Trichloroethylene (TCE) is a common environmental contaminant that can induce occupational dermatitis medicamentosa-like TCE (ODMLT), where the liver damage is the most common complication. The study aims to uncover the underlying mechanism of TCE-sensitization-induced liver damage by targeting specific exosomal microRNAs (miRNAs). Among the enriched serum exosomal miRNAs of ODMLT patients, miR-205–5p had a significant correlation coefficient with the liver function damage indicators. Moreover, retinoic acid receptor-related orphan receptor α (RORα) was identified as a direct target of miR-205–5p via specific binding. Further experiments showed that kupffer cells (KCs) underwent M1 phenotypic and functional changes in liver injury induced by TCE which were alleviated by reducing the expression of miR-205–5p. However, this alleviation was reversed by the RORα antagonist SR1001. In vitro experiments showed that miR-205–5p promoted M1 polarization of macrophages and enhanced the secretion of inflammatory factors by regulating RORα. An increase in RORα reversed the polarization direction of M1-type macrophages and reduced the secretion of proinflammatory factors. In addition, pretreatment of mice with SR1078, a specific RORα agonist, effectively blocked M1 polarization of KCs and reduced the severity of TCE-induced liver injury. Our study uncovers that miR-205–5p regulates KC M1 polarization by targeting RORα in immune liver injury induced by TCE sensitization, providing new insight into the molecular mechanisms and new therapeutic targets for ODMLT.

Abstract Background The prevalence of abnormal weight is on the rise, presenting serious health risks and socioeconomic problems. Nonetheless, there is a lack of studies on the medical cost savings that can be attained through the mitigation of abnormal weight. The aim of this study was to estimate the impact of abnormal weight on healthcare costs in China. Methods The study employed a 4-wave panel data from China Family Panel Studies (CFPS) between 2012 and 2018 (11,209 participants in each wave). Inpatient, non-inpatient and total healthcare costs were outcome variables. Abnormal weight is categorized based on body mass index (BMI). Initially, the two-part model was employed to investigate the impact of overweight/obesity and underweight on healthcare utilisation and costs, respectively. Subsequently, the estimated results were utilised to calculate the overweight/obesity attributable fraction (OAF) and the underweight attributable fraction (UAF). Results In 2018, healthcare costs per person for overweight and obese population were estimated to be $607.51 and $639.28, respectively, and the underweight population was $755.55. Desertification and salt stress are major causes of terrestrial ecosystem loss worldwide, and the Gobi, representing a salt-stressed area in inland China, has a major impact on the ecosystems and biodiversity of its surrounding environment. The restoration of the Gobi Desert is an important way to control its expansion, but there are few studies on the evaluation of restoration. In this study, soils under different restoration scenarios, namely, soils in restored areas (R1, R2), semi-restored areas (SR1, SR2), and unrestored control areas (C1, C2), were used to investigate differences in microbial diversity and physicochemical properties. The results showed that the soil was mainly dominated by particles of 4–63 μm (26.45–37.94%) and >63 μm (57.95–72.87%). Across the different restoration levels, the soil pH (7.96–8.43) remained basically unchanged, salinity decreased from 9.23–2.26 to 0.24–0.25, and water content remained constant (10.98–12.27%) except for one restored sample in which it was higher (22.32%). The effective Al, Cu, and Zn in the soil increased, but only slightly. Total organic matter (TOM) decreased from 3.86–5.20% to 1.31–1.47%, and total organic nitrogen (TON) decreased from 0.03–0.06% to 0.01–0.02%, but the difference in total organic carbon (TOC) was not significant. High-throughput testing revealed that the bacterial population of the restored area was dominated by A4b (6.33–9.18%), MND1 (4.94–7.39%), and Vicinamibacteraceae (7.04–7.39%). Regarding archaea, samples from the restored areas were dominated by Marine Group II (76.17–81.49%) and Candidatus Nitrososphaera (6.07–9.75%). PCoA showed that the different restoration levels were the main cause of the differences between the samples. Additionally, salinity was the dominant factor that induced this difference, but it was inhibited by the restoration and targeted enrichment of some of these functional genera. Desert restoration should therefore focus on conserving water rather than adding nutrients. Planting salt- and drought-tolerant vegetation will contribute to the initial restoration of the desert and the restoration of the microbiological content of the soil as it migrates over time, creating a cycle of elements. Restoration stimulates and enhances the microbial diversity of the soil via beneficial microorganisms.

