Abstract Parkinson’s disease (PD) is a prevalent type of neurodegenerative disorder. AVE0991, a non-peptide analogue of Ang-(1–7), by which the progression of PD has been discovered to be ameliorated, but the specific mechanism whereby AVE0991 modulates the progression of PD re-mains unclear. The mice overexpressing human α-syn (A53T) were established to simulate PD pathology, and we also constructed an in vitro model of mouse dopaminergic neurons overexpressing hα-syn (A53T). The [18F] FDG-PET/CT method was employed to assess FDG uptake in human α-syn (A53T) overexpressing mice. Levels of lnc HOTAIRM1 and miR-223-3p were detected via qRT-PCR. Flow cytometry was deployed to assay cell apoptosis. Here, we found that AVE0991 improved behaviour disorders and decreased α-syn expression in the substantia nigra of mice with Parkinson’s disease. AVE0991 inhibited the apoptosis of dopaminergic neurons overexpressing hα-syn (A53T) via lncRNA HOTAIRM1. MiR-223-3p binds to HOTAIRM1 as a ceRNA and directly targets α-syn. Moreover, miR-223-3p level in peripheral blood was found negatively correlated with the α-syn. Our present study shows that the angiotensin-(1–7) analogue AVE0991 targeted at the HOTAIRM1/miR-223-3p axis to degrade α-synuclein in PD mice, and showed neuroprotection in vitro.

ObjectiveTo investigate the expression of transcription factor forkhead box protein O (FOXO) genes in gastric cancer tissues and their correlation and clinical significance with Helicobacter pylori (Hp) infection. MethodsThe expression levels of FOXOs genes (including FOXO1, FOXO3, FOXO4, and FOXO6) were detected by immunohistochemistry in cancer tissues and paracancerous tissues from 41 gastric cancer patients with Hp (−) and 29 gastric cancer patients with Hp (+) , as well as in gastric tissues from 30 healthy individuals. The correlation between FOXOs expression and Hp infection, clinical pathological features was analyzed. The relationship between FOXOs expression and survival prognosis of gastric cancer patients was analyzed using the Kaplan-Meier plotter. 【Objective】Grain thickness is one of the controlling factors affecting rice yield and taste quality of rice. QTL mapping for grain thickness in rice was conducted by using recombinant inbred lines (RIL) with the indica-japonica cross (V20B/CPSLO17) genetic background and major QTLs of grain thickness were obtained, providing a scientific basis for the discovery of new grain thickness genes and the development of functional molecular markers.【Method】Based on the highdensity genetic linkage map and the indica-japonica cross genetic background of RIL populations, combined with the grain thickness phenotype data of RIL populations under three different ecological environments (Guiyang, Guizhou in 2020; Guiding, Guizhou in 2021; Sanya, Hainan in 2021), QTL mapping and genetic effect analysis were performed by the ICIM-ADD method of IciMapping 4.0 software.【Result】The study showed that the grain thickness trait in rice exhibited a continuous unimodal distribution under all three ecological environments and was not affected by planting environmental factors significantly. Five grain thickness QTLs (qGT3-1, qGT5-1, qGT5-2, qGT8-1, and qGT10-1) were detected on chromosomes 3, 5, 8, and 10 in rice under the three different ecological environments. The beneficial alleles of these five QTLs were all from the parent V20B, with LOD values ranging from 3.431 to 14.081 and a range of phenotypic contribution rates from 5.479% to 26.483%. Two QTLs (qGT5-1 and qGT5-2) had a phenotypic contribution rate greater than 10%, with qGT5-2 being the only one repeatedly detected in two planting environments (Guiyang, Guizhou in 2020 and Sanya, Hainan in 2021), explaining 26.483% and 14.933% of the population's phenotypic variance, respectively. The physical distance of QTL qGT5-2 on the chromosome was about 3.9 kb, with only one candidate gene (LOC_Os05g07920); the physical distance of qGT8-1 was about 2.3 kb, with only one candidate gene (LOC_Os08g10360).【Conclusion】Grain thickness in rice is a quantitative genetic trait regulated by multiple genes. qGT5-2 is major QTL with stable heredity and high contribution rate for grain thickness, which has significant application potential in the discovery of grain thickness regulatory genes and the breeding of new high-quality and high-yield rice varieties.

Abstract Nanobodies (Nbs), the smallest antigen-binding fragments with high stability and affinity derived from the variable domain of naturally occurring heavy-chain-only antibodies in camelids, have been shown as an efficient way to improve the specificity to tumors for photodynamic therapy (PDT). Nonetheless, the rapid clearance of Nbs in vivo restricts the accumulation and retention of the photosensitizer at the tumor site causing insufficient therapeutic outcome, especially in large-volume tumors. Herein, we develop photodynamic conjugates, MNB-Pyra Nbs, through site-specific conjugation between 7D12 Nbs and type I photosensitizer MNB-Pyra (morpholine-modified nile blue structure connected to pyrazolinone) in a 1:2 ratio. The photosensitizers with long-term retention can be released at the tumor site by reactive oxygen species cleavage after illumination, accompanied with fluorescence recovery for self-reporting the occurrence of PDT. Ultimately, a single dose of MNB-Pyra Nbs demonstrate highly effective tumor suppression with high biosafety in the large-volume tumor models after three rounds of PDT. This nanobody conjugate provides a paradigm for the design of precise long-time retention photosensitizers and is expected to promote the development of PDT.

Given the side effects associated with synthetic antihypertensive drugs, there is a growing need among researchers to investigate angiotensin-converting enzyme (ACE) inhibitory peptides derived from food protein as safer therapeutic alternatives. This study used seabuckthorn (Hippophae rhamnoides L.) seed meal as the raw material, and the protein was extracted by alkaline extraction and acid precipitation. After enzymatic digestion, peptides with molecular weight less than 3 kDa were selected for study. The screened peptide had an IC50 value of 4.358 mg/mL on ACE with a non-competitive inhibition mechanism and good inhibition stability. By employing infrared (IR) analysis, exclusively β-fold and β-helix structures were identified in the hydrolysate, while no other structural motifs were detected. X-ray diffraction revealed that it had an irregular amorphous structure. The peptide contains 17 amino acids that are both highly acidic and hydrophobic, with glutamic acid ranking first in terms of the number of individual amino acids. Compared with the database (NCBI, Uniport), ten peptides with ACE inhibitory activity were detected, and molecular docking showed the mechanism of each peptide inhibiting ACE, FRVAWTEKNDGQRAPLANN, LIISVAYARVAKKLWLCNMIGDVT-TEQY, VIRSRASDGCLEVKEFEDIPP, AGGGG-GGGGGGSRRL, LQPREGPAGGTT-ALREELSLGPEAALDTPPAGP, DDEARINQLFL, FAVSTLTSYDWSDRDDATQGR-KL, RQLSLEGSGLGVEDLKDN, GGGGGGGGGGGGGGGIGGGGGGGGGGGAR, and KEALGEGCFGNRIDRIGD. According to the results, AGGGGGG-GGGGSRRL is more stable in binding to ACE and may have better inhibitory activity. It has been shown that seabuckthorn protein can be an alternative source of ACE inhibitory peptides.

