Abstract Pulmonary fibrosis is a chronic interstitial lung disease with no curative therapeutic treatment, leading to significant mortality. The aims of this study were to investigate the regulatory mechanisms of mitophagy in the progression of pulmonary fibrosis. Through bioinformatics analysis, we identified the downregulation of long-chain fatty acyl-CoA synthetase 1 (ACSL1) as being associated with the severity of pulmonary fibrosis. A pulmonary fibrosis model was established through bleomycin (BLM) exposure both in vivo and in vitro. Mitoquinone (MitoQ) pretreatment significantly decreased redox damage, stabilized mitochondrial membrane potential (MMP), improved mitochondrial dynamics, and activated PINK1/Parkin-mediated mitophagy, thereby alleviating pulmonary fibrosis. In vitro, overexpression of ACSL1 mitigated mitochondrial damage and restored PINK1/Parkin-mediated mitophagy under BLM exposure. In contrast, ACSL1 inhibition exacerbated pulmonary fibrosis, and these adverse effects could not be reversed by MitoQ treatment. Taken together, our study reveals a novel mechanism underlying the pathogenesis of pulmonary fibrosis and suggests a potential therapeutic target for its treatment.

Abstract VEXAS syndrome, an uncommon yet severe autoimmune disorder stemming from a mutation in the UBA1 gene, is the focus of this paper. The overview encompasses its discovery, epidemiological traits, genetic underpinnings, and clinical presentations. Delving into whether distinct genotypes yield varied clinical phenotypes in VEXAS patients, and the consequent adjustment of treatment strategies based on genotypic and clinical profiles necessitates thorough exploration within the clinical realm. Additionally, the current therapeutic landscape and future outlook are examined, with particular attention to the potential therapeutic roles of IL-6 inhibitors and JAK inhibitors, alongside an elucidation of prevailing limitations and avenues for further research. This study contributes essential theoretical groundwork and clinical insights for both diagnosing and managing VEXAS syndrome.

Solar radiation plays a pivotal role in urban microclimate, significantly, impacting building energy consumption and renewable energy utilization. Utilizing a comprehensive three- and-a- half-year dataset of high-precision urban microclimate, this study proposes a three-step approach to solar radiation prediction: 1) Data Preprocessing and Characterization: The microclimate data are meticulously preprocessed and categorized, with solar radiation values categorized into three resolutions: total daily solar radiation, hourly solar radiation, and 30-min solar radiation. 2) Feature Selection: Various climatic parameters are carefully selected for solar radiation prediction at different resolutions. 3) Machine Learning Models: Three machine learning models, namely Long Short-Term Memory (LSTM), Random Forest (RF) and Support Vector Regression (SVR), are employed for solar radiation prediction. The results show that all three models exhibit prediction accuracy with closely matched errors. Particularly noteworthy findings include SVR’s superior performance in predicting total daily solar radiation with a minimal error rate of about 6.0% and an impressive coefficient of determination of 0.9789. LSTM exhibits adaptability, achieving R2 values of 0.9906 and 0.9956 for hourly and 30-min radiation predictions, respectively. This study offers valuable insights into solar radiation and its implications for building energy research. It underscores that large datasets enable accurate solar radiation predictions, even when excluding certain parameters like daily maximum temperature and relative humidity.

Abstract Non-small cell lung cancer (NSCLC) is characterized by a high incidence rate and poor prognosis worldwide. A deeper insight into the pathogenesis of NSCLC and identification of novel therapeutic targets are essential to improve the prognosis of NSCLC. In this study, we revealed that fibrinogen-like protein 1 (FGL1) promotes proliferation, migration, and invasion of NSCLC cells. Mechanistically, we found that Stat3 acts as a transcription factor and can be recruited to the FGL1 promoter, enhancing FGL1 promoter activity. Lysine-specific demethylase 4A (KDM4A) interacts with Stat3 and facilitates the removal of methyl groups from H3K9me3, thereby enhancing Stat3-mediated transcription of FGL1. Furthermore, we observed that Stat3 and KDM4A promote NSCLC cell proliferation, migration, and invasion partly by upregulating FGL1 expression. Additionally, the expression of FGL1 was significantly higher in cancer tissues (n = 90) than in adjacent non-cancerous tissues (n = 90). Furthermore, patients with high FGL1 expression had a shorter overall survival (OS) compared to those with low FGL1 expression. We measured the expression levels of FGL1 on circulating tumor cells (CTCs) in 65 patients and found that patients with a dynamic decrease in FGL1 expression on CTCs exhibited a better therapeutic response. These findings suggest that the dynamic changes in FGL1 expression can serve as a potential biomarker for predicting treatment efficacy in NSCLC. Overall, this study revealed the significant role and regulatory mechanisms of FGL1 in the development of NSCLC, suggesting its potential as a therapeutic target for patients with NSCLC. Future studies should provide more personalized and effective treatment options for patients with NSCLC to improve clinical outcomes.

Ultraviolet mutagenesis is a safe and efficient method to induce mutations in laver. It has the advantages of non-pollution, high efficiency, easy operation, and low cost. It has been primarily used for mutagenesis of the filament, protoplast, or blade of cultivated Pyropia species, but is rarely used to induce genetic mutants from germinating conchospore. The germination period of conchospores is the period of meiosis in Pyropia haitanensis. The four progeny cells of germinating conchospores are linearly arranged forming a meiotic tetrad. The tetrad cells that undergo genetic recombination can determine the developmental pattern and segregation of traits in the thallus. In this study, short-wave ultraviolet (UV-C) irradiation with different doses (50, 100, 200, 300, 400, 500, and 600 J/m2) was used to induce color mutants during the germination of conchospores in P. haitanensis. The results showed that low-dose irradiation (50 J/m2) promoted the germination of conchospores, while irradiation doses above 100 J/m2 inhibited the germination and growth of tetrad germlings. Therefore, low-doses of UV-C irradiation were used in the production to promote the germination of conchospores and improve the production efficiency of laver. In the dose range of 50–400 J/m2, the frequency of color chimeras increased with increasing irradiation intensity. When the dose was 300 J/m2 and 400 J/m2, and the pigmentation mutation rate was 15.22% and 17.18%, respectively, and the death rate of conchospores was 51.70% and 61.00%, respectively. In the dose range of 50–500 J/m2, with the increase in UV-C irradiation dose, the proportion of 2-color sectored chimera showed a trend of first decreasing and then increasing, and the proportion of 3- and 4-color sectored chimera showed a trend of first increasing and then decreasing. Among them, the regenerated color chimeras that appear were generally 2- and 3-color sectored chimera, yet the proportion of 4-color sectored chimera was the least. When the irradiation intensity reached 500 J/m2, the majority of the conchospores died, and the death rate was 83.98%, and the frequency of color mutants was significantly lower than that of 300 J/m2 and 400 J/m2 dose groups. These results indicate that the mutagenesis effect was the best when the dose was 300 or 400 J/m2, which was convenient to obtain abundant genetic recombination and genetic variation in progeny cells. In addition, UV-C irradiation also had a significant effect on the early development of conchospores and phenotypes of pigmentation mutant arranged in tetrad germlings, which was mainly manifested in the large number of color-sectored blades developed from the irradiated conchospores. Simultaneously, UV-C irradiation retarded the development of cells at the top of conchospore germlings, inhibited the development of cells at the middle base toward the poles, and increased the lateral development, resulting in the decrease in blade aspect ratios. Somatic cell germlings with single colored pigmentation were also isolated from color sectored chimeras by enzymatic hydrolysis. The color species of the five monochromatic mutants obtained were basically consistent with the pigment variant sectors observed on color-sectored thallus, indicating that the obtained color mutants were derived from a single mutant cell on the maternal color-sectored thallus. In conclusion, UV-C irradiations can effectively mutate the conchospores of P. haitanensis, and appropriate irradiation doses (300 or 400 J/m2) can obtain a certain number of color-sectored thallus. This study provides a novel pathway for the preparation of artificial color mutants and mutation breeding in P. haitanensis.

