Abstract Objective Obesity is a common feature in women with polycystic ovary syndrome (PCOS) and potentially significantly influences reproductive function. However, opinions are divided as to which factor is a more appropriate obesity predictor of reproductive outcomes. The aim of this study was to investigate the discriminatory capability of anthropometric measures in predicting reproductive outcomes in Chinese women with PCOS. Methods A total of 998 women with PCOS from PCOSAct were included. Logistic regression models were used to compute the odds ratios (ORs) and 95% confidence interval (95% CIs) to assess the effect of anthropometric measures, including body mass index (BMI), waist circumference (WC), hip circumference (HC), the waist‒hip ratio (WHR) and the waist‒height ratio (WHtR), on reproductive outcomes. The discrimination abilities of the models were assessed and compared based on the area under the receiver operating characteristic curve (AUC), Akaike’s information criterion (AIC) and integrated discrimination improvement (IDI). Results Among PCOS women, there was a graded association between anthropometric measures and predicted reproductive outcomes across quintiles of anthropometric measures, including a linear association among WHR, BMI and reproductive outcomes and among waist circumference, WHtR and live birth, pregnancy, and ovulation. However, only a linear association was noted between the hip and ovulation. C-statistic comparisons and IDI analyses revealed a trend towards a significant superiority of BMI for ovulation and WHR for live birth, pregnancy and conception in the models. Combining obesity variables improved discrimination in the multivariable models for reproductive outcomes. Conclusions Our findings support that BMI is a better predictor of ovulation and that the WHR is a better predictor of live birth, pregnancy and conception, whereas the combination of obesity variables contributes to the discrimination of reproduction.

High purity and ultrafine DAAF (u-DAAF) is an emerging insensitive charge in initiators. Although there are many ways to obtain u-DAAF, developing a preparation method with stable operation, accurate control, good quality consistency, equipment miniaturization, and minimum manpower is an inevitable requirement to adapt to the current social technology development trend. Here reported is the microfluidic preparation of u-DAAF with tunable particle size by a passive swirling microreactor. Under the guidance of recrystallization growth kinetics and mixing behavior of fluids in the swirling microreactor, the key parameters (liquid flow rate, explosive concentration and crystallization temperature) were screened and optimized through screening experiments. Under the condition that no surfactant is added and only experimental parameters are controlled, the particle size of recrystallized DAAF can be adjusted from 98 nm to 785 nm, and the corresponding specific surface area is 8.45 m2·g−1 to 1.33 m2·g−1. In addition, the preparation method has good batch stability, high yield (90.8%–92.6%) and high purity (99.0%–99.4%), indicating a high practical application potential. Electric explosion derived flyer initiation tests demonstrate that the u-DAAF shows an initiation sensitivity much lower than that of the raw DAAF, and comparable to that of the refined DAAF by conventional spraying crystallization method. This study provides an efficient method to fabricate u-DAAF with narrow particle size distribution and high reproducibility as well as a theoretical reference for fabrication of other ultrafine explosives.

Abstract Nonalcoholic fatty liver disease (NAFLD) is a group of chronic liver disease which ranges from simple steatosis (NAFL) to non-alcoholic steatohepatitis (NASH) and is characterized by lipid accumulation, inflammation activation, fibrosis, and cell death. To date, a number of preclinical studies or clinical trials associated with therapies targeting fatty acid metabolism, inflammatory factors and liver fibrosis are performed to develop effective drugs for NAFLD/NASH. However, few therapies are cell death signaling-targeted even though the various cell death modes are present throughout the progression of NAFLD/NASH. Here we summarize the four types of cell death including apoptosis, necroptosis, pyroptosis, and ferroptosis in the NAFLD and the underlying molecular mechanisms by which the pathogenic factors such as free fatty acid and LPS induce cell death in the pathogenesis of NAFLD. In addition, we also review the effects of cell death-targeted therapies on NAFLD. In summary, our review provides comprehensive insight into the roles of various cell death modes in the progression of NAFLD, which we hope will open new avenues for therapeutic intervention.

In recent years, the extensive use of plastic packaging has caused serious environmental pollution problems. With the increase in people's environmental awareness, the development of new biodegradable food packaging materials to replace traditional petroleum-based plastics has become a hot research topic. Cellulose nanofibrils (CNF) are widely favored by researchers due to its excellent properties such as high specific surface area, elastic modulus, strength, and gas barrier properties. This paper summarizes the preparation methods of CNF and CNF composite films, discusses the mechanical property, barrier property, antibacterial property, degradable property, and responsive property as food packaging materials. Finally, the challenges faced by CNF in the field of food packaging are proposed, aiming to provide a reference for the future production of CNF food packaging materials to replace traditional petroleum-based materials.

Background: Immunotherapy has emerged as a novel strategy for cancer treatment following surgery, radiotherapy, and chemotherapy. Immune checkpoint blockade and Chimeric antigen receptor (CAR)-T cell therapies have been successful in clinical trials. Cancer cells evade immune surveillance by hijacking inhibitory pathways via overexpression of checkpoint genes. The Cluster of Differentiation 47 (CD47) has emerged as a crucial checkpoint for cancer immunotherapy by working as a “don’t eat me” signal and suppressing innate immune signaling. Furthermore, CD47 is highly expressed in many cancer types to protect cancer cells from phagocytosis via binding to SIRPα on phagocytes. Targeting CD47 by either interrupting the CD47-SIRPα axis or combing with other therapies has been demonstrated as an encouraging therapeutic strategy in cancer immunotherapy. Antibodies and small molecules that target CD47 have been explored in pre- and clinical trials. However, formidable challenges such as the anemia and palate aggregation cannot be avoided because of the wide presentation of CD47 on erythrocytes. Aim of view: This review summarizes the current knowledge on the regulation and function of CD47, and provides a new perspective for immunotherapy targeting CD47. It also highlights the clinical progress of targeting CD47 and discusses challenges and potential strategies. Key scientific concepts of review: This review provides a comprehensive understanding of targeting CD47 in cancer immunotherapy, it also augments the concept of combination immunotherapy strategies by employing both innate and adaptive immune responses.

Abstract Heart failure (HF) is a significant global public health concern with a high readmission rate, posing a serious threat to the health of the elderly population. While several studies have used machine learning (ML) to develop all-cause readmission risk prediction models for elderly patients with HF, few have integrated ML-selected features with those chosen by human experts to assess HF patients readmission. A retrospective analysis of 8396 elderly HF patients hospitalized at the Affiliated Hospital of North Sichuan Medical College from January 1, 2018 to December 31, 2021 was conducted. Variables selected by XGBoost, LASSO regression, and random forest constituted the machine group, while the human expert group comprised variables chosen by two experienced cardiovascular professors. The variables selected by both groups were combined to form a human–machine collaboration group. Model performance was evaluated using the area under the receiver operating characteristic curve (AUC). The SHapley Additive exPlanations (SHAP) method was used to elucidate the importance of each predictive feature, explain the impact of individual features on the model, and provide visual representation. A total of 73 features were included for model development. The human–machine collaboration model, utilizing CatBoost, achieved an AUC of 0.83617, an F1-score of 0.73521, and a Brier score of 0.16536 on the validation set. This model demonstrated superior predictive performance compared to those created solely by human experts or machine. The SHAP plot was then used to visually display the feature analysis of the human–machine collaboration model, revealing HGB, NT-proBNP, smoking history, NYHA classification, and LVEF as the 5 most important features. This study indicate that the human–machine collaboration model outperforms those relying solely on human expert selection or machine algorithm at predicting all-cause readmission in elderly HF patients. The application of the SHAP method enhanced the interpretability of the model outcomes, aiding clinicians in accurately pinpointing risk factors associated with HF readmission. This advancement enables the formulation of tailored treatment strategies, offering a more personalized approach to patient care.

