BACKGROUND: Strawberry fruits are perishable with a short post-harvest life. Cinnamyl alcohol dehydrogenase (CAD) is the key enzyme for lignin biosynthesis strengthening plant cell wall. A systematic characterization of strawberry CAD family is absent and their involvement in fruit firmness is largely elusive. OBJECTIVE: Current work aims for a genome-wide identificationof CAD family and its expression correlation with fruit firmness in strawberry varieties. METHODS: A genome-scale identification and molecular characterization for CADs were performed in the commercial strawberry (Fragaria×ananassa) and woodland strawberry (F. vesca). qPCR analysis of CAD homoeologs in three cultivars varying with fruit firmness revealed candidate CAD members positively correlated with lignin content and fruit firmness. RESULTS: A total of 14 and 24 CAD loci were identified in the genomes of F.vesca var. Hawaii4 and F. ×ananassa cv. Camarosa, respectively.Phylogenetic analysis supported a division of this family into three classes. Class I FvCAD each has four homoeologs in commercial strawberry, while those of Class II and Class III have only one or two homoeologs. Except for FvCAD2 and -6, there exits at least one pair of CADs sharing ∼97% or above amino acid identity between F. vesca and F. ×ananassa.The flesh firmness and lignin content varied greatly among strawberry germplasm. Distinct dynamic changes in fruit lignin content were observed before the large green stage, but fruit firmness displayed a similar decrease profile during fruit development in three varieties. Of the eight genes detected in F.×ananassa, FvCAD3 and -12 did not display a F. vesca-biased expression pattern during fruit development.FvCAD4 of Class I was expressed at levels positively correlated with variation in fruit lignin content at white stage.Transcript abundance of five Class IIgenes including FvCAD3, -8, -10, -11, and -12 was positively correlated with lignin content and fruit firmness, with FvCAD10 and -11 (FaCAD in previous publication) reaching an extremely significant correlation with the genetic variation in fruit firmness across three varieties. CONCLUSION: Strawberry Class II CADs were significantly correlated with the genetic variation in fruit firmness, which might expand the potential choices for improving strawberry shelf life.

BACKGROUND: Bet v 1 family identified as one major plant food allergen class, is highly homologous to pathogenesis-related protein 10 (PR-10), but its biological function involved in disease resistance is still unclear. OBJECTIVE: This study aims to investigate whether Bet v 1 potential allergens participate in the resistance of berry crops against fungal pathogen. METHODS: Allergenicity of Bet v 1 proteins in strawberry (Fragaria) was evaluated by bioinformatics methods. Their expression in response to anthracnose and between susceptible and resistance varieties was analyzed. RESULTS: 19 Bet v 1 homologous proteins were identified and 15 of them were considered as allergen candidates. RNA-seq analysis indicated most of these Fra a 1s expressed in fruits could be largely induced by the invasion of anthracnose pathogen Colletotrichum. The mRNA level of fruit major allergen Fra a 1.05 in the resistant variety Shenyang (SY) was 20∼50 fold higher compared with those in the susceptible cultivar and two diploid wild species. Immunoblotting using Birch (Betula pendula) Bet v 1 allergen-specific IgG antibody confirmed the large-scale accumulation of potential cross-reactive antigens in SY fruit. CONCLUSIONS: Strawberry Bet v 1 potential allergens exhibit their correspondence with anthracnose resistance that might be instructive to future breeding strategies.

Strawberry anthracnose caused by Colletotrichum spp. is one of the most serious diseases in the strawberry fields of China. In total, 196 isolates of Colletotrichum were obtained from leaves, stolons, and crowns of strawberry plants with anthracnose symptoms in eastern China and were characterized based on morphology, internal transcribed spacer (ITS), and β-tubulin (TUB2) gene sequences. All 196 isolates were identified as the Colletotrichum gloeosporioides species complex. In total, 62 strains were further identified at the species level by phylogenetic analyses of multilocus sequences of ITS, glyceraldehyde-3-phosphate dehydrogenase (GAPDH), actin (ACT), Apn2-Mat1-2 intergenic spacer and partial mating type (ApMat), calmodulin (CAL), and TUB2. Three species from the C. gloeosporioides species complex were identified: Colletotrichum siamense, C. fructicola, and C. aenigma. Isolates of C. siamense were tolerant to high temperatures, with a significantly larger colony diameter than the other two species when grown above 36°C. The inoculation of strawberry plants confirmed the pathogenicity of all three species. C. siamense isolates resulted in the highest disease severity. The in vitro sensitivities of C. siamense and C. fructicola isolates to azoxystrobin and three demethylation-inhibitor (DMI) fungicides (difenoconazole, tebuconazole, and prochloraz) were determined. Both species were sensitive to DMI fungicides but not to azoxystrobin. C. siamense isolates were more sensitive to prochloraz, while C. fructicola isolates were more sensitive to difenoconazole and tebuconazole. The present study provides valuable information for the effective management of strawberry anthracnose.

