3. Comparison of physico-chemical characteristics of myrtle at different ripening stages

Abstract

The fruits of myrtle at different stages of development, namely green (G), colour break (C) and mature (M), were used to investigate the changes of fruit quality and anthocyanin composition during its development. Five anthocyanin components, such as delphinidin (Dp), cyanidin (Cy), pelargonidin (Pg), peonidin (Pn) and malvidin (Mv), were detected. Among them, the content of Dp glucoside was the highest (2.12 μg · g−1) and the content of Pn glucoside was the lowest (0.17 μg · g−1) at the green stage. The content of Cy glucoside was the highest (120.94 and 1,200.03 μg · g−1), and the content of Mv glucoside was the lowest (19.50 and 62.72 μg · g−1) at the colour break and mature stages. The single fruit weight, vertical diameter and transverse diameter at the mature stage were significantly higher than those at the colour break and the green stages by 0.87 g, 1.27 mm, 1.53 mm and 1.54 g, 3.4 mm, 3.55 mm, respectively. Fruit quality, the contents of soluble sugar and carotenoids, gradually increased with the development of fruit; and the contents were the highest at the mature stage (17.68% and 16.90 μg · L−1). The contents of titratable acidity, total phenol, flavonoids, chlorophyll a and total chlorophyll gradually decreased with the development of fruit, the content was the lowest at the mature stage (2.71 mmol · 100 g−1, 9.29 μg · g−1, 0.1%, 0.37 μg · L−1, 0.9 μg · L−1) and the content was the highest at the green stage (5.25 mmol · 100 g−1, 21.07 μg · g−1, 0.17%, 8.86 μg · L−1, 15.47 μg · L−1). The difference between the green period and the colour break period was significant.

Abstract

Dendrobium officinale Kimura & Migo is a perennial herbaceous plant of the genus Dendrobium in the family of Orchidaceae with high medicinal value. Melatonin (MT) is an indole-like tryptamine with functions such as regulating plant growth and development. This experiment investigated the effects of different concentrations of MT on the growth and development of protocorms of D. officinale protocorm-like bodies (PLBs). The results showed that the changes in morphological indicators such as color, cluster size, and surface changes were more significant under 75 µM MT than those of 0 µM (CK), and the appearance of white on the PLB surface was expedited, which was more conducive to the proliferation of PLBs. MT treatment of 100 µM inhibited the differentiation of adventitious buds, and the contents of photosynthetic pigments, polysaccharides, and flavonoids were significantly increased. Moreover, as compared with CK, the activities of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) in PLBs increased significantly, while the content of malondialdehyde (MDA) decreased gradually with 75 µM or less. In conclusion, a concentration of 75 µM melatonin can enhance the rapid propagation rate of D. officinale in vitro, providing insights into the effects of melatonin on the growth of tissue-cultured D. officinale seedlings. Abstract

The hydroponic method was used to study the photosynthetic characteristics of navel orange "No.52" seedlings under Cu stress. The study shows that chlorophyll (Chl.) content under 0.1 μmol・L −1 of Cu increases significantly, but decreases under 5 μmol・L −1 , 20 μmol・L −1 and 40 μmol・L −1 of Cu. Carotenoid content decreases with increasing Cu concentration. Net photosynthetic rate and light saturation point reduces under Cu stress as Cu concentration exceeds 5 μmol・L −1 . 0.1 μmol・L −1 of Cu improves, but 5 μmol・L −1 and 20 μmol・L −1 of Cu decrease transpiration rate and stomatal conductance of orange. These parameters do not change significantly under 40 μmol・L −1 of Cu. At the same time, high Cu concentration (5 μmol・L −1 , 20 μmol・L −1 and 40 μmol・L −1 )

Abstract

ABSTRACTDendrobium cariniferumis a valuable ornamental and medicinal plant rich with polysaccharides, alkaloid, and other bioactive compounds, which are potential raw materials for pharmacological utilization. In this study, an efficient protocol for the rapid propagation of D. cariniferumwas developed. By using the tissue culture protocol, the effects of pH, hormone combinations, temperatures, light intensity, culture time protocorm proliferation, seedlings rooting, and accumulation of biomass with bioactive compounds were investigated. The experiments showed that the medium [1/2 MS + activated carbon1.0 g/L+ agar strip 7.5 g/L + sucrose 25 g/L] effectively promoted the germination of D. cariniferumseeds. The optimal culture conditions were found at pH 5.7, temperature 23 ± 2°C, and light intensity of 1000 Lx in the protocorm proliferation stage. Adding 1.5 g/L peptone in the medium effectively promoted the seedling rooting. The optimal culture conditions for accumulation of bioactive compounds (polysaccharides and alkaloids) of seedlings were found at temperature of 25 ± 2°C, light intensity of 1500–2000 Lx after the 60-day (d). Our study constructed a rapid propagation system in vitro for D. cariniferum, as well as the methods for efficient accumulation of active substances in seedling culture, which will serve as guidance for industrial production of D. cariniferumseedlings for both medicinal raw materials and ornamental plants. In addition, our study provided a new idea that we can directly use the high bioactive compound seedlings to extract medicinal components in industry conditions without transferring to the field.

