Among the approaches in three-dimensional (3D) single molecule localization microscopy, there are several point spread function (PSF) engineering approaches, in which depth information of molecules is encoded in 2D images. Usually,the molecules are excited sparsely in each raw image. The consequence is that the temporal resolution has to be sacrificed. In order to improve temporal resolution and ensure localization accuracy, we propose a method, SH-CS, based on light needle excitation, detection system with single helix-point spread function (SH-PSF), and compressed sensing (CS). Although the SH-CS method still has a limitation about the molecule density, it is suited for relatively dense molecules. For each light needle scanning position, an SH image of excited molecules is processed with CS algorithm to decode their axial information. Simulations demonstrated, for random distributed 1 ~ 15 molecules in depth range of 4 {\mu}m, the axial localization accuracy is 12.1 nm ~ 73.5 nm. The feasibility of this method is validated by experimental data.

Stimulated Emission Depletion Microscopy (STED) can achieve a spatial resolution as high as several nanometers. As a point scanning imaging method, it requires 3D scanning to complete the imaging of 3D samples. The time-consuming 3D scanning can be compressed into a 2D one in the non-diffracting Bessel-Bessel STED (BB-STED) where samples are effectively excited by an optical needle. However, the image is just the 2D projection, i.e., there is no real axial resolution. Therefore, we propose a method to encode axial information to axially dense emitters by using a detection optical path with single-helix point spread function (SH-PSF), and then predicted the depths of the emitters by means of deep learning. Simulation demonstrated that, for a density 1~ 20 emitters in a depth range of 4 nm, an axial precision of ~35 nm can be achieved. Our method also works for experimental data, and an axial precision of ~63 nm can be achieved.

Fourier light-field microscopy (FLFM) uses a micro-lens array (MLA) to segment the Fourier Plane of the microscopic objective lens to generate multiple two-dimensional perspective views, thereby reconstructing the three-dimensional(3D) structure of the sample using 3D deconvolution calculation without scanning. However, the resolution of FLFM is still limited by diffraction, and furthermore, dependent on the aperture division. In order to improve its resolution, a Super-resolution optical fluctuation Fourier light field microscopy (SOFFLFM) was proposed here, in which the Sofi method with ability of super-resolution was introduced into FLFM. SOFFLFM uses higher-order cumulants statistical analysis on an image sequence collected by FLFM, and then carries out 3D deconvolution calculation to reconstruct the 3D structure of the sample. Theoretical basis of SOFFLFM on improving resolution was explained and then verified with simulations. Simulation results demonstrated that SOFFLFM improved lateral and axial resolution by more than sqrt(2) and 2 times in the 2nd and 4th order accumulations, compared with that of FLFM.

The last two decades have witnessed a dramatic growth of wearable sensor technology, mainly represented by flexible, stretchable, on-skin electronic sensors that provide rich information of the wearer's health conditions and surroundings. A recent breakthrough in the field is the development of wearable chemical sensors based on surface-enhanced Raman spectroscopy (SERS) that can detect molecular fingerprints universally, sensitively, and noninvasively. However, while their sensing properties are excellent, these sensors are not scalable for widespread use beyond small-scale human health monitoring due to their cumbersome fabrication process and limited multifunctional sensing capabilities. Here we demonstrate a highly scalable, wearable SERS sensor based on an easy-to-fabricate, low-cost, ultrathin, flexible, stretchable, adhesive, and bio-integratable gold nanomesh. It can be fabricated in any shape and worn on virtually any surface for label-free, large-scale, in-situ sensing of diverse analytes from low to high concentrations (10 nM - 1 mM). To show the practical utility of the wearable SERS sensor, we test the sensor for the detection of sweat biomarkers, drugs of abuse, and microplastics. Is it possible to modulate the inherent properties of a single light beam, namely amplitude, phase and polarization, simultaneously, by merely its phase? Here, we solve this scientific problem by unifying all these three properties of a single light beam using phase vectorization and phase version of Malus's law. Full-property spatial light modulator is therefore developed based on the unification of these fundament links, which enables pixel-level polarization, amplitude and phase manipulation of light beams in a real-time dynamic way. This work not only implies that the amplitude, phase and polarization of a single light beam are interconnected, but also offers a solid answer on how to modulate these three natures of a single light beam simultaneously, which will deepen our understanding about the behavior of light beam, and facilitating extensive developments in optics and relate fields.

