5p. Methods: Combined use of diet induced obese mice with human-derived oleic acid-exposed Hep G2 cells revealed new NAFLD roles of LysTTT-5′tRF and miR-194-5p. Results: Unlike lean animals, dietary-induced NAFLD mice showed concurrent hepatic decrease of both LysTTT-5′tRF and miR-194-5p levels, which were restored following miR-132 antisense oligonucleotide treatment which suppresses hepatic steatosis. Moreover, exposing human-derived Hep G2 cells to oleic acid for 7 days co-suppressed miR-194-5p and LysTTT-5′tRF levels while increasing lipid accumulation. Inversely, transfecting fattened cells with a synthetic LysTTT-5′tRF mimic elevated mRNA levels of the metabolic regulator β-Klotho while decreasing triglyceride amounts by 30% within 24 h. In contradistinction, antisense suppression of miR-194-5p induced accumulation of its novel target, the NAFLD-implicated lipid droplet-coating PLIN2 protein. Further, two out of 15 steatosis-alleviating screened drug-repurposing compounds, Danazol and Latanoprost, elevated miR-194-5p or LysTTT-5′tRF levels. Conclusion: Our findings highlight the different yet complementary roles of miR-194-5p and LysTTT-5′tRF and offer new insights into the complex roles of small non-coding RNAs and the multiple pathways involved in NAFLD pathogenesis.

Abstract Recently it has become possible to de novo design high affinity protein binding proteins from target structural information alone. There is, however, considerable room for improvement as the overall design success rate is low. Here, we explore the augmentation of energy-based protein binder design using deep learning. We find that using AlphaFold2 or RoseTTAFold to assess the probability that a designed sequence adopts the designed monomer structure, and the probability that this structure binds the target as designed, increases design success rates nearly 10-fold. We find further that sequence design using ProteinMPNN rather than Rosetta considerably increases computational efficiency.

Abstract Animals select habitats based on food, water, space, and cover. Each of those components are essential to the ability of an individual to survive and reproduce in a particular habitat. Selection of resources is linked to reproductive fitness and individuals likely vary in how they select resources relative to their reproductive state: during pregnancy, while provisioning young when nutritional needs of the mother are high, but offspring are vulnerable to predation, or if they lose young to mortality. We investigated the effects of reproductive state on selection of resources by maternal female desert bighorn sheep (Ovis canadensis nelsoni) by comparing selection during the last trimester of gestation, following parturition when females were provisioning dependent young, and if the female lost an offspring. We captured, and recaptured each year, 32 female bighorn sheep at Lone Mountain, Nevada, during 2016–2018. Captured females were fit with GPS collars and those that were pregnant received vaginal implant transmitters. We used a Bayesian approach to estimate differences in selection between females provisioning and not provisioning offspring, as well as the length of time it took for females with offspring to return levels of selection similar to that observed prior to parturition. Females that were not provisioning offspring selected areas with higher risk of predation, but greater nutritional resources than those that were provisioning dependent young. When females were provisioning young immediately following parturition, females selected areas that were safe from predators, but had lower nutritional resources. Females displayed varying rates of return to selection strategies associated with access to nutritional resources as young grew and became more agile and less dependent on mothers. We observed clear and substantial shifts in selection of resources associated with reproductive state, and females exhibited tradeoffs in favor of areas that were safer from predators when provisioning dependent young despite loss of nutritional resources to support lactation. As young grew and became less vulnerable to predators, females returned to levels of selection that provided access to nutritional resources to restore somatic reserves lost during lactation.

