Abstract Extracellular matrix (ECM) assembly/disassembly is a critical regulator for airway epithelial development and remodeling. Airway organoid is widely used in respiratory research, yet there is limited study to indicate the roles and mechanisms of ECM organization in epithelial growth and differentiation by using in vitro organoid system. Moreover, most of current Matrigel-based airway organoids are in basal-out orientation where accessing the apical surface is challenging. We present a human apical-out airway organoid using a biochemically defined hybrid hydrogel system. During human nasal epithelial progenitor cells (hNEPCs) differentiation, the gel gradually degrade, leading to the organoid apical surfaces facing outward. The expression and activity of ECM-degrading enzymes, matrix metalloproteinases (MMP7, MMP9, MMP10 and MMP13) increases during organoid differentiation, where inhibition of MMPs significantly suppresses the normal ciliation, resulting in increased goblet cell proportion. Moreover, a decrease of MMPs is found in goblet cell hyperplastic epithelium in inflammatory mucosa. This system reveals essential roles of epithelial-derived MMPs on epithelial cell fate determination, and provides an applicable platform enabling further study for ECM in regulating airway development in health and diseases.

The aim of this experiment is to explore the effect of sodium sulfate (Na2SO4) on methane reduction in the rumen, and its impact on anaerobic methane-oxidizing archaea (ANME). Using mixed rumen fluid from four Angus cattle fistulas, this study conducted an in vitro fermentation. Adding Na2SO4 to the fermentation substrate resulted in sulfur concentrations in the substrate of 0.4%, 0.6%, 0.8%, 1.0%, 1.2%, 1.4%, 1.6%, 1.8%, 2.0%, 2.2%, and 2.4%. The gas production rate and methane yield were measured using an in vitro gas production method. Subsequently, the fermentation fluid was collected to determine the fermentation parameters. The presence of ANME in the fermentation broth, as well as the relationship between the number of bacteria, archaea, sulfate reducing bacteria (SRB), ANME, and the amount of Na2SO4 added to the substrate, were measured using qPCR. The results showed that: (1) the addition of Na2SO4 could significantly reduce CH4 production and was negatively correlated with CO2 production; (2) ANME-1 and ANME-2c did exist in the fermentation broth; (3) the total number of archaea, SRB, ANME-1, and ANME-2c increased with the elevation of Na2SO4. The above results indicated that Na2SO4 could mitigate methane production via sulfate-dependent anaerobic methane oxidation (S-DAMO) in the rumen. In the future management of beef cattle, including sodium sulfate in their diet can stimulate S-DAMO activity, thereby promoting a reduction in methane emissions.

With the increase in human activities and the acceleration of urbanization, over-exploitation of natural resources has led to a decline in ecosystem services (ESs), subsequently affecting the achievement of sustainable development goals (SDGs). As the key ecological zone of Qinghai-Tibet Plateau, the stability and enhancement of ESs in the Hehuang Valley are crucial for achieving SDGs and biodiversity conservation. This study quantifies nine SDGs for the Hehuang Valley in the last twenty years. Four ecological models were utilized to compute key ESs: net primary productivity (NPP), water yield, soil retention, and sand fixation. Panel data were analyzed using a coupling coordination model to quantify the relationship between ESs and sustainable development level (SDL) in each county. Additionally, the Geographically and Temporally Weighted Regression (GTWR) model was employed to examine the correlation between ESs and SDL. The results indicate the following: (1) During the period, NPP and water yield first increased and then decreased. The capacity for soil retention and sand fixation showed an overall increase, highlighting substantial variability among counties in their ability to deliver these ESs. (2) The SDL of counties in the Hehuang Valley increased, with Xining City showing slightly higher SDL than other counties. (3) The overall coupling coordination degree among NPP, water yield, soil retention, sand fixation, and SDL in the Hehuang Valley exhibited an upward trend in the last twenty years. SDL demonstrated the highest coordination degree with NPP, followed by soil retention, water yield, and sand fixation. (4) Most counties in the Hehuang Valley exhibited a lag in SDL relative to NPP, water yield, and soil retention in the last twenty years. In the early stage, sand fixation and SDL were primarily lagging in SDL, while in the late stages, sand fixation lagged behind SDL. (5) During the period, there was an increasing negative correlation observed between the four ESs and SDL. The positive contribution of NPP and sand fixation in some counties gradually shifted to a negative effect, and the negative effect of water yield and soil retention on SDL intensified. The impact of human activities on ecosystem function hindered local SDL. This study offers scientific theoretical backing and practical recommendations for promoting SDL and biodiversity conservation in the Hehuang Valley.

The severe wildfires that have ravaged Guangdong province, China, present a significant threat to the local ecosystem, socio-economics, and public health. Effective risk assessment is essential for early warning and timely prevention in wildfire management, thereby mitigating disaster losses. In this study, we compiled a dataset comprising 11,507 historical wildfire incidents in Guangdong Province spanning a decade (2011–2021) and developed a deep learning-based model to predict the likelihood of wildfire occurrence in the region. In addition to analyzing risk characteristics throughout the year, we also trained separate models for different seasons and analyzed the discrepancies in the contribution of driven factors to wildfire occurrence across seasons. Furthermore, the performance of our deep learning-based model was compared with that of traditional machine learning algorithms. The experimental results revealed that: (1) Factors such as relative humidity, temperature, NDVI, and precipitation exerted significant influence on wildfire occurrence. (2) The impact of wildfire driving factors varied across different seasons. (3) Our deep learning model outperformed traditional machine learning models, achieving a superior performance with an AUC of 0.962.

Generally, the soil requires to be compacted in highway construction. The expansive soil is a special type of soil that is highly susceptible to variations in water content, which affects the degree of compaction at the same compaction energy. In the present study, a series of wet compaction tests and dry compaction tests were carried out in the laboratory. Laboratory test results show that dry compaction will produce a higher optimum water content and a higher maximum dry unit weight compared to wet compaction, because its matric suction is smaller. Field compaction tests were also conducted, the results showed that there might be a risk of under-compacting soils during construction caused by different water content change path in actual field conditions.

Wildfire is one of the most severe natural disasters globally, profoundly affecting natural ecology, economy, and health and safety. Precisely predicting the spread of wildfires has become an important research topic. Current fire spread prediction models depend on inputs from a variety of geographical and environmental variables. However, unlike the ideal conditions simulated in the laboratory, data gaps often occur in real wildfire scenarios, posing challenges to the accuracy and robustness of predictions. It is necessary to explore the extent to which different missing items affect prediction accuracy, thereby providing rational suggestions for emergency decision-making. In this paper, we tested how different conditions of missing data affect the prediction accuracy of existing wildfire spread models and quantified the corresponding errors. The final experimental results suggest that it is necessary to judge the potential impact of data gaps based on the geographical conditions of the study area appropriately, as there is no significant pattern of behavior yet identified. This study aims to simulate the impact of data scarcity on the accuracy of wildfire spread prediction models in real scenarios, thereby enabling researchers to better understand the priority of different environmental variables for the model and identify the acceptable degree of missing data and the indispensable data attributes. It offers new insights for developing spread prediction models applicable to real-world scenarios and rational assessment of the effectiveness of model outcomes.