Strawberry mottle virus (SMoV) is one of the main RNA viruses that profoundly affects the growth of strawberries worldwide. The rapid on-site detection of SMoV described here can be applied to produce virus-free strawberry seedlings. Reverse transcriptase recombinase polymerase amplification (RT-RPA) was combined with lateral flow (LF) strip to rapidly detect SMoV. The detection limit was 500 fg of RNA under optimized conditions. The SMoV-RT-RPA-LF assay was optimal with a combination of 2 μL reverse primer (5 μM) and 0.6 μL probe (10 μM) in a 50 μL RT-RPA reaction mixture for isothermal amplification at 40 ℃ for 15 min. In addition, 100 suspected samples were collected from different regions in the Shanghai suburbs. The SMoV-RT-RPA-LF assay showed that 3 of these 100 samples were positive for SMoV, which was in good concordance with the reverse transcription polymerase chain reaction (RT-PCR) results. The primers and probe had a unique specificity to SMoV because there was no cross-reaction with other strawberry viruses. This study provides an effective technique for the rapid on-site detection of SMoV to ensure a virus-free strawberry nursery.

Intragenesis is an all-native engineering technology for crop improvement. Using an intragenic strategy to bring genes from wild species to cultivated strawberry could expand the genetic variability. A robust regeneration protocol was developed for the strawberry cv. ‘Shanghai Angel’ by optimizing the dose of Thidiazuron and identifying the most suitable explants. The expression cassette was assembled with all DNA fragments from F. vesca, harboring a sugar transporter gene FvSTP8 driven by a fruit-specific FvKnox promoter. Transformed strawberry was developed through an Agrobacterium-mediated strategy without any selectable markers. Other than PCR selection, probe-based duplex droplet digital PCR (ddPCR) was performed to determine the T-DNA insert. Four independent transformed shoots were obtained with a maximum of 5.3% efficiency. Two lines were confirmed to be chimeras, while the other two were complete transformants with six and 11 copies of the intragene, respectively. The presence of a vector backbone beyond the T-DNA in these transformants indicated that intragenic strawberries were not obtained. The current work optimized the procedures for producing transformed strawberry without antibiotic selection, and accurately determined the insertion copies by ddPCR in the strawberry genome for the first time. These strategies might be promising for the engineering of ‘Shanghai Angel’ and other cultivars to improve agronomic traits.

Strawberry is a highly efficient and economical horticultural crop plant, and strawberry fruits are easy to soften after ripening and decay after harvest, which severely impacts the economic benefits. Expansins are plant cell-wall loosening proteins involved in the process of fruit softening, loosening cell walls and reducing fruit firmness. In this study, 35 FvEXPs genes were identified in the F. vesaca genome. These genes were divided into four subfamilies (27 FvEXPAs, 5 FvEXPBs, 1 FvEXLAs, and 2 FvEXLBs) and were unevenly distributed on 7 chromosomes. Gene structure and motif analysis showed the conserved structure and motif in same subgroup, however, the different motifs and structures may reveal functional divergence of multigene family members of FvEXPs in different developmental stages of fruits. The expression profiling by RNA-seq and qRT-PCR analysis revealed that the FvEXP genes have distinct expression patterns among different stages of strawberry development and ripening. Among them, 3 genes (FvEXPA9, FvEXPA12, and FvEXPA27) were highly expressed in the ripening stage, FvEXPA9 and FvEXPA12 were especially highly expressed in turning stage, whereas FvEXPA27 was especially highly expressed in red stage. Our study provides a better understanding of the FvEXP genes, which may benefit strawberry biotechnological breeding and genetic modification for improving fruit quality and delaying fruit softening.

