SQUAMOSA promoter-binding protein-like (SPL) transcription factors are very important for the plant growth and development. Here 15 RoSPLs were identified in Rubus occidentalis. The conserved domains and motifs, phylogenetic relationships, posttranscriptional regulation, and physiological function of the 92 SPL family genes in Fragaria vesca, Malus domestica, Prunus persica, R. occidentalis, and Pyrus pyrifolia were analyzed. Sequence alignment and phylogenetic analysis showed the SPL proteins had sequence conservation, some FvSPLs could be lost or developed, and there was a closer relationship between M. domestica and P. pyrifolia, F. vesca and R. occidentalis, respectively. Genes with similar motifs clustering together in the same group had their functional redundancy. Based on the function of SPLs in Arabidopsis thaliana, these SPLs could be involved in vegetative transition from juvenile to adult, morphological change in the reproductive phase, anthocyanin biosynthesis, and defense stress. Forty-eight SPLs had complementary sequences of miR156, of which nine PrpSPLs in P. persica and eight RoSPLs in R. occidentalis as the potential targets of miR156 were reported for the first time, suggesting the conservative regulatory effects of miR156 and indicating the roles of miR156-SPL modules in plant growth, development, and defense response. It provides a basic understanding of SPLs in Rosaceae plants.

The feeding preference of pear psylla (Cacopsylla chinensis) varies among pear cultivars. To clarify whether leaf structural factors are related to C. chinensis resistance, eleven pear cultivars of three pear species and their hybrids in China were studied. The population size of C. chinensis in plots of different cultivars was investigated, and leaf paraffin sections of each pear cultivar were observed. Correlation analysis results showed that the thickness of palisade tissue (PT) and the thickness ratio (TR) of PT to leaf (L) were the main factors influencing resistance to C. chinensis. The leaves of cultivar Wujiuxiang and those species from France and the USA exhibited the thickest PT and the highest TR; these three cultivars demonstrated much higher resistance to C. chinensis than did the other cultivars. The cultivar Gold Nijisseiki, with a large C. chinensis population size, showed low PT and TR. The findings of this study may provide valuable information to pear growers, experts and scientists regarding plant protection, cultivation and breeding when selecting suitable varieties to culture from the perspective of pest control.

Soil fungi play a very important role in the soil ecological environment. In agricultural production, long-term monoculture and continuous cropping lead to changes in fungal community diversity. However, the effects of long-term monoculture and continuous cropping on strawberry plant health and fungal community diversity have not been elucidated. In this study, using high-throughput sequencing (HTS), we compared the fungal community and diversity of strawberry rhizosphere soil after various durations of continuous cropping (0, 2, 4, 6, 8, 10 and 12 years). The results showed that soil fungal diversity increased with consecutive cropping years. Specifically, the soil-borne disease pathogens Fusarium and Guehomyces were significantly increased after strawberry continuous cropping, and the abundance of nematicidal (Arthrobotrys) fungi decreased from the fourth year of continuous cropping. The results of correlation analysis suggest that these three genera might be key fungi that contribute to the changes in soil properties that occur during continuous cropping. In addition, physicochemical property analysis showed that the soil nutrient content began to decline after the fourth year of continuous cropping. Spearman's correlation analysis showed that soil pH, available potassium (AK) and ammonium nitrogen (NH4+-N) were the most important edaphic factors leading to contrasting beneficial and pathogenic associations across consecutive strawberry cropping systems.

Long-term monoculture leads to continuous cropping (CC) problems, which complicate agricultural production, both locally and abroad. This study contrasted the different bacterial community compositions, physicochemical properties and enzyme activities of strawberry soil subjected to CC, CC rhizosphere (CCR), non-CC (NCC) and non-CC rhizosphere (NCCR) treatments. The soil physicochemical properties and enzyme activities were significantly reduced after long-term CC. In addition, five variation trends were observed for the 11 major bacterial genera in the soil. Sphingomonas was the only stable group among all treatments. The proportions of Novosphingobium, Rhodoplanes, Povalibacter, Cellvibrio and Stenotrophobacter decreased after CC. The relative abundances of Pelagibius, Thioprofundum and Allokutzneria increased only in the CC treatment. Nitrospira were more abundant in rhizosphere soil than in non-rhizosphere soil. The relative abundance of Bacillus increased after CC. Redundancy analysis revealed that Bacillus, Pelagibius and Allokutzneria had significant negative correlations with the soil physicochemical properties and enzyme activities. Therefore, these genera may be the key bacteria influenced by the physicochemical properties and enzyme activities altered by replanting. These results indicate that long-term CC of strawberry leads to less favourable rhizosphere soil conditions, which can be understood as a stress-induced response of the bacterial community diversity. Further research is needed to determine how the quality of soil is reduced by the shift in the diversity of the soil bacterial community.