Abstract Traumatic brain injury (TBI) is a major cause of morbidity and mortality globally. We unveiled the diagnostic value of serum NLRP3, metalloproteinase‐9 (MMP‐9) and interferon‐γ (IFN‐γ) levels in post‐craniotomy intracranial infections and hydrocephalus in patients with severe craniocerebral trauma to investigate the high risk factors for these in patients with TBI, and the serological factors predicting prognosis, which had a certain clinical predictive value. Study subjects underwent bone flap resection surgery and were categorized into the intracranial infection/hydrocephalus/control (without postoperative hydrocephalus or intracranial infection) groups, with their clinical data documented. Serum levels of NLRP3, MMP‐9 and IFN‐γ were determined using ELISA kits, with their diagnostic efficacy on intracranial infections and hydrocephalus evaluated by receiver operating characteristic curve analysis. The independent risk factors affecting postoperative intracranial infections and hydrocephalus were analysed by logistic multifactorial regression. The remission after postoperative symptomatic treatment was counted. The intracranial infection/control groups had significant differences in Glasgow Coma Scale (GCS) scores, opened injury, surgical time and cerebrospinal fluid leakage, whereas the hydrocephalus and control groups had marked differences in GCS scores, cerebrospinal fluid leakage and subdural effusion. Serum NLRP3, MMP‐9 and IFN‐γ levels were elevated in patients with post‐craniotomy intracranial infections/hydrocephalus. The area under the curve values of independent serum NLRP3, MMP‐9, IFN‐γ and their combination for diagnosing postoperative intracranial infection were 0.822, 0.722, 0.734 and 0.925, respectively, and for diagnosing hydrocephalus were 0.865, 0.828, 0.782 and 0.957, respectively. Serum NLRP3, MMP‐9 and IFN‐γ levels and serum NLRP3 and MMP‐9 levels were independent risk factors influencing postoperative intracranial infection and postoperative hydrocephalus, respectively. Patients with hydrocephalus had a high remission rate after postoperative symptomatic treatment. Serum NLRP3, MMP‐9 and IFN‐γ levels had high diagnostic efficacy in patients with postoperative intracranial infection and hydrocephalus, among which serum NLRP3 level played a major role.

Abstract Background This study aimed to investigate the feasibility and precision of using a 3D-printed template for femoral tunnel placement in guiding the optimal positioning of the Internal anatomical stop and Low tension maintenance (IDEAL) bone tunnel during single-bundle anterior cruciate ligament (ACL) reconstruction. Methods A retrospective analysis was conducted on 40 patients who underwent arthroscopic single-bundle ACL reconstruction at our hospital between April 2021 and November 2021. In the direct vision group, the IDEAL bone tunnel was positioned using radiofrequency localization directly visualized at the stump. In the 3D-printed positioning group, preoperative CT scans and Digital Imaging and Communications in Medicine (DICOM) data were employed. Following the Quadrant method by Bernard, the femoral tunnel’s depth was set at 25% and its height at 29%. Postoperative plain CT scans enabled the reconstruction of 3D models for both groups. The accuracy of femoral tunnel placement was then compared. Results The central locations of the bone tunnels in the direct vision group were at a mean depth of 25.74 ± 1.84% and a height of 29.22 ± 2.97%. In the 3D printing localization group, these values were 25.39 ± 2.98% for depth and 28.89 ± 2.50% for height, respectively. No significant differences were found in tunnel positioning between the groups. Both groups demonstrated statistically significant improvements in International Knee Documentation Committee Subjective Knee Form (IKDC) and Lysholm scores postoperatively, with no significant differences observed 12 months post-surgery. Conclusion The findings of this study suggest that 3D printing-assisted arthroscopic IDEAL point femoral tunnel positioning and conventional arthroscopic positioning are feasible and effective for ACL reconstruction. Using 3D printing technology to design femoral anchor points in ACL reconstruction allows for the customization of anterior fork reconstruction and precise bone tunnel positioning, supporting the goal of individualized and accurate reconstruction.

As a superfruit, wolfberry has extremely high nutritional value, and how to enhance the accessibility of its nutrients is the core of current research. This study focused on exploring the kinetic model of Lactobacillus plantarum CCFM1050 fermentation of wolfberry and the potential alterations of antioxidant activity and volatile flavor compounds induced by lactic acid fermentation. we monitored cell counts, product formation, and substrate changes over a 72-h period of wolfberry fermentation. A kinetic model was developed to illustrate cell growth, substrate consumption, and product accumulation during wolfberry pulp fermentation. Phenolic substance analysis revealed a significant increase in total phenol and flavonoid content in wolfberry pulp during fermentation, reaching 1.16 and 1.15 times, respectively, compared to pre-fermentation levels. The elevated levels of phenolic substances led to a substantial increase in DPPH and ABTS free radical scavenging rates in fermented wolfberry pulp, reaching 67.16% and 32.10%, respectively. Volatile components of samples were analyzed using the HS-GC-IMS method, and fingerprints of wolfberry pulp before and after fermentation were established. A total of 51 compounds were identified, including 12 alcohols, seven aldehydes, two acids, eight esters, and 12 ketones, contributing to an enhanced flavor profile in the fermented wolfberry pulp. This study is helpful for understanding the kinetic changes in the lactic acid fermentation of wolfberry, the changes of antioxidant active substances and VOCs, and provides guidance for the industrial processing of wolfberry.

Bacterial infections are a great threat to human health, and the irrational use of antibiotics has largely compromised the efficacy of antibiotic therapy due to the emergence of drug‐resistant pathogens. It is known that synthesizing new antibiotics is difficult and time‐consuming. In this case, developing antibiotics‐independent antibacterial approaches is of great importance and significance. In the past decade, various functional nanomaterials have shown great potentials in the treatment of bacterial infections. Among these nanomaterials, transition metal carbides or nitrides, namely MXene, have attracted much attention. As the novel 2D nanosheets, MXene can serve either as a direct antibacterial agent due to its intrinsic antibacterial activity and photothermal effect, or as an efficient carrier to load photosensitizers and photocatalysts for photodynamic and photocatalytic therapy. In recent few years, the number of literatures regarding MXene‐based antibacterial therapy has increased rapidly. Thus, it is the time to systematically summarize the applications of MXene in the treatment of bacteria, especially those with drug resistance. Herein, it is aimed to summarize the preparation methods for MXene and provide a comprehensive understanding of its properties and applications in antibacterial therapy. Also, its use for bacterial detection and the challenges for practical use are discussed.

Litsea cubeba essential oil (LCEO) has garnered widespread attention due to its robust biological activity. However, challenges such as high volatility, limited water solubility, and low bioavailability impede its application. Nano-emulsion encapsulation technology offers an effective solution to these issues. In this study, we prepared litsea cubeba essential oil nano-emulsion (LCEO-NE) for the first time using whey protein (WP) as the emulsifier through an ultrasonic-assisted method, achieving high efficiency with minimal energy consumption. Transmission electron microscopy and dynamic light scattering analyses revealed that the nanoparticles were uniformly spherical, with a particle size of 183.5 ± 1.19 nm and a zeta potential of −35.5 ± 0.95 mV. Stability studies revealed that LCEO-NE exhibited excellent thermal and salt stability, maintaining its integrity for up to four weeks when stored at 4 °C and 25 °C. In vitro digestion assays confirmed the digestibility of LCEO-NE. Furthermore, evaluation of the DPPH, ABTS, and antimicrobial activities revealed that LCEO-NE displayed superior bacteriostatic and antioxidant properties compared to LCEO. Scanning electron microscopy elucidated that its bacteriostatic effect involved the disruption of bacterial microstructure. Hemocompatibility and cytotoxicity assays demonstrated the safety of LCEO-NE within the effective concentration range. This research supports the utilization of nanoparticles for encapsulating LCEO, thereby enhancing its stability and bioactivity, and consequently expanding its applications in the food and pharmaceutical industries.