Yak (Bos grunniens) is an important economic animal species on the Qinghai-Tibet Plateau. Yaks grazed in the cold season often suffer from nutritional stress, resulting in low production performance. This situation can be improved by properly feeding the grazing yaks in the cold season; however, there is still little information about the effect of different feeding levels on the intestinal microflora and metabolites of yaks. Therefore, this study aimed to explore the effect of feeding different doses of concentrate supplements on rumen bacterial communities and metabolites in grazing yaks during the cold season. Feed concentrate supplementation significantly improved the production performance and rumen fermentation status of grazing yaks during the cold season, and switched the type of ruminal fermentation from acetic acid fermentation to propionic acid fermentation. Ruminal fermentation parameters and ruminal bacterial abundance correlated strongly. At the phylum level, the abundance of Firmicutes increased with increasing concentrate supplementation, while the opposite was true for Bacteroidota. At the genus level, the abundance of Christensenellaceae_R-7_group, NK4A214_group, Ruminococcus, norank_f__Eubacterium_coprostanoligenes_group, norank_f__norank_o__ Clostridia_UCG-014, Lachnospiraceae_NK3A20_group, Acetitomaculum, and Family_XIII_AD3011_group increased with increasing concentrate supplementation, while the abundance of Rikenellaceae_RC9_gut_ group decreased. Dietary concentrate supplementation altered the concentration and metabolic mode of metabolites in the rumen, significantly affecting the concentration of metabolites involved in amino acid and derivative metabolism (e.g., L-aspartic acid, L-glutamate, and L-histidine), purine metabolism (e.g., guanine, guanosine, and hypoxanthine), and glycerophospholipid metabolism (e.g., phosphatidate, phosphatidylcholine, and phosphocholine), and other metabolic pathways. The strong correlation between yak rumen microorganisms and metabolites provided a more comprehensive understanding of microbial community composition and function. This study showed significant changes in the composition and abundance of bacteria and metabolites in the rumen of cool season grazing yaks fed with concentrate supplements. Changes in ruminal fermentation parameters and metabolite concentration also showed a strong correlation with ruminal bacterial communities. These findings will be helpful to formulate supplementary feeding strategies for grazing yaks in the cold season from the perspective of intestinal microorganisms.

For assessing the influence of vegetation variation in upper reaches of Luanhe River (URLR) on streamflow, we first computed the equation between underlying surface parameter (ω) and the Normalized Difference Vegetation Index (NDVI). Then we introduced the vegetation information into the Budyko equation and established a modified Budyko equation. Finally, the impact of vegetation change on streamflow in URLR were estimated using the modified Budyko equation. Guanidinoacetic acid (GAA) functions as a precursor for creatine synthesis in the animal body, and maintaining ample creatine reserves is essential for fostering rapid growth. This study aimed to explore the impact of GAA supplementation on growth performance, rumen fermentation, blood indices, nutrient digestion, and nitrogen metabolism in Angus steers through two experiments: a feeding experiment (Experiment 1) and a digestive metabolism experiment (Experiment 2). In Experiment 1, thirty-six Angus steers (485.64 ± 39.41 kg of BW) at 16 months of age were randomly assigned to three groups: control (CON), a conventional dose of GAA (CGAA, 0.8 g/kg), and a high dose of GAA (HGAA, 1.6 g/kg), each with twelve steers. The adaptation period lasted 14 days, and the test period was 130 days. Weighing occurred before morning feeding on days 0, 65, and 130, with rumen fluid and blood collected before morning feeding on day 130. Experiment 2 involved fifteen 18-month-old Angus steers (575.60 ± 7.78 kg of BW) randomly assigned to the same three groups as in Experiment 1, with a 7-day adaptation period and a 3-day test period. Fecal and urine samples were collected from all steers during this period. Results showed a significantly higher average daily gain (ADG) in the CGAA and HGAA groups compared to the CON group (p = 0.043). Additionally, the feed conversion efficiency (FCE) was significantly higher in the CGAA and HGAA groups than in the CON group (p = 0.018). The concentrations of acetate and the acetate:propionate ratio were significantly lower in the CGAA and HGAA groups, while propionate concentration was significantly higher (p < 0.01). Serum concentration of urea (UREA), blood ammonia (BA), GAA, creatine, and catalase (CAT) in the CGAA and HGAA groups were significantly higher than in the CON group, whereas malondialdehyde (MDA) concentrations were significantly lower (p < 0.05). Digestibility of dry matter (DM) and crude protein (CP) and the nitrogen retention ratio were significantly higher in the CGAA and HGAA groups than in the CON group (p < 0.05). In conclusion, dietary addition of both 0.8 g/kg and 1.6 g/kg of GAA increased growth performance, regulated rumen fermentation and blood indices, and improved digestibility and nitrogen metabolism in Angus steers. However, higher doses of GAA did not demonstrate a linear stacking effect.

This study was conducted to explore the potential effect of Yucca schidigera extract (YSE) on the metabolism of beef cattle. Thirty Angus crossbreed steers were selected, with an initial mean body weight of 506.6 ± 33.3 kg, and assigned to two treatments: a diet with no additives (CON group) and a diet supplemented with 1.75 g/kg of YSE (YSE group) (on a dry matter basis). The experiment lasted for 104 days, with 14 days for adaptation. The results showed that adding YSE could significantly improve the average daily gain (ADG) from 1 to 59 d (15.38%) (p = 0.01) and 1 to 90 d (11.38%) (p < 0.01), as well as dry matter digestibility (DMD) (0.84%) (p < 0.05). The contents of alanine aminotransferase, aspartate aminotransferase, and bilirubin and the total antioxidant capacity were increased and blood urea was reduced in the YSE group, compared to the CON group (p < 0.05). Both the glycerophospholipids and bile acids, including phosphocholine, glycerophosphocholine, PC(15:0/18:2(9Z,12Z)), PE(18:0/20:3(5Z,8Z,11Z)), PE(18:3(6Z,9Z,12Z)/P-18:0), LysoPC(15:0), LysoPC(17:0), LysoPC(18:0), LysoPC(20:5(5Z,8Z,11Z,14Z,17Z)), deoxycholic acid, glycocholic acid, and cholic acid, were upregulated by the addition of YSE. In summary, YSE may improve the ADG by increasing the blood total antioxidant capacity and glycerophospholipid synthesis, maintaining steers under a healthy status that is beneficial for growth. Furthermore, YSE may also increase the expression of bile acid synthesis, thereby promoting DMD, which, in turn, offers more nutrients available for growth.

The complex mechanical characteristics of the Xiyu conglomerate significantly influence the resistance and deformation features of its caverns’ surrounding rock, thereby constraining the construction of related water diversion tunnels. This paper introduces an improved SegFormer framework developed for the detection of mesoscale geomaterial structures. Computerized tomography (CT) scan images of the Xiyu conglomerate were employed to establish a high-precision numerical model. From the results of segmentation, the proposed algorithm outperformed UNet, HRNet, and the original SegFormer neural network. The segmentation results were used to calculate the porosity, and biaxial compression numerical simulation experiments based on the real structure were carried out using the particle flow code (PFC). We observed the failure process of the model and obtained the shear strength of the Xiyu conglomerate. We explored the causes and influencing factors of the anisotropy of the Xiyu conglomerate from the microstructure perspective and provide a micro-observation basis for establishing an anisotropic mechanical model.

