Abstract

Nanomaterials have become integral in various aspects of agricultural practices, including the development of nano-fertilizers for optimized crop nutrition. This study explores the application of green-synthesized iron (Fe) and molybdenum (Mo) nanoparticles, as well as their composites, using a guava leaf extract (GLE). The focus is on assessing their impact on nitrogen fixation and growth in tomato plants (Solanum lycopersicum). The nanoparticles were characterized through Fourier Transform Infrared Spectroscopy, Ultraviolet Diffused Reflectance Spectroscopy, Raman Spectroscopy, and X-ray diffraction analysis. The experiment involved two application methods (soil and direct plant spraying) with varying nanoparticle concentrations. Results indicate that the 1% composite nanoparticles applied to the soil and 3% Mo directly on plants yield the most favorable growth and nitrogen uptake in S. lycopersicum. Notably, the 1% composite treatment demonstrated significant enhancement in shoot length, number of branches, and shoot diameter at all three growth stages. Conversely, the 3% Mo treatment when applied directly to plants exhibited optimal results showing substantial shoot length, number of branches, and shoot diameter. Post-experimental soil nutrient analysis further revealed the nuanced effects of nanoparticle applications with 1% composite treatments enhancing nutrient availability compared to control and other concentrations. This research contributes to the evolving field of agri-nanotechnology emphasizing the importance of nanoparticle concentration and application method in influencing plant development and nutrient uptake, paving the way for sustainable agricultural practices.

34. Changes in the prevalence of hepatitis B and C viral infections in Sindh province, Pakistan: Findings from two sero‐surveys in 2007 and 2019.

Abstract

Pakistan harbours a large burden of hepatitis B virus (HBV) and hepatitis C virus (HCV) infection. We utilised repeat sero‐surveys to assess progress achieved towards hepatitis elimination in Pakistan. Multilevel logistic regression evaluated the change in HBV infection (HBV surface antigen (HBsAg)‐positive) prevalence and HCV exposure (HCV antibody (HCV‐Ab)‐positive) prevalence between two sero‐surveys from 2007 and 2019 for Sindh province and associated risk factors. Adjusted odds ratios (aORs) were estimated and population‐attributable fractions (PAF) for modifiable risk factors for HCV exposure. The 2007 and 2019 surveys included 8855 and 6672 individuals. HBsAg prevalence decreased from 2.6% (95% confidence intervals (95% CI): 2.2–2.9) in 2007 to 1.1% (95% CI: 0.8–1.3) in 2019, while HCV‐Ab prevalence increased from 5.1% (95% CI: 4.6%–5.5%) to 6.2% (95% CI: 5.6%–6.8%). The age and gender‐adjusted HBsAg prevalence decreased by 80% (aOR = 0.2, 95% CI: 0.1–0.4) among children and 60% (aOR = 0.4, 95% CI: 0.3–0.6) among adults over 2007–2019, while HCV‐Ab prevalence decreased by 60% (aOR = 0.4, 95%CI:0.2–0.7) in children and increased by 40% (aOR = 1.4, 95% CI: 1.2–1.7) in adults. HCV‐Ab prevalence was lower in adults with secondary (aOR = 0.6, 95% CI: 0.5–0.8) and higher (aOR = 0.5, 95%CI:0.3–0.8) education compared to illiterates and higher among adults reporting blood transfusion (aOR = 1.7, 95% CI: 1.2–2.4), family history of hepatitis (aOR = 2.5, 95% CI: 1.9–3.3), past year medical injection (aOR = 2.1, 95% CI: 1.6–2.7), being tattooed (aOR = 1.4, 95% CI: 1.0–1.9) and shaved by traditional barber (aOR = 1.2, 95% CI: 1.0–1.5). Modifiable risk factors accounted for 45% of HCV exposure, with medical injection(s) accounting for 38% (95%CI,25.7–48.4%). Abstract