Abstract Background Inequality of opportunity (IOp) stemming from social circumstances exists in outpatient service utilization for the multimorbid elderly in China. However, little is known regarding the magnitude of the IOp and its composition. Therefore, this study aims to measure the IOp in outpatient expenditure and provide potential pathways for policy reform by assessing the contribution of each circumstance. Methods This study included 3527 elderly aged ≥ 65 years with multimorbidity from the Chinese Longitudinal Healthy Longevity Study conducted in 2017–2018. An ordinary least squares regression model was used to analyze the circumstance-influencing factors of outpatient expenditure. The parametric approach was performed to quantify the IOp in outpatient expenditure and the Shapley value decomposition method was employed to determine the contribution of each circumstance. By extracting heterogeneity in the residual of the circumstance-dependent equation of outpatient expenditure across circumstance groups divided based on cluster analysis, we captured the effect of unobserved circumstances. Results Except for pension and distance to health facilities, all the associations between circumstance and outpatient expenditure were statistically significant. The inequality caused by circumstances accounted for 25.18% of the total inequality. The decomposition results revealed that the reimbursement rate contributed 82.92% of the IOp, followed by education duration (4.55%), household registration (3.21%), household income (3.18%), pension (1.49%), medical insurance (1.26%), physical labor (0.99%), unobserved circumstances (0.86%), distance to health facilities (0.83%) and region (0.71%). Conclusions The priority of policy enhancement is to effectively improve the outpatient reimbursement benefit for treating chronic diseases. Additional crucial actions include enhancing the health literacy of the multimorbid elderly to promote the shift from medical needs to demands and accelerating the construction of rural capacity for providing high-quality healthcare to the elderly with multimorbidity.

Abstract A novel α-tocopherol-producing bacteria strain was isolated and identified as Bacillus tequilensis QH1 (QH1). The effects of QH1 strain inoculation alone (QH), combined with cell wall degradation enzyme (QC), or Lactiplantibacillus plantarum subsp. plantarum YM3 and cell wall degradation enzyme (QLC) as the additives on the fermentation characteristics, structural carbohydrates, and α-tocopherol content of rice straw and corn stover silages were investigated, and without additives as the control (CON), in 4 replicates. After silages were kept in a laboratory cabinet (26–34 ℃) and the dark for 42 d, experimental silos (polyethylene plastic bags) were opened for analysis. More and more clinical evidence shows that occupational medicamentose-like dermatitis due to trichloroethylene (OMDT) patients often present immune kidney damage. However, the exact mechanisms of cell-to-cell transmission in TCE-induced immune kidney damage remain poorly understood. The present study aimed to explore the role of high mobility group box-1 (HMGB 1) in glomerular endothelial cell-podocyte transmission. 17 OMDT patients and 34 controls were enrolled in this study. We observed that OMDT patients had renal function injury, endothelial cell activation and podocyte injury, and these indicators were associated with serum HMGB 1. To gain mechanistic insight, a TCE-sensitized BALB/c mouse model was established under the interventions of sirtuin 1 (SIRT 1) activator SRT 1720 (0.1 ml, 5 mg/kg) and receptor for advanced glycation end products (RAGE) inhibitor FPS-ZM 1 (0.1 ml, 1.5 mg/kg). We identified HMGB 1 acetylation and its endothelial cytoplasmic translocation following TCE sensitization, but SRT 1720 abolished the process. RAGE was located on podocytes and co-precipitated with extracellular acetylated HMGB 1, promoting podocyte injury, while SRT 1720 and FPS-ZM 1 both alleviated podocyte injury. The results demonstrate that interventions to upstream and downstream pathways of HMGB 1 may weaken glomerular endothelial cell-podocyte transmission, thereby alleviating TCE-induced immune renal injury.