The transport of sugar is essential for plant growth and development, tightly correlated with crop productivity and quality. SWEET (Sugar Will Eventually be Exported Transporters) is a family of sugar effluxer first reported a decade ago, whose significance has been reported in a dozen of plant species. Strawberry is an important crop developing striking fruits. Up to date, the involvement of SWEETs in strawberry is completely elusive. Current work focuses on the genome-wide identification of SWEET transporter coding genes in diploid strawberry (Fragaria vesca) and the expression analysis of octoploid strawberry (F. × ananassa) SWEET homoeologs in different vegetative and reproductive organs. A total of 20 candidate SWEET loci were identified in Fragaria vesca V4 genome via a local blast using the MtN3_slv motif (PF03083) against the protein database of FvH4.0.a2 version. Phylogenetic analysis indicated this family falling into four clades. These genes are distributed in all seven chromosomes except for Fvb1, and most members contain six exons divided by five introns, coding a protein with 235 to 310 aa and around 7 transmembrane helices. The spatio-temporal expression profiles of SWEET genes in different parts of fruit (cortex, pith and achenes) of cv. Benihoppe were explored and a set of candidate SWEET genes invoved in sugar transport in strawberry fruits were identified, such as FvSWEET1, -2a, -7, -9a, -9c, -10, and -11. Our results provide a good foundation for further functional investigation of SWEETs in strawberry fruit quality formation and other developmental processes as well as responses to pathogens and abiotic stresses. The information of SWEETs in strawberry also has a reference value for other Rosaceae plants.

We systematically investigate ellipticity dependence of high-order harmonic generation of Ar and ${\mathrm{N}}_{2}$ in intense elliptically polarized laser fields. The experimental normalized ratios of low-order harmonic intensity to high-order harmonic intensity increase with ellipticity for both Ar and ${\mathrm{N}}_{2}$, and quantitatively depend on targets and trajectory paths. The experimental results are well reproduced by a nonadiabatic semiclassical simulation and explained by trajectory-based analysis. In addition, the influence of nuclear distance on the ratios is theoretically investigated. Our work reveals that the difference between atoms and molecules can be attributed to the influence of different ionic potentials, which depends on the molecular structure (internuclear distance) and alignment, on the evolution of the photoelectron.

Gut microbiota plays a key role in the maintenance of human health. In most cases, carbohydrates can be fermented by gut microbiota as substrates. By such degradation reaction, the abundances, activities and metabolite production of gut microbiota will be regulated and thus prevent the occurrence of human diseases such as glycolipid metabolism disorders, enteric abnormality, aging and neurodegeneration, cancers and depression. In this paper, we reviewed the roles of carbohydrates in promoting human health through gut microbial regulation, as well as further studies on the regulatory mechanism and potential application of carbohydrates, which need to be strengthened in the future.

Sugar from plant photosynthesis is a basic requirement for life activities. Sugar transporters are the proteins that mediate sugar allocation among or within source/sink organs. The transporters of the major facilitator superfamily (MFS) targeting carbohydrates represent the largest family of sugar transporters in many plants. Strawberry (Fragaria × ananassa Duchesne) is an important crop appreciated worldwide for its unique fruit flavor. The involvement of MFS sugar transporters (STs) in cultivated strawberry fruit sugar accumulation is largely unknown. In this work, we characterized the genetic variation associated with fruit soluble sugars in a collection including 154 varieties. Then, a total of 67 ST genes were identified in the v4.0 genome integrated with the v4.0.a2 protein database of F. vesca, the dominant subgenome provider for modern cultivated strawberry. Phylogenetic analysis updated the nomenclature of strawberry ST homoeologs. Both the chromosomal distribution and structural characteristics of the ST family were improved. Semi-RT-PCR analysis in nine tissues from cv. Benihoppe screened 34 highly expressed ST genes in fruits. In three varieties with dramatically differing fruit sugar levels, qPCR integrated with correlation analysis between ST transcript abundance and sugar content identified 13 sugar-correlated genes. The correlations were re-evaluated across 19 varieties, including major commercial cultivars grown in China. Finally, a model of the contribution of the sugar transporter system to subcellular sugar allocation in strawberry fruits was proposed. Our work highlights the involvement of STs in controlling strawberry fruit soluble sugars and provides candidates for the future functional study of STs in strawberry development and responses and a new approach for strawberry genetic engineering and molecular breeding.

We experimentally investigated the high-order harmonic generation (HHG) from aligned O2 and N2 molecules in a linearly polarized laser field, and presented the dependence of the harmonic spectrum on the driving laser intensity and molecular alignment angle. The minimum position of HHG of O2 varies with changing the laser intensity, which is caused by multi-orbital interference. However, the location of the observed minimum structure in N2 harmonic spectrum remained unchanged upon changing the laser intensity. The mechanism of the spectral minimum for N2 case is regarded as a Cooper-like minimum in HHG associated with the molecular electronic structure. This work indicates that harmonic spectroscopy can effectively uncover information about molecular structure and electron dynamics.