With the increase in energy demand, environmental issues such as carbon emissions are becoming more and more prominent. China will scale its intended nationally determined contributions by adopting more vigorous policies and measures. China aims to have CO2 emissions peak before 2030 and achieve carbon neutrality before 2060. The current challenge and priority of China’s high-quality development is to ensure a harmonious balance between the ecological environment and the economy. The South-to-North Water Diversion Project passes through Beijing, Tianjin, Henan, and Hebei, which were chosen as the study sites. The carbon emission data was from the China Carbon Emission Database 2000–2019. Decoupling modeling using statistical yearbook data from four provinces and municipalities. KMO and Bartlett’s test used SPSS 27 software. The selection of indicators was based on relevance. Analyses were performed using the extended STIRPAT model and ridge regression. Moreover, projections of carbon peaks in the study area for 2020–2035 under different rates of change were simulated by the scenario analysis method. The results show that: (1) The decoupling analysis of the four provinces and cities from 2000-2019 gradually shifts to strong decoupling; (2) Resident population, energy structure, and secondary industry as a proportion of GDP significantly impact carbon emissions; (3) From 2000–2035, Beijing and Henan experienced carbon peaks. The peak time in Beijing was 96.836 million tons in 2010. The peak time in Henan was 654.1004 million tons in 2011; (4) There was no peak in Hebei from 2000–2035.

Similar to hydraulic fracturing (HF), the coalescence and fracture of cracks are induced within a rock under the action of an ultrasonic field, known as ultrasonic fracturing (UF). Investigating UF is important in both hard rock drilling and oil and gas recovery. A three-dimensional internal laser-engraved crack (3D-ILC) method was introduced to prefabricate two parallel internal cracks within the samples without any damage to the surface. The samples were subjected to UF. The mechanism of UF was elucidated by analyzing the characteristics of fracture surfaces. The crack propagation path under different ultrasonic parameters was obtained by numerical simulation based on the Paris fatigue model and compared to the experimental results of UF. The results show that the 3D-ILC method is a powerful tool for UF research. Under the action of an ultrasonic field, the fracture surface shows the characteristics of beach marks and contains powder locally, indicating that the UF mechanism includes high-cycle fatigue fracture, shear and friction, and temperature load. The two internal cracks become close under UF. The numerical result obtained by the Paris fatigue model also shows the attraction of the two cracks, consistent with the test results. The 3D-ILC method provides a new tool for the experimental study of UF. Compared to the conventional numerical methods based on the analysis of stress–strain and plastic zone, numerical simulation can be a good alternative method to obtain the crack path under UF.

With diversification of yak breeding, it is important to understand the effects of feed type on the rumen, especially microbiota and metabolites. Due to the unique characteristics of yak, research on rumen microbes and metabolites is limited. In this study, the effects of two diet types on rumen eukaryotic microflora and metabolites were evaluated using the Illumina MiSeq platform and liquid chromatography-mass spectrometry (LC-MS). All identified protozoa belonged to Trichostomatia. At the genus level, the relative abundance of Metadinium and Eudiplodinium were significantly (p < 0.05) higher in the roughage group than that of concentrate group, while the concentrate group harbored more Isotricha. Ascomycota, Basidiomycota, and Neocallimastigomycota were the main fungal phyla, and the Wallemia, Chordomyces, Chrysosporium, Cladosporium, Scopulariopsis, and Acremonium genera were significantly (p < 0.05) more abundant in the roughage group than the concentrate group, while the concentrate group harbored more Aspergillus, Neocallimastix, Thermoascus, and Cystofilobasidium (p < 0.05). Metabolomics analysis showed that feed type significantly affected the metabolites of rumen protein digestion and absorption (L-proline, L-phenylalanine, L-tryosine, L-leucine, L-tryptophan, and β-alanine), purine metabolism (hypoxanthine, xanthine, guanine, guanosine, adenosine, and adenine), and other metabolic pathway. Correlation analysis revealed extensive associations between differential microorganisms and important metabolites. The results provide a basis for comprehensively understanding the effects of feed types on rumen microorganisms and metabolites of yaks. The findings also provide a reference and new directions for future research.

The treatment of severe cases usually requires multimodality hemodynamic monitoring approaches, particularly for tissue and organ perfusion tracking. Currently, only a few studies have investigated renal perfusion status at the bedside. Ultrasound has become increasingly utilized to guide the hemodynamic management of severe patients. Similarly, intrarenal Doppler (IRD) is widely used to assess renal perfusion from both the intrarenal artery and vein perspectives. The renal resistive index (RRI), which reflects the renal arterial blood flow profile, is often applied to predict the reversibility of renal dysfunction and to titrate hemodynamic support. Intrarenal venous flow (IRVF) patterns and the renal venous stasis index (RVSI), which reflects the intrarenal vein blood flow profile, are now being used to assess intravenous congestion. They may also be useful in predicting the risk of acute kidney injury and avoiding fluid overload. IRD can provide diverse and supplemental information on renal perfusion and may help to establish the early diagnosis in severe patients. This review focused on the specific operational methods, influencing factors, and applications of IRD in hemodynamics.

AbstractNear-surface imaging structures often plays a significant role in the field of environmental and engineering geophysics. Early-arrival waveform inversion (EWI) is state-of-the-art method to imaging near-surface structures due to its high resolution. However, the method faces with cycle-skipping issue which might lead to an unexpected local minimum. Envelope inversion (EI) could deal with this issue which contributes to the ultralow-frequency information extracted from the envelope but has a low resolution. We have developed a curvelet-based joint waveform and envelope inversion (CJWEI) method for inverting imaging near-surface velocity structures. By inverting two types of data, we are able to recover the low- and high-wavenumber structures and mitigate the cycle-skipping problem. Curvelet transform was used to decompose seismic data into different scales and provide a multiscale inversion strategy to further reduce non-uniqueness of waveform inversion efficiently. With synthetic test and real data application, we demonstrate that our method can constrain the anomalies and hidden layers in the shallow structure more efficiently as well as is robust in terms of noise. The proposed multiscale joint inversion offers a computational efficiency and high precision to imaging fine-scale shallow underground structures.

The discourse about urban planning, urban design, urban management, and many other basic urban planning issues are related to identifying the spatial heterogeneity in the main urban area (MUA). Road networks are important infrastructure for social and economic development and also an important driving factor for urban land use. Therefore, the topological characteristics of the urban road network can be used as an important basis for the space of the division of the MUA. This study takes the MUA space in the Harbin Ring Expressway as the research object. The findings of this research establish a city model to identify the spatial heterogeneity, through a method of “Spatial essential attribute—Spatial autocorrelation analysis—Establishment of urban model.” In this model, spatial essential attributes are obtained from the analysis of space syntax. The spatial autocorrelation analysis is a method to calculate spatial clustering, and building the urban model is a kind of computer visualization method. Using this method, this research constructs a spatial heterogeneity model (SHM) of the urban built-up area in Harbin, which consists of three layers of spatial circles and two prominent regional structures. Then, it analyzes the spatial distribution of infrastructure and population agglomeration through nuclear density. We gain three conclusions: first, in addition to educational and greening facilities, the spatial distribution of infrastructure is correlated with the regional structure of SHM in Harbin’s MUA. Second, there are differences in the spatial attraction of population agglomeration between the different areas in the SHM of Harbin’s MUA. Finally, the new identification model of the SHM is scientific, but it has some advantages and demerits. Using spatial topological values to construct the SHM, it provides a new city model in urban spatial heterogeneity region recognition. Through the research results, the identification method of the SHM improves the traditional method which identifies urban heterogeneity areas according to people’s subjective wishes in urban planning. Therefore, the SHM helps relevant practitioners and scholars practice urban planning, urban analysis, urban protection, and urban management.