Abstract Multiple sclerosis (MS) is a chronic autoimmune disease of the central nervous system and a leading cause of neurological disability in young adults. Clinical presentation and disease course are highly heterogeneous. Typically, disease progression occurs over time and is characterized by the gradual accumulation of disability. The risk of developing MS is driven by complex interactions between genetic and environmental factors, including the gut microbiome. How the commensal gut microbiota impacts disease severity and progression over time remains unknown. In a longitudinal study, disability status and associated clinical features in 58 MS patients were tracked over 4.2 ± 0.98 years, and the baseline fecal gut microbiome was characterized via 16S amplicon sequencing. Progressor status, defined as patients with an increase in Expanded Disability Status Scale (EDSS), were correlated with features of the gut microbiome to determine candidate microbiota associated with risk of MS disease progression. We found no overt differences in microbial community diversity and overall structure between MS patients exhibiting disease progression and non-progressors. However, a total of 41 bacterial species were associated with worsening disease, including a marked depletion in Akkermansia, Lachnospiraceae, and Oscillospiraceae, with an expansion of Alloprevotella, Prevotella-9, and Rhodospirillales. Analysis of the metabolic potential of the inferred metagenome from taxa associated with progression revealed enrichment in oxidative stress-inducing aerobic respiration at the expense of microbial vitamin K2 production (linked to Akkermansia), and a depletion in SCFA metabolism (linked to Oscillospiraceae). Further, as a proof of principle, statistical modeling demonstrated that microbiota composition and clinical features were sufficient to predict disease progression. Additionally, we found that constipation, a frequent gastrointestinal comorbidity among MS patients, exhibited a divergent microbial signature compared with progressor status. These results demonstrate a proof of principle for the utility of the gut microbiome for predicting disease progression in MS in a small well-defined cohort. Further, analysis of the inferred metagenome suggested that oxidative stress, vitamin K2, and SCFAs are associated with progression, warranting future functional validation and mechanistic study.

Abstract Feeding behavior, the most fundamental physiological activity, is controlled by two opposing groups of factors, orexigenic and anorexigenic factors. The sulfakinin family, an insect analogue of the mammalian satiety factor cholecystokinin (CCK), has been shown to suppress food intake in various insects. Nevertheless, the mechanisms through which sulfakinin regulates feeding behavior remain a biological question. This study aimed to elucidate the signaling pathway mediated by the anorexigenic peptide sulfakinin in Bombyx mori. We identified the Bombyx mori neuropeptide G protein-coupled receptor A9 (BNGR-A9) as the receptor for sulfakinin through functional assays. Stimulation with sulfakinin triggered a swift increase in intracellular IP3, Ca2+, and a notable enhancement of ERK1/2 phosphorylation, in a manner sensitive to a Gαq-specific inhibitor. Treatment with synthetic sulfakinin resulted in decreased food consumption and average body weight. Additionally, administering synthetic sulfakinin to silkworms significantly elevated hemolymph trehalose levels, an effect markedly reduced by pre-treatment with BNGR-A9 dsRNA. Consequently, our findings establish the sulfakinin/BNGR-A9 signaling pathway as a critical regulator of feeding behavior and hemolymph trehalose homeostasis in Bombyx mori, highlighting its roles in the negative control of food intake and the positive regulation of energy balance.

Ganoderma is a large and diverse genus containing fungi that cause white rot to infect a number of plant families. This study describes G. phyllanthicola and G. suae as new species from Southwest China, based on morphological and molecular evidence. Ganoderma phyllanthicola is characterized by dark brown to purplish black pileus surface with dense concentric furrows, pale yellow margin, irregular pileipellis cells, small pores (5–7 per mm) and ellipsoid to sub-globose basidiospores (8.5–10.0 × 6.0–7.5 µm). Ganoderma suae is characterized by reddish brown to oxblood red pileus surface and lead gray to greyish-white pore surface, heterogeneous context, wavy margin and almond-shaped to narrow ellipsoid basidiospores (8.0–10.5 × 5.0–7.0 μm). The phylogeny of Ganoderma is reconstructed with multi-gene sequences: the internal transcribed spacer region (ITS), the large subunit (nrLSU), translation elongation factor 1-α gene (TEF-1α) and the second subunit of RNA polymerase II (RPB2). The results show that G. suae and G. phyllanthicola formed two distinct line-ages within Ganoderma. Descriptions, illustrations and phylogenetic analyses results of the two new species are presented.

Abstract Background Epididymal tumors, especially malignant tumors, have low incidence and are rare in our clinical work. However, they may progress quickly and have poor prognosis. For such rare clinical cases with extremely low incidence rates, and as they are prone to misdiagnosis and missed diagnosis and have a very poor prognosis, clinical workers need to pay special attention and consider the possibility of primary epididymal malignant tumors. Case report A 63-year-old Chinese male patient from Asia was admitted due to scrotal pain. Upon examination, an abnormal lesion was found in the right epididymal region. After thorough evaluation, surgical resection was performed, and the postoperative pathological result confirmed the presence of epididymal adenocarcinoma. After further ruling out secondary lesions, primary epididymal adenocarcinoma was considered. Right retroperitoneal lymph node dissection was performed under laparoscopic for treatment, and 1/11 lymph node metastasis was detected after surgery. The patient is currently under close follow-up. Conclusions The number of clinical cases of primary epididymal malignant tumors is very limited, there is currently no standardized diagnosis and treatment process, and there is a lack of systematic evaluation methods regarding the effectiveness of different treatment options such as chemotherapy, radiotherapy, immunotherapy, and targeted therapy. In addition, the outcome is difficult to predict. In this article, we reviewed relevant literature and systematically elaborated on the diagnosis and treatment of epididymal malignant tumors, hoping to provide useful information for relevant experts.

Pangenomes are replacing single reference genomes to capture all variants within a species or clade, but their analysis predominantly leverages graph-based methods that require multiple high-quality genomes and computationally intensive multiple-genome alignments. K-mer decomposition is an alternative to graph-based pangenomes. However, how to directly use k-mers for the population genetic analyses is unknown. Here, we developed a novel strategy that uses the variants of k-mer count in the genome for population analyses. To test the effectivity of this method, we compared it directly to the SNP-based method on the analysis of population structure and genetic diversity of 267 Saccharomyces cerevisiae strains within two simulated datasets and a real sequence dataset. The population structure identified with k-mers recapitulates that obtained using SNPs, indicating the effectiveness of k-mer-based approach, and higher genetic diversity within real dataset supported k-mers contained more genetic variants. Based on k-mer frequency, we found not only SNP but also some insertion/deletion and horizontal gene transfer (HGT) fragments related to the adaptive evolution of S. cerevisiae. Our study creates a framework for the alignment- and reference-free (ARF) method in population genetic analyses, which will be more pronounced in the species with no complete genome or highly diverged species.

Peanut is a plant characterised by belowground fruiting that absorbs nutrients not only through its roots but also through its pods. However, little is currently known regarding the species of bacteria that contribute to nutrient absorption and utilisation in this plant's pod and root zones. This study examined the effects of root and pod area nitrogen (N) fertiliser application on peanut rhizosphere and geocarposphere microbial communities and functions. Using two peanut cultivars [nodulated Huayu 22 (H) and non-nodulated NN-1 (B)], we applied the following four treatments: no N fertiliser (HT1, BH1); N applied to geocarposphere soil (HT2, BT2); N applied to rhizosphere soil (HT3, BT3), and N applied to both rhizosphere and geocarposphere soil (HT4, BT4). The results revealed that compared with HT1 and BT1, the HT3, HT4, BT3, and BT4 treatments promoted increases in total plant accumulated N of 11.2, 30.1, 38.5, and 9.9%, respectively. Moreover, N input contributed to an increase in the abundance of bacteria colonising the surrounding pods, which differed significantly from bacteria colonising the rhizosphere. Among the top four bacterial phyla detected, we recorded a significant increase in the relative abundances of Proteobacteria and Gemmatimonadetes in response to treatments HT2 and HT4, whereas the highest relative abundances of Acidobacteria and Actinobacteria were detected in HT3 plants. Regarding cultivar B, we detected increases in the relative abundances of Bacteroidetes and Gemmatimonadetes in response to the BT2 and BT4 treatments, and in the relative abundance of Actinobacteria in BT3 treated soil. The findings of FAPROTAX functional analysis revealed clear differences among the T2, T4, and T3 treatments of two peanut cultivars concerning the functional groups with the highest relative abundances. These findings will make a considerable contribution to enhancing our understanding of the effects of N fertilisation on soil microbial structure and function in the rhizosphere and geocarposphere of peanuts and can provide a basis for identifying beneficial bacteria for promoting N utilisation and yield enhancement.

