Abstract

This communication will provide the latest information about the progress of the “Research Project for the Study of Georgian Grapes and Wine Culture”, managed by the National Wine Agency of Georgia since 2014. Local and foreign institutions continue to work together with the aim of stimulating multidisciplinary scientific research activity on Georgian viticulture and viniculture and to reconstruct their development from Neolithic civilizations to the present. The project is multidisciplinary in nature, merging contributions from archaeology, history, ethnography, molecular genetics, biomolecular archaeology, palaeobotany, ampelography, enology, climatology and other scientific fields.

Abstract

Major surgical societies advised using non-operative management of appendicitis and suggested against laparoscopy during the COVID-19 pandemic. The hypothesis is that a significant reduction in the number of emergent appendectomies was observed during the pandemic, restricted to complex cases. The study aimed to analyse emergent surgical appendectomies during pandemic on a national basis and compare it to the same period of the previous year. This is a multicentre, retrospective, observational study investigating the outcomes of patients undergoing emergent appendectomy in March–April 2019 vs March–April 2020. The primary outcome was the number of appendectomies performed, classified according to the American Association for the Surgery of Trauma (AAST) score. Secondary outcomes were the type of surgical technique employed (laparoscopic vs open) and the complication rates. One thousand five hundred forty one patients with acute appendicitis underwent surgery during the two study periods. 1337 (86.8%) patients met the inclusion criteria: 546 (40.8%) patients underwent surgery for acute appendicitis in 2020 and 791 (59.2%) in 2019. According to AAST, patients with complicated appendicitis operated in 2019 were 30.3% vs 39.9% in 2020 (p = 0.001). We observed an increase in the number of post-operative complications in 2020 (15.9%) compared to 2019 (9.6%) (p < 0.001). The following determinants increased the likelihood of complication occurrence: undergoing surgery during 2020 (+ 67%), the increase of a unit in the AAST score (+ 26%), surgery performed > 24 h after admission (+ 58%), open surgery (+ 112%) and conversion to open surgery (+ 166%). In Italian hospitals, in March and April 2020, the number of appendectomies has drastically dropped. During the first pandemic wave, patients undergoing surgery were more frequently affected by more severe appendicitis than the previous year's timeframe and experienced a higher number of complications. Abstract

Major surgical societies advised using non-operative management of appendicitis and suggested against laparoscopy during the COVID-19 pandemic. The hypothesis is that a significant reduction in the number of emergent appendectomies was observed during the pandemic, restricted to complex cases. The study aimed to analyse emergent surgical appendectomies during pandemic on a national basis and compare it to the same period of the previous year. This is a multicentre, retrospective, observational study investigating the outcomes of patients undergoing emergent appendectomy in March–April 2019 vs March–April 2020. The primary outcome was the number of appendectomies performed, classified according to the American Association for the Surgery of Trauma (AAST) score. Secondary outcomes were the type of surgical technique employed (laparoscopic vs open) and the complication rates. One thousand five hundred forty one patients with acute appendicitis underwent surgery during the two study periods. 1337 (86.8%) patients met the inclusion criteria: 546 (40.8%) patients underwent surgery for acute appendicitis in 2020 and 791 (59.2%) in 2019. According to AAST, patients with complicated appendicitis operated in 2019 were 30.3% vs 39.9% in 2020 (p = 0.001). We observed an increase in the number of post-operative complications in 2020 (15.9%) compared to 2019 (9.6%) (p 24 h after admission (+ 58%), open surgery (+ 112%) and conversion to open surgery (+ 166%). In Italian hospitals, in March and April 2020, the number of appendectomies has drastically dropped. During the first pandemic wave, patients undergoing surgery were more frequently affected by more severe appendicitis than the previous year's timeframe and experienced a higher number of complications. Abstract