45. In vitro regeneration of Abelmoschus esculentusL. cv. Wufu: Influence of anti-browning additives on phenolic secretion and callus formation frequency in explants

Abstract

Okra (Abelmoschus esculentusL.) belongs to the Malvaceae family and is a difficult species to manipulate and regenerate in vitro. In this study, factors that influence secretion of phenolic compounds, callus induction, and subsequent regeneration in okra were optimized. Different concentrations of anti-browning additives, such as activated charcoal (AC) and citric acid (CA) were tested alone or in combination with ascorbic acid (AA) in MS medium to evaluate their ability to control phenolic secretion from hypocotyl and cotyledon explants. Among the anti-browning additives, 200 mg·L -1AC and 10 mg·L -1CA + AA (1:1) were best at controlling phenolic secretion from hypocotyl and cotyledon explants in callus induction medium. Hypocotyl explants were more suitable for callus initiation and organogenesis than cotyledon explants. Moreover, among twelve types of media tested for callus culture, the addition of 0.5 mg·L -12,4-dicholorophenoxy acetic acid (2,4-D) and 1.5 mg·L -1benzyladenine (BA) to the growth media was the most effective for hypocotyl explants, whereas MS + 1.5 mg·L -1α-naphthylacetic acid (NAA) and 0.5 mg·L -16-benzyloaminopurine (BAP) was effective for cotyledon explants. Improved shoot regeneration was achieved with 2 mg·L -1BAP and 0.1 mg·L -1indole butyric acid (IBA). The maximum number of strong and healthy roots was obtained on media that included 2 mg·L -1IBA and 200 mg·L -1AC. Rooted plantlets were successfully acclimatized in earthen pots, and the rooted plantlets had normal morphology and growth characteristics. The present study offers a potential solution for controlling phenolic secretion during okra regeneration using additives and evaluates the manipulation of plant growth regulators for effective regeneration of the Wufu okra cultivar.

Abstract

A leaf blight disease with an incidence level of about 50% was found on Robusta banana in Guangdong province of China in September 2020. The early symptom appeared as pale gray to black brown, irregular, small, necrotic lesions mainly on the top 3–5 leaves. Severely infected leaves were withered and necrotic. Two representative fungus strains, strain L1 and strain L2, were isolated from affected banana leaves, and morphological and molecular identification analysis confirmed that the two fungi were both Alternaria jacinthicola. Many Alternaria species have been reported to cause wilting, decay, leaf blight and leaf spots on plants and lead to serious economic losses in their production, including A. alternata, causing leaf blight and leaf sport diseases on banana. The Koch's postulates of A. jacinthicola causing the leaf blight disease was further fulfilled, which confirmed that it is the causal agent of this disease. Abstract

As an emerging technology, shoot encapsulation has been employed in germplasm conservation, distribution, and micropropagation of elite plant species. However, the production of synthetic seeds of sweet potato via non-zygotic embryogenesis requires a large number of embryos per cultured callus suspension and is labour-intensive. Here, we reported a simple method of encapsulating in vitro derived vegetable sweet potato nodal segments with sodium alginate, calcium chloride (CaCl2), and Murashige and Skoog (MS) salts. The nodes encapsulated with 4% sodium alginate (w/v) and 100 mM CaCl2 were the most suitable for propagation. They had uniform spherical beads and took the least number of days to shoot and root emergence. These plantlets produced more leaves, roots, and long shoots. Further evaluation of the MS salts concentration revealed that the plantlets encapsulated and grown with ½ MS salts had the least days to shoot and root emergence. They also had a longer shoot, the highest conversion rate (99%), and the least leaf abscission (17%). Thus, the sweet potato nodal segments encapsulated with 4% sodium alginate, 100 mM CaCl2, and ½ MS salts could be used as excellent material for micropropagation, germplasm conservation, and exchange of sweet potato planting materials. 49. Comparative Studies on the Quality and Lycopene Content of Pomelo (Citrus grandisOsbeck) Cultivars

Abstract

Three cultivars of pomelo “Guanximiyou” (GX) and its variants “Hongroumiyou” (HR) and “Sanhongmiyou” (SH) were selected as experimental materials to evaluate the quality and edible value of the fruit. In this study, the fruit quality and lycopene contents were investigated. The results show that among the 3 cultivars used, the contents of soluble solids, vitamin C, and total phenol in the juice sac of SH were the highest, followed by HR and GX. SH has the largest fruit shape index, soluble sugar content: GX > HR > SH, and titratable acid content: GX < HR < SH. From our findings, the content of lycopene was the highest in SH, followed by HR and GX. SH has the greatest potential for production due to its appreciable content of lycopene and fruit quality.