BackgroundCDGSH iron-sulfur domain 2 (CISD2), an iron-sulfur protein with a [2Fe-2S] cluster, plays a pivotal role in the progression of various cancers, including Diffuse Large B-cell Lymphoma (DLBCL). However, the mechanisms by which CISD2 regulates the occurrence and development of DLBCL remain to be fully elucidated.MethodsThe potential role of CISD2 as a predictive marker in DLBCL patients treated with the R-CHOP regimen was investigated through bioinformatics analysis and clinical cohort studies. DLBCL cell lines (SUDHL-4 and HBL-1) were employed in this research. Adenoviral (AV) plasmids were used to either silence or overexpress CISD2 in these DLBCL cell lines. Additionally, the induction of ferroptosis in DLBCL cell lines was assessed. Various parameters, including cell proliferation, intracellular free iron levels, lipid peroxides, reactive oxygen species (ROS), and mitochondrial membrane potential (MMP), were measured. Furthermore, the expression of proteins associated with ferroptosis and ferritinophagy was analyzed. Drug-resistant DLBCL cell lines were developed by gradually increasing doxorubicin (DOX) concentration over 6 months. The biological role of CISD2 in these drug-resistant DLBCL cell lines was subsequently assessed.ResultsElevated CISD2 levels were found to be associated with decreased sensitivity of DLBCL patients to the R-CHOP regimen, as indicated by bioinformatics and clinical cohort analysis. Silencing CISD2 significantly reduced cell proliferation, increased iron accumulation, depleted glutathione (GSH), and elevated malondialdehyde (MDA) levels, alongside the accumulation of ROS and increased MMP. Additionally, BECN1 and NCOA4 expressions were upregulated, while p62, FTH1, and GPX4 expressions were downregulated. Conversely, overexpression of CISD2 reversed these effects. Treatment of DLBCL cell lines with Erastin led to decreased CISD2 levels. Notably, in drug-resistant DLBCL cell lines, CISD2 knockdown promoted ferroptosis and ferritinophagy, restoring sensitivity to DOX and enhancing the efficacy of Erastin treatment.ConclusionOur findings suggest that CISD2 may play a role in the drug resistance observed in DLBCL patients. Inhibition of CISD2 could enhance ferroptosis and ferritinophagy, potentially improving the sensitivity of DLBCL cells to DOX treatment.

This study explored the effect of sesamol on the stability of walnut oil based on the changes of endogenous characteristics in the oxidation process, which provided a theoretical reference for the application of natural antioxidants in walnut oil. A total of 300 mg/kg sesamol (SP), compound antioxidant AC (sesamol 353.62 mg/kg, citric acid 149.60 mg/kg, and BHA 76.33 mg/kg) and 35% sesame oil (35%-SO) were added to walnut oil respectively; in addition, 200 mg/kg t-butylhydroquinone (TBHQ), butylated hydroxyanisole (BHA), and citric acid were used as controls and blank walnut oil to study their effects on peroxide value, acid value, carbonyl compounds, conjugated olefins, phenols, flavonoids, sterols, vitamin E, β-carotene, and 51 fatty acids of walnut oil and their correlation between endogenous antioxidant components. The results showed that the addition of SP, 35%-SO, and AC could inhibit the increase of peroxide value, acid value, and carbonyl compounds in walnut oil, and could inhibit the decrease of β-carotene, total phenols, total sterols, and vitamin E. SP and 35%-SO could inhibit the decrease of total flavonoids, and several antioxidants could inhibit the decrease of endogenous antioxidant components in walnut oil. At the same time, it can better inhibit the change of unsaturated fatty acids in walnut oil. By the end of oxidation, the unsaturated fatty acids of blank walnut oil decreased by 10.31%, but AC, SP, and 35%-SO treatment groups increased by 10.90, 5.09 and 4.13%, respectively. Indicating that it had a certain protective effect on unsaturated fatty acids in walnut oil. There was a certain correlation between the endogenous substances of walnut oil. so the addition of several antioxidants can enhance the endogenous antioxidants of walnut oil, inhibit the oxidation of unsaturated fatty acids, and inhibit the increase of carbonyl compounds, codienes, acid value, and peroxide value. SP and AC have better antioxidant effects on walnut oil and improve the stability of walnut oil.

IntroductionThe impacts of plastic pollution on soil ecosystems have emerged as a significant global environmental concern. The progress in understanding how plastic pollution affects soil microbial communities and ecological functions is essential for addressing this issue effectively.MethodsA bibliometric analysis was conducted on the literature from the Web of Science Core Collection database to offer valuable insights into the dynamics and trends in this field.ResultsTo date, the effects of plastic residues on soil enzymatic activities, microbial biomass, respiration rate, community diversity and functions have been examined, whereas the effects of plastic pollution on soil microbes are still controversial.DiscussionTo include a comprehensive examination of the combined effects of plastic residue properties (Type, element composition, size and age), soil properties (soil texture, pH) at environmentally relevant concentrations with various exposure durations under field conditions in future studies is crucial for a holistic understanding of the impact of plastic pollution on soil ecosystems. Risk assessment of plastic pollution, particularly for nanoplasctics, from the perspective of soil food web and ecosystem multifunctioning is also needed. By addressing critical knowledge gaps, scholars can play a pivotal role in developing strategies to mitigate the ecological risks posed by plastic pollution on soil microorganisms.

IntroductionCasuarina equisetifolia is a common protective forest in coastal areas. However, artificial C. equisetifolia forests cannot self-renew, mainly due to the accumulation of allelochemicals. Endophytic bacteria may alleviate the root growth inhibition caused by allelochemicals in C. equisetifolia seedlings. B. amyloliquefaciens and B. aryabhattai were endophytic bacteria with strong allelopathy in C. equisetifolia root. The allelopathy mechanism of these two endophytes and their interaction with C. equisetifolia remains to be studied.MethodsWhole-genome sequencing of B. amyloliquefaciens and B. aryabhattai isolated from the roots of allelochemical-accumulating C. equisetifolia was performed using Illumina Hiseq and PacBio single-molecule sequencing platforms. Sterile seedlings of C. equisetifolia were treated with either individual or mixed bacterial cultures through root drenching. Transcriptional and metabolomics analyses were conducted after 3 days of infection.Results and discussionWhole-genome sequencing of Bacillus aryabhattai and Bacillus amyloliquefaciens showed that the two strains contained various horizontal gene transfer elements such as insertion sequence, prophage and transposon. In addition, these two strains also contain numerous genes related to the synthesis and catabolism of allelochemicals. After these two strains of bacteria were individually or mixed infected with C. equisetifolia, metabolomics and transcriptomic analysis of C. equisetifolia showed the 11 important secondary metabolite biosynthesis among them alkaloids biosynthesis, phenylpropanoid and terpenes biosynthesis and related genes were putatively regulated. Correlation analysis revealed that 48 differentially expressed genes had strong positive correlations with 42 differential metabolites, and 48 differentially expressed genes had strong negative correlations with 36 differential metabolites. For example, CMBL gene showed positive correlations with the allelochemical (-)-Catechin gallate, while Bp10 gene showed negative correlations with (-)-Catechin gallate.ConclusionThe intergenerational accumulation of allelochemicals may induce horizontal gene transfer in endogenic bacteria of Casuarina equisetifolia root. Endophytic Bacillus plays an allelopathic role by assisting the host in regulating gene expression and the production and/or variety of allelochemicals. This comprehensive study sheds light on the intricate genetic and metabolic interactions between Bacillus endophytes and C. equisetifolia. These findings provide insights into endophyte-mediated allelopathy and its potential uses in plant biology and forest sustainability.

Plants are integral components of ecosystems and key sources of food, medicine, and other resources for human societies. The interactions between micro(nano)plastics and plants have garnered significant attention in recent years due to the pervasive nature of plastic pollution and its potential impact on terrestrial and aquatic ecosystems. This study aims to analyze the current understanding, critical knowledge gaps and future perspectives on the interactions between plants and plastic residues, including microplastics, nanoplastics, microfiber, and microbeads. Data was gathered from the Web of Science Core Collection database, with 1049 documents indexed from 2009 to 2023 for further analysis. Co-citation analysis combined with co-word network analysis was utilized. The findings indicate a notable increase in publication productivity on plastic-plant interactions over the past decade, with China, India, Italy, Korea, and Spain as the core research countries in the field. Chinese universities and research institutions, particularly Naikai University and the Chinese Academy of Sciences, are the major research drivers. Weitao Liu from Naikai University was the most productive author up to 2023. Science of the Total Environment, Environmental Pollution, and Journal of Hazardous Materials were the top three journal that published the most articles. The most frequently cited article titled “Microplastics can change soil properties and affect plant performance” published in Environmental Science & Technology in 2019. The co-citation network highlights the interconnectedness of plant-plastic interactions, while burst analysis and thematic mapping suggest that future research will focus on the impact of emerging contaminants like microplastics and nanoplastics on soil health in the plastisphere. More long-scale and long-term interdisciplinary studies including plant species and polymer types at field condition are needed to a better understanding the plant-plastic interactions. This study offers a thorough and unbiased real-time analysis of plant-plastic interactions, highlighting current trends and outlining future research directions and priorities.