Turbulence plays a very important role in determining the transport of energy and particles in tokamaks. This work is devoted to studying the chaotic diffusion in multi-scale turbulence in the context of the nonlinear wave-particle interaction. Turbulent waves with different scales of characteristic wavelengths can interact with the same group of charged particles when their phase velocity is close to the velocities of the charged particles. A multi-wavenumber standard mapping is developed to model the chaotic diffusion in multi-scale turbulence. The diffusion coefficient is obtained by calculating the correlation functions analytically. It is found that the contribution of the largest scale turbulence dominates the deviation from the quasi-linear diffusion coefficient. Increasing the overlap parameters of the smaller scale turbulence by just the increasing the wavenumber cannot make the diffusion coefficient to be the quasi-linear diffusion coefficient for a finite wave amplitude. Especially, in two-scale turbulence, the diffusion coefficient is mostly over the quasi-linear diffusion coefficient in the large wavenumber (of the smaller scale turbulence) limit. As more scales of components are added in the turbulence, the diffusion coefficient approaches the quasi-linear diffusion coefficient. Light beam with optical vortices can propagate in free space only with integer orbital angular momentum. Here, we invert this scientific consensus theoretically and experimentally by proposing light beams carrying natural non-integer orbital angular momentum. These peculiar light beams are actually special solutions of wave function, which possess optical vortices with the topological charge l+0.5, where l is an integer. Owing to the interaction of phase and polarization singularity, these vortex beams with fractional topological charge can maintain their amplitude and vortex phase even when they propagate to an infinite distance. This work demonstrates another state of optical vortices in free space, which will fundamentally inject new vigor into optics, and other relate scientific fields.

Vector polarization induced by a change in scalar phase has been far beyond our understanding of the relationship between polarization and phase in classical optics due to the entanglement of inherent polarized modes of light beams. To overcome this limitation, we establish this miraculous relationship by the principle of phase vectorization, which can transform three particular phases into linear, circular and elliptical polarizations. A polarized-spatial light modulator based on the principle of phase vectorization can be achieved using a phase-only spatial light modulator, which not only enables pixel-level polarization manipulation of light beams in a real-time dynamic way but also simultaneously retains complete phase control. This work demonstrates the phase-to-polarization link and reports the creation of polarized-spatial light modulator, which will transform our view of the natural properties of light beam and pave the way for the era of vector optics.

Recently, a type of curved light beams, photonic hooks (PHs), was theoretically predicted and experimentally observed. The production of photonic hook (PH) is due to the breaking of structural symmetry of a plane-wave illuminated microparticle. Herein, we presented and implemented a new approach, of utilizing the symmetry-broken of the microparticles in material composition, for the generation of PHs from Janus microcylinders. Finite element method based numerical simulation and energy flow diagram represented theoretical analysis were used to investigate the field distribution characteristics and formation mechanism of the PHs. The full width at half-maximum (FWHM) of the PH (~0.29$\lambda$) is smaller than the FWHM of the photonic nanojet (~0.35$\lambda$) formed from a circular microcylinder with the same geometric radius. By changing the refractive index contrasts between upper and lower half-cylinders, or rotating the Janus microcylinder relative to the central axis, the shape profiles of the PHs can be efficiently modulated. The tunability of the PHs through simple stretching or compression operations, for the Janus microcylinder constituted by one solid inorganic half-cylinder and the other flexible polymer half-cylinder, was studied and discussed as well.

A light beam propagating with an infinite anti-diffracting distance requires infinite power to preserve its shape. However, the fundamental barrier of finite power in free space has made the problem of diffraction insurmountable over the past few decades. Here, to overcome this limitation, we report an approach that employs the multiple energy oscillation mechanism, thereby permitting the creation of a light beam with an ultralong anti-diffracting distance in free space. The anti-diffracting distance is no longer restricted by finite power in free space but instead depends on the number of energy oscillations. This unprecedented propagation behavior is attributed to a new understanding of non-diffractive light beam: when an anti-diffracting light beam completely discharges its energy, it cannot recharge again. A versatile optical pen is therefore developed to manipulate the number, amplitude, position and phase of energy oscillations for an arbitrary numerical aperture of a focusing lens so that energy recharge can occur in free space and multiple energy oscillations can be realized. A light beam with a tunable number of energy oscillations is eventually generated in free space and propagates along a wavy trajectory. This work will enable extending non-diffractive light beams to an expanded realm and facilitate extensive developments in optics and other research fields, such as electronics and acoustics.
