Abstract The aim is to address the limitations of conventional sliding mode control for PMSM, namely the problems of sliding mode oscillation, load disturbance, and poor control accuracy. An improved sliding mode control strategy for PMSM is proposed based on the novel convergence rate and extended state observer. Firstly, an integral sliding mode velocity loop controller is designed based on the novel convergence rate to solve the sliding mode vibration problem. The stability of the proposed control strategy is derived by the Lyapunov criterion. Secondly, an extended state observer capable of estimating the disturbance torque is designed for the purpose of disturbance compensation. The disturbance torque estimate is then used as a feed-forward signal for the sliding mode velocity loop controller. A fast second-order sliding mode controller is used for the current loop to avoid the potential introduction of noise and disturbances. The simulation results show that compared to alternative control strategies, the proposed control strategy enables the PMSM to respond effectively to start-up and load disturbances, significantly reduces the sliding mode jitter, and improves both the torque estimation accuracy and the system control accuracy.

Prokaryotes play a crucial role in marine ecosystem health and drive biogeochemical processes. The northern Ninety East Ridge (NER) of the Indian Ocean, a pivotal yet understudied area for these cycles, has been the focus of our study. We employed high-throughput 16S rRNA gene sequencing to analyze 35 water samples from five stations along the ridge, categorized into three depth- and dissolved oxygen-level-based groups. Our approach uncovered a clear stratification of microbial communities, with key bioindicators such as Prochlorococcus MIT9313, Sva0996 marine group, and Candidatus Actinomarina in the upper layer; Ketobacter, Pseudophaeobacter, Nitrospina, and SAR324 clade in the middle layer; and Methylobacterium-Methylorubrum, Sphingomonas, Sphingobium, and Erythrobacter in the deep layer. Methylobacterium-Methylorubrum emerged as the most abundant bacterial genus, while Nitrosopumilaceae predominated among archaeal communities. The spatial and depth-wise distribution patterns revealed that Ketobacter was unique to the northern NER, whereas Methylobacterium-Methylorubrum, UBA10353, SAR324 clade, SAR406, Sva0996_marine_group, Candidatus Actinomarina were ubiquitous across various marine regions, exhibiting niche differentiation at the OTU level. Environmental factors, especially dissolved oxygen (DO), silicate, nitrate, and salinity, significantly influence community structure. These findings not only reveal the novelty and adaptability of the microbial ecosystem in the northern NER but also contribute to the broader understanding of marine microbial diversity and its response to environmental heterogeneity.

Abstract Recombinant adeno-associated viruses (rAAVs) have emerged as promising gene therapy vectors due to their proven efficacy and safety in clinical applications. In non-human primates (NHPs), rAAVs are administered via suprachoroidal injection at a higher dose. However, high doses of rAAVs tend to increase additional safety risks. Here, we present a novel AAV capsid (AAVv128), which exhibits significantly enhanced transduction efficiency for photoreceptors and retinal pigment epithelial (RPE) cells, along with a broader distribution across the layers of retinal tissues in different animal models (mice, rabbits, and NHPs) following intraocular injection. Notably, the suprachoroidal delivery of AAVv128-anti-VEGF vector completely suppresses the Grade IV lesions in a laser-induced choroidal neovascularization (CNV) NHP model for neovascular age-related macular degeneration (nAMD). Furthermore, cryo-EM analysis at 2.1 Å resolution reveals that the critical residues of AAVv128 exhibit a more robust advantage in AAV binding, the nuclear uptake and endosome escaping. Collectively, our findings highlight the potential of AAVv128 as a next generation ocular gene therapy vector, particularly using the suprachoroidal delivery route.

With inspiration from natural systems, various soft actuators and robots have been explored in recent years with versatile applications in biomedical and engineering fields. Soft active materials with rich stimulus-responsive characteristics have been an ideal candidate to devise these soft machines by using different manufacturing technologies. Among these technologies, three-dimensional (3D) printing shows advantages in fabricating constructs with multiple materials and sophisticated architectures. In this Review, we aim to provide an overview of recent progress on 3D printing of soft materials, robotics performances, and representative applications. Typical 3D printing techniques are briefly introduced, followed by state-of-the-art advances in 3D printing of hydrogels, shape memory polymers, liquid crystalline elastomers, and their hybrids as soft actuators and robots. From the perspective of material properties, the commonly used printing techniques and action-generation principles for typical printed constructs are discussed. Actuation performances, locomotive behaviors, and representative applications of printed soft materials are summarized. The relationship between printing structures and action performances of soft actuators and robots is also briefly discussed. Finally, the advantages and limitations of each soft material are compared, and the remaining challenges and future directions in this field are prospected.

Tetrazole is widely utilized as a bioisostere for carboxylic acid in the field of medicinal chemistry and drug development, enhancing the drug-like characteristics of various molecules. Typically, tetrazoles are introduced from their nitrile precursors through late-stage functionalization. In this work, we propose a novel strategy involving the use of diversely protected, unprecedented tetrazole aldehydes as building blocks. This approach facilitates the incorporation of the tetrazole group into multicomponent reactions or other chemistries, aiding in the creation of a variety of complex, drug-like molecules. These innovative tetrazole building blocks are efficiently and directly synthesized using a Passerini three-component reaction (PT-3CR), employing cost-effective and readily available materials. We further showcase the versatility of these new tetrazole building blocks by integrating the tetrazole moiety into various multicomponent reactions (MCRs), which are already significantly employed in drug discovery. This technique represents a unique and complementary method to existing tetrazole synthesis processes. It aims to meet the growing demand for tetrazole-based compound libraries and novel scaffolds, which are challenging to synthesize through other methods.

Abstract Background Neoadjuvant therapy followed by surgery has become the standard of care for locally advanced esophageal squamous cell carcinoma (ESCC) and accurate pathological response assessment is critical to assess the therapeutic efficacy. However, it can be laborious and inconsistency between different observers may occur. Hence, we aim to develop an interpretable deep-learning model for efficient pathological response assessment following neoadjuvant therapy in ESCC. Methods This retrospective study analyzed 337 ESCC resection specimens from 2020–2021 at the Pudong-Branch (Cohort 1) and 114 from 2021–2022 at the Puxi-Branch (External Cohort 2) of Fudan University Shanghai Cancer Center. Whole slide images (WSIs) from these two cohorts were generated using different scanning machines to test the ability of the model in handling color variations. Four pathologists independently assessed the pathological response. The senior pathologists annotated tumor beds and residual tumor percentages on WSIs to determine consensus labels. Furthermore, 1850 image patches were randomly extracted from Cohort 1 WSIs and binarily classified for tumor viability. A deep-learning model employing knowledge distillation was developed to automatically classify positive patches for each WSI and estimate the viable residual tumor percentages. Spatial heatmaps were output for model explanations and visualizations. Results The approach achieved high concordance with pathologist consensus, with an R^2 of 0.8437, a RAcc_0.1 of 0.7586, a RAcc_0.3 of 0.9885, which were comparable to two senior pathologists (R^2 of 0.9202/0.9619, RAcc_0.1 of 8506/0.9425, RAcc_0.3 of 1.000/1.000) and surpassing two junior pathologists (R^2 of 0.5592/0.5474, RAcc_0.1 of 0.5287/0.5287, RAcc_0.3 of 0.9080/0.9310). Visualizations enabled the localization of residual viable tumor to augment microscopic assessment. Conclusion This work illustrates deep learning's potential for assisting pathological response assessment. Spatial heatmaps and patch examples provide intuitive explanations of model predictions, engendering clinical trust and adoption (Code and data will be available at https://github.com/WinnieLaugh/ESCC_Percentage once the paper has been conditionally accepted). Integrating interpretable computational pathology could help enhance the efficiency and consistency of tumor response assessment and empower precise oncology treatment decisions.

