Low permeation flux and rapid flux decline are challenging membrane technology in wastewater treatment. Here we used a metal–ligand complexation protocol to construct novel forward osmosis (FO) membrane for efficient and sustainable phosphorus removal. Disodium and tetrasodium salts of ethylenediaminetetraacetic acid (EDTA-2Na, EDTA-4Na) were separately grafted onto the conventional polyamide (PA) membranes via chemical complexation using Fe3+ as a bridge. These EDTA salts substantially improved the membrane properties. Consequently, the water fluxes produced by the EDTA-2Na and EDTA-4Na modified membranes increased by 109% (11.1 LMH) and 140% (12.7 LMH), respectively, relative to that of the pristine membrane (5.3 LMH) with 0.5 M NaCl as the draw solution. Meanwhile, the EDTA-4Na modified membrane surpassed the naked PA membrane in performance with higher water recovery efficiencies and solute rejections after long-term experiments against a mixture of bovine serum albumin and HPO42−. Moreover, the water flux recovery ratio of the naked PA membrane was only 51%, while that of the EDTA-4Na modified membrane reached over 93% after three cycles of fouling experiments. This study provides a new paradigm to develop FO membrane via a metal–ligand complexation protocol for efficient and sustainable wastewater treatment. Novel FO membranes synthesized through a complexation protocol are used for phosphorus removal and fouling resistance in forward osmosis.

Chlorophyll (chl) degradation plays a vital role during green plant growth and development, including nutrient metabolism, fruit and seed maturation, and phototoxic detoxification. STAY-GREEN (SGR) is a plant-specific regulator involved in chl degradation. Previous studies showed that SlSGR1 functioned in chl degradation and lycopene accumulation during fruit ripening of tomato (Solanum lycopersicum). However, little is known about SlSGR-LIKE (SlSGRL) gene, which is a homolog of SlSGR1. We cloned the SlSGRL gene and created transgenic tomato plants overexpressing (OE) SlSGRL. Expression analysis showed that SlSGRL was up-regulated by abscisic acid (ABA). Our data showed that SlSGRL-OE lines exhibited earlier leaf yellowing than wild-type (WT) lines under ABA treatment. Yeast two-hybrid (Y2H) assay revealed that SlSGRL interacted with pheophytin pheophorbide hydrolase (SlPPH) and light-harvesting complex a2 (SlLHCa2) to promote the chl degradation. Further analysis demonstrated that ABA-INSENSITIVE5 (SlABI5) and SlABI5-LIKE regulated SlSGRL expression by directly binding to the sequence (-611 to -582) of the SlSGRL promoter that included an ABRE cis-element. We proposed that SlSGRL, which was regulated by SlABI5/SlABI5-LIKE, mainly acted in ABA-induced chl degradation via interacting with SlPPH and SlLHCa2.

Acids are extensively used in contemporary industries; however, time-consuming and environmentally unfriendly processes hinder their isolation from wastes containing various ionic species. While membrane technology can overcome these challenges by efficiently extracting analytes of interest, the associated processes typically exhibit inadequate ion-specific selectivity. In this regard, we rationally designed a membrane with uniform angstrom-sized pore channels and built-in charge-assisted hydrogen bond donors that preferentially conduct monobasic acids while exhibiting negligible conductance for other compounds. The ionic selectivity originates from the size screening ability of angstrom-sized channels between protons and other hydrated cations, while the built-in charge-assisted hydrogen bond donor enables the screening of acids by exerting host-guest interactions to varying extent, acting as an anion filter. Furthermore, in contrast to conventional cationic membranes, the strong hydrogen-bonding interactions between the guanidinium moieties and their counter anions resulted in charge transfer from the anions to the membrane skeleton, which switched the effective pore surface charge from positive to negative, thereby facilitating the transport of protons. Ion transport measurements indicated that the resulting membrane exhibited exceptional permeation for protons over other cations and anions of monobasic acids over those of polybasic acids with selectivities up to 4334 and 183, respectively, demonstrating prospects for monobasic acid extraction from waste streams. These findings will aid the design of advanced multifunctional membranes for sophisticated separation.