A new nematode species of the genusPhasmarhabditiswas isolated from the body surface of a slug (Philomycus bilineatusBenson, PB). Morphological and molecular analyses confirmed this nematode as a new species. The nematode was namedPhasmarhabditis zhejiangensissp. nov. (Nematoda: Rhabditidae) and is dioecious. In males, the open bursa with genital papillae is characterized by the formula 1-1-1-2-1-3, and the spicule length is 58μm. In female, the vulva is located approximately in the middle of the body. The nematode belongs to papillosa group because of its tail shape pointed with filiform tip. The phasmids are rod-shaped. The posterior anus is slightly swollen.P.zhejiangensiswas further characterized by internal transcribed spacer (ITS), 18S rDNA and 28S rDNA sequences. After the sequencing results were compared with sequences available from the National Center for Biotechnology Information (NCBI), the maximum similarities of ITS, 18S and 28S sequences were 89.81%, 96.22% and 95.28%, respectively. Background In strawberry cultivation, continuous cropping (CC) obstacles seriously threaten production. A patented soil amendment (SA) can effectively relieve the CC obstacles to strawberry cultivation, but knowledge of the recovery mechanisms underlying this phenomenon is limited. Results In this study, transcriptomic profiling of strawberry roots in soil with and without the SA was conducted using RNA-Seq technology to reveal gene expression changes in response to SA treatment. In total, 188 differentially expressed genes (DEGs), including 144 upregulated and 44 downregulated DEGs, were identified. SA treatment resulted in genotype-dependent responses, and the response pattern, including an overall increase in the expression of nutrient transport genes and a decrease in the expression of defense response genes, may be a possible mechanism underlying recovery strategies in strawberry roots after the application of the SA to CC soil. We also found that 9 Hsp genes involved in plant defense pathways were all downregulated in the SA-treated roots. Conclusions This research indicated that strawberry plants reallocated defense resources to development when SA treatment alleviated the stress caused by a CC soil environment. The present study provides an opportunity to reveal the fundamental mechanisms of the tradeoff between growth and defense in strawberry.

Intercropping Persian walnut (Juglans regia L.) and wheat (Triticum aestivum L.) have been widely applied in the Xinjiang Uygur Autonomous Region in Northwest China as a means of reducing soil and water losses and improving both land-use efficiency and economic returns. To understand how changes in soil conditions and nematode community structure can contribute to the evaluation of wheat–walnut intercropping systems from the view of soil fauna, we studied the soil nematode community in wheat rhizosphere soil under both monoculture and intercropping systems for 2 years. The results showed that the pH and total nitrogen and organic matter contents in intercropping systems with walnut trees were decreased compared with those of system with wheat alone. The nematode communities differed significantly between intercropping and monoculture plots, e.g., Rhabditis and Dorylaimus were dominant only in monocultures, whereas Tylenchus was dominant only in intercropping systems. Moreover, intercropping systems resulted in decreased nematode abundance, increased proportions of plant-feeding nematodes, and decreased omnivores/predators, particularly in the second year (2012). The decrease in diversity indices (H′) and ecological indices (WI, EI, and SI) of the nematode communities indicated high disturbance and low soil fertility in intercropping systems. Overall, wheat intercropping with walnut had a significant negative effect on wheat rhizosphere soil conditions.