Background: Phalaenopsis is an important ornamental flowering plant that belongs to the Orchidaceae family and is cultivated worldwide. Phalaenopsis has a long juvenile phase; therefore, it is important to understand the genetic elements regulating the transition from vegetative phase to reproductive phase. In this study, FLOWERING LOCUS T (FT) homologs in Phalaenopsis were cloned, and their effects on flowering were analyzed. Results: A total of five FT-like genes were identified in Phalaenopsis. Phylogenetic and expression analyses of these five FT-like genes indicated that some of these genes might participate in the regulation of flowering. A novel FT-like gene, PhFT-1, distantly related to previously reported FT genes in Arabidopsis and other dicot crops, was also found to be a positive regulator of flowering as heterologous expression of PhFT-1 in Arabidopsis causes an early flowering phenotype. Conclusions: Five FT homologous genes from Phalaenopsis orchid were identified, and PhFT-1 positively regulates flowering.

During hundreds of years of medication, it is believed that the steamed Panax notoginseng (SPN) can enrich and regulate the blood, replenish the body, and improve the health. The aim of this study was to optimize the steaming conditions of SPN which are related to the hematopoietic effect. In the study, network pharmacology and pharmacological experiments were used to predict and verify the potential hematopoietic active ingredients of SPN. Three variables including the steaming time (2-10 h), steaming temperature (90-130°C), and different producing areas of PN were investigated by using single-factor analysis. Box-Behnken design response surface methodology (BBD-RSM) was performed to explore the optimized steaming conditions which are responsible for the hematopoietic effect of SPN. Furthermore, the hematopoietic effect of the optimized SPN was evaluated. Results demonstrated that ginsenoside Rd, Rh1, Rh4, Rk3, and 20(S)-Rg3 can significantly increase blood routine parameters and expressions of hematopoietic factors in anemia mice. The total contents of the five ginsenosides were selected as evaluation indexes of the response surface method. We found that the PN from Wenshan steamed at 120°C for 5 h could significantly increase the levels of blood routine parameters and hematopoietic factor expression compared with the model group. The study not only provides data support for the determination of hematinic effect-related markers for SPN but also gives a scientific reference for the processing of SPN which has a better hematopoietic effect. The underlying mechanisms require further research.

In traditional Chinese medicine, the feces of flying squirrels have long been used to promote blood circulation and relieve bodily stasis. However, the excrement of flying squirrels may harbor zoonotic agents that could be hazardous to public health. To understand the occurrence of bacterial and parasitic infections in this species, we investigated selected zoonotic pathogens including Leptospira and Blastocystis in the urine and feces of flying squirrels in China. Urine and fecal samples from flying squirrels were collected from a family-owned flying squirrel farm located in Enshi County, Hubei Province in China. Leptospira and Blastocystis DNA was extracted from the urine and feces of flying squirrels, and used as targets for PCR amplification, using different specific primers. PCR amplification and DNA sequencing showed that 4.4% (3/69) of flying squirrels were positive for Leptospira, while 30.4% (21/69) of the animals were positive for Blastocystis. Notably, 1.4% (1/69) of flying squirrels were found to be co-infected with Leptospira and Blastocystis. Sequence analyses allowed for the detection of 3 Blastocystis subtypes (ST1, ST3 and ST13), and mixed infections of Blastocystis subtype 1 and subtype 3 were found in 4.4% (3/69) of flying squirrels. Phylogenetic analysis of the 16S ribosomal RNA gene (rrs2), the flagellin B gene (flaB), and outer membrane lipoprotein lipL32 gene (LipL32) sequences indicated that the Leptospira species detected in the study was L. interrogans. We concluded that flying squirrels from central China were infected with Leptospira and Blastocystis, suggesting that these animals can be a source of infection for their owners, and using fresh excrement from this animal as traditional medicine could be risky to human health. To the best of our knowledge, this is the first report of Leptospira and Blastocystis infection in flying squirrels from Enshi County, China. Our findings provide new data on the epidemiology of these pathogens in this region.