A vast amount of energy can be extracted from the untapped low-grade heat from sources below 100 °C and the Gibbs free energy from salinity gradients. Therefore, a process for simultaneous and direct conversion of these energies into electricity using permselective membranes was developed in this study. These membranes screen charges of ion flux driven by the combined salinity and temperature gradients to achieve thermo-osmotic energy conversion. Increasing the charge density in the pore channels enhanced the permselectivity and ion conductance, leading to a larger osmotic voltage and current. A 14-fold increase in power density was achieved by adjusting the ionic site population of covalent organic framework (COF) membranes. The optimal COF membrane was operated under simulated estuary conditions at a temperature difference of 60 K, which yielded a power density of ≈231 W m

Rubisco, which consists of eight large subunits (RBCLs) and eight small subunits (RBCSs), is a major photosynthetic enzyme that is sensitive to chilling stress. However, it is largely unclear how plants maintain high Rubisco content under low temperature conditions. Here, we report that tomato WHIRLY1 (SlWHY1) positively regulates the Rubisco level under chilling stress by directly binding to the promoter region of SlRbcS1, resulting in the activation of SlRbcS1 expression. SlRbcS1-overexpressing lines had higher Rubisco contents and were more resistant to chilling stress compared with the wild type. Quantitative real-time PCR analyses showed that, among the five RbcS genes, only SlRbcS1 expression is up-regulated by chilling treatment. These results indicate that SlWHIRLY1 specifically enhances the levels of SlRbcS1 and confers tolerance to chilling stress. The amino acid sequence of SlRBCS1 shows 92.67% identity with those of another two RBCS proteins and three residues are specifically found in SlRBCS1. However, mutation of these residues to alanine in SlRBCS1 does not influence its function during cold adaptation. Thus, we conclude that high levels of Rubisco, but not the specific residues in SlRBCS1, play important roles in tolerance to chilling stress in tomato.

Soil deficiencies with respect to the availability of macronutrients such as nitrogen, phosphorus, and potassium seriously affect the growth, yield, and grain quality of maize (Zea mays L.). To improve the utilization efficiency of mineral elements in maize, we wanted to find the key genes that regulate the growth of maize roots under nutrient-deficient conditions. Maize plants were subjected to nitrogen, phosphorus, and potassium deficiency stress and their roots were collected and analyzed using transcriptome sequencing. GO and KEGG analyses of the differentially expressed genes (DEGs) were performed, and qPCR was used to verify the reliability of the transcriptome data. When maize was subjected to any of the three nutrient-deficiencies mentioned in Methods, the growth and root vitality of its roots were inhibited. 1255, 1082, and 324 genes specifically expressed when the maize was subjected to N, P, and K deficiencies, respectively, and all three treatments shared 575 DEGs. Genes that are associated with nutrient utilization, hormones, and transcription factors differentially expressed under different types of nutrient-deficiency stress. We speculated that MRP2, bZIP77, and bZIP53 play a positive regulatory role in maize root growth in an environment suffering from nutrient deficiencies. The molecular mechanism by which maize root growth responds to nutrient stress is complicated. NPF7.3, GlpT4, HAK24, and HAK5, MRP2, bZIP77, and bZIP53 can be used as candidates’ genes that regulate maize root growth under nitrogen, phosphorus, and potassium deficiency stress.

In plants, C2H2-type zinc finger proteins play important roles in multiple processes, including plant growth and development, as well as biotic and abiotic responses. In the present study, based on the presence of the C2H2 domain (CX2~4CX3FX5LX2HX3~5H), 112 C2H2-type zinc finger proteins were predicted in tomato. Through gene and protein structures analyses and phylogenetic analysis, the 112 C2H2-type zinc finger proteins were divided into five subfamilies. Members of the same subfamily shared similarities in gene and protein structures, while members of different subfamilies contained different numbers of the C2H2 domain. The tissue expression pattern analysis showed that 24 C2H2-type zinc finger proteins are constitutively expressed in all tissues, indicating that they may play important roles in the growth and development of all tissues. In addition, under chilling (4 °C), heat (42 °C), high salinity (200 Mm NaCl), and osmotic (20% PEG) stresses, members of C2H2-type zinc finger family were induced to varying degrees, which suggested that these genes were involved in multiple abiotic stress responses. This study will provide theoretical basis for further research of C2H2-type zinc finger proteins in tomato.

Water contaminated with low concentrations of pollutants is more difficult to clean up than that with high pollutant content levels. Membrane separation provides a solution for removing low pollutant content from water. However, membranes are prone to fouling, losing separation performances over time. Here we synthesized—a neutral (IM-NH2) and positively-charged (IL-NH2) imidazole derivatives to chemically functionalize membranes. With distinct properties, these imidazole grafts could tailor membrane physicochemical properties and structures to benefit forward osmosis (FO) processes for the removal of 20 − 100 ppm of Safranin O dye − a common dye employed in the textile industry. The water fluxes produced by IM-NH2 and IL-NH2 -modified membranes increased by 67% and 122%, respectively, with DI water as the feed compared to that with the nascent membrane. A 39% flux increment with complete dye retention (∼ 100%) was achieved for the IL-NH2-modified membrane against 100ppm of Safranin O dye. Regardless of the dye concentration, the IL-NH2 -modified membrane exhibited steadily higher permeation performance than the original membrane in long-term experiments. Reproducible experimental results were obtained with the IL-NH2 -modified membrane after cleaning with DI water, demonstrating the good antifouling properties and renewability of the newly developed membrane.

