2. Analysis of mRNA expression profile in the treatment of diabetic foot ulcer healing by tibial cortex transverse distraction

Abstract To investigate mRNA Expression profile and associated signaling pathways in the treatment of diabetic foot ulcer healing by tibial cortex transverse distraction. Tissue samples were collected from the wound edge before and after the surgery. After reference genome transcriptome sequencing and subsequent bioinformatics analysis, the differentially expressed genes and related pathways were explored, and functional analysis of important genes and pathways was conducted. qRT-PCR was used to verify the significantly expressed genes-HLA-DRB1, HLA-DRB5, CXCL5 and IGFL1. There were 2441 significantly up-regulated and 3904 significantly down-regulated genes in the postoperative group. The qRT-PCR results showed the expression of HLA-DRB1, HLA-DRB5 and CXCL5 was consistent with the transcriptional sequencing results. CXCL5 and CXCL6 differentially up-regulated genes are involved in the process of neovascularization, and HLA-DRB1 is involved in the improvement of the degree of diabetic peripheral nerve degeneration. Pathway analysis showed that differential genes were most significantly enriched in Adherens junction, Inflammatory mediator regulation of TRP channels and Wnt signaling pathway. Inflammatory mediator regulation of TRP channels is involved in the improvement of peripheral neurodegeneration, VEGF signaling pathway is involved in the process of neovascularization, and Wnt signaling pathway is involved in the process of bone healing. Significantly up-regulated CXCL5 and CXCL6 and enriched VEGF signaling pathway analyzed are involved in postoperative lower limb neovascularization. The HLA-DRB1 and the enriched Inflammatory mediator regulation of TRP channels may be related to the improvement of postoperative peripheral neurodegeneration. The differentially expressed genes and related pathways can provide objective basis for further mechanism study.

Abstract Direct current microgrid (DCMG) clusters are gaining popularity in power systems due to their simplicity and high efficiency. However, DCMG clusters are susceptible to minor disturbances due to low system inertia. This paper proposes a method to enhance the small‐signal stability of a DCMG cluster by optimizing the main control parameters of the system. This paper presents a small‐signal state‐space model of a DCMG cluster system at the system level, considering a multi‐bus network topology. Then, the control parameters that significantly affect the small‐signal stability of the DCMG are selected using the participation factor method. To enhance the system damping, the Pareto‐optimal frontier of the bi‐objective problem was determined using the elite non‐dominated sorting genetic algorithm (NSGA‐II). The optimal compromise is determined by using the fuzzy membership function method to extract it from the generated Pareto optimal front. The proposed method has been verified on a three‐sub DCMG test system with droop control.

Abstract The global scenario on PVC plasticizer is experiencing a drastic change from petroleum-based, toxic di-(2-ethylhexyl) phthalate (DEHP) toward renewable, non-toxic bio-alternatives. However, replacing diisodecyl phthalate (DIDP), a DEHP analogue specifically intended for plasticizing PVC automotive upholstery, with bio-alternative remains a challenge as few bio-plasticizer volatilizes from PVC as slowly as DIDP, a crucial aspect compulsorily required by automotive industry. Here, we demonstrate that covalently attaching two short esters at the α-position of all components of a traditional epoxidized fatty acid methyl ester via a two-step, hydrogen-to-ester nucleophilic substitution in a one-pot procedure yields an epoxidized fatty acid tri-ester bio-plasticizer with remarkably suppressed volatilization from PVC, and hence an extremely low fogging value comparable to DIDP. With this strategy in hand, DIDP, long deemed irreplaceable despite its toxicity and non-renewable nature, may ultimately be phased out. Graphical Abstract

Nucleic acids play a decisive role in vital life processes, such as growth, heredity, and evolution, and exploring different types of nucleic acid detection techniques has always been a research hotspot worldwide. Compared to traditional nucleic acid analysis methods, surface-enhanced Raman spectroscopy (SERS) detection has advantages, such as high sensitivity, good specificity, narrow spectral bands, minimal sample requirement, simple operation, and resistance to interference from autofluorescence and water in biological samples. The clustered regularly interspaced short palindromic repeats (CRISPR)/Cas system has gained widespread attention in the fields of gene editing and biosensors. Combining the sequence-specific recognition capability of the CRISPR/Cas system with the high sensitivity fingerprinting properties of SERS technology offers a simple, rapid, and sensitive method towards nucleic acid detection. Here, common SERS Nanotags methods for nucleic acid detection are introduced, including “sandwich assay”, “signal switch assay” and HCR signal amplification. Then, the building mechanism of CRISPR-associated SERS detection is summarized. CRISPR/Cas12a activated by crRNA can recognize and lock the complementary target double-stranded DNA (dsDNA), followed by cis-cleavage of the dsDNA and trans-cleavage of the adjacent linker ssDNA. The loss of SERS signal based on aggregation is finally used for highly sensitive detection of the target DNA. Lastly, some typical applications through the integration of CRISPR/Cas and SERS technology are introduced, such as HIV-1 dsDNA, HBV DNA and AFB1 etc.

