Purposes The economic dispatch of Integrated Energy Systems (IES) is a key research topic of energy technology reform, inherently a complex Mixed-Integer Nonlinear Programming problem. Traditional optimization methods have high computational complexity and struggle to adapt to the source-load uncertainty in IES coupled with renewable energy systems. Utilizing Deep Reinforcement Learning to decompose and accelerate the original problem enhances the efficiency of the IES economic dispatch model. Methods To address these issues, in this paper, an IES optimization dispatch framework based on an improved Proximal Policy Optimization (PPO) algorithm is proposed. The PPO algorithm is used to approximate some variables of the nonlinear constraints in the original model, converting them into linear constraints to speed up the solution. Findings The effectiveness and efficiency of this method over others are validated through case studies, predicting significant computational advantages in large-scale IES optimization problems.

IntroductionPathologists rely on whole slide images (WSIs) to diagnose cancer by identifying tumor cells and subtypes. Deep learning models, particularly weakly supervised ones, classify WSIs using image tiles but may overlook false positives and negatives due to the heterogeneous nature of tumors. Both cancerous and healthy cells can proliferate in patterns that extend beyond individual tiles, leading to errors at the tile level that result in inaccurate tumor-level classifications.MethodsTo address this limitation, we introduce NATMIL (Neighborhood Attention Transformer Multiple Instance Learning), which utilizes the Neighborhood Attention Transformer to incorporate contextual dependencies among WSI tiles. NATMIL enhances multiple instance learning by integrating a broader tissue context into the model. Our approach enhances the accuracy of tumor classification by considering the broader tissue context, thus reducing errors associated with isolated tile analysis.ResultsWe conducted a quantitative analysis to evaluate NATMIL’s performance against other weakly supervised algorithms. When applied to subtyping non-small cell lung cancer (NSCLC) and lymph node (LN) tumors, NATMIL demonstrated superior accuracy. Specifically, NATMIL achieved accuracy values of 89.6% on the Camelyon dataset and 88.1% on the TCGA-LUSC dataset, outperforming existing methods. These results underscore NATMIL’s potential as a robust tool for improving the precision of cancer diagnosis using WSIs.DiscussionOur findings demonstrate that NATMIL significantly improves tumor classification accuracy by reducing errors associated with isolated tile analysis. The integration of contextual dependencies enhances the precision of cancer diagnosis using WSIs, highlighting NATMILs´ potential as a robust tool in pathology.

Peptide-functionalized hydrogel is one of commonly used biomaterials to introduce hydrogel-induced vessel regeneration. Despite many reports about the discoveries of high-active peptides (or ligands) for regeneration, the study on the conjugating methods for the hydrogel functionalization with peptides is limited. Here, we compared the vasculogenic efficacy of the peptide-functionalized hydrogels prepared by two commonly used conjugating methods, 1-ethyl-3-(3-dimethylamino propyl) carbodiimide (EDC) and Click methods, through cell models, organ-on-chips models, animal models, and RNA sequencing analysis. Two vascular-related cell types, the human umbilical vein endothelial cells (HUVECs) and the adipose-derived stem cells (ADSCs), have been cultured on the hydrogel surfaces prepared by EDC/Click methods. It showed that the hydrogels prepared by Click method supported the higher vasculogenic activities while the ones made by EDC method compromised the peptide activities on hydrogels. The vasculogenesis assays further revealed that hydrogels prepared by Click method promoted a better vascular network formation. In a critical ischemic hindlimb model, only the peptide-functionalized hydrogels prepared by Click method successfully salvaged the ischemic limb, significantly improved blood perfusion, and enhanced the functional recoveries (through gait analysis and animal behavior studies). RNA sequencing studies revealed that the hydrogels prepared by Click method significantly promoted the PI3K-AKT pathway activation compared to the hydrogels prepared by EDC method. All the results suggested that EDC method compromised the functions of the peptides, while Click method preserved the vascular regenerating capacities of the peptides on the hydrogels, illustrating the importance of the conjugating method during the preparation of the peptide-functionalized hydrogels.

Abstract Background The alteration of the prognostic nutritional index (PNI) or the utilization of distinct anesthesia strategies has been linked to the prognosis of various cancer types, but the existing evidence is limited and inconclusive, particularly for colorectal cancer (CRC). Our objective was to evaluate the association between PNI change and progression free survival (PFS) and overall survival (OS) in patients treated with CRC surgery after propofol-based or sevoflurane-based anesthesia. Methods We conducted a retrospective analysis of 414 patients with CRC who underwent surgical resection. Among them, 165 patients received propofol-based total intravenous anesthesia (TIVA-P), while 249 patients received sevoflurane-based inhalation anesthesia (IA-S). Hepatitis E virus (HEV) is an important cause of acute hepatitis, however, is highly neglected and largely underreported. This study aimed to describe the detailed epidemiology of hepatitis E (HE) through a 10-year surveillance. A community-based active hepatitis surveillance was conducted between November 2007 and October 2017 in 11 townships of Dongtai City in China, involving 355,673 residents. Serum samples were obtained from patients presenting with hepatitis symptoms for more than 3 days. Serum alanine aminotransferase (ALT) levels greater than 2.5 times the upper limit of normal (ULN) were considered acute hepatitis. Samples were subsequently tested for IgG and IgM anti-HEV antibodies, HEV RNA, and hepatitis B surface antigen (HBsAg). The data indicated the incidence of HE fluctuated downward from 2007 to 2017, with an average annual age-standardized incidence of 17.50 per 100,000, exceeding the 10.26 per 100,000 in the National Notifiable Disease Report System (NNDRS). The incidence was notably higher among males (20.95 per 100,000) and individuals aged 50–69 years (37.47 per 100,000). Genotype 4 (HEV-4) was the predominantly circulating genotype during the study period. Furthermore, the study revealed the incidence of hepatitis with HEV and hepatitis B virus (HBV) co-infection was 4.99 per 100,000. The active surveillance system identified a higher incidence of HE compared to NNDRS, with a decreased prevalence over a 10-year period. While efforts are still needed to prevent HE in high-risk populations, including individuals with hepatitis B and the elderly.

Workover operations significantly impact the service life and gas production capacity of coalbed methane (CBM) wells and are crucial for optimizing resource exploitation. To investigate workover operations’ impact on coal seam reservoirs, the authors designed a series of experiments and obtained the following results: (1) The workover operation induced a phase transition in the solid-liquid composition produced by the CBM well, indicating changes in the coal reservoir’s internal structure. (2) During the stable production stage before and after the workover, the proportion of Na+, Cl−, Ca2+, and Total Dissolved Solids (TDS) in the water samples showed a downward trend as a whole, while the HCO3−; after the workover, the Na+, Cl−, Ca2+, and TDS all increased suddenly, while the HCO3− decreased. (3) While inorganic minerals predominated in the precipitation material during the stable production stage pre-workover, their proportion decreased post-workover, with a noticeable shift in their qualitative composition. (4) It is an indisputable fact that workover operations cause physical and chemical damage to coal seam reservoirs. During workover operation, how to avoid damage and conduct benign reconstruction to the reservoir will be the direction of our future efforts. The experimental results provide valuable insights that can guide the optimization of CBM workover operations and inform the strategic planning of subsequent drainage activities.

In order to improve fuel economy to meet the standard for passenger car oil, a new formulation with good viscosity–temperature performance for gasoline engine oil is required. In this study, coal-to-liquid (CTL) base oil, with a high viscosity index and good low-temperature performance, was selected as the base oil to develop the gasoline engine oil. A systematic study on the molecular interaction between the CTL base oil and the viscosity index improver (VII), including three kinds of hydrogenated styrene diene copolymers (HSD-type) and four kinds of ethylene propylene copolymers (OCP-type), was conducted. It was found that in general, in CTL base oil, the HSD-type VII exhibited a much higher viscosity index, a significantly lower shear stability index, a higher thickening ability, and a lower cold-cranking simulator (CCS) viscosity than that of OCP-type VII. Moreover, when comparing CTL base oil with mineral oil 150N, the combination of CTL base oil and the VII displayed a lower CCS viscosity than that of mineral oil, suggesting it had better low-temperature performance and was able to quickly form a protective oil film on the surface, which was beneficial for the cold start. The functional group distribution state of the VII in base oil was analyzed using synchrotron radiation micro-infrared microscope (SR Micro-IR) technology, which revealed that HSD-1 had a better molecular interaction with CTL6 than 150N because of the better uniformity of the C=C group distribution. Based on this, a SP 0W-20 gasoline engine oil was developed by the combination of CTL base oil and the HSD-1 viscosity index improver, together with an additive package, a polymethacrylate pour point depressant, and a non-silicone defoamer, which showed excellent low-temperature performance, thermal oxidation stability, and detergency performance compared to the reference oil.