The co-existence of hydroxylamine (NH2OH) and nitrite (NO2–-N) can aggravate the difficulty of wastewater treatment. The roles of hydroxylamine (NH2OH) and nitrite (NO2–-N) in accelerating the elimination of multiple nitrogen sources by a novel isolated strain of Acinetobacter johnsonii EN-J1 were investigated in this study. The results demonstrated that strain EN-J1 could eliminate 100.00% of NH2OH (22.73 mg/L) and 90.09% of NO2–-N (55.32 mg/L), with maximum consumption rates of 1.22 and 6.75 mg/L/h, respectively. Prominently, the toxic substances NH2OH and NO2–-N could both facilitate nitrogen removal rates. Compared with the control treatment, the elimination rates of nitrate (NO3–-N) and NO2–-N were enhanced by 3.44 and 2.36 mg/L/h after supplementation with 10.00 mg/L NH2OH, and those of ammonium (NH4+-N) and NO3–-N were improved by 0.65 and 1.00 mg/L/h after the addition of 50.00 mg/L NO2–-N. Furthermore, the nitrogen balance results indicated that over 55.00% of the initial total nitrogen was transformed into gaseous nitrogen by heterotrophic nitrification and aerobic denitrification (HN-AD). Ammonia monooxygenase (AMO), hydroxylamine oxidoreductase (HAO), nitrate reductase (NR), and nitrite reductase (NIR), which are essential for HN-AD, were detected at levels of 0.54, 0.15, 0.14, and 0.01 U/mg protein, respectively. All findings confirmed that strain EN-J1 could efficiently execute HN-AD, detoxify NH2OH and NO2–-N, and ultimately promote nitrogen removal rates.

Moso bamboo (Phyllostachys Pubescens) expansion into adjacent forests has been widely reported to affect plant diversity and its association with mycorrhizal fungi in subtropical China, which will likely have significant impacts on soil respiration. However, there is still limited information on how Moso bamboo expansion changes soil respiration components and their linkage with microbial community composition and activity. Based on a mesh exclusion method, soil respirations derived from roots, arbuscular mycorrhizal (AM) mycelium, and free-living microbes were investigated in a pure Moso bamboo forest (expanded), an adjacent broadleaved forest (non-expanded), and a mixed bamboo-broadleaved forest (expanding). Our results showed that bamboo expansion decreased the cumulative CO2 effluxes from total soil respiration, root respiration and soil heterotrophic respiration (by 19.01%, 30.34%, and 29.92% on average), whereas increased those from AM mycelium (by 78.67% in comparison with the broadleaved forests). Bamboo expansion significantly decreased soil organic carbon (C) content, bacterial and fungal abundances, and enzyme activities involved in C, N and P cycling whereas enhanced the interactive relationships among bacterial communities. In contrast, the ingrowth of AM mycelium increased the activities of β-glucosidase and N-acetyl-β-glucosaminidase and decreased the interactive relationships among bacterial communities. Changes in soil heterotrophic respiration and AM mycelium respiration had positive correlations with soil enzyme activities and fungal abundances. In summary, our findings suggest that bamboo expansion decreased soil heterotrophic respiration by decreasing soil microbial activity but increased the contribution of AM mycelial respiration to soil C efflux, which may potentially increase soil C loss from AM mycelial pathway.

Niclosamide is a drug used to treat parasitic infections. Recent studies have shown that niclosamide may have a wide range of clinical applications and can be used to treat cancer and other diseases. However, its application is also limited by its water solubility and safety, and drug resistance to cancer. To solve these problems, some studies have shown that niclosamide can be used in combination with chemotherapeutic drugs, targeted drugs, radiotherapy, and immunotherapy to enhance the anti-tumor effect. This review summarizes the drug combination strategies and therapeutic effect of niclosamide, to provide a reference for the combination therapy of niclosamide and wider application of antitumor drugs.