The nematodes of genus Oscheius are insect parasites with a potential role as biological control agents. The composition of gut microbiota and its potential assistant role in the complex pathogenic mechanism of nematodes have been poorly illustrated. In this study, the intestinal bacteria associated with dauer juveniles of the nematode Oscheius chongmingensis Tumian were classified by 16S rDNA high‐throughput sequencing. The raw reads were assigned to 845 operational taxonomic units (OTUs) after quality filtering. The results showed that the genus Ochrobactrum, with a proportion of 59.82%, was the most abundant genus, followed by 7.13% Bacillus, 4.7% Albidiferax, 4.26% Acinetobacter, and 3.09% Rhodococcus. The two dominant bacteria, Ochrobactrum and Bacillus, were further isolated by culturing on NBTA and LB medium respectively, and then identified as Ochrobactrum tritici and Bacillus cereus by morphological and 16S rDNA sequence analysis. Furthermore, the entomopathogenicity of these two bacterial species was studied. The results showed that O. tritici caused 93.33% mortality within 144 hr in the 4th‐instar larvae of Galleria mellonella treated with 2 × 109 CFU/ml, whereas B. cereus showed 100% mortality at a concentration of 3.3 × 107 CFU/ml within 48 hr. These findings, especially the presence of O. tritici, which had not been found in other nematode species in the genus Oscheius, indicate that the associated nematode O. chongmingensis may have particular utility as a biocontrol agent., The nematodes of genus Oscheius are insect parasites with a potential role as biological control agents. We identified and analyzed the gut bacterial community of the dauer juveniles of O. chongmingensis Tumian.

WRKY transcription factors play important roles in many plant developmental processes and adaptation to the environment. However, little knowledge is available about the WRKY gene family in cultivated strawberry (Fragaria × ananassa Duch.), an important soft fruit worldwide. In this study, a total of 47 WRKY gene members were identified and renamed on the basis of their order on the chromosomes. According to their evolutionary events and conserved structure, the 47 FaWRKYs were divided into three major groups with several subgroups. A cis-element analysis showed that all FaWRKYs possessed at least one stress response-related cis-element. Comprehensive analysis, including phylogenetic analysis and expression profiling, based on real-time qPCR analysis in root, stem, leaf and fruit was performed on group III FaWRKY genes. The phylogenetic tree of the WRKY III genes in cultivated strawberry, wild Strawberry, Arabidopsis, tomato, and rice was divided into five clades. Additionally, the expression profiles of the FaWRKY genes in response to continuous cropping were further investigated based on RNA-seq data. FaWRKY25, FaWRKY32, and FaWRKY45, which are group III FaWRKY genes, were upregulated after continuous cropping. The level of reactive oxygen species (ROS) and the expression levels of PR1 and peroxidase were higher in continuous cropping (CC) than in non-continuous cropping (NCC). The results indicated that group III FaWRKYs might play an important role in continuous cropping. These results provide a foundation for genetic improvements for continuous cropping tolerance in cultivated strawberry.

Soil ecology plays an important role in the growth and health of plants. Research suggests that long-term monocropping may lead to soil ecological disorders. In this study, we aimed to understand the reasons for the decrease in plant productivity stemming from long-term monoculture cropping. Greenhouse studies were conducted to determine the cause of continuous cropping (CC) obstacles in soils under 12 years of continuous strawberry (Fragaria × ananassa Duch.) production. The data suggested that CC led to three phases of changes in abiotic and biotic soil factors. In phase I (CC for 2–6 years), significant changes were observed only in soil physicochemical properties, such as the pH, total nitrogen (TN), ammonium nitrogen (NH4+-N), available phosphorus (AP), available potassium (AK) and soil organic matter (SOM), which obviously changed from the second year to the sixth year. In phase II (CC for 6–8 years), two biotic factors, key fungi such as Fusarium, Humicola and Arthrobotrys and soil nematodes, i.e., populations and communities of nematodes, started to change significantly in terms of their abundance. In phase III (CC for >8 years), the accumulation of phenolic acids, i.e., p-hydroxybenzoic acid (p-HBA), ferulic acid (FA), p-coumaric acid (p-CA) and cinnamic acid (CA), significantly inhibited crop growth, and the abundance of key bacteria, including Bacillus, Sphingomonas and Sphingopyxis, started to change significantly from the eighth CC year. The results in this study provide useful information for solving CC obstacles in strawberry production.

Soil nematode community in jujube ( Ziziphus jujuba Mill.) rhizosphere soil under monoculture and jujube/wheat ( Triticum aestivum Linn.) intercropping systems were investigated for three years in Hetian arid area, Xinjiang Uygur Autonomous Region, northwest of China. The results showed that the density of soil nematodes in jujube orchards became larger when wheat was planted between the lines of jujube trees, mainly due to the higher numbers of Rhabditis and Acrobeloides . The bacterivores of cp-1 and cp-2 ( c – p , colonizer–persister) guilds, and fungivores of cp-2 guilds were much more in jujube/wheat intercropping system than those in jujube monoculture system, while other nematode groups had no significant difference between the two ecosystems. Although Shannon–Weaver index ( H′ ), genus dominant index ( Ig ) and species richness ( SR ), showed no differences between the two systems, still lower maturity index ( MI ) and modified maturity index ( ∑MI ) with higher PPI / MI presented enriched but unstable soil food web in jujube/wheat intercropping system. Higher nematode channel ratio ( NCR ) and Wasilewska index ( WI ) in the intercropping system indicated that bacterial decomposition channel was dominant channel with higher level of soil total nitrogen content in both layers but lower organic matters in surface layer. We concluded that jujube/wheat intercropping system is good for young tree growing, but jujube monoculture system is preferred if cultivated land is sufficient.