COVID-19 has undergone multiple mutations, with the Omicron variant proving to be highly contagious and rapidly spreading across many countries. The United States was severely hit by the Omicron variant. However, it was still unclear how Omicron transferred across the United States. Here, we collected daily COVID-19 cases and deaths in each county from 1 December 2021 to 28 February 2022 as the Omicron wave. We adopted space-time scan statistics, the Hoover index, and trajectories of the epicenter to quantify spatiotemporal patterns of the Omicron wave of COVID-19. The results showed that the highest and earliest cluster was located in the Northeast. The Hoover index for both cases and deaths exhibited phases of rapid decline, slow decline, and relative stability, indicating a rapid spread of the Omicron wave across the country. The Hoover index for deaths was consistently higher than that for cases. The epicenter of cases and deaths shifted from the west to the east, then southwest. Nevertheless, cases were more widespread than deaths, with a lag in mortality data. This study uncovers the spatiotemporal patterns of Omicron transmission in the United States, and its underlying mechanisms deserve further exploration.

Changes in dietary composition affect the rumen microbiota in ruminants. However, information on the effects of dietary concentrate-to-forage ratio changes on yak rumen bacteria and metabolites is limited. This study characterized the effect of three different dietary concentrate-to-forage ratios (50:50, C50 group; 65:35, C65 group; 80:20, C80 group) on yak rumen fluid microbiota and metabolites using 16S rRNA gene sequencing and liquid chromatography-mass spectrometry (LC-MS) analyses. Rumen fermentation parameters and the abundance of rumen bacteria were affected by changes in the dietary concentrate-to-forage ratio, and there was a strong correlation between them. At the genus level, higher relative abundances of norank_f__F082, NK4A214_group, Lachnospiraceae_NK3A20_group, Acetitomaculum, and norank_f__norank_o__Clostridia_UCG-014 were observed with a high dietary concentrate-to-forage ratio (P < 0.05). Combined metabolomic and enrichment analyses showed that changes in the dietary concentrate-to-forage ratio significantly affected rumen metabolites related to amino acid metabolism, protein digestion and absorption, carbohydrate metabolism, lipid metabolism, and purine metabolism. Compared with the C50 group, 3-methylindole, pantothenic acid, D-pantothenic acid, and 20-hydroxy-leukotriene E4 were downregulated in the C65 group, while spermine and ribose 1-phosphate were upregulated. Compared to the C50 group, Xanthurenic acid, tyramine, ascorbic acid, D-glucuronic acid, 6-keto-prostaglandin F1a, lipoxin B4, and deoxyadenosine monophosphate were upregulated in the C80 group, while 3-methylindole and 20-hydroxy-leukotriene E4 were downregulated. All metabolites (Xanthurenic acid, L-Valine, N-Acetyl-L-glutamate 5-semialdehyde, N-Acetyl-L-glutamic acid, Tyramine, 6-Keto-prostaglandin F1a, Lipoxin B4, Xanthosine, Thymine, Deoxyinosine, and Uric acid) were upregulated in the C80 group compared with the C65 group. Correlation analysis of microorganisms and metabolites provided new insights into the function of rumen bacteria, as well as a theoretical basis for formulating more scientifically appropriate feeding strategies for yak.

Chronic rhinosinusitis with nasal polyps (CRSwNP) is characterized by heterogeneous inflammatory endotypes of unknown etiology. Invariant natural killer T (iNKT) cells are multifunctional innate T cells that exhibit Th1-, Th2-, and Th17-like characteristics. We investigated functional relationships between iNKT cells and inflammatory subtypes of CRSwNP. Eighty patients with CRSwNP and thirty-two control subjects were recruited in this study. Flow cytometry was used to analyze the frequencies and functions of iNKT cells and their subsets in peripheral blood mononuclear cells (PBMCs) and tissues. Polyp tissue homogenates were used to study the multifunctionality of iNKT cells. iNKT cells were significantly increased in polyps (0.41%) than in control mucosa (0.12%). iNKT cells were determined in the paucigranunlocytic (n=20), eosinophilic (n=22), neutrophilic (n=23), and mixed granulocytic (n=13) phenotypes of CRSwNP. The percentages of iNKT cells and HLA-DR+PD-1+ subsets were lower in eosinophilic or mixed granulocytic polyps than those of other phenotypes. iNKT cells and subsets were enriched in polyp tissues than in matched PBMCs. The evaluation of surface markers, transcription factors, and signature cytokines indicated that the frequencies of iNKT2 and iNKT17 subsets were significantly increased in eosinophilic and neutrophilic polyps, respectively, than in the paucigranulocytic group. Moreover, the production of type 2 (partially dependent on IL-7) and type 17 (partially dependent on IL-23) iNKT cells could be stimulated by eosinophilic and neutrophilic homogenates, respectively. Our study revealed that type 2 and type 17 iNKT cells were involved in eosinophilic and neutrophilic inflammation, respectively, in CRSwNP, while different inflammatory microenvironments could modulate the functions of iNKT cells, suggesting a role of iNKT cells in feedback mechanisms and local inflammation.

AbstractWith the development of hydraulic engineering, more hydropower stations are built in the complex geological condition. This work proposed a case study on stilling basin in complex fractured dam foundation using 3D nonlinear analysis. A detail implementation of the numerical mesh generation from profile to solid geometry using boundary representation (B-Rep) is illustrated. Considering the main factors like self-weight, hydrodynamic pressure, and uplift pressure, the stress and deformation results of stilling basin in complex fractured foundation are obtained. According to the calculation and analysis, the design stilling basin can meet the engineering requirements of hydraulic structure. However, it is suggested to carry out the necessary foundation treatment for the sericitized slate as well as fracture plane. The study will provide valuable reference in the analysis of related engineering.

Hotpot is a widely popular cooking method for sheepmeat in China. This study measured the sensory responses of 720 untrained Chinese consumers to Australian sheepmeat cooked using a hotpot technique with methods based on Meat Standards Australia protocols. Shoulder and leg cuts of 108 lambs and 109 yearlings were scored on tenderness, juiciness, flavour and overall liking with linear mixed effects models used to analyse the influence of muscle type and animal factors on these scores. On average, shoulder cuts were more palatable than legs cuts for all sensory traits (p < 0.01) and lambs compared to yearlings (p < 0.05). Intramuscular fat and muscularity were identified as strong drivers of eating quality (p < 0.05), with greater palatability for both cuts as intramuscular fat increased (range 2.5 to 7.5%), and muscularity decreased (as measured through loin weight adjusted for hot carcase weight). Consumers were unable to detect differences between animal sire type and sex in sheepmeat hotpot. These findings suggest shoulder and leg cuts performed comparatively well in hotpot compared to previously tested sheepmeat cooking methods and emphasise the importance of balanced selection for quality and yield traits to ensure that consumer satisfaction is maintained.