RT-qPCR remains a vital approach for molecular biology studies aimed at quantifying gene expression in a range of physiological or pathological settings. However, the use of appropriate reference genes is essential to attain meaningful RT-qPCR results. Anastatus japonicus Ashmead (Hymenoptera: Helicopteridae) is an important egg parasitoid wasp and natural enemy of fruit bugs and forest caterpillars. While recent transcriptomic studies have analyzed gene expression profiles in A. japonicus specimens, offering a robust foundation for functional research focused on this parasitoid, no validated A. japonicus reference genes have yet been established, hampering further research efforts. Accordingly, this study sought to address this issue by screening for the most stable internal reference genes in A. japonicus samples to permit reliable RT-qPCR analyses. The utility of eight candidate reference genes (ACTIN, TATA, GAPDH, TUB, RPL13, RPS6, EF1α, RPS3a) was assessed under four different conditions by comparing developmental stages (larvae, pupae, adults), tissues (abdomen, chest, head), sex (male or female adults), or diapause states (diapause induction for 25, 35, 45, or 55 days, or diapause termination). RefFinder was used to calculate gene stability based on the integration of four algorithms (BestKeeper, Normfinder, geNorm, and ΔCt method) to determine the optimal RT-qPCR reference gene. Based on this approach, RPS6 and RPL13 were found to be the most reliable reference genes when assessing different stages of development, while ACTIN and EF1α were optimal when comparing adults of different sexes, RPL13 and EF1α were optimal when analyzing different tissues, and TATA and ACTIN were optimal for different diapause states. These results provide a valuable foundation for future RT-qPCR analyses of A. japonicus gene expression and function under a range of experimental conditions.

BackgroundThe national essential public health service (NEPHS) has been in operation for more than a decade. Numerous studies examined the utilization of NEPHS by migrants and the factors that influence it, but few examined the effect of NEPHS awareness and utilization on the health of inhabitants, particularly migrants. The purpose of this study is to ascertain the level of awareness and utilization of NEPHS, as well as to examine their health-improving effects on migrants.MethodsBased on the data from the 2017 China Migrants Dynamic Survey, linear probability model, ordered logit model and the propensity score matching methods were employed to investigate impact of awareness and utilization of NEPHS on the health among Chinese migrants. Mediating effect model were used to identify the mechanism of the impact of NEPHS on health.ResultsThe findings indicated that migrants' awareness and utilization of NEPHS are still insufficient. After adjusting for other factors, the study discovered that increased awareness and use of NEPHS had a beneficial influence on migrants' self-rated health. Further heterogeneity analysis revealed significant disparities in the health consequences of NEPHS awareness and utilization across subgroups. The effect of increased awareness and usage of NEPHS on health is stronger for middle-aged and elderly people, women, and low-educated migrants with urban household registration. The estimated results of the mediating effect model supported the mechanism that increased NEPHS awareness among the floating population could encourage its utilization and further improve the floating population's health.ConclusionsGiven that migrants' NEPHS utilization is still low and that NEPHS utilization has a positive effect on health, some targeted strategies, such as a variety of new media communication methods, health education related to occupational disease and tuberculosis prevention, and targeted NEPHS projects for specific groups, such as men, young and middle-aged groups, those with a high level of education, and rural migrants, should be conducted to improve the health of migrants.