Ethnopharmacological relevance In Chinese folk medicine, Ligustrum robustum (Roxb.) Blume has been widely used as a healthy tea beverage for improvement in obesity and lipidemic metabolic disorders. Aim of the study: We aimed to investigate the effect of L. robustum extract (LRE) on metabolic syndrome in high-fat diet (HFD)-fed mice and to explore the underlying role of gut microbiota during the treatment. Materials and methods The ground dried leaves of L. robustum (Roxb.) Blume were extracted with ethanol and then purified by a resin column. The composition of L. robustum extract (LRE) was analyzed by high-performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS). C57BL/6 J mice fed with HFD were treated with LRE for 16 weeks. RT-qPCR and morphological staining were utilized to reveal the impact of LRE on hepatic glucolipid metabolism and gut integrity. The next-generation sequencing of 16 S rDNA was applied for analyzing the gut microbial community of fecal samples. Results LRE, mainly composed of ligupurpuroside A and aceteoside, alleviated insulin resistance, improved hepatic metabolism, enhanced intestinal integrity, and suppressed inflammatory responses in HFD-fed mice. Moreover, LRE treatment reshaped the gut microbiota structure by increasing the levels of genera Streptococcus, Lactobacillus, and Mucispirillum and decreasing the populations of Alistipes and Lachnospiraceae NK4A136 group in HFD-fed mice. The alteration of gut microbiota was associated with several metabolic pathways of gut bacteria. Spearman's correlation analysis further confirmed the links between the changed intestinal bacteria and multiple disease indices. Conclusions LRE prevented gut microbiota dysbiosis and metabolic disorder in HFD-fed mice, which helps to promote the application in LRE-mediated prevention from metabolic syndrome as a gut microbial regulator.

Tree peony (Paeonia section Moutan DC.) is an excellent woody oil crop, and the cloning and functional analysis of genes related to fatty acid (FA) metabolism from this organism has not been reported. Lysophosphatidic acid acyltransferase (LPAAT), which converts lysophosphatidic acid (LPA) to phosphatidic acid (PA), catalyzes the addition of fatty acyl moieties to the sn-2 position of the LPA glycerol backbone in triacylglycerol (TAG) biosynthesis. This project reports a putative lysophosphatidic acid acyltransferase gene PrLPAAT1 isolated from Paeonia rockii. Our data indicated that PrLPAAT1 has 1047 nucleotides and encodes a putative 38.8 kDa protein with 348 amino acid residues. Bioinformatic analysis demonstrated that PrLPAAT1 contains two transmembrane domains (TMDs). Subcellular localization analysis confirmed that PrLPAAT1 is a plasma membrane protein. Phylogenetic analysis revealed that PrLPAAT1 shared 74.3 and 65.5% amino acid sequence identities with the LPAAT1 sequences from columbine and grape, respectively. PrLPAAT1 belongs to AGPAT family, and may have acyltransferase activity. PrLPAAT1 was ubiquitously expressed in diverse tissues, and PrLPAAT1 expression was higher in the flower and developing seed. PrLPAAT1 is probably an important component in the FA accumulation process, especially during the early stages of seed development. PrLPAAT1 overexpression using a seed-specific promoter increased total FA content and the main FA accumulation in Arabidopsis transgenic plants.

Leeches are frequently used in traditional Chinese medicine. However, they are potentially hazardous to human and animal health by transmitting several pathogens. Studies of diseases transmitted by leeches are scarce. The purpose of this study was to analyze the pathogens carried in pond-farmed medicinal leech in China. Leeches were collected from 6 farms in Hubei Province in central China. DNA was extracted from the internal organ of leeches to analyze the origin of blood meal. Leech genera were confirmed through amplification of 18S rRNA and mitochondrial gene cytochrome oxidase I (COI) gene by PCR and host animal species were identified through amplification of mitochondrial cytochrome b gene. Species of Ehrlichia in the leech specimens were screened with PCR using specific primers. PCR amplification and DNA sequencing showed that 620 leeches were Hirudinaria sp. Ehrlichia DNA was detected in 39 specimens from 2 farms. We obtained a total of 65 sequences of the cytB gene from 620 leech internal organ samples including sequences of human (n = 5), rat (n = 1), domestic pig (n = 10), duck (n = 23), goose (n = 12) and buffalo (n = 14). Phylogenetic analysis of the rrs and groEL gene sequences showed that Ehrlichia detected in the study were closely related to Ehrlichia sp. in ticks from Korea and Japan. To the best of our knowledge, this is the first report on Ehrlichia DNA being detected from leeches. Our findings provided new data on Ehrlichia spp. and farmed leech species in China.

Study the use of cellulose as a coating material on the base paper coating production of food packaging paper, using NMMO direct dissolution method to dissolved cellulose. Through a series of single-factor experiments to compare the best formula for making cellulose coated paper, and test the main properties of the cellulose coated paper produced. The cellulose concentration is 5%, the mass ratio of cellulose to chitosan is 4:1, and the plasticizer glycerin concentration is 3%, the resulting cellulose coated paper to achieve the best performance. The coating amount of 2g / ㎡, moisture permeability coefficient of 480.23g/ ㎡·24h, oxygen permeability coefficient of 7.8g·mm/ ㎡·24h, oil permeability 0.026g·mm / ·d, tensile strength of 7012N/m.