The Yellow River Basin assumes an important ecological and economic function in China. The study of carbon emissions from land use in the nine provinces (regions) of the pathway is important to achieve carbon reduction. Based on the dynamic data of land use, energy, and economic changes in nine provinces (regions) for the past 30 years from 1990 to 2018, this study analyzed the spatial and temporal evolution characteristics of land-use carbon emissions by using the carbon emission coefficient method in the IPCC inventory method and evaluating the low-carbon development model of the nine provinces (regions) by land-use carbon emission intensity. Finally, the LMDI model was used to analyze the factors influencing land-use carbon emissions. The results showed that: (1) in the past 30 years, the net carbon emissions have shown a continuously increasing trend, and the difference in the spatial distribution of carbon emissions in different periods was obvious. The carbon sink effect was not significant enough to offset the carbon emissions generated. (2) The continuously decreasing carbon emission intensity values per unit of GDP indicate that the coordination between land-use and economic development was getting better. (3) The factors of population size, economic size, and land-use structure accelerated land-use carbon emissions, whereas land-use efficiency limited land-use carbon emissions. Accordingly, this paper puts forward some corresponding policy suggestions.

Previous studies of beef cattle feed efficiency have primarily concentrated on the ruminal microbiota because it plays a key role in energy production and nutrient supply in the host. Although the small intestine is the important site of post-ruminal digestion and absorption of nutrients, only a few studies have explored the relationship between the microbial populations in the small intestine and feed efficiency. Moreover, variations in GIT metabolites contribute to differences in feed efficiency. Microbial carbonization based on carbon-capturing bacteria induced calcium carbonate precipitation has become a new construction material for soil improvement. This paper presents a new technique for dust control in sand using Streptomyces microflavus induced calcium carbonate precipitation. This kind of carbon-capturing bacteria can capture and transform CO2 into carbonate then precipitate calcium carbonate crystals through the microbial carbonization process, which then acts as cementitious materials to bind sand particles. In this way, loose sand particles can be bridged and increase their hardness, consequently forming a consolidated layer for dust control. The Shore hardness of the consolidated layer using Streptomyces microflavus could be improved to 25 HD with a calcium carbonate content of 5.5% after 7 times of carbonization treatment. The microstructure, Shore hardness, and obtained calcium carbonate content were also compared with sand samples treated by Paenibacillus mucilaginosus. These results reveal that the consolidated sand layer using Streptomyces microflavus is sufficient to against wind erosion or rainfall erosion and has better geomechanical performance than Paenibacillus mucilaginosus.

Feed cost is the greatest expense during cattle production; therefore, reducing it is critical to increasing producer profits. In ruminants, the microbial population is important to nutrient digestion and absorption in the rumen. The objective of this study was to investigate the relationships among rumen bacteria, rumen metabolites, and the residual feed intake (RFI) phenotype of beef cattle. Twelve Angus heifers were selected to be sampled and divided into high RFI (HRFI; n = 6) group and low RFI (LRFI; n = 6) group according to their RFI classification determined during the feedlot-finishing period. After the ruminal liquid samples were collected at slaughter, Illumina MiSeq sequencing of the 16S rRNA V3-V4 region and liquid chromatography-mass spectrometry (LC-MS) were performed to determine their bacterial composition and metabolites, respectively. At the phylum level, the relative abundance of Proteobacteria was higher in the LRFI group than in the HRFI group (P < 0.01). At the family level, the relative abundances of Rikenellaceae (P < 0.01), Ruminococcaceae, Bacteroidales_S24-7_group, and Lachnospiraceae (P < 0.05) were significantly higher in the LRFI group. At the genus level, the relative abundances of Rikenellaceae_RC9_gut_group and Ruminiclostridium_1 were higher in the LRFI group (P < 0.01), as were the relative abundances of norank_f__Bacteroidales_S24-7_group, Lachnospiraceae_ND3007_group, and Lachnospiraceae_NK3A20_group (P < 0.05). Moreover, the genera Rikenellaceae_RC9_gut_group, Ruminococcaceae_NK4A214_group, Christensenellaceae_R-7_group, Ruminococcaceae_UCG-010, Lachnospiraceae_ND3007_group, Ruminiclostridium_1, and Lachnospiraceae_NK3A20_group were negatively associated with the RFI; both foundational and key species are associated with feed efficiency phenotype. In addition, rumen metabolomics analysis revealed that the RFI was associated with significantly altered concentrations of rumen metabolites involved in protein digestion and absorption, Linoleic acid metabolism, Lysine degradation, and Fatty acid degradation. Correlation analysis revealed the potential relationships between the significantly differential ruminal metabolites and the genera ruminal bacteria. The present study provides a better understanding of rumen bacteria and metabolites of beef cattle with different RFI phenotypes and the relationships among them, which are potentially important for the improvement of beef cattle feed efficiency.

AbstractSlopes facing reservoir water often collapse, affecting reservoir capacity. It is important to understand the factors affecting such collapses and predict the locations of collapse after impoundment for practical purposes. However, the mechanism of bank collapse in mountainous areas after reservoir impoundment remains unclear. To address this issue, field geotechnical property surveys, laboratory tests, and numerical simulations were employed to evaluate bank collapse susceptibility. The results showed that (i) bank collapse mainly occurs in slopes with a small cohesion and internal friction angle of less than 30°; (ii) the main effects of wave action on bank collapse are scour, erosion, and cyclic impact on the bank slope: wave erosion occurs in the bank slope with a wave height of less than 1.0 m, cohesion of less than 90 kPa, and slope angle of less than 30°, and wave impaction mainly occurs at the area with a wave height of more than 2.0 m and a slope angle of more than 30°; (iii) for valley reservoirs, softening of water on the slope rock and soil mainly reduces cohesion, which commonly occurs within 30 days; the cyclic action of wave impact load mainly damages the structure and reduces the internal friction angle. The influence of waves on bank collapse is mainly related to the wave height and wave velocity. The antiwave impact broken energy of rock and soil is an important index to evaluate whether bank collapse occurs in the long term, which is related to the strength, structure, and impact resistance. The present research is helpful to better understand the mechanism and evolution process of reservoir bank collapse.

AbstractThe behavioral characteristics of multiple cracks are important factors leading to material failures. However, to date, the fracturing and interactions of internal cracks have received minimal research due to the challenges in creating real internal cracks in brittle solids without incurring any damages to the surfaces. Therefore, this study investigated the fracturing processes and interactions between two parallel penny-shaped internal cracks under tensile stress conditions using both experimental and numerical simulation methods. Glass was used as the experimental material in this study. The results showed that the vertical spacing d between the two cracks was an important factor which affected both the crack interactions and the failure loads. When the d was small, attraction and coalescence occurred between the two cracks. However, when the d was large, the fracturing was observed to occur in the plane where one crack was located. A crack propagation arc appeared in all of the examined specimens. The loads of the crack initiation and the strength levels of the specimens were determined to be positively related to the crack spacing d. It was found that the larger the d was, the larger the crack initiation load and tensile strength values would be. In the present study, the crack paths in the numerical simulations based on the MTS criterion were determined to be consistent with the experiment results.

The laminated joints of transversely isotropic rock mass show strong discontinuity, inhomogeneity, anisotropy, and nonlinearity. This paper proposes a novel rigid block discrete element method (RB-DEM) to simulate the transversely isotropic rock mass. The balls in traditional DEM simulation are replaced by rigid blocks generated using FEM mesh. The contact on the laminar joints can be described accurately. The rigid block discrete element method (RB-DEM) results are consistent with the laboratory test. The effects of joint parameters, joint spacing, and stiffness ratio on the uniaxial compression properties of transversely isotropic rock mass are discussed. This study provides a novel and effective tool for the analysis of transversely isotropic rock mass.