Abstract Background There are few widely accepted and operationally feasible models for predicting the mortality risk of patients in surgical intensive care unit (SICU). Although serum anion gap (AG) is known to be correlated with severe metabolic acidosis, no investigations have been reported about the association between AG level and the outcome during hospitalization in SICU. This study aimed to explore the predictive power of AG for 90-day all-cause mortality in SICU. Methods Data of the eligible patients in SICU from 2008 to 2019 was obtained from the Medical Information Mart for Intensive Care IV version 2.0 (MIMIC-IV v2.0) database. Baseline clinical data of the selected patients was compared in different groups stratified by the outcome during their admission via univariate analysis. Restricted cubic spline (RCS) was drawn to confirm the relationship of AG and the short-term mortality. Kaplan-Meier survival curve was plotted in different AG level groups. Univariate and multivariate Cox analyses were performed, and Cox proportional-hazards models were built to investigate an independent role of AG to predict 90-day all-cause mortality risk in SICU. Receiver operating characteristics (ROC) curves analysis was performed to evaluate the predictive value of AG on the 90-day prognosis of patients. Results A total of 6,395 patients were enrolled in this study and the 90-day all-cause mortality rate was 18.17%. Background: N-glycosylation is one of the most common posttranslational modifications in humans, and these alterations are associated with kidney diseases. Methods: A novel technological approach, single-cell N-acetyllactosamine sequencing (scLacNAc-seq), was applied to simultaneously detect N-glycosylation expression and the transcriptome at single-cell resolution in three human kidney tissues from zero-time biopsy. Cell clusters, glycation abundance in each cell cluster, functional enrichment analysis, cell-cell crosstalk, and pseudotime analysis were applied. Results: Using scLacNAc-seq, 24,247 cells and 22 cell clusters were identified, and N-glycan abundance in each cell was obtained. Transcriptome analysis revealed a close connection between capillary endothelial cells (CapECs) and parietal epithelial cells (PECs). PECs and CapECs communicate with each other through several pairs of ligand receptors (e.g., TGFB1-EGFR, GRN-EGFR, TIMP1-FGFR2, VEGFB-FLT1, ANGPT2-TEK, and GRN-TNFRSF1A). Finally, a regulatory network of cell-cell crosstalk between PECs and CapECs was constructed, which is involved in cell development. Conclusions: We here, for the first time, constructed the glycosylation profile of 22 cell clusters in the human kidney from zero-time biopsy. Moreover, cell-cell communication between PECs and CapECs through the ligand-receptor system may play a crucial regulatory role in cell proliferation.

Wheat (Triticum aestivum L.) is one of the most important food crops worldwide and provides the staple food for 40% of the world’s population. Increasing wheat production has become an important goal to ensure global food security. The grain yield of wheat is a complex trait that is usually influenced by multiple agronomically important traits. Thus, the genetic dissection and discovery of quantitative trait loci (QTL) of wheat-yield-related traits are very important to develop high-yield cultivars to improve wheat production. To analyze the genetic basis and discover genes controlling important agronomic traits in wheat, a recombinant inbred lines (RILs) population consisting of 180 RILs derived from a cross between Xinong822 (XN822) and Yannong999 (YN999), two well-adapted cultivars, was used to map QTL for plant height (PH), spike number per spike (SNS), spike length (SL), grain number per spike (GNS), spike number per plant (SN), 1000- grain weight (TGW), grain length (GL), grain width (GW), length/width of grain (GL/GW), perimeter of grain (Peri), and surface area of grains (Sur) in three environments. A total of 64 QTL were detected and distributed on all wheat chromosomes except 3A and 5A. The identified QTL individually explained 2.24–38.24% of the phenotypic variation, with LOD scores ranging from 2.5 to 29. Nine of these QTL were detected in multiple environments, and seven QTL were associated with more than one trait. Additionally, Kompetitive Allele Specific PCR (KASP) assays for five major QTL QSns-1A.2 (PVE = 6.82), QPh-2D.1 (PVE = 37.81), QSl-2D (PVE = 38.24), QTgw-4B (PVE = 8.78), and QGns-4D (PVE = 13.54) were developed and validated in the population. The identified QTL and linked markers are highly valuable in improving wheat yield through marker-assisted breeding, and the large-effect QTL can be fine-mapped for further QTL cloning of yield-related traits in wheat.

The end clearance and radial clearance of the gear pump are the most important factors affecting its internal leakage and viscous friction loss, and also the most important factors affecting the total efficiency of a high-speed gear pump. The reasonable value of radial clearance and end clearance is the decisive condition for the high-efficiency performance of high-speed gear pumps. Based on a certain type of high-speed gear pump with a speed of 6000 r/min, the influence of end clearance and radial clearance on internal leakage and viscous friction loss of the gear pump is analyzed under the rated working condition of outlet pressure of 8 MPa. The flow field simulation model of the high-speed gear pump with oil flowing in radial clearance and end clearance is established in PumpLinx. The oil leakage flow rate and pump shaft power under different radial and end clearances are analyzed. The results show that under the condition of 6000 r/min, the phenomenon of insufficient oil filling in the tooth groove is produced, and the radial clearance is 0.16 mm when the total efficiency of the high-speed gear pump is the largest. Under the optimal radial clearance, the working characteristics of gear pumps with different end clearances are analyzed. The end clearance is 0.03 mm when the total efficiency of high-speed gear pumps is the largest. In this paper, the selection of radial and end clearance of high-speed gear pump and the internal flow of different clearances are analyzed, which has important theoretical significance and engineering practical value.

Urban green development is a way of economic growth and social development aiming at efficiency, harmony and sustainability, and in recent years urban green development has become an important trend for future urban development. In this study, Hefei City is selected as the study area, and a comprehensive green-development model is constructed by applying the system dynamics method, which integrates five important subsystems, namely, economy, environment, people’s livelihood, S and T and resources. Through simulation analysis, this study reveals the dynamic trend of green development in Hefei City. The analysis results show that economic growth provides the foundation for green development, scientific and technological innovation promotes the development of green technology, and that sustained investments in people’s livelihoods and environmental protection improves people’s quality of life and a city’s eco-friendliness. This study aims to promote the coordinated economic, social and environmental development of Hefei to ensure that Hefei is steadily moving towards high-quality and sustainable development goals; it effectively reveals the current situation, forecasts the trend of green development in Hefei, and also provides a reference for the urban green development of other cities.

Shale is a common rock type that is associated with underground engineering projects, and several important factors, such as bedding structure, confining pressure, and the loading and unloading path, significantly influence the anisotropy of shale. Triaxial monotonic loading tests and triaxial incremental cyclic loading and unloading tests of shale under three kinds of confining pressures and five types of bedding inclination angles (θ) were thus performed to investigate the anisotropy of shale in terms of mechanical behavior, acoustic emission (AE), and energy evolution, and reveal the mechanism by which shale anisotropy is weakened. The results show that (1) the compressive strength and elastic modulus of shale decrease and then increase as the θ increases, and that both σ3 and incremental cyclic loading and unloading reduce the anisotropy in terms of the compressive strength and elastic modulus of shale, with the ratio of plastic strain to total strain reaching its maximum at a θ of 60° during each loading and unloading cycle. (2) The failure modes of shale with θ of 0°, 30°, and 90° under triaxial monotonic loading are similar to the counterparts under triaxial incremental cyclic loading and unloading, while the failure modes of shale with θ of 45° and 60° differ significantly under the two loading conditions, and interestingly, the degree to which the bedding plane participates in shale crack evolution under incremental cyclic loading and unloading is considerably lower than that under triaxial monotonic loading. (3) The cumulative AE count and AE b-value of shale first decrease and then increase as the θ increases, while the Felicity ratio decreases as the number of cycles increases. (4) As the θ increases, the total energy density U0 and the parameter m, which reflects the accumulation rate of elastic energy, first decrease and then increase, with both reaching a minimum at a θ of 60°. (5) The mode by which cyclic loading and unloading leads to failure in shale with a θ of 60° is similar to that at a θ of 0° and is the main mechanism by which shale anisotropy weakening occurs as a result of cyclic loading and unloading. The results provide experimental support and a theoretical basis for safer and more efficient underground engineering projects that involve shale.