Abstract Volatile solid additives have attracted increasing attention in optimizing the morphology and improving the performance of currently dominated non‐fullerene acceptor‐based organic solar cells (OSCs). However, the underlying principles governing the rational design of volatile solid additives remain elusive. Herein, a series of efficient volatile solid additives are successfully developed by the crossbreeding effect of chalcogenation and iodination for optimizing the morphology and improving the photovoltaic performances of OSCs. Five benzene derivatives of 1,4‐dimethoxybenzene (DOB), 1‐iodo‐4‐methoxybenzene (OIB), 1‐iodo‐4‐methylthiobenzene (SIB), 1,4‐dimethylthiobenzene (DSB) and 1,4‐diiodobenzene (DIB) are systematically studied, where the widely used DIB is used as the reference. The effect of chalcogenation and iodination on the overall property is comprehensively investigated, which indicates that the versatile functional groups provided various types of noncovalent interactions with the host materials for modulating the morphology. Among them, SIB with the combination of sulphuration and iodination enabled more appropriate interactions with the host blend, giving rise to a highly ordered molecular packing and more favorable morphology. As a result, the binary OSCs based on PM6:L8‐BO and PBTz‐F:L8‐BO as well as the ternary OSCs based on PBTz‐F:PM6:L8‐BO achieved impressive high PCEs of 18.87%, 18.81% and 19.68%, respectively, which are among the highest values for OSCs.

【Objective】Grain size is one of the main agronomic traits affecting rice yield. A recombinant inbred line population derived from indica rice variety V20B and slender javanica rice variety CPSLO17 was used for QTL mapping of brown rice grain size. It aims to explore the genetically-stable main effect QTLs, providing new genetic resources and scientific basis for developing high-quality and high-yield rice varieties.【Method】Based on the high-density genetic linkage map of V20B/CPSLO17 genetic background, the brown rice grain size phenotype data of 150 recombinant inbred lines under four environments (Guiyang, Guizhou in 2019, Guiyang, Guizhou in 2020, Guiding, Guizhou in 2021, and Sanya, Hainan in 2021) were used for QTL scanning by the ICIM-ADD method of IciMapping 4.0 software.【Result】The grain size of brown rice from parent V20B was significantly larger than that from CPSLO17, and the grain sizes of brown rice from recombinant inbred lines showed significant differences among the four environments, all exhibiting a continuous unimodal distribution. A total of 11 QTLs related to brown rice grain size distributed on 7 chromosomes were found in the four environments, with their LOD values ranging from 3.00 to 14.57. Four QTLs including qRGS5, qRGS7.1, qRGS7.2, and qRGS11.3 had a phenotypic variance explained (PVE) over 10%, of which the qRGS5 was repeatedly detected in all four environments, with its PVE of 29.71%, 28.77%, 17.27% and 12.50%, respectively. The synergistic alleles of three QTLs (qRGS2.1, qRGS2.2 and qRGS12) derived from the parent CPSLO17, while the other eight synergistic alleles (qRGS4.1, qRGS5, qRGS7.1, qRGS7.2, qRGS8, qRGS11.1, qRGS11.2 and qRGS11.3) derived from the parent V20B, and the synergistic alleles of QTL qRGS5 detected in all four environments derived from the parent V20B.【Conclusion】The difference of brown rice grain size between parents V20B and CPSLO17 is extremely significant. The brown rice grain size trait is affected by the environments. The QTL qRGS5 is a genetically-stable main effect QTL that affects grain size in brown rice, which has practical value in new gene discovery for brown rice grain size and molecular breeding for high-yield and high-quality rice.

Alluvial parent material soil is an important soil type found on the Qinghai–Tibet Plateau (QTP) in China. However, due to the limited age data for alluvial soils, the relationship between alluvial geomorphological processes and soil pedogenic processes remains unclear. In this study, three representative alluvial parent material profiles on the Buha River alluvial plain in the Qinghai Lake Basin, northeast QTP, were analyzed using the optical luminescence (OSL) dating method. Combined with physical and chemical analyses of the soil, we further analyzed the pedogenic process of alluvial soil. The alluvial parent material of the Buha alluvial plain predominately yielded ages between 11.9 and 9.1 ka, indicating that the alluvial soil began to form during the early Holocene. The development of the alluvial soil on the first-order terrace presents characteristics of entisol with multiple burial episodes, mainly between 8.5 and 4.0 ka, responding to the warm and humid middle Holocene and high lake levels.

The stable carbon isotope composition of total organic matter (δ13Corg) has been utilized in aeolian sediments, serving as an indicator for reconstructing terrestrial paleoenvironments. The Qinghai Lake (QHL) Basin is a climate-sensitive region of significant importance in paleoclimatic reconstruction. However, the reconstructed climatic variations based on δ13Corg in aeolian sediments in the QHL Basin in the northeastern Qinghai-Tibet Plateau (QTP) are lacking, and their paleoclimatic significance remains poorly understood. By conducting δ13Corg measurements on the Niaodao (ND) aeolian profile near QHL, we reconstructed the paleoclimate changes of 11 ka–present. The variation range of the δ13Corg values in the ND profile indicated the terrestrial ecosystems were not the sole contributor to lacustrine organic matter. The δ13Corg values are an indicator of historical temperature changes in the study area, exhibiting similar trends with the reconstruction of Chinese summer temperatures, East Asian air temperature, global temperature, and Northern Hemisphere summer insolation at 37° N. The temperature increased with high frequency and amplitude oscillations, with strong aeolian activity and low total organic carbon accumulation during the Early Holocene. The temperature was maintained at a high and stable level, with the weakest aeolian activity and intensified pedogenesis during the Middle Holocene. The temperature decreased at a high rate, with renewed aeolian activity and weak pedogenesis during the Late Holocene.

Objectives. Sepsis, caused by the body’s response to infection, poses a life-threatening condition and represents a significant global health challenge. Characterized by dysregulated immune response to infection, sepsis may lead to organ dysfunction and failure, ultimately resulting in high mortality rates. The liver plays a crucial role in sepsis, yet the role of differentially expressed genes in septic patients remains unclear in hepatocellular carcinoma (HCC). In this study, we aim to investigate the significance of differentially expressed genes related to sepsis in the occurrence and prognosis of tumors in HCC. Methods. We conducted analyses by obtaining gene transcriptome data and clinical data of HCC cases from The Cancer Genome Atlas (TCGA). Furthermore, we obtained transcriptomic sequencing results of septic patients from the Gene Expression Omnibus (GEO) database, identified intersecting differentially expressed genes between the two, and performed survival analysis on the samples using LASSO and Cox regression analysis. Combining analyses of tumor mutation burden (TMB) and immune function, we further elucidated the mechanisms of sepsis-related genes in the prognosis and treatment of HCC. Results. We established a prognostic model consisting of four sepsis-related genes: KRT20, PAEP, CCR3, and ANLN. Both the training and validation sets showed excellent outcomes in the prognosis of tumor patients, with significantly longer survival times observed in the low-risk group based on this model compared to the high-risk group. Furthermore, analyses, such as differential analysis of tumor mutation burden, immune function analysis, GO/KEGG pathway enrichment analysis, and drug sensitivity analysis, also demonstrated the potential mechanisms of action of sepsis-related genes. Conclusions. Models constructed based on sepsis-related genes have shown excellent predictive ability in prognosis and differential analysis of drug sensitivity among tumor patients. These predictive models can enhance patient prognosis and inform the creation of early treatment protocols for sepsis, consequently aiding in the prevention of sepsis-induced HCC development through the modulation of the overall immune status. This may play a crucial role in patient management and immunotherapy, providing valuable reference for subsequent research.