Thus it is concluded that the barcode warehouse management system not only highly improves the level of inventory management and reduces the labor intensity, but also decreases the total storage cost and improves inventory management efficiency with its application in warehouse management of printing and packaging enterprises. Simple, safe, quick and efficient barcode warehouse management system is becoming more proficient, and it will play an increasingly significant role in the warehouse management of printing and packaging enterprises.

In this paper, the sliding window, reasons and TCP algorithms of congestion control such as slow start, congestion avoidance, fast retransmit and fast recovery are discussed. Auxin has been regarded as the main signal molecule coordinating the growth and ripening of fruits in strawberry, the reference genomic system for Rosaceae. The mechanisms regulating auxin biosynthesis in strawberry are largely elusive. Recently, we demonstrated that two YUCCA genes are involved in flower and fruit development in cultivated strawberry. Here, we show that the woodland strawberry (Fragaria vesca L.) genome harbors nine loci for YUCCA genes and eight of them encode functional proteins. Transcription pattern in different plant organs was different for all eight FvYUCs. Functionality of the FvYUC6 gene was studied in transgenic strawberry overexpressing FvYUC6, which showed typical high-auxin phenotypes. Overexpression of FvYUC6 also delayed flowering and led to complete male sterility in F. vesca. Additionally, specific repression of FvYUC6 expression by RNA interference significantly inhibited vegetative growth and reduced plant fertility. The development of leaves, roots, flowers, and fruits was greatly affected in FvYUC6-repressed plants. Expression of a subset of auxin-responsive genes was well correlated with the changes of FvYUC6 transcript levels and free indole-3-acetic acid levels in transgenic strawberry. These observations are consistent with an important role of FvYUC6 in auxin synthesis, and support a main role of the gene product in vegetative and reproductive development in woodland strawberry.

Carbonic anhydrases (CAs, EC 4.2.1.1) are metalloenzymes found throughout the phylogenetic tree. The β-class carbonic anhydrases (β-CAs) are the predominating class of CAs in plants. Growing evidence underscores the importance of β-CAs in plant immunity and environmental adaptation in addition to their roles in photosynthesis. However, many fundamental problems in Arabidopsis βCAs expression remain unsolved. Here we examined the transcript abundance of AtβCAs in different tissues of Arabidopsis thaliana, and the accumulation of mRNA in response to CO2 and darkness. Histochemical analysis was performed to study the promoter activity of AtβCAs during post-germination seedling growth and in mature plants. All six members of the AtβCA subfamily showed a response to changed CO2 level and darkness, but each member showed a specific dynamic pattern. Although expression of each AtβCA was unique, in general most AtβCAs were synchronously expressed in green leaves since 5 days after germination until flowering. AtβCA1 and AtβCA2 were most highly expressed in leaves but AtβCA2 displayed weaker expression in roots. The level of AtβCA3 transcripts was highest in flowers, while AtβCA5 was most widely expressed and might be involved in more processes than other members. AtβCA6 was unique for increased expression in darkness and no expression in either the anther or pistil. The present study provides useful information for further functional investigation.

We investigate experimentally multi-orbital effects in high-order harmonic generation (HHG) from aligned CO2 and N2O molecules by intense femtosecond laser fields with linear and elliptical polarizations. For either of the aligned molecules, a minimum in the harmonic spectrum is observed, the position of which shifts to lower-order harmonics when decreasing the intensity or increasing the ellipticity of the driving laser. This indicates that the minimum originates from the dynamic interference of different channels, of which the tunneling ionization and recombination are contributed via different molecular orbitals. The results show that both the highest occupied molecular orbital (HOMO) and low-lying HOMO-2 in CO2 (or HOMO-1 in N2O) contribute to the molecular HHG in both linearly and elliptically polarized strong laser fields. Our study would pave a way for understanding multi-electron dynamics from polyatomic molecules irradiated by strong laser fields.

We perform an experimental study of the multi-orbital effect on the high-order harmonic generation (HHG) from aligned N2 molecules in both linearly and elliptically polarized intense laser fields. Measured by a home-built extreme ultraviolet (XUV) flat grating spectrometer with the pump–probe method, the angular distributions of different orders of HHG are obtained, which show distinctive behaviors for harmonics in the plateau and the cut-off regions. The ellipticity dependence of HHG is investigated by aligning the molecular axis parallel or perpendicular to the laser polarization. Our results indicate that both the highest occupied molecular orbital (HOMO) as well as the lower one (HOMO-1) contribute to the HHG of N2 molecules, in either linearly or elliptically polarized intense laser field. The study paves the way for understanding the ultrafast electron dynamics of molecules exposed to an intense laser field.