Public education programs on the dangers of high salt intake, and population level interventions to reduce the salt content in foods are possible strategies to address this problem. In Jamaica, there are limited data on the levels of salt consumption and the population’s knowledge and practices with regards to salt consumption. This study therefore aims to obtain baseline data on salt consumption, salt content in foods sold in restaurants, and evaluate knowledge, attitudes, and practices of Jamaicans regarding salt consumption. Methods The study is divided into four components. Component 1 will be a secondary analysis of data on urinary sodium from spot urine samples collected as part of a national survey, the Jamaica Health and Lifestyle Survey 2016-2017. Component 2 will be a survey of chain and non-chain restaurants in Jamaica, to estimate the sodium content of foods sold in restaurants. Component 3 is another national survey, this time on a sample 1,200 individuals to obtain data on knowledge, attitudes and practices regarding salt consumption and estimation of urinary sodium excretion. Component 4 is a validation study to assess the level of agreement between spot urine sodium estimates and 24-hour urinary sodium from 120 individuals from Component 3. Discussion This study will provide important baseline data on salt consumption in Jamaica and will fulfil the first components of the World Health Organization SHAKE Technical Package for Salt Reduction. The findings will serve as a guide to Jamaica’s Ministry of Health and Wellness in the development of a national salt reduction program. Findings will also inform interventions to promote individual and population level sodium reduction strategies as the country seeks to achieve the national target of a 30% reduction in salt consumption by 2025.

Chinook salmon (Oncorhynchus tshawytscha) populations express diverse early life history pathways that increase habitat utilization and demographic resiliency. Extensive anthropogenic alterations to freshwater habitats along with hatchery and harvest impacts have led to marked reductions in early life history diversity across much of the species’ range. The recent removal of two Elwha River dams between 2011 and 2014 restored access to over 90% of the available habitat that had been inaccessible to Chinook salmon since the early 1900s. This provided an opportunity to investigate how renewed access to this habitat might affect life history diversity. As exotherms, egg-to-fry development, juvenile growth, and movement are influenced by water temperatures. We used spatially and temporally explicit Elwha River water temperature and Chinook salmon spawning location data, in conjunction with spawn timing, emergence, growth, and movement models, to predict observed timing and sizes of juvenile Chinook salmon captured in three rotary screw traps in the mainstem and two tributaries during four trap years. This effort allowed us to test hypotheses regarding Elwha River Chinook salmon early life history, identify potential problems with the data, and predict how emergence and growth would change with increased spawning in the upper watershed. Predicted Chinook salmon emergence timing and predicted dates that juveniles reached 65 mm differed by as much as 2 months for different river locations due to large differences in thermal regimes longitudinally in the mainstem and between tributaries. For 10 out of the 12 trap–year combinations, the model was able to replicate important characteristics of the out-migrant timing and length data collected at the three traps. However, in most cases, there were many plausible parameter combinations that performed well, and in some cases, the model predictions and observations differed. Potential problems with the data and model assumptions were identified as partial explanations for differences and provide avenues for future work. We show that juvenile out-migrant data combined with mechanistic models can improve our understanding of how differences in temperature, spawning extent, and spawn timing affect the emergence, growth, and movement of juvenile fish across diverse riverine habitats.

Summary: Background: While vaccines have established utility against COVID-19, phase 3 efficacy studies have generally not comprehensively evaluated protection provided by previous infection or hybrid immunity (previous infection plus vaccination). Individual patient data from US government-supported harmonized vaccine trials provide an unprecedented sample population to address this issue. We characterized the protective efficacy of previous SARS-CoV-2 infection and hybrid immunity against COVID-19 early in the pandemic over three-to six-month follow-up and compared with vaccine-associated protection. Methods: In this post-hoc cross-protocol analysis of the Moderna, AstraZeneca, Janssen, and Novavax COVID-19 vaccine clinical trials, we allocated participants into four groups based on previous-infection status at enrolment and treatment: no previous infection/placebo; previous infection/placebo; no previous infection/vaccine; and previous infection/vaccine. The main outcome was RT-PCR-confirmed COVID-19 >7–15 days (per original protocols) after final study injection. We calculated crude and adjusted efficacy measures. Findings: Previous infection/placebo participants had a 92% decreased risk of future COVID-19 compared to no previous infection/placebo participants (overall hazard ratio [HR] ratio: 0.08; 95% CI: 0.05–0.13). Among single-dose Janssen participants, hybrid immunity conferred greater protection than vaccine alone (HR: 0.03; 95% CI: 0.01–0.10). Too few infections were observed to draw statistical inferences comparing hybrid immunity to vaccine alone for other trials. Vaccination, previous infection, and hybrid immunity all provided near-complete protection against severe disease. Interpretation: Previous infection, any hybrid immunity, and two-dose vaccination all provided substantial protection against symptomatic and severe COVID-19 through the early Delta period. Thus, as a surrogate for natural infection, vaccination remains the safest approach to protection. Funding: National Institutes of Health.