Trichloroethylene (TCE) hypersensitivity syndrome (THS), called occupational medicamentosa-like dermatitis due to TCE (OMDT) in China, is a fatal occupational disorder caused by TCE exposure. Visceral damage, including kidney injury, is one of the major complications. Necroptosis is a regulated cell death form linked to local inflammatory response. This study aimed to investigate whether renal cell necroptosis was involved in TCE-induced kidney injury. A Balb/c mouse model of TCE sensitization was utilized to study mechanisms of modulation of TCE-induced renal necroptosis. Renal histology (using light and transmission electron microscopy) and renal tubular impairment indexes, including α1-microglobulin (α1-MG), and β2-microglobulin (β2-MG), were evaluated. In addition, tissue expression of necroptosis-related proteins, including tumor necrosis factor (TNF)-α, TNF receptor 1 (TNFR1), receptor-interacting protein kinase 3 (RIPK3), p-RIK3, mixed lineage kinase domain-like protein (MLKL), and p-MLKL, were also evaluated. The study here confirmed TCE sensitization caused damage to renal tubules and renal tubular epithelial cell (RTEC) necroptosis. In mice treated with R7050 (a specific TNFα antagonist), it was also seen that inhibition of TNFα expression could effectively inhibit RTEC necroptosis and improve renal function in the TCE-sensitized mice. Taken together, these results help to define a novel mechanism by which RTEC necroptosis plays a key role in TCE-induced kidney damage.

Biological nitrogen removal is primarily conducted by bacteria and fungi rather than actinomycetes. However, accumulations of nitrite and hydroxylamine could significantly impair the biological nitrogen removal process. A strain of Streptomyces mediolani, termed EM-B2, was isolated from a cow dung fermentation biogas digester. The strain removed more than 99% of ammonium and 78% of total nitrogen in the presence of glucose and under environmental conditions of 30 °C, a carbon/nitrogen ratio of 15, 7.4 mg/L dissolved oxygen and a pH range of 7.5–9.0. Maximal removal rates were 2.29 mg/L/h for ammonium, 1.90 mg/L/h for nitrate and 2.01 mg/L/h for nitrite. The removal efficiencies of hydroxylamine and total nitrogen peaked at 81.48% and 60.38%, respectively. Notably, hydroxylamine and nitrite were never detected during the heterotrophic nitrification and aerobic denitrification. Nitrate rather than nitrite was accumulated from the process of hydroxylamine oxidation. These findings indicate that S. mediolani strain EM-B2 performs heterotrophic nitrification and aerobic denitrification, and can be used to remove hydroxylamine and nitrite from wastewater.

Trichloroethylene (TCE) is a serious health hazard for workers with daily exposure, causing occupational medicamentosa-like dermatitis due to TCE (OMDT) and glomerular damage. Recent studies suggest that mTORC1 signaling is activated in various glomerular disorders; however, the role of mTORC1 signaling in TCE-induced glomerular damage remains to be explored. In the present study, 6 OMDT patients were enrolled and a TCE-sensitized mouse model was established to investigate molecular mechanisms underlying the glomerular damage associated with OMDT. Glomerular damage was assessed by levels of urine nephrin, H&E staining, and renal function test. Ultrastructural change of podocyte was investigated by transmission electron microscopy. The podocyte-related molecules including nephrin, α-actinin-4, and integrin β1 were visualized by immunofluorescence. The activation of mTORC1 signaling was confirmed by Western blot. Glomerular apoptosis was examined by the TUNEL test and Western blotting. Expression and location of cathepsin L (CTSL) were assessed by RT-PCR and immunofluorescence. Our results showed that TCE sensitization caused damage to glomerular structural integrity and also increased the activation of mTORC1 signaling, which was accompanied by podocyte loss, hypertrophy, and glomerular apoptosis. Importantly, we also found that over-expressed CTSL was mainly located in podocyte and CTSL inhibition could partially block the activation of mTORC1 signaling. Thus, our findings suggested a novel mechanism whereby hyperactive mTOR signaling contributes to TCE sensitization–induced and immune-mediated glomerular damage via CTSL activation.