Single-cell analysis provides an overwhelming strategy for revealing cellular heterogeneity and new perspectives for understanding the biological function and disease mechanism. Moreover, it promotes the basic and clinical research in many fields at a single-cell resolution. A digital polymerase chain reaction (dPCR) is an absolute quantitative analysis technology with high sensitivity and precision for DNA/RNA or protein. With the development of microfluidic technology, digital PCR has been used to achieve absolute quantification of single-cell gene expression and single-cell proteins. For single-cell specific-gene or -protein detection, digital PCR has shown great advantages. So, this review will introduce the significance and process of single-cell analysis, including single-cell isolation, single-cell lysis, and single-cell detection methods, mainly focusing on the microfluidic single-cell digital PCR technology and its biological application at a single-cell level. The challenges and opportunities for the development of single-cell digital PCR are also discussed.

IntroductionMany patients who undergo assisted reproductive technology (ART) suffer from recurrent implantation failure (RIF). The addition of hyaluronic acid (HA) to the transfer medium is one of several methods to improve pregnancy outcomes. We investigated whether HA could improve the live birth and clinical pregnancy rates of RIF patients.MethodsThis study included 248 RIF patients, who were divided into two groups: the control (CTL) group (n=137), which received transfer medium without HA, and the HA group (n=111), which received transfer medium with HA. The two groups were compared according to the ART outcome.ResultsThe primary outcomes were the clinical pregnancy and live birth rates. Secondary outcomes include a positive urine pregnancy test, the implantation, ongoing pregnancy, multiple pregnancy, clinical miscarriage, and ectopic pregnancy rates, foetal or congenital defects, obstetric complications, infant birth weight and any related adverse events. Regarding the primary outcomes, the clinical pregnancy rate was significantly higher in the HA group than in the control group, and there was no significant difference in the live birth rate (LBR) between the HA and control groups. Regarding the secondary outcomes, the implantation, multiple pregnancy and ectopic pregnancy rates were similar between the two groups.DiscussionOur findings supported the conclusion that HA can improve the clinical pregnancy rate of patients with RIF undergoing FET cycles, but the live birth rate was not significantly improved with the addition of HA to the traditional transfer medium.

Abstract Recurrent implantation failure (RIF) is a rather thorny problem in the clinical practice of assisted reproductive technology. Due to the complex aetiology of RIF, its pathogenesis is far from fully understood, and there is no effective treatment available. Here, We explored the regulatory mechanism of the four half-domains of LIM domain 1 (FHL1), which is significantly downregulated in the endometrium of RIF patients, in blastocyst-epithelial adhesion. Indeed, FHL1 expression was dramatically increased in normal female mid-secretory endometrial epithelial cells and was abnormally reduced in RIF patients. Furthermore, FHL1 overexpression promoted blastocyst-epithelial adhesion, and interfering with FHL1 expression in the mouse uterus significantly inhibited embryo implantation. Mechanistically, FHL1 did not regulate HOXA10 mRNA expression but increased HOXA10 protein stability and activated HOXA10, thereby promoting its regulation of downstream gene expression and the β3 integrin/FAK pathway. Meanwhile, FHL1 regulates HOXA10 function by increasing HOXA10 deacetylation through enhanced binding of HOXA10 and SIRT2. SIRT2-specific inhibitors can significantly inhibit this effect. In the endometrial epithelial cells of RIF patients, the correlation between FHL1 and HOXA10 and its downstream target genes has also been verified. Finally, our data indicated FHL1 is a regulatory molecule that promotes blastocyst-epithelial adhesion. Altogether, downstream dysfunction due to aberrant FHL1 expression is an important molecular basis for embryo implantation failure in patients with RIF and to provide new potential therapeutic targets.

Abstract Plant immunity is activated upon perception of pathogens and often affects growth when it is constitutively active. It is still a challenge to balance plant immunity and growth in disease resistance breeding. Here, we demonstrated that soybean (Glycine max) polyamine oxidase (GmPAO) confers resistance to multiple Phytophthora pathogens, but has no obvious adverse impact on agronomic traits. GmPAO produces H2O2 by oxidizing spermidine and spermine. Phytophthora sojae induces an increase in these two substrates, and thus promotes GmPAO-mediated polyamine catabolism specifically during infection. Interestingly, we found that the two substrates showed higher accumulation in transgenic soybean lines overexpressing GmPAO than in WT and CK after inoculation with P. sojae to ensure H2O2 production during infection, rather than directly inhibit P. sojae. In these transgenic soybean plants, the significantly enhanced resistance to different P. sojae isolates was achieved; PAMP-induced H2O2 accumulation was enhanced by GmPAO overexpression. Moreover, transient expression of GmPAO also significantly improved Nicotiana benthamiana resistance to Phytophthora capsici and Phytophthora parasitica in agroinfiltration assays. Our results provide a novel approach to allow rapid defense responses in plants upon pathogen infection while minimizing growth penalties under normal conditions, with a clear mechanism in which plant promotes H2O2 production via pathogen-activated substrates.

BackgroundPostoperative recurrence of hepatocellular carcinoma (HCC) is associated with low survival rates. While HCC treatment options have expanded substantially, they are accompanied by several challenges. This study assessed the outcomes of repeated hepatectomy (RH) for postoperative intrahepatic recurrence of HCC among patients undergoing initial hepatectomy (IH) as well as independent risk factors for HCC recurrence among patients undergoing repeated hepatectomy (RH).MethodsClinical data from 84 patients undergoing both IH and RH and 66 recurrent HCC patients who had received radiofrequency ablation (RFA) from July 2011 to September 2017 were retrospectively reviewed. The following groups were compared: (1) RH Group A (n = 84), (2) IH Group (n = 84, same individuals as RH Group A), (3) RH Group B (n = 45/84 from RH Group A), and (4) RFA Group (n = 66). The clinical pathology and operative characteristics of the patients in RH Group A were compared to those in the IH Group. Meanwhile, the clinical pathology and pre- and post-treatment features of the patients in RH Group B were compared to those in the RFA Group. The tumor-free survival time was compared between patients in RH Group A and the IH Group as well as between patients in RH Group B and the RFA Group. The independent risk factors for the 1-year postoperative tumor-free survival of RH Group A patients were investigated using univariate and multivariate analysis.ResultsMeasures of clinical pathology, including AFP, Child-Pugh score, HBV-DNA, tumor number, liver cirrhosis, tumor differentiation, surgical approach, and TNM stage differed significantly between patients in RH Group A and the IH Group (all P < 0.05), with the exception of tumor number and tumor size (both P > 0.05). No significant differences were found in these measures between the patients in RH Group B and the RFA Group (all P > 0.05). While patients in the RH Group A had a longer operation time than those in the IH Group (4.35 ± 1.25 h vs. 3.55 ± 0.92 h, P < 0.001), the level of intraoperative bleeding was similar (400.00 ± 199.25 ml vs. 359.40 ± 213.37 ml, P = 0.204). RH Group B patients had a longer hospitalization time than those in the RFA Group (6.5 ± 0.8 d vs. 5.5 ± 1.1 d, P < 0.001), however, the difference in hospitalization costs was not statistically significant (29,009 ± 3,806 CNY vs. 29,944 ± 3,752 CNY, P = 0.202). Five-day post-operative serum biomarker levels, including direct bilirubin (DB) and albumin (ALB), were significantly higher in RH Group B than in the RFA Group (all P < 0.05), with the exception of ALT, AST, and total bilirubin (TB) (all P > 0.05). Patients in RH Group A had a lower tumor-free survival time than those in the IH Group (median: 12 vs. 22 months, P < 0.001), and patients in the RH Group B had a significantly higher tumor-free survival time than those in the RFA group (median: 15 months vs. 8 months, P < 0.001). Age ≥50 y, Child-Pugh class A, and negative HBV-DNA were independent risk factors that positively impacted the 1-year postoperative tumor-free survival rate of postoperative intrahepatic recurrent HCC patients undergoing RH (P < 0.001, respectively).ConclusionDue to the potential of harm related to relapse of recurrent HCC for cancer patients, RH is a superior option. RH could offer better outcomes for recurrent HCC patients undergoing IH. Compared with lesion pathology, the better target organ of the liver will be key to ameliorating tumor-free survival for recurrent HCC patients undergoing RH.