With the popularization and application of auxiliary transportation devices such as diesel monorail cranes and trackless rubber-wheeled vehicles in underground coal mines, the frequency of transporting and refilling diesel fuel in underground mines is getting higher and higher, leakage of diesel fuel and volatile diesel vapor during transportation and refueling will pollute the underground air environment, and may cause combustion and explosion accidents in serious cases. In this paper, for the environmental characteristics of diesel transportation and refueling process in underground coal mine, we constructed the evaporation and diffusion model of leaked diesel in underground fluid environment, established the prediction method of peak diesel vapor volume fraction based on wind speed and evaporation rate, investigated the distribution law of diesel vapor volume fraction under the conditions of different ambient wind speeds and diesel evaporation rates, and put forward fire prevention and explosion-proof measures for the mixed diesel vapor-gas environment in underground coal mine. The results show that in the underground flow environment, diesel vapor diffusion presents obvious subsiding, layering and segmentation characteristics, and the wind flow has a promoting effect on diesel vapor diffusion. When the diesel was leaking for 20 s, the vapour spread to 10 m downwind of the leak, and when the diesel was leaking for 40 s, the vapour spread to 15 m downwind of the leak, and when the diesel was leaking for 70 s, the volume fraction of diesel vapour in the chamber reached a peak and tended to stabilize, and the peak volume fraction of diesel vapour was located at a point 1 m downwind of the leak. Measures such as lowering the ambient temperature of the chamber, reducing the leakage area and increasing the air velocity can be taken to prevent the diesel vapour-gas mixture from burning and exploding.

Objective Urban rail transit generates a considerable amount of energy consumption during operation, with traction energy consumption of train operation offering significant optimization potential. Therefore, there is an urgent need to study a train traction energy-saving control strategy. Method Firstly, a dynamic equation of train operation is established, and the constraint conditions for the train operation process are defined. Secondly, the optimization strategy for train traction energy consumption is decomposed to establish the objective function.Based on the genetic algorithm model, the allocation of train operation energy consumption in different intervals and the optimal cruising speed within intervals are solved. Then, through the cross mutation process of the genetic algorithm, the optimal energy-saving effect of the driving speed recommendation curve is obtained. Finally, a simulation model is built using Matlab software, with real-line train parameters and operational data to simulate the inter-station operation process of the train. Result & Conclusion Experimental results show that the control strategy based on the genetic algorithm model, compared to the conventional fixed working condition sequence control strategies, significantly improves traction energy-saving indicators.

Abstract Background Advanced unresectable gastric cancer (GC) patients were previously treated with chemotherapy alone as the first-line therapy. However, with the Food and Drug Administration’s (FDA) 2022 approval of programmed cell death protein 1 (PD-1) inhibitor combined with chemotherapy as the first-li ne treatment for advanced unresectable GC, patients have significantly benefited. However, the significant costs and potential adverse effects necessitate precise patient selection. In recent years, the advent of deep learning (DL) has revolutionized the medical field, particularly in predicting tumor treatment responses. Our study utilizes DL to analyze pathological images, aiming to predict first-line PD-1 combined chemotherapy response for advanced-stage GC. Methods In this multicenter retrospective analysis, Hematoxylin and Eosin (H&E)-stained slides were collected from advanced GC patients across four medical centers. Treatment response was evaluated according to iRECIST 1.1 criteria after a comprehensive first-line PD-1 immunotherapy combined with chemotherapy. Three DL models were employed in an ensemble approach to create the immune checkpoint inhibitors Response Score (ICIsRS) as a novel histopathological biomarker derived from Whole Slide Images (WSIs). Results Analyzing 148,181 patches from 313 WSIs of 264 advanced GC patients, the ensemble model exhibited superior predictive accuracy, leading to the creation of ICIsNet. The model demonstrated robust performance across four testing datasets, achieving AUC values of 0.92, 0.95, 0.96, and 1 respectively. It is well known that there are several vertical flight phases for the aircraft in the flight. The cruise flight phase takes most of the flight time, flight distance and fuel consumption, so it is important and necessary to develop the flight management system(FMS) vertical trajectory prediction algorithm for the cruise flight phase, aiming to improve the economy, comfort and safety of the flight. In order to meet the versatility of the vertical trajectory prediction algorithm between different types of aircraft in the cruise phase and improve the accuracy and reliability of the vertical trajectory prediction, the algorithm of vertical trajectory prediction for cruise flight phase is proposed in this paper. Firstly, the cruise speed profile is calculated, and the realistic atmospheric model of prediction is constructed. Then the cruise fuel flow data is calculated based on the aircraft model of first-principles, and the vertical trajectory prediction for the cruise phase is given by designing the vertical trajectory prediction algorithm logic. Finally, the effectiveness and accuracy of the algorithm are verified by ground simulation and air flight test. The results show that the vertical trajectory prediction algorithm of FMS cruise phase based on the aircraft model of firstprinciples can predict the cruise trajectory of the aircraft, and the prediction accuracy error is less than 1%.

Abstract Escherichia coli Nissle 1917 (EcN) is an important chassis strain widely used for the development of live biotherapeutic products (LBPs). EcN strain naturally harbors two cryptic plasmids with unknown function. During the development of LBPs using EcN strain, the cryptic plasmids were cured usually to avoid plasmid incompatibility or alleviate metabolic burdens associated with these cryptic plasmids. While the cryptic plasmids curing in EcN may appear to be a routine procedure, the comprehensive impact of cryptic plasmids curing on the EcN strain remains incompletely understood. In the present study, the effects of cryptic plasmids curing on EcN were investigated using transcriptome sequencing. The results revealed that only a small number of genes showed significant changes in mRNA levels after cryptic plasmid curing (4 upregulated and 6 downregulated genes), primarily involved in amino acid metabolism. Furthermore, the flu gene showed the most significant different expression, encoding Antigen 43 (Ag43) protein, a Cah family adhesin. Mass spectrometry analysis further confirmed the significant increase in Ag43 expression. Ag43 is commonly present in Escherichia coli and mediates the bacterial autoaggregation. However, despite the upregulation of Ag43 expression, no Ag43-mediated cell self-sedimentation was observed in the cured EcN strain. These findings contribute to making informed decisions regarding the curing of the cryptic plasmids when Escherichia coli Nissle 1917 is used as the chassis strain.

BackgroundRosmarinic acid (RA), a natural phenolic acid, exhibits promising anti-cancer properties. The abnormal expression of microRNA (miRNA) regulates the gene expression and plays a role as an oncogenic or tumor suppressor in TNBC. However, the biological role of RA in miR-30a-5p on BCL2L11 during MDA-MB-231 induced breast cancer stem-like cells (BCSCs) progression and its regulatory mechanism have not been elucidated.ObjectiveTo investigate whether RA inhibited the silencing effect of miR-30a-5p on the BCL2L11 gene and promoted apoptosis in BCSCs.Materials and MethodsWe assessed the migration, colony formation, proliferation, cell cycle, and apoptosis of BCSCs after RA treatment using the wound-healing assay, colony formation assay, CCK-8 assay, and flow cytometry, respectively. The expression of mRNA and protein levels of BCL-2, Bax, BCL2L11, and P53 genes in BCSCs after RA treatment was obtained by real-time polymerase chain reaction and Western blot. Differential miRNA expression in BCSCs was analyzed by high-throughput sequencing. Targetscan was utilized to predict the targets of miR-30a-5p. The dual luciferase reporter system was used for validation of the miR-30a-5p target.ResultsWound-healing assay, colony formation assay, CCK-8 assay, and cell cycle assay results showed that RA inhibited migration, colony formation and viability of BCSCs, and cell cycle arrest in the G0-G1 phase. At the highest dose of RA, we noticed cell atrophy, while the arrest rate at 100 μg/mL RA surpassed that at 200 μg/mL RA. Apoptotic cells appeared early (Membrane Associated Protein V FITC+, PI−) or late (Membrane Associated Protein V FITC+, PI+) upon administration of 200 μg/mL RA, Using high-throughput sequencing to compare the differences in miRNA expression, we detected downregulation of miR-30a-5p expression, and the results of dual luciferase reporter gene analysis indicated that BCL2L11 was a direct target of miR-30a-5p.ConclusionRA inhibited the silencing effect of miR-30a-5p on the BCL2L11 gene and enhanced apoptosis in BCSCs.

Drawing on two theoretical frameworks–positive psychology theory and conservation of resources theory–the relationship and underlying mechanism between team-level service-oriented human resource practice (SO-HRP) bundles and team performance were explored by constructing a team-level serial mediation model. Survey data were collected from 424 employees at 80 branches of five financial holding companies in Northern Taiwan. The team-level serial-mediated model and hypotheses were examined using SPSS version 24 and the PROCESS macro for SPSS. The results indicate that team psychological capital (PsyCap) and team work engagement serially mediated the relationship between SO-HRP bundles and team performance. These results imply that managers should plan and implement SO-HRP bundles in detail, reinforce team PsyCap, and increase engagement within their financial service teams to enhance team performance.

Radiation is one of the standard therapies for pediatric high-grade glioma (pHGG), of which the prognosis remains poor. To gain an in-depth understanding of biological consequences beyond the classic DNA damage, we treated 9 patient-derived orthotopic xenograft (PDOX) models, including one with DNA mismatch repair (MMR) deficiency, with fractionated radiations (2 Gy/day x 5 days). Extension of survival time was noted in 5 PDOX models (P < 0.05) accompanied by γH2AX positivity in >95 % tumor cells in tumor core and >85 % in the invasive foci as well as ∼30 % apoptotic and mitotic catastrophic cell death. The model with DNA MMR (IC-1406HGG) was the most responsive to radiation with a reduction of Ki-67(+) cells. Altered metabolism, including mitochondria number elevation, COX IV activation and reactive oxygen species accumulation, were detected together with the enrichment of CD133+ tumor cells. The latter was caused by the entry of quiescent G0 cells into cell cycle and the activation of self-renewal (SOX2 and BMI1) and epithelial mesenchymal transition (fibronectin) genes. These novel insights about the cellular and molecular mechanisms of fractionated radiation in vivo should support the development of new radio-sensitizing therapies.