Background Polypoidal choroidal vasculopathy (PCV) is a hemorrhagic fundus disease that can lead to permanent vision loss. Predicting the treatment response to anti-VEGF monotherapy in PCV is consistently challenging. We aimed to conduct a prospective multicenter study to explore and identify the imaging biomarkers for predicting the anti-VEGF treatment response in PCV patients, establish predictive model, and undergo multicenter validation.Methods This prospective multicenter study utilized clinical characteristics and images of treatment naïve PCV patients from 15 ophthalmic centers nationwide to screen biomarkers, develop model, and validate its performance. Patients from Peking Union Medical College Hospital were randomly divided into a training set and an internal validation set. A nomogram was established by univariate, LASSO regression, and multivariate regression analysis. Patients from the other 14 centers served as an external test set. Area under the curve (AUC), sensitivity, specificity, and accuracy were calculated. Decision curve analysis (DCA) and clinical impact curve (CIC) were utilized to evaluate the practical utility in clinical decision-making.Findings The eye distribution for the training set, internal validation set, and external test set were 66, 31, and 71, respectively. BackgroundGastric ulcer (GU), a globally prevalent disease, represents a significant burden to human health. Bletilla ochracea Schltr. (BOS), an herbal medicine, shows promising therapeutic potential in the treatment of chronic GU.MethodsThis study utilized a rat model of chronic gastric ulceration induced by acetic acid to evaluate the protective effects of Bletilla ochracea Schltr. (BOS) on gastric tissue through the analysis of gross morphological and histopathological changes. Non-targeted metabolomic techniques were employed to identify differential metabolites, followed by the use of metabolic analysis software to enrich the pathways associated with these metabolites, thereby revealing the potential mechanisms underlying the anti-gastric ulcer effects of BOS.ResultsThe results suggest that the primary mechanism underlying BOS regulation of GU involves modulation of endogenous metabolites, including dimethylglycine, l-2,4-diaminobutyric acid, uridine propionic acid and l-asparagine. These diverse metabolites may have anti-inflammatory, antioxidant and reparative properties. In addition, KEGG enrichment analysis indicated potential anti-GU effects of BOS through diverse pathways such as energy metabolism, immune metabolism and amino acid metabolism.ConclusionThe study demonstrates BOS protective effects on GU in rats, potentially through modulating key metabolites and pathways, highlighting its therapeutic potential and warranting further investigation for clinical applications.

Supervisor ostracism is commonplace in work settings. Previous studies have mainly focused on the influence of supervisor ostracism on employees’ work behavior and performance, while the impact of supervisor ostracism on employees’ family harmony has garnered less attention. Based on the affective events theory, we analyzed data collected from 3 time points and 343 employees in the Chinese context. We found that supervisor ostracism significantly undermines employee-family harmony, which is mediated by workplace anger. In addition, our study demonstrated that high emotional regulation alleviates the relationship between workplace anger and family undermining, reducing the destructive consequences for the family. Our results indicated that individual differences in emotional regulation are responsible for moderating the mediating effects of workplace ange between supervisor ostracism and family undermining, shedding light on future research directions and providing practical implications for solving work-family conflict.

9. Machine Learning for Dynamic Prognostication of Patients With Hepatocellular Carcinoma Using Time-Series Data: Survival Path Versus Dynamic-DeepHit HCC Model

Objectives: Patients with intermediate or advanced hepatocellular carcinoma (HCC) require repeated disease monitoring, prognosis assessment and treatment planning. In 2018, a novel machine learning methodology “survival path” (SP) was developed to facilitate dynamic prognosis prediction and treatment planning. One year after, a deep learning approach called Dynamic Deephit was developed. The performance of the two state-of-art models in dynamic prognostication have not been compared. Methods: We trained and tested the SP and Dynamic DeepHit models in a large cohort of 2511 HCC patients using time-series data. The time-series data were converted into data of time slices, with an interval of three months. The time-dependent c-index for OS at given prediction time ( t = 1, 6, 12, 18 months) and evaluation time (∆ t = 3, 6, 9, 12, 18, 24, 36, 48 months) were compared. Results: The comparison between SP model and Dynamic DeepHit-HCC model showed the latter had significant better performance at the time of initial admission. The time-dependent c-index of Dynamic DeepHit-HCC model gradually decreased with the extension of time (from 0.756 to 0.639 in the training set; from 0.787 to 0.661 in internal testing set; from 0.725 to 0.668 in multicenter testing set); while the time-dependent c-index of SP model displayed an increased trend (from 0.665 to 0.748 in the training set; from 0.608 to 0.743 in internal testing set; from 0.643 to 0.720 in multicenter testing set). When the prediction time comes to 6 months or later since initial treatment, the survival path model outperformed the dynamic DeepHit model at late evaluation times (∆ t > 12 months). Conclusions: This research highlighted the unique strengths of both models. The SP model had advantage in long term prediction while the Dynamic DeepHit-HCC model had advantages in prediction at near time points. Fine selection of models is needed in dealing with different scenarios.