After adopting a combined approach of data-driven methods and machine learning, the prediction of material performance and the optimization of composition design can significantly reduce the development time of materials at a lower cost. In this research, we employed four machine learning algorithms, including linear regression, ridge regression, support vector regression, and backpropagation neural networks, to develop predictive models for the electrical performance data of titanium alloys. Our focus was on two key objectives: resistivity and the temperature coefficient of resistance (TCR). Subsequently, leveraging the results of feature selection, we conducted an analysis to discern the impact of alloying elements on these two electrical properties.The prediction results indicate that for the resistivity data prediction task, the radial basis function kernel-based support vector machine model performs the best, with a correlation coefficient above 0.995 and a percentage error within 2%, demonstrating high predictive capability. For the TCR data prediction task, the best-performing model is a backpropagation neural network with two hidden layers, also with a correlation coefficient above 0.995 and a percentage error within 3%, demonstrating good generalization ability. The feature selection results using random forest and Xgboost indicate that Al and Zr have a significant positive effect on resistivity, while Al, Zr, and V have a significant negative effect on TCR. The conclusion of the composition optimization design suggests that to achieve both high resistivity and TCR, it is recommended to set the Al content in the range of 1.5% to 2% and the Zr content in the range of 2.5% to 3%.

Abstract Objectives Patients with vascular anomalies (VAs) who receive oral sirolimus may be at high risk of infectious complications. Antibiotic prophylaxis with trimethoprim-sulfamethoxazole (TMP-SMZ) has been advocated. However, there have been few evidence-based analyses on this topic. This study assessed the effect of prophylactic TMP-SMZ on the incidence of infections in VA patients receiving sirolimus monotherapy. Methods A retrospective, multicenter chart review was performed on all VA patients receiving sirolimus treatment from August, 2013 to January, 2021. Results Before January 2017, 112 patients were treated with sirolimus without antibiotic prophylaxis. In the subsequent period, 195 patients were treated with TMP-SMZ for at least 12 months during sirolimus therapy. The percentage of patients with at least one serious infection during the initial 12 months of sirolimus treatment did not differ between the groups (difference, 1.1%; 95% CI − 7.0–8.0%). We observed no difference in the incidence of individual infection or total adverse events between the groups. The rate of sirolimus discontinuation due to adverse events did not differ significantly between groups. Conclusions We demonstrated that prophylactic TMP-SMZ does not decrease the incidence of infection or improve tolerance in VA patients receiving sirolimus monotherapy.

IntroductionThere is a clinical challenge in diagnosing tuberculous pleurisy accurately and promptly, highlighting the urgent need for a rapid and sensitive diagnostic method. This study aimed to evaluate the diagnostic accuracy of metagenomic next-generation sequencing (mNGS) and GeneXpert Mycobacterium tuberculosis (MTB) for identifying tuberculous pleurisy and analyzing the microbial profiles of both tuberculous and non-tuberculous pleural effusions.MethodsThe study enrolled 31 patients with suspected tuberculous pleurisy, of which 15 were confirmed to have tuberculous pleurisy and subsequently allocated to the tuberculous pleurisy group (TP group), while the remaining 16 individuals were assigned to the non-tuberculous pleurisy group (NTP group). mNGS and GeneXpert MTB were performed on pleural effusion samples, and the diagnostic accuracy of both tests was compared. We employed established formulas to compute crucial indicators, including sensitivity, specificity, missed diagnosis rate, misdiagnosed rate, positive predictive value (PPV), and negative predictive value (NPV).ResultsThe results showed that both tests had high specificity (100%) and positive predictive value (100%) for detecting tuberculous pleurisy, along with comparable sensitivity (46.67% for mNGS and 40.0% for GeneXpert MTB). Further analysis of the combined efficacy of mNGS and GeneXpert MTB showed that the combined test had a sensitivity of 66.67% and a specificity of 100%. mNGS analysis revealed that MTB was detected in 7 out of 15 patients with tuberculous pleural effusions, while non-tuberculous pleural effusions were associated with a diverse range of microbial genera and species. The most frequently detected genera at the microbial genus level in the NTP group were Microbacterium spp. (6/16), Prevotella spp. (5/16), and Campylobacter spp. (5/16).DiscussionThese findings suggest that mNGS and GeneXpert MTB are useful diagnostic tools for identifying patients with tuberculous pleurisy, and mNGS can provide valuable insights into the microbial profiles of both tuberculous and non-tuberculous pleural effusions.

ObjectiveWe aimed to investigate the predictive value of the CT findings combined with serum potassium levels for primary aldosteronism (PA) subtype diagnosis, with a particular interest in sex differences.MethodsIn this retrospective study, we eventually included 482 PA patients who underwent successful adrenal venous sampling (AVS) and had available data. We diagnosed the subjects as having either unilateral (n = 289) or bilateral PA (n = 193) based on AVS. We analyzed the concordance rate between AVS and adrenal CT combined with serum potassium and performed a logistic regression analysis to assess the prevalence of unilateral PA on AVS.ResultsThe total diagnostic concordance rate between CT findings and AVS was 51.5% (248/482). The prevalence of hypokalemia in men and women was 47.96% (129/269) and 40.85% (87/213), respectively. The occurrence of unilateral lesions on CT and hypokalemia was significantly associated with an increased prevalence of unilateral PA [odds ratio (OR) 1.537; 95% confidence interval (CI) 1.364–1.731; p < 0.001]. In male participants, G2 (bilateral lesion on CT and normokalemia), G3 (unilateral lesion on CT and normokalemia), G4 (bilateral normal on CT and hypokalemia), G5 (bilateral lesion on CT and hypokalemia), and G6 (unilateral lesion on CT and hypokalemia) were significantly increased for the prevalence of unilateral PA on AVS (G2: OR 4.620, 95% CI 1.408–15.153; G3: OR 6.275, 95% CI 2.490–15.814; G4: OR 3.793, 95% CI 1.191–12.082; G5: OR 16.476, 95% CI 4.531–59.905; G6: OR 20.101, 95% CI 7.481–54.009; all p < 0.05), compared with G1 (patients with bilateral normal on CT and normokalemia). However, among female participants, we found an increased likelihood for unilateral PA in patients with unilateral lesions on CT and hypokalemia alone (OR 10.266, 95% CI 3.602–29.259, p < 0.001), while no associations were found in other groups (all p > 0.05). Recently, both ensemble clustering and semi-supervised clustering have emerged as important paradigms of traditional clustering. Ensemble clustering seeks to integrate multiple clustering results from different methods or the same methods with different parameters. Semi-supervised clustering involves using a small amount of class membership information in some samples for the learning process. Meanwhile, Semi-Supervised Ensemble Clustering (SSEC) has attracted increasing attention due to its high performance. However, most SSEC algorithms are configured based on partitional clustering techniques, and there are few attempts on hierarchical clustering techniques. Even in existing hierarchy-based SSEC algorithms, prior knowledge is not sufficiently used and is often applied to create primary partitions. To address these problems, we propose a Semi-supervised Hierarchical Ensemble Clustering framework based on a novel Similarity metric and stratified feature Sampling, which we call SHECSS. SHECSS uses the information of all primary partitions according to their strength to calculate the similarity between samples. Also, SHECSS is equipped with a stratified feature sampling mechanism that can improve the diversity of primary partitions and deal with high-dimensional data. Here, the primary partitions are created based on multiple hierarchical clustering techniques, and the target partition is configured by a consensus function based on the clusters clustering policy. Experimental results show the effectiveness and efficiency of SHECSS compared to representative clustering methods.