Sea cucumber (Apostichopus japonicus) is a significant aquatic economic species globally. However, the advancement of the sea cucumber aquaculture industry is impeded by its high susceptibility to elevated temperatures. To elucidate the impact of high temperature on physiological changes, gene expression, and metabolic responses in sea cucumbers, we conducted transcriptional profiling of A. japonicus under heat stress. Our results revealed 3691 differentially expressed genes (DEGs), comprising 1629 up-regulated and 2062 down-regulated genes, between the control and treatment groups. Analysis of the Kyoto Encyclopedia of Genes and Genomes (KEGG), Gene Ontology (GO), and Protein-Protein Interaction Networks (PPI) indicated that high-temperature stress triggered the expression of stress defense-related genes while inhibiting the expression of genes associated with development and metabolism. Furthermore, 30 out of 39 DEGs were annotated to heat shock protein genes, underscoring the crucial role of these genes in the regulatory mechanism of A. japonicus in response to heat stress. In summary, our findings provide fundamental insights for further investigation into the molecular mechanisms in A. japonicus under high-temperature stress and may contribute to the development of novel strategies for identifying heat-tolerant candidate genes through the analysis of their transcriptional changes in the future.

Abstract Strengthening aquatic resilience to prevent adverse shifts is critical for preserving global freshwater biodiversity and advancing Sustainable Development Goals. Nonetheless, understanding the long‐term trends and underlying causes of lake ecosystem resilience at a global scale remains elusive. Here, we employ an innovative framework, integrating satellite‐derived water quality indices with early warning signals and machine learning techniques, to investigate the dynamics of resilience in 1,049 lakes worldwide during 2000–2018. Our results indicate that 46.7% of lakes are experiencing a significant decline in resilience, particularly since the early 2010s, closely associated with higher human population density and anthropogenic eutrophication. In contrast, most lakes situated in alpine regions exhibit an increase in resilience, probably benefiting from climate warming and wetting. Together, this study provides a novel way to monitor lake resilience and predict undesired transitions, and reveals a widespread erosion in the ability of lakes to withstand stressors associated with global change.

Abstract Background Streptomyces lincolnensis is well known for producing the clinically important antimicrobial agent lincomycin. The synthetic and regulatory mechanisms on lincomycin biosynthesis have been deeply explored in recent years. However, the regulation involved in primary metabolism have not been fully addressed. Results SLCG_7083 protein contains a Per-Arnt-Sim (PAS) domain at the N-terminus, whose homologous proteins are highly distributed in Streptomyces. The inactivation of the SLCG_7083 gene indicated that SLCG_7083 promotes glucose utilization, slows mycelial growth and affects sporulation in S. lincolnensis. Comparative transcriptomic analysis further revealed that SLCG_7083 represses eight genes involved in sporulation, cell division and lipid metabolism, and activates two genes involved in carbon metabolism. Conclusions SLCG_7083 is a PAS domain-containing regulator on morphological development and glucose utilization in S. lincolnensis. Our results first revealed the regulatory function of SLCG_7083, and shed new light on the transcriptional effects of SLCG_7083-like family proteins in Streptomyces.

Abstract Background The role of cholesterol metabolism in gastric cancer (GC) and its implications for tumor characteristics and immunotherapy response remain poorly understood. In this study, our aim was to investigate this role, identify associated metabolic subtypes, and assess their clinical implications in GC. Methods We conducted a comprehensive analysis of cholesterol metabolism genes (CMGs) using transcriptomic data from TCGA and GEO. Based on 23 representative CMGs, we classified GC into metabolic subtypes. We evaluated clinical features and immune cell infiltration between these subtypes. Additionally, we identified a CMG signature and assessed its clinical relevance in GC. We retrospectively enrolled thirty-five GC patients receiving chemotherapy plus a PD-1 inhibitor to assess the CMG signature using multiplex immunohistochemistry. Results Our analysis revealed two cholesterol metabolism subtypes in GC: Cholesterol Metabolism Type 1 (CMT1) and Cholesterol Metabolism Type 2 (CMT2). These subtypes exhibited distinct patterns: CMT1 indicated heightened cholesterol biosynthesis, while CMT2 showed abnormal cholesterol transport. CMT2 was associated with unfavorable clinical features, enriched malignant pathways, and a pro-tumor immune microenvironment. Furthermore, we developed a five-CMG prognostic signature (ABCA1, NR1H3, TSPO, NCEH1, and HMGCR) that effectively predicted the prognosis of patients with GC and their response to chemotherapy plus a PD-1 inhibitor. This signature was validated in a clinical cohort using multiplex immunohistochemistry. Conclusion Our results highlight the effectiveness of cholesterol metabolism patterns as biomarkers for predicting the prognosis and immunotherapy response in GC. The expression of cholesterol metabolism genes and the assessment of cholesterol metabolism patterns have the potential to predict the outcome of immunotherapy and guide treatment strategies.

Summary: We evaluated the diagnostic performance of a multimodal deep-learning (DL) model for ovarian mass differential diagnosis. This single-center retrospective study included 1,054 ultrasound (US)-detected ovarian tumors (699 benign and 355 malignant). Patients were randomly divided into training (n = 675), validation (n = 169), and testing (n = 210) sets. The model was developed using ResNet-50. Three DL-based models were proposed for benign-malignant classification of these lesions: single-modality model that only utilized US images; dual-modality model that used US images and menopausal status as inputs; and multi-modality model that integrated US images, menopausal status, and serum indicators. After 5-fold cross-validation, 210 lesions were tested. We evaluated the three models using the area under the curve (AUC), accuracy, sensitivity, and specificity. The multimodal model outperformed the single- and dual-modality models with 93.80% accuracy and 0.983 AUC. The Multimodal ResNet-50 DL model outperformed the single- and dual-modality models in identifying benign and malignant ovarian tumors.

Skin ulceration syndrome of sea cucumbers is one of the most serious diseases in intensive aquaculture, and it is the most effective way of preventing the spread of this disease to detect the abnormal behavior of sea cucumbers in time and take corresponding measures. However, the detection and tracking of multi-object is a hard problem in sea cucumber behavior analysis. To solve this problem, this paper first proposes a novel one-stage algorithm SUS-YOLOv5 for multi-object detection and tracking of sea cucumbers. The proposed SUS-YOLOv5 optimizes the maximum suppression algorithm in the overlapping region of the object detection box. Next, the SE-BiFPN feature fusion structure is proposed to enhance the transmission efficiency of feature information between deep and shallow layers of the network. Then, a MO-Tracking algorithm is proposed integrated with DeepSORT to achieve real-time multi-object tracking. Experimental results show that the mAP@0.5 and mAP@0.5:0.95 of the proposed object detector reach 95.40% and 83.80%, respectively, which are 3.30% and 4.10% higher than the original YOLOv5s. Compared with the traditional SSD, YOLOv3, and YOLOv4, the mAP of SUS-YOLOv5 is improved by 5.49%, 1.57%, and 3.76%, respectively. This research can realize the multi-object detection and tracking, which lays the foundation for the prediction of skin ulceration syndrome in sea cucumbers and has a certain practical application value for improving the intelligence level of aquaculture.

BackgroundThis study aimed to investigate the effects of adjuvant beam radiation therapy (ABRT) on overall survival (OS) in patients with primary single intracranial atypical meningioma (AM), with a focus on age-related outcomes.MethodsWe conducted a retrospective study using data from SEER database. Our cohort consisted of patients diagnosed with a primary single intracranial AM tumor and had undergone surgery. The primary endpoint was OS. For survival analysis, univariable and multivariable Cox regression analysis were performed. A multivariable additive Cox model was used to assess the functional relationship between age and OS in patients with or without ABRT.ResultsOf the 2,759 patients included, 1,650 underwent gross total resection and 833 received ABRT. Multivariable Cox analysis indicated that ABRT did not significantly influence OS across the entire cohort. According to the multivariable generalized additive Cox model, the relative risk of all-cause mortality increased with advancing age in both ABRT-yes and ABRT-no group. ABRT-yes had a lower relative risk than ABRT-no when age ≤ 55 years old while a higher relative risk when age > 55 years old. Subsequent multivariable Cox analysis showed that ABRT was associated with a significant lower risk for all-cause mortality in patients with age ≤ 55 years old while a significant higher risk in patients with age > 55 years old.ConclusionOur study found that ABRT enhanced OS in younger primary single intracranial AM patients. But we also revealed a negative correlation between OS and ABRT in older patients.