Abstract Deep learning‐based image registration (DLIR) has been widely developed, but it remains challenging in perceiving small and large deformations. Besides, the effectiveness of the DLIR methods was also rarely validated on the downstream tasks. In the study, a multi‐scale complexity‐aware registration network (MSCAReg‐Net) was proposed by devising a complexity‐aware technique to facilitate DLIR under a single‐resolution framework. Specifically, the complexity‐aware technique devised a multi‐scale complexity‐aware module (MSCA‐Module) to perceive deformations with distinct complexities, and employed a feature calibration module (FC‐Module) and a feature aggregation module (FA‐Module) to facilitate the MSCA‐Module by generating more distinguishable deformation features. Experimental results demonstrated the superiority of the proposed MSCAReg‐Net over the existing methods in terms of registration accuracy. Besides, other than the indices of Dice similarity coefficient (DSC) and percentage of voxels with non‐positive Jacobian determinant (|Jϕ|≤0), a comprehensive evaluation of the registration performance was performed by applying this method on a downstream task of multi‐atlas hippocampus segmentation (MAHS). Experimental results demonstrated that this method contributed to a better hippocampus segmentation over other DLIR methods, and a comparable segmentation performance with the leading SyN method. The comprehensive assessment including DSC, |Jϕ|≤0, and the downstream application on MAHS demonstrated the advances of this method.

Acute kidney injury (AKI) constitutes a significant public health issue. Sepsis accounts for over 50 % of AKI cases in the ICU. Recent findings from our research indicated that the PRD1-BF1-RIZ1 homeodomain protein 16 (PRDM16) inhibited the progression of diabetic kidney disease (DKD). However, its precise role and regulatory mechanism in sepsis-induced AKI remain obscure. This study reveals that lipopolysaccharide (LPS) and cecum ligation and puncture (CLP) instigated PRDM16 expression in Boston University mouse proximal tubule (BUMPT) cells and mouse kidneys, respectively. Functionally, PRDM16 curtailed LPS-induced ferroptosis. Mechanistically, PRDM16 associates with the promoter regions of nuclear factor-erythroid 2-related factor-2 (NRF2) and augments its expression, subsequently enhancing glutathione peroxidase 4 (GPX4) expression. Additionally, PRDM16 directly engages with the promoter regions of GPX4, stimulating its expression. Notably, these observations were corroborated in human renal tubular epithelial (HK-2) cells. Furthermore, the ablation of PRDM16 from kidney proximal tubules in mice inhibited NRF2 and GPX4 expression, leading to decreased glutathione (GSH)/oxidized glutathione (GSSG) ratio, increased Fe2+ and reactive oxygen species (ROS) production, exacerbated ferroptosis, and AKI progression. Conversely, PRDM16 knock-in exhibited the opposite effects. Ultimately, adenovirus (ADV)-PRDM16 plasmid or poly (lactide-glycolide acid) (PLGA)-encapsulated formononetin not only mitigated sepsis-induced AKI but also alleviated liver, cardiac, and lung injury. In summary, PRDM16 inhibits ferroptosis via the NRF2/GPX4 axis or GPX4 to prevent sepsis-induced multi-organ injury, including AKI. PLGA-encapsulated formononetin presents a promising therapeutic approach.

Serious bubble clogging in flow-field channels will hinder the water supply to the electrode of proton exchange membrane water electrolyzer (PEMWE), deteriorating the cell performance. In order to address this issue, the dual-layer flow field design has been proposed in our previous study. In this study, the VOF (volume of fluid) method is utilized to investigate the effects of different degassing layer and base heights on the bubble behavior in channel and determine the time for the bubbles to detach from the electrode surface. However, it is very time-consuming to get the optimal combination of base layer and degassing layer heights due to the large number of potential cases, which needs to be calculated through computation-intensive physical model. Therefore, machine learning methods are adopted to accelerate the optimization. A data-driven surrogate model based on deep neural network (DNN) is developed and successfully trained using data obtained by the physical VOF method. Based on the highly efficient surrogate, genetic algorithm (GA) is further utilized to determine the optimal heights of base layer and degassing layer. Finally, the reliability of the optimization was validated by bubble visualization in channel and electrochemical characterization in PEMWE through experiments.

Abstract Creatures, such as Venus flytrap and hummingbirds, capable of rapid predation through snap‐through transition, provide paradigms for the design of soft actuators and robots with fast actions. However, these artificial “snappers” usually need contact stimulations to trigger the flipping. Reported here is a constrained anisotropic poly(N‐isopropylacrylamide) hydrogel showing fast snapping upon light stimulation. This hydrogel is prepared by flow‐induced orientation of nanosheets (NSs) within a rectangular tube. The precursor containing gold nanoparticles is immediately exposed to UV light for photopolymerization to fix the ordered structure of NSs. Two ends of the slender gel are clamped to form a buckle with bistability nature, which snaps to the other side upon laser irradiation. Systematic experiments are conducted to investigate the influences of power intensity and irradiation angle of the laser, as well as thickness and buckle height of the gel, on the snapping behaviors. The fast snapping is further used to kick a plastic bead and control the switch state. Furthermore, synergetic or oscillated snapping of the gel with two buckles of opposite directions is realized by inclined irradiation of a laser or horizontal irradiation with two lasers, respectively. Such light‐steered snapping of hydrogels should merit designing soft robots, energy harvests, etc.

Layered double hydroxides (LDHs) are a class of promising cathode materials for supercapacitors. However, the bad cycling performance has always been the Achilles’ heel of LDHs‐based supercapacitors. In this contribution, a nonelectrochemical active element Scandium (Sc) is doped into NiCo‐LDHs to greatly improve the intrinsic cycling performance of active materials. The trimetallic NiCoSc‐LDHs exhibit an ultralong cycle lifespan with 40 000 charge–discharge cycles (exceeding 50 days) and a high specific capacity of 196 mAh g−1 (1695 F g−1). Sc doping greatly changes the degradation mode of NiCo‐LDHs from rapid decay in thousands of cycles to a two‐stage performance evolution, which consists of a superlong activation process of about 10 000 cycles and then extremely slow degradation. Moreover, Sc doping enhances the electrochemical activity of Ni3+, so as to not only avoid its Jahn–Teller distortion, but also perform as a structural stabilizer to alleviate the local strain of host layer. The assembled asymmetric supercapacitor delivers an ultralong cycling lifespan with 101% capacity retention even after 50 000 cycles. This work presents a new pathway to significantly improve the electrochemical performance, especially the cycling stability of LDH‐based electrodes for high‐performance supercapacitors.

The prevalence of calcific aortic valve disease (CAVD) remains substantial while there is currently no medical therapy available. Forkhead box O1 (FOXO1) is known to be involved in the pathogenesis of cardiovascular diseases, including vascular calcification and atherosclerosis; however, its specific role in calcific aortic valve disease remains to be elucidated. In this study, we identified FOXO1 significantly down-regulated in the aortic valve interstitial cells (VICs) of calcified aortic valves by investigating clinical specimens and GEO database analysis. FOXO1 silencing or inhibition promoted VICs osteogenic differentiation in vitro and aortic valve calcification in Apoe−/− mice, respectively. We identified that FOXO1 facilitated the ubiquitination and degradation of RUNX2, which process was mainly mediated by SMAD-specific E3 ubiquitin ligase 2 (SMURF2). Our discoveries unveil a heretofore unacknowledged mechanism involving the FOXO1/SMURF2/RUNX2 axis in CAVD, thereby proposing the potential therapeutic utility of FOXO1 or SMURF2 as viable strategies to impede the progression of CAVD.