Abstract Background The progression of ductal carcinoma in situ (DCIS) into invasive ductal carcinoma (IDC) is prevented by normal breast myoepithelial cells. Studies have suggested that EMT-associated genes were enriched in IDC in contrast to DCIS. This paper explored the relationship and potential mechanism between myoepithelial cells and EMT-associated genes in facilitating the transformation from DCIS to breast cancer. Methods EMT markers and myoepithelial phenotypic markers in IDC, DCIS, and healthy breast tissue were characterized using immunohistochemical assay. Both in vivo and in vitro models were created to mimic the various cell–cell interactions in the development of invasive breast cancer. Results We found that EMT markers were more abundant in invasive carcinomas than DCIS and adjacent normal breast tissue. Meanwhile, TGF-β1 regulated the morphology of MCF-7 (epithelial cells substitute) migration and EMT markers during the transformation from DCIS to invasive breast cancer. Additionally, TGF-β1 also regulated invasion, migration and cytokines secretion of MDA-MB-231 (myoepithelial cells substitute) and epithelial cells when co-cultured with MCF-7 both in vitro and in vivo. Conclusions In conclusion, these findings demonstrated that both EMT phenotypes and cancer-associated myoepithelial cells may have an impact on the development of invasive breast cancer.

Capsaicin (CAP) and Yucca schidigera extract (YSE) are two types of plant extracts that can change rumen fermentation. This study was conducted to investigate whether supplementation of beef cattle diets with CAP and YSE for 90 days would affect rumen fermentation and microflora. Forty-five healthy Angus steers (initial body weight = 510.54 ± 41.27 kg) were divided into three groups: control (CON), CAP, and YSE. Ammonia nitrogen (NH3-N) and total volatile fatty acid (TVFA) concentrations were significantly higher in the YSE group than in the CON group and significantly lower in the CAP group than in the CON group. At the phylum level, YSE increased the relative abundances of Bacteroidota and Patescibacteria and reduced that of Bacillota. At the genus level, CAP and YSE both increased the relative abundances of genera subordinate to Bacteroidota and decreased the relative abundances of genera subordinate to Bacillota. Our study shows that YSE and CAP have different effects on rumen fermentation and microflora after long-term supplementation.

The wildfire prediction model is crucial for accurate rescue and rapid evacuation. Existing models mainly adopt regular grids or fire perimeters to describe the wildfire landscape. However, these models have difficulty in explicitly demonstrating the local spread details, especially in a complex landscape. In this paper, we propose a wildfire spread model with an irregular graph network (IGN). This model implemented an IGN generation algorithm to characterize the wildland landscape with a variable scale, adaptively encoding complex regions with dense nodes and simple regions with sparse nodes. Then, a deep learning-based spread model is designed to calculate the spread duration of each graph edge under variable environmental conditions. Comparative experiments between the IGN model and widely used fire simulation models were conducted on a real wildfire in Getty, California, USA. The results show that the IGN model can accurately and explicitly describe the spatiotemporal characteristics of the wildfire spread in a novel graph form while maintaining competitive simulation refinement and computational efficiency (Jaccard: 0.587, SM: 0.740, OA: 0.800).

A mobile laser scanning (MLS) system can acquire railway scene information quickly and provide a data foundation for regular railway inspections. The location of the catenary support device in an electrified railway system has a direct impact on the regular operation of the power supply system. However, multi-type support device data accounts for a tiny proportion of the whole railway scene, resulting in its poor characteristic expression in the scene. Therefore, using traditional point cloud filtering or point cloud segmentation methods alone makes it difficult to achieve an effective segmentation and extraction of the support device. As a result, this paper proposes an automatic extraction algorithm for complex railway support devices based on MLS point clouds. First, the algorithm obtains hierarchies of the pillar point clouds and the support device point clouds in the railway scene through high stratification and then realizes the noise that was point-cloud-filtered in the scene. Then, the center point of the pillar device is retrieved from the pillar corridor by a neighborhood search, and then the locating and initial extracting of the support device are realized based on the relatively stable spatial topological relationship between the pillar and the support device. Finally, a post-processing optimization method integrating the pillar filter and the voxelized projection filter is designed to achieve the accurate and efficient extraction of the support device based on the feature differences between the support device and other devices in the initial extraction results. Furthermore, in the experimental part, we evaluate the treatment effect of the algorithm in six types of support devices, three types of support device distribution scenes, and two types of railway units. The experimental results show that the average extraction IoU of the multi-type support device, support device distribution scenes, and railway unit were 97.20%, 94.29%, and 96.11%, respectively. In general, the proposed algorithm can achieve the accurate and efficient extraction of various support devices in different scenes, and the influence of the algorithm parameters on the extraction accuracy and efficiency is elaborated in the discussion section.

The unique high/cold environment of the Qinghai–Tibet Plateau (QTP) limits the natural distribution of the population living there and threatens local residents’ health. Thus, exploring the quality of human settlements in this area is of great significance. In this study, 5 first-level indicators and 25 second-level indicators were initially selected, and the entropy TOPSIS method was used to determine the weight of each indicator and evaluate the quality of the human settlements in each county of the QTP. Then, the coefficient of variation and spatial autocorrelation were used to analyze the spatial differences in human settlement quality. Finally, the obstacle degree model was used to identify those obstacles that affect the quality of the human settlements in the QTP. This study has gathered important findings. (1) The human settlement quality in these counties can be divided into 18 high-level areas, 45 mid- and high-level areas, 44 mid-level areas, 79 mid- and low-level areas, and 28 low-level areas. (2) In terms of spatial patterns, the north is higher than the south, the east is slightly higher than the west, and the surrounding area is higher than the interior. (3) In the clustering model, the high–high clustering trend is mainly concentrated in the north of the QTP, whereas the south-central part of the QTP and the zone where Tibet, Qinghai, and Sichuan meet exhibit obvious low–low clustering. (4) The variability of human settlement quality occurs in the order of Sichuan < Yunnan < Gansu < Xinjiang Autonomous Region < Tibet Autonomous Region < Qinghai. (5) The main first-level obstacles affecting human settlement quality in the counties of the QTP are living conditions, construction level of public service facilities, and infrastructure. The main second-level obstacles are the number of living service facilities, the number of residential districts, and the density of the road networks.

Abstract Background Adherent junction associated protein 1 (AJAP1), a typical molecule of adherent junctions, has been found to be a tumor suppressor in many cancer types. Aberrant activation of β-catenin has been demonstrated to be associated with malignant biological properties of tumors including breast cancer. This study aimed to investigate the function and mechanism of AJAP1-mediated β-catenin activity of breast cancer lines in vitro and in breast cancer patients. Methods AJAP1 and β-catenin expressions in breast cancer tissues and cell lines were detected by immunohistochemistry, western blotting and qRT-PCR. The EGF/EGFR axis-mediated AJAP1 attenuated β-catenin nuclear location was measured by western blotting, immunofluorescence assay, co-immunoprecipitation, luciferase assay and ubiquitination assays. Furthermore, the function of AJAP1 and β-catenin regulated breast cancer progression was explored both in vivo and in vitro. Results It was found that AJAP1 had a high negative correlation with β-catenin nuclear expression and was a novel tumor suppressor in breast cancer. AJAP1 loss can mediate β-catenin accumulated in cytoplasm and then transferred it to the nucleus, activating β-catenin transcriptional activity and downstream genes. Additionally, β-catenin can reverse the invasion, proliferation ability and tumorigenicity of the depletion of AJAP1 caused both in vivo and in vitro. Besides, EGF/EGFR also involved in the process of AJAP1-depiction induced β-catenin transactivation to the nucleus. More importantly, EGFR depletion/AJAP1 knocked down promoted the progression of breast cancer by regulating the activity of β-catenin nuclear transactivation. Conclusion This study demonstrated that AJAP1 acted as a putative tumor suppressor while β-catenin nuclear localization positively fed back on EGF/EGFR-attenuated AJAP1 expression in breast cancer, which might be beneficial to develop new therapeutic targets for decreasing nuclear β-catenin-mediated malignancy in breast cancer.