The essential oil from the root tubers of Lindera aggregata was analyzed by GC-FID and GC-MS. Thirty-five compounds were identified and the main components of the essential oil were α-longifolene (15.13%), bornyl acetate (11.49%) followed by α-eudesmol (9.14%), α-pinene (7.88%), β-phellandrene (6.31%), β-terpinene (5.67%) and α-terpeninol (5.11%). During the screening program for new agrochemicals from Chinese medicinal herbs and wild plants, essential oil of L. aggregata was found to possess insecticidal activity against two grain storage insects, Sitophilus zeamais and Tribolium castaneum. The essential oil of L. aggregata possessed contact toxicity against S. zeamais and T. castaneum adults with LC50 values of 61.65 and 18.47 μg/adult, respectively. The oil also showed pronounced fumigant toxicity against S. zeamais and T. castaneum (LC50 = 23.04 and 14.69 mg/L air, respectively).

During our screening program for new agrochemicals from the local wild plants and Chinese medicinal herbs, the essential oil of Cyperus rotundus rhizomes was found to possess strong insecticidal activities against the booklice, Liposcelis bostrychophila Badonnel. Essential oil of C. rotundus rhizomes was obtained by hydrodistillation and analyzed by Gas chromatography-Mass spectrometry. A total of 35 components of the essential oil of C. rotundus were identified. The main components of the essential oil were α-cyperone (29.38%), cyperene (13.97%), caryophyllene oxide (6.71%) and β-selinene (6.47%). Bioactivities-directed chromatographic separation on repeated silica gel columns led to the isolation of two compounds: cyperene and α-cyperone. α-Cyperone exhibited pronounced contact toxicity against L. bostrychophila (LC50 = 41.32 μg/cm2) and was more toxic than cyperene (LC50= 50.08 μg/cm2) while the essential oil acting against the booklice showed an LC50 value of 102.11 μg/cm2. Purpose: To investigate the chemical composition and insecticidal activity of the essential oil of the aerial parts of Ainsliaea fragrans against maize weevils ( Sitophilus zeamais) . Methods : The essential oil of A. fragrans aerial parts was obtained by hydrodistillation and analyzed by gas chromatography (GC) and gas chromaotography-mass spectrometry (GC-MS). Contact toxicity of the essential oil and its major constituents was determined by topical application against S. zeamais. Results : A total of 30 components of the essential oil were identified. The major constituents were myristicin (41.3 %), elemicine (11.9 %), cis-isosafrole (11.5 %), borneol (9.1 %) and caryophyllene (8.8 %). The essential oil of A. fragrans exhibited contact toxicity against S. zeamais with LC 50 value of 50.7 μg/adult. Elemicine possessed the strongest contact toxicity (LC 50 = 13.5 μg/adult) while cis-isosafrole, myristicin, caryophyllene and borneol had LC 50 values of 31.2 μg/adult, 43.4 μg/adult, 57.9 μg/adult, and 98.4 μg/adult, respectively. Conclusion: The study indicates that the essential oil of A. fragrans aerial parts and its major constituents have a potential for development into natural insecticides for the control of grain storage insects. Water-distilled essential oil from Clinopodium chinense (Labiatae) aerial parts at the flowering stage was analyzed by gas chromatography-mass spectrometry. Thirty-five compounds, accounting for 99.18% of the total oil, were identified, and the main components of the essential oil of C. chinense were spathulenol (18.54%), piperitone (18.9%), caryophyllene (12.04%), and bornyl acetate (8.14%). Based on bioactivity-directed fractionation, bornyl acetate, caryophyllene, and piperitone were identified from the essential oil. The essential oil possessed fumigant toxicity against booklice (Liposcelis bostrychophila) with a 50% lethal concentration (LC50) value of 423.39 μg/liter, while the isolated constituents, bornyl acetate and piperitone, had LC50 values of 351.69 and 311.12 μg/liter against booklice, respectively. The essential oil also exhibited contact toxicity against L. bostrychophila with an LC50 value of 215.25 μg/cm(2). Bornyl acetate, caryophyllene, and piperitone exhibited acute toxicity against booklice with LC50 values of 321.42, 275.00, and 139.74 μg/cm(2), respectively. The results indicated that the essential oil and its isolated constituents have potential for development into natural insecticides or fumigants for control of insects in stored grains.