Long noncoding RNA (lncRNA) CBR3-AS1 (termed as CBR3-AS1) has been reported to be upregulated in several cancers including osteosarcoma. Its positive impact on the proliferation, migration, and invasion of osteosarcoma cells has been unveiled; nevertheless, whether it also affects the stemness and epithelial-mesenchymal transition (EMT) of osteosarcoma cells is unclear. The purpose for this study was to explore the effects of CBR3-AS1 on the stemness and EMT of osteosarcoma cells as well as its underlying mechanism. qRT-PCR and western blot were applied to detect target gene expression. Function assays were conducted to evaluate the effect of genes on the stemness and EMT of osteosarcoma cells. Mechanism assays were done to verify the association among different genes. In vivo assays were also performed. The obtained data showed that CBR3-AS1 demonstrated a high expression in osteosarcoma cells. CBR3-AS1 could promote stemness and EMT of osteosarcoma cells as well as osteosarcoma tumor growth. Mechanically, CBR3-AS1 sponged miR-140-5p and recruited DDX54 to upregulate NUCKS1, thus activating the mTOR signaling pathway. Furthermore, NUCKS1 could facilitate stemness and EMT of osteosarcoma cells. In summary, this study reveals that CBR3-AS1 exerts an oncogenic role in osteosarcoma through modulating the network of the miR-140-5p/DDX54-NUCKS1-mTOR signaling pathway.

Here, the authors developed an alternative energy-efficient strategy to use of an acyl-ACP transacylase to produce medium chain oleochemicals in E. coli.

Mung bean (Vigna radiata) production has been greatly threatened by numerous diseases. Infection with these pathogens causes extensive changes in gene expression and the activation of hormone signal transduction. Quantitative real-time PCR (qRT-PCR) is the most common technique used for gene expression validation. Screening proper reference genes for mung bean under pathogen infection and hormone treatment is a prerequisite for ensuring the accuracy of qRT-PCR data in mung bean disease-resistance research. In this study, six candidate reference genes (Cons4, ACT, TUA, TUB, GAPDH, and EF1α) were selected to evaluate the expression stability under four soil-borne disease pathogens (Pythium myriotylum, Pythium aphanidermatum, Fusarium oxysporum, and Rhizoctonia solani) and five hormone treatments (SA, MeJA, ETH, ABA, and GA3). In the samples from different treatments, the Ct value distribution of the six candidate reference genes was different. Under the condition of hormone treatment, the Ct value ranged from a minimum of 17.87 for EF1α to a maximum of 29.63 for GAPDH. Under the condition of pathogen infection, the Ct value ranged from a minimum of 19.43 for EF1α to a maximum of 31.82 for GAPDH. After primer specificity analysis, it was found that GAPDH was not specific, so the five reference genes Cons4, ACT, TUA, TUB, and EF1α were used in subsequent experiments. The software products GeNorm, NormFinder, BestKeeper and RefFinder were used for qRT-PCR data analysis. In general, the best candidates reference genes were: TUA for SA, ABA, GA3, and Pythium myriotylum treatment; TUB for ETH treatment; ACT for MeJA and Fusarium oxysporum treatment; and EF1α for Pythium aphanidermatum and Rhizoctonia solani treatment. The most stably expressed genes in all samples were TUA, while Cons4 was the least stable reference gene. Finally, the reliability of the reference gene was further validated by analysis of the expression profiles of four mung bean genes (Vradi0146s00260, Vradi0158s00480, Vradi07g23860, and Vradi11g03350) selected from transcriptome data. Our results provide more accurate information for the normalization of qRT-PCR data in mung bean response to pathogen interaction.

Due to the extreme water sensitivity of shale, the excavation of shale underground engineering is prone to major disaster accidents such as roof falls and collapses. However, current investigations have failed to fully explain the mechanisms by which water content affects shale damage behaviors. In this study, the acousto-mechanical properties and failure behaviors of laminated shale under different confining pressures σ3 are investigated with the aid of AE monitoring for three different water content states. The results show that the shale strength decreases with the increase of the water content, but it increases as the confining pressure σ3 increases. For the shale, the change in the wetting angle and the distance between the centroids of the two adjacent particles inside the bedding plane is more prominent than the surrounding shale matrix, and the swelling pressure is generated among the clay minerals, which are the two main mechanisms for the bedding-participating failure and the shale softening after immersion. Moreover, with the increase of the water content and σ3, the damage mode of shale specimens gradually changes from tension damage to shear damage. Controlled by bedding, shale failure shows significant suddenness without clear acoustical precursors. This study provides experimental and theoretical bases for the stability analysis of shale underground engineering.

Abstract Counterfeit leather products infringe the intellectual property rights of the business, cause enormous economic loss, and negatively influence the business enthusiasm for innovation. However, traditional anti-counterfeiting materials for leather products suffer from complicated fabrication procedures, photobleaching, and high volatile organic compound (VOC) emissions. Here, a sustainable and invisible anti-counterfeiting ink composed of waterborne polyurethane and water-dispersible lanthanide-doped upconversion nanoparticles (UCNPs) featuring ease of preparation, high photostability, non-toxicity, low VOC emissions, and strong adhesion strength for leather products is designed and synthesized. After decorating on the surface of leather products, the obtained patterns are invisible under normal light conditions. Upon irradiation at 808 nm, the invisible patterns can be observed by naked eyes due to the visible light emitted by 808 nm excited UCNPs. Our approach described here opens a new pathway to realize the long-term, stable anti-counterfeiting function of leather products. Graphical Abstract

The mutant efficiency and hatching ratio are two key factors that significantly affect the construction of genome-modified mutant insects. In the construction of CRISPR/Cas9-mediated dsLmRNase2–/– mutant locusts, we found that the tanned eggs which experienced a 20-min contact with the oocyst exhibited a higher success rate compared to fresh newly-laid eggs that were less tanned. However, the heritable efficiency of the dsLmRNase2 deletion to the next generation G1 progeny was similar between adults derived from the tanned or less tanned engineered eggs. Further, the similar effective mutant ratios in the normally developed eggs and G0 adults of tanned and less tanned eggs also indicated that tanning did not reduce the absolute mutation efficiency induced by CRISPR/Cas9. Moreover, we found that the syncytial division period, which was longer than the time for tanning, conferred a window period for microinjection treatment with efficient mutation in both tanned and less tanned eggs. We further found that tanned eggs exhibited a higher hatching rate due to a reduced infection rate following microinjection. Both the anti-pressure and ultrastructure analyses indicated that the tanned eggs contained compressed eggshells to withstand increased external pressure. In summary, tanned eggs possess stronger defense responses and higher efficiency of genome editing, providing an improved model for developing Cas9-mediated gene editing procedures in locusts.