This study was conducted to investigate the growth performance, carcass traits, blood parameters, rumen enzymes, and fattening earnings of beef cattle when substituting corn stalk silage with corn silage or corn grain in finishing rations. Forty-five Bohai Black steers were selected and fattened in a three-phase (4 weeks-4 weeks-16 weeks) way with one of three diets based on corn silage (CS), corn stalk silage (SS) without/with equivalent corn grain supplement (SSC), respectively. During the 24-week trial, individual feed intake and body weight were recorded every four weeks. By the end, blood and rumen fluid were sampled, and all the cattle were slaughtered to evaluate carcass performance. There were no significant differences found in the body weight gain, daily feed intake or feed efficiency among different dietary treatments over the whole finishing period except that the cattle fed CS achieved higher weight gain and feed efficiency in Phase 2 than those fed SS or SSC along with a lower feed intake than that of cattle fed SSC. No significant effect was found on the hot carcass weight, chilled carcass weight, dressing percentage, aging loss, loin eye area, and the weights of chuckeye, ribeye, striploin, and tenderloin. The cattle fed CS showed lower blood concentrations of ALT and glucose along with a higher ratio of AST and ALT than those fed SS. The cattle fed SSC also presented a higher activity of avicelase in the rumen fluid and their fattening earnings were approximately $27.50 less than those of the cattle fed CS or SS. These results suggest that substituting corn stalk silage with corn silage or corn grain could not improve animal performance and it is more economical to substitute corn stalk silage with corn silage rather than supplement equivalent corn grain in a high-concentrate finishing ration.

Rock resistant coefficient is a key parameter in rock engineering and can reflect the resistance of surrounding rocks. According to the load-structure method, there are many different formulas in the determination of rock resistant coefficient using elasticity or plasticity theory. Based on the generalized Hoek-Brown failure criterion, this paper develops a closed-form formula for calculating the rock resistant coefficient. The sensibility of different parameters on the rock resistant coefficient is also analyzed. The fact shows that the formula is sensitive to a, σc, and Kd comparing with mb and s especially when they are in a low value, laying a foundation for the determination of rock resistant coefficient in similar lining tunnels.

An earthquake-induced landslide, mainly affected by seismic movement, is a frequent large-scale geological hazard in hydraulic engineering. This paper proposed a rate-dependent strain-softened micromechanical contact model and implemented it in discrete element method code, namely, PFC. Using the PFC-FLAC coupling scheme, the Hongshiyan earthquake landslide is analyzed as a case study. The influence of the strain rate, damping, and topographic effect is discussed. The results indicate that the rate-dependent micromechanical model can give a reasonable seismic-induced failure process compared with the in situ situation and provide a numerical technique for earthquake-induced landslide analysis and rockfall hazard prediction.

Because of complex mechanical properties and deformation mechanism, gravel soil mixture is an important medium of concern in the field of geotechnical engineering. Based on the continuous-discontinuous coupling algorithm formed by the structure shell elements and the particle discrete element method (PFC3D), the soft servo loading of the sample is realized, and the typical triaxial compression test data are used to calibrate mesoscale parameters. Furthermore, the numerical tests under different rock contents and confining pressure are carried out. The change rule of shear strength of mixed medium is discussed. It shows that the continuous-discontinuous coupling algorithm achieves a good effect in reflecting the deformation process. Under the mechanism of the flexible servo, the failure mode of the sample takes on drum-failure mode, and the internal of the damaged sample forms an obvious asymmetric X-shaped shear band. With the increase of stone content, the internal friction angle and cohesive force distribution of the sample have certain discreteness, but on the whole, the internal friction angle and cohesive force increase with the increase of stone content. The results can provide a reference for the parameter determination of mixed mediums such as gravel soil.

Exploring the compositional characteristics of rumen eukaryotic community can expand our understanding of their role in rumen function and feed efficiency. In this study, we applied metatranscriptomics to characterize the active rumen eukaryotic community (protozoa and fungi) in beef cattle (n = 48) of three breeds [Angus (AN), Charolais (CH), and Kinsella Composite (KC)] and with divergent residual feed intake (RFI). The composition of active rumen eukaryotic microbiota was evaluated based on enriched 18S rRNAs from the metatranscriptomic datasets. At the phylum level, a total of four protozoal taxa (Ciliophora, Parabasalia, unclassified SAR, and unclassified Alveolata), six fungal taxa (Neocallimastigomycota, Basidiomycota, unclassified Fungi, Mucoromycota, Ascomycota, and Chytridiomycota), and one sister group of kingdom Fungi (unclassified Opisthokonta) were detected with relative abundances higher than 0.01% and in at least 50% of animals within each breed. Among these, Ciliophora, Parabasalia, unclassified Opisthokonta, and Neocallimastigomycota were the top four active eukaryotic phyla. At the genus level, a total of 8 ciliated protozoa, 5 flagellated protozoa, 5 anaerobic fungi, and 10 aerobic fungi taxa were detected, with unclassified Trichostomatia, Tetratrichomonas, unclassified Neocallimastigaceae, and Pleurotus being the most predominant taxa of ciliated protozoa, flagellated protozoa, anaerobic fungi, and aerobic fungi, respectively. Differential abundance analysis revealed that breed had a significant effect on the phylogenetic lineages of rumen eukaryotes, and seven fungal taxa were more abundant (linear discriminant analysis score > 2 with P < 0.05) in the rumen of KC steers than in the rumen of AN and CH steers. Although principal coordinate analysis (PCoA) revealed that the ruminal active eukaryotic profiles were not distinguishable between high- and low-RFI groups, the diversity indices, including Faith’s phylogenetic diversity (PD), observed operational taxonomic units (OTUs), and Shannon index of rumen eukaryotes were higher in low-RFI steers than those in high-RFI steers. Meanwhile, the abundance of genus Entodinium and the kingdom Fungi was higher in low-RFI steers than that in high-RFI steers. This information on active rumen eukaryotic microbiota and identified differential abundance of taxa between high- and low-RFI animals suggests the possibility of improving feed efficiency through altering rumen eukaryotic microbiota.

Rock mass classification is important in preliminary design of geotechnical engineering projects. Using the columnar jointed basalt at the foundation of Baihetan Hydropower Station as an example, this paper presents a classification scheme of the columnar jointed rock. Unlike many common rock masses, an obvious characteristic of columnar jointed rock is that it is discontinuous in geometry while continuous in mechanics. Due to the inapplicability of existing rock mass classification systems, a classification scheme, combined with rock mass integrity, weak plane tightness, and permeability, is proposed. The new classification system has five grades with quantitative factors, which takes into account the features of columnar joints. As an easy-to-use scheme and case study, it would be helpful as a reference in the rock mass classification of similar problems.