Translational control is critical for cell fate transitions during development, lineage specification, and tumorigenesis. Here, we show that the transcription factor double homeobox protein 4 (DUX4), and its previously characterized transcriptional program, broadly regulates translation to change the cellular proteome. DUX4 is a key regulator of zygotic genome activation in human embryos, whereas misexpression of DUX4 causes facioscapulohumeral muscular dystrophy (FSHD) and is associated with MHC-I suppression and immune evasion in cancer. We report that translation initiation and elongation factors are disrupted downstream of DUX4 expression in human myoblasts. Genome-wide translation profiling identified mRNAs susceptible to DUX4-induced translation inhibition, including those encoding antigen presentation factors and muscle lineage proteins, while DUX4-induced mRNAs were robustly translated. Endogenous expression of DUX4 in human FSHD myotubes and cancer cell lines also correlated with reduced protein synthesis and MHC-I presentation. Our findings reveal that DUX4 orchestrates cell state conversion by suppressing the cellular proteome while maintaining translation of DUX4-induced mRNAs to promote an early developmental program.

Summary: The transcription factor DUX4 regulates a portion of the zygotic gene activation (ZGA) program in the early embryo. Many cancers express DUX4 but it is unknown whether this generates cells similar to early embryonic stem cells. Here we identified cancer cell lines that express DUX4 and showed that DUX4 is transiently expressed in a small subset of the cells. DUX4 expression activates the DUX4-regulated ZGA transcriptional program, the subsequent 8C-like program, and markers of early embryonic lineages, while suppressing steady-state and interferon-induced MHC class I expression. Although DUX4 was expressed in a small number of cells under standard culture conditions, DNA damage or changes in growth conditions increased the fraction of cells expressing DUX4 and its downstream programs. Our demonstration that transient expression of endogenous DUX4 in cancer cells induces a metastable early embryonic stem cell program and suppresses antigen presentation has implications for cancer growth, progression, and immune evasion.

The NASA North Atlantic Aerosols and Marine Ecosystems Study (NAAMES) ship and aircraft field campaign deployed to the western subarctic Atlantic between the years 2015 and 2018. One of the primary goals of NAAMES is to improve the understanding of aerosol–cloud interaction (ACI) over the Atlantic Ocean under different seasonal regimes. ACIs currently represent the largest source of uncertainty in global climate models. During three NAAMES field campaigns (NAAMES-1 in November 2015, NAAMES-2 in May 2016, and NAAMES-3 in September 2017), multiple 10 h science flights were conducted using the NASA C-130 aircraft to measure marine boundary layer aerosol and cloud properties. The standard flight pattern includes vertical spirals where the C-130 transitioned from high altitude to low altitude (and vice versa), collecting in situ measurements of aerosols, trace gases, clouds, and meteorological parameters as a function of altitude. We examine the data collected from 37 spirals during the three NAAMES field campaigns, and we present a comprehensive characterization of the vertical profiles of aerosol properties under different synoptic conditions and aerosol regimes. The vertical distribution of submicron aerosol particles exhibited strong seasonal variation, as well as elevated intra-seasonal variability depending on emission sources and aerosol processes in the atmospheric column. Pristine marine conditions and new particle formation were prevalent in the wintertime (NAAMES-1) due to low biogenic emissions from the surface ocean and reduced continental influence. Higher concentrations of submicron aerosol particles were observed in the spring (NAAMES-2) due to strong phytoplankton activity and the arrival of long-range-transported continental plumes in the free troposphere with subsequent entrainment into the marine boundary layer. Biomass burning from boreal wildfires was the main source of aerosol particles in the region during the late summer (NAAMES-3) in both the marine boundary layer and free troposphere.

