Abstract

We aim to investigate the mechanism of acetowhitening upon which the colposcopic diagnosis of cervical cancer is based. The changes in light scattering induced by acetic acid in intact cervical cancer cells and cellular components were studied using elastic light-scattering spectroscopy. After adding acetic acid to intact cancer cell culture samples (cell suspensions and attached monolayer cell cultures), a slight decrease in small-angle forward scattering was observed, while the large-angle scattering increased by a factor of 5-9, indicating that acetowhitening signals are mainly contributed from small-sized intracellular scattering structures. The cellular components of different sizes and masses were isolated to investigate their individual contribution to the changes of light scattering induced by acetic acid. The study provided the evidence that the cellular components of diameter smaller than 0.2 [micro] m in the cytoplasm are the major contributors to the acetowhitening effect in whole cells, while the light scattering from the mitochondria are not sensitive to the acetic acid. 24. Miniaturized three-dimensional endoscopic imaging system based on active stereovision

Abstract

A miniaturized three-dimensional endoscopic imaging system is presented. The system consists of two imaging channels that can be used to obtain an image from an object of interest and to project a structured light onto the imaged object to measure the surface topology. The structured light was generated with a collimated monochromatic light source and a holographic binary phase grating. The imaging and projection channels were calibrated by use of a modified pinhole camera. The surface profile was extracted by use of triangulation between the projected feature points and the two channels of the endoscope. The imaging system was evaluated in three-dimensional measurements of several objects with known geometries. The results show that surface profiles of the objects with different surfaces and dimensions can be obtained at high accuracy. The in vivo measurements at tissue sites of human skin and an oral cavity demonstrated the potential of the technique for clinical applications. OCIS codes: 170.2150, 170.3890, 150.6910, 170.0110.

Abstract

Our primary goal in this study is to demonstrate that near-infrared Raman spectroscopy is feasible as a rapid and reagentless analytic method for clinical diagnostics. Raman spectra were collected on human sera by use of a 785-nm excitation laser and a single-stage holographic spectrometer. A partial-least-squares method was used to predict the analyte concentrations of interest. The prediction errors of total protein, albumin, triglyceride, and glucose in human sera ranged from 1.0% to 10%, which are highly acceptable for clinical diagnosis, of their mean physiological levels. For investigating the potential application of near-infrared Raman spectroscopy in screening the therapeutical drugs and substances of abuse the concentrations of acetaminophen, ethanol, and codeine in water solution were measured in the same fashion. The errors of the Raman tests for acetaminophen and ethanol are lower than their toxic levels in human serum, and the sensitivity for detection of codeine fails to reach its toxic level.

Abstract

To understand the mechanisms of important lipid-related biological processes and diseases, it is highly demanded to study the dynamics of lipids in living biological system with high spatiotemporal resolution. However, in vivo quantitative analysis of lipid synthesis and lipolysis has been technically difficult to achieve by conventional lipid extraction and fluorescent staining methods. Recently, SRS microscopy has emerged as a powerful tool to probe small molecules with alkyne (C≡C) or deuterium (C-D) labeling in cell-silent region. The Raman tags have been used for the quantitative study of lipids in cells. In this study, we investigated metabolic dynamics of lipid droplets (LDs) by tracing the alkyne-tagged fatty acid 17-ODYA and deuterium-labeled saturated and unsaturated fatty acids PA-D31 & OA-D34 in living C. elegans. Specifically, we developed a hyperspectral SRS microscope system for LDs characterization. The system can sequentially excite and probe the stimulated Raman scattering-induced CH2 stretching of endogenous lipids information (2863 cm-1), C≡C stretching from 17-ODYA (2125 cm-1) and C-D stretching from deuterium-labeled fatty acids (2117 cm-1). We first examined the concentration levels of fatty acids in E. coli OP50. Two major lipid metabolic processes, namely uptake and turnover, were further studied in adult C. elegans. We imaged alkyne-tagged and deuterated fatty acids using SRS and traced their uptake, transportation, incorporation and turnover over time. Additionally, several other treatments including starvation were also conducted to study their effects on metabolic dynamics of pulse labeled 17-ODYA, PA-D31 and OA-D34. © 2018 SPIE.

Abstract

Kinesin-1 is a kind of motor protein responsible for intracellular transportation and has been studied in a variety of tissues. However, its roles in cartilage development are not clear. In this study, a kinesin-1 heavy chain (Kif5b) knockout mouse model is used to study the functions of kinesin-1 in the cartilage development. We developed a multimodal nonlinear optical (NLO) microscope system integrating stimulated Raman scattering (SRS), second harmonic generation (SHG) and two-photon excited fluorescence (TPEF) to investigate the morphological and biomedical characteristics of fresh tibial cartilage from normal and mutant mice at different developmental stages. The combined forward and backward SHG imaging resolved the fine structure of collagen fibrils in the extracellular matrix of cartilage. Meanwhile, the chondrocyte morphology in different zones of cartilage was visualized by label-free SRS and TPEF images. The results show that the fibrillar collagen in the superficial zone of cartilage in postnatal day 10 and 15 (P10 and P15) knockout mice was significantly less than that of control mice. Moreover, we observed distorted morphology and disorganization of columnar arrangement of chondrocytes in the growth plate cartilage of mutant mice. This study reveals the significant roles of kinesin-1 in collagen formation and chondrocyte morphogenesis.