Abstract Backgrounds Adrenal venous sampling (AVS) represents the gold standard for classifying primary aldosteronism (PA). However, AVS is a technically demanding, expensive and invasive procedure. Computed tomography (CT) scans is recommended as the initial study of classification diagnosis by the current guidelines. In addition, postural stimulation test (PST) has been used to provide additional subtype diagnostic information. Objective This work aimed to evaluate the diagnostic utility of the adrenal CT combined with PST in the classification diagnosis of PA. Methods We analyzed PA patients who underwent AVS from November 2017 to February 2022 at a single center. Subtype classification of PA was determined by AVS. We analyzed the concordance rate between AVS outcomes, adrenal CT, and PST, and explored the value of adrenal CT combined with PST for predicting laterality of PA. Results Total 531 PA patients were included in the present study. The concordance rate between AVS and the adrenal CT was 51.0%(271/531). Receiver operating characteristic (ROC) curve of PST showed that the area under curve (AUC) was 0.604 [95% confidence interval (CI): 0.556, 0.652], the optimal cut-off value was 30%. The sensitivity, specificity, positive predictive value (PPV), negative predictive value (NPV), positive likelihood ratio (+LR), and negative likelihood ratio (−LR) of PST for diagnosis bilateral PA on AVS was 72.8, 46.2%, 0.48, 0.71, 1.35, and 0.59, respectively. The prevalence of unilateral PA on AVS in patients with unilateral lesion on CT and negative PST, unilateral lesion on CT and positive PST, bilateral normal or lesions on CT and negative PST, and bilateral normal or lesions on CT and positive PST was 82.4% (108/131), 59.9% (91/152), 50.7% (37/73), and 44.6% (78/175), respectively. The sensitivity, specificity, PPV, NPV, +LR, and -LR of adrenal CT combined with PST for the diagnosis of unilateral PA were 34.4, 89.4%, 0.82, 0.49, 3.25, and 0.73, respectively. Conclusions The combination of CT findings and PST can improve the accuracy of predicting laterality of PA.

ObjectiveThe lymph node posterior to the right recurrent laryngeal nerve (LN-prRLN) is a crucial component of the central lymph nodes (LNs). We aimed to evaluate multimodal predictive factors of LN-prRLN metastasis in patients with papillary thyroid carcinomas (PTCs), including the clinical data, pathologic data, and preoperative sonographic characteristics of PTCs.MethodsA total of 403 diagnosed PTC patients who underwent unilateral, sub-total, or total thyroidectomy with central neck dissection were enrolled in this retrospective study. The clinical data, pathologic data, conventional ultrasound (US) and contrast-enhanced ultrasound (CEUS) characteristics of PTCs were collected and evaluated for predicting LN-prRLN metastasis.ResultsIn this study, 96 PTC patients with LN-prRLN metastasis and 307 PTC patients without LN-prRLN metastasis were included. Univariate analysis demonstrated that PTC patients with LN-prRLN metastasis more often had younger age, larger size, multifocal cancers, A/T < 1, well-margins, microcalcification, petal-like calcification, internal vascularity, centripetal perfusion pattern and surrounding ring enhancement. Multivariate logistic regression analysis revealed that the CEUS centripetal perfusion pattern, central LN detected by ultrasound and LN-arRLN metastasis were independent characteristics for predicting LN-prRLN metastasis in PTC patients.ConclusionAccording to our research, it is essential for clinicians to thoroughly dissect central LNs, particularly LN-prRLNs.

Drug resistance substantially compromises antibiotic therapy and poses a serious threat to public health. Fusidic acid (FA) is commonly used to treat staphylococcal infections, such as pneumonia, osteomyelitis and skin infections. However, Gram-negative bacteria have natural resistance to FA, which is almost restrained in cell membranes due to the strong interactions between FA and phospholipids. Herein, we aim to utilize the strong FA–phospholipid interaction to pre-form a complex of FA with the exogenous phospholipid. The FA, in the form of an FA–phospholipid complex (FA-PC), no longer interacts with the endogenous membrane phospholipids and thus can be delivered into bacteria cells successfully. We found that the water solubility of FA (5 µg/mL) was improved to 133 µg/mL by forming the FA-PC (molar ratio 1:1). Furthermore, upon incubation for 6 h, the FA-PC (20 µg/mL) caused a 99.9% viability loss of E. coli and 99.1% loss of P. aeruginosa, while free FA did not work. The morphology of the elongated bacteria cells after treatment with the FA-PC was demonstrated by SEM. The successful intracellular delivery was shown by confocal laser scanning microscopy in the form of coumarin 6-PC (C6-PC), where C6 served as a fluorescent probe. Interestingly, the antibacterial effect of the FA-PC was significantly compromised by adding extra phospholipid in the medium, indicating that there may be a phospholipid-based transmembrane transport mechanism underlying the intracellular delivery of the FA-PC. This is the first report regarding FA-PC formation and its successful reversing of Gram-negative bacteria resistance to FA, and it provides a platform to reverse transmembrane delivery-related drug resistance. The ready availability of phospholipid and the simple preparation allow it to have great potential for clinical use.

Plaque biofilms play critical roles in the development of dental caries. Mechanical plaque control methods are considered to be most effective for plaque removal, such as brushing teeth or using flosser. Recently, water flosser has been paid much attention. Here, we tested the ability of a water flosser to remove the adhered sucrose and the dental plaque biofilms formed by Streptococcus mutans, Streptococcus sanguinis, and Actinobacillus viscosus. We found that the residual sucrose concentration was 3.54 mg/mL in the control group, 1.75 mg/mL in the syringe group (simulating the ordinary mouthwash), and 0 mg/mL in water flosser group. In addition, the residual bacterial concentration was 3.6 × 108 CFU/mL in the control group, 1.6 × 107 CFU/mL in the syringe group, and only 5.5 × 105 CFU/mL in the water flosser group. In summary, water flosser is effective for cleaning the teeth, which may have significant potential in preventing dental caries and maintaining oral health.

Sea buckthorn is a deciduous shrub with strong vitality, which belongs to the plant with homology of medicine and food. According to previous studies, sea buckthorn has a large amount of protein in its leaves and seeds, which belongs to high-quality plant protein resources. Peptides can be obtained from sea buckthorn protein or raw materials through a series of operations such as enzymatic hydrolysis, electrolysis, acid-base catalysis and so on. This paper summarizes the existing literature and data to compare the principles and characteristics of different extraction methods of sea buckthorn protein and polypeptide, analyzes the amino acid composition and molecular weight of protein and polypeptide, and introduces their hypoglycemic, antioxidant, sobering and anti-inflammatory activities. Although sea buckthorn protein and polypeptide have many kinds of activities, the application of compound method to prepare protein and polypeptide is less, and the research of structure and activity is not deep enough. It's necessary to strengthen the research from above aspects to promote the development and utilization of sea buckthorn protein resources.

A ring-shaped Mg–8.5Gd–4Y–1Zn–0.4Zr (wt%) alloy was manufactured via centrifugal casting and ring-rolling process. The effects of accumulative ring-rolling reduction amount on the microstructure, texture, and tensile properties of the alloy were investigated. The results indicate that the microstructure of centrifugal cast alloy consists of equiaxed grains and network-like eutectic structure present at grain boundaries. The ring-rolled alloy exhibits a characteristic bimodal microstructure composed of fine dynamic recrystallized (DRXed) grains with weak basal texture and coarse un-DRXed grains with strong basal texture, along with the presence of LPSO phase. With increasing amount of accumulative ring-rolling reduction, the coarse un-DRXed grains are refined via the formation of increasing amount of fine DRXed grains. Meanwhile, the dynamic precipitation of Mg5RE phase occurs, generating a dispersion strengthening effect. A superior combination of strength and ductility is achieved in the ring-rolled alloy after an accumulative rolling reduction of 80%. The tensile strength of this ring-rolled alloy after peak aging is further enhanced, reaching 511 MPa, while keeping a reasonable ductility. The salient strengthening mechanisms identified include the grain boundary strengthening of fine DRXed grains, dispersion strengthening of dynamic precipitated Mg5RE phase, short fiber strengthening of LPSO lamellae/rods, and precipitation strengthening of nano-sized prismatic β′ precipitates and basal γ′ precipitates.