Abstract Electric field-induced second harmonic generation allows electrically controlling nonlinear light-matter interactions crucial for emerging integrated photonics applications. Despite its wide presence in materials, the figures-of-merit of electric field-induced second harmonic generation are yet to be elevated to enable novel device functionalities. Here, we show that the polar skyrmions, a topological phase spontaneously formed in PbTiO3/SrTiO3 ferroelectric superlattices, exhibit a high comprehensive electric field-induced second harmonic generation performance. The second-order nonlinear susceptibility and modulation depth, measured under non-resonant 800 nm excitation, reach ~54.2 pm V−1 and ~664% V−1, respectively, and high response bandwidth (higher than 10 MHz), wide operating temperature range (up to ~400 K) and good fatigue resistance (>1010 cycles) are also demonstrated. Through combined in-situ experiments and phase-field simulations, we establish the microscopic links between the exotic polarization configuration and field-induced transition paths of the skyrmions and their electric field-induced second harmonic generation response. Our study not only presents a highly competitive thin-film material ready for constructing on-chip devices, but opens up new avenues of utilizing topological polar structures in the fields of photonics and optoelectronics.

Abstract Steering soft robots in a self-regulated manner remains a grand challenge, which often requires continuous symmetry breaking and recovery steps for persistent motion. Although structural morphology is found significant for robotic functions, geometric topology has rarely been considered and appreciated. Here we demonstrate a series of knotbots, namely hydrogel-based robots with knotted structures, capable of autonomous rolling and spinning/rotating motions. With symmetry broken by external stimuli and restored by self-regulation, the coupling between self-constraint-induced prestress and photothermal strain animates the knotbots continuously. Experiments and simulations reveal that nonequilibrium processes are regulated dynamically and cooperatively by self-constraints, active deformations, and self-shadowing effect of the photo-responsive gel. The active motions enable the knotbots to execute tasks including gear rotation and rod climbing. This work paves the way to devise advanced soft robots with self-regulated sustainable motions by harnessing the topology.

Abstract The northeastern Indian Ocean exhibits distinct hydrographic characteristics influenced by various local and remote forces. Variations in these driving factors may alter the physiochemical properties of seawater, such as dissolved oxygen levels, and affect the diversity and function of microbial communities. How the microbial communities change across water depths spanning a dissolved oxygen gradient has not been well understood. Here we employed both 16S rDNA amplicon and metagenomic sequencing approaches to study the microbial communities collected from different water depths along the E87 transect in the northeastern Indian Ocean. Samples were collected from the surface, Deep Chlorophyll Maximum (DCM), Oxygen Minimum Zone (OMZ), and bathypelagic layers. Proteobacteria were prevalent throughout the water columns, while Thermoproteota were found to be abundant in the aphotic layers. A total of 675 non-redundant metagenome-assembled genomes (MAGs) were constructed, spanning 21 bacterial and 5 archaeal phyla. The community structure and genomic information provided by this dataset offer valuable resources for the analysis of microbial biogeography and metabolism in the northeastern Indian Ocean.

To study the impact of faults on the enrichment and mobility of shale oil in the Gulong area, representative rock samples were selected in this paper. Based on geochemical data and chemical kinetics methods, coupled with shale oil enrichment and mobility analysis techniques, the shale oil generation quantity and in situ oil content were evaluated from the perspectives of shale oil generation and micro migration, and the mobility of shale oil was revealed. At the same time, the hydrocarbon expulsion efficiency (HEE) of shale was qualitatively and quantitatively characterized, combined with the development of faults. The research results indicate that the study area mainly develops organic-rich felsic (ORF)/organic-containing felsic (OCF) shale, their proportion in both wells exceeds 65%, and the resource amount is the largest in this type of lithofacies. The development of a fault controls the enrichment of shale oil, and the in situ oil content and oil saturation index (OSI) of the shale in well Y58, which is close to the fault, are significantly worse than those in well S2. Well Y58 has 9.52 mg/g and 424.83 mg/g TOC respectively, while well S2 has 11.34 mg/g and 488.73 mg/g TOC respectively. The fault enhanced the migration of shale oil, increasing the efficiency of oil expulsion. As a result, the components with weak polarity or small molecules, such as saturated hydrocarbons and low carbon number n-alkanes, are prone to migration, reducing the mobility of shale oil.

Microbial communities within oxygen minimum zones (OMZs) play crucial roles in the marine biogeochemical cycling. Arabian Sea (AS) has one of the largest OMZs among the global oceans, however, knowledge about the microbial ecology of the AS OMZ remained limited. In the present study, 44 water samples collected from six stations across the AS, spanning from the deep chlorophyll maximum (DCM) layer to 4000m depth were analyzed. High-throughput sequencing of 16S rRNA genes revealed the structural diversity of bacterial and archaeal communities, influenced primarily by depth and dissolved oxygen (DO) levels. Distinct community compositions were observed across different oxygen gradients, with shifts in the relative abundance of key taxa. Notably, Desulfosarcinaceae, UBA10353, Nitrospina, SUP05, Sva0996_marine_group, Microtrichaceae, and Nitrosopumilus emerged as bioindicator taxa in the AS hypoxic zones. Co-occurrence network analysis identified SAR324, Alteromonadaceae, and Sphingomonadaceae as keystone taxa. The spatial and depth-wise distribution patterns revealed that Desulfosarcinaceae was predominantly found in the hypoxic zones of the Arabian Sea, whereas UBA10353, Nitrospina, SUP05, Microtrichaceae and SAR324 were ubiquitous across AS, Bay of Bengal (BOB), and Eastern Tropical North Pacific (ETNP) OMZs, with OTU-level niche differentiation observed for the latter two. Functional profiling using FAPROTAX predicted higher metabolic potential for nitrogen and sulfur in the OMZ compared to other layers of the AS. Our findings provide valuable insights into the distribution, structure, and diversity of microbial communities in the AS OMZ, highlighting the ecological roles of key taxa in hypoxic environments. The established sequence database offers a foundation for further research into the complex interactions within these microbial ecosystems.

High-efficiency electromagnetic interference (EMI) shielding composite films (EMI shielding effectiveness (SE) value > 100 dB) with a small thickness are imminently desired for eliminating the serious electromagnetic wave pollution. Herein, asymmetric sandwiched silver-coated four-needle zinc oxide whisker/nanofibrillated cellulose/Ti3C2Tx (F-ZnO&Ag/NFC/Ti3C2Tx) composite films consisting of the loose F-ZnO&Ag sedimentary layer and compact upper Ti3C2Tx layer were successful fabricated by a facile one-pot vacuum filtration method in combination with gravity-induced delamination. Arising from the favorable electrical conductivity and unique asymmetric sandwich structure, electromagnetic waves can be trapped and dissipated in the F-ZnO&Ag skeletons and strongly multiply reflected between top Ti3C2Tx layer and bottom F-ZnO&Ag layers. Thereby, F-ZnO&Ag/NFC/Ti3C2Tx-30.4 exhibits the ultrahigh averaged EMI SET of 103.9 dB (the maximal value of 123.8 dB) at a low thickness of merely 209 μm, which is far superior to that of almost all the reported NFC composites at the similar thickness to date. In the real EMI shielding application test, composite films are confirmed to block signal transmission and switch off the toy car system. Such simple and high-efficiency asymmetric sandwich structure design makes a great progress towards preparing and adjusting thin EMI shielding composite films for the promising application prospects in advanced electronics and communication technology.