We demonstrate subwavelength resolution, full-field, and lensless terahertz ptychography via longitudinal shifts of the sample. An extended illumination is adopted to realize full-field recording. Compared to a typical scheme, this implementation requires fewer diffraction patterns, which improves the imaging throughput. To suppress the incoherent infrared background, an optical chopper is introduced and a preprocessing algorithm based on digital phase-locked is proposed. By combining the ideas of multi-plane phase retrieval and ptychographic reconstruction, a reconstruction algorithm is proposed to simultaneously retrieve the object and the probe. Due to the lensless single-beam mode and full-field illumination, the sample is placed as close to the detector as possible, and the aperture of the whole sensor is utilized to achieve the highest numerical aperture. Note that the proposed approach could be applied to other wavebands as well. We apply this method to a biological material, a polymer sample, and a resolution target. The uneven illumination, as well as the diffraction effects associated with detector edges, is separated from the object reconstruction. The secondary veins of the dragonfly forewing, as small as ∼20 µm, are observed, and the lateral resolution achieves 88 µm (0.74λ).

Although mungbean (Vigna radiata (L.) R. Wilczek) is an important legume crop, its seed yield is relatively low. To address this issue, here 196 accessions with 3,607,508 SNP markers were used to identify quantitative trait nucleotides (QTNs), QTN-by-environment interactions (QEIs), and their candidate genes for seed length (SL), seed width, and 100-seed weight (HSW) in two environments. As a result, 98 QTNs and 20 QEIs were identified using 3VmrMLM, while 95, >10,000, and 15 QTNs were identified using EMMAX, GEMMA, and CMLM, respectively. Among 809 genes around these QTNs, 12 were homologous to known seed-development genes in rice and Arabidopsis thaliana, in which 10, 2, 1, and 0 genes were found, respectively, by the above four methods to be associated with the three traits, such as VrEmp24/25 for SL and VrKIX8 for HSW. Eight of the 12 genes were significantly differentially expressed between two large-seed and two small-seed accessions, and VrKIX8, VrPAT14, VrEmp24/25, VrIAR1, VrBEE3, VrSUC4, and Vrflo2 were further verified by RT-qPCR. Among 65 genes around these QEIs, VrFATB, VrGSO1, VrLACS2, and VrPAT14 were homologous to known seed-development genes in A. thaliana, although new experiments are necessary to explore these novel GEI-trait associations. In addition, 54 genes were identified in comparative genomics analysis to be associated with seed development pathway, in which VrKIX8, VrABA2, VrABI5, VrSHB1, and VrIKU2 were also identified in genome-wide association studies. This result provided a reliable approach for identifying seed-size-related genes in mungbean and a solid foundation for further molecular biology research on seed-size-related genes.

The ECOWAS region has one of Africa's highest potentials for energy production, including both non-renewable (oil, gas, and uranium) and renewable sources (hydroelectric, solar energy, wind energy). Despite this significant potential, the region deals with a number of issues that affect its energy strategy. A review and analysis of the social, political, and economic factors influencing regional energy policy are provided in this paper, along with an assessment and forecast of energy policy in the ECOWAS. The analysis of regional energy policy then takes into account demand management, clean energy production, regional energy trade, and hydrocarbon exploration and production. The results show that the ECOWAS nations have started their transition to a renewable energy-based economy. These policies have long-term implications on the world's energy system and have the potential to improve the region's energy policy, even if the consequences are not immediately noticeable.

Adenomyosis is a common benign uterine lesion that is associated with female infertility, reduced clinical pregnancy rate and high miscarriage risk. While it has been known that the impaired endometrial receptivity is implicated in infertility in patients with adenomyosis, the underlying mechanism remains unclear. In the present study, we showed that intracellular protein level of IL-33 was downregulated in the endometrium of patients with adenomyosis, and IL-33 expression status was shown to be positively correlated with that of HOXA10, an endometrial receptivity marker. The subsequent analysis indicated IL-33 overexpression led to the increase of HOXA10 expression and enhancement of embryo implantation in vitro, which was accompanied with induction of STAT3 phosphorylation. Meanwhile, cryptotanshinone, a potent STAT3 inhibitor, was found to significantly suppress the increase of HOXA10 expression and embryo implantation caused by IL-33 overexpression in vitro, revealing the critical role of STAT3 activity. Consistently, the positive relationship between IL33 and HOXA10 expression in the endometrium was verified in the analysis of adenomyosis mouse model.

Faba bean is an important legume crop consumed as a vegetable or snack food, and its green cotyledons could present an attractive color for consumers. A mutation in SGR causes stay-green in plants. In this study, vfsgr was identified from a green-cotyledon-mutant faba bean, SNB7, by homologous blast between the SGR of pea and the transcriptome of faba bean. Sequence analysis revealed that a SNP at position 513 of the CDS of VfSGR caused a pre-stop codon, resulting in a shorter protein in the green-cotyledon faba bean SNB7. A dCaps marker was developed according to the SNP that caused the pre-stop, and this marker was completely associated with the color of the cotyledon of faba bean. SNB7 stayed green during dark treatment, while the expression level of VfSGR increased during dark-induced senescence in the yellow-cotyledon faba bean HST. Transient expression of VfSGR in Nicotiana. benthamiana leaves resulted in chlorophyll degradation. These results indicate that vfsgr is the gene responsible for the stay-green of faba bean, and the dCaps marker developed in this study provides a molecular tool for the breeding of green-cotyledon faba beans.

Abstract Background Trisomy 18 syndrome (Edwards syndrome, ES) is a type of aneuploidy caused by the presence of an extra chromosome 18. Aneuploidy is the leading cause of early pregnancy loss, intellectual disability, and multiple congenital anomalies. The research of trisomy 18 is progressing slowly, and the molecular characteristics of the disease mechanism and phenotype are still largely unclear. Results In this study, we used the commercial Chromium platform (10× Genomics) to perform sc-ATAC-seq to measure chromatin accessibility in 11,611 single umbilical cord blood cells derived from one trisomy 18 syndrome patient and one healthy donor. We obtained 13 distinct major clusters of cells and identified them as 6 human umbilical cord blood mononuclear cell types using analysis tool. Compared with the NC group, the ES group had a lower ratio of T cells to NK cells, the ratio of monocytes/DC cell population did not change significantly, and the ratio of B cell nuclear progenitor and megakaryocyte erythroid cells was higher. The differential genes of ME-0 are enriched in Human T cell leukemia virus 1 infection pathway, and the differential peak genes of ME-1 are enriched in apopotosis pathway. We found that CCNB2 and MCM3 may be vital to the development of trisomy 18. CCNB2 and MCM3, which have been reported to be essential components of the cell cycle and chromatin. Conclusions We have identified 6 cell populations in cord blood. Disorder in megakaryocyte erythroid cells implicates trisomy 18 in perturbing fetal hematopoiesis. We identified a pathway in which the master differential regulatory pathway in the ME-0 cell population involves human T cell leukemia virus 1 infection, a pathway that is dysregulated in patients with trisomy 18 and which may increase the risk of leukemia in patients with trisomy 18. CCNB2 and MCM3 in progenitor may be vital to the development of trisomy 18. CCNB2 and MCM3, which have been reported to be essential components of the cell cycle and chromatin, may be related to chromosomal abnormalities in trisomy 18.

Previous studies have shown that shale oil mobility depends on the relative content of free oil and adsorbed oil. However, the research on how to establish a shale oil mobility evaluation is relatively insufficient. This study aims to use pyrolysis data before and after extraction to accurately identify the content of free oil and adsorbed oil, analyze the influencing factors of shale oil mobility, characterize the hydrocarbon generation and expulsion process, and evaluate shale oil mobility. We utilized an integrated mineralogical and geochemical dataset from the PS18-1 well in the Liutun Sag, Dongpu Depression, Bohai Bay Basin. The results show that the adsorption capacity of type I organic matter (OM) on shale oil is greater than that of type II OM, the OM abundance is of great significance to shale oil mobility, and that quartz and feldspar can promote shale oil mobility. The Tmax corresponding to the threshold of hydrocarbon expulsion is 438~440 °C, and the oil saturation index (OSI) is about 158 mg/g TOC. There are four small intervals: a (3257 m~3260 m), b (3262 m~3267 m), c (3273 m~3278 m), and d (3281 m~3282 m) meeting the conditions of hydrocarbon expulsion. Large-scale hydrocarbon expulsion occurred in interval a, a small amount of hydrocarbon expulsion in interval b, a large amount of hydrocarbon expulsion in interval c, and almost no hydrocarbon expulsion in interval d. Based on the crossplot of S1 and TOC, combined with other parameters such as OSI, hydrocarbon generation potential (HGP), and free and adsorbed oil, we established an evaluation chart of shale oil mobility and divided it into five categories: A, B, C, D, and E. While categories A and C have good mobility and great resource potential, categories B and D have relatively poor mobility and medium resource potential, and category E has little mobility and is an invalid resource.