Abstract Background Sarcopenia is associated with increased morbidity and mortality. The systemic immune-inflammation index (SII) has been correlated to a variety of disorders. The present study conducted a systematic review and meta-analysis to investigate the relationship between SII and sarcopenia. Methods A literature search was performed in Web of Science, PubMed, Embase, Cochrane Library, CINAHL, China National Knowledge Infrastructure, Chinese Biomedical Literature Database, Wanfang Database, and VIP Chinese Science and Technology Database, from inception to March 2024. Then, the literature quality was assessed. After the heterogeneity test, a random effects or fixed effects model was applied to establish the forest plot, and investigate the relationship between SII and sarcopenia. Then, the sensitivity analysis and publication bias were examined. Results Nine articles, which included 18,634 adults, were analyzed. Sarcopenic adults had higher SII levels, when compared to non-sarcopenic adults (standardized mean difference [SMD] = 0.66, 95% confidence interval [CI] = 0.22 − 0.19, p = 0.003). The high SII level was associated to the increased risk of sarcopenia (odds ratio = 1.52, 95% CI = 1.09–2.13, p = 0.01). In addition, the subgroup analysis revealed that the SII levels were higher in the sarcopenic group, when compared to the non-sarcopenic group, in elderly adults, as well as in adults with or without gastrointestinal disorders. The analysis was robust with a low risk of publication bias. Conclusions SII is closely associated to sarcopenia. Sarcopenic adults had elevated SII levels. The high SII level increased the risk of sarcopenia. Large scale multi-center prospective studies are required to validate these study findings.

Powder bed fusion (PBF), along with subsequent heat treatment, plays a crucial role in enhancing the impact toughness of FeCoNiCrMo0.2 high-entropy alloys (HEAs) and expanding their potential applications in field for high-speed deformation. In this study, the dynamic-mechanical properties and microstructure of the as-built PBF–FeCoNiCrMo0.2 HEAs and quenched PBF–FeCoNiCrMo0.2 HEAs heated at 600–750 °C for 8 h were investigated. As the heating temperature increases, the columnar grains and cellular structures undergo coarsening and the dislocation density gradually decreases. Moreover, higher heating temperatures facilitate the precipitation of Mo-rich second phases. This occurrence can be primarily attributed to the segregation of Mo at the boundaries of the as-built specimens. Consequently, in the quenched specimens, the μ phases are predominantly distributed along the boundaries of the cellular structures. In the quenched Q700 specimen (heated at 700 °C for 8h), the size and volume fraction of the μ phases are measured to be 63.4 nm and 3.59%, respectively. Additionally, under the same condition, the Q700 specimens also exhibited a relatively slight increase in the size of the cellular structures. The impact energy absorption, dynamic yield strength, and dynamic compressive strength of the Q700 quenched specimens were found to be 210.1 MJ/m3, 1066 MPa, and 1649 MPa at a strain rate of 1840 s−1. These values represent a dramatic improvement of 38.1%, 52.7%, and 14.7% higher in comparison to those of the as-built specimens. Under impact deformation, the presence of the μ phases plays a significant role in impeding the movement of dislocations by acting as a pinning agent for the boundaries of the cellular structures, thereby enhancing the strength. Simultaneously, the cellular structures were significantly elongated to form the deformation bands and to coordinate the impact deformation, leading to good impact energy absorption. In combination, the synergistic strengthening and toughening effect of the μ phases and the elongated cellular structures contribute to a remarkable improvement in the impact toughness of the PBF-HEAs.

Abstract Background Climate change is seriously affecting human survival and development, and the anxiety caused by it is becoming increasingly prominent. How to alleviate people’s climate change anxiety, improve the ecological environment, and promote the formation of green lifestyles among people, especially young people, is an important topic that deserves to be explored. This study examined the relationship between climate change anxiety and pro-environmental behaviors and the underlying psychological mechanism in the adolescents. Methods This study explored the crucial role of future self-continuity (FSC) between climate change anxiety (CCA) and pro-environmental behaviors (PEB) in adolescents and examined the moderating role of green self-efficacy (GSE). In this study, a total of 1,851 middle and high school students from five schools were selected for questionnaire survey. Results The results showed that (1) in both middle and high school grades, there was a significant negative correlation between climate change anxiety and pro-environmental behaviors; future self-continuity was significantly positively correlated with pro-environmental behaviors; green self-efficacy was negatively correlated with climate change anxiety and positively correlated with pro-environmental behaviors; (2) climate change anxiety negatively predicted pro-environmental behaviors, and compared with middle school grades, high school grade adolescents’ climate change anxiety was significantly predicted pro-environmental behaviors. Future self-continuity mediated the relationship between climate change anxiety and pro-environmental behaviors in both grades. (3) green self-efficacy moderated the second half of the pathway of the mediation model only in middle grades. Specifically in middle school, future self-continuity did not significantly predict pro-environmental behaviors at low green self-efficacy level, but positively predicted pro-environmental behaviors at high green self-efficacy level. In high school, future self-continuity did not significantly predict pro-environmental behaviors in either high or low green self-efficacy level. Conclusion This study suggests that there is a moderated mediation model between adolescents’ climate change anxiety and pro-environmental behaviors, with different mediating and moderating effects among adolescents in various grades. This is of great significance in alleviating climate anxiety among adolescents and cultivating their pro-environmental behaviors.

Abstract Background PIEZO1 works differently in different cancers and at different stages of development. The objective of the current study was to explore the function and underlying mechanism of PIEZO1 in lung adenocarcinoma (LUAD) cells. Methods Different LUAD cell lines were treated with PIEZO1 inhibitor (GsMTx4) and agonist (Yoda1), and the expression of PIEZO1 in LUAD cells was detected using real‐time quantitative PCR (RT‐qPCR) and western blotting. The effects of PIEZO1 on invasion, migration and epithelial‐mesenchymal transition (EMT) markers protein expression of LUAD cells were detected using the MTT assay, flow cytometry, transwell assay, wound‐healing assay, and western blotting. Reactive oxygen species (ROS) agonists (BAY 87–2243) and inhibitors (NAC) and Wnt/β‐catenin pathway inhibitors (iCRT3) were selected to treat A549 cells to investigate the mechanism of PIEZO1 on ROS production and Wnt/β‐catenin expression in A549 cells. Results In A549, NCI‐H1395, and NCI‐H1975 cells, GsMTx4 promoted cell proliferation, invasion, migration, upregulated EMT‐related marker protein expression, and inhibited cell apoptosis, while Yoda1 exerted effects opposite to those of GsMTx4. In A549 cells, GsMTx4 can reduce ROS production, it also inhibited ROS production, apoptosis, and downregulated proapoptotic markers induced by BAY 87–2243. Importantly, BAY 87–2243 blocked the effect of GSMTX4‐induced Wnt/β‐catenin overexpression. Similarly, Yoda1 can reduce the effect of NAC. In addition, iCRT3 can block the upregulation of EMT‐related marker proteins by GsMTx4, and increase apoptosis and decrease cell invasion and migration. Conclusion In summary, PIEZO1 acts as a cancer suppressor by regulating the ROS/Wnt/β‐catenin axis, providing a new perspective on the role of mechanosensitive channel proteins in cancer.