Abstract Mg3(Sb,Bi)2 is a promising thermoelectric material suited for electronic cooling, but there is still room to optimize its low-temperature performance. This work realizes >200% enhancement in room-temperature zT by incorporating metallic inclusions (Nb or Ta) into the Mg3(Sb,Bi)2-based matrix. The electrical conductivity is boosted in the range of 300–450 K, whereas the corresponding Seebeck coefficients remain unchanged, leading to an exceptionally high room-temperature power factor >30 μW cm−1 K−2; such an unusual effect originates mainly from the modified interfacial barriers. The reduced interfacial barriers are conducive to carrier transport at low and high temperatures. Furthermore, benefiting from the reduced lattice thermal conductivity, a record-high average zT > 1.5 and a maximum zT of 2.04 at 798 K are achieved, resulting in a high thermoelectric conversion efficiency of 15%. This work demonstrates an efficient nanocomposite strategy to enhance the wide-temperature-range thermoelectric performance of n-type Mg3(Sb,Bi)2, broadening their potential for practical applications.

Abstract Background The percutaneous iliosacral screw technique represents a global standard fixation method for unstable fractures of the posterior pelvic ring. However, the inaccurate positioning of iliosacral screws is associated with a significant risk of severe intra-operative complications. Therefore, this study aimed to investigate the relationship between the skin entry point of the transverse iliosacral screw of the first sacral vertebral body and the anterior superior iliac spine and the greater trochanter of the femur using computed-tomography-guided validation. Methods Overall, 91 consecutive patients admitted to a tertiary referral center in China for posterior pelvic ring fixation via the “triangulation method” using computed-tomography-guided validation between January 1, 2020, and December 31, 2020, were included in this retrospective observational cohort study. Modeling and simulated iliosacral screw placement were performed using the Mimics software. The distance between the three points of interest was measured, and their relationship in a rectangular coordinate system was determined. Patients were categorized according to gender, body mass index, and femoral rotation angle to investigate the factors affecting the positional relationship between the three points. Results An equilateral triangular relationship was observed between the positioning points of the transverse iliosacral screw, anterior iliac spine, and greater trochanter. Additionally, 95% of the entry points were within a circle radius centered 12 mm at the apex of an equilateral triangle comprising the anterior superior iliac spine and the greater trochanter as the base. The entry point in the femoral external rotation was more dorsal than that in the internal femoral rotation. Furthermore, the entry point in females was more rostral than that in males, and the entry point in overweight patients was more dorsal than that in normal-weight patients. Conclusions The skin entry point of the percutaneous iliosacral screw can be located by drawing an equilateral triangle from the anterior superior iliac spine and the greater trochanter as the base to the dorsum end of the patient’s head. In summary, this retrospective cohort study validated the safety and efficacy of the “triangulation methods” for percutaneous fixation of unstable posterior pelvic ring injuries.

Abstract The SN2 nucleophilic substitution reaction is a vital organic transformation used for drug and natural product synthesis. Nucleophiles like cyanide, oxygen, nitrogen, sulfur, or phosphorous replace halogens or sulfonyl esters, forming new bonds. Isocyanides exhibit unique C-centered lone pair σ and π* orbitals, enabling diverse radical and multicomponent reactions. Despite this, their nucleophilic potential in SN2 reactions remains unexplored. We have uncovered that isocyanides act as versatile nucleophiles in SN2 reactions with alkyl halides. This yields highly substituted secondary amides through in situ nitrilium ion hydrolysis introducing an alternative bond break compared to classical amide synthesis. This novel 3-component process accommodates various isocyanide and electrophile structures, functional groups, scalability, late-stage drug modifications, and complex compound synthesis. This reaction greatly expands chemical diversity, nearly doubling the classical amid coupling’s chemical space. Notably, the isocyanide nucleophile presents an unconventional Umpolung amide carbanion synthon (R-NHC(-) = O), an alternative to classical amide couplings.

The ischemia–reperfusion process of a donor heart during heart transplantation leads to severe mitochondrial dysfunction, which may be the main cause of donor heart dysfunction after heart transplantation. Pyruvate carboxylase (PC), an enzyme found in mitochondria, is said to play a role in the control of oxidative stress and the function of mitochondria. This research examined the function of PC and discovered the signaling pathways controlled by PC in myocardial IRI. We induced IRI using a murine heterotopic heart transplantation model in vivo and a hypoxia–reoxygenation cell model in vitro and evaluated inflammatory responses, oxidative stress levels, mitochondrial function, and cardiomyocyte apoptosis. In both in vivo and in vitro settings, we observed a significant decrease in PC expression during myocardial IRI. PC knockdown aggravated IRI by increasing MDA content, LDH activity, TUNEL-positive cells, serum cTnI level, Bax protein expression, and the level of inflammatory cytokines and decreasing SOD activity, GPX activity, and Bcl-2 protein expression. PC overexpression yielded the opposite findings. Additional research indicated that reducing PC levels could block the Wnt/β-catenin pathway and glutamine metabolism by hindering the movement of β-catenin to the nucleus and reducing the activity of complex I and complex II, as well as ATP levels, while elevating the ratios of NADP+/NADPH and GSSG/GSH. Overall, the findings indicated that PC therapy can shield the heart from IRI during heart transplantation by regulating glutamine metabolism through the Wnt/β-catenin pathway.

Human–machine interactions (HMIs) have penetrated into various academic and industrial fields, such as robotics, virtual reality, and wearable electronics. However, the practical application of most human–machine interfaces faces notable obstacles due to their complex structure and materials, high power consumption, limited effective skin adhesion, and high cost. Herein, we report a self-powered, skin adhesive, and flexible human–machine interface based on a triboelectric nanogenerator (SSFHMI). Characterized by its simple structure and low cost, the SSFHMI can easily convert touch stimuli into a stable electrical signal at the trigger pressure from a finger touch, without requiring an external power supply. A skeleton spacer has been specially designed in order to increase the stability and homogeneity of the output signals of each TENG unit and prevent crosstalk between them. Moreover, we constructed a hydrogel adhesive interface with skin-adhesive properties to adapt to easy wear on complex human body surfaces. By integrating the SSFHMI with a microcontroller, a programmable touch operation platform has been constructed that is capable of multiple interactions. These include medical calling, music media playback, security unlocking, and electronic piano playing. This self-powered, cost-effective SSFHMI holds potential relevance for the next generation of highly integrated and sustainable portable smart electronic products and applications.

Background: This study investigated the impact of salvianolic acids, derived from Danshen, on melanoma cell growth. Specifically, we assessed the ability of salvianolic acid A (Sal A) to modulate melanoma cell proliferation. Methods: We used human melanoma A2058 and A375 cell lines to investigate the effects of Sal A on cell proliferation and death by measuring bromodeoxyuridine incorporation and lactate dehydrogenase release. We assessed cell viability and cycle progression using water soluble tetrazolium salt-1 (WST-1) mitochondrial staining and propidium iodide. Additionally, we used a phospho-kinase array to investigate intracellular kinase phosphorylation, specifically measuring the influence of Sal A on checkpoint kinase-2 (Chk-2) via western blot analysis. Results: Sal A inhibited the growth of A2058 and A375 cells dose-responsively and induced cell cycle arrest at the G2/M phase. Notably, Sal A selectively induces Chk-2 phosphorylation without affecting Chk-1, thereby degrading Chk-2-regulated genes Cdc25A and Cdc2. However, Sal A does not affect the Chk1-Cdc25C pathway. Conclusions: Salvianolic acids, especially Sal A, effectively hinder melanoma cell growth by inducing Chk-2 phosphorylation and disrupting G2/M checkpoint regulation.