In Africa, switching to renewable energy sources with low carbon emissions is becoming more popular. In the Economic Community of West African states (ECOWAS), the low-carbon energy transition must overcome significant obstacles, including those posed by policies put in place and their implementation. In order to replace the current fossil fuel-driven economy with low-carbon development that also advances the regional aims and Sustainable Development Goals (SDGs), this paper analyzes the current policy landscape in the region. A total of 75 policies from four categories (energy access, energy efficiency, renewable energy, climate change) were chosen for the policy review. Multicriteria decision analysis and a thorough review of the literature have shed light on how well the policies of the ECOWAS countries could promote low-carbon development and what the main challenges to overcome are. The last step was the comparison of the progress of the two key policies in the region: the National Energy Efficiency Action Plan (NEEAP) and National Renewable Energy Action Plan (NREAP). The results illustrate the amount of work still needing to be done in the region, with only 26 of the 75 policies receiving a score above 50. However, the prospect of a better energy strategy that adequately considers the difficulties of the energy transition is possible, given the progress made by the region’s nations since the creation of national action plans for energy efficiency (EE) and renewable energy (RE). Our analysis has also shown that countries like Senegal have obtained quite promising results, as evidenced by the best scores of 76.88 and 73.25, respectively, obtained by its NREAP and NEEAP policies.

WRKY-, PHD-, and MYB-like proteins are three important types of transcription factors in mungbeans, and play an important role in development and stress resistance. The genes’ structures and characteristics were clearly reported and were shown to contain the conservative WRKYGQK heptapeptide sequence, Cys4-His-cys3 zinc binding motif, and HTH (helix) tryptophan cluster W structure, respectively. Knowledge on the response of these genes to salt stress is largely unknown. To address this issue, 83 VrWRKYs, 47 VrPHDs, and 149 VrMYBs were identified by using comparative genomics, transcriptomics, and molecular biology methods in mungbeans. An intraspecific synteny analysis revealed that the three gene families had strong co-linearity and an interspecies synteny analysis showed that mungbean and Arabidopsis were relatively close in genetic relationship. Moreover, 20, 10, and 20 genes showed significantly different expression levels after 15 days of salt treatment (p < 0.05; Log2 FC > 0.5), respectively. Additionally, in the qRT-PCR analysis, VrPHD14 had varying degrees of response to NaCl and PEG treatments after 12 h. VrWRKY49 was upregulated by ABA treatment, especially in the beginning (within 24 h). VrMYB96 was significantly upregulated in the early stages of ABA, NaCl, and PEG stress treatments (during the first 4 h). VrWRKY38 was significantly upregulated by ABA and NaCl treatments, but downregulated by PEG treatment. We also constructed a gene network centered on the seven DEGs under NaCl treatment; the results showed that VrWRKY38 was in the center of the PPI network and most of the homologous Arabidopsis genes of the interacted genes were reported to have response to biological stress. Candidate genes identified in this study provide abundant gene resources for the study of salt tolerance in mungbeans.

The medium and long-term demand prediction for water resources is critical to the country’s macro-allocation of water resources and the formulation of a long-term regional economic development strategy. However, the official prediction report of China’s water resources was created ten years ago, and its results are far from accurate in practice making it difficult to direct China’s water resource planning in the coming ten years. By introducing the demand forecasting method for the first time, this study built a mathematical prediction model that integrates “data constraint + mathematical prediction + actuarial prediction”. The conclusions are as follows: (1) The new method developed in this research is more accurate than previous prediction methods and has a higher degree of matching with actual data. Despite the problem of trend extrapolation, this model can accurately understand the long-term trend. However, the amount of data must be calculated. (2) China’s consumption of total water, agricultural water, industrial water, domestic water, and ecological water will predict to be 552.9 billion m3, 319.1 billion m3, 95.7 billion m3, 95.5 billion m3 and 42.6 billion m3 in 2030, respectively, as well as 504.7 billion m3, 281.1 billion m3, 61.4 billion m3, 101.4 billion m3, and 60.8 billion m3 in 2050, respectively, indicating obvious structural changes. (3) China’s industrial structure evolves frequently; therefore, water resource forecasting must account for changes in industrial structure as well as geographical variances. In North China, agricultural water demand will fall, but ecological water investment will rise. The planning and distribution of the nation’s water resources will significantly be influenced theoretically and practically by the development of this methodology study. This method is universal and can be used for long-term prediction of future water demand in other countries.

YUCCA, belonging to the class B flavin-dependent monooxygenases, catalyzes the rate-limiting step for endogenous auxin synthesis and is implicated in plant-growth regulation and stress response. Systematic analysis of the YUCCA gene family and its stress response benefits the dissection of regulation mechanisms and breeding applications. In this study, 12 YUCCA genes were identified from the mungbean (Vigna radiata L.) genome and were named based on their similarity to AtYUCCAs. Phylogenetic analysis revealed that the 12 VrYUCCAs could be divided into 4 subfamilies. The evidence from enzymatic assays in vitro and transgenetic Arabidopsis in vivo indicated that all the isolated VrYUCCAs had biological activity in response to IAA synthesis. Expression pattern analysis showed that functional redundancy and divergence existed in the VrYUCCA gene family. Four VrYUCCAs were expressed in most tissues, and five VrYUCCAs were specifically highly expressed in the floral organs. The response toward five stresses, namely, auxin (indole-3-acetic acid, IAA), salinity, drought, high temperatures, and cold, was also investigated here. Five VrYUCCAs responded to IAA in the root, while only VrYUCCA8a was induced in the leaf. VrYUCCA2a, VrYUCCA6a, VrYUCCA8a, VrYUCCA8b, and VrYUCCA10 seemed to dominate under abiotic stresses, due to their sensitivity to the other four treatments. However, the response modes of the VrYUCCAs varied, indicating that they may regulate different stresses in distinct ways to finely adjust IAA content. The comprehensive analysis of the VrYUCCAs in this study lays a solid foundation for further investigation of VrYUCCA genes’ mechanisms and applications in breeding.

Bending resistance is regarded as a significant factor in achieving the widespread application of copper foils. This paper proposed a novel double-cross rolling process, which improved the bending fatigue life of copper foils by as much as 101% in comparation with conventional rolling. In this study, an investigation was conducted into the effects of double-cross rolling on the microstructure and texture of copper foils and the strengthening mechanism during the bending test was discussed. Results show that double-cross rolling could change the deformation behavior on bright and matt surfaces, resulting in equiaxed grains on both surfaces and some fine grains. Both surfaces were dominated by Brass and S textures, which can be attributable to from non-uniform and shear deformation respectively. Weak Goss and Cube textures resulted from the fine grains remaining after heat treatment. After double-cross rolling, the strain induced subgrains of copper foils facilitated the migration of grain boundary and grain rotation, thus improving bending property.

