4. Applicability of Thermal Dissipation Method in Sap Flow Measurement of Karst Shrub Community.

Abstract

Although the thermal dissipation probe (TDP) method is prominent in forest transpiration studies, the accurate estimation of sap flow density in particular trees with different wood structures remains highly variable. To verify the applicability of the TDP method in the measurement of the transpirational water consumption of shrub communities in degraded karst areas in southern China, Granier's original formula was validated and corrected using the isolated stem segment method with Celtis sinensis, Xylosma congesta, Triadica sebifera, Paliurus ramosissimus, Pyracantha fortuneana, Sageretia thea, and Phanera championii. Finally, the correction coefficients were analyzed in relation to wood structural parameters. Our study reveals that the sap flow density estimated using Granier's original formula exhibited varying degrees of overestimation or underestimation compared to the measured sap flow density. The differences ranged from −78.9% to 114.2%, highlighting the need for formula correction. Consequently, we adjusted the coefficients α and β of the original formula (n = 3–5), resulting in a revised range of 0.0047–0.0509 g cm−2 s−1 and 0.5882–1.9821 (R2 = 0.81–0.97), respectively. The conduit diameter was significantly correlated with the correction coefficient α. It is currently unknown whether the results obtained for a particular species can be applied to different growth environments and more prominent individuals of the same species. 5. Growth and Transcriptional Regulation of Camellia sinensisPlanted in Understory Mode Revealed by Transcriptomic, Metagenomic, and Machine-Vision Analyses

Abstract

Camellia sinensisis a significant economic and medical plant. The plant is short and thrives in shaded environments, making it suitable for cultivation under forest canopies. However, the mechanisms governing the growth of C. sinensisin understory conditions need to be fully understood. The study aims to delve into the growth regulatory mechanisms of C. sinensisin understory mode and the impact of the environment on its growth efficiency and bioactive compound synthesis. Through physiological measurements, transcriptomics, metagenomics, and machine vision analysis, a systematic investigation of C. sinensischaracteristics in different environments was conducted. Transcriptome data comparisons unveiled key gene expression changes, and the role of these genes in biosynthetic pathways was validated using quantitative Polymerase Chain Reaction (qPCR). Concurrently, metagenomic analysis of soil microbial communities revealed the environmental effects on microbial diversity. C. sinensisin understory mode exhibited higher stomatal density and smaller pore sizes under low light and humidity conditions; phenolic and flavonoid compounds were identified as the main regulatory pathways, with enhanced expression of key genes such as Dihydroflavonol-4-Reductase(DFR), Anthocyanidin Reductase(ANR), and Leucoanthocyanidin Reductase(LAR), which promoted the synthesis of tea polyphenols and lignin, the abundance of Acidobacteria might be related to the growth of C. sinensisin understory mode. Furthermore, machine vision models indicated that C. sinensisdisplayed higher growth efficiency in the understory mode environment. This research discovered the characteristics of C. sinensisin understory mode and elucidated its growth efficiency in understory mode by modulating phenolic and flavonoid metabolism key genes and promoting the accumulation of secondary metabolites. Differences in soil microbial communities could also affect vegetation coverage and other aspects. These findings provide a scientific basis for optimizing understory mode cultivation and highlight the crucial importance of multidisciplinary approaches in understanding plant adaptability.