Abstract Background Hepatic encephalopathy (HE) is closely associated with inflammatory responses. However, as a crucial regulator of the immune and inflammatory responses, the role of leucine-rich repeat kinase 2 (LRRK2) in the pathogenesis of HE remains unraveled. Herein, we investigated this issue in thioacetamide (TAA)-induced HE following acute liver failure (ALF). AbstractThe saddleback silver-biddy Gerres limbatus (Cuvier 1830) is distributed in Indo-West Pacific Oceans and associated with shallow coastal marine waters and estuaries. In this study, the complete mitochondrial genome of G. limbatus was firstly documented, which is 16,730 bp in length, including 13 protein-coding genes, 22 transfer RNA genes and 2 ribosomal RNA genes. The overall base composition of the mitochondrial genome is 26.42% A, 28.68% C, 27.32% T, and 17.58% G. The Maximum Likelihood phylogenetic tree was constructed based on COI gene of the 31 species from the family Gerreidae, with Heteroclinus puellarum and Hypopterus macropterus as outgroups. It revealed that G. erythrourus was placed as the sister group to G. limbatus.

15. SHISA3 Reprograms Tumor‐Associated Macrophages Toward an Antitumoral Phenotype and Enhances Cancer Immunotherapy

Abstract The main challenge for immune checkpoint blockade (ICB) therapy lies in immunosuppressive tumor microenvironment (TME). Repolarizing M2‐like tumor‐associated macrophages (TAMs) into inflammatory M1 phenotype is a promising strategy for cancer immunotherapy. Here, this study shows that the tumor suppressive protein SHISA3 regulates the antitumor functions of TAMs. Local delivery of mRNA encoding Shisa3 enables cancer immunotherapy by reprogramming TAMs toward an antitumoral phenotype, thus enhancing the efficacy of programmed cell death 1 (PD‐1) antibody. Enforced expression of Shisa3 in TAMs increases their phagocytosis and antigen presentation abilities and promotes CD8+ T cell‐mediated antitumor immunity. The expression of SHISA3 is induced by damage/pathogen‐associated molecular patterns (DAMPs/PAMPs) in macrophages via nuclear factor‐κB (NF‐κB) transcription factors. Reciprocally, SHISA3 forms a complex with heat shock protein family A member 8 (HSPA8) to activate NF‐κB signaling thus maintaining M1 polarization of macrophages. Knockout Shisa3 largely abolishes the antitumor efficacy of combination immunotherapy with Toll‐like receptor 4 (TLR4) agonist monophosphoryl lipid A (MPLA) and PD‐1 antibody. It further found that higher expression of SHISA3 in antitumoral TAMs is associated with better overall survival in lung cancer patients. Taken together, the findings describe the role of SHISA3 in reprogramming TAMs that ameliorate cancer immunotherapy.

The efficacy of PD-1 therapy in non-small cell lung cancer (NSCLC) patients remains unsatisfactory. Activating the STING pathway is a promising strategy to improve PD-1 inhibitor efficacy. Here, we found tetrandrine (TET), an anti-tumor compound extracted from a medicinal plant commonly used in traditional Chinese medicine, has the ability to inhibit NSCLC tumor growth. Mechanistically, TET induces nuclear DNA damage and increases cytosolic dsDNA, thereby activating the STING/TBK1/IRF3 pathway, which in turn promotes the tumor infiltration of dendritic cells (DCs), macrophages, as well as CD8+ T cells in mice. In vivo imaging dynamically monitored the increased activity of the STING pathway after TET treatment and predicted the activation of the tumor immune microenvironment. We further revealed that the combination of TET with αPD-1 monoclonal antibody (αPD-1 mAb) yields significant anti-cancer effects by promoting CD8+ T cell infiltration and enhancing its cell-killing effect, which in turn reduced the growth of tumors and prolonged survival of NSCLC mice. Therefore, TET effectively eliminates NSCLC cells and enhances immunotherapy efficacy through the activation of the STING pathway, and combining TET with anti-PD-1 immunotherapy deserves further exploration for applications.