With the normalization of epidemic prevention and control, the expression of the public’s demand for health information on online platforms continues to increase, while knowledge hiding behavior has seriously hindered the communication and dissemination of epidemic prevention knowledge and has a negative impact on public communication and access to health information in the socialized Q&A communities. Therefore, further stimulating diving users' activity and reducing their knowledge hiding behavior have become the key to the sustainable development of epidemic prevention and control and communities. Based on the social cognition theory, from the perspective of individual cognition and external environment, this study constructs a theoretical model of the influencing factors of users’ knowledge hiding behavior in the socialized Q&A communities in the post-epidemic era and puts forward relevant assumptions. 151 effective questionnaires are collected and an empirical analysis is carried out by using the structural equation model. The results show that outcome expectation, community atmosphere, and requesting negatively affect knowledge hiding behavior; self-efficacy, outcome expectation, and community atmosphere negatively affect the three different types of knowledge hiding behavior, which are evasive hiding, playing dumb, and rationalized hiding; community atmosphere positively affects outcome expectation, which plays a significant intermediary effect between community atmosphere and knowledge hiding behavior. The research content and relevant conclusions of this study deepen and expand the connotation and extension of knowledge hiding behavior in the negative performance of Q&A communities. From the perspective of practical application, it can also effectively reduce knowledge hiding behavior, grasp the development direction of public health needs, and strengthen the dissemination of epidemic prevention and control knowledge.

Background. Uncut Roux-en-Y gastrojejunostomy, recently developed in China, is useful in the treatment of distal gastric cancer. This study is aimed at comparing laparoscopic gastric jejunum uncut Roux-en-Y anastomosis with conventional anastomosis in the surgical treatment of distal gastric malignancy. Methods. In this retrospective study, the clinical data of 178 patients and their follow-up records were analyzed. 112 cases (uncut group) were the observation group for stomach jejunum uncut Roux-en-Y anastomosis, the control group for the stomach, 66 cases (conventional group) were for jejunum Roux-en-Y anastomosis and Billroth I and Billroth II anastomosis. A comparison between the two groups was conducted based on the general situation of the patients, TNM stage, and one-year survival rate. Results. There was no significant difference reported between the two groups in terms of the general situation and TNM stage. A comparison on postoperative complications between the two groups revealed that the postoperative bleeding was 0.9% and 6.1%, the bile reflux gastritis was 1.8% and 9.1%, the anastomotic leakage was 0.0% and 3.0%, the delayed gastric emptying was 0.9% and 7.6%, and the overall complications was at 3.6% and 25.8%, which was significantly lower in the observation group than in the control group, and the difference was statistically significant. Notably, there was no significant difference in 1-year survival rate between the two groups. Conclusion. Laparoscopic gastric jejunal uncut Roux-en-Y anastomosis significantly reduces the risk of postoperative complications of the digestive tract. Its operation is easy and exhibits an effective curative effect.

Abstract Background The WRKY proteins are a superfamily of transcription factors and members play essential roles in the modulation of diverse physiological processes, such as growth, development, senescence and response to biotic and abiotic stresses. However, the biological roles of the majority of the WRKY family members remains poorly understood in soybean relative to the research progress in model plants. Results In this study, we identified and characterized GmWRKY40, which is a group IIc WRKY gene. Transient expression analysis revealed that the GmWRKY40 protein is located in the nucleus of plant cells. Expression of GmWRKY40 was strongly induced in soybean following infection with Phytophthora sojae, or treatment with methyl jasmonate, ethylene, salicylic acid, and abscisic acid. Furthermore, soybean hairy roots silencing GmWRKY40 enhanced susceptibility to P. sojae infection compared with empty vector transgenic roots. Moreover, suppression of GmWRKY40 decreased the accumulation of reactive oxygen species (ROS) and modified the expression of several oxidation-related genes. Yeast two-hybrid experiment combined with RNA-seq analysis showed that GmWRKY40 interacted with 8 JAZ proteins with or without the WRKY domain or zinc-finger domain of GmWRKY40, suggesting there were different interaction patterns among these interacted proteins. Conclusions Collectively, these results suggests that GmWRKY40 functions as a positive regulator in soybean plants response to P. sojae through modulating hydrogen peroxide accumulation and JA signaling pathway.

Pancreatic cancer is one of the most lethal cancers and its prognosis is extremely poor. Clarification of molecular mechanisms and identification of prognostic biomarkers are urgently needed. Though we previously found that LGMN was involved in pancreatic carcinoma progression, the upstream regulation of LGMN remains unknown. We used reliable software to search for the potential transcription factors that may be related with LGMN transcription, we found that ELK1 could be a new regulator of LGMN transcription that binded directly to the LGMN promoter. Moreover, knocking down of ELK1 reduced pancreatic cancer cells proliferation, invasion and survival, while LGMN restored the malignancy of pancreatic cancer in vitro and in vivo. Overexpression of ELK1 further increased cancer cells proliferation, invasion and survival. Clinically, ELK1 and LGMN were positively correlated with clinical stage, degree of differentiation and Lymph node infiltration. ELK1 and LGMN were identified as independent prognostic factors for overall survival. The patients with low expression of ELK1/LGMN survived an average of 29.65 months, whereas those with high expression of ELK1/LGMN survived an average of 16.67 months. In conclusive, our results revealed a new mechanism by which ELK1 promoted the progression of pancreatic cancer via LGMN and conferred poor prognosis.

When performing the diffuse optical tomography (DOT) of the breast, the mismatch between the forward model and the experimental conditions will significantly hinder the reconstruction accuracy. Therefore, the reference measurement is commonly used to calibrate the measured data before the reconstruction. However, it is complicated to customize corresponding reference phantoms based on the breast shape and background optical parameters of different subjects in clinical trials. Furthermore, although high-density (HD) DOT configuration has been proven to improve imaging quality, a large number of source-detector (SD) pairs also increase the difficulty of multi-channel correction. To enhance the applicability of the breast DOT, a data self-calibration method based on an HD parallel-plate DOT system is proposed in this paper to replace the conventional relative measurement on a reference phantom. The reference predicted data can be constructed directly from the measurement data with the support of the HD-DOT system, which has nearly a hundred sets of measurements at each SD distance. The proposed scheme has been validated by Monte Carlo (MC) simulation, breast-size phantom experiments, and clinical trials, exhibiting the feasibility in ensuring the quality of the DOT reconstruction while effectively reducing the complexity associated with relative measurements on reference phantoms.

Purpose: The stimulator of interferon genes (STING) pathway plays a crucial role in antitumor immunity, and it is strictly regulated by many types of post-translational modifications. However, the contribution of acetylation involved in the regulation of STING to endometrial tumorigenesis remains unclear. Methods: We attempted to identify the key role of STING in endometrial carcinoma (EC) tissue and cell lines and explore its epigenetic regulation mechanism by HDACs that are critically involved in EC. We used IHC and qRT-PCR to detect the protein level and mRNA level of STING expression in endometrial carcinoma tissues, then explored the potential role of STING in tumor proliferation and apoptosis by CCK8 and flow cytometry, and identified the STING effect in the tumorigenicity by a mouse xenograft assay. We explored the possible relationship of acetylation alteration in STING regulation by ChIP analysis and Co-IP, and we knocked out STING in ECC1 and Ishikawa cells using CRISPR-Cas9 to further confirm the critical role of STING restoration induced by HDAC3 inhibitor RGFP-966 in the proliferation and apoptosis. Results: We found that STING expression was largely decreased and worked as an important regulator of cell proliferation and apoptosis; either activated or overexpressed STING, with both pharmacological and genetic approaches, largely blocked cell proliferation and induced apoptosis in EC. Moreover, STING expression was deregulated by both β-estradiol and HDAC3. Mechanically, we determined that HDAC3 can interact with β-estradiol-ERα and induce deacetylation of histone 3 lysine 4 at the STING promoter, thereby decreasing STING expression. Inhibition of HDAC3 increased STING expression, thereby inhibiting tumorigenesis. Conclusion: This study reveals a novel molecular mechanism by which HDAC3 inhibits STING transcription via β-estradiol-ERα and provides a promising therapy (a combination of HDAC and STING) for combating endometrial cancer.