Similar to other pear psylla species in Europe and America, Cacopsylla chinensis (Yang and Li) is one of the most important pests that causes yield loss in commercial pear orchards in China. To investigate effective essential oils as alternatives to conventional pesticides against C. chinensis, 26 essential oils derived from commonly used Chinese spices and medicinal herbs were screened for insecticidal activity. Among these, the essential oil from Perilla frutescens (L.) Britton leaves was the top performer; it exhibited strong and acute toxicity against pear psylla, with an LD50 value of 0.63 μg per adult. Then, we tested the constituents of the essential oil and its toxicity in the field. Field trials showed a 72% corrected reduction in the first-second-instar population 7 d after spraying P. frutescens leaf oil solution at a concentration of 1 mg/ml and a 47% corrected reduction at days 3 and 14. This report is the first to document the application of essential oil from P. frutescens leaves to control C. chinensis under field conditions. Our results suggest that P. frutescens oil can be considered a novel potential pesticide for C. chinensis control in pear orchards.

The essential oil of Stachys riederi var. japonica (Family: Lamiaceae) was extracted by hydrodistillation and determined by GC and GC-MS. A total of 40 components were identified, representing 96.01% of the total oil composition. The major compounds in the essential oil were acetanisole (15.43%), anisole (9.43%), 1,8-cineole (8.07%), geraniol (7.89%), eugenol (4.54%), caryophyllene oxide (4.47%), caryophyllene (4.21%) and linalool (4.07%). Five active constituents (acetanisole, anisole, 1,8-cineole, eugenol and geraniol) were identified by bioactivity-directed fractionation. The essential oil possessed fumigant toxicity against maize weevils (Sitophilus zeamais) and booklice (Liposcelis bostrychophila), with LC50 values of 15.0 mg/L and 0.7 mg/L, respectively. Eugenol and anisole exhibited stronger fumigant toxicity than the oil against booklice. 1,8-Cineole showed stronger toxicity, and anisole as well as eugenol exhibited the same level of fumigant toxicity as the essential oil against maize weevils. The essential oil also exhibited contact toxicity against S. zeamais adults and L. bostrychophila, with LC50 values of 21.8 µg/adult and 287.0 µg/cm2, respectively. The results indicated that the essential oil of S. riederi var. japonica and its isolates show potential as fumigants, and for their contact toxicity against grain storage insects.

Pear psyllids are among the most damaging pests in pear orchards, but little knowledge exists of psyllid species in cultivated pear orchards in China. In this study, DNA sequence analyses of the 16S rDNA and cytochrome oxidase I (COI) DNA regions were performed to identify pear psyllids from 28 regions of 20 provinces in China and to classify their genetic relationships to understand the origin of the species. The results showed that Cacopsylla chinensis Yang et Li (Hemiptera: Psyllidae) was found in most pear orchards in China, but Cacopsylla qianli (Hemiptera: Psyllidae) was found in only the cities of Guiyang (Guizhou province) and Xiangyang (Hubei province). The results for the 16S rDNA and COI regions were similar. Based on the nucleotide sequences and phylogenetic analyses of 16S rDNA and COI, C. chinensis could be divided into three groups: lineages I, II, and III. Based on 16S rDNA and COI, lineage II showed approximately 4% and 3% difference from lineage I, and lineage III showed approximately 12% and 9% difference from lineage I, respectively. C. chinensis lineage I was found in most provinces of China, while C. chinensis lineage II samples were mainly found in the Bohai rim region of China, and lineage III samples were found in Northeast China. The results of this study will provide information to pear producers regarding effective control measures to prevent further damage from pear psyllids.