Abstract Background The C2H2 zinc finger protein family plays important roles in plants. However, precisely how C2H2s function in Opisthopappus (Opisthopappus taihangensis and Opisthopappus longilobus) remains unclear. Results In this study, a total of 69 OpC2H2 zinc finger protein genes were identified and clustered into five Groups. Seven tandem and ten fragment repeats were found in OpC2H2s, which underwent robust purifying selection. Of the identified motifs, motif 1 was present in all OpC2H2s and conserved at important binding sites. Most OpC2H2s possessed few introns and exons that could rapidly activate and react when faced with stress. The OpC2H2 promoter sequences mainly contained diverse regulatory elements, such as ARE, ABRE, and LTR. Under salt stress, two up-regulated OpC2H2s (OpC2H2-1 and OpC2H2-14) genes and one down-regulated OpC2H2 gene (OpC2H2-7) might serve as key transcription factors through the ABA and JA signaling pathways to regulate the growth and development of Opisthopappus species. Conclusion The above results not only help to understand the function of C2H2 gene family but also drive progress in genetic improvement for the salt tolerance of Opisthopappus species.

Abstract Chinese consumers have shown a strong preference for foreign brands (FBs) for many years. However, in recent years, rising patriotism has spurred Chinese consumers to source locally, indicating a shift in preference for patriotic brands (PBs). FBs operating in the world’s second-largest market are now framing their marketing strategies to appeal more to Chinese consumers. This study examines two culturally mixed co-branded product (CMCP) framing strategies: foreign × host culture (FB × PB) and host × foreign culture (PB × FB). The results show that the effects of product fit and cultural congruence on co-branding attitude for PB × FB is stronger than that of FB × PB, and the influence of brand fit on co-branding attitude for FB × PB is stronger than that of PB × FB. Background: Cancer-associated cachexia is a multi-organ disorder associated with progressive weight loss due to a variable combination of anorexia, systemic inflammation and excessive energy wasting. Considering the importance of immunoregulation in cachexia, it still lacks a complete understanding of the immunological mechanisms in cachectic progression. Aim of review: Our aim here is to describe the complex immunoregulatory system in cachexia. We summarize the effects and translational potential of the immune system on the development of cancer-associated cachexia and we attempt to conclude with thoughts on precise and integrated therapeutic strategies under the complex immunological context of cachexia. Key Scientific Concepts of Review: This review is focused on three main key concepts. First, we highlight the inflammatory factors and additional mediators that have been identified to modulate this syndrome. Second, we decipher the potential role of immune checkpoints in tissue wasting. Third, we discuss the multilayered insights in cachexia through the immunometabolic axis, immune-gut axis and immune-nerve axis.

Background In the UK, one in four patients are in work at the time of their hip or knee replacement surgery. These patients receive little support about their return to work (RTW). There is a need for an occupational support intervention that encourages safe and sustained RTW which can be integrated into National Health Service practice. We developed a two-arm intervention trial, based on a feasibility study, to assess whether an occupational support intervention (the OPAL (Occupational support for Patients undergoing Arthroplasty of the Lower limb) intervention) is effective in supporting a reduced time to full, sustained RTW compared with usual care in patients undergoing hip and knee replacement.Methods and analysis This is a multicentre, individually randomised controlled superiority trial comparing the OPAL intervention to usual care. 742 working adults listed for elective primary hip or knee replacement, who intend to RTW, will be randomised to the OPAL intervention or usual care. The intervention comprises: (1) multimedia information resources; and (2) support from a designated RTW coordinator. The primary outcome is time until ‘full’ sustained RTW without sick leave for a consecutive 4-week period. Secondary outcomes are: time to any RTW, measures of functional recovery, number of ‘sick days’ between surgery and ‘full’ sustained RTW and the use of workplace modifications to facilitate their return. A health economic evaluation and a mixed methods process evaluation will assess cost-effectiveness and the implementation, fidelity and acceptability of the intervention, respectively. Outcomes will be collected at baseline, 3, 6, 9 and 12-month follow-up time points, as well as a monthly RTW questionnaire.Ethics and dissemination Dissemination will focus on supporting the wider adoption and implementation of the intervention (if effective) and will target groups for whom the results will be relevant. The de Winter electrocardiogram (ECG) pattern indicates severe stenosis or occlusion of the left anterior descending artery (LAD). We present a 72-year-old female with 1.5-h chest pain. Angiography and optical coherence tomography (OCT) revealed 90 % LAD stenosis with plaque rupture, but no ST-segment elevation in the precordial leads. The de Winter pattern, characterized by upsloping ST-segment depression in V1-V6, appeared only in lead V2. Following successful percutaneous coronary intervention (PCI), the de Winter pattern disappeared. This case underscores the significance of the de Winter pattern in one precordial lead, necessitating prompt angiography and PCI for improved patient outcomes.

Summary: Aortic dissection is characterized pathologically by aortic medial degeneration (AMD) where disturbance of mitochondrial dynamics may be involved. Stearic acid (SA) can promote mitochondrial fusion and improve mitochondrial function. Here, we established an AMD mouse model through oral administration of β-aminopropionitrile (BAPN) and a cellular model by treating primary vascular smooth muscle cells (VSMCs) with Angiotensin-II to explore the potential role of SA in AMD. Our results showed SA reduced AMD and prolonged survival of BAPN-treated mice. Excessive mitochondrial fission was observed during AMD both in vivo and in vitro, and SA reduced mitochondrial fission and increased fusion. Additionally, SA promoted expression of contractile phenotype markers of VSMCs. At the molecular level, SA reduced AMD by inhibiting JNK/MAPK signaling. Our study suggests SA can promote mitochondrial fusion and increase the contractile phenotype of VSMCs by inhibiting JNK/MAPK signaling, thereby reducing AMD formation and possibly the consequent risk of aortic dissection.

To explore the characteristics of the dynamic shear modulus of river-phase (as opposed to estuarine) floodplain interbedded soil, undisturbed interbedded soil from the floodplain of the Yangtze River in Nanjing was subjected to strain-controlled cyclic triaxial tests to investigate how the initial effective confining pressure (σʹm), consolidation ratio (kc), and degree of consolidation (U) influence the maximum dynamic shear modulus Gmax and the dynamic shear modulus ratio G/Gmax. The results show that for this soil, G decreases with increasing strain amplitude, and for a given strain amplitude, G increases with increasing σʹm, kc, and U. Compared with soil from the Yangtze estuary, kc has a greater effect on Gmax of the floodplain interbedded soil. Finally, a modified Martin-Davidenkov model is proposed for predicting G/Gmax of river-phase floodplain interbedded soil under different σʹm, kc, and U.

Abstract Visual observation and therapeutic intervention against tumors hold significant appeal for tumor treatment, particularly in meeting the demands of intraoperative navigation. From a clinical perspective, the naked‐eye visualization of tumors provides a direct and convenient approach to identifying tumors and navigating during surgery. Nevertheless, there is an ongoing need to develop effective solutions in this frontier. Genetically engineered microorganisms are promising as living therapeutics for combatting malignant tumors, leveraging precise tumor targeting and versatile programmed functionalities. Here, genetically modified Escherichia coli (E. coli) MG1655 bacterial cells are introduced, called MelaBac cells, designed to express tyrosinase continuously. This bioengineered melanogenesis produces melanin capable of pigmenting both subcutaneous CT26 xenografts and chemically induced colorectal cancer (CRC). Additionally, MelaBac cells demonstrate the initiation of photonic hyperthermia therapy and immunotherapy against tumors, offering promising selective therapeutic interventions with high biocompatibility.