Fires emit sufficient sulfur to affect local and regional air quality and climate. This study analyzes SO2 emission factors and variability in smoke plumes from US wildfires and agricultural fires, as well as their relationship to sulfate and hydroxymethanesulfonate (HMS) formation. Observed SO2 emission factors for various fuel types show good agreement with the latest reviews of biomass burning emission factors, producing an emission factor range of 0.47–1.2 g SO2 kg−1 C. These emission factors vary with geographic location in a way that suggests that deposition of coal burning emissions and application of sulfur-containing fertilizers likely play a role in the larger observed values, which are primarily associated with agricultural burning. A 0-D box model generally reproduces the observed trends of SO2 and total sulfate (inorganic + organic) in aging wildfire plumes. In many cases, modeled HMS is consistent with the observed organosulfur concentrations. However, a comparison of observed organosulfur and modeled HMS suggests that multiple organosulfur compounds are likely responsible for the observations but that the chemistry of these compounds yields similar production and loss rates as that of HMS, resulting in good agreement with the modeled results. We provide suggestions for constraining the organosulfur compounds observed during these flights, and we show that the chemistry of HMS can allow organosulfur to act as an S(IV) reservoir under conditions of pH > 6 and liquid water content >10−7 g sm−3. This can facilitate long-range transport of sulfur emissions, resulting in increased SO2 and eventually sulfate in transported smoke.

Data are presented from intercomparisons between two research aircraft, the FAAM BAe-146 and the NASA Lockheed P3, and between the BAe-146 and the surface-based DOE (Department of Energy) ARM (Atmospheric Radiation Measurement) Mobile Facility at Ascension Island (8∘ S, 14.5∘ W; a remote island in the mid-Atlantic). These took place from 17 August to 5 September 2017, during the African biomass burning (BB) season. The primary motivation was to give confidence in the use of data from multiple platforms with which to evaluate numerical climate models. The three platforms were involved in the CLouds–Aerosol–Radiation Interaction and Forcing for Year 2017 (CLARIFY-2017), ObseRvations of Aerosols above CLouds and their intEractionS (ORACLES), and Layered Atlantic Smoke and Interactions with Clouds (LASIC) field experiments. Comparisons from flight segments on 6 d where the BAe-146 flew alongside the ARM facility on Ascension Island are presented, along with comparisons from the wing-tip-to-wing-tip flight of the P3 and BAe-146 on 18 August 2017. The intercomparison flight sampled a relatively clean atmosphere overlying a moderately polluted boundary layer, while the six fly-bys of the ARM site sampled both clean and polluted conditions 2–4 km upwind. We compare and validate characterisations of aerosol physical, chemical and optical properties as well as atmospheric radiation and cloud microphysics between platforms. We assess the performance of measurement instrumentation in the field, under conditions where sampling conditions are not as tightly controlled as in laboratory measurements where calibrations are performed. Solar radiation measurements compared well enough to permit radiative closure studies. Optical absorption coefficient measurements from all three platforms were within uncertainty limits, although absolute magnitudes were too low ( Mm−1) to fully support a comparison of the absorption Ångström exponents. Aerosol optical absorption measurements from airborne platforms were more comparable than aircraft-to-ground observations. Scattering coefficient observations compared adequately between airborne platforms, but agreement with ground-based measurements was worse, potentially caused by small differences in sampling conditions or actual aerosol population differences over land. Chemical composition measurements followed a similar pattern, with better comparisons between the airborne platforms. Thermodynamics, aerosol and cloud microphysical properties generally agreed given uncertainties.