Based on the oil and gas exploration practice in the Songliao Basin, combined with the latest exploration and development data such as seismic, well logging and geochemistry, the basic geological conditions, oil and gas types and distribution characteristics, reservoir-forming dynamics, source-reservoir relationship and hydrocarbon accumulation model of the whole petroleum system in shallow and medium strata in the northern part of Songliao Basin are systematically studied. The shallow–medium strata in northern Songliao Basin have the conditions for the formation of whole petroleum system, with sufficient oil and gas sources, diverse reservoir types and well-developed transport system, forming a whole petroleum system centered on the source rocks of the Cretaceous Qingshankou Formation. Different types of oil and gas resources in the whole petroleum system are correlated with each other in terms of depositional system, lithologic association and physical property changes, and they, to a certain extent, have created the spatial framework with orderly symbiosis of shallow–medium conventional oil reservoirs, tight oil reservoirs and shale oil reservoirs in northern Songliao Basin. Vertically, the resources are endowed as conventional oil above source, shale oil/tight oil within source, and tight oil below source. Horizontally, conventional oil, tight oil, interlayer-type shale oil, and pure shale-type shale oil are developed in an orderly way, from the margin of the basin to the center of the depression. Three hydrocarbon accumulation models are recognized for the whole petroleum system in northern Songliao Basin, namely, buoyancy-driven charging of conventional oil above source, retention of shale oil within source, and pressure differential-driven charging of tight oil below source.

Abstract The scaffold protein IRS-1 is an essential node in insulin/IGF signaling. It has long been recognized that the stability of IRS-1 is dependent on its endomembrane targeting. However, how IRS-1 targets the intracellular membrane, and what type of intracellular membrane is actually targeted, remains poorly understood. Here, we found that the phase separation-mediated IRS-1 puncta attached to endoplasmic reticulum (ER). VAPB, an ER-anchored protein that mediates tethers between ER and membranes of other organelles, was identified as a direct interacting partner of IRS-1. VAPB mainly binds active IRS-1 because IGF-1 enhanced the VAPB-IRS-1 association and replacing of the nine tyrosine residues of YXXM motifs disrupted the VAPB-IRS-1 association. We further delineated that the Y745 and Y746 residues in the FFAT-like motif of IRS-1 mediated the association with VAPB. Notably, VAPB targeted IRS-1 to the ER and subsequently maintained its stability. Consistently, ablation of VAPB in mice led to downregulation of IRS-1, suppression of insulin signaling, and glucose intolerance. The amyotrophic lateral sclerosis (ALS)-derived VAPB P56S mutant also impaired IRS-1 stability by interfering with the ER-tethering of IRS-1. Our findings thus revealed a previously unappreciated condensate-membrane contact (CMC), by which VAPB stabilizes the membraneless IRS-1 signalosome through targeting it to ER membrane.

The synthesis of alternative fuels from biomass has emerged to be a vital strategy to maintain sustainable development. Long-chain levulinate esters (LEs) are a class of good biofuel candidates because they have similar structures to biodiesels and superior heating values. The synthesis of long-chain LEs with effective heterogeneous catalysts has always been challenging. In this work, a few heterogeneous phosphotungstate catalysts were facilely prepared, among which the V3+ exchanged phosphotungstic acid catalyst (VPW) was the most active for the alcoholysis of furfuryl alcohol (FA) and 1-hexanol into 1-hexyl levulinate (HL). A maximum HL yield of 63% was achieved at 180 °C. The VPW catalyst could be reused without obvious decrement of activity. The system was also applicable to the alcoholysis of FA with other hexanols and 1-octanol into various long-chain LEs.

In many oceanic regions, anthropogenic warming will coincide with iron (Fe) limitation. Interactive effects between warming and Fe limitation on phytoplankton physiology and biochemical function are likely, as temperature and Fe availability affect many of the same essential cellular pathways. However, we lack a clear understanding of how globally significant phytoplankton such as the picocyanobacteria Synechococcus will respond to these co-occurring stressors, and what underlying molecular mechanisms will drive this response. Moreover, ecotype-specific adaptations can lead to nuanced differences in responses between strains. In this study, Synechococcus isolates YX04-1 (oceanic) and XM-24 (coastal) from the South China Sea were acclimated to Fe limitation at two temperatures, and their physiological and proteomic responses were compared. Both strains exhibited reduced growth due to warming and Fe limitation. However, coastal XM-24 maintained relatively higher growth rates in response to warming under replete Fe, while its growth was notably more compromised under Fe limitation at both temperatures compared with YX04-1. In response to concurrent heat and Fe stress, oceanic YX04-1 was better able to adjust its photosynthetic proteins and minimize the generation of reactive oxygen species while reducing proteome Fe demand. Its intricate proteomic response likely enabled oceanic YX04-1 to mitigate some of the negative impact of warming on its growth during Fe limitation. Our study highlights how ecologically-shaped adaptations in Synechococcus strains even from proximate oceanic regions can lead to differing physiological and proteomic responses to these climate stressors.

Purpose: To investigate the pharmacokinetics and tissue distribution of VGB racemate and its single enantiomers, and explore the potential of clinic development for single enantiomer S-VGB. Methods: In the pharmacokinetics study, male Sprague-Dawley rats were gavaged with VGB racemate or its single enantiomers dosing 50, 100 or 200 mg/kg, and the blood samples were collected during 12 h at regular intervals. In the experiment of tissue distribution, VGB and its single enantiomers were administered intravenously dosing 200 mg/kg, and the tissues including heart, liver, spleen, lung and kidney, eyes, hippocampus, and prefrontal cortex were separated at different times. The concentrations of R-VGB and S-VGB in the plasma and tissues were measured using HPLC. Results: Both S-VGB and R-VGB could be detected in the plasma of rats administered with VGB racemate, reaching Cmax at approximately 0.5 h with t1/2 2–3 h. There was no significant pharmacokinetic difference between the two enantiomers when VGB racemate was given 200 mg/kg and 100 mg/kg. However, when given at the dose of 50 mg/kg, S-VGB presented a shorter t1/2 and a higher Cl/F than R-VGB, indicating a faster metabolism of S-VGB. Furthermore, when single enantiomer was administered respectively, S-VGB presented a slower metabolism than R-VGB, as indicated by a longer t1/2 and MRT but a lower Cmax. Moreover, compared with the VGB racemate, the single enantiomers S-VGB and R-VGB had shorter t1/2 and MRT, higher Cmax and AUC/D, and lower Vz/F and Cl/F, indicating the stronger oral absorption and faster metabolism of single enantiomer. In addition, regardless of VGB racemate administration or single enantiomer administration, S-VGB and R-VGB had similar characteristics in tissue distribution, and the content of S-VGB in hippocampus, prefrontal cortex and liver was much higher than that of R-VGB. Conclusions: Although there is no transformation between S-VGB and R-VGB in vivo, those two enantiomers display certain disparities in the pharmacokinetics and tissue distribution, and interact with each other. These findings might be a possible interpretation for the pharmacological and toxic effects of VGB and a potential direction for the development and optimization of the single enantiomer S-VGB.