Anthracnose caused by Colletotrichum spp. is one of the most destructive diseases of cultivated strawberry (Fragaria × ananassa Duchesne) worldwide. The correlation between NBS–LRR genes, the largest class of known resistance genes, and strawberry anthracnose resistance has been elusive. BLAST search in NCBI identified 94 FvNBSs in the diploid genome of strawberry Fragaria vesca, with 67 of the TIR–NBS–LRR type. At least 36 FvNBSs were expressed, with 25% being non-coding genes. Two F. vesca ecotypes, HLJ and YW, showed great variations in both morphological and physiological responses upon C. gloeosporioides infection. qRT-PCR revealed that 5 of the 12 leaf-expressed FvNBSs displaying opposite transcription responses to C. gloeosporioides infection in two ecotypes. These results showed that the transcriptional responses of several FvNBSs were involved in the ecotype-specific responses to C. gloeosporioides in F. vesca. These FvNBSs hold potential in characterizing molecular components and developing novel markers associated with anthracnose resistance in strawberry.

Design method of main circuit parameter based on theoretical calculation and engineering practice is developed for permanent magnetic direct-drive wind power generating system with dual PWM converter topology. DC bus voltage, DC bus filter capacitor, rating value of power electronic devices and gird-side LCL filter parameters are calculated for an experimental wind power generating system. Hardware platform is built using calculated parameters, and the experimental results show that the design method is viable and expected design goal is achieved.

Capacitor matching in single-phase capacitor-run induction motor (SPCRIM) is investigated in this paper. Combining symmetrical components method with resultant current method, capacitor value is calculated. Relationship between turn ratio of main winding and auxiliary winding, main winding current, auxiliary winding current and capacitor value is analyzed by simulation. SPCRIM is optimized on the ground of the theoretically calculated capacitor value and turn ratio of the main and auxiliary windings with FEA (Finite Element Analysis). Results show that calculated capacitor value contributes to the determination of turn ratio of the main winding and auxiliary winding, and with appropriately calculated capacitor value and turn ratio, small winding current can be got.

The disease symptoms recognized as 'Anthracnose' are caused by Colletotrichum spp. and lead to large-scale strawberry (Fragaria×ananassa Duchesne) losses worldwide in terms of both quality and production. Little is known regarding the mechanisms underlying the genetic variations in the strawberry-Colletotrichum spp. interaction. In this work, Colletotrichum gloeosporioides (C. gloeosporioides) infection was characterized in two varieties exhibiting different susceptibilities, and the involvement of salicylic acid (SA) was examined. Light microscopic observation showed that C. gloeosporioides conidia germinated earlier and faster on the leaf surface of the susceptible cultivar compared with the less-susceptible cultivar. Several PR genes were differentially expressed, with higher-amplitude changes observed in the less-susceptible cultivar. The less-susceptible cultivar contained a higher level of basal SA, and the SA levels increased rapidly upon infection, followed by a sharp decrease before the necrotrophic phase. External SA pretreatment reduced susceptibility and elevated the internal SA levels in both varieties, which were sharply reduced in the susceptible cultivar upon inoculation. The less-susceptible cultivar also displayed a more sensitive and marked increase in the transcripts of NB-LRR genes to C. gloeosporioides, and SA pretreatment differentially induced transcript accumulation in the two varieties during infection. Furthermore, SA directly inhibited the germination of C. gloeosporioides conidia; NB-LRR transcript accumulation in response to SA pretreatment was both dose- and cultivar-dependent. The results demonstrate that the less-susceptible cultivar showed reduced conidia germination. The contribution of SA might involve microbial isolate-specific sensitivity to SA, cultivar/tissue-specific SA homeostasis and signaling, and the sensitivity of R genes and the related defense network to SA and pathogens.

Plant calcium-dependent protein kinases (CDPKs) play vital roles in calcium signal transduction during various developmental processes and during responses to biotic and abiotic stresses. Here, we isolated and characterized a CDPK gene designated FvCDPK1 from a wild diploid strawberry accession Heilongjiang-3 (Fragaria vesca L.). The FvCDPK1 gene contains 12 exons and 11 introns, and the sequences of most exons are highly conserved in higher plants. The full-length cDNA of FvCDPK1 contains 1,825 nucleotides with an open reading frame of 1,653 bp encoding a polypeptide of 550 amino acids. The deduced FvCDPK1 protein contains the basic features of typical plant CDPKs: a catalytic kinase domain and a regulatory calmodulin-like domain containing four EF-hand calcium-binding motifs. Phylogenetic analysis confirmed that FvCDPK1 belongs to the plant CDPK family. When transiently expressed in onion epidermal cells, the FvCDPK1-GFP fusion protein was found to be localized in the nucleus. Expression analysis indicated that FvCDPK1 was expressed in fruits at different developmental and ripening stages, as well as in several tissues such as roots, runners, flowers, leaves, and meristems. Moreover, expression levels of FvCDPK1 were higher in meristems than in other vegetative tissues. Under abiotic stress conditions, however, FvCDPK1 was found to be upregulated upon abscisic acid, NaCl, cold-, or high-temperature treatments. Taken together, our data suggest that FvCDPK1 might play a role in various responses to abiotic stresses in strawberry.