Large-scale slopes at the banks of reservoirs pose a serious threat to the safety of hydropower stations. The fluctuation of the reservoir water level is a key factor in the slope stability. However, the parameters to describe the relationship among water content, matric suction, and soil strength are difficult to measure using unsaturated soil strength theory. To solve this problem, a simple FEM-LEM-combined scheme considering pore pressure, seepage force, and strength weakening is presented to calculate the safety factor. A numerical study on the impact of reservoir water level fluctuations on stability of a glaciofluvial deposit slope is implemented. Two typical profiles are used to estimate the stability of the glaciofluvial deposit slope in response to rising and lowering water levels. The results indicate that this method proposed a simple and efficient tool for water level-induced slope stability analysis.

To study the strength, deformation, and failure patterns of columnar-jointed-rock-mass (CJRM) under unloading conditions, triaxial unloading tests using the CJRM-like material samples are carried out, and acoustic wave (AW) velocities are simultaneously recorded. Based on stress-strain curves and AW velocities under different initial confining pressures and unloading rates, the stress-strain characteristics, strength, and deformation parameters, failure modes, and variation of the AW velocity are analyzed. Test results show that the CJRM may exhibit intense volume expansion during the unloading process. With the increase of the unloading and its rate, the volume expansion becomes more serious and the failure mode becomes more complicated. By reducing the unloading (rate), a phenomenon of unloading relaxation is observed and the quality of CJRM is significantly improved. The AW velocity of CJRM shows a strong correlation with the volume strain, which verifies the effectiveness of applying AW velocity for assessing the rock quality. It is hoped that the research results may provide a reference for the construction and operation of the Baihetan Hydropower Project.

Abstract Background Androgen receptor (AR) is expressed in 60%~ 70% oestrogen receptor (ER)-negative breast cancer (BC) cases and promotes the growth of this cancer subtype. Expression of prostate-derived Ets factor (PDEF), a transcription factor, is highly restricted to epithelial cells in hormone-regulated tissues. MYC and its negative regulator MAD1 play an important role in BC progression. Previously, we found that PDEF expression is strongly correlated with AR expression. However, the relationship between AR and PDEF and the function of PDEF in ER-negative BC proliferation are unclear. Methods AR and PDEF expression in ER-negative BC tissues and cell lines was determined by performing immunohistochemistry or western blotting. Protein expression levels and location were analysed by performing western blotting, RT-qPCR and immunofluorescence staining. Co-immunoprecipitation and chromatin immunoprecipitation assays were performed to validate the regulation of AR–PDEF–MAD1–MYC axis. Moreover, the effect of AR and PDEF on BC progression was investigated both in vitro and in vivo. Results We found that PDEF was overexpressed in ER-negative BC tissues and cell lines and appeared to function as an oncogene. PDEF expression levels were strongly correlated with AR expression in ER-negative BC, and PDEF transcription was positively regulated by AR. PDEF upregulated MYC-mediated gene transcription by promoting MAD1 degradation in ER-negative BC. Finally, we found that compared with the inhibition of AR expression alone, simultaneous inhibition of AR and PDEF expression further suppressed tumour proliferation both in vitro and in vivo. Conclusions Our data highlight the role of the AR–PDEF–MAD1–MYC axis in BC progression and suggest that PDEF can be used as a new clinical therapeutic target for treating ER-negative BC.

Six rumen-fistulated Limousin×Jinnan steers were fed diets supplemented with either 0% (0NR), 1% (1NR), or 2% (2NR) nitrate (dry matter basis) regimens in succession. Rumen fluid was taken after two-week adaptation for evaluation of in vitro methane production, methanogen abundance, and composition measurements. Food security is essential for human survival and sustainable development. Due to rapid urbanization and industrialization, the farmland loss in Yangtze River Delta Urban Agglomeration (YRDUA) has threatened food security. Thus, this study intended to quantify the farmland loss and assess its impacts on food security in the YRDUA from 2000 to 2020 at three scales based on the Google Earth Engine platform. Our results show that the area of farmland in YRDUA is decreasing at a rather high speed in the past 20 years and the trend is getting worse. At the urban agglomeration scale, there was a farmland deficit of 0.44 million ha in 2000, followed by larger farmland deficits in 2010 and 2020. At the city scale, Shanghai had the largest scarcity of farmland. At the urban subgroup scale, Subgroup I in the west and Subgroup II in the north always maintained an oversupply of farmland, while Subgroup III in the east and Subgroup IV in the south faced serious food security problems. Our study suggests that farmland must be protected in YRDUA in order to ensure food self-sufficiency and promote regional sustainability.

The continuous supply of ecosystem services is the foundation of the sustainable development of human society. The identification of the supply–demand relationships and risks of ecosystem services is of considerable importance to the management of regional ecosystems and the effective allocation of resources. This paper took the Yihe River Basin as the research area and selected water yield, carbon sequestration, food production, and soil conservation to assess changes in the supply and demand of ecosystem services and their matching status from 2000 to 2018. Risk identification and management zoning were also conducted. Results show the following: (1) The spatial distribution of the four ecosystems service supply and demand in the Yihe River Basin was mismatched. The food production supply levels in the middle and lower reaches and the upstream water yield, carbon sequestration, and soil conservation supply levels were high. However, most of the areas with high demand for ecosystem services were concentrated downstream. (2) From 2000 to 2018, the supply of water yield and carbon sequestration in the Yihe River Basin decreased, while that of food production and soil conservation increased. The demand for the four ecosystem services also increased. (3) Water yield faced considerable supply–demand risks. Fifty percent of the sub-basins were at a high-risk level, and the risk areas were concentrated in the middle and lower reaches. The three remaining services were mainly at low-risk levels. The Yihe River Basin was divided into eight types of supply–demand risk spatial management zones based on the ecosystem service supply and demand levels, which will help promote refined regional ecosystem management and sustainable development. The supply and demand assessment of ecosystem services from a risk perspective can integrate the information of natural ecosystems and socio-economic systems and provide scientific support for watershed spatial management.

These experiments were conducted to evaluate the effect of excessive sulfur on rumen fermentation, microflora, and epithelial barrier function in steers through in vitro gas production and animal feeding experiments. Nine and four levels of sulfur addition were evaluated in in vitro ruminal fermentation and animal feeding experiment, respectively. The results showed that increasing the level of sulfur in substrates decreased the total gas and methane production linearly, while increasing the production of hydrogen sulfide gas (p < 0.01). Volatile fatty acid concentrations, especially that of butyric acid, were increased by extra sulfur (p < 0.01). Sulfur content in the diet had no significant effect (p > 0.05) on most of the rumen microbes, except for Desulfovibrio, one of the major sulfate-reducing bacteria (SRB) in the rumen, whose population increased by adding extra sulfur (p < 0.001). The changes in the morphology of rumen epithelium and thickening of the total epithelial layer were mainly attributed to the increase in the acanthosis cell layer and stratum basale (p < 0.05). Further, the relative expressions of two tight junction protein regulating genes, CLDN-1 and TJP1, were reduced (p < 0.05). Excessive sulfur in the diet can change the type of rumen fermentation, sulfate metabolism and SRB population, and the rumen epithelial barrier function. The results of this study demonstrated that sulfur can be used as a methane inhibitor with the mechanism that SRB competitively used protons to produce hydrogen sulfide. However, a higher level of sulfur in the diet could increase the inflammatory reaction of the rumen epithelium which may affect nutrient absorption.