Plants develop in a microbe-rich environment and must interact with a plethora of microorganisms, both pathogenic and beneficial. Indeed, such is the case of Pseudomonas, and its model organisms P. fluorescens and P. syringae, a bacterial genus that has received particular attention because of its beneficial effect on plants and its pathogenic strains. The present study aims to compare plant-beneficial and pathogenic strains belonging to the P. syringae species to get new insights into the distinction between the two types of plant-microbe interactions. In assays carried out under greenhouse conditions, P. syringae pv. syringae strain 260-02 was shown to promote plant-growth and to exert biocontrol of P. syringae pv. tomato strain DC3000, against the Botrytis cinerea fungus and the Cymbidium Ringspot Virus. This P. syringae strain also had a distinct volatile emission profile, as well as a different plant-colonization pattern, visualized by confocal microscopy and gfp labeled strains, compared to strain DC3000. Despite the different behavior, the P. syringae strain 260-02 showed great similarity to pathogenic strains at a genomic level. However, genome analyses highlighted a few differences that form the basis for the following hypotheses regarding strain 260-02. P. syringae strain 260-02: (i) possesses nonfunctional virulence genes, like the mangotoxin-producing operon Mbo; (ii) has different regulation pathways, suggested by the difference in the autoinducer system and the lack of a virulence activator gene; (iii) has genes encoding DNA methylases different from those found in other P. syringae strains, suggested by the presence of horizontal-gene-transfer-obtained methylases that could affect gene expression.

Abstract

Mgaloblishvili, a Vitis vinifera cultivar, exhibits unique resistance traits against Plasmopara viticola, the downy mildew agent. This offers the unique opportunity of exploring the molecular responses in compatible and incompatible plant-pathogen interaction. In this study, whole transcriptomes of Mgaloblishvili, Pinot noir (a V. vinifera susceptible cultivar), and Bianca (a resistant hybrid) leaves, inoculated and non-inoculated with the pathogen, were used to identify P. viticola effector-encoding genes and plant susceptibility/resistance genes. Multiple effector-encoding genes were identified in P. viticola transcriptome, with remarkable expression differences in relation to the inoculated grapevine cultivar. Intriguingly, five apoplastic effectors specifically associated with resistance in V. vinifera. Gene coexpression network analysis identified specific modules and metabolic changes occurring during infection in the three grapevine cultivars. Analysis of these data allowed, for the first time, the detection in V. vinifera of a putative P. viticola susceptibility gene, encoding a LOB domain-containing protein. Finally, the de novo assembly of Mgaloblishvili, Pinot noir, and Bianca transcriptomes and their comparison highlighted novel candidate genes that might be at the basis of the resistant phenotype. These results open the way to functional analysis studies and to new perspectives in molecular breeding of grapevine for resistance to P. viticola.

Between August 2011 and June 2013, wood samples from diseased vines showing dead spur and cordons, shoot dieback associated with sunken necrotic bark lesions and brown to black vascular streaking were collected from numerous diseased vineyards. Several fungal species were isolated from the margin between healthy and symptomatic tissue. Three species of Botryosphaeriaceae, namely, Diplodia seriata, Neofusicoccum australe, and N. vitifusiforme and one species of Diaporthaceae, namely, Diaporthe neotheicola, were observed to be associated with the decline of old vines. Other fungal species were recovered from diseased wood, namely, Alternaria alternata, Botryotinia fuckeliana (anamorph of Botrytis cinerea), Acremonium spp., Aspergillus spp., and Fusarium spp. In addition, Penicillium spp. inter- and intra-species diversity were assessed based on virtual RFLP gel analyses and identification of restriction enzymes able to distinguish fungi strains within species based on determination of single nucleotide polymorphism (SNP) lineages within cluster members based on the sequencing of the internal transcribed spacer (ITS1-5.8S-ITS2) region of the rDNA.

Entre agosto de 2011 y junio de 2013, se recolectaron muestras de madera de vid enfermas que mostraban espolones y cordones muertos, desvirones de tallos asociados con lesiones de corteza necrótica hundidas y rayas vasculares de color café a negro de numerosos viñedos enfermos. Se aislaron varias especies de hongos del margen entre tejido sano y sintomático. Tres especies de Botryosphaeriaceae, a saber, Diplodia seriata, Neofusicoccum australe, y N. vitifusiforme y una especie de Diaporthaceae, a saber, Diaporthe neotheicola, se observaron a ser asociado con la disminución de viñas viejas. Abstract