Abstract Human cervicovaginal fluid (CVF) is a complex, functionally important and glycan rich biological fluid, fundamental in mediating physiological events associated with reproductive health. Using a comprehensive glycomic strategy we reveal an extremely rich and complex N-glycome in CVF of pregnant and non-pregnant women, abundant in paucimannose and high mannose glycans, complex glycans with 2–4 N-Acetyllactosamine (LacNAc) antennae, and Poly-LacNAc glycans decorated with fucosylation and sialylation. N-glycosylation profiles were observed to differ in relation to pregnancy status, microbial composition, immune activation, and pregnancy outcome. Compared to CVF from women experiencing term birth, CVF from women who subsequently experienced preterm birth showed lower sialylation, which correlated to the presence of a diverse microbiome, and higher fucosylation, which correlated positively to pro-inflammatory cytokine concentration. This study is the first step towards better understanding the role of cervicovaginal glycans in reproductive health, their contribution to the mechanism of microbial driven preterm birth, and their potential for preventative therapy.

Abstract Natural environments provide a myriad of health benefits, yet the role of species diversity within these spaces remains underexplored. Bird diversity may yield mental health benefits for humans, through birdsong or feelings of connection to nature. In an initial effort to establish whether bird diversity may be linked with human health in a US context and to test the consistency in such trends from year to year, we combine widely available community (aka citizen) science data (eBird) estimating bird diversity across the state of Michigan with anxiety/mood disorder hospitalisation records (2008–18). We found a negative, significant association between bird species diversity and anxiety/mood disorder hospitalisations (β = −0.36, 95% CI = −0.69 to −0.04). The relationship between bird diversity and hospitalisations found at this scale is significant, given the potential for biodiversity to affect severe mental health outcomes. Thus, these initial findings should be further explored in studies with finer resolution of exposure to bird species and longitudinal or experimental designs that account for other demographic characteristics, risk factors and other neighbourhood features. If future studies confirm these findings, there are important implications for urban greening efforts, some of which are explicitly focused on increasing bird habitat.

IntroductionPreterm birth is one of the major causes of neonatal morbidity and mortality across the world. Both term and preterm labour are preceded by inflammatory activation in uterine tissues. This includes increased leukocyte infiltration, and subsequent increase in chemokine and cytokine levels, activation of pro-inflammatory transcription factors as NF-κB and increased prostaglandin synthesis. Prostaglandin F2α (PGF2α) is one of the myometrial activators and stimulators.MethodsHere we investigated the role of PGF2α in pro-inflammatory signalling pathways in human myometrial cells isolated from term non-labouring uterine tissue. Primary myometrial cells were treated with G protein inhibitors, calcium chelators and/or PGF2α. Nuclear extracts were analysed by TranSignal cAMP/Calcium Protein/DNA Array. Whole cell protein lysates were analysed by Western blotting. mRNA levels of target genes were analysed by RT-PCR.ResultsThe results show that PGF2α increases inflammation in myometrial cells through increased activation of NF-κB and MAP kinases and increased expression of COX-2. PGF2α was found to activate several calcium/cAMP-dependent transcription factors, such as CREB and C/EBP-β. mRNA levels of NF-κB-regulated cytokines and chemokines were also elevated with PGF2α stimulation. We have shown that the increase in PGF2α-mediated COX-2 expression in myometrial cells requires coupling of the FP receptor to both Gαq and Gαi proteins. Additionally, PGF2α-induced calcium response was also mediated through Gαq and Gαi coupling.DiscussionIn summary, our findings suggest that PGF2α-induced inflammation in myometrial cells involves activation of several transcription factors – NF-κB, MAP kinases, CREB and C/EBP-β. Our results indicate that the FP receptor signals via Gαq and Gαi coupling in myometrium. This work provides insight into PGF2α pro-inflammatory signalling in term myometrium prior to the onset of labour and suggests that PGF2α signalling pathways could be a potential target for management of preterm labour.