Enhancing the alkalinity of the ocean is a promising approach for CO2 removal by promoting marine carbon sequestration. Olivine is a key candidate material for enhancing alkalinity owing to its release of silicates when dissolved in seawater. These released compounds serve as crucial nutrients for phytoplankton such as diatoms to foster their growth, which in turn accelerates olivine dissolution and further enhances carbon sequestration. In this study, we investigated the short-term synergistic CO2 removal effects of an olivine–diatom coculture system. Over a 6-day incubation period, the olivine dissolution was 92% to 144% higher in the olivine–diatom groups compared with the olivine-only groups. The olivine-only groups achieved a CO2 removal efficiency of 5.15% to 5.49%, while the olivine–diatom groups achieved a CO2 removal efficiency of 8.84% to 14.44%. Adding olivine was found to increase the total alkalinity by 70 to 100 μM and the diatom abundance by 26.4% to 58.4%. Diatom growth and the fixed carbon content were greatly enhanced, particularly during the later silicate-depleted stage when the Si:C ratio significantly exceeded that of groups without olivine. This mutually beneficial olivine–diatom coculture system offers a highly efficient CO2 removal strategy for addressing climate change. The results of this study contribute to our understanding of carbonate and biological carbon pump processes.

Automatic segmentation of pulmonary vessels is a fundamental and essential task for the diagnosis of various pulmonary vessels diseases. The accuracy of segmentation is suffering from the complex vascular structure. In this paper, an Improved Residual Attention U-Net (IRAU-Net) aiming to segment pulmonary vessel in 3D is proposed. To extract more vessel structure information, the Squeeze and Excitation (SE) block is embedded in the down sampling stage. And in the up sampling stage, the global attention module (GAM) is used to capture target features in both high and low levels. These two stages are connected by Atrous Spatial Pyramid Pooling (ASPP) which can sample in various receptive fields with a low computational cost. By the evaluation experiment, the better performance of IRAU-Net on the segmentation of terminal vessel is indicated. It is expected to provide robust support for clinical diagnosis and treatment.

Synechococcus are widely distributed in the global ocean, from the pelagic zone to coastal waters. However, little is known about Synechococcus in coastal seawater due to limitations in isolation and culture conditions. In this study, a combination of metagenomic sequencing technology, flow cytometry sorting, and multiple displacement amplification was used to investigate Synechococcus in the coastal seawater of Xiamen Island. The results revealed 18 clades of Synechococcus and diverse metabolic genes that appear to contribute to their adaptation to the coastal environment. Intriguingly, some metabolic genes related to the metabolism of carbohydrates, energy, nucleotides, and amino acids were found in 89 prophage regions that were detected in 16,258 Synechococcus sequences. The detected metabolic genes can enable prophages to contribute to the adaptation of Synechococcus hosts to the coastal marine environment. The detection of prophages also suggests that the cyanophages have infected Synechococcus. On the other hand, the identification of 773 genes associated with antiviral defense systems indicates that Synechococcus in Xiamen have evolved genetic traits in response to cyanophage infection. Studying the community diversity and functional genes of Synechococcus provides insights into their role in environmental adaptation and marine ecosystems.

The Ag@MgO nanoparticles (NPs), consisting of silver NPs with a size range of 6–15 nm well-dispersed on the surface of MgO nanoparticles, were synthesized using a simple and efficient in situ synthesis method. These NPs exhibited superior antibacterial activity compared to bare MgO NPs. Subsequently, they were incorporated into poly(butylene succinate-co-terephthalate) (PBST) matrix to prepare antibacterial food packaging films. The effects of Ag@MgO NPs morphology on the compatibility, mechanical, water vapor barrier, UV blocking and antibacterial properties of PBST matrix were evaluated. Unmodified Ag@MgO NPs caused agglomeration and poor mechanical properties. However, when the Ag@MgO NPs were surface modified and interacted with the PBST matrix, the dispersion of nanoparticles and crystallinity improved, resulting in enhanced tensile strength and barrier properties of the composite films. Furthermore, the well-formed of Ag@MgO NPs in the films played a positive role in controlling the growth of microorganisms. Upon incorporation of the modified Ag@MgO NPs, the PBST nanocomposite films exhibited a 23.5% increase in the tensile strength and a 63% decrease in the water vapor permeability compared to PBST films. Moreover, the UV blocking performances were enhanced, and high antibacterial rates of 99.8% and 99.7% were achieved against E. coli and S. aureus, respectively. All the results indicate that the PBST incorporating modified Ag@MgO NPs have excellent potential for use in food packaging applications.

In recent years, the Alzheimer’s disease (AD) epidemic has become one of the largest global healthcare crises. Besides, the available systemic therapies for AD are still inadequate. Due to the insufficient therapeutic options, new treatment strategies are urgently needed to achieve a satisfactory therapeutic effect. Marine bio-resources have been accepted as one of the most economically viable and sustainable sources with potential applications for drug discovery and development. In this study, a marine cyanobacteria–Synechococcus sp. XM-24 was selected as the object of research, to systematically investigate its therapeutic potential mechanisms for AD. The major active compounds derived from the Synechococcus sp. biomass were identified via pyrolysis-gas chromatography-mass spectrometry (GC-MS), and 22 compounds were identified in this strain. The most abundant chemical compounds was (E)-octadec-11-enoic acid, with the peak area of 30.6%. Follow by tridecanoic acid, 12-methyl- and hexadecanoic acid, with a peak area of 23.26% and 18.23%, respectively. GC-MS analysis also identified indolizine, isoquinoline, 3,4-dihydro- and Phthalazine, 1-methyl-, as well as alkene and alkane from the strain. After the chemical toxicity test, 10 compounds were finally collected to do the further analysis. Then, network pharmacology and molecular docking were adopted to systematically study the potential anti-AD mechanism of these compounds. Based on the analysis, the 10 Synechococcus-derived active compounds could interact with 128 related anti-AD targets. Among them, epidermal growth factor receptor (EGFR), vascular endothelial growth factor A (VEGFA) and mitogen-activated protein kinase 3 (MAPK3) were the major targets. Furthermore, the compounds N-capric acid isopropyl ester, (E)-octadec-11-enoic acid, and 2H-Pyran-2,4(3H)-dione, dihydro-6-methyl- obtained higher degrees in the compounds-intersection targets network analysis, indicating these compounds may play more important role in the process of anti-AD. In addition, Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis showed that these active compounds exert the anti-AD effects mainly through PI3K-Akt signaling pathway, neuroactive ligand-receptor interaction and ras signaling pathway. Our study identified Synechococcus-derived bioactive compounds have the potential for application to AD by targeting multiple targets and related pathways, which will provide a foundation for future research on applications of marine cyanobacteria in the functional drug industry.