Phytoplasmas of the stolbur group infect a wide range of wild and cultivated plants in several areas worldwide. In Europe they are transmitted by polyphagous planthoppers of the Cixiidae family. Based on actual and in silico RFLP analyses of 16S rDNA nucleotide sequences subgroups have been described in the stolbur group. In vineyards, grapevine-affecting stolbur phytoplasmas are associated with bois noir (BN) disease and are transmitted by Hyalesthes obsoletus Signoret, but in wine-growing areas where H. obsoletus is absent, the presence of stolbur phytoplasmas could implies the existence of alternative vectors. Taxonomy of stolbur phytoplasmas on 16S rDNA gene is still poorly studied, and from virtual RFLP analyses there is growing evidence of a discrete amount of variability suggesting delineation of a number of subgroups. Selecting 16Sr DNA sequences from 26 stolbur phytoplasmas infecting diverse plant species such as grapevine, potatoes, corn, and rhododendron in different areas worldwide, and sharing 99% sequence identity, made it possible to distinguish at least 6 different clusters, and many subclusters, based upon nucleotide sequences within the ca. 1,240 bp; actual RFLP patterns confirmed the presence of nine new subgroups in grapevine plants with BN from Northern and Central Italy, Hungary, Serbia and Iran. Key enzymes for distinguishing among these subgroups were AluI, BfaI, BstUI, together with MboII, FauI, Tsp509I, and Hpy188I. However when compared to its closest relatives, stolbur phytoplasma 16Sr DNA is 97.6% identical to the 16Sr DNA of ‘Candidatus Phytoplasma australiense’, 96.5% and 95.3% identical to ‘Ca. P. graminis’ and ‘Ca. P. caricae’, respectively. Due to its 16S identity with ‘Ca. P. australiense’, the designation of stolbur phytoplasma as a ‘Candidatus’ species will be not possible without a comparative analysis of non ribosomal loci. Biological complexity of stolbur phytoplasmas, indicated by the existence of numerous herbaceous hosts and diverse insect vectors, has stimulated studies on molecular markers of stolbur phytoplasma strains. Characterization of stolbur phytoplasmas in Italian vineyards was performed by multilocus sequence analysis of tuf, hlyC, trxA-truB, cbiQ-glyA, and rplS-csdB genes based on PCR-RFLP assays. Tuf gene was selected since two tuf gene sequence variants of stolbur phytoplasmas were found consistently associated with different herbaceous hosts and natural ecologies in Italian vineyards. For each of the other genes it was possible to define two distinct SNP lineages: hlyC SNP genetic lineages were consistent with those identified on the basis of tuf gene sequences; SNP lineages of trxA-truB, cbiQ-glyA, and rplS-csdB were not consistent with tuf-hlyC SNP lineages. Several SNPs of tuf, hlyC, trxA-truB, cbiQ-glyA, and rplS-csdB genes were positioned within recognition sites of restriction enzymes, and were employed for multilocus sequence analyses that grouped the stolbur phytoplasma strains from grapevines in six SNP genetic lineages. Intriguingly, distribution patterns indicated a different prevalence of these SNP lineages in the geographic areas investigated. Furthermore, the finding that the host specificities of stolbur phytoplasma lineages delineated based on hlyC and tuf genes, points to their possible involvement in the interaction of phytoplasmas with specific hosts. Moreover genotyping of the SecY locus revealed 27 stolbur genotypes in the Euro-Mediterranean areas, that formed two major clusters which are congruent with the two tuf groups. The interaction between phytoplasma membrane proteins and host proteins may influence biological and ecological properties of phytoplasmas such as specificity of plants host and insect vectors or symptom induction in plants. Such phytoplasma proteins can be highly variable; therefore their study should present opportunities for further development of phytoplasma genomic markers. Abstract

"Bois noir" (BN) is a grapevine yellows disease, associated with phytoplasma strains related to 'Candidatus Phytoplasma solani', that causes severe losses to viticulture in the Euro-Mediterranean basin. Due to the complex ecological cycle of its etiological agent, BN epidemiology is only partially known, and no effective control strategies have been developed. Numerous studies have focused on molecular characterization of BN phytoplasma strains, to identify molecular markers useful to accurately describe their genetic diversity, geographic distribution and host range. In the present study, a multiple gene analysess were carried out on 16S rRNA, tuf, vmp1, and stamp genes to study the genetic variability among 18 BN phytoplasma strains detected in diverse regions of the Republic of Macedonia. Restriction fragment length polymorphism (RFLP) assays showed the presence of one 16S rRNA (16SrXII-A), two tuf (tuf-type a, tuf-type b), five vmp1 (V2-TA, V3, V4, V14, V18), and three stamp (S1, S2, S3) gene patterns among the examined strains. Based on the collective RFLP patterns, seven genotypes (Mac1 to Mac7) were described as evidence for genetic heterogeneity, and highlighting their prevalence and distribution in the investigated regions. Phylogenetic analyses on vmp1 and stamp genes underlined the affiliation of Macedonian BN phytoplasma strains to clusters associated with distinct ecologies.