Human stressors block, eliminate, and simplify habitat mosaics, eroding landscapes’ life history diversity and thus biological resilience. One goal of restoration is to alleviate human stressors that suppress life history diversity, but life history responses to these efforts are still coming into focus. Sicklefin (Macrhybopsis meeki) and sturgeon chub (M. gelida) historically occurred throughout the Missouri River (MR), in some tributaries, and Mississippi River downstream of the MR. They have been species of U.S. state-level conservation concern and U.S. Endangered Species Act listing candidates since the 1990s. We applied analytical approaches from occupancy modeling to correlation to monitoring data spanning 23 years to assess relationships between occupancy and time, space, environmental factors, habitat, and other species. Sicklefin chub occupancy appeared higher in the early to mid-2000s and mid-to-late 2010s. A potential decline in occupancy occurred for sturgeon chub in the mid-to-late 2010s. Spatially, chub occupancy was depressed for 159 to 438 km downstream of MR dams. Among macrohabitats, inside bends had relatively high occupancy for both species; secondary connected channels had relatively high values for sturgeon chub. Co-occurrence was likely between sicklefin and sturgeon chub and between chubs and shovelnose sturgeon (Scaphirhybchus platorybchus) and channel catfish (Ictalurus punctatus). The observed co-occurrence of chubs and pallid sturgeon (Scaphirhynchus albus; PS) was potentially higher than expected for adult PS. For juvenile PS, co-occurrence was lower than expected in the Lower MR and potentially higher than expected in the Upper MR, warranting future research. Results from this research suggest management for the improvement of sicklefin and sturgeon chub populations may benefit other MR fish populations.

Abstract Ecosurveillance has proliferated in recent years, generating vast amounts of data on the natural environment. Ecosurveillance also has significant potential impacts on humans; therefore, researchers and policymakers need new conceptual tools to anticipate and mitigate any negative effects. Surveillance studies is an interdisciplinary field in the social sciences, providing a number of insights and practical lessons for predicting and managing the complex impacts (positive and negative, intended and unintended) of surveillance tools and practices. We draw on surveillance studies literature to propose two tools to guide designers and practitioners of ecosurveillance—a ‘red flag checklist’ to anticipate potential problems, and a ‘considerations guide’ to inform design decisions across a wide range of ecosurveillance systems. These tools will help ensure that the coming era of ecosurveillance is guided by responsible and ethical practices towards wildlife and humans alike, while also realizing the potential of these technologies for improving environmental outcomes. BackgroundMenstrual cups (MCs) are increasingly used to collect cervicovaginal secretions to characterise vaginal mucosal immunology, in conjunction with high vaginal swabs (HVS) for metataxonomics, particularly in HIV transmission studies. We hypothesised that both methods of collecting bacterial biomass are equivalent for 16S rRNA gene sequencing.Material and MethodsCervicovaginal fluid (CVF) samples from 16 pregnant women with HIV-1 (PWWH) were included to represent the major vaginal bacterial community state types (CST I-V). Women underwent sampling during the second trimester by liquid amies HVS followed by a MC (Soft disc™) and samples were stored at -80°C. Bacterial cell pellets obtained from swab elution and MC (500 µL, 1 in 10 dilution) were resuspended in 120 µL PBS for DNA extraction. Bacterial 16S rRNA gene sequencing was performed using V1-V2 primers and were analysed using MOTHUR. Paired total DNA, bacterial load, amplicon read counts, diversity matrices and bacterial taxa were compared by sampling method using MicrobiomeAnalyst, SPSS and R.ResultsThe total DNA eluted from one aliquot of diluted CVF from an MC was similar to that of a HVS (993ng and 609ng, p=0.18); the mean bacterial loads were also comparable for both methods (MC: 8.0 log10 16S rRNA gene copies versus HVS: 7.9 log10 16S rRNA gene copies, p=0.27). The mean number of sequence reads generated from MC samples was lower than from HVS (MC: 12730; HVS:14830, p=0.05). The α-diversity metrices were similar for both techniques; MC Species Observed: 41 (range 12-96) versus HVS: 47 (range 16-96), p=0.15; MC Inverse Simpson Index: 1.98 (range 1.0-4.0) versus HVS: 0.48 (range 1.0-4.4), p=0.22). The three most abundant species observed were: Lactobacillus iners, Lactobacillus crispatus and Gardnerella vaginalis. Hierarchical clustering of relative abundance data showed that samples obtained using different techniques in an individual clustered in the same CST group.ConclusionThese data demonstrate that despite sampling slightly different areas of the lower genital tract, there was no difference in bacterial load or composition between methods. Both are suitable for characterisation of vaginal microbiota in PWWH. The MC offers advantages, including a higher volume of sample available for DNA extraction and complimentary assays.