Waterlogging occurs when field soil is saturated with water induced by extensive rainfall or improper irrigation, which is a severe abiotic stress influencing wheat plant growth and yield production. At the germination stage, waterlogging usually induces rot of seeds and reduced germination rate and seedling survival. Development of tolerant wheat varieties is the most efficient approach to improve seed germination and mitigate the damages caused by waterlogging. In this study, we screened 432 wheat accessions at germination stage by waterlogging treatment, and identified 27 tolerant accessions with a germination rate of over 80% after treatment. To identify quantitative trait loci (QTL) for waterlogging tolerance, two segregation populations were developed by crossing waterlogging-tolerant cultivars Shannong 135 and Huaimai 18 with sensitive cultivars Siyang 936 and CD1840, respectively. Three QTL qWlg5A, qWlg7B and qWlg2D for waterlogging tolerance were detected on chromosomes 5A, 7B and 2D through bulked segregation analysis genotyped by wheat 55K SNP array. Two, one, and two kompetitive allele specific PCR (KASP) assays linked with qWlg5A, qWlg7B and qWlg2D were developed and validated in the two populations, respectively. The identified waterlogging tolerant germplasm lines, the QTL for waterlogging tolerance and the high-throughput KASP markers, were highly valuable in improving waterlogging tolerance in wheat-marker-assisted breeding.

Positron Annihilation Technique is very sensitive to vacancy defects and small vacancy clusters and has been used as supplementary for TEM in radiation damage research. In this paper, we evaluated the displacement damage in solids induced by positrons and its effect on defects measurement, which has not been considered carefully in most PAT application. Displacement damage could only be produced when kinetic energy of positron exceeds a threshold determined by average displacement energy and the mass of atoms. Based on the latest models of displacement damage and numerical Mott elastic differential cross section of positrons interacting with atoms, displacement cross section for fast positron in solids was calculated and compared with that for electrons. In the energy range from 1 MeV to 100 MeV, electrons will produce more displacement defects than positrons do, especially in high-Z materials. Number of total defects depends on the primary displacement cross section and the size of damage cascades. To get distributions of defects and terminating position of positrons in materials, a Monte Carlo tool with full damage model was developed and validated. From the results of Monte Carlo modeling, the defects -generated by mono-energy positron beam have shallower distribution than the stopping position of positrons, indicating separation of defects and positrons and this fact benefits defects measurement because displacement damage produced by fast probing positron beam have little affection on the measurement. But for the positron beams from radioactive sources (such as 22Na and 64Cu) with continuous energy spectrum (from 0 to Emax), the depth profile of defects and terminated position of positrons overlap with each other and this fact indicated that defects measurement in materials will be affected by extra defects produced by positrons. The density of defects produced by positron beam is quite low with a typical value of about 0.01–0.1 cm−3 per positron in low-Z materials, which could be comparable with the defect density limitation if enough accumulated positrons are implanted into materials during measurement. For commonly used structural metals (such as Fe, Mn, Cu and metals with Z number higher than 40), using positron from 22N and 64Cu directly to measure defects density is quite safe because the energy is smaller than the displacement threshold.

Abstract Background Few prospective birth cohort studies are available on the effects of prenatal and early-life exposures on food allergy and eczema among Chinese children. The aim of this study was to evaluate the influence of prenatal and early-life exposures on food allergy and eczema during the first year of life in a prospective birth cohort study. Methods This study was based on a prospective, observational birth cohort of 976 mother-child pairs in three Streets in Changsha, China from January to December 2015. Data on prenatal, early-life exposures and allergic outcomes were obtained from questionnaires collected at birth, and 1, 3, 6, 8, and 12 months of age. Multivariate logistic regression models were performed to estimate the effects of prenatal and early-life exposures on food allergy and eczema. Results Common risk factors for food allergy and eczema in infancy were parental history of allergy, while moderate eggs consumption (3–4 times/week) during pregnancy was protective for both of them compared with low consumption (≤ 2 times/week). Factors only associated with food allergy were maternal aquatic products consumption during pregnancy, number of older siblings and age of solid food introduction, whereas factors only associated with eczema were maternal milk or milk products consumption during pregnancy, maternal antibiotic exposure during pregnancy, season of birth and antibiotic exposure through medication during the first year of life. Conclusion Our study suggests that factors associated with food allergy and eczema are multifaceted, which involving hereditary, environmental and nutritional exposures. Furthermore, differential factors influence the development of food allergy and eczema in infants.

Abstract Background Cottonseed oil and protein content as well as germination traits are major indicators of seed quality. However, the responses of these indicators to plant density and mepiquat chloride (MC) are still uncertain. To investigate plant density and MC effects on cottonseed yield and main quality parameters, we conducted a two-year field experiment including four plant densities (1.35, 2.55, 3.75 and 4.95 plants·m− 2) and two doses of MC (0 and 135 g·hm− 2) in Dafeng, Jiangsu Province, in 2013 and 2014. Results The application of MC reduced plant height, fruit branch length and fruiting branch number under different plant densities, resulting in a lower and more compact plant canopy. Cottonseed yield showed a nonlinear increase as plant density increasing and achieved the highest value at 3.75 plants·m− 2, regardless of MC application. No significant interactions were found between plant density and MC for cottonseed yield and quality parameters. The 100-seed weight, cottonseed oil content and vigor index significantly decreased as plant density increased, while these parameters significantly increased with MC applying under different plant densities. Seed vigor index was positively correlated with 100-seed weight and seed oil content across different plant densities and MC treatments. Conclusions Thus, application of MC could realize a win-win situation between cottonseed yield and main quality parameters under various densities; and plant density of 3.75 plants·m− 2 combined with 135 g·hm− 2 of MC applying is optimal for high cottonseed yield and quality in this cotton production area.

Abstract Background Serratia marcescens is a chitinolytic bacterium that can potentially be used for consolidated bioprocessing to convert chitin to value-added chemicals. Currently, S. marcescens is poorly characterized and studies on intracellular metabolic and regulatory mechanisms would expedite development of bioprocessing applications. Results In this study, our goal was to characterize the metabolic profile of S. marcescens to provide insight for metabolic engineering applications and fundamental biological studies. Hereby, we constructed a constraint-based genome-scale metabolic model (iSR929) including 929 genes, 1185 reactions and 1164 metabolites based on genomic annotation of S. marcescens Db11. The model was tested by comparing model predictions with experimental data and analyzed to identify essential aspects of the metabolic network (e.g. 138 essential genes predicted). The model iSR929 was refined by integrating RNAseq data of S. marcescens growth on three different carbon sources (glucose, N-acetylglucosamine, and glycerol). Significant differences in TCA cycle utilization were found for growth on the different carbon substrates, For example, for growth on N-acetylglucosamine, S. marcescens exhibits high pentose phosphate pathway activity and nucleotide synthesis but low activity of the TCA cycle. Conclusions Our results show that S. marcescens model iSR929 can provide reasonable predictions and can be constrained to fit with experimental values. Thus, our model may be used to guide strain designs for metabolic engineering to produce chemicals such as 2,3-butanediol, N-acetylneuraminic acid, and n-butanol using S. marcescens.

Background/Aims: End-stage renal disease (ESRD), characterized by progressive loss of rental function during the disease course, has been reported to be correlated with immune dysregulation. To date, a majority of previous studies on immune response to ESRD have been focused on the T-cell response. This prospective study was to assess the B-cell receptor (BCR) heavy chain repertoire in ESRD patients. Materials and methods: A total of 10 ESRD patients and six healthy controls were prospectively enrolled in this study. BCR immunoglobulin heavy chain (IGH) repertoire in the peripheral blood from ESRD patients and healthy individuals were analyzed by means of next generation sequencing (NGS) in combination with multiplex PCR, Illumina sequencing, and the international ImMunoGeneTics database (IMGT). Results: Abnormal BCR complementary-determining region 3 (CDR3) sequences were identified in relation to ESRD. This paper explores how the sequence of positive or negative comments and the degree of product involvement affect the purchase intention of consumers through a $2\times 2$ experiment. The results demonstrated that the sequence of positive or negative supplementary comments will affect the consumer’s purchase intention, while the degree of product involvement will adjust the relationship between them. Specifically, the conclusions include three points: (1) compared to initial comments, supplementary comments have a greater impact on a consumer’s purchase intention; (2) when the initial comments are positive but the supplementary ones are negative, consumers who have a high degree of product involvement would have a greater purchase intention than those who demonstrate a low degree of product involvement; (3) In the case of negative initial comments / positive supplementary comments, consumers with low product involvement are more likely to purchase than consumers who have a high product involvement.
