Abstract

We present the first limit on $g_{A\gamma}$ coupling constant using the Bragg-Primakoff conversion based on an exposure of 1107.5 kg days of data from the CDEX-1B experiment at the China Jinping Underground Laboratory. The data are consistent with the null signal hypothesis, and no excess signals are observed. Limits of the coupling $g_{A\gamma}<2.08\times10^{-9}$ GeV$^{-1}$ (95\% C.L.) are derived for axions with mass up to 100 eV/$c^2$. Within the hadronic model of KSVZ, our results exclude axion mass $>5.3~\rm{eV}/c^2$ at 95\% C.L., Comment: 7 pages, 5 figures

Abstract

We present the first results of the search for sub-MeV fermionic dark matter absorbed by electron targets of Germanium using the 205.4~kg$\cdot$day data collected by the CDEX-10 experiment, with the analysis threshold of 160~eVee. No significant dark matter (DM) signals over the background are observed. Results are presented as limits on the cross section of DM--electron interaction. We present new constraints of cross section in the DM range of 0.1--10 keV/$c^2$ for vector and axial-vector interaction. The upper limit on the cross section is set to be $\rm 5.5\times10^{-46}~cm^2$ for vector interaction, and $\rm 1.8\times10^{-46}~cm^2$ for axial-vector interaction at DM mass of 5 keV/$c^2$., Comment: 6 pages, 4 figures

Abstract

We report new constraints on light dark matter (DM) boosted by blazars using the 205.4 kg day data from the CDEX-10 experiment located at the China Jinping Underground Laboratory. Two representative blazars, TXS 0506+56 and BL Lacertae are studied. The results derived from TXS 0506+56 exclude DM-nucleon elastic scattering cross sections from $4.6\times 10^{-33}\ \rm cm^2$ to $1\times10^{-26}\ \rm cm^2$ for DM masses between 10 keV and 1 GeV, and the results derived from BL Lacertae exclude DM-nucleon elastic scattering cross sections from $2.4\times 10^{-34}\ \rm cm^2$ to $1\times10^{-26}\ \rm cm^2$ for the same range of DM masses. The constraints correspond to the best sensitivities among solid-state detector experiments in the sub-MeV mass range., Comment: 7 pages, 4 figures

Abstract

Dark matter (DM) is a major constituent of the Universe. However, no definite evidence of DM particles (denoted as ``$\chi$") has been found in DM direct detection (DD) experiments to date. There is a novel concept of detecting $\chi$ from evaporating primordial black holes (PBHs). We search for $\chi$ emitted from PBHs by investigating their interaction with target electrons. The examined PBH masses range from 1$\times$10$^{15}$ to 7$\times$10$^{16}$ g under the current limits of PBH abundance $f_{PBH}$. Using 205.4 kg$\cdot$day data obtained from the CDEX-10 experiment conducted in the China Jinping Underground Laboratory, we exclude the $\chi$--electron ($\chi$--$e$) elastic-scattering cross section $\sigma_{\chi e} \sim 5\times10^{-29}$ cm$^2$ for $\chi$ with a mass $m_{\chi}\lesssim$ 0.1 keV from our results. With the higher radiation background but lower energy threshold (160 eV), CDEX-10 fill a part of the gap in the previous work. If ($m_{\chi}$, $\sigma_{\chi e}$) can be determined in the future, DD experiments are expected to impose strong constraints on $f_{PBH}$ for large $M_{PBH}$s., Comment: 9 pages, 6 figures, 3 tables. Abstract

Recently a dark matter-electron (DM-electron) paradigm has drawn much attention. Models beyond the standard halo model describing DM accelerated by high energy celestial bodies are under intense examination as well. In this Letter, a velocity components analysis (VCA) method dedicated to swift analysis of accelerated DM-electron interactions via semiconductor detectors is proposed and the first HPGe detector-based accelerated DM-electron analysis is realized. Utilizing the method, the first germanium based constraint on sub-GeV solar reflected DM-electron interaction is presented with the 205.4 kg$\cdot$day dataset from the CDEX-10 experiment. In the heavy mediator scenario, our result excels in the mass range of 5$-$15 keV/$c^2$, achieving a 3 orders of magnitude improvement comparing with previous semiconductor experiments. In the light mediator scenario, the strongest laboratory constraint for DM lighter than 0.1 MeV/$c^2$ is presented. The result proves the feasibility and demonstrates the vast potential of the VCA technique in future accelerated DM-electron analyses with semiconductor detectors., Comment: 7 pages, 4 figures. Abstract