Abstract Background Gastrointestinal Neuroendocrine Neoplasms (GI-NENs) often result in liver metastases, and the role of Primary Tumor Resection (PTR) in managing GI-NENs with liver metastases (GI-NENLM) is still debated. This study aimed to investigate the potential benefits of PTR in treating GI-NENLM by analyzing data from the Surveillance, Epidemiology, and End Results Program (SEER) and the First Affiliated Hospital of Sun Yat-sen University (FAH). Methods The SEER Registry 17 database and the FAH clinical pathology database were used to collect clinicopathology data for GI-NENLM diagnosed between 2010 and 2019 and between 2011 and 2022, respectively. Propensity score matching (PSM) was used to match the clinicopathological characteristics of patients from both cohorts. Inverse probability weighting (IPTW) was used to weigh the PTR and non-PTR groups. The primary endpoint was overall survival (OS). Results After matching, 155 patients from the SEER database were matched to the FAH cohort. PTR was significantly associated with better prognosis in PSM-matched/unmatched SEER cohorts (P

Urban rail transit plays a very important role in cities’ social and economic development. To ensure the safe and stable operation of urban rail transit operation equipment and facilities, it is necessary to monitor a large number of safety hazard statuses and data and improve the over-centralisation of traditional monitoring. This paper designs a scheme for storing, validating and monitoring the safety hazard status of urban rail transit operation equipment and facilities based on blockchain technology. The safety hazards of equipment and facilities during the operation stage of urban rail transit are listed using the literature analysis method and the case study method. The European RAMS (reliability, availability, maintainability and safety) standard method is used to determine the safety hazard status of equipment and facilities by availability index. Based on the features of the consensus mechanism, smart contract and other features of blockchain technology, this paper designs an overall scheme for storing, verifying and monitoring the safety hazard status of equipment and facilities. This scheme provides a practical operation method for evaluating the safety hazard status of rail transit equipment and facilities, which is conducive to the safety rectification of the entire urban rail transit.

Abstract Objectives Chronic obstructive pulmonary disease (COPD) is one of the main causes of morbidity and mortality in the world. However, there are some patients who are not diagnosed early and correctly through routine methods because of inconspicuous or serious symptoms. This study aims to assess the diagnostic role of long non-coding RNA (lncRNA) in COPD. Methods We searched literature from electronic databases, after excluding non-COPD literature, the bibliometric analysis was performed, and VOSviewer software was used to represent the data analyzed. Literature evaluating the diagnostic test accuracy of lncRNA for COPD was eligible, and the QUADAS-2 checklist was used to evaluate the quality. The pooled sensitivity (SEN), specificity (SPE), diagnostic odds ratio (DOR), and summary receiver operating characteristic curve (sROC) were used to analyze the overall diagnostic performance. Subgroup and meta-regression analyses were performed to explore the heterogeneity, and a funnel plot was assessed for publication bias. Also, lncRNAs related to COPD were identified and explored for their potential biological function. Results An increased annual growth rate of literature on this subject from 2016 focused on COPD, humans, RNA, and lncRNA. The meta-analysis enrolled 17 literature indicated that the SEN, SPE, and DOR differentiating COPD patients from normal controls (NCs) were 0.86 (95% CI [0.80, 0.90]), 0.78 (95% CI [0.67, 0.86]), and 21.59 (95% CI [11.39, 40.91]), respectively. Meanwhile, lncRNAs had the ability to distinguish acute exacerbations of COPD (AECOPD) patients from COPD; the SEN, SPE, and DOR were 0.75 (95% CI [0.62, 0.85]), 0.81 (95% CI [0.71, 0.89]), and 13.02 (95% CI [7.76, 21.85]), respectively. The area under the sROC were calculated to be greater than 0.8 at least. Subgroup and meta-regression analysis showed that the types of specimens and dysregulated lncRNAs might affect the diagnostic accuracy. The funnel plot showed there was a certain publication bias. 41 lncRNAs related to COPD were identified and mainly located in the nucleus and cytoplasm, associated with proliferation, invasion, and prognosis. These lncRNA-binding proteins were involved in the spliceosome, Rap1 signaling pathway, MAPK signaling pathway, and so on. Conclusion LncRNA suggests potential diagnostic biomarkers and therapeutic targets for COPD patients.

In this study, a liquid chromatographic method was developed for the fast determination of lincomycin, polymyxin and vancomycin in a preservation solution for transplants. A Kinetex EVO C18 (150 × 4.6 mm, 2.6 µm) column was utilized at 45 °C. Gradient elution was applied using a mixture of mobile phases A and B, both including 30 mM phosphate buffer at pH 2.0 and acetonitrile, at a ratio of 95:5 (v/v) for A and 50:50 (v/v) for B. A flow rate of 1.0 mL/min, an injection volume of 20 µL and UV detection at 210 nm were used. A degradation study treating the three antibiotics with 0.5 M hydrochloric acid, 0.5 M sodium hydroxide and 3% H2O2 indicated that the developed method was selective toward lincomycin, polymyxin, vancomycin and their degradation products. Other ingredients of the preservation solution, like those from the cell culture medium, did not interfere. The method was validated with good sensitivity, linearity, precision and accuracy. Furthermore, lincomycin, polymyxin and vancomycin were found to be stable in this preservation solution for 4 weeks when stored at −20 °C.

The coastal Casuarina equisetifolia is the most common tree species in Hainan’s coastal protection forests. Sequencing the genomes of its allelopathic endophytes can allow the protective effects of these bacteria to be effectively implemented in protected forests. The goal of this study was to sequence the whole genomes of the endophytes Bacillus amyloliquefaciens and Bacillus aryabhattai isolated from C. equisetifolia root tissues. The results showed that the genome sizes of B. amyloliquefaciens and B. aryabhattai were 3.854 Mb and 5.508 Mb, respectively. The two strains shared 2514 common gene families while having 1055 and 2406 distinct gene families, respectively. The two strains had 283 and 298 allelochemical synthesis-associated genes, respectively, 255 of which were shared by both strains and 28 and 43 of which were unique to each strain, respectively. The genes were putatively involved in 11 functional pathways, including secondary metabolite biosynthesis, terpene carbon skeleton biosynthesis, biosynthesis of ubiquinone and other terpene quinones, tropane/piperidine and piperidine alkaloids biosynthesis, and phenylpropanoid biosynthesis. NQO1 and entC are known to be involved in the biosynthesis of ubiquinone and other terpenoid quinones, and rfbC/rmlC, rfbA/rmlA/rffH, and rfbB/rmlB/rffG are involved in the biosynthesis of polyketide glycan units. Among the B. aryabhattai-specific allelochemical synthesis-related genes, STE24 is involved in terpene carbon skeleton production, atzF and gdhA in arginine biosynthesis, and TYR in isoquinoline alkaloid biosynthesis. B. amyloliquefaciens and B. aryabhattai share the genes aspB, yhdR, trpA, trpB, and GGPS, which are known to be involved in the synthesis of carotenoids, indole, momilactones, and other allelochemicals. Additionally, these bacteria are involved in allelochemical synthesis via routes such as polyketide sugar unit biosynthesis and isoquinoline alkaloid biosynthesis. This study sheds light on the genetic basis of allelopathy in Bacillus strains associated with C. equisetifolia, highlighting the possible use of these bacteria in sustainable agricultural strategies for weed management and crop protection.

Abstract Diffuse large B-cell lymphoma (DLBCL) is the most common lymphoma in adults. This study aimed to determine the prognostic significance of endoplasmic reticulum (ER) stress-related genes in DLBCL. ER stress-related genes were obtained from the molecular signatures database. Gene expression data and clinical outcomes from the gene expression omnibus and TCGA datasets were collected, and differentially expressed genes (DEGs) were screened out. Gene ontology enrichment analysis, the kyoto encyclopaedia of genes and genomes pathway analysis, and geneset enrichment analysis were used to analyse the possible biological function of ER stress-related DEGs in DLBCL. Protein–protein interaction network construction using the STRING online and hub genes were identified by cytoHubba on Cytoscape software. The significant prognosis-related genes were screened, and the differential expression was validated. The immune microenvironment assessment of significant genes were evaluated. Next, the nomogram was built using univariate and multivariate Cox regression analysis. 26 ER stress-related DEGs were screened. Functional enrichment analysis showed them to be involved in the regulation of the endoplasmic reticulum mainly. NUPR1 and TRIB3 were identified as the most significant prognostic-related genes by comparison with the GSE10846, GSE11318, and TCGA datasets. NUPR1 was correlated with a good prognosis and immune infiltration in DLBCL; on the other hand, high expression of TRIB3 significantly correlated with a poor prognosis, which was an independent prognostic factor for DLBCL. In summary, we identified NUPR1 and TRIB3 as critical ER stress-related genes in DLBCL. NUPR1 might be involved in immune infiltration in DLBCL, and TRIB3 might serve as a potential therapeutic target and prognostic factor in DLBCL.