With the advancement of tumor subtype-specific treatments, precise histopathologic distinction between adenocarcinoma (ADC) and squamous cell carcinoma (SCC) is of significant clinical importance. Nevertheless, the current markers are insufficiently precise in poorly differentiated tissue. This study aimed to establish a histology-specific immunomarker combination to subclassify non-small cell lung cancer (NSCLC) specimens. Based on previous work, we assessed the differential expression of anterior gradient 2 (AGR2) and keratin 5 (KRT5) in ADC and SCC by analyzing public datasets and postoperative specimens. Subsequently, we established a train set (n = 188) and a validation set (n = 42) comprised of NSCLC surgical specimens for training and verifying the subtype-identification capabilities of the two biomarkers separately and in combination, and contrasted the diagnostic utility of AGR2-KRT5 with that of the classic immunomarker combination, TTF1-P40. Differential expression of the two genes was statistically significant in ADC and SCC samples, both at the mRNA and protein levels. The specificity and sensitivity of AGR2 to detect ADC in the training set were 97.0% and 94.4%, while the sensitivity and specificity of KRT5 to determine SCC were 93.9% and 98.9%, respectively. The accuracies of AGR2-KRT5 in ADC, SCC, and across all samples were 93.3%, 92.0% and 92.6% respectively. In the validation cohort, the predictive accuracy of AGR2-KRT5 was up to 100% for ADC and 86.7% for SCC. Compared with TTF1-P40 in ADC samples, AGR2-KRT5 had 8.4% higher accuracy. In summary, the AGR2-KRT5 immunomarker combination reliably distinguished SCC from ADC, and was more accurate than TTF1-P40 in ADC.

Chlorophyll (Chl) degradation is one of the most obvious signs of leaf senescence and fruit ripening. Stay-green (SGR) homologs that can remove magnesium from Chl a are the most important components in Chl degradation pathway in green plants. SGR homologs are not only universally involved in Chl breakdown during the senescence of green organs, but also play crucial roles in other organs during plant growth and development, such as fruit mature and nodule development. In this review, we focus on the diverse functions of SGR homologs in plant growth and development. A better understanding of SGR would be helpful for providing a theoretical basis for further illustrating the regulatory mechanism of SGR homologs.

Drought stress in plants leads to inhibition of photosynthesis and respiration, accumulation of reactive oxygen species (ROS), and reprogramming of gene expression. Here, we established that the tomato (Solanum lycopersicum) WHIRLY2 (SlWHY2) gene, which encodes a mitochondrial single-stranded DNA-binding protein, was significantly induced by drought stress. Under drought conditions, SlWHY2 RNAi plants showed more wilting and lower fresh weight, chlorophyll content, quantum yield of photosystem I (PSI; YI), and maximal photochemical efficiency of PSII (Fv/Fm) than the wild type (WT). Drought treatment also caused the SlWHY2 RNAi lines to accumulate more ROS than the WT, and the silenced lines had lower AOX (alternative oxidase) activity. As expected, the mitochondrial membrane potential (MMP) was less stable in the SlWHY2 RNAi lines. The expression levels of seven genes in the mitochondrial genome (SYCF15, NAD7, NAD4, COS2, COX1, COX2, and COX3) were decreased even more in the SlWHY2 RNAi lines than they were in the WT under drought stress. SlWHY2 interacted directly in vivo and in vitro with SlRECA2, a mitochondrial recombinase A that is important for mitochondrial DNA recombination and repair. These results suggest that SlWHY2 plays an essential role in maintaining mitochondrial function and enhancing drought tolerance in tomato.