Abstract Taking advantage of the high specific surface area of the nanoparticles, boron nitride (BN) nanoparticles were incorporated into the semi-solidified aluminium-copper alloy Al–5Cu–Mn (ZL201) system during the casting process, and its properties and enhancement mechanism were studied. The results shown that the BN in the new composite material is more uniformly distributed in the second phase (Al2Cu), which can promote grain refinement and enhance the bonding with the aluminium-based interface, and the formation of stable phases such as AlB2, AlN, CuN, etc. makes the tensile strength and hardness of the material to be significantly improved (8.5%, 10.2%, respectively). The mechanism of the action of BN in Al2Cu was analyzed by establishing an atomic model and after calculation: BN can undergo strong adsorption on the surface of Al2Cu (0 0 1), and the adsorption energy is lower at the bridge sites on the two cut-off surfaces, which makes the binding of BN to the aluminum base more stable. Abstract Hierarchical compartmentalization, a hallmark of both primitive and modern cells, enables the concentration and isolation of biomolecules, and facilitates spatial organization of biochemical reactions. Coacervate-based compartments can sequester and recruit a large variety of molecules, making it an attractive protocell model. In this work, we report the spontaneous formation of core-shell cell-sized coacervate-based compartments driven by spontaneous evaporation of a sessile droplet on a thin-oil-coated substrate. Our analysis reveals that such far-from-equilibrium architectures arise from multiple, coupled segregative and associative liquid-liquid phase separation, and are stabilized by stagnation points within the evaporating droplet. The formation of stagnation points results from convective capillary flows induced by the maximum evaporation rate at the liquid-liquid-air contact line. This work provides valuable insights into the spontaneous formation and maintenance of hierarchical compartments under non-equilibrium conditions, offering a glimpse into the real-life scenario.

Abstract Objective This study aims to translate the Pectus Excavatum Evaluation Questionnaire(PEEQ) into Chinese, and to comprehensively assess subjective outcomes in quality of life of children with pectus excavatum. Methods The PEEQ was translated from English to Chinese as according to the PRO translation guidelines. Structural validity and reliability of the questionnaire were examined by validated factor analysis and Cronbach’s alpha coefficient analysis respectively. Results The results of the validation factor analysis for the Chinese PEEQ parent’s and child’s questionnaires demonstrated that the fit indicators for each dimension met the required criteria. The overall Cronbach’s alpha coefficient of parent’s and child’s questionnaires were 0.840 and 0.854. Both the item-level content validity index (I-CVI) and scale-level content validity index (S-CVI) of each sub-questionnaire were 1. Conclusion The Chinese version of the PEEQ parent’s questionnaire is suitable as a proxy assessment for patients with PE, but the child’s questionnaire needs further adjustments.

Abstract Exfoliation corrosion (EFC) on extruded Mg-4Li-1Ca alloys as bone implant materials has not been reported. This study aimed to elucidate the degradation mechanism of a corrosion type-exfoliation occurred on an extruded Mg-4Li-1Ca alloy under the interaction of glucose (Glu) and bovine serum albumin (BSA) in Hank’s solution. The alloy microstructure, chemical composition, texture, stress distribution and Volta potential maps were characterized using metallographic microscopy, scanning electron microscopy and electron backscattered diffraction, and scanning Kelvin Probe. The alloy corrosion behavior was investigated via electrochemical testing and hydrogen evolution rate methods. X-ray diffraction, Fourier transform infrared spectrometry, X-ray photoelectron spectrometer, and fluorescence microscopy were used to analyze the corrosion products and the adsorption of the organic molecules. The experimental results indicated that the anodic second phase (Mg2Ca) were distributed in lamellar and rod-like patterns along the extrusion direction of the alloy, a lower Volta potential for Mg2Ca, texture and residual stresses also promote EFC. An increase in Glu concentration exacerbated the EFC, whereas BSA inhibited the EFC of the alloy. Higher Glu concentrations in BSA in turn inhibited EFC since Glu can complex with amino acids in proteins to form Glu-amino acid amides, which promoted protein adsorption and thereby forming a dense protective film on the alloy surface. The preferential dissolution of the Mg2Ca phase particles led to the complexation and adsorption of Glu and protein. This study enriches the understanding of the corrosion types in biomedical Mg-4Li-1Ca alloy and paving the way for enhancing the EFC resistance of the alloy.

Despite supervisors playing a crucial role in the cultivation of postgraduate students, the impact of supervisors’ leadership on postgraduate students’ well-being is poorly understood. Based on Self-Determination Theory (SDT), this study explores the immediate and delayed effect of supervisors’ humble leadership on postgraduate students’ well-being, together with the mediating effects of basic psychological need satisfaction and the moderating effects of power distance orientation. Using a survey experiment (Study 1) and multi-timepoint questionnaire (Study 2) design, this paper finds that supervisors’ humble leadership influences postgraduate students’ well-being through its effect on basic psychological need satisfaction. Power distance orientation moderates the relationships between supervisors’ humble leadership, basic psychological need satisfaction, and postgraduate students’ well-being; specifically, humble leadership leads to higher basic psychological need satisfaction and well-being in students with high power distance orientation compared to those with low power distance orientation. This research validates the application of SDT in higher education and clarifies how supervisors’ humble leadership influences postgraduate students’ well-being, providing practical guidance for its improvement.