ObjectiveTo explore epidemiological characteristics and re-positivity incidence of imported and local infections with severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) Delta variant during a period of coronavirus disease 2019 (COVID-19) epidemic in Fujian province. MethodsFrom National Information System of Disease Prevention and Control and Information System of Public Health Emergency Management, we extracted the information on 69 genome sequencing-confirmed imported infections and 264/212 genome sequencing-/epidemiologically confirmed local infections with SARS-CoV-2 Delta variant registered in Fujian province from March 2021 through February 2022. Descriptive statistics was used to analyze disease characteristics and incubation period of the SARS-CoV-2 Delta variant infections. The 'R0' and 'EpiEstim' packages of R software were used to estimate the serial interval (SI) and the time-dependent reproduction number (Rt). Influencing factors of the re-positivity were analyzed with logistic regression.ResultsThe period of COVID-19 epidemic induced by SARS-CoV-2 Delta variant was approximately from March 2021 to February 2022 in Fujian province, with a continuous incidence of sporadic imported cases and local outbreak of the infections. The majority of the imported cases were seafarers (28.99%), students (15.94%) and commercial service personnel (14.49%) from Southeast Asia, Japan, the United States, the United Kingdom and other regions and entered Fujian province via Xiamen Customs. The local epidemics due to rapid transmission of Delta variant occurred mainly in schools and factories in Putian and Xiamen city. The SI of the local infections was fitted to Gamma distribution, with the mean and standard deviation of 2.28 and 2.06, respectively. For the 149 infector-infectee pairs of local infections during the local outbreak，the maximum value of Rt was 7.35 and the median incubation period was 6.50 days. The incidence of re-positivity was 37.98% for all the Delta variant infections. The results of multivariate regression analysis revealed that the local infections with Delta variant were at a significantly higher risk of re-positivity ( odds ratio = 2.47, 95% confidence interval: 1.25 – 4.87) compared to the imported infections. No associates of gender, age, occupation and clinical type with the re-positivity were observed. ConclusionThe period of COVID-19 epidemic due to SARS-CoV-2 Delta variant infections in Fujian province was approximately from March 2021 to February 2022, with continuously sporadic imported and local infections and the local epidemic was under effective control rapidly. The incidence rate of re-positivity was 37.98% among all infections with Delta variant.

Polyvinyl alcohol (PVA) hydrogel is favored by researchers due to its good biocompatibility, high mechanical strength, low friction coefficient, and suitable water content. The widely distributed hydroxyl side chains on the PVA molecule allow the hydrogels to be branched with various functional groups. By improving the synthesis method and changing the hydrogel structure, PVA-based hydrogels can obtain excellent cytocompatibility, flexibility, electrical conductivity, viscoelasticity, and antimicrobial properties, representing a good candidate for articular cartilage restoration, electronic skin, wound dressing, and other fields. This review introduces various preparation methods of PVA-based hydrogels and their wide applications in the biomedical field.

BackgroundS100A8, a calcium-binding protein belonging to the S100 family, is involved in immune responses and multiple tumor pathogens. Diffuse large B-cell lymphoma (DLBCL) is one of the most common types of B-cell lymphoma and remains incurable in 40% of patients. However, the role of S100A8 and its regulation of the immune response in DLBCL remain unclear.MethodsThe differential expression of S100A8 was identified via the GEO and TCGA databases. The prognostic role of S100A8 in DLBCL was calculated using the Kaplan-Meier curve. The function enrichment of differentially expressed genes (DEGs) was explored through GO, KEGG, GSEA, and PPI analysis. In our cohort, the expression of S100A8 was verified. Meanwhile, the biological function of S100A8 was applied after the inhibition of S100A8 in an in vitro experiment. The association between S100A8 and immune cell infiltration and treatment response in DLBCL was analyzed.ResultsS100A8 was significantly overexpressed and related to a poor prognosis in DLBCL patients. Function enrichment analysis revealed that DEGs were mainly enriched in the IL-17 signaling pathway. Our cohort also verified this point. In vitro experiments suggested that inhibition of S100A8 should promote cell apoptosis and suppress tumor growth. Single-cell RNA sequence analysis indicated that S100A8 might be associated with features of the tumor microenvironment (TME), and immune infiltration analyses discovered that S100A8 expression was involved in TME. In terms of drug screening, we predicted that many drugs were associated with preferable sensitivity.ConclusionElevated S100A8 expression is associated with a poor prognosis and immune infiltration in DLBCL. Inhibition of S100A8 could promote cell apoptosis and suppress tumor growth. Meanwhile, S100A8 has the potential to be a promising immunotherapeutic target for patients with DLBCL.

Peptides are widely used as natural bio-small molecules because of their various pharmacological activities such as enhancing immunity, promoting wound healing, and improving inflammation. Alcoholic heart injury has become one of the major health problems worldwide, and alcohol consumption is now the main cause of alcoholic cardiomyopathy. In this study, deer heart peptides were extracted from deer hearts by enzymatic digestion and the antioxidant activity of deer heart peptides extracted at different times was evaluated by three in vitro antioxidant methods, and the active peptide with the best enzymatic effect has been selected for in vivo animal experiments. The anti-inflammatory and antioxidant properties of deer heart enzymatic extracts were evaluated in in vivo experiments in mice. In this study, mice were orally gavaged with white wine (12 mL/kg body weight) to induce a mouse model of cardiac injury, while mice were orally administered a single dose of 100 mg/kg/bw and 200 mg/kg/bw of deer heart enzyme digest and were examined for body weight, dietary intake, water intake, and coat gloss, as well as for general behaviors, adverse effects, and mortality. Histology, serum, anti-inflammatory factors, and oxidative stress parameters were subsequently assessed. In all modeled mice, no four-way or any significant behavioral changes were observed in all groups, but in the modeled group, mice showed weight loss, decreased diet and water intake, and decreased cardiac index. For in vivo tests, the extract inhibited the anti-inflammatory activity with a significant decrease in inflammatory factors of TNF-α, IL-6, and IL-1β in cardiac tissues, a significant increase in serum levels of both CAT and SOD, an increase in MDA content, and a remarkable increase in the level of the marker CK in the cardiac myocardial enzyme profile. Significant improvement in myocardial disorders by deer heart peptide could be observed from heart tissue sections. The present study emphasizes the anti-inflammatory and antioxidant activity of deer heart peptide, an enzymatic digest of deer heart, which provides empirical as well as supportive role for the anti-inflammatory properties of traditional medicine.

A single-row plasma tube array (PTA) experimental system is established to improve the backscattering attenuation of a metal plate covered by a plasma tube array. The backscattering test system is utilized in a microwave anechoic chamber to examine the effects of gas composition, pressure, tube diameter, and discharge power on the backscattering attenuation of a metal plate using a plasma tube array. The electron density is obtained via microwave diagnosis. The backscattering attenuation mechanism in different frequency bands is revealed via numerical simulation. The results show that the reasonable selection of PTA parameters achieves strong attenuation in different frequency bands. The strong attenuation frequency bands of Ar–Hg PTA are in low frequency (1.5–3.5 GHz) and high frequency (13–17 GHz), while that of Ne–Hg discharge is in medium frequency (6.4–11.7 GHz). When the pressure is 0.5 and 1 Torr, the PTA shows a low, medium, and high multi-band distribution for the backscattering strong attenuation region. The backscattering strong attenuation region shows a low and high dual-band distribution, while the pressure is 2–4 Torr. As the tube diameter increases, the strong attenuation region maintains the dual-band, but it changes from low and high frequency bands to medium frequency (6-12 GHz), where the backscattering attenuation mechanism is collisional absorption when the frequency of plasma electron oscillation is close to that of electro-magnetic waves. The backscattering attenuation mechanism in the low frequency band involves the periodic structure of PTA generating local surface plasmon to absorb electromagnetic waves.