Cacopsylla chinensis (Yang and Li) (Hemiptera: Psyllidae) is an important pest of pear in China. As an alternative to conventional chemical pesticides, botanicals including essential oils and their constituents could provide an eco-friendly and nonhazardous control method. In this study, the essential oil of clove buds (Syzygium aromaticum) was obtained by hydrodistillation. Five constituents, accounting for 99.89% of the oil, were identified by gas chromatography-mass spectrometry, and the major constituents were eugenol (88.61%) and eugenol acetate (8.89%), followed by β-caryophyllene (1.89%). In a laboratory bioassay, clove essential oil, commercial eugenol (99.00%) and β-caryophyllene (98.00%) exhibited strong contact toxicity against the summerform adults of C. chinensis with LD50 values of 0.730, 0.673, and 0.708 µg/adult, and against the nymphs with LD50 values of 1.795, 1.668, and 1.770 µg/nymph, respectively. In contrast, commercial eugenol acetate (98%) had LD50 values of 9.266 µg/adult and 9.942 µg/nymph. In a field trial, clove essential oil caused significant population reductions of 73.01% (4.80 mg/ml), 66.18% (2.40 mg/ml) and 46.56% (1.20 mg/ml), respectively. Our results demonstrated that clove essential oil and its constituents have potential as a source of natural insecticides.

The aim of this research was to evaluate contact toxicity and repellency of the essential oil of Pogostemon cablin (Blanco) Bentham leaves against German cockroaches (Blattella germanica) (L.) and to isolate any active constituents. Essential oil of P. cablin leaves was obtained by hydrodistillation and analyzed by gas chromatography (GC) and gas chromatography-mass spectrometry (GC-MS). Twenty-three components were identified in the essential oil, and the main constituents were patchoulol (41.31%), pogostone (18.06%), α-bulnesene (6.56%), caryophyllene (5.96%), and seychellene (4.32%). Bioactivity-directed chromatographic separation of the essential oil led to the isolation of pogostone, patchoulol, and caryophyllene as active compounds. The essential oil of P. cablin leaves exhibited acute toxicity against male B. germanica adults with an LC50 value of 23.45 μg per adult. The constituent compound, pogostone (LC50 = 8.51 μg per adult) showed stronger acute toxicity than patchoulol (LC50 = 207.62 μg per adult) and caryophyllene (LC50 = 339.90 μg per adult) against the male German cockroaches. The essential oil of P. cablin leaves and the three isolated constituents exhibited strong repellent activity against German cockroaches at a concentration of 5 ppm. The results indicated that the essential oil of P. cablin leaves and its major constituents have good potential as a source for natural insecticides and repellents.

Water-distilled essential oil from the dried bulbs of Allium chinense (Liliaceae) was analyzed by gas chromatography–mass spectrometry (GC–MS). Eighteen compounds, accounting for 98.4% of the total oil, were identified and the main components of the essential oil of A. chinense were methyl allyl trisulfide (30.7%), dimethyl trisulfide (24.1%), methyl propyl disulfide (12.8%) and dimethyl disulfide (9.6%) followed by methyl allyl disulfide (3.4%) and methyl propyl trisulfide (3.6%). The essential oil exhibited contact toxicity against the booklice ( Liposcelis bostrychophila ) with an LC 50 value of 441.8 μg/cm 2 while the two major constituents, dimethyl trisulfide and methyl propyl disulfide had LC 50 values of 153.0 μg/cm 2 and 738.0 μg/cm 2 against the booklice, respectively. The essential oil of A. chinense possessed strong fumigant toxicity against the booklice with an LC 50 value of 186.5 μg/l while methyl allyl trisulfide (LC 50 = 90.4 μg/l) and dimethyl trisulfide (LC 50 = 114.2 μg/l) exhibited stronger fumigant toxicity than methyl propyl disulfide (LC 50 = 243.4 μg/l) and dimethyl disulfide (LC 50 = 340.8 μg/l) against the booklice. The results indicated that the essential oil and its major constituents have potential for development into natural insecticides or fumigants for control of insects in stored grains.