Abstract Rheumatoid arthritis (RA) is a common, chronic, and highly disabling autoimmune disease characterized by difficult treatment, long disease duration, and easy recurrence. The development and application of high‐sensitivity theranostic probes for RA that will facilitate precise monitoring of disease progression and enable effective treatment are currently hotspots in the field of RA theranostics. In this study, mZMI@HA, a dual‐triggered theranostics nanoprobe, is constructed based on near‐infrared persistent luminescence nanoparticles (NIR‐PLNPs) for precise RA treatment and therapeutic evaluation. This is the first reported use of high‐sensitivity autofluorescence‐free imaging based on NIR‐PLNPs for precise RA treatment and therapeutic evaluation. Compared with the NIR fluorescence imaging probe‐indocyanine green, the signal‐to‐background ratio of persistent luminescence (PersL) imaging is improved nearly 14‐fold. Using PersL imaging to guide photothermal therapy and controllable drug release through NIR/pH‐responsiveness, the progress of collagen‐induced RA is relieved. Additionally, the therapeutic evaluation of RA by PersL imaging is consistent with clinical micro‐computed tomography and histological analyses. This study demonstrates the potential of NIR‐PLNPs for high‐sensitivity imaging‐guided RA treatment, providing a new strategy for RA precise theranostics.

Aeolian loess is a widespread deposit in mid-latitude arid and semiarid regions that has been extensively investigated in arid Central Asia and the Chinese Loess Plateau. The northeastern Tibetan Plateau is crucial to the release, transport, and accumulation of aeolian sediments because of its vast environmental differences, but relatively little research has been conducted on the loess here. To further deepen our understanding of the loess source in this region, we selected the Menyuan Basin as the study area for this loess tracing research. Through our investigation of the indicators and distribution characteristics of rare earth elements in the Yahecun (YHC) loess-paleosol profile of the Menyuan Basin, the following conclusions were drawn: 1) the indices and distribution curves of rare earth elements in the diverse strata of the YHC profile were not significantly distinct and were comparable to those of loess from other locations in northwestern China, revealing a general association with loess in arid northwestern China; 2) the influence of chemical weathering and particle diameter on rare earth elements in the loess of the study area was minimal, and Menyuan loess rare earth elements contained considerable information regarding their source; and 3) a comparison of the (La/Yb)N, δEu, (La/Sm)N, and (Gd/Yb)N parameters of potential source areas indicates that arid Central Asia, dominated by the Qaidam Desert, is the primary source area of the Menyuan loess, and dust particles released from these deserts are transported to the Menyuan Basin by the Westerlies and deposited to form loess. Additionally, the Badain Jaran Desert is an essential source of material for the Menyuan loess by winter winds, and some Tengger Desert materials are also transported to the Menyuan Basin by winter winds, but the material transported from the Tengger Desert has a negligible contribution.

Abstract Background The predictive role of blood-based tumor mutation burden (bTMB) for selecting advanced nonsmall cell lung cancer (NSCLC) patients who might benefit from immune checkpoint inhibitors (ICIs) is still under debate. Therefore, the purpose of this meta-analysis was to evaluate the efficacy of programmed cell death 1 (PD-1) /programmed cell death ligand 1 (PD-L1) inhibitors versus that of standard-of-care therapy in patients with NSCLC who were bTMB high and bTMB low. Methods PubMed, Embase, Cochrane, the Web of Science, and ClinicalTrials.gov were searched systematically from inception to February 2021 for studies of PD-1/PD-L1 inhibitors (durvalumab OR atezolizumab OR avelumab OR pembrolizumab OR Nivolumab) that provided hazard ratios (HRs) for overall survival (OS) or progression-free survival (PFS), or odds ratios (ORs) for objective response rate (ORR) in both bTMB high and bTMB low groups. Results A total of 2338 patients with advanced or metastatic NSCLC from six randomized controlled trials, which all used chemotherapy (CT) as a control, were included in this study. Abstract To minimise the profile of Satcom‐on‐the‐Move ground terminals, in this study, a two‐dimensional beam‐scanning continuous transverse stub antenna that is based on a compact linear source generator (LSG) is presented. A periodic anti‐symmetric two‐step (ASTS) structure cascaded with a broadband network is used to construct a single‐layer compact LSG. The concept of a parallel‐plate waveguide polarisation twister is used to design the ASTS unit that acts as a TE10‐to‐transverse electromagnetic wave converter, to which chamfers and T‐type grooves are added to improve the output plane wave performance over the entire frequency band. A prototype array operating in the band 10.95–12.75 GHz is designed, fabricated, and measured, and the experimental results agree well with the simulated ones. A beam‐scanning range of ±61° in elevation is achieved at the central frequency. Although only 29 of 36 slots (ϕ = 0°) contribute to the radiation, the antenna aperture efficiency is still over 60% due to the superior performance of the single‐layer minimised LSG.

Dynamic task allocation of unmanned aerial vehicle swarms for ground targets is an important part of unmanned aerial vehicle (UAV) swarms task planning and the key technology to improve autonomy. The realization of dynamic task allocation in UAV swarms for ground targets is very difficult because of the large uncertainty of swarms, the target and environment state, and the high real-time allocation requirements. Hence, dynamic task allocation of UAV swarms oriented to ground targets has become a key and difficult problem in the field of mission planning. In this work, a dynamic task allocation method for UAV swarms oriented to ground targets is comprehensively and systematically summarized from two aspects: the establishment of an allocation model and the solution of the allocation model. First, the basic concept and trigger scenario are introduced. Second, the research status and the advantages and disadvantages of the two allocation models are analyzed. Third, the research status and the advantages and disadvantages of several common dynamic task allocation algorithms, such as the algorithm based on market mechanisms, intelligent optimization algorithm, and clustering algorithm, are evaluated. Finally, the specific problems of the current UAV swarm dynamic task allocation method for ground targets are highlighted, and future research directions are established. This work offers important reference significance for fully understanding the current situation of UAV swarm dynamic task allocation technology.

PurposeTesticular yolk sac tumor (TYST) is a rare malignant germ cell tumor that mainly occurs in young men. Due to the low incidence of yolk sac tumors, there is a lack of prospective cohort studies with large samples. We aimed to develop a nomogram to predict cancer-specific survival (CSS) in patients with TYST.Materials and methodsPatient information was downloaded from the Surveillance, Epidemiology and End Results (SEER) database. We enrolled all patients with TYST from 2000 to 2018, and all patients were randomly divided into a training set and a validation set. Univariate and multivariate Cox proportional hazards regression models were used to identify independent risk factors for patients. We constructed a nomogram based on the multivariate Cox regression model to predict 1-, 3-, and 5-year CSS in patients with TYST. We used a series of validation methods to test the accuracy and reliability of the model, including the concordance index (C-index), calibration curve and the area under the receiver operating characteristic curve (AUC).Results619 patients with TYST were enrolled in the study. Univariate and multivariate Cox regression analysis showed that age, T stage, M stage and chemotherapy were independent risk factors for CSS. A nomogram was constructed to predict the patient's CSS. The C-index of the training set and the validation set were 0.901 (95%CI: 0.859–0.847) and 0.855 (95%CI: 0.865–0.845), respectively, indicating that the model had excellent discrimination. The AUC showed the same results. The calibration curve also indicated that the model had good accuracy.ConclusionsIn this study, we constructed the nomogram for the first time to predict the CSS of patients with TYST, which has good accuracy and reliability and can help doctors and patients make clinical decisions.