Staphylea bumalda is a rare medicine and edible shrub native to the temperate regions of Asia, possessing significant medicinal potential. In this study, the components of S. bumalda tender leaves and buds extract (SBE) were analyzed and identified by HPLC and LC/MS method, and the safety of SBE was evaluated through mouse acute toxicity models. The protective effects of SBE on dextran sulfate sodium (DSS)-induced ulcerative colitis (UC) in mice were investigated in terms of inflammatory factor levels, oxidative stress, and gut microorganisms. Results showed that hyperoside, kaempferol-3-O-rutinoside, isorhoifolin, and rutin were the main components of the extract, and SBE demonstrated good safety in experimental mice. SBE could alleviate weight losing, disease activity index (DAI) raising, and colon shortening in mice. Pathological section results showed that the inflammatory cell infiltration decreased significantly, and the number of goblet cells increased significantly in the SBE group. After SBE treatment, interleukin-6 (IL-6), interleukin-1β (IL-1β), and tumor necrosis factor-α (TNF-α) levels in serum were significantly decreased, and the levels of myeloperoxidase (MPO) and nitric oxide (NO) in colon tissues were significantly decreased. SBE inhibited gut inflammation by increasing Lactobacillus. In summary, SBE played a therapeutic role in UC mice by relieving colon injury, reducing inflammatory factor levels, and maintaining gut flora homeostasis. SBE is expected to become an auxiliary means to participate in the prevention and treatment of UC.

29. Use of Waste Slag and Rubber Particles to Make Mortar for Filling the Joints of Snow-Melting Concrete Pavement

Waste slag and rubber particles are commonly used to modify concrete, offering benefits such as reduced cement consumption and lower greenhouse gas emissions during cement production. In this study, these two environmentally friendly, sustainable waste materials were proposed for the preparation of mortar intended for snow-melting pavements. A series of experiments were conducted to evaluate the performance of the material and to determine whether its compressive and flexural strengths meet the requirements of pavement specifications. The mortar’s suitability for snow-melting pavements was assessed based on its thermal conductivity, impermeability, and freeze–thaw resistance. The results indicate that slag, when used in different volume fractions, can enhance the compressive and flexural strength of the mortar. Slag also provides excellent thermal conductivity, impermeability, and resistance to freeze–thaw cycles, contributing to the overall performance of snow-melting pavements. When the slag content was 20%, the performance was optimal, with the compressive strength and flexural strength reaching 58.5 MPa and 8.1 MPa, respectively. The strength loss rate under freeze–thaw cycles was 8.03%, the thermal conductivity reached 2.2895 W/(m * K), and the impermeability pressure value reached 0.5 MPa. Conversely, the addition of rubber particles was found to decrease the material’s mechanical and thermal properties. However, when used in small amounts, rubber particles improved the mortar’s impermeability and resistance to freeze–thaw cycles. When the rubber content was 5% by volume, the impermeability pressure value reached 0.5 MPa, which was 166.7% lower than that of ordinary cement mortar. Under freeze–thaw cycles, the strength loss rate of the test block with a rubber content of 25% volume fraction was 9.83% lower than that of ordinary cement mortar.

Sex reversal and sex ratio bias are common in mollusks, and temperature is one of the most important environmental factors influencing gonadal development and differentiation. Here, we obtained the full-lengh cDNAs of Hsdmrt1 and Hsfoxl2 and found that they were predominantly expressed in the gonads. Furthermore, Hsdmrt1 and Hsfoxl2 showed a sexually dimorphic expression pattern, with Hsdmrt1 biased toward males and Hsfolx2 favoring females. The expression of Hsfoxl2 was inhibited by Hsdmrt1. The sex ratios of 1–3-year-old mussels H. schlegelii were assessed, and the sex ratio bias was from male to female, accompanied by a significant decrease in log10 (Hsdmrt1/Hsfoxl2). And log10(Hsdmrt1/Hsfoxl2) value outside the interval [-2,1] was considered a stable sexual state. The sex reversal in H. schlegelii was reproduced by simulating temperatures, in which male mussels at 27 °C underwent sex reversal to become females; females at 19°C underwent sex reversal to males, and the log10(Hsdmrt1/Hsfoxl2) value was altered to be in the [-2,1] interval. These results support the hypothesis that an imbalance in the expression of either gene leads to a sex reversal and that this may be regulated by temperature. This study deepens our understanding of the mechanisms of sex reversal and gonadal development in bivalves.