Infectious bronchitis virus (IBV) is responsible for avian infectious bronchitis, a disease prevalent in countries with intensive poultry farming practices. Given the presence of multiple genotypic strains in China, identifying the regionally dominant genotypes is crucial for the implementation of effective prevention and control measures. This study focuses on the IBV strain CK/CH/WJ/215, isolated from a diseased commercial chicken flock in China in 2021. The CK/CH/WJ/215 isolate was genetically characterized through complete S1 sequence analysis. Phylogenetic comparisons were made with prevalent vaccine strains (H120, LDT3-A, and 4/91). Glycosylation patterns in the S1 protein were also analyzed. Pathogenicity was assessed in 7-day-old specific-pathogen-free chicks, monitoring morbidity, mortality, and tissue tropisms. Phylogenetic analysis clustered the CK/CH/WJ/215 isolate within the GI-19 lineage. Identity with the vaccination strains H120, LDT3-A, and 4/91 was low (75.7%, 78.6%, and 77.5% respectively). Novel glycosylation sites at positions 138 and 530 were identified compared to H120 and LDT-A. The isolate demonstrated nephropathogenic characteristics, causing 100% morbidity and 73.3% mortality in SPF chicks, with broader tropisms in tissues including trachea, lungs, kidneys, and bursa of Fabricius. Comprehensive genetic and pathological investigations revealed significant differences between the CK/CH/WJ/215 isolate and common vaccine strains, including novel glycosylation sites and a strong multiorgan infective capability. These findings are crucial for understanding the evolutionary dynamics of IBV and developing more effective prevention and control strategies.

Metabolic diseases are a group of disorders caused by metabolic abnormalities, including obesity, diabetes, non-alcoholic fatty liver disease, and more. Increasing research indicates that, beyond inherent metabolic irregularities, the onset and progression of metabolic diseases are closely linked to alterations in the gut microbiota, particularly gut bacteria. Additionally, fecal microbiota transplantation (FMT) has demonstrated effectiveness in clinically treating metabolic diseases, notably diabetes. Recent attention has also focused on the role of gut viruses in disease onset. This review first introduces the characteristics and influencing factors of gut viruses, then summarizes their potential mechanisms in disease development, highlighting their impact on gut bacteria and regulation of host immunity. We also compare FMT, fecal filtrate transplantation (FFT), washed microbiota transplantation (WMT), and fecal virome transplantation (FVT). Finally, we review the current understanding of gut viruses in metabolic diseases and the application of FVT in treating these conditions. In conclusion, FVT may provide a novel and promising treatment approach for metabolic diseases, warranting further validation through basic and clinical research.

Background: Limb remote ischemic postconditioning (LRIP) and paeoniflorin (PF) both can ameliorate cerebral ischemia reperfusion (I/R) injury. At present, whether LRIP combined with PF can achieve better therapeutic effect is unknown. Purpose: This study explored the alleviating effect and mechanism of LRIP in combination with PF on cerebral I/R injury in rats. Methods: Middle cerebral artery occlusion (MCAO) surgery was performed on rats except Sham group. Then PF (2.5 mg/kg, 5 mg/kg, 10 mg/kg) was administrated by intraperitoneal injection 10 min before the start of reperfusion. LRIP was operated on the left femoral artery at 0 h of reperfusion. Behavioral testing was used to assess neurological impairment, while TTC staining was used to examine infarct volume. Protein expression of MyD88, TRAF6, p38-MAPK and phosphorylation of p47phox in neutrophils from rat peripheral blood were tested by Western blot. Rat bone marrow neutrophils were extracted and incubated for 24 h with serum from rats after LRIP combined with PF. p38 MAPK inhibitor group was administrated SB203580 while the nicotinamide adenine dinucleotide phosphate (NADPH) oxidase inhibitor group was administrated Apocynin. Neutrophils were stimulated by fMLP (10 μM). Reactive oxygen species (ROS) production and protein expression of MyD88, TRAF6, p38 MAPK, and p47phox (ser 304 and ser 345) were detected. Results: LRIP combined with PF (5 mg/kg) reduced cerebral infarct volume, ameliorated neurological deficit score (NDS), decreased fMLP-stimulated ROS release and downregulated the protein expression of MyD88, TRAF6, p38-MAPK and phosphorylation of p47phox (ser 304 and ser 345) in neutrophils. Conclusion: The protective effect of LRIP combined with PF on cerebral I/R injury was better than either alone. Taken together, we provided solid evidence to demonstrate that the combination of LRIP and PF had potential to alleviate cerebral I/R injury, which was regulated by MyD88-TRAF6-p38 MAPK pathway and neutrophil NADPH oxidase pathway.

The measurement of resistivity in a compressed material within a diamond anvil cell presents significant challenges. The high-pressure experimental setup makes it difficult to directly measure the size changes induced by pressure in the three crystallographic directions of the sample. In this study, we introduce a novel and effective method that addresses these technical challenges. This method is anticipated to offer a valuable foundation for high-pressure investigations on quantum materials, particularly those with anisotropic layered structures.

Abstract Continental alkaline magmatism has been suggested to play a significant role in releasing deep mantle carbon into the atmosphere, which can greatly impact the global climate. However, the temporal variations of alkaline magmatism and their potential to modulate climate over geologic time remain poorly constrained. The detrital zircon record is a frequently used proxy for tracking secular variations in particular magmatism. Here, we use a novel machine‐learning technique to discriminate zircon from carbonatites, kimberlites, and other alkaline rocks. A global compilation of detrital zircon yields continental alkaline magmatic flare‐ups between 1,050−850, 650−500, 250−200, and 50−0 Ma. Our estimates indicate relatively elevated contributions of total magmatic carbon outgassing from alkaline magmatism during the aforementioned magmatic flare‐ups. We infer that anomalous alkaline magmatism may influence global warming during specific intervals of geologic time, but when they are not that voluminous or persistent extensive arc magmatism may drive warming conditions.

BackgroundThis study aimed to determine the diagnostic accuracy of CA125, HE4, systemic immune-inflammation index (SII), prognostic nutritional index (PNI), fibrinogen-to-albumin ratio (FAR), neutrophil-to-lymphocyte ratio (NLR), platelet-to-lymphocyte ratio (PLR), monocyte-to-lymphocyte ratio (MLR), and the combination of the six inflammatory-nutritional markers for ovarian cancer (OC) to identify the best diagnostic indicator for OC early diagnosis. An extensive study was performed to establish the connection between these indicators and the pathological aspects of OC.MethodsA total of 170 individuals were included in this study, with 87 diagnosed with OC and 83 with benign ovarian tumors (BOTs). The diagnostic abilities of the variables were evaluated by calculating sensitivity, specificity, and area under the ROC curves. Through the use of DCA, we evaluated the variables’ clinical value in the discrimination of ovarian masses.ResultsAll markers showed significant diagnostic power for OC. CA125, HE4, SII, FAR, and MLR levels significantly increased from the BOTs group to the early-stage OC group. The advanced-stage OC group had significantly lower PNI values compared to the early-stage OC group but significantly higher levels of CA125, HE4, SII, NLR, and FAR. Moreover, the OC group with lymph node metastasis exhibited significantly higher levels of CA125, HE4, SII, NLR, PLR, and FAR, in contrast to the non-metastatic group, while PNI levels were significantly lower. Categorical factors, such as histological grade and pathological classification, showed noticeable discrepancies in CA125 and HE4 levels. NLR was significantly different among the pathological type groups. Among the six inflammatory-nutritional markers, the FAR displayed the greatest diagnostic value. In the analysis of logistic regression, it was observed that a combination marker containing all six inflammatory-nutritional markers exhibited a notably higher AUC value (0.881; 95% CI, 0.823 - 0.926) than any of the individual marker.ConclusionPNI, NLR, PLR, MLR, SII, and FAR showed excellent diagnostic performance for OC. The combination of these markers demonstrated a superior diagnostic capability compared to each individual one. The systemic inflammatory indicators may be helpful to diagnose OC.

The effect of the fine content (FC) and the relative density (Dr) on the deformation properties of saturated coral sand has been explored through a series of drained cyclic triaxial tests. An important finding is that both FC and Dr significantly affect the development of cumulative volumetric strain (εvd,irN) and dynamic elastic modulus (Ed,N). Steady state cumulative volumetric strain (εvd,irs) increases with FC and decreases with Dr. When FC increases from 0 % to 30 %, the increment in εvd,irs is about 0.69 %, 0.41 %, 0.16 % under Dr=30 %, 45 %, 70 %, respectively. Furthermore, the development rate of εvd,irN increased with FC. Another significant finding is that the increase of the FC leads to a certain decay of the initial dynamical shear modulus (Ed,1) and the steady state dynamical elastic modulus (Ed,s). When FC increases from 0 % to 30 %, the reduction in Ed,1 is about 16.38 MPa, 12.39 MPa, 10.31 MPa and the reduction in Ed,s is about 6.96 MPa, 5.44 MPa, 3.24 MPa under Dr = 30 %, 45 %, 70 %, respectively. A dimensionless parameter, the relative dynamic elastic modulus (Er), is introduced to characterize the development of Ed,N with N. It is found that the growth rate of Er increased with FC, but it is independent on Dr. The equivalent skeleton void ratio (esk*) was introduced based on the binary medium theory. It is found that εvd,irs shows a power-function growth trend with an increase in esk*, while Ed,1 and Ed,s decrease linearly with the increase of esk*. A predictive model between εvd,irN/εvd,irs, Er, and esk* was established, and the applicability of the model was verified based on data from the literature.