Background: Zea mays (maize) is a globally significant cereal crop with diverse applications in food, feed, and industrial products. However, maize cultivation is often challenged by environmental stressors such as heavy metal toxicity and drought stress (DS). Heavy metals like cadmium (Cd) and lead (Pb) can accumulate in soil through industrial activities and improper waste disposal, posing significant threats to plant growth and development. Drought stress further exacerbates these challenges by limiting water availability and affecting physiological processes in plants. This study explores the impact of Cd and Pb toxicity, as well as DS, on maize growth and development, and investigates the potential mitigating effects of various treatments, including gibberellic acid (GA3), biochar (BC), rhizobacteria (RB), and their combinations. Methods: The experiment involved maize plants subjected to different stress conditions: cadmium (Cd) at concentrations of 0, 6, and 12 ppm, lead (Pb) at 0 and 400 ppm, and drought stress (DS). Treatments included the application of 10 ppm GA3, 0.75% BC, a combined treatment of 10 ppm GA3 and 0.75% BC, rhizobacteria (RB), and a combined treatment of 0.5% BC and RB. The study measured germination rates, shoot and root lengths, and biochemical parameters such as shoot and root protein, phenolics, and chlorophyll contents under these conditions. Results: In the absence of Cd stress (0 Cd), the application of 10 ppm GA3 and 0.75% BC significantly enhanced germination rates by 72% and 76%, respectively, compared to the control, with the combined treatment exhibiting the highest enhancement of 86%. Under Cd stress (6 ppm Cd), GA3 and BC individually improved germination by 54% and 57%, respectively, with the combined treatment showing the largest increase of 63%. Drought stress influenced germination, with notable improvements observed with the application of 0.5% BC (50% increase) and RB (49% increase). Similar trends were observed in shoot and root lengths, where the combined treatment of GA3 and BC resulted in the most significant improvements. The treatments positively influenced shoot and root protein, phenolics, and chlorophyll contents, particularly under stress conditions. Conclusion: These findings highlight the potential of combined treatments, such as the application of GA3 and BC or BC with RB, in alleviating the adverse effects of heavy metals (Cd and Pb) and drought stress in maize cultivation. The combined treatments not only improved germination rates but also significantly enhanced shoot and root growth, as well as important biochemical parameters under stress conditions. This suggests that GA3 and BC, alone or in combination with RB, can play a crucial role in enhancing maize resilience to environmental stressors. The study highlights the importance of exploring sustainable agricultural practices to mitigate the impacts of heavy metal toxicity and drought stress. Future research should focus on long-term field trials to validate these findings and further investigate the mechanistic pathways involved in stress mitigation by these amendments, as well as their economic feasibility and environmental impact on a larger scale to ensure their practical applicability in real-world agricultural settings. 42. Enhanced Citric Acid Production through Aspergillus niger : Insights from Fermentation Studies Using Sugarcane Molasses.

Abstract

The production of citric acid, a vital agricultural commodity utilized across various industries such as food, beverages, pharmaceuticals, agriculture, detergents, and cosmetics, predominantly relies on microbial fermentation, with Aspergillus niger accounting for approximately 90% of global production. In this study, we aimed to optimize the key factors influencing citric acid production, with a focus on strains, fermentation techniques, and carbon sources, particularly sugarcane molasses. A. niger, sourced from the Botany department/Biotechnology laboratories at Govt. College of Science, Lahore, was employed for citric acid production. The process involved inoculum preparation through spore collection from 3 to 5 days of cultured PDA slants. The fermentation medium, comprising cane molasses with a 15% sugar concentration, was meticulously prepared and optimized for various factors, including magnesium sulfate, potassium ferrocyanide, time of addition of potassium ferrocyanide, ammonium oxalate, and calcium chloride. Our optimization results shed light on the significant impact of different factors on citric acid production. For instance, the addition of 0.4 g/L magnesium sulfate led to a maximum yield of 75%, while 2 g/L potassium ferrocyanide, added at 24 h, achieved a yield of 78%. Remarkably, ammonium oxalate, at a concentration of 10 g/L, resulted in a notable 77% yield. Conversely, the addition of calcium chloride exhibited negligible effects on citric acid production, with the control group yielding more at 78%. Our study underscores the potential for optimizing factors to enhance citric acid production by A. niger in submerged fermentation. These findings highlight the pivotal role of magnesium sulfate, potassium ferrocyanide, and ammonium oxalate in augmenting citric acid yields while emphasizing the minimal impact of calcium chloride. Ultimately, these insights contribute to advancing our understanding of microbial citric acid biosynthesis, providing valuable implications for industrial applications and future research endeavors. Abstract