Abstract Our study presents the genetic landscape betel quid chewing‐associated tongue carcinomas (BQ‐TCs). We compared the genetic landscape and mutational signatures of 15 BQ‐TCs, five nonbetel quid chewing‐associated tongue carcinomas (nBQ‐TCs), and 82 tongue carcinomas in general population from the TCGA (TCGA‐TCs) project. The highlights of this research mainly include: (a) The genetic landscape of BQ‐TC was characterized with frequent mutations in RASA1 gene and in CpG islands throughout the genome. (b) The BQ‐TC had a distinct mutational signature from that of nBQ‐TC and tongue carcinomas in the general population, and this signature was associated with the mutations in RASA1 and in CpG islands. (c) Our study indicates that betel quid (BQ) chewing classifies a distinct group of tongue carcinoma. The BQ chewing might not contribute to the tumorigenesis of tongue carcinomas as a mutagen.

To gain better insight into the regulatory networks of anthocyanin biosynthesis, an integrated analysis of the metabolome and transcriptome in purple and green leaves of Tetrastigma hemsleyanum was conducted. Transcript and metabolite profiles were archived by RNA-sequencing data analysis and LC-ESI-MS/MS, respectively. There were 209 metabolites and 4211 transcripts that were differentially expressed between purple and green leaves. Correlation tests of anthocyanin contents and transcriptional changes showed 141 significant correlations (Pearson correlation coefficient >0.8) between 16 compounds and 14 transcripts involved in the anthocyanin biosynthesis pathway. Some novel genes and metabolites were discovered as potential candidate targets for the improvement of anthocyanin content and superior cultivars.

Cross-linking is an effective way to enhance biodegradable poly(propylene carbonate) (PPC) from CO2 and propylene oxide (PO). Cross-linked PPC can be prepared by one-step terpolymerization of multifunctional third monomers with CO2 and PO. However, few such third monomers are available. Each molecule of maleic anhydride oligomer (MAO) contains more than two cyclic anhydride groups. Here, we use it to synthesize PPC with cross-linked networks by adding a small quantity of MAO (0.625−5 wt% of PO) in CO2/PO copolymerization that was catalyzed by zinc glutarate. The formation of networks in the prepared copolymers was confirmed by the presence of gel in copolymers combined Fourier transform infrared spectroscopy (FT-IR), 1H NMR, and the improved mechanical properties. The 5% weight-loss degradation temperatures and maximum weight-loss degradation temperatures greatly increase up to 289.8 °C and 308.8 °C, respectively, which are remarkably high when compared to those of PPC. The minimum permanent deformation of the copolymers closes to 0, while that of PPC is 173%. The maximum tensile strength of the copolymers is 25.5 MPa higher than that of PPC, reaching 38.4 MPa, and it still has some toughness with the elongation at break of 25%. The above phenomena indicate that MAO that was inserted in PPC chains play a cross-linking role, which results in enhanced thermal stability, dimensional stability, and mechanical strength, comprehensively.

The thermoplastic poly(propylene carbonate) (PPC) containing cross-linked networks was one-pot synthesized by copolymerization of carbon dioxide, propylene oxide (PO), maleic anhydride (MA), and furfuryl glycidyl ether (FGE). The copolymers were characterized by Fourier transform infrared spectroscopy (FT-IR), gel permeation chromatography (GPC), differential scanning calorimetry (DSC), and thermogravimetric analysis (TGA) measurements. The thermal and dimensional stability of the copolymers were improved. When the MA and FGE load increased from 1 mol% to 4 mol% of PO, the copolymers contained the gel contents of 11.0%–26.1% and their yields were about double that of the PPC. The 5% weight-loss degradation temperatures (Td,-5%) and the maximum weight-loss degradation temperatures (Td,max) increased from 149.7–271.3 °C and from 282.6–288.6 °C, respectively, corresponding to 217.1 °C and 239.0 °C of PPC. Additionally, the hot-set elongation tests showed that the copolymers exhibited elasticity and dimensional stability with the minimum permanent deformation of 6.5% which was far less than that of PPC of 157.2%, while the tensile strengths were a little lower than that of PPC because of the following two conflicting factors, cross-links and flexibility of the units formed by the introduced third monomers, MA and FGE. In brief, we provide a novel method of one-pot synthesis of PPC containing cross-linked networks. According to this idea, the properties would be more extensively regulated by changing the cross-linkable monomers.