Melanization reaction, resulting from the activation of prophenoloxidase, is a vital immune response in insects for encapsulating and killing the invasive organisms. Prophenoloxidase needs to be proteolytically activated by its upstream prophenoloxidase-activating protease (PAP) in melanization. Identification and characterization of PAPs facilitates the understanding of the molecular mechanisms involved in insect immunity. We here cloned a full-length cDNA for a serine protease, named as SP105, from Asian corn borer, Ostrinia furnacalis (Guenée). The open reading frame of SP105 encodes 424-amino acid residue protein with a 19-residue signal peptide. Sequence comparison indicates that SP105 is most similar to Manduca sexta PAP3, a defined prophenoloxidase-activating protease. qRT-PCR analysis showed that SP105 mRNA levels increased significantly after a bacterial injection. Recombinant SP105 directly cleaved and activated Asian corn borer prophenoloxidase and therefore acted as the prophenoloxidase-activating protease. Additionally, SP105 formed SDS-stable complexes with a serine protease inhibitor, serpin-3, and its activity in activating prophenoloxidase was efficiently inhibited by serpin-3. Our work thus illustrated a prophenoloxidase-activating protease and revealed its regulation by serpin-3. The results would allow further advances in the understanding of the melanization in Asian corn borer and other insects.

The screening of Chinese medicinal herbs for insecticidal principles showed that the essential oil of Echinops grijsii Hance roots possessed significant larvicidal activity against mosquitoes. The essential oil was extracted via hydrodistillation and its constituents were determined by gas chromatography-mass spectrometry (GC-MS) analysis. GC-MS analyses revealed the presence of 31 components, with 5-(3-buten-1-yn-1-yl)-2,2'-bithiophene (5-BBT, 27.63%), αterthienyl (α-T, 14.95%),1,8-cineole (5.56%) and cis-β-ocimene (5.01%) being the four major constituents. Based bioactivity-directed chromatographic separation of the essential oil led to the isolation of 5-BBT, 5-(4-isovaleroyloxybut-1-ynyl)-2,2'-bithiophene (5-IBT) and αT as active compounds. The essential oil of E. grijsii exhibited larvicidal activity against the fourth instar larvae of Aedes albopictus, Anopheles sinensis and Culex pipiens pallens with LC50 values of 2.65 μg/mL, 3.43 μg/mL and 1.47 μg/mL, respectively. The isolated thiophenes, 5-BBT and 5-IBT, possessed strong larvicidal activity against the fourth instar larvae of Ae. albopictus(LC50 = 0.34 μg/mL and 0.45 μg/mL, respectively) and An. sinensis(LC50 = 1.36 μg/mL and 5.36 μg/mL, respectively). The two isolated thiophenes also had LC50 values against the fourth instar larvae of C. pipiens pallens of 0.12 μg/mL and 0.33 μg/mL, respectively. The findings indicated that the essential oil of E. grijsii roots and the isolated thiophenes have an excellent potential for use in the control of Ae.albopictus, An. sinensis and C. pipiens pallens larvae and could be used in the search for new, safer and more effective natural compounds as larvicides.

The aim of this research was to determine chemical composition and repellent and insecticidal activities of the essential oil of Kaempferia galanga L. rhizomes against the booklouse, Liposcelis bostrychophila Badonnel, and to isolate insecticidal or repellent constituents from the oil. The essential oil was obtained by hydrodistillation and analyzed by gas chromatography-mass spectrometry. Twenty-eight components of the oil were identified. The major compounds in the oil were ethyl-rho-methoxycinnamate (38.6%), ethyl cinnamate (23.2%), 1,8-cineole (11.5%), trans-cinnamaldehyde (5.3%), and borneol (5.2%). Based on bioactivity-guided fractionation, four active constituents were isolated from the oil and identified as 1,8-cineole, ethyl cinnamate, ethyl rho-methoxycinnamate, and trans-cinnamaldehyde. The essential oil exhibited contact toxicity against the booklouse with an LC50 value of 68.6 microg/cm2. Ethyl cinnamate (LC50 = 21.4 microg/cm2) exhibited stronger contact toxicity than ethyl rho-methoxycinnamate and trans-cinnamaldehyde (LC50 = 44.6 and 43.4 microg/cm2, respectively) while 1,8-cineole showed weak acute toxicity. The essential oil also possessed fumigant toxicity against the booklouse with a LC50 value of 1.5 mg/liter air. 1,8-Cineole and trans-cinnamaldehyde (LC50 = 1.1 and 1.3 mg/liter, respectively) possessed stronger fumigant toxicity against the booklouse than ethyl cinnamate and ethyl rho-methoxycinnamate (LC50 = 10.2 and 10.2 mg/liter air, respectively). trans-Cinnamaldehyde was strongly repellent to booklice, whereas ethyl cinnamate and ethyl rho-methoxycinnamate were weakly repellent and 1,8-cineole did not repel booklice. The results indicate that the essential oil and its constituent compounds have potential for development into natural insecticides or fumigants and repellents for control of insects in stored grains.