Neferine (Nef) might possess anti-depressive properties; however, its therapeutic effects are yet to be elucidated. Therefore, in this study, we aimed to explore the anti-depressant property of Nef using a mouse model of chronic stress-induced depression. Fifteen depression-prone mice were randomly selected and divided into three groups, namely, the model, Nef, and fluoxetine (Flu) groups. We observed that in tail suspension and forced swimming tests, the Nef and Flu treatments significantly decreased the immobility time of the depressed mice, and increased their sucrose preference indices. Moreover, both Nef and Flu treatments induced significant increases in the levels of anti-depressant neurotransmitters, including dopamine (DA), serotonin (5-HT), and norepinephrine (NE), and also reduced pathological damage to the hippocampus of the depressed mice. Incidentally, Illumina MiSeq sequencing analysis demonstrated that the relative abundance of Lactobacillus in the intestinal microbiota of depressed mice was restored after Nef/Flu treatment. Moreover, colonic Lactobacillus abundance was positively correlated with the levels of DA, 5-HT, and NE in the hippocampus of the mice. In conclusion, Nef improved monoamine neurotransmitter secretion and modulated the intestinal flora structure, particularly the abundance of Lactobacillus. Hence, it showed considerable anti-depressant potential, and might be a prospective anti-depressant therapeutic agent.

Sea buckthorn peel is the by-product of the sea buckthorn processing, which contains many bioactive compounds. In this paper, sea buckthorn high methoxyl pectin (SBHMP) was obtained, with a yield of 8% and a light-colored. The SBHMP was a high methoxyl with a degree of esterification of 57.75% and uronic acid content of 65.35%. The structural and morphological characterization of SBHMP were analyzed by high-performance liquid chromatography, Fourier-transform infrared spectroscopy, and scanning electron microscopy. Results showed that SBHMP presented a sheet and layered stacked morphological, and was mainly composed of galacturonic acid, arabinose, galactose, rhamnose, and mannose, which indicated that SBHMP mainly consisted of homogalacturonan (HG) and rhamnogalacturonan-I (RG-I) type pectin polysaccharides. In addition, SBHMP also presented significant gel, thickening, and emulsifying properties. The results exhibited that SBHMP could form jelly-like gels under acid and high sucrose conditions, presenting a shear-thinning behavior and increasing apparent viscosity with the enhancement of pectin and sucrose contents. Besides, SBHMP could form oil-in-water emulsions with pectin concentrations of 1.0–3.0%. When the SBHMP concentrations were 2.0 and 3.0%, the emulsions were stable during 7 days of storage. Findings in this paper demonstrated the potential of SBHMP to be a food thickener and emulsifier and support the in-depth utilization of sea buckthorn by-products.

Vascular dementia is a type of dementia from brain damage caused by cerebrovascular lesions and vascular risk factors. Prevotella histicola is a species of Prevotella, belonging to the category of obligate anaerobe. The purpose of our work was to study the protection of Prevotella histicola on cognitive function in rats subjected to vascular dementia (VaD) and investigate underlying molecular mechanisms. The rats were randomly divided into three groups: control group, 2VO group and 2VO + Prevotella histicola group. The VaD rats (the 2VO group and 2VO + Prevotella histicola group) were generated by bilateral common carotid artery occlusion (2VO). Rats in the 2VO+ Prevotella histicola group were administered with Prevotella histicola twice daily. In comparison with the rats in the 2VO group, rats in the 2VO + Prevotella histicola group presented an enhanced cognitive ability, increased synapse-associated protein expression, a downregulation of proinflammatory factors and an upregulation of neurotrophic factors. The relevant mechanism of the protective effect of Prevotella histicola may be associated with the inhibition of glial cell-associated inflammation by regulating phosphorylation of CaMKII. In conclusion, Prevotella histicola attenuates neurological impairments via regulating synapse-associated protein expression and the liberation of inflammatory elements in vascular dementia rats. The findings above might benefit the development of Prevotella histicola transplantation as a promising treatment of VaD.

Passion fruit is known to be sensitive to drought, and in order to study the physiological and biochemical changes that occur in passion fruit seedlings under drought stress, a hypertonic polyethylene glycol (PEG) solution (5%, 10%, 15%, and 20%) was used to simulate drought stress in passion fruit seedlings. We explored the physiological changes in passion fruit seedlings under drought stress induced by PEG to elucidate their response to drought stress and provide a theoretical basis for drought-resistant cultivation of passion fruit seedlings. The results show that drought stress induced by PEG had a significant effect on the growth and physiological indices of passion fruit. Drought stress significantly decreased fresh weight, chlorophyll content, and root vitality. Conversely, the contents of soluble protein (SP), proline (Pro), and malondialdehyde (MDA) increased gradually with the increasing PEG concentration and prolonged stress duration. After nine days, the SP, Pro and MDA contents were higher in passion fruit leaves and roots under 20% PEG treatments compared with the control. Additionally, with the increase in drought time, the activities of antioxidant enzymes such as peroxidase (POD), superoxide dismutase (SOD) and catalase (CAT) showed an increasing trend and then a decreasing trend, and they reached the highest value at the sixth day of drought stress. After rehydration, SP, Pro and MDA contents in the leaves and roots of passion fruit seedlings was reduced. Among all the stress treatments, 20% PEG had the most significant effect on passion fruit seedlings. Therefore, our study demonstrated sensitive concentrations of PEG to simulate drought stress on passion fruit and revealed the physiological adaptability of passion fruit to drought stress.

Abstract Despite remarkable breakthrough made by virtue of “polymerized small‐molecule acceptor (PSMA)” strategy recently, the limited selection pool of high‐performance polymer acceptors and long‐standing challenge in morphology control impede their further developments. Herein, three PSMAs of PYDT‐2F, PYDT‐3F, and PYDT‐4F are developed by introducing different fluorine atoms on the end groups and/or bithiophene spacers to fine‐tune their optoelectronic properties for high‐performance PSMAs. The PSMAs exhibit narrow bandgap and energy levels that match well with PM6 donor. The fluorination promotes the crystallization of the polymer chain for enhanced electron mobility, which is further improved by following n‐doping with benzyl viologen additive. Moreover, the miscibility is also improved by introducing more fluorine atoms, which promotes the intermixing with PM6 donor. Among them, PYDT‐3F exhibits well‐balanced high crystallinity and miscibility with PM6 donor; thus, the layer‐by‐layer processed PM6/PYDT‐3F film obtains an optimal nanofibril morphology with submicron length and ≈23 nm width of fibrils, facilitating the charge separation and transport. The resulting PM6/PYDT‐3F devices realizes a record high power conversion efficiency (PCE) of 17.41% and fill factor of 77.01%, higher than the PM6/PYDT‐2F (PCE = 16.25%) and PM6/PYDT‐4F (PCE = 16.77%) devices.

Sea buckthorn proanthocyanidins (SBP) are the most important antioxidant components of sea buckthorn, which are widely used in functional foods and cosmetics. Studies have shown that SBP have significant protective effects on macrophages against oxidative stress induced by hydrogen peroxide (H2O2). However, the mechanism remains uncertain. In the present study, we explored the effects of SBP on mitochondrial function and the mechanism of their protective effects against oxidative stress in cells. Our results showed that SBP could increase mitochondrial membrane potential, inhibit mPTP opening, reduce mitochondrial swelling, and enhance mitochondrial synthesis and metabolism. Thus, they alleviated oxidative damage and protected the cells against mitochondrial function. Western blot analysis showed that SBP had a protective effect on RAW264.7 cells by activating the AMPK-PGC1α-Nrf2 pathway. These results showed that SBP alleviated mitochondrial damage and dysfunction caused by oxidative stress. This study revealed the mechanism of SBP in reducing oxidative damage and provided a theoretical basis for further research on natural bioactive compounds to exert antioxidant activity and prevent arteriosclerosis and other diseases.