Broad-Complex (BrC) is a downstream target of both 20-hydroxyecdysone and juvenile hormone signalling. BrC regulates morphogenetic changes between nymphal instars in hemimetabolans, whereas it controls pupal commitment, pupal morphogenesis and inhibits adult differentiation in holometabolans. Among five BrC cDNAs (Z1-Z4 and Z6) identified in the Colorado potato beetle, we found in this work that Z1, Z2 and Z6 were mainly expressed at the last (fourth) instar and prepupal stages, whereas the levels of Z3 and Z4 increased during the penultimate (third) instar stage, peaked at the last instar larval phase and gradually decreased at the prepupal and pupal periods. When knocking down all BrC isoforms by RNA interference (RNAi) at the penultimate instar stage, around 20% of the resultant larvae remained as moribund beetles. These moribund BrC RNAi larvae were completely or partially wrapped in old cuticle. Likewise, a portion of larvae treated for a single double-stranded RNA of Z3, Z4 or Z6 displayed a degree of similar aberrancies, increasing in the order of isoforms Z6

Dietary delivery of bacterially expressed double-stranded RNA (dsRNA) has a great potential for management ofLeptinotarsa decemlineata. An important first step is to discover possible RNA-interference (RNAi)-target genes effective against larvae, especially the old larvae. In the present paper, five putativeBroad-Complex(BrC) cDNAs (Z1-Z4, andZ6) were identified inL. decemlineata. The expression of the fiveLdBrCisoforms was suppressed by juvenile hormone signaling, whereas the transcription was upregulated by 20-hydroxyecdysone signaling at the fourth (final) instar larval stage. Feeding of bacterially expressed dsBrC(derived from a common fragment of the fiveLdBrCvariants) in the third- and fourth-instar larvae successfully knocked down the target mRNAs. For the fourth-instarLdBrCRNAi hypomorphs, they had a higher larval mortality compared with the controls. Moreover, most dsBrC-fed beetles did not pupate normally. After removal of the apolysed larval cuticle, a miniature adult was found. The adult head, compound eyes, prothorax, mesothorax, metathorax were found on the dorsal view. Distinct adult cuticle pigmentation was seen on the prothorax. The mouthparts, forelegs, midlegs, and hindlegs could be observed on the ventral view of the miniature adults. For the third-instarLdBrCRNAi specimens, around 20% moribund beetles remained as prepupae and finally died. Therefore,LdBrCis among the most attractive candidate genes for RNAi to control the fourth-instar larvae inL. decemlineata.

Severe environments disturb the folding or assembly of newly synthesized proteins, resulting in accumulation of misfolded or unfolded proteins in the endoplasmic reticulum (ER) as well as cytotoxic aggregation of abnormal proteins. Therefore, ER stress is evoked due to disturbed ER homeostasis. Alternative oxidase (AOX) plays an important role in coping with various abiotic stresses and plant growth. Our previous study has reported that PpAOX is involved in the regulation of salt tolerance in moss Physcomitrella patens (P. patens), but its biological functions in modulating ER stress remain unknown. Here we report that the gametophyte of P. patens displays severe growth inhibition and developmental deficiency under tunicamycin (Tm, an elicitor of ER stress)-induced ER stress conditions. PpAOX and selected ER stress response-like genes in P. patens were induced under Tm treatment. PpAOX knockout (PpAOX KO) plants exhibited decreased resistance to Tm-induced ER stress, whereas PpAOX-overexpressing lines (PpAOX OX) plants were more tolerant to Tm-induced ER stress. Data showed that PpAOX contributes to redox homeostasis under Tm treatment. In addition, we observed that PpAOX completely restores the Tm-sensitive phenotype of Arabidopsis AOX1a mutant (Ataox1a). Taken together, our work reveals a functional link between PpAOX and ER stress tolerance regulation in P. patens.

The NAC (NAM, ATAF1/2, and CUC2) transcription factor family participates in responses to various kinds of environmental stimuli in plants. However, the roles of NAC protein in cold resistance, especially in the cold resistance of tomatoes, are not completely understood. This study examined the roles of a tomato (Solanum lycopersicum) NAC transcription factor (SlNAC35) in resisting chilling using transgenic tomatoes. GUS staining and expression analysis revealed that SlNAC35 expression was induced at 4 °C, thereby suggesting its involvement in plant responses to chilling stress. Moreover, transgenic lines over-expressing SlNAC35 exhibited high chlorophyll content, fresh mass, and low accumulation of reactive oxygen species and membrane damage under chilling stress. These results indicated that SlNAC35 overexpression enhanced the chilling tolerance of transgenic tomatoes. High expressions of cold tolerance markers SlCOR518 and SlCOR413IM1 were observed under chilling stress in transgenic lines. This observation suggested that SlNAC35 overexpression enhanced the chilling tolerance of transgenic lines by involving the c-repeat binding factor-cold stress response (CBF-COR) signaling pathway and by regulating SlCOR expression.