Animal welfare standards are used within the food industry to demonstrate efforts in reaching higher welfare on farms. To verify compliance with those standards, inspectors conduct regular on-farm animal welfare assessments. Conducting these welfare assessments can, however, be time-consuming and prone to human bias. The emergence of Digital Livestock Technologies (DLTs) offers new ways of monitoring farm animal welfare and can alleviate some of the challenges related to animal welfare assessments by collecting data automatically and more frequently. Whilst automating welfare assessments with DLTs may be promising, little attention has been paid to farmers’ perceptions of the challenges that could prevent successful implementation. This study aims to address this gap by focusing on the trial of a DLT (a 3D machinelearning camera) to automate mobility and body condition scoring on 11 dairy cattle farms. Semi-structured, in-depth interviews were conducted with farmers, technology developers and a stakeholder involved in a farm assurance scheme (N14). Findings suggest that stakeholders perceived important benefits to the use of the camera in this context, from building consumer trust by increasing transparency to improved management efficiency. There was also a potential for greater consistency in data collection and thus for enhanced fairness across the UK dairy sector, particularly on the issue of lameness prevalence. However, stakeholders also raised important concerns, such as a lack of clarity around data ownership, reliability, and use, and the possibility of some farmers being penalised (e.g., if the technology failed to work). More clarity should thus be given to farmers in relation to data governance and evidence provided in terms of technical performance and accuracy. The findings of this study highlighted the need for more inclusive approaches to ensure farmers’ concerns are adequately identified and addressed. These approaches can help minimise negative consequences to farmers and animal welfare, whilst maximising the potential benefits of automating welfare-related data collection.

DUX4 activates the first wave of zygotic gene expression in the early embryo. Mis-expression of DUX4 in skeletal muscle causes facioscapulohumeral dystrophy (FSHD), whereas expression in cancers suppresses IFNγ induction of major histocompatibility complex class I (MHC class I) and contributes to immune evasion. We show that the DUX4 protein interacts with STAT1 and broadly suppresses expression of IFNγ-stimulated genes by decreasing bound STAT1 and Pol-II recruitment. Transcriptional suppression of interferon-stimulated genes (ISGs) requires conserved (L)LxxL(L) motifs in the carboxyterminal region of DUX4 and phosphorylation of STAT1 Y701 enhances interaction with DUX4. Consistent with these findings, expression of endogenous DUX4 in FSHD muscle cells and the CIC-DUX4 fusion containing the DUX4 CTD in a sarcoma cell line inhibit IFNγ induction of ISGs. Mouse Dux similarly interacted with STAT1 and suppressed IFNγ induction of ISGs. These findings identify an evolved role of the DUXC family in modulating immune signaling pathways with implications for development, cancers, and FSHD.

Purpose: To describe primary ocular toxoplasmosis infection related to ingestion of undercooked venison. Observations: This single site, retrospective case series reviewed 4 patients with primary ocular toxoplasmosis that was acquired by ingesting undercooked venison. De-identified data was collected regarding baseline patient characteristics including age, sex, past medical and ocular history, onset of symptoms, visual acuity (VA), response to treatment, and workup. All patients with acquired toxoplasmosis had similar chronology of systemic and ocular symptoms. Exposure occurred in October or November and systemic symptoms developed within 2 weeks, followed by ocular symptoms an average of 2.6 months later. Average age at onset was 56 ± 13 (age ± SD) years old and all were male. Average initial and final VA were 20/50 and 20/50, respectively. Positive anti-toxoplasma IgM and IgG serologies were found in all cases. All patients were treated with trimethoprim/sulfamethoxazole and achieved rapid improvement. Complications occurred in 50% of cases and included epiretinal membrane, cystoid macular edema, vitreoretinal traction, and neovascularization. Conclusions and importance: Consumption of undercooked venison is a source of primary ocular toxoplasmosis even in immunocompetent hosts and has a clear chronology. A presentation of retinochoroiditis during the winter months should prompt questioning for exposure to wild game.