IntroductionThe outbreak of the Coronavirus Disease 2019 pandemic has presented significant difficulties for healthcare workers worldwide, resulting in a higher tendency to quit their jobs. This study aims to investigate the correlation between organizational support, work-family-self balance, job satisfaction, and turnover intention of healthcare professionals in China’s public hospitals.MethodsA cross-sectional survey was conducted on 5,434 health workers recruited from 15 public hospitals in Foshan municipality in China’s Guangdong province. The survey was measured by organizational support, work-family-self balance, job satisfaction, and turnover intention using a five-point Likert scale. The association between organizational support, work-family-self balance, job satisfaction, and turnover intention was investigated using Pearson correlation analysis and mediation analysis through the PROCESS macro (Model 6).ResultsOrganizational support indirectly affected turnover intention through three pathways: the mediating role of work-family-self balance, job satisfaction, and the chain mediating role of both work-family-self balance and job satisfaction.ConclusionHealth administrators and relevant government sectors should provide sufficient organizational support, enhance work-family-self balance and job satisfaction among healthcare workers, and consequently reduce their turnover intentions.

32. Corrosion resistance of Cu-Fe deformation processed in situ alloy in chloride ion environment

To assess the corrosion resistance of Cu-Fe deformation in situ alloys in a chloride ion environment, Cu-Fe alloys with varying Fe contents (5 wt%, 10 wt%, and 14 wt%) were prepared using vacuum induction melting, and the impact of Fe content on the corrosion resistance was examined. The corrosion morphology and corrosion products were analyzed, and the corrosion rate, corrosion period, dynamic potential polarization curves, electrochemical parameters, and electrochemical impedance spectra with different Fe contents were determined. However, the corrosion resistance of Cu-Fe alloys initially increased with an increase in Fe content before decreasing, with Cu-10 wt% Fe alloys (95% reduction rate) exhibiting the best corrosion resistance. As the Fe content increased, the amount of primary Fe phase gradually increased and became more densely distributed. This led to an increase in the dense oxide film on the surface, thereby enhancing the corrosion resistance of the material. Moreover, with a further increase in Fe, the primary Fe phase exhibited coarsening and non-uniform distribution. This resulted in the oxide film becoming looser, leading to a decreased corrosion resistance of the alloy.

Natural bioactive compounds and plant constituents are considered to have a positive anti-inflammatory effect. This study aimed to establish a screening technique for anti-inflammatory function in foods based on label-free Raman imaging. A visible anti-inflammatory analysis method based on coherent anti-Stokes Raman scattering (CARS) was established with an LPS-induced RAW264.7 cell model. Dynamic changes in proteins and lipids were determined at laser pump light wavelengths of 2956 cm−1 and 2856 cm−1, respectively. The method was applied to a plant-based formula (JC) with anti-inflammatory activity. Q-TOF-MS and HPLC analyses revealed the main active constituents of JC as quercetin, kaempferol, l-glutamine, and sodium copper chlorophyllin. In in vitro and in vivo verification experiments, JC showed significant anti-inflammatory activity by regulating the TLR4/NF-κB pathway. In conclusion, this study successfully established a label-free and visible method for screening anti-inflammatory constituents in plant-based food products, which will facilitate the evaluation of functional foods.

Purpose: The study investigates the value of the BRAFV600E mutation in determining the aggressiveness of papillary thyroid cancer (PTC) and its correlation with ultrasound features. Methods: The study selected 176 patients with BRAFV600E mutation and 80 without the mutation who underwent surgery at Guangxi Medical University Cancer Hospital. Clinical and pathological data were collected, focusing on BRAFV600E mutations and associated ultrasonic features. Correlation analysis, as well as univariate and multivariate logistic regression analysis, were conducted to identify independent risk factors for BRAFV600E mutation. The results were verified using a nomogram model. Results: The analysis results indicate that the BRAFV600E mutation correlates with tumor size, nodule size, taller-than-wide shape, margin, and shape of papillary thyroid cancer. The receiver operating characteristic curve was used to analyze the diagnostic effect of these features on BRAFV600E mutation. The results showed that nodule size had the most significant area under the curve (AUC = 0.665). Univariate and multivariate logistic regression analyses revealed that taller-than-wide shape ≥1, ill-defined margin, irregular shape, nodule size (≤1.40 cm), TT4 (>98.67 nmol/L), and FT3 (