Some insect growth disruptors (IGDs), such as pyriproxyfen and halofenozide, may be used to control Leptinotarsa decemlineata. However, their mechanism of action remains elusive. Methoprene-tolerant (Met) mediates juvenile hormone (JH) signal to control numerous essential physiological processes. In the present paper, we identified a Met gene (LdMet). LdMet protein was a typical basic helix-loop-helix/Per-Arnt-Sim (bHLH-PAS) transcription factor with a bHLH domain, two PAS domains (PAS-A and PAS-B) and a region called PAS associated C terminal (PAC). Eight conserved amino acids critical for JH binding were located in PAS-B and PAC domains. The temporal expression pattern of LdMet was in accordance with the variation of circulating JH titers. Feeding of juvenoid methoprene or pyriproxyfen, or provide for JH dose-dependently stimulated the expression of LdMet. RNA interference-mediated knockdown of two JH degradation genes increased the transcription of LdMet, while silencing of a JH biosynthesis gene repressed the transcription. Conversely, ingestion of an ecdysteroid agonist halofenozide or 20-hydroxyecdysone (20E) reduced the mRNA levels of LdMet, in a dose-dependent manner, whereas knockdown of either ecdysteroidogenesis or 20E signaling genes increased the mRNA accumulation. Providing that the expression of LdMet can be disturbed by methoprene, pyriproxyfen and halofenozide, LdMet may be a potential target of these IGDs in L. decemlineata larvae.

In the tobacco hornworm Manduca sexta, juvenile hormone (JH) is critical for the control of species-specific size. However, whether the basic helix-loop-helix/Per-Arnt-Sim domain receptor methoprene-tolerant (Met) is involved remains unconfirmed. In the present paper, we found that RNA interference (RNAi)-aided knockdown of Met gene (LdMet) lowered the larval and pupal fresh weights and shortened the larval development period in the Colorado potato beetle Leptinotarsa decemlineata. Dietary introduction of JH into the LdMet RNAi larvae rescued neither the decreased weights nor the reduced development phase, even though JH ingestion by control larvae extended developmental time and caused large pupae. Moreover, the transcript levels of five genes involved in prothoracicotropic hormone and cap 'n' collar isoform C/Kelch-like ECH associated protein 1 pathways were upregulated in the LdMet silenced larvae. Ecdysteroidogenesis was thereby activated; 20-hydroxyecdysone (20E) titer was increased; and 20E signaling pathway was elicited in the LdMet RNAi larvae. Therefore, JH, acting through its receptor Met, inhibits PTTH production and release before the attainment of critical weight. Once the critical weight is reached, JH production and release are averted; and the hemolymph JH is removed. The elimination of JH allows the brain to release PTTH. PTTH subsequently stimulates ecdysteroid biosynthesis and release to start larval-pupal transition in L. decemlineata.

Two Drosophila melanogaster E-twenty-six domain transcription factor isoforms (E74A and E74B) act differentially at the start of the 20-hydroxyecdysone (20E) signalling cascade to regulate larval-pupal metamorphosis. In the present paper, we identified the two isoforms (LdE74A and LdE74B) in Leptinotarsa decemlineata. During the larval development stage, the mRNA transcript levels of the two LdE74 isoforms were correlated with circulating 20E titres. In vitro midgut culture and in vivo dietary supplementation with 20E revealed that the presence of 20E induced expression peaks of both LdE74A and LdE74B, with similar patterns observed for the two isoforms. Moreover, the mRNA transcript levels of both LdE74A and LdE74B isoforms were significantly downregulated in the L. decemlineata ecdysone receptor RNA interference (RNAi) specimens, but not in the LdE75 RNAi beetles. Ingestion of 20E reduced the larval fresh weights and shortened the larval development period, irrespective of knockdown of LdE74 or not. RNAi of LdE74 did not affect 20E-induced expression of the Ecdysone induced protein 75-hormone receptor 3-fushi tarazu factor 1 (E75-HR3-FTZ-F1) transcriptional cascade. Thus, it seems that LdE74 mediates 20E signalling independent of the E75-HR3-FTZ-F1 transcriptional cascade. Furthermore, silencing of both LdE74 isoforms caused failure of ecdysis. Most of the LdE74 RNAi beetles remained as prepupae. The LdE74 RNAi prepupae exhibited adult character-like forms underneath after removal of the apolysed larval cuticle. Their appendages such as antennae, legs and wings were shorter than those of control larvae. Only a few LdE74 RNAi larvae finally became deformed pupae, with shortened antennae and legs. Therefore, LdE74 is required for larval-pupal metamorphosis and appendage growth in L. decemlineata.