BackgroundType A aortic dissection (TAAD) is a cardiovascular emergency condition with high mortality rate. Hybrid total aortic arch replacement using endovascular graft for the descending aorta repair results in favorable outcomes and has been recommended as an alternative procedure for the higher-risk category patients. Our institution started applying the upper ministernotomy incision technique for the hybrid procedures back in 2018.MethodsWe collected patients who underwent hybrid total arch replacement (HTAR) via ministernotomy (96) and total arch replacement with frozen elephant trunk (TAR + FET) procedures (99), between 2018 and 2021. The baseline information, intraoperative and postoperative characteristics have been compared. Kaplan-Meier analysis was used for survival evaluation. Abstract Despite recent progress in subtype classification for small cell lung carcinoma (SCLC), little is known about the biomarker for triple‐negative (ASCL1, NEUROD1, and POU2F3 negative) tumors. The long‐term survival, adjuvant chemotherapy (ACT) response, and immune milieu in different SCLC subtypes have also not been well established. Here, we retrospectively collected a large cohort of 192 primary SCLC tumors and reported that ASCL1‐, NEUROD1‐ and POU2F3‐dominant subtypes counted for 61.38%, 19.31%, and 6.21%, respectively. Subtype intra‐tumoral heterogeneity and co‐expression at the single‐cell level existed substantially. Adverse stress, such as the long-term restriction of food intake and activity in intensive production, leads to a depression-like mental state in sows. Mood disorder, such as depression, is a widely concerned animal welfare issue. However, little is known about the biological mechanisms that underlie mood disorders in pigs. This study is the first attempt to establish a pig depression model by acute stress. A total of 16 adult Bama pigs were divided into the control and model groups, with 8 pigs (half male and half female) per group. The pigs in the model group were restrained for 24 h in a dark and ventilated environment, with food and water deprivation. After the restraint, behavioral tests (feed intake, sucrose preference test, open field test, and novel object test) were used to evaluate apparent indicators. The levels of COR and ACTH in the serum and the levels of 5-HT, NE, and BDNF in the hippocampus and medial prefrontal cortex were detected using ELISA to identify the physiological state. After acute stress, pigs exhibited decreased feed intake and sucrose preference, increased serum COR levels, decreased hippocampal 5-HT levels, and exhibited more fear. Finally, the model was evaluated according to the weight of the test indicators. The overall score of the model was 0.57, indicating that modeling was feasible. Although the reliability and stability require further verification, this novel model revealed typical depression-like changes in behavior and provided a potential method to establish a model of depression in pigs.

Reactive nitrogen gas (Nr) originating from the soil is highly susceptible to the influence of agricultural water and fertilizer management practices, making it a critical gas that can substantially contribute to atmospheric pollution. Understanding the emission patterns of Nr in the surface soil under reclaimed water (RW) irrigation is essential for guiding RW irrigation practices and informing policies aimed at reducing Nr emissions. In this study conducted in Beijing, North China, field sites were selected with different irrigation management strategies and corresponding fertilizer application methods. The two types of irrigation water quality comprised RW irrigation and groundwater (UW) irrigation, whereas the two fertilizer types utilized were calcium ammonium nitrate with a high nitrate nitrogen content and ammonium sulfate with ammonium nitrogen. Nr emissions and concentrations in the soil profile (0–30 cm depth) were monitored throughout key agricultural events, including cultivation, irrigation, fertilization, and harvest, spanning the period from 2020 to 2021. It is observed that RW irrigation effectively reduces cumulative ammonia (NH3) emissions compared to groundwater (UW) treatment. However, it results in increased concentrations of nitrous oxide (N2O) (12.33%−73.82%) and nitric oxide (NO) (13.74%−36.59%) in the soil within the depth range of 0–30 cm. Of particular importance is the notable increase in the abundance of soil denitrifying and ammonia-oxidizing archaea (AOA-amoA) genes due to RW irrigation, indicating the potential for elevated N2O and NO emissions. Furthermore, it is noteworthy that the soil Nr emissions and concentrations are more strongly affected by nitrogen type rather than water quality. Conversely, the inclusion of nitrate has proven to be effective in mitigating N2O and NO emissions in RW-irrigated farmland.

BackgroundThe purpose of this study was to investigate the prognostic significance of serum albumin to creatinine ratio (ACR) in patients receiving heart transplantation of end-stage heart failure.MethodsFrom January 2015 to December 2020, a total of 460 patients who underwent heart transplantation were included in this retrospective analysis. According to the maximum Youden index, the optimal cut-off value was identified. Kaplan-Meier methods were used to describe survival rates, and multivariable analyses were conducted with Cox proportional hazard models. Meanwhile, logistic regression analysis was applied to evaluate predictors for postoperative complications. The accuracy of risk prediction was evaluated by using the concordance index (C-index) and calibration plots.ResultsThe optimal cut-off value was 37.54 for ACR. With the growing shortage of organs, improvements in donor organ protection are needed to meet the increasing demands for transplantation. Here, the aim was to investigate the protective effect of cinnamaldehyde against ischemia–reperfusion injury (IRI) in donor hearts exposed to prolonged cold ischemia. Donor hearts were harvested from rats pretreated with or without cinnamaldehyde, then subjected to 24 h of cold preservation and 1 h of ex vivo perfusion. Hemodynamic changes, myocardial inflammation, oxidative stress, and myocardial apoptosis were evaluated. The PI3K/AKT/mTOR pathway involved in the cardioprotective effects of cinnamaldehyde was explored through RNA sequencing and western blot analysis. Intriguingly, cinnamaldehyde pretreatment remarkably improved cardiac function through increasing coronary flow, left ventricular systolic pressure, +dp/dtmax, and −dp/dtmax, decreasing coronary vascular resistance and left ventricular end-diastolic pressure. Moreover, our findings indicated that cinnamaldehyde pretreatment protected the heart from IRI by alleviating myocardial inflammation, attenuating oxidative stress, and reducing myocardial apoptosis. Further studies showed that the PI3K/AKT/mTOR pathway was activated after cinnamaldehyde treatment during IRI. The protective effects of cinnamaldehyde were abolished by LY294002. In conclusion, cinnamaldehyde pretreatment alleviated IRI in donor hearts suffering from prolonged cold ischemia. Cinnamaldehyde exerted cardioprotective effects through the activation of the PI3K/AKT/mTOR pathway.

Abstract Developing low-cadmium (Cd) rice cultivars has emerged as a promising avenue for food safety in Cd-contaminated farmlands. The root-associated microbiomes of rice have been shown to enhance rice growth and alleviate Cd stress. However, the microbial taxon-specific Cd resistance mechanisms underlying different Cd accumulation characteristics between different rice cultivars remain largely unknown. This study compared low-Cd cultivar XS14 and hybrid rice cultivar YY17 for Cd accumulation with five soil amendments. The results showed that XS14 was characterized by more variable community structures and stable co-occurrence networks in the soil-root continuum compared to YY17. The stronger stochastic processes in assembly of the XS14 (~25%) rhizosphere community than that of YY17 (~12%) suggested XS14 may have higher resistance to changes in soil properties. Microbial co-occurrence networks and machine learning models jointly identified keystone indicator microbiota, such as Desulfobacteria in XS14 and Nitrospiraceae in YY17. Meanwhile, genes involved in sulfur cycling and nitrogen cycling were observed among the root-associated microbiome of these two cultivars, respectively. Microbiomes in the rhizosphere and root of XS14 showed a higher diversity in functioning, with the significant enrichment of functional genes related to amino acid and carbohydrate transport and metabolism, and sulfur cycling. Our findings revealed differences and similarities in the microbial communities associated with two rice cultivars, as well as bacterial biomarkers predictive of Cd-accumulation capacity. Thus, we provide new insights into taxon-specific recruitment strategies of two rice cultivars under Cd stress and highlight the utility of biomarkers in offering clues for enhancing crop resilience to Cd stresses in the future.