In strawberry (Fragaria × ananassa Duch.), auxin has been recognized as the main signal molecule coordinating the growth and initiation of ripening of fruits. The molecular mechanism regulating auxin biosynthesis in strawberry remains unknown. This project reports two YUCCA flavin monooxygenase genes FaYUC1-2 isolated from cultivated strawberry. FaYUC1 and FaYUC2 are most homologous to AtYUC6 and AtYUC4, respectively. Significant expression of FaYUC1-2 is found in vegetative meristems and reproductive organs, with overlapping but distinct patterns. During fruit development, both transcripts of FaYUC1 and FaYUC2 in achenes reach a peak around large green fruit (G2) stage, but the sudden rise in FaYUC2 transcript level is much steeper and begins earlier than that in FaYUC1. FaYUC2 is also obviously expressed in the receptacles from green fruits, hinting another auxin source for receptacle development, other than achenes. FaYUC1 over-expression Arabidopsis exhibits typical auxin hyper-accumulation phenotype in many aspects, such as the narrow and downward curled leaves, strong apical dominance, short and hairy root. It is also severely sterile, due to the disruption of floral meristems initiation and floral organs development. Transgenic analysis indicates that strawberry YUC gene may hold conserved role in auxin biosynthesis like their homologs in other plants. Integrated with the spatiotemporal expression features, these results led us to propose that FaYUC1-2 may involve in many developmental processes including flower and fruit development in strawberry. Key message This paper is the first report of isolation and characterization of strawberry auxin biosynthesis genes. And their conserved functions in auxin biosynthesis were confirmed after ectopic expression.

'Shenmei 3#' strawberry (Fragaria × ananassa Duchesne) offers growers in Shanghai and other areas with relatively mild winter and spring climates a short-day cultivar that is early fruiting and produces fruit that has a distinctively sweet flavor and is resistant to powdery mildew (caused by Podosphaera macularis). Fruits of 'Shenmei 3#' ripen about one week later than 'Toyonoka', a Japanese variety that has been the standard short-day cultivar in Southeast China along the seaside, especially in the Chang Jiang River drainage area.

The surface morphology and chemistry of CBp obtained by pyrolysis of waste tyres at 500 and 700 °C, respectively was studied compared with a commercial tyre carbon blacks by laser particle size analyzer, X-ray diffractogram (XRD) and electron spectroscopy for chemical analysis (ESCA). The distribution of CBp aggregates was the mixed particle distribution of commercial carbon blacks added to tyres in fabrication. The concentration of inorganic compounds and carbonaceous deposits (the organic compounds deposited on the surface of the CBp) depends on the pyrolysis temperature. The chemical nature of the CBp from pyrolysis at 700 °C was found to be closer to the commercial tyre carbon blacks than the CBp from pyrolysis at 500 °C.

A relatively large laboratory-scale sample of a commercial type of PVC material usually used in building, flooring, home furnishings, clothing, etc., is thermally decomposed under carefully controlled conditions. Dehydrochlorination of PVC material at high temperature (up to 600 °C) has been studied. The results show that dehydrochlorination beginning at the temperature of 150 °C can be divided into three stages: low-temperature moderation in the temperature interval 150−180 °C, medium-temperature acceleration in the interval 180−250 °C, and high-temperature moderation in the interval 250−280 °C. The release of chlorine as HCl from PVC materials, at the same temperature, is highest in oxygen, and it increases with an increase of oxygen content. The release of chlorine as HCl is higher by about 5−10% in hydrogen than in nitrogen. Yttria-stabilized zirconia thin films deposited by radio-frequency sputtering from a 3 mol% Y 2 O 3 ZrO 2 target show primarily tetragonal orientation. Annealing in air further reduces the fraction of monoclinic phase and enhances the crystallinity of the tetragonal phase. Films annealed at 700 °C for 2 h showed a phase composition which was greater than 95% tetragonal.