The potential of teflubenzuron was assessed in a series of laboratory studies in order to achieve consistent, long-term, integrated management of the Colorado potato beetle, Leptinotarsa decemlineata (Say). Teflubenzuron exhibited excellent stomach toxicity to the larvae. Its larvicidal activity was comparable with those of cyhalothrin, chlorantraniliprole and spinosad. Moreover, the teflubenzuron-fed larvae consumed less foliage, grew slower, and needed a longer period to develop, in a dose dependent manner. Most of these larvae died during larval-larval molting, larval-pupal ecdysis or adult emergence. Furthermore, chitin contents in body carcass (without midgut) and integument specimen of the teflubenzuron-fed larvae significantly decreased, whereas the chitin amount in the midgut peritrophic matrix was not affected. In addition, uridine diphosphate-N-acetylglucosamine-pyrophosphorylase gene (LdUAP1), which was mainly responsible for chitin biosynthesis in ectodermally-derived tissues, was suppressed after teflubenzuron ingestion, in contrast to its partner LdUAP2 for chitin formation in the midgut peritrophic matrix. In a word, by inhibition of chitin production in ectodermally-derived tissues, teflubenzuron is an effective benzoylurea insecticide to L. decemlineata larvae. It can be a valuable tool in effective integrated pest management and insecticide resistance management programs against L. decemlineata.

WHIRLY transcription factors play critical roles in responses to biotic and abiotic stresses, but their other biological functions remain unclear. In this study, SlWHY2, a member of the WHIRLY family, was isolated from Solanum lycopersicum. The role of SlWHY2 was studied using transgenic tobacco plants. Real-time quantitative polymerase chain reaction analysis showed that SlWHY2 expression was induced by polyethylene glycol, NaCl, salicylic acid, hydrogen peroxide, and bacterial pathogens. SlWHY2 overexpression in tobacco caused enhanced tolerance to drought stress, as indicated by lower accumulation of malondialdehyde and relative electrolyte leakage, as well as higher relative water content and activities of superoxide dismutase and ascorbate peroxidase. Moreover, higher expression of cytochrome oxidase 1 (NtCOX1) and open reading frame 1 (NtORF1) were observed under drought in the transgenic lines. This suggested that overexpression of SlWHY2 enhanced tolerance to drought stress by regulating the transcription of mitochondrial genes and stabilizing mitochondrial function. Transgenic tobacco also displayed greater resistance to Pseudomonas solanacearum infection as evidenced by lower reactive oxygen species content and higher expression of defence-related genes. Overall, these findings suggest that SlWHY2 acts as a positive regulator in response to biotic and abiotic stresses.

Condenser tube tubes are widely applied in condenser in thermal power plants. Leakage often occurs in condenser tube groups due to the quality of condenser media or high serving stress. Condenser tube leakage not only brings economic losses to power stations, but even influences the safety operation of the power generation units. This paper simulates and experimentally studies the ferromagnetic heat exchanger tubes based on the detection mechanism of far field eddy current. The results show that the position of the far field is with the increase of the wall thickness and the inner diameter of the tubes, and the detection sensitivity of the far field is high for the same size defect, but the difference of the magnetic flux intensity varies equally when the defects with the same size are located at the inner and outer walls. This is consistent with the experimental results of real tube defect detection and meets the principle of far field eddy current detection. It is concluded that far field eddy current testing technology can better determine and identify ferromagnetic tube defects, and has a good application prospect for online nondestructive testing and safety performance evaluation of service tubes