BackgroundDiffuse large B-cell lymphoma (DLBCL) is the most common B-cell lymphoma in adults. CDGSH iron sulfur domain 2 (CISD2) is an iron–sulfur protein and plays a critical role of cell proliferation. The aberrant expression of CISD2 is associated with the progression of multiple cancers. However, its role in DLBCL remains unclear.MethodsThe differential expression of CISD2 was identified via public databases, and quantitative real-time PCR (qRT-PCR) and western blot were used to identifed the expression of CISD2. We estimated the impact of CISD2 on clinical prognosis using the Kaplan-Meier plotter. Meanwhile, the drug sensitivity of CISD2 was assessed using CellMiner database. The 100 CISD2-related genes from STRING obtained and analyzed using the LASSO Cox regression. A CISD2 related signature for risk model (CISD2Risk) was established. The PPI network of CISD2Risk was performed, and functional enrichment was conducted through the DAVID database. The impacts of CISD2Risk on clinical features were analyzed. ESTIMATE, CIBERSORT, and MCP-counter algorithm were used to identify CISD2Risk associated with immune infiltration. Subsequently, Univariate and multivariate Cox regression analysis were applied, and a prognostic nomogram, accompanied by a calibration curve, was constructed to predict 1-, 3-, and 5-years survival probabilities.ResultsCISD2 was upregulated in DLBCL patients comparing with normal controls via public datasets, similarly, CISD2 was highly expressed in DLBCL cell lines. Abstract Triple-negative breast cancer (TNBC) is a subtype of breast cancer with poor outcome and lacks of approved targeted therapy. Overexpression of epidermal growth factor receptor (EGFR) is found in more than 50% TNBC and is suggested as a driving force in progression of TNBC; however, targeting EGFR using antibodies to prevent its dimerization and activation shows no significant benefits for TNBC patients. Here we report that EGFR monomer may activate signal transducer activator of transcription-3 (STAT3) in the absence of transmembrane protein TMEM25, whose expression is frequently decreased in human TNBC. Deficiency of TMEM25 allows EGFR monomer to phosphorylate STAT3 independent of ligand binding, and thus enhances basal STAT3 activation to promote TNBC progression in female mice. Moreover, supplying TMEM25 by adeno-associated virus strongly suppresses STAT3 activation and TNBC progression. Hence, our study reveals a role of monomeric-EGFR/STAT3 signaling pathway in TNBC progression and points out a potential targeted therapy for TNBC.

Unsteady aerodynamic prediction at high angles of attack is of great importance to the design and development of advanced fighters. In this paper, a weighted feature fusion model (WFFM) that combines the state-space model and neural networks is proposed to build an unsteady aerodynamic model for the precise simulation and control of post-stall maneuvers. In the proposed model, the influences of the physical model on neural networks are considered and adjusted by introducing a standardization layer and a new weighting method. A long short-term memory (LSTM) network is used to fuse two mappings: one from flight states to aerodynamic loads, and the other from low-fidelity data to high-fidelity data. Data from wind tunnel oscillation experiments at high angles of attack using a new kind of wire-driven parallel robot and the traditional tail support are used for verifying the proposed aerodynamic model. The output of the WFFM is also compared with predictions from other models, such as the state-space model, single LSTM model, and feature fusion model not including a feature weighting layer. Results demonstrate improved accuracy of the proposed model in the interpolation and extrapolation tests. Furthermore, the WFFM is applied to the flight simulation of F-16 with different control inputs. Compared with conventional models, the WFFM shows improved accuracy and better generalization capability.

Marine anti-fouling coatings represent an efficient approach to prevent and control the marine biofouling. However, a significant amount of antifouling agent is added to improve the static antifouling performance of the coatings, which leads to an issue whereby static antifouling performance conflicts with eco-friendly traits. Herein, this work reports an in situ reduction synthesis of silver nanoparticles (AgNPs) within polymers to produce composite coatings, aiming to solve the aforementioned issue. Firstly, urushiol-based benzoxazine monomers were synthesized by the Mannich reaction, using an eco-friendly natural product urushiol and n-octylamine and paraformaldehyde as the reactants. Additionally, AgNPs were obtained through the employment of free radicals formed by phenolic hydroxyl groups in the urushiol-based benzoxazine monomers, achieved by the in situ reduction of silver nitrate in benzoxazine. Then, the urushiol-based benzoxazine/AgNPs composite coatings were prepared by the thermosetting method. AgNPs exhibit broad-spectrum and highly efficient antimicrobial properties, with a low risk to human health and a minimal environmental impact. The composite coating containing a small amount of AgNPs (≤1 wt%) exhibits effective inhibition against various types of bacteria and marine microalgae in static immersion, thereby displaying outstanding antifouling properties. This organic polymer and inorganic nanoparticle composite marine antifouling coating, with its simple preparation method and eco-friendliness, presents an effective solution to the conflict between static antifouling effectiveness and environmental sustainability in marine antifouling coatings.

Introduction: The goal of this study is to compare the prognostic performance of NETPET scores, based on gallium-68 DOTANOC (68Ga-DOTANOC) and fluorine-18 fluorodeoxyglucose (18F-FDG) Positron Emission Tomography-Computed Tomography (PET-CT), and PET-CT metabolic parameters in metastatic gastrointestinal neuroendocrine tumors (GI-NET), while constructing and validating a nomogram derived from dual-scan PET-CT. Methods: In this retrospective study, G1–G3 GI-NET patients who underwent 68Ga-DOTANOC and 18F-FDG PET scans were enrolled and divided into training and internal validation cohorts. Three grading systems were constructed based on NETPET scores and standardized uptake value maximum (SUVmax). LASSO regression selected variables for a multivariable Cox model, and nomograms predicting progression-free survival (PFS) and overall survival (OS) were created. The prognostic performance of these systems was assessed using time-dependent receiver-operating characteristic (ROC) curves, concordance index (C-index), and other methods. Nomogram evaluation involved calibration curves, decision curve analysis (DCA), and the aforementioned methods in both cohorts. Results: In this study, 223 patients (130 males; mean age ± SD: 52.6 ± 12 years) were divided into training (148) and internal validation (75) cohorts. Dual scans were classified based on NETPET scores (D1–D3). Single 68Ga-DOTANOC and 18F-FDG PET-CT scans were stratified into S1-S3 and F1-F3 based on SUVmax. The NETPET score-based grading system demonstrated the best OS and PFS prediction (C-index, 0.763 vs. 0.727 vs. 0.566). Nomograms for OS and PFS exhibited superior prognostic performance in both cohorts (all AUCs > 0.8). Conclusions: New classification based on NETPET score predicts patient OS/PFS best. PET-CT-based nomograms show accurate OS/PFS forecasts.

In paleolimnology, ecological trajectories of biological communities are typically reconstructed using a sediment core from the deepest part of a lake. However, the representativeness of chironomid assemblages from the profundal zone and its influence on ecological response to environmental change in large, deep lakes remain poorly understood. To address this knowledge gap, we conducted a study on subfossil chironomids in Fuxian Lake, including thirty-five surface sediment samples and two sediment cores from different water depths. Our findings reveal a distinct bathymetric distribution pattern of subfossil chironomid assemblages attributed to limited homogenization in the large, deep lake. The profundal assemblages exhibited the lowest richness representativeness, while nearshore assemblages showed the least similarity to the entire-lake assemblage. Consequently, the response pattern of chironomid assemblage to the environmental change varied with depth. Two notable shifts in the mid-depth core, occurring in 1979 CE and 2005 CE, were primarily attributed to eutrophication processes. However, the chironomid assemblage from the deepest zone exhibited only one significant change in 2005 CE, likely indicating exacerbated hypoxia in the profundal zone. Our combined analysis of surface samples and sediment cores underscores that the profundal zone, where bottom water is characterized by low temperature and anoxia, is unsuitable as a coring site in large, deep lakes due to its extremely simplified species composition. Instead, mid-depth zones should be prioritized as candidate coring sites in large, deep lakes, given their relatively high richness representativeness, head capsule concentration, and stable sedimentary conditions. Specifically, we recommended retrieving sedimentary cores around the water depth of 50 m when conducting subfossil chironomid analysis in Fuxian Lake.

Difficult and labour-intensive transportation up and down the steps is a challenge in small spaces. This work focused on the smooth performance requirements about each joint structure of robot foot. We have made research on the hand and foot structure of the transport part, which simplifies its posture calculation in the designed bipedal transport robot model. Methods for determination of each joint angle of the hand and foot are presented, including the use of standard D-H method to establish a mathematical model and perform forward and inverse kinematic analysis to derive the angles of each joint, where the matrix was coded by Python software to ensure the accuracy of the calculated results. The joints angles are assigned by the Step function, and the motion of the bipedal robot in the case of single access is simulated by the inverse kinematic derivation formula. Simulation results show that the structure of the robot's joints meets the requirement of maintaining the stability of the robot when going up and down the stairs.

Abstract Background Existing studies for ferroptosis and prognosis in colorectal cancer (CRC) were limited. In this study, we aim to investigate the prognostic role of ferroptosis markers in patients with CRC and exploration of its micro‐environmental distributions. Methods Immunohistochemical staining was performed for CRC patients’ tissue microarray. Selection and prognostic validation of markers were based on mRNA data from the cancer genome atlas (TCGA) database. Gene Set Enrichment Analysis (GSEA) was performed to indicate relative immune landmarks and hallmarks. Ferroptosis and immune contexture were examined by CIBERSORT. Survival outcomes were analyzed by Kaplan‐Meier analysis and cox analysis. Results A panel of 42 genes was selected. Through mRNA expression difference and prognosis analysis, GPX4, NOX1 and ACSL4 were selected as candidate markers. By IHC, increased GPX4, decreased NOX1 and decreased FACL4 indicate poor prognosis and worse clinical characteristics. Ferroptosis score based on GPX4, NOX1 and ACSL4 was constructed and validated with high C‐index. Low ferroptosis score can also demonstrate the better progression free survival and better adjuvant chemotherapy (ACT) responsiveness. Moreover, tumor with low ferroptosis score tend to be infiltrated with more CD4+ T cells, CD8+ T cells and less M1 macrophage. Finally, we found that IFN‐γ was potentially the central molecule at the crossroad between ferroptosis and onco‐immune response. Conclusion Ferroptosis plays important role on CRC tumor progression, ACT response and prognosis. Ferroptosis contributes to immune‐supportive responses and IFN‐γ was the central molecule for this process.