A key and urgent scientific issue is how to characterize the complex intergranular contact state and mechanical behavior evolution of sand–fines mixed materials and then show how this state contributes to the static and dynamic properties of such materials. Both theoretical analyses and experimental data suggest that the mixes have different intergranular contact states as the fines content (FC) increases, and the mixed materials can be classified as sand-like, in-transition, or fines-like. The capability of the Rahman semi-empirical formula to predict the threshold fines content FCth—which distinguishes the regime of “fines in sand” (sand-dominated behavior) from that of “sand in fines” (fines-dominated behavior)—is verified using the basic material and mechanical properties of mixed materials from the literature. Developed from the method for determining the theoretical minimum void ratio, a new method that allows unified characterization of the critical intergranular contact state parameters FCin-min and FCin-max for in-transition soils is established based on the binary packing model, and FCin-min and FCin-max can be determined simply through explicit expressions that use some material physical indices of pure-sand and pure-fines materials. The proposed procedure offers significant advantages for evaluating the critical intergranular contact state and mechanical properties of sand–fines mixed materials in geotechnical engineering practice.

Objective To analyze the risk factors of bleeding after subcutaneous injection of low-molecular-weight heparin in elderly patients with unstable angina pectoris and weakness. Methods Elderly patients with unstable angina pectoris and weakness who were hospitalized in the Department of Geriatrics and the Department of Cardiology of Nanjing Hospital Affiliated to Nanjing Medical University(Nanjing First Hospital) from June 2020 to June 2023, they were randomly divided into subcutaneous hemorrhage group(n=90) and control group(n=90) according to whether subcutaneous hemorrhage occurred.General data such as age, gender, body mass index(BMI), as well as combined diseases and injection methods were compared between the two groups.Multivariate logistic regression was performed to analyze the risk factors for subcutaneous hemorrhage after subcutaneous injection of low-molecular-weight heparin. Abstract DNA is a danger signal sensed by cGAS to engage signaling through STING to activate innate immune functions. The best-studied downstream responses to STING activation include expression of type I interferon and inflammatory genes, but STING also activates other pathways, including apoptosis. Here, we report that STING-dependent induction of apoptosis in macrophages occurs through the intrinsic mitochondrial pathway and is mediated via IRF3 but acts independently of gene transcription. By intersecting four mass spectrometry datasets, we identify SAM68 as crucial for the induction of apoptosis downstream of STING activation. SAM68 is essential for the full activation of apoptosis. Still, it is not required for STING-mediated activation of IFN expression or activation of NF-κB. Mechanistic studies reveal that protein trafficking is required and involves SAM68 recruitment to STING upon activation, with the two proteins associating at the Golgi or a post-Golgi compartment. Collectively, our work identifies SAM68 as a STING-interacting protein enabling induction of apoptosis through this DNA-activated innate immune pathway.

Sulforaphane, a naturally specialized metabolite, plays significant roles in human disease prevention and plant defense. Myrosinase (MY) is a key gene responsible for the catalysis of sulforaphane formation, but the molecular mechanisms through which MY regulates sulforaphane biosynthesis in plants remains largely unknown. Here, we discovered that the change of sulforaphane content in broccoli sprouts caused by exogenous selenite treatments is positively related to BoMY expression. BoMY overexpression in the Arabidopsis thaliana tgg1 mutants could dramatically increase myrosinase activity and sulforaphane content in the rosette leaves of 35S::BoMY/tgg1 and rescue its phenotypes. Moreover, an obvious increase of myrosinase activity and sulforaphane content was displayed in transgenic BoMY-overexpressed broccoli lines. In addition, a 2 033 bp promoter fragment of BoMY was isolated. Yeast one-hybrid (Y1H) library screening experiment uncovered that one bHLH transcription factor, BoFAMA, could directly bind to BoMY promoter to activate its expression, which was further evidenced by Y1H assay and dual-luciferase reporter assay. BoFAMA is a selenite-responsive transcription factor that is highly expressed in broccoli leaves; its protein is solely localized to nucleus. Additionally, genetic evidence suggested that the knockdown of FAMA gene in Arabidopsis thaliana could significantly decrease sulforaphane yield by inhibiting the expression of myrosinase genes. Interestingly, exogenous selenite supply could partially restore the low level of sulforaphane content in transgenic Arabidopsis FAMA-silencing plants. Our findings uncover a novel function of FAMA-MY module in the regulation of selenite-mediated sulforaphane synthesis and provide a new insights into the molecular mechanism by which selenite regulates the accumulation of sulforaphane in plants.

Abstract Chlamydia trachomatis (CT) is the most common sexually transmitted infections globally, and CT infection can enhance HPV persistence. Epidemiological analysis has shown that patients with CT/HPV coinfection have a higher risk of developing cervical cancer and exhibit more rapid progression to cervical cancer than patients with HPV infection alone. However, the mechanism has not been fully elucidated. Here, we report that CT infection supports HPV persistence by further suppressing the functions of Langerhans cells (LCs); in particular, CT further activates the PI3K pathway and inhibits the MAPK pathways in LCs, and these pathways are frequently involved in the regulation of immune responses. CT/HPV coinfection also impairs LC functions by reducing the antigen-presenting ability and density of LCs. Moreover, CT/HPV coinfection can alter T-cell subsets, resulting in fewer CD4 + and CD8 + T cells and more infiltrating Tregs. Moreover, CT/HPV coinfection decreases the CD4 + /CD8 + T cell ratio to below 1, coinfection also induces greater T lymphocytes’ apoptosis than HPV infection, thus impairing cell-mediated immunity and accelerating the progress to cervical cancer.

Abstract Chronic obstructive pulmonary disease (COPD) is a leading aging related cause of global mortality. Small airway narrowing is recognized as an early and significant factor for COPD development. Senescent fibroblasts were observed to accumulate in lung of COPD patients and promote COPD progression through aberrant extracellular matrix (ECM) deposition and senescence-associated secretory phenotype (SASP). On the basis of our previous study, we further investigated the the causes for the increased levels of miR-377-3p in the blood of COPD patients, as well as its regulatory function in the pathological progression of COPD. We found that the majority of up-regulated miR-377-3p was localized in lung fibroblasts. Inhibition of miR-377-3p improved chronic smoking-induced COPD in mice. Mechanistically, miR-377-3p promoted senescence of lung fibroblasts, while knockdown of miR-377-3p attenuated bleomycin-induced senescence in lung fibroblasts. We also identified ZFP36L1 as a direct target for miR-377-3p that likely mediated its pro senescence activity in lung fibroblasts. Our data reveal that miR-377-3p is crucial for COPD pathogenesis, and may serve as a potential target for COPD therapy.