This paper presents an AI(Artificial Intelligence)-powered method for enhancing digital creative design through image stylization. To achieve this, we introduce the Content-Style Alignment Module (CSAM), which includes the Dual-Stream Content-Style Processing Block (DS-CSPB), Content-Style Matching Attention Block (CS-MAB), and Content-Style Space-Aware Interpolation Block (CS-SAIB). DS-CSPB removes style information from content descriptors using whitening transformation while preserving semantic structures. CS-MAB reorganizes each content descriptor with its most relevant style descriptor, ensuring optimal style adaptation for content semantics. CS-SAIB aligns content and style descriptors in the same space, enabling diverse semantic distributions in content images to match various style patterns. Moreover, we introduce the Multifaceted Optimization Loss (MOL). This loss comprises multiple components: The relaxed Earth Mover Distance (rEMD) loss enhances color and texture distributions on content images. The Moment Matching (MM) loss reduces visual artifacts caused by cosine distance. The differentiable Color Histogram (CH) loss efficiently addresses color blending issues, preserving image naturalness. The content loss ensures no significant deformation or distortion during stylization. The reconstruction loss constrains all encoder-decoder features to the VGG feature space, maintaining shared spaces between content and style descriptors. We conducted extensive comparative and ablation experiments, which demonstrated superior performance in image stylization, resulting in high-quality stylized images. Additionally, we provide a comprehensive review of current research in image stylization, effectively bridging the gap in this area.

Stereo vision measurement is widely applied in tasks such as autonomous driving and 3D scene reconstruction. Accurately obtaining the disparity of stereo images relies on effective stereo matching algorithms. Compared with the traditional algorithm, the stereo matching algorithm based on convolutional neural networks (CNNs) demonstrates higher accuracy. In this paper, we propose Cs-Net, a coarse-to-fine stereo matching framework that incorporates structural information filtering, aiming to obtain accurate disparity maps. The proposed framework specifically addresses the challenge of accurate disparity estimation, and improves stereo matching in ill-posed regions, such as texture-less and reflective surfaces. To effectively tackle this challenge, the proposed framework incorporates several key modules. First, a contextual attention feature extraction module is introduced, which plays a crucial role in obtaining context information for ill-posed region. Second, a structural attention weight generation module is designed to alleviate the stereo matching errors caused by lack of structural information, and the structure boundary generated by the proposed module is proved to be related to stereo matching errors. Furthermore, a two-stage cost aggregation module is used to regularize the initial cost volume and effectively aggregate the depth information to alleviate matching errors. In the ablation experiments studies, compared to baseline algorithm (GwcNet), Cs-Net can improve D3 and EPE metrics by 14.4% and 0.16 px on the KITTI2015 validation dataset, respectively. Additionally, in the reflective regions of the KITTI2012 benchmark, compared to baseline algorithm, the D3 and D5 metrics of Cs-Net reduced by 15.3% and 20.1%. Additionally, on the DriveStereo dataset, Cs-Net exhibited significant reductions in the D3 and EPE metrics compared to the baseline algorithm, achieving a decrease of 23.5% and 0.09 px, respectively.

Abstract Background Currently, programmed death ligand-1 (PD-L1) expression has been widely applied in clinical trials and real-world clinical practice as a major biomarker for the efficacy of immune-checkpoint inhibitors. The purpose of this study is to reveal the distribution and concordance of PD-L1 expression in a large-scale consecutive cohort from East-Asian patients with non-small cell lung cancer (NSCLC). Methods PD-L1 testing was conducted using 22C3 assays, and cases were categorized into the high, low, and no expression of PD-L1 based on the tumor proportion score (TPS). Abstract Background Postoperative delirium (POD) is a common complication after hip fracture surgery that is associated with various short- and long-term outcomes. The mechanism of POD may be associated with the oxidative stress process. Uric acid has been shown to provide a neuroprotective effect in various neurodegenerative diseases through its antioxidant properties. However, it is unclear whether lower preoperative serum uric acid levels are associated with the development of POD after hip fracture surgery. Therefore, this study assessed the association of lower preoperative uric acid levels in patients with POD during hospitalization. Methods This is a matched retrospective case-control study that included 96 older patients (≥65 y) who underwent hip fracture surgery. POD was diagnosed using the Diagnostic and Statistical Manual of Mental Disorders, Fifth Edition. Patients diagnosed with POD (cases) were matched 1:1 with patients without POD (controls) on the basis of age, sex, and anesthesia type. The relationship between preoperative uric acid and POD was analyzed by multivariable analysis. Results The POD and non-POD groups each had 48 patients. In the univariate analysis, lower log preoperative serum uric acid value, higher neutrophil-to-lymphocyte ratio, and cerebrovascular disease were more likely in patients with POD than in those with no POD. Multivariable conditional logistic regression analysis showed that lower log preoperative serum uric acid (adjusted odds ratio [aOR], 0.028; confidence interval [CI], 0.001–0.844; p = 0.040), higher neutrophil-to-lymphocyte ratio (aOR, 1.314; 95% CI, 1.053–1.638; p = 0.015), and increased surgery duration (aOR, 1.034; 95% CI, 1.004–1.065; p = 0.024) were associated with increased risk of POD. Conclusions Lower preoperative serum uric acid levels may be an independent risk factor for POD after adjustment for possible confounding factors. However, large prospective studies are needed to confirm this finding.

The graphene-modified composite poly(lactic acid) (PLA)/poly(butylene adipate-co-terephthalate) (PBAT) monofilaments were prepared by melt spinning. The effects of graphene (GR) contents on the microstructural, thermal, dynamic mechanical, and mechanical properties and the degradation behavior of the graphene-modified composite PLA/PBAT monofilaments were studied. Scanning electron microscopy (SEM) confirmed the improved compatibility of the PLA particles in the PBAT matrix, while the transmission electron microscopy (TEM) demonstrated the size and morphology of the GR distribution in the polymer. As the GR content increased, dynamic mechanical analysis (DMA) clarified a decrease of the difference of glass transition temperature (Tg) between PLA and PBAT. In addition, the mechanical properties of the composite monofilaments were significantly enhanced at a GR content of 0.2 wt%. Weight loss rate of the composite monofilaments under soil and sea degradation conditions were obviously related to the GR content.

Bacteria of the genus Sulfurovum within the class Campylobacteria are widespread in global oceans and are predominant in sulfide-rich environments. However, little is known about their adaptation to such harsh environments owing to their resistance to cultivation. In this study, we obtained three pure cultures of this genus from marine coastal environments and compared them with those obtained from the deep sea. Phylogenetic analysis of 16S rRNA gene sequences indicated that they represent two novel species of the genus, sharing 95.9%–96.1% sequence similarities to Sulfurovum aggregans Monchim33T. Based on the polyphasic classification results, the type strains XTW-4T and zt1-1T were proposed to represent two new species: Sulfurovum xiamenensis sp. nov. and Sulfurovum zhangzhouensis sp. nov., respectively. These coastal isolates were also obligate chemoautotrophs featuring molecular hydrogen as an electron donor and molecular oxygen, thiosulfate, or elemental sulfur as the sole electron acceptor. Comparative genomic analyses based on 11 Sulfurovum species further revealed a clear differentiation between hydrothermal vent and non-vent origins. The non-vent Sulfurovum can use thiosulfate as an electron acceptor but lacks denitrification pathways, whereas the vent bacteria can respire nitrate through complete denitrification pathways. Moreover, the non-vent Sulfurovum contained a nitrogen fixation pathway, implying their adaptation to nitrogen source-deficit niches. In addition, non-vent Sulfurovum species adapted to a higher oxygen concentration via multiple antioxidative defense mechanisms. These phenotypic and genotypic features help us to understand the ecological role of Sulfurovum bacteria in marine ecosystems.