To evaluate the efficacy and safety of Chinese medicine (CM) improving pregnancy outcomes after surgery for endometriosis-associated infertility.A multicenter, randomized, double-blind placebo parallel controlled clinical trial was designed. A total of 202 patients who had laparoscopy for endometriosis-associated infertility with qi stagnation and blood stasis syndrome were included and randomly divided into the CM treatment group and placebo control group at a ratio of 1:1 using a central block randomization from May 2014 to September 2017, 101 patients in each group. The two groups received continuous intervention at 1-5 days after surgery, for 6 menstrual cycles. Before ovulation, the CM group was treated Huoxue Xiaoyi Granule (); after ovulation, Bushen Zhuyun Granule ( was involved. The control group was treated with placebo. Transvaginal ultrasonography was performed every menstrual cycle during the treatment, and female hormone levels in the follicular and luteal phases were measured during the 1st, 3rd and 6th menstrual cycles. The analysis was continued until pregnancy. The primary outcomes were clinical pregnancy rate and pregnancy outcome, and the secondary outcomes were follicular development and endometrial receptivity. Safety evaluations were performed before and after treatment.(1) Clinical pregnancy and live birth rates: the clinical pregnancy and live birth rates of the CM group were significantly higher than those of the placebo group [44.6% (45/101) vs. 29.7% (30/101), 34.7% (35/101) vs. 20.8% (21/101), both P0.05]. (2) Follicle development: the incidence of dominant follicles, rate of cumulative cycle ovulation, and rate of cumulative cycle mature follicle ovulation were significantly higher in the CM group than those in the placebo group [93.8% (350/373) vs. 89.5% (341/381), 80.4% (275/342) vs. 69.1% (253/366), 65.8% (181/275) vs 56.1% (142/253), P0.05 or P0.01]). The incidence of cumulative cycle luteinized unruptured follicle syndrome was significantly lower in the CM group than in the placebo group [11.7% (40/342) vs. 17.8% (65/366), P0.05). (3) Endometrial receptivity: after treatment, both endometrial types and endometrial blood flow types in the CM group were mainly types A and B, while those in the placebo group were mainly types B and C, with a significant difference between the two groups (both P0.05). (4) Adverse events: the incidence of adverse events between the two groups was not significantly different (P0.05).Strategies for activating blood circulation-regulating Gan (Liver)-tonifying Shen (Kidney) sequential therapy can effectively improve the clinical pregnancy rate and live birth rate of endometriosis-associated infertility with qi stagnation and blood stasis after laparoscopy, improve follicular development, promote ovulation, improve endometrial receptivity, while being a safe treatment option. It is challenging to purify wastewater containing heavy metals at low concentrations. Forward osmosis (FO) is promising to remove most of contaminants from wastewater. However, the lack of suitable membranes disables FO to achieve its full potential. Here we designed positively charged PA-Fe-H2O and PA-Fe-Bet membranes for efficient chromium (CrO42-) removal through micellar-enhanced FO (MEFO) separation. These membranes were developed by functionalizing the ready-made polyamine (PA) membrane via eco-friendly complexation between Fe3+ and carboxyl groups of the PA layer and/or betaine (Bet). A Fe3+-bridged hydration or quaternary ammonium layer was formed which imbued the membrane with desirable properties for FO separation. The water permeability of the PA-Fe-H2O and PA-Fe-Bet membranes was increased by 92% and 142%, respectively, compared to that of the PA membrane when 0.5 M MgCl2 as the draw solution. The PA-Fe-Bet membrane greatly improved CrO42- removal efficiency by increasing the water flux from 6.2 to 12.0 LMH along with complete CrO42- retention in the cetylpyridinium chloride (CPC) facilitated MEFO process. Moreover, this membrane exhibited enhanced antifouling properties when exposed to CPC solutions. The environmentally benign synthesis approach, high CrO42- removal efficiency and fouling resistance of the PA-Fe-Bet membrane demonstrate its potential for contaminants removal.

A series of Eu3+ or Tb3+ doped Ba2Ca(BO3)2 phosphors were synthesized by a high temperature solid state method, and the luminescence properties are investigated. Ba2Ca(BO3)2:Tb3+ can show an obvious green emission, and the peak locates at 551 nm, which corresponds to the 5D4→7F5 transition of Tb3+. Ba2Ca(BO3)2:Eu3+ can present the characteristic emission of Eu3+, and the peak locates at 600 nm, which is ascribed to the 5D0→7F2 transition of Eu3+. In order to achieve the emission-tunable phosphors, the Eu3+/Tb3+ co-doped Ba2Ca(BO3)2 are synthesized. When tuning the Eu3+ or Tb3+ concentration, Ba2Ca(BO3)2:Eu3+, Tb3+ can both show the tunable emission, which may be induced by the energy transfer from Tb3+ to Eu3+.

Over-expression of SlJA2 decreased the accumulation of SA, which resulted in significant physiological and gene expression changes in transgenic tobacco plants, leading to the decreased heat tolerance of transgenic tobacco. NAC family, the largest transcription factors in plants, responses to different environmental stimuli. Here, we isolated a typical NAC transcription factor (SlJA2) from tomato and got transgenic tobacco with SlJA2 over-expression. Expression of SlJA2 was induced by heat stress (42 °C), chilling stress (4 °C), drought stress, osmotic stress, abscisic acid, and salicylic acid. Over-expression of SlJA2 decreased the accumulation of salicylic acid by regulating expression of salicylic acid degradation gene under heat stress. Compared to WT plants, stomatal apertures and water loss increased in transgenic plants, and the damage of photosynthetic apparatus and chlorophyll breakdown were more serious in transgenic plants under heat stress. Meanwhile, more H2O2 and O2 ·− were accumulated transgenic plants and proline synthesis was restricted, which resulted in more serious oxidative damage compared to WT. qRT-PCR analysis showed that over-expression of SlJA2 could down-regulate genes involved in reactive oxygen species scavenging, proline biosynthesis, and response to heat stress. All the above results indicated that SlJA2 may be a negative regulator responded to plant’s heat tolerance. Thus, this study provides new insight into roles of NAC family member in plant response to abiotic stress.

