2. Diffusion of Real-Time Information in Social-Physical Networks

Abstract

We study the diffusion behavior of real-time information. Typically, real-time information is valuable only for a limited time duration, and hence needs to be delivered before its "deadline." Therefore, real-time information is much easier to spread among a group of people with frequent interactions than between isolated individuals. With this insight, we consider a social network which consists of many cliques and information can spread quickly within a clique. Furthermore, information can also be shared through online social networks, such as Facebook, twitter, Youtube, etc. We characterize the diffusion of real-time information by studying the phase transition behaviors. Capitalizing on the theory of inhomogeneous random networks, we show that the social network has a critical threshold above which information epidemics are very likely to happen. We also theoretically quantify the fractional size of individuals that finally receive the message. Finally, the numerical results indicate that under certain conditions, the large size cliques in a social network could greatly facilitate the diffusion of real-time information., Comment: add one more figure

Abstract

We consider a cyber-physical system consisting of two interacting networks, i.e., a cyber-network overlaying a physical-network. It is envisioned that these systems are more vulnerable to attacks since node failures in one network may result in (due to the interdependence) failures in the other network, causing a cascade of failures that would potentially lead to the collapse of the entire infrastructure. The robustness of interdependent systems against this sort of catastrophic failure hinges heavily on the allocation of the (interconnecting) links that connect nodes in one network to nodes in the other network. In this paper, we characterize the optimum inter-link allocation strategy against random attacks in the case where the topology of each individual network is unknown. In particular, we analyze the "regular" allocation strategy that allots exactly the same number of bi-directional inter-network links to all nodes in the system. We show, both analytically and experimentally, that this strategy yields better performance (from a network resilience perspective) compared to all possible strategies, including strategies using random allocation, unidirectional inter-links, etc., Comment: 13 pages, 6 figures. 4. Conjoining Speeds up Information Diffusion in Overlaying Social-Physical Networks

Abstract

We study the diffusion of information in an overlaying social-physical network. Specifically, we consider the following set-up: There is a physical information network where information spreads amongst people through conventional communication media (e.g., face-to-face communication, phone calls), and conjoint to this physical network, there are online social networks where information spreads via web sites such as Facebook, Twitter, FriendFeed, YouTube, etc. We quantify the size and the critical threshold of information epidemics in this conjoint social-physical network by assuming that information diffuses according to the SIR epidemic model. One interesting finding is that even if there is no percolation in the individual networks, percolation (i.e., information epidemics) can take place in the conjoint social-physical network. We also show, both analytically and experimentally, that the fraction of individuals who receive an item of information (started from an arbitrary node) is significantly larger in the conjoint social-physical network case, as compared to the case where the networks are disjoint. These findings reveal that conjoining the physical network with online social networks can have a dramatic impact on the speed and scale of information diffusion., Comment: 14 pages, 4 figures

The protease inhibitor indinavir causes raises blood sugar levels without a compensatory increase in insulin secretion, according to a study of 12 HIV patients. The drug also caused insulin resistance. This could explain why some HIV patients who take the drug develop hyperglycemia and other metabolic abnormalities.

Abstract

Hepatitis B infection, a sexually active homosexual lifestyle, and injection drug use increase the risk of hepatitis G infection (HGV) in HIV patients. Hepatitis G virus is thought to be transmitted by the same routes as HIV. Researchers tested 157 HIV-infected patients for genetic evidence of HGV, and found that 22% were infected. Forty-nine percent of patients who tested positive for the hepatitis B surface antigen (HBsAg) were infected with HGV. Homosexuality was as likely to be associated with HGV infection as injection drug use, as both increase the risk of disease transmission.

Abstract

The viral load of HIV in the blood may be related to weight loss in HIV-associated wasting. Wasting is a complication of advanced HIV disease in which patients progressively lose body weight, muscle mass, and strength. Researchers examined 33 HIV-infected patients, and found that loss of weight was strongly associated with the concentration of HIV in the blood. Higher degrees of weight loss were also associated with lower CD4 white blood cell counts, indicative of more advanced HIV disease. Antiviral drug therapy to reduce viral load may be effective in treating HIV-associated wasting.

33. Tumour characteristics provide evidence for germline mismatch repair missense variant pathogenicity

Abstract

BackgroundPathogenic variants in mismatch repair (MMR) genes (MLH1, MSH2, MSH6and PMS2) increase risk for Lynch syndrome and related cancers. We quantified tumour characteristics to assess variant pathogenicity for germline MMR genes.MethodsAmong 4740 patients with cancer with microsatellite instability (MSI) and immunohistochemical (IHC) results, we tested MMR pathogenic variant association with MSI/IHC status, and estimated likelihood ratios which we used to compute a tumour characteristic likelihood ratio (TCLR) for each variant. Predictive performance of TCLRin combination with in silicopredictors, and a multifactorial variant prediction (MVP) model that included allele frequency, co-occurrence, co-segregation, and clinical and family history information was assessed.ResultsCompared with non-carriers, carriers of germline pathogenic/likely pathogenic (P/LP) variants were more likely to have abnormal MSI/IHC status (p<0.0001). Among 150 classified missense variants, 73.3% were accurately predicted with TCLRalone. Models leveraging in silicoscores as prior probabilities accurately classified >76.7% variants. Adding TCLRas quantitative evidence in an MVP model (MVP +TCLRPred) increased the proportion of accurately classified variants from 88.0% (MVP alone) to 98.0% and generated optimal performance statistics among all models tested. Importantly, MVP +TCLRPredresulted in the high yield of predicted classifications for missense variants of unknown significance (VUS); among 193 VUS, 62.7% were predicted as P/PL or benign/likely benign (B/LB) when assessed according to American College of Medical Genetics and Genomics/Association for Molecular Pathology guidelines.ConclusionOur study demonstrates that when used separately or in conjunction with other evidence, tumour characteristics provide evidence for germline MMR missense variant assessment, which may have important implications for genetic testing and clinical management.

Abstract

Background Reliable in vitro islet quality assessment assays that can be performed routinely, prospectively, and are able to predict clinical transplant outcomes are needed. In this paper we present data on the utility of an assay based on cellular oxygen consumption rate (OCR) in predicting clinical islet autotransplant (IAT) insulin independence (II). IAT is an attractive model for evaluating characterization assays regarding their utility in predicting II due to an absence of confounding factors such as immune rejection and immunosuppressant toxicity. Methods Membrane integrity staining (FDA/PI), OCR normalized to DNA (OCR/DNA), islet equivalent (IE) and OCR (viable IE) normalized to recipient body weight (IE dose and OCR dose), and OCR/DNA normalized to islet size index (ISI) were used to characterize autoislet preparations (n = 35). Correlation between pre-IAT islet product characteristics and II was determined using receiver operating characteristic analysis. Results Preparations that resulted in II had significantly higher OCR dose and IE dose (p<0.001). These islet characterization methods were highly correlated with II at 6–12 months post-IAT (area-under-the-curve (AUC) = 0.94 for IE dose and 0.96 for OCR dose). FDA/PI (AUC = 0.49) and OCR/DNA (AUC = 0.58) did not correlate with II. OCR/DNA/ISI may have some utility in predicting outcome (AUC = 0.72). Conclusions Commonly used assays to determine whether a clinical islet preparation is of high quality prior to transplantation are greatly lacking in sensitivity and specificity. While IE dose is highly predictive, it does not take into account islet cell quality.