Abstract The exotic states X(3872) and $$Z_c(3900)$$ Z c ( 3900 ) have long been conjectured as isoscalar and isovector $${\bar{D}}^*D$$ D ¯ ∗ D molecules. In this work, we first propose the triangle diagram mechanism to investigate their productions in B decays as well as their heavy quark spin symmetry partners, $$X_2(4013)$$ X 2 ( 4013 ) and $$Z_c(4020)$$ Z c ( 4020 ) . We show that the large isospin breaking of the ratio $${\mathcal {B}}[B^+ \rightarrow X(3872) K^+]/{\mathcal {B}}[B^0 \rightarrow X(3872) K^0] $$ B [ B + → X ( 3872 ) K + ] / B [ B 0 → X ( 3872 ) K 0 ] can be attributed to the isospin breaking of the neutral and charged $${\bar{D}}^*D$$ D ¯ ∗ D components in their wave functions. For the same reason, the branching fractions of $$Z_c(3900)$$ Z c ( 3900 ) in B decays are smaller than the corresponding ones of X(3872) by at least one order of magnitude, which naturally explains its non-observation. A hierarchy for the production fractions of X(3872), $$Z_c(3900)$$ Z c ( 3900 ) , $$X_2(4013)$$ X 2 ( 4013 ) , and $$Z_c(4020)$$ Z c ( 4020 ) in B decays, consistent with all existing data, is predicted. Furthermore, with the factorization ansatz we extract the decay constants of X(3872), $$Z_c(3900)$$ Z c ( 3900 ) , and $$Z_c(4020)$$ Z c ( 4020 ) as $${\bar{D}}^*D^{(*)}$$ D ¯ ∗ D ( ∗ ) molecules via the B decays, and then calculate their branching fractions in the relevant $$B_{(s)}$$ B ( s ) decays, which turn out to agree with all existing experimental data. The mechanism we proposed is useful to elucidate the internal structure of the many exotic hadrons discovered so far and to extract the decay constants of hadronic molecules, which can be used to predict their production in related processes.

The effects of large granular slow-controlled release fertiliser prepared by a double coating of sulfur and sodium alginate on peanut growth, nitrogen fertiliser utilisation, and soil microbial community were investigated through peanut pot experiments, with a view to providing a theoretical and practical basis for the development of large granular slow-controlled release fertiliser. The results showed that the homemade large granular fertiliser could promote the root development of peanuts, and the root volume increased by 45.10% compared with the uncoated fertiliser at the fruiting stage. At the same time, the soil NH4+-N and NO3--N content were reduced at the seedling stage and increased at the fruiting stage to achieve the fertiliser's slow and controlled release effect. A significant contribution to the net photosynthetic rate was made for growth development and yield in the middle and late stages. Pod dry weight was significantly higher at the blooming stage than uncoated fertiliser, 4.8% higher at the fruiting stage, and 22.9% higher in nitrogen use efficiency (NUE). In terms of microbial bacterial communities, the large granular slow-release fertiliser promoted the diversity of the treated bacterial communities to some extent, with little difference in the relative abundance of soil bacterial communities. These results showed that a one-time application of homemade large granular slow-release fertiliser positively affected peanuts in terms of yield increase, promotion of nitrogen uptake and improved nitrogen utilisation under nitrogen application with urea equivalent, but the overall effect on soil microbial community was small.

The application of liquid crystal (LC)-based reconfigurable intelligent surface (RIS)-assisted transmitters in visible light communication (VLC) systems can guide and amplify the light emitted by light emitting diodes (LEDs). In this paper, we investigate the VLC system employing the RIS-assisted transmitter, where user fairness is maximized by optimizing the refractive index of the RIS module. To address the non-convex problem, we adopt the particle swarm optimization (PSO) algorithm. Simulation results illustrate that equipping the RIS-assisted transmitter in the system and using the proposed algorithm can significantly improve user fairness.

Abstract Background Health security is a critical issue which involves multiple dimensions. It has received increasing attention in recent years, especially in China. In order to improve the national health level, China has made many efforts, such as the “Healthy China 2030” plan proposed several years ago. However, due to the complexity of its national conditions and the difficulty of index design, the results of these efforts are not significant. Therefore, it is necessary to construct a new measurement index system. Methods Based on the questionnaire of “Health China 2030”, we have collected a total of 3,000 participants from all 31 provinces, autonomous regions, and municipalities in China. We used statistical methods such as multiple correspondence analysis and rank-ordered effect analysis to process the data. The balance index is constructed by a series of actions such as weight division, order calculation and ranking. Results Through multiple correspondence analysis, we can find that there was a close relation in the correspondence space between the satisfaction degrees 1, 2, and 3, while a far distance from satisfaction degrees 4 and 5. There were four positive and four negative indices separately based on the average expected level and four clusters after ordinal rank cluster analysis. Generally speaking, there are no prominent discrepancies across gender and residential areas. Conclusions We created and examined balanced indicators for health security in China based on the “Health China 2030” questionnaire. The findings of this study give insight into the overall situation of health security in China and indicate opportunities for improvement.

With the continuous development of the automobile industry, fast and efficient body styling methods have become crucial. Although traditional hand-drawn sketches and 3D modeling are feasible, they often consume lots of time, manpower and financial resources when faced with complex design requirements. Therefore, a quick method for car body modeling has become a hot topic in automotive design. This paper proposes a three-dimensional automotive wireframe model generation and surface reconstruction method based on a single image in any view. Firstly, 3D wireframe model generation for a single car image in any view. By the MobileNet V2 network trained based on the established 2D car body library USMV3085, the visual angle of a specific single car image in any view can be identified, following the relevant wireframe model can be generated with the weight coefficients computed by constrained least squares method according to the key points and the corresponding rotated key points of the 3D wireframe library. Secondly, the corresponding 3D surface is reconstructed automatically. With the skeleton line firstly computed by the Skeleton Generation Algorithm for every 2D projected split part of the 3D wireframe model, corresponding 3D boundary relation pairs and control polygons are obtained, following the 3D surface is reconstructed integrating all split parts. Compared to traditional computer-aided shape design, this automatic method greatly shortens the research and development cycle and has certain reference significance in car body design.

Follicular dendritic cell sarcoma (FDCS) is a rare tumor, which mainly originates from follicular dendritic cells (FDCs) in the lymph nodes. Sometimes FDCS can arise from outside the lymph nodes. FDCS is an extremely rare malignant tumor in intraperitoneal or retroperitoneal tissue. We gathered the detailed clinical data of three patients diagnosed with FDCS in the abdomen. The clinical observations and histopathologic and immunohistochemical features of FDCS were analyzed. The patients included two men and one woman aged 55 ~ 61 years old. The mesentery of the small intestine and colon was involved in case 1, spleen in case 2, and retroperitoneal tissues in case 3. Two patients presented with abdominal masses, and one presented with no obvious symptoms. Histology showed ovoid to spindle neoplastic cells arranged in fascicles and storiforms with inflammatory infiltrate as well as whorled patterns in some areas. Immunohistochemical staining was positive for CD21, CD23, CD35, and SSTR2. FDCS exhibits no characteristic clinical manifestations. Morphologically, FDCS can have overlapping features with many other entities, leading to misdiagnosis. The use of histopathology supplemented with FDC markers, such as CD21, CD23, and CD35, is useful for diagnosis and differential diagnosis.

Abstract Background In cellular activities, essential proteins play a vital role and are instrumental in comprehending fundamental biological necessities and identifying pathogenic genes. Current deep learning approaches for predicting essential proteins underutilize the potential of gene expression data and are inadequate for the exploration of dynamic networks with limited evaluation across diverse species. Results We introduce ECDEP, an essential protein identification model based on evolutionary community discovery. ECDEP integrates temporal gene expression data with a protein–protein interaction (PPI) network and employs the 3-Sigma rule to eliminate outliers at each time point, constructing a dynamic network. Next, we utilize edge birth and death information to establish an interaction streaming source to feed into the evolutionary community discovery algorithm and then identify overlapping communities during the evolution of the dynamic network. SVM recursive feature elimination (RFE) is applied to extract the most informative communities, which are combined with subcellular localization data for classification predictions. We assess the performance of ECDEP by comparing it against ten centrality methods, four shallow machine learning methods with RFE, and two deep learning methods that incorporate multiple biological data sources on Saccharomyces. Cerevisiae (S. cerevisiae), Homo sapiens (H. sapiens), Mus musculus, and Caenorhabditis elegans. ECDEP achieves an AP value of 0.86 on the H. sapiens dataset and the contribution ratio of community features in classification reaches 0.54 on the S. cerevisiae (Krogan) dataset. Conclusions Our proposed method adeptly integrates network dynamics and yields outstanding results across various datasets. Furthermore, the incorporation of evolutionary community discovery algorithms amplifies the capacity of gene expression data in classification.