Introduction In the occurrence and development of heat stroke (HS), factors such as hyperthermia, ischemia and hypoxia are essential to the central nervous system (CNS) inflammatory response, but the main mechanism underlying CNS inflammation remains unclear. The aim of the study was to observe the polarization of microglia in response to heat-induced early nerve injury and to explore its possible mechanism of action. Material and methods To establish a heatstroke animal model in Beagle dogs, 18 Beagle dogs were divided into control (group A) and experimental groups (group B, group C and group D) according to a random numbers table. The animals in the experimental groups were placed on an electric blanket of an animal body temperature maintaining apparatus. The temperature was set at 40 ±0.5°C, and the rectal temperature was monitored every 5 min until the target body temperature was reached. Once the target temperature was reached, the dogs were transferred to an environment of 26 ±0.5°C and 60 ±0.5% humidity. Western blot analysis was used to detect the expression of microglia-specific markers CD45, iNOS, arginase, and CD206 in normal and heat-damaged brain tissues at different time points (1 h, 6 h, 24 h). The expression of CD45 and arginase was further determined by co-localization with immunofluorescence. Results CD45 and iNOS protein expression was detected in group A. The two protein markers in group B were significantly higher than those in group A (p < 0.05), and the protein markers in group C were still higher than those in group A (p 0.05). Arginase and CD206 protein expression was also detected in group A. Levels of the two protein markers in group B were higher than those in group A (p < 0.05), and the protein marker levels in group C were even higher than those in group A (p < 0.05). Further analysis of the two groups of protein markers in group D showed significantly higher levels than those in group A (p < 0.001). Immunofluorescence co-loca­lization of CD45 and arginase showed significantly increased fluorescence density at 6 h and 24 h after thermal injury (p < 0.001). Conclusions After heat-induced disease, microglia were found to be active in the brain tissues of dogs. The microglia activated in the early 1-6 h of CNS injury were mainly the M1 type, which were then converted to the M2 type after 6 h. The 24 h M2 type was dominant. The relationship between M1/M2 polarization trends and early brain injury in heat-induced disease may be a key to understanding CNS injury in heat-induced disease.

Insulin therapy plays an essential role in the treatment of diabetes mellitus. However, frequent injections required to effectively control the glycemic levels lead to substantial inconvenience and low patient compliance. In order to improve insulin delivery, many efforts have been made, such as developing the nanoparticles (NPs)-based release systems and oral insulin. Although some improvements have been achieved, the ultimate results are still unsatisfying and none of insulin-loaded NPs systems have been approved for clinical use so far. Recently, nano‒protein interactions and protein corona formation have drawn much attention due to their negative influence on the in vivo fate of NPs systems. As the other side of a coin, such interactions can also be used for constructing advanced drug delivery systems. Herein, we aim to provide an insight into the advance and flaws of various NPs-based insulin delivery systems. Particularly, an interesting discussion on nano‒protein interactions and its potentials for developing novel insulin delivery systems is initiated.

Worldwide, the demand for power load forecasting is increasing. A multi-step power-load forecasting model is established based on Informer, which takes the historical load data as the input to realize the prediction of the power load in the future. The constructed model abandons the common recurrent neural network to deal with time-series problems, and uses the seq2seq structure with sparse self-attention mechanism as the main body, supplemented by specific input and output modules to deal with the long-range relationship in the time series, and makes effective use of the parallel advantages of the self-attention mechanism, so as to improve the prediction accuracy and prediction efficiency. The model is trained, verified and tested by using the power-load dataset of the Taoyuan substation in Nanchang. Compared with RNN, LSTM and LSTM with the attention mechanism and other common models based on a cyclic neural network, the results show that the prediction accuracy and efficiency of the Informer-based power-load forecasting model in 1440 time steps have certain advantages over cyclic neural network models.

Abstract Background Approximately 10% of patients would develop symptomatic intracranial hemorrhage (sICH) after endovascular therapy. The aim of our study was to explore the ability of hypersensitive C-reactive protein-albumin ratio (HAR) in predicting sICH after endovascular therapy. Methods From April 2016 to December 2018, 334 consecutive patients with anterior circulation infarction undergoing endovascular therapy were enrolled in our study. sICH was defined using Heidelberg bleeding classification after endovascular therapy. Multiple regression analysis was used to investigate the potential risk factors of sICH after endovascular therapy. We used receiver operating characteristic curve analysis and nomogram analysis to assess the overall discriminative ability of the HAR in predicting sICH after endovascular therapy. Results Among these 334 patients enrolled, 37 (11.1%) patients with anterior circulation infarction were identified with sICH after endovascular therapy. Univariate logistic regression analysis demonstrated that patients with higher levels of HAR may be inclined to develop sICH (odds ratio, 10.994; 95% confidence interval, 4.567–26.463; P = 0.001). This association remained significant even after adjustment for potential confounders. Also, a cutoff value of 0.526× 10− 3 for HAR was detected in predicting sICH (area under curve, 0.763). Procyanidin is an important polyphenol for its health-promoting properties, however, the study of procyanidin in sea buckthorn was limited. In this paper, sea buckthorn procyanidin (SBP) was obtained through a green isolation and enrichment technique with an extraction rate and purity of 9.1% and 91.5%. The structure of SBP was analyzed using Ultraviolet–visible spectroscopy (UV–vis), Fourier-transform infrared spectroscopy (FT-IR), and liquid chromatography-mass spectrometry (LC-MS/MS). The results show that SBP is an oligomeric procyanidin, mainly composed of (−)-epicatechin gallate, procyanidin B, (+)-gallocatechin-(+)-catechin, and (+)-gallocatechin dimer. SBP showed superior scavenging capacity on free radicals. Furthermore, the cleaning rate of the ABTS radical was 4.8 times higher than vitamin C at the same concentration. Moreover, SBP combined with vitamin C presented potent synergistic antioxidants with combined index values below 0.3 with concentration rates from 5:5 to 2:8. SBP also provided significant protection against oxidative stress caused by hydrogen peroxide (H2O2) on RAW264.7 cells. These findings prove the potential of SBP as a natural antioxidant in food additives and support the in-depth development of sea buckthorn resources.

As society develops and aging populations increase, the incidence of arteriosclerosis, a seriously harmful cardiovascular disease (CVD) which mostly results from endothelial cellular oxidative damage, has continuously risen. Procyanidins from sea-buckthorn is a powerful antioxidant, although its protective effect on the cardiovascular system is not yet clearly understand. In this study, oxidative damaged HUVECs induced by palmitate acid (PA) were used as a model and the regulatory effect of procyanidins from sea-buckthorn (SBP) on HUVECs were investigated. The results showed SBP can be used for 12 h by HUVECs and had no detective cytotoxicity to them under 400 μg/L. Also, different concentrations of SBP can increase mitochondrial membrane potential and NO level and decrease LDH leakage in a dose-effect relationship, indicating SBP can improve oxidative damage. In addition, western blots and qPCR results showed SBP regulation on oxidative injured HUVECs is probably through p38MAPK/NF-κB signal pathway. This study revealed the molecular mechanism of procyanidins in decreasing endothelial oxidative damage, providing a theoretical foundation for further research on natural bioactive compounds to exert antioxidant activity in the body and prevent and improve cardiovascular diseases.