SUMOylation is an important post-translational modification process that regulates different cellular functions in eukaryotes. SIZ/PIAS-type SAP and Miz1 (SIZ1) proteins exhibit SUMO E3 ligase activity, which modulates SUMOylation. However, SIZ1 in tomato has been rarely investigated. In this study, a tomato SIZ1 gene (SlSIZ1) was isolated and its molecular characteristics and role in tolerance to drought stress are described. SlSIZ1 was up-regulated by cold, sodium chloride (NaCl), polyethylene glycol (PEG), hydrogen peroxide (H2 O2 ) and abscisic acid (ABA), and the corresponding proteins were localized in the nucleus. The expression of SlSIZ1 in Arabidopsis thaliana (Arabidopsis) siz1-2 mutants partially complemented the phenotypes of dwarf, cold sensitivity and ABA hypersensitivity. SlSIZ1 also exhibited the activity of SUMO E3 ligase to promote the accumulation of SUMO conjugates. Under drought stress, the ectopic expression of SlSIZ1 in transgenic tobacco lines enhanced seed germination and reduced the accumulation of reactive oxygen species. SlSIZ1 overexpression conferred the plants with improved growth, high free proline content, minimal malondialdehyde accumulation and increased accumulation of SUMO conjugates. SlSIZ1 is a functional homolog of Arabidopsis SIZ1 with SUMO E3 ligase activity. Therefore, overexpression of SlSIZ1 enhanced the tolerance of transgenic tobacco to drought stress.

Ascorbate (AsA) is an important antioxidant that can scavenge reactive oxygen species to protect plant cells against oxidative stress. Guanosine 5'-diphosphate (GDP)-L-galactose phosphorylase (GGP) is a key enzyme in the AsA biosynthetic pathway. To investigate the functions of GGP in AsA synthesis and oxidative stress tolerance in tomato, antisense lines with a reduced expression of SlGGP were obtained. Photobleaching after treatment of leaf disks with methyl viologen was more severe in transgenic lines compared to wild type (WT) plants. Moreover, compared with the WT plants, the transgenic plants showed a higher content of hydrogen peroxide, superoxide anion, malondialdehyde, as well as ion leakage, but a lower content of AsA and chlorophylls, ascorbate peroxidase activity, net photosynthetic rate, and maximal photochemical efficiency of photosystem II. Results of real-time quantitative polymerase chain reaction show that suppression of the SlGGP gene in the transgenic plants reduced their oxidative stress tolerance.

Trehalose is proposed to serve multiple physiological roles in insects. However, its importance remains largely unconfirmed. In the present paper, we knocked down either a trehalose biosynthesis gene (trehalose-6-phosphate synthase, LdTPS) or each of three degradation genes (soluble trehalases LdTRE1a, LdTRE1b or membrane-bound LdTRE2) in Leptinotarsa decemlineata by RNA interference (RNAi). Knockdown of LdTPS decreased trehalose content and caused larval and pupal lethality. The LdTPS RNAi survivors consumed a greater amount of foliage, obtained a heavier body mass, accumulated more glycogen, lipid and proline, and had a smaller amount of chitin compared with the controls. Ingestion of trehalose but not glucose rescued the food consumption increase and larval mass rise, increased survivorship, and recovered glycogen, lipid and chitin to the normal levels. In contrast, silencing of LdTRE1a increased trehalose content and resulted in larval and pupal lethality. The surviving LdTRE1a RNAi hypomorphs fed a smaller quantity of food, had a lighter body weight, depleted lipid and several glucogenic amino acids, and contained a smaller amount of chitin. Neither trehalose nor glucose ingestion rescued these LdTRE1a RNAi defects. Silencing of LdTRE1b caused little effects. Knockdown of LdTRE2 caused larval death, increased trehalose contents in several tissues and diminished glycogen in the brain-corpora cardiaca-corpora allata complex (BCC). Feeding glucose but not trehalose partially rescued the high mortality rate and recovered glycogen content in the BCC. It seems that trehalose is involved in feeding regulation, sugar absorption, brain energy supply and chitin biosynthesis in L. decemlineata larvae.