The Middle Permian Qixia Formation in the Shuangyushi area, northwestern Sichuan Basin, develops shoal-facies dolomite reservoirs. To pinpoint promising reservoirs in the Qixia Formation, deep thin shoal-facies dolomite reservoirs were predicted using the techniques of pre-stack Kirchhoff-Q compensation for absorption, inverse Q filtering, low-to high-frequency compensation, forward modeling, and facies-controlled seismic meme inversion. The results are obtained in six aspects. First, the dolomite reservoirs mainly exist in the middle and lower parts of the second member of Qixia Formation (Qi2 Member), which coincide with the zones shoal cores are developed. Second, the forward modeling shows that the trough energy at the top and bottom of shoal core increases with increasing shoal-core thickness, and weak peak reflections are associated in the middle of shoal core. Third, five types of seismic waveform are identified through waveform analysis of seismic facies. Type-I and Type-II waveforms correspond to promising facies (shoal core microfacies). Fourth, vertically, two packages of thin dolomite reservoirs turn up in the sedimentary cycle of intraplatform shoal in the Qi2 Member, and the lower package is superior to the upper package in dolomite thickness, scale and lateral connectivity. Fifth, in plane, significantly controlled by sedimentary facies, dolomite reservoirs laterally distribute with consistent thickness in shoal cores at topographical highs and extend toward the break. Sixth, the promising prospects are the zones with thick dolomite reservoirs and superimposition of horst–graben structural traps.

Objective: Differentially expressed genes (DEGs) in lung adenocarcinoma (LUAD) tumor stem cells were screened, and the biological characteristics of NR5A2 gene were investigated. Methods: The expression and prognosis of NR5A2 in human LUAD were predicted and analyzed through bioinformatics analysis from a human cancer database. Gene expression and clinical data of LUAD tumor and normal lung tissues were obtained from The Cancer Genome Atlas (TCGA) database, and DEGs associated with lung cancer tumor stem cells (CSCs) were screened. Univariate and multivariate Cox regression models were used to screen and establish prognostic risk prediction models. The immune function of the patients was scored according to the model, and the relative immune functions of the high- and low-risk groups were compared to determine the difference in survival prognosis between the two groups. In addition, we calculated the index of stemness based on the transcriptome of the samples using one-class linear regression (OCLR). Results: Bioinformatics analysis of a clinical cancer database showed that NR5A2 was significantly decreased in human LUAD tissues than in normal lung tissues, and the decrease in NR5A2 gene expression shortened the overall survival and progression-free survival of patients with LUAD. Conclusion: The NR5A2 gene may regulate LUAD tumor stem cells through selective splicing mutations, thereby affecting the survival and prognosis of patients with lung cancer, and the NR5A2 gene may regulate CSCs through single nucleotide polymorphism.

In order to expand the application range of casting aluminum alloy ZL105, the stirring fusion casting method was used to add carbon nanotubes (CNTs) with different content and aspect ratio into the ZL105 aluminum matrix. And then the effect of the reinforcement on the mechanical properties of the alloy was compared and analyzed. The research results show that the tensile strength and hardness of the carbon nanotube composites with different contents will be improved, but to a certain extent the elongation of the composite material will be reduced, and there is an optimal addition amount. The mechanical properties of composite materials prepared by adding CNTs with relatively small length and diameter are better. There are different forms of reinforcement mechanisms for CNTs to reinforce cast aluminum alloys, and the improvement of composite material performance is the result of the combined effect of multiple strengthening methods. The research has made a meaningful exploration for the realization of carbon nanotube reinforced aluminum matrix composites under the casting method.

Furthermore, the transcriptomic analysis revealed that blue light treatment induced up-regulation of genes in photosynthesis (BrHY5-2, BrCOP1 and BrDET1) and the methylerythritol 4-phosphate pathways (BrGGPS, BrDXS and BrHDR) upstream of the carotenoid metabolic pathway. Carotenoid metabolomic analysis revealed that the accumulation of several orange and red carotenoids (lycopene, zeaxanthin, β-carotene, lutein, and β-cryptoxanthin) after blue light treatment contributed to the deepening of the leaf coloration, suggesting that short-term blue light treatment could be used to boost nutritional quality. The light signal gene BrHY5-2 participated in the blue light-induced transcriptional regulation of carotenoid biosynthesis in OHCC. Overexpression of BrHY5-2 in Arabidopsis significantly increased the total carotenoid content and the sensitivity to blue light. The above findings revealed new insights about blue-light-induced carotenoid synthesis and accumulation in OHCC lines. They suggested a new engineering approach to increase the nutritional value of vegetables.

39. Optimization on the Preparation Process of Whole Egg Cake Premixed Powder and Its Flavor Analysis

The effects of whole egg powder, cake oil and sugar additive amount on the specific volume, texture characteristics and sensory scores of the cake were evaluated using single factor experiment. Based on the single factor experiment, response surface methodology was used to optimize the formula of the cake premixed powder with the sensory score as the response value. The results showed that the cake had the highest sensory score of 87.12 when the formula of the premixed powder was 65% whole egg powder, 20% cake oil and 57.5% of white granulated sugar. The specific volume and texture characteristics of the cake under the optimal formula were determined as follows: Specific volume was 3.69 mL/g, hardness was 438.54±19.31 g, elasticity was 0.91±0.02, and cohesiveness was 0.69±0.01, which were all better than those before response surface optimization. The cakes prepared with the optimal formula were bright yellow and had small internal pores, soft and elastic taste, and a unique cake aroma, sweet but not greasy. The electronic nose was used to analyze the flavor of the optimal cake, and the results found that ethanol, amine, fluorine and hydrocarbon were the main flavor substances in the cake. Further flavor analysis indicated that the optimal cake contained nine pleasant flavor substances, among which 1-nonanal and (E,E)-2,4-decadien-1-al were at relatively high levels in the optimal cake, thus presenting a pleasant aroma as a whole.