The genus Trichoderma holds economic significance due to its widespread distribution and diverse applications, including biological control, enzyme production, and various biotechnological uses. The accurate identification of Trichoderma species is crucial given their close association with human activities. Despite previous efforts in classification, a comprehensive analysis combining morphological and molecular approaches is necessary. This study focuses on the isolation of four Trichoderma species from industrial wastewater in Pakistan, expanding on the known diversity in the region; isolation involved collecting samples from industrial wastewater effluents at specific sites in Punjab, Pakistan. Trichoderma strains were cultured and purified on solid media, with subsequent biomass production for bisorptional activity. Morphological characterization included colony features and microscopic examinations. DNA extraction, polymerase chain reaction (PCR), and sequencing of the internal transcribed spacer (ITS) region were conducted for molecular analysis. Phylogenetic analysis was performed using the Maximum Likelihood Algorithm. The study identified three Trichoderma species, viz. T. citrinoviride, T. erinaceum, and T. longibrachiatum. Each species was characterized morphologically and supported by molecular–phylogenetic analysis. Illustrations of microscopic features and a phylogenetic tree based on the ITS-nrDNA region were recorded. T. citrinoviride and T. longibrachiatum, isolated from steel mill and tanneries wastewater, respectively, were differentiated based on morphological characteristics such as phialides and conidia. The combination of morphological and molecular techniques enhances the accuracy of species identification. The study highlights the significance of Trichoderma in industrial wastewater environments and underscores the need for continued research in this area. Future research should focus on exploring the ecological roles and potential applications of the newly identified Trichoderma species. Additionally, further investigations into the biotechnological potential of these species, including enzyme production and bioremediation capabilities, would contribute to their practical applications. Abstract

Ad5 is a common cause of respiratory disease and an occasional cause of gastroenteritis and conjunctivitis, and seroconversion before adolescence is common in humans. To gain some insight into how Ad5 infection affects the immune system of rhesus macaques (RM) 18 RM were infected with a host-range mutant Ad5 (Ad5hr) by 3 mucosal inoculations. There was a delay of 2 to 6 weeks after the first inoculation before plasmacytoid dendritic cell (pDC) frequency and function increased in peripheral blood. Primary Ad5hr infection suppressed IFN-γ mRNA expression, but the second Ad5hr exposure induced a rapid increase in IFN-gamma mRNA in peripheral blood mononuclear cells (PBMC). Primary Ad5hr infection suppressed CCL20, TNF and IL-1 mRNA expression in PBMC, and subsequent virus exposures further dampened expression of these pro-inflammatory cytokines. Primary, but not secondary, Ad5hr inoculation increased the frequency of CXCR3+ CD4+ T cells in blood, while secondary, but not primary, Ad5hr infection transiently increased the frequencies of Ki67+, HLADR+ and CD95+/CCR5+ CD4+ T cells in blood. Ad5hr infection induced polyfunctional CD4 and CD8+ T cells specific for the Ad5 hexon protein in all of the animals. Thus, infection with Ad5hr induced a complex pattern of innate and adaptive immunity in RM that included transient systemic CD4+ T cell activation and suppressed innate immunity on re-exposure to the virus. The complex effects of adenovirus infection on the immune system may help to explain the unexpected results of testing Ad5 vector expressing HIV antigens in Ad5 seropositive people.
