Abstract

Age-related macular degeneration (AMD) is the leading cause of blindness in elderly people, with limited treatment options available for most patients. AMD involves the death of retinal pigment epithelium (RPE) and photoreceptor cells, with mitochondria dysfunction being a critical early event. In the current study, we utilized our unique resource of human donor RPE graded for AMD presence and severity to investigate proteome-wide dysregulation involved in early AMD. Organelle-enriched fractions of RPE were isolated from donors with early AMD (n = 45) and healthy age-matched controls (n = 32) and were analyzed by UHR-IonStar, an integrated proteomics platform enabling reliable and in-depth proteomic quantification in large cohorts. A total of 5941 proteins were quantified with excellent analytical reproducibility, and with further informatics analysis, many biological functions and pathways were found to be significantly dysregulated in donor RPE samples with early AMD. Several of these directly pinpointed changes in mitochondrial functions, e.g., translation, ATP metabolic process, lipid homeostasis, and oxidative stress. These novel findings highlighted the value of our proteomics investigation by allowing a better understanding of the molecular mechanisms underlying early AMD onset and facilitating both treatment development and biomarker discovery.

8. A mechanism of gene evolution generating mucin function

Abstract

How novel gene functions evolve is a fundamental question in biology. Mucin proteins, a functionally but not evolutionarily defined group of proteins, allow the study of convergent evolution of gene function. By analyzing the genomic variation of mucins across a wide range of mammalian genomes, we propose that exonic repeats and their copy number variation contribute substantially to the de novo evolution of new gene functions. By integrating bioinformatic, phylogenetic, proteomic, and immunohistochemical approaches, we identified 15 undescribed instances of evolutionary convergence, where novel mucins originated by gaining densely O-glycosylated exonic repeat domains. Our results suggest that secreted proteins rich in proline are natural precursors for acquiring mucin function. Our findings have broad implications for understanding the role of exonic repeats in the parallel evolution of new gene functions, especially those involving protein glycosylation.

37. ShieldScatter: Improving IoT Security with Backscatter Assistance

Abstract

The lightweight protocols and low-power radio technologies open up many opportunities to facilitate Internet-of-Things (IoT) into our daily life, while their minimalist design also makes IoT devices vulnerable to many active attacks due to the lack of sophisticated security protocols. Recent advances advocate the use of an antenna array to extract fine-grained physical-layer signatures to mitigate these active attacks. However, it adds burdens in terms of energy consumption and hardware cost that IoT devices cannot afford. To overcome this predicament, we present ShieldScatter, a lightweight system that attaches battery-free backscatter tags to single-antenna devices to shield the system from active attacks. The key insight of ShieldScatter is to intentionally create multi-path propagation signatures with the careful deployment of backscatter tags. These signatures can be used to construct a sensitive profile to identify the location of the signals' arrival, and thus detect the threat. We prototype ShieldScatter with USRPs and ambient backscatter tags to evaluate our system in various environments. The experimental results show that even when the attacker is located only 15 cm away from the legitimate device, ShieldScatter with merely three backscatter tags can mitigate 97% of spoofing attack attempts while at the same time trigger false alarms on just 7% of legitimate traffic.

38. Does monetary policy affect the stock pledge of listed companies? Evidence from Chinese listed companies

Abstract

Based on the financing motivation of equity pledges, this study explores whether and how monetary policy adjustment affects the stock pledge behaviour of listed companies. The empirical findings show that monetary policy adjustments significantly affect the stock pledge of listed companies; that is, a tightening monetary policy increases the stock pledge phenomenon of listed companies, and an expansionary monetary policy reduces the stock pledge phenomenon. Heterogeneity analysis revealed that monetary policy had a greater impact on the stock pledges of non-state-owned enterprises, enterprises with weak government–enterprise relationships and enterprises without bank–enterprise relationship. The results of mechanism tests suggested that a quantitative monetary policy only affects the stock pledge behaviour of listed companies through financing costs and that a price-based monetary policy affects the stock pledge behaviour of listed companies through financing costs and financing scale. This article expands the literature on the impact of monetary policies on equity pledges and enriches studies on macroeconomic policies and microfirm behaviour.