In our screening program for insecticidal activity of the essential oils/extracts derived from some Chinese medicinal herbs and spices, garlic (Allium sativum L.) essential oil was found to possess strong insecticidal activity against overwintering adults of Cacopsylla chinensis Yang et Li (Hemiptera: Psyllidae). The commercial essential oil of A. sativum was analyzed by gas chromatography-mass spectrometry. Sixteen compounds, accounting for 97.44% of the total oil, were identified, and the main components of the essential oil of A. sativum were diallyl trisulfide (50.43%), diallyl disulfide (25.30%), diallyl sulfide (6.25%), diallyl tetrasulfide (4.03%), 1,2-dithiolane (3.12%), allyl methyl disulfide (3.07%), 1,3-dithiane (2.12%), and allyl methyl trisulfide (2.08%). The essential oil of A. sativum possessed contact toxicity against overwintering C. chinensis, with an LC50 value of 1.42 microg per adult. The two main constituent compounds, diallyl trisulfide and diallyl disulfide, exhibited strong acute toxicity against the overwintering C. chinensis, with LC50 values of 0.64 and 11.04 /g per adult, respectively.

Water-distilled essential oil from the aerial parts of Dipsacus japonicus Miq. (Dipsacaceae) at the flowering stage was analysed by gas chromatography-mass spectrometry (GC-MS). Forty-six compounds, accounting for 96.76% of the total oil, were identified and the main compounds of the essential oil were linalool (11.78%), trans-geraniol (8.58%), 1,8-cineole (7.91%), β-caryophyllene (5.58%), α-terpineol (5.32%), β-selinene (5.15%), and spathulenol (5.04%). The essential oil of D. japonicus possessed contact toxicity against two grain storage insects, Sitophilus zeamais and Tribolium castaneum adults, with LD50 values of 18.32 μg/ adult and 13.45 μg/adult, respectively. The essential oil of D. japonicus also exhibited pronounced fumigant toxicity against S. zeamais (LC50 = 10.11 mg/l air) and T. castaneum adults (LC50 = 5.26 mg/l air). Of the three major compounds, 1,8-cineole exhibited stronger fumigant toxicity than the crude essential oil against S. zeamais and T. castaneum adults with LC50 values of 2.96 mg/l air and 4.86 mg/l air, respectively

Water-distilled essential oil from Phlomis umbrosa Turcz. (Lamiaceae) flowering aerial parts was analyzed by gas chromatography-mass spectrometry (GC-MS). Forty-one compounds, accounting for 97.0 % of the oil, were identified and the main components of the essential oil of P. umbrosa flowering aerial parts were geranial (16.5 %), linalool (13.3 %), cis-geraniol (7.4 %), β-caryophyllene (6.3 %), β-myrcene (6.2 %), 1,8-cineol (5.3 %), 4-terpineol (5.1 %) and myristicin (5.0 %). The essential oil of P. umbrosa possessed contact and fumigant toxicity against two grain storage insects, Sitophilus zeamais and Tribolium castaneum adults (for contact toxicity LD values were 23.76 µg/adult and 10.52 µg/adult; for fumigant toxicity LC50 values were 8.15 mg/L and 5.62 mg/L air against S. zeamais and T. castaneum, respectively).

During a screening program for new agrochemicals from Chinese medicinal herbs, the ethanol extract of Notopterygium incisum rhizomes was found to possess strong nematicidal activity against the two species of nematodes, Bursaphelenchus xylophilus and Meloidogyne incognita. Based on bioactivity-guided fractionation, the four constituents were isolated from the ethanol extract and identified as columbianetin, falcarindiol, falcarinol, and isoimperatorin. Among the four isolated constituents, two acetylenic compounds, falcarindiol and falcarinol (2.20-12.60 μg/mL and 1.06-4.96 μg/mL, respectively) exhibited stronger nematicidal activity than two furanocoumarins, columbianetin, and isoimperatorin (21.83-103.44 μg/mL and 17.21-30.91 μg/mL, respectively) against the two species of nematodes, B. xylophilus and M. incognita. The four isolated constituents also displayed phototoxic activity against the nematodes. The results indicate that the ethanol extract of N. incisum and its four isolated constituents have potential for development into natural nematicides for control of plant-parasitic nematodes.