Femtosecond laser machining offers high precision and minimal thermal impact, making it a promising technique for processing hard and brittle materials like single-crystal diamonds (SCDs). In this study, the femtosecond laser machining process for SCD material was systematically optimized to improve both machining efficiency and quality. Initial single-factor experiments were conducted to explore the effects of key process parameters—laser power, scanning speed, and number of scans—on machining performance. Subsequently, response surface methodology (RSM)-based experiments designed using the Box–Behnken method were employed to comprehensively refine the process. A regression model was developed to analyze the data, and the interaction effects of the parameters were thoroughly evaluated. The validated model identified an optimal set of parameters, resulting in a significant improvement in machining performance. This research provides a comprehensive framework for optimizing femtosecond laser machining processes, offering valuable insights critical for the production of advanced lightweight components in industries such as aerospace, optical instruments, and high-performance electronics.

Botryosphaeria dothidea, a notorious plant pathogen, is responsible for causing chestnut rot during postharvest storage. This research aimed to assess the antifungal properties of perilla essential oil (PEO) and perillaldehyde (PAE) against B. dothidea. PEO’s and PAE’s inhibitory effects on B. dothidea were investigated using an agar dilution method, a fumigation method, and an in vivo assay in chestnuts and shell buckets. Based on the results of gas chromatography-mass spectrometry, it was confirmed that the main component of PEO was elemicin. The antifungal mechanism of PEO and PAE against B. dothidea was investigated by conducting staining experiments of the fungal cell wall and cell membrane. PEO and PAE strongly inhibit the mycelial growth of B. dothidea in a dose-dependent manner. The inhibitory mechanism is mainly related to the destruction of the integrity of the fungal cell wall and plasma membrane. Notably, PEO retains its antifungal efficacy against B. dothidea in chestnuts, effectively prolonging their storage life. These findings indicate that PEO and PAE are nontoxic, eco-friendly botanical fungicides, holding promise for controlling postharvest chestnut rot.

Abstract Retinoblastoma is the most frequent intraocular malignancy in children. Little is known on the molecular basis underlying the biological and clinical behavior of this cancer. Here, using gene expression profiles, we demonstrate the existence of two major retinoblastoma subtypes that can be divided into six subgroups. Subtype 1 has higher expression of cone related genes and higher percentage of RB1 germline mutation. By contrast, subtype 2 tumors harbor more genes with ganglion/neuronal features. The dedifferentiation in subtype 2 is associated with stemness features including low immune infiltration. Gene Otology analysis demonstrates that immune response regulations and visual related pathways are the key molecular difference between subtypes. Subtype 1b has the highest risk of invasiveness across all subtypes. The recognition of these molecular subtypes shed a light on the important biological and clinical perspectives for retinoblastomas.

Phosphatase of regenerating liver 3 (PRL3) is a specific tumor antigen overexpressed in a broad range of adult cancer types. However, its physiological expression in pediatric embryonal and mesenchymal tumors and its association with clinical outcomes in children is unknown. We sought to profile the expression of PRL3 in pediatric tumors in relation to survival outcomes, expression of angiogenesis markers, and G-protein-coupled receptor (GPCR)-mitogen-activated protein kinase (MAPK) signaling targets. PRL3-zumab, a first-in-class humanized antibody, was administered in a dose escalation schedule in a first-in-child clinical trial to study toxicity, pharmacokinetics, and clinical outcomes. Among 64 pediatric tumors, PRL3 was most frequently expressed in neuroblastoma (100%), rhabdomyosarcoma and non-rhabdomyosarcoma soft tissue sarcomas (71%), and renal sarcomas (60%) but absent in paired normal tissues. PRL3 was expressed in 75% of relapsed tumors and associated with shorter median event-free survival. Microarray profiling of PRL3-positive tumors showed elevation of angiogenin, TIMP1 and TIMP2, and GPCR-MAPK signaling proteins that commonly interacted with PRL3. The first use of PRL3-zumab in a pediatric patient saw no adverse events. A 28.6% reduction in maximum target lesion diameter was achieved when PRL3-zumab was administered concurrently with hypofractionated radiation. These findings support wider exploration of PRL3 expression in embryonal and mesenchymal tumors and further clinical application of PRL3-zumab in pediatric patients.