AbstractMeconopsis bella Prain 1894 (M. bella) is a rare herb within the family Papaveraceae of which unique and gorgeous purple flowers are blooming in the flowering phase. In this study, we reported the complete chloroplast (cp) genome of M. bella, which was mainly distributed on the Qinghai-Tibet plateau. The complete chloroplast genome sequence of M. bella was 153,073 bp in size and was characterized by a typical quadripartite structure consisting of a large single-copy (LSC) region of 83,562 bp, a small single-copy (SSC) region of 178,33 bp and two identical inverted repeats (IR) regions of 25,839 bp. The genome contained 133 genes, including 88 protein-encoding genes, eight ribosomal RNA genes, and 37 transfer RNA genes. Phylogenetic analysis based on the maximum-likelihood (ML) method showed that M. bella was closely related to M. paniculate and M. pinnatifolia within the genus Meconopsis.

Abstract The 2023 update of the Chinese Society of Clinical Oncology (CSCO) Clinical Guidelines for Gastric Cancer focuses on standardizing cancer diagnosis and treatment in China, reflecting the latest advancements in evidence‐based medicine, healthcare resource availability, and precision medicine. These updates address the differences in epidemiological characteristics, clinicopathological features, tumor biology, treatment patterns, and drug selections between Eastern and Western gastric cancer patients. Key revisions include a structured template for imaging diagnosis reports, updated standards for molecular marker testing in pathological diagnosis, and an elevated recommendation for neoadjuvant chemotherapy in stage III gastric cancer. For advanced metastatic gastric cancer, the guidelines introduce new recommendations for immunotherapy, anti‐angiogenic therapy and targeted drugs, along with updated management strategies for human epidermal growth factor receptor 2 (HER2)‐positive and deficient DNA mismatch repair (dMMR)/microsatellite instability‐high (MSI‐H) patients. Additionally, the guidelines offer detailed screening recommendations for hereditary gastric cancer and an appendix listing drug treatment regimens for various stages of gastric cancer. The 2023 CSCO Clinical Guidelines for Gastric Cancer updates are based on both Chinese and international clinical research and expert consensus to enhance their applicability and relevance in clinical practice, particularly in the heterogeneous healthcare landscape of China, while maintaining a commitment to scientific rigor, impartiality, and timely revisions.

Abstract Objective This trial was to investigate the effect of different treatment methods on the clinical efficacy and fertility outcome of patients with adenomyosis. Methods In total, 140 patients with adenomyosis were evenly and randomly allocated into group A (laparoscopic surgery), group B (laparoscopic surgery combined with gonadotropin-releasing hormone analogs [GnRH-a]), group C (ultrasound-guided percutaneous radiofrequency ablation), and group D (ultrasound-guided percutaneous radiofrequency ablation combined with GnRH-a). On the 3rd day after surgery, patients in group B and group D were subcutaneously injected with GnRH-a (Leuprorelin Acetate SR for Injection) at 3.75 mg/time, once every 4 weeks, for a total of 3 months. The therapeutic effects of the 4 groups were compared, including menstrual volume, dysmenorrhea score, uterine volume, clinical efficacy, luteinizing hormone (LH), estradiol (E2), and follicle-stimulating hormone (FSH) levels, CA125 levels, recurrence, pregnancy status, and pregnancy outcomes. Results After treatment, the menstrual volume of 4 groups was lowered, dysmenorrhea, Visual Analog Scale (VAS) score, LH, FSH, E2, and CA125 levels were reduced, and uterine volume was decreased. The menstrual volume, VAS score, levels of LH, FSH, E2, and CA125, and uterine volume were reduced in groups B, C, and D compared with group A, and the decrease was more significant in group D. The total effective rate of group D was 100.00%, which was higher than that of group A (71.43%), group B (80.00%), and group C (82.86%). After one year of drug withdrawal, the recurrence of hypermenorrhea, dysmenorrhea, uterine enlargement, and excessive CA125 in group D was significantly lower than that in groups A, B and C, and the recurrence in groups B and C was significantly lower than that in group A (P

Abstract Graphite is considered to be the most auspicious anode candidate for potassium ion batteries. However, the inferior rate performances and cycling stability restrict its practical applications. Few studies have investigated the modulating the graphitization degree of graphitic materials. Herein, a nitrogen‐doped carbon‐coated carbon fiber composite with tunable graphitization (CNF@NC) through etching growth, in‐situ oxidative polymerization, and subsequent carbonization process is reported. The prepared CNF@NC with abundant electrochemical active sites and a rapid K+/electron transfer pathway, can effectively shorten the K+ transfer distance and promote the rapid insertion/removal of K+. Amorphous domains and short‐range curved graphite layers can provide ample mitigation spaces for K+ storage, alleviating the volume expansion of the highly graphitized CNF during repeated K+ insertion/de‐intercalation. As expected, the CNF@NC‐5 electrode presents a high initial coulombic efficiency (ICE) of 69.3%, an unprecedented reversible volumetric capacity of 510.2 mA h cm−3 at 0.1 A g−1 after 100 cycles with the mass‐capacity of 294.9 mA h g−1. The K+ storage mechanism and reaction kinetic analysis are studied by combining in‐situ analysis and first‐principles calculation. It manifests that the K+ storage mechanism in CNF@NC‐5 is an adsorption‐insertion‐insertion mechanism (i.e., the “1+2” model). The solid electrolyte interphase (SEI) film forming is also detected.

ABSTRACT Smallpox is a highly contagious human disease caused by the variola virus. Although the disease was eliminated in 1979 due to its highly contagious nature and historical pathogenicity, with a mortality rate of up to 30%, this virus is an important candidate for biological weapons. Currently, vaccines are the critical measures to prevent this virus infection and spread. In this study, we designed a peptide vaccine using immunoinformatics tools, which have the potential to activate human immunity against variola virus infection efficiently. The design of peptides derives from vaccine-candidate proteins showing protective potential in vaccinia WR strains. Potential non-toxic and nonallergenic T-cell and B-cell binding and cytokine-inducing epitopes were then screened through a priority prediction using special linkers to connect B-cell epitopes and T-cell epitopes, and an appropriate adjuvant was added to the vaccine construction to enhance the immunogenicity of the peptide vaccine. The 3D structure display, docking, and free energy calculation analysis indicate that the binding affinity between the vaccine peptide and Toll-like receptor 3 is high, and the vaccine receptor complex is highly stable. Notably, the vaccine we designed is obtained from the protective protein of the vaccinia and combined with preventive measures to avoid side effects. This vaccine is highly likely to produce an effective and safe immune response against the variola virus infection in the body.IMPORTANCEIn this work, we designed a vaccine with a cluster of multiple T-cell/B-cell epitopes, which should be effective in inducing systematic immune responses against variola virus infection. Besides, this work also provides a reference in vaccine design for preventing monkeypox virus infection, which is currently prevalent.

Summary: Using noninvasive biomarkers to identify high-risk individuals prior to endoscopic examination is crucial for optimization of screening strategies for esophageal squamous cell carcinoma (ESCC). We conducted a nested case-control study based on two community-based screening cohorts to evaluate the warning value of serum metabolites for esophageal malignancy. The serum samples were collected at enrollment when the cases had not been diagnosed. We identified 74 differential metabolites and two prominent perturbed metabolic pathways, and constructed Metabolic Risk Score (MRS) based on 22 selected metabolic predictors. The MRS generated an area under the receiver operating characteristics curve (AUC) of 0.815. The model performed well for the within-1-year interval (AUC: 0.868) and 1-to-5-year interval (AUC: 0.845) from blood draw to diagnosis, but showed limited ability in predicting long-term cases (>5 years). In summary, the MRS could serve as a potential early warning and risk stratification tool for establishing a precision strategy of ESCC screening.