Abstract Background Little is known about the association between gut microbiota and intestinal injury under a state of chronic cerebral hypoperfusion (CCH). Here, the effects of gut microbiota and short-chain fatty acids (SCFAs), as important metabolic products, on intestinal function and potential mechanisms after CCH were investigated. Methods Rats were subjected to bilateral common carotid artery occlusion (BCCAo) to induce CCH. The gut microbiota and metabolites of SCFAs were assessed by 16S rRNA sequencing and targeted metabolomics, respectively. Transcriptomic analysis of colon tissues was also conducted. Subsequently, potential molecular pathways and differentially expressed genes were verified by western blot, immunoprecipitation, and immunofluorescence analyses. Furthermore, the integrity of the colonic barrier was evaluated by hematoxylin and eosin and mucin 2 staining and expression levels of tight junction proteins. Besides, colonic inflammation was further assessed by flow cytometry and expression levels of inflammatory cytokines. In addition, colonic mitochondrial dysfunction was analyzed via membrane potential, reactive oxygen species, electron transport chain (ETC) activities, adenosine triphosphate content, and mitochondrial ultrastructure. Results CCH modified gut microbial composition and microbial metabolism of SCFAs, which may be associated with inhibition of mitochondrial ETC activities and oxidative phosphorylation, leading to dysregulation of mitochondrial energy metabolism. Furthermore, CCH induced differentiation of pathogenic Th17 cells, promoted the formation of complexes of interferon regulatory factor 4 and signal transducer and activator of transcription 3 (STAT3), and increased the phosphorylation of STAT3. This was associated with an impairment of colonic barrier function and chronic colonic inflammation. In contrast, FMT and SCFA replenishment ameliorated CCH-induced gut microbial dysbiosis by increasing the intestinal content of Ruminococcus_sp_N15_MGS_57 and modulating microbial metabolism of SCFAs by increasing acetic acid contents associated with an improvment of the balance between Tregs and Th17 cells, mitochondrial ETC activities, and oxidative phosphorylation to prevent colonic inflammation and dysregulation of mitochondrial energy metabolism. Conclusion These findings indicate that FMT and SCFA replenishment present a promising therapeutic strategy against colonic dysfunction under a state of chronic cerebral ischemia.

The Chinese lacquer composite films were prepared by modifying raw lacquer with polyethylene oxide. The film was characterized via Fourier-transform infrared spectroscopy, thermogravimetric analysis, and scanning electron microscopy. The infrared spectra confirmed the interaction between the polyethylene oxide and urushiol. The heat-resistance of the film was found to have decreased due to the presence of polyethylene oxide via thermogravimetric analysis. Additional pores and wrinkles were observed in the scanning electron microscopy image of polyethylene oxide modified lacquer films. The mechanical properties were tested according to the national standard. The results indicated that the gloss and flexibility of the modified film was enhanced by the presence of polyethylene oxide. When the ratio of polyethylene oxide was 3%, the gloss was increased from 59.8 to 81.6 and the flexibility changed from 15 mm to 1 mm. The alkaline-resistance, hardness, and adhesion were also increased via the modification of polyethylene oxide.

This study aimed to unveil the detailed role and new mechanism of circ-LIMK1 in lung adenocarcinoma. Real-time quantitative polymerase chain reaction was performed to analyze the expression of circ-LIMK1, miR-512-5p, and HMGA1. 3-(4,5)-Dimethylthiahiazo(-z-y1)-3,5-di-phenytetrazoliumromide assay was employed to test the half maximal inhibitory concentration of cisplatin (DDP). Western blot was used to measure the expression of HMGA1, multidrug resistance protein 1, mitochondrial 37S ribosomal protein, and vascular endothelial growth factor A. Colony formation assay, flow cytometry, transwell assay, and tube formation assay were performed to analyze cell functions. Animal models were established to assay the role of circ-LIMK1 in vivo. The expression of circ-LIMK1 was up-regulated in DDP-resistant tumor tissues and cells. Knockdown of circ-LIMK1 reduced DDP resistance, impaired cancer cell growth, migration, invasion, and angiogenesis. circ-LIMK1 targeted miR-512-5p, and HMGA1 was targeted by miR-512-5p. MiR-512-5p absence could restore the repressive effects of circ-LIMK1 knockdown on lung adenocarcinoma cell phenotypes. Overexpression of HMGA1 could restore the inhibitory effects of miR-512-5p enrichment on lung adenocarcinoma cell malignant phenotypes. Knockdown of circ-LIMK1 could reduce growth of DDP-resistant tumors in vivo. Collectively, circ-LIMK1 regulated DDP resistance in lung adenocarcinoma by targeting miR-512-5p/HMGA1 axis.

Objective To determine the effect of rapamycin(Rapa) on JAK2, ABCA3, and the immune checkpoint PD-1/PD-L1 in exosomes derived from JAK2 V617F positive HEL cells. Methods Human erythroleukemia HEL cells (JAK2 V617F mutation-positive) were cultured in vitro, and rapamycin was added at concentrations of 10, 50, and 100 nmol/L. The control group was established and cell proliferation inhibition rate was detected by CCK-8. Based on the inhibition rate of cell proliferation, cells intervened with 10 and 50 nmol/L Rapa were selected. Exosomes were extracted using a kit and identified by Western blot and flow cytometry. JAK2, ABCA3, and PD-1/PD-L1 mRNA changes in exosomes were detected by fluorescent quantitative PCR. The expression of exosome PD-1/PD-L1 protein was determined by flow cytometry. Results The exosomes extracted with the exosome kit all expressed characteristic CD9, CD63, and CD81 proteins, which were consistent with the general characteristics of exosomes. JAK2 mRNA can be detected in exosomes from HEL cells; Rapa reduced exosome production by HEL cells, and dose-dependently decreased gene expression of JAK2, ABCA3, and PD-L1 in exosomes. In addition, Rapa inhibited exosomal PD-L1 protein expression and had no significant effect on PD-1 protein expression. Conclusion HEL cells may transmit JAK2 mutant gene signals via exosomes. Rapa can reduce the production of exosomes and inhibits JAK2 mutated signals delivered by exosomes and suppresses PD-L1 protein expression.

Abstract An air-supplement plasma synthetic jet (PSJ) actuator increases the air supplemental volume in the recovery stage and improves the jet energy by attaching a check valve to the chamber of a conventional actuator. To explore the flow control effect and mechanism of the air-supplement actuator, via particle image velocimetry experiments in a low-speed wind tunnel, the flow field and boundary layer characteristics of a two-dimensional airfoil surface under different actuation states were compared for different attack angles and jet orifices. The experimental results show that, compared with the conventional actuation state, the jet energy of the air-supplement PSJ is higher and the indirect mixing effect of the counter-vortex sequence produced by the jet-mainstream interaction is stronger. Furthermore, the boundary layer mixing effect is better, which can further suppress flow separation and improve the critical flow separation attack angle. Moreover, increasing the jet momentum coefficient can enhance the flow control effect. The findings of this study could provide guidance for the flow control application of air-supplement PSJs.

Objective: We aimed to explore the correlations between and possible mechanisms of common environmental endocrine disruptors, phthalates, and ovarian dysfunction in endometriosis.Methods: Subjects were included in the case group (n = 107) who were diagnosed with endometriosis by postoperative pathology in Fujian Maternal and Child Hospital from February 2018 to February 2021, and the women who were excluded from endometriosis by surgery were as the control group (n = 70). The demographic information of the subjects were evaluated by questionnaire, and the clinical characteristics were evaluated by medical records and 3-year follow-up results. Gas chromatography‒mass spectrometry was used to quantify 10 metabolites of phthalates, including dimethyl ortho-phthalate (DMP), mono-n-methyl phthalate (MMP), dioctyl ortho-phthalate (DEP), mono-ethyl phthalate (MEP), di-n-butyl ortho-phthalate (DBP), mono-butyl phthalate (MBP), benzylbutyl phthalate (BBzP), mono-benzyl; phthalate (MBzP), diethylhexyl phthalate (DEHP) and mono-ethylhexyl phthalate (MEHP), in the urine samples of the subjects. Furthermore, a total of 54 SD rats were exposed to DEHP 0, 5, 50, 100, 250, 500, 1,000, 2000, and 3,000 mg/kg/day for 2 weeks. The SD rats’ body weight, oestrus cycle changes, and serum anti-mullerian hormone (AMH) levels were evaluated. After sacrifice, the mass index of the rat uterus and bilateral ovaries were calculated.