CDEX-50 is a next-generation project of the China Dark Matter Experiment (CDEX) that aims to search for dark matter using a 50-kg germanium detector array. This paper comprises a thorough summary of the CDEX-50 dark matter experiment, including an investigation of potential background sources and the development of a background model. Based on the baseline model, the projected sensitivity of weakly interacting massive particle (WIMP) is also presented. The expected background level within the energy region of interest, set to 2--2.5 keVee, is $\sim$0.01 counts keVee$^{-1}$ kg$^{-1}$ day$^{-1}$. At 90\% confidence level, the expected sensitivity to spin-independent WIMP-nucleon couplings is estimated to reach a cross-section of 5.1 $\times$ 10$^{-45}$ cm$^{2}$ for a WIMP mass of 5 GeV/c$^{2}$ with an exposure objective of 150 kg$\cdot$year and an analysis threshold of 160 eVee. Childhood obesity appears to be associated with an increased prevalence of sleep apnea syndrome. Sleep apnea is an inestimable risk factor for thrombosis, hypertension, cardiomyopathy and many other diseases. Therefore, exploring the relationship between childhood obesity and sleep apnea syndrome will help to understand the potential link between the two and provide research directions for future disease prevention and treatment. However, no studies have confirmed whether there is a causal relationship between childhood obesity and sleep apnea syndrome.Methods: The IEU OpenGWAS project provided the GWAS-aggregated data for childhood obesity and sleep apnea syndrome. Inverse-variance weighted (IVW) was used as the main method to evaluate the causal relationship between childhood obesity and sleep apnea syndrome. Single nucleotide polymorphisms (SNPs) were regarded as instrumental variables, and the screening threshold was P < 5.0× 10− 6. Leave-one-out method was performed to confirm the robustness of the results.Results: IVW analysis confirmed a causal relationship between genetic susceptibility to childhood obesity and an increased risk of sleep apnea syndrome [odds ratio (OR)=1.12, 95% confidence interval (CI): 1.02– 1.23, P=0.016]. However, two-sample MR results also showed no causal relationship between genetic susceptibility to sleep apnea syndrome and an increased risk of childhood obesity (OR=1.50, 95% CI: 0.95– 2.38, P=0.083). The intercept of MR-Egger regression was close to 0, which implies that there are no confounding factors in the analysis to affect the results of two-sample MR analysis. The leave-one-out results show that the bidirectional two-sample MR analysis results were robust.Conclusion: There is a causal relationship between genetic susceptibility to childhood obesity and increased risk of sleep apnea syndrome. Abstract

We operated a p-type point contact high purity germanium (PPCGe) detector (CDEX-1B, 1.008 kg) in the China Jinping Underground Laboratory (CJPL) for 500.3 days to search for neutrinoless double beta ($0\nu\beta\beta$) decay of $^{76}$Ge. A total of 504.3 kg$\cdot$day effective exposure data was accumulated. The anti-coincidence and the multi/single-site event (MSE/SSE) discrimination methods were used to suppress the background in the energy region of interest (ROI, 1989$-$2089 keV for this work) with a factor of 23. A background level of 0.33 counts/(keV$\cdot$kg$\cdot$yr) was realized. The lower limit on the half life of $^{76}$Ge $0\nu\beta\beta$ decay was constrained as $T_{1/2}^{0\nu}\ > \ {1.0}\times 10^{23}\ \rm yr\ (90\% \ C.L.)$, corresponding to the upper limits on the effective Majorana neutrino mass: $\langle m_{\beta\beta}\rangle < $3.2$-$7.5$\ \mathrm{eV}$., Comment: 11 pages, 12 figures, 2 tables. We present novel constraints on boosted light dark matter particles (denoted as ``$\chi$'') from evaporating primordial black holes (PBHs) using 205.4 kg$\cdot$day data from the China Jinping Underground Laboratory's CDEX-10 p-type point contact germanium detector with a 160 eVee analysis threshold. $\chi$ from PBHs with masses ranging from 1$\times$10$^{15}$ g to 7$\times$10$^{16}$ g are searched in this work. In the presence of PBH abundance compatible with present bounds, our result excludes the $\chi$-nucleon elastic-scattering cross section region from 3.4$\times$10$^{-32}$ cm$^{2}$ to 2.3$\times$10$^{-29}$ cm$^{2}$ for $\chi$ of 1 keV to 24 MeV from PBHs with masses of 5$\times$10$^{15}$ g, as well as from 1.1$\times$10$^{-28}$ cm$^{2}$ to 7.6$\times$10$^{-28}$ cm$^{2}$ for $\chi$ of 1 keV to 0.6 MeV from PBHs with masses of 7$\times$10$^{16}$ g. If the $\chi$-nucleon elastic-scattering cross section can be determined in the future, the abundance of PBHs may be severely constrained by $\chi$ evaporation. With the lower threshold (160 eVee) of the CDEX-10 experiment compared to the previously used experiments, this work allows for a better reach at soft spectra produced by heavier PBHs, which demonstrates the vast potential of such a technical route to pursue $\chi$ from larger PBHs with a low threshold., Comment: 8 pages, 6 figures. Abstract

The $^{13}$C($\alpha$,$n$)$^{16}$O reaction is the main neutron source for the slow-neutron-capture (s-) process in Asymptotic Giant Branch stars and for the intermediate (i-) process. Direct measurements at astrophysical energies in above-ground laboratories are hindered by the extremely small cross sections and vast cosmic-ray induced background. We performed the first consistent direct measurement in the range of $E_{\rm c.m.}=$0.24 MeV to 1.9 MeV using the accelerators at the China Jinping Underground Laboratory (CJPL) and Sichuan University. Our measurement covers almost the entire i-process Gamow window in which the large uncertainty of the previous experiments has been reduced from 60\% down to 15\%, eliminates the large systematic uncertainty in the extrapolation arising from the inconsistency of existing data sets, and provides a more reliable reaction rate for the studies of the s- and i-processes along with the first direct determination of the alpha strength for the near-threshold state.

Abstract

We investigate exotic neutrino interactions using the 205.4 kg$\cdot$day dataset from the CDEX-10 experiment at the China Jinping Underground Laboratory. New constraints on the mass and couplings of new gauge bosons are presented. Two nonstandard neutrino interactions are considered: a $U(1)_{B-L}$ gauge-boson-induced interaction between an active neutrino and electron/nucleus, and a dark-photon-induced interaction between a sterile neutrino and electron/nucleus via kinetic mixing with a photon. This work probes an unexplored parameter space involving sterile neutrino coupling with a dark photon. New laboratory limits are derived on dark photon masses below $1~{\rm eV}/c^{2}$ at some benchmark values of $\Delta m_{41}^{2}$ and $g^{\prime2}{\rm{sin}}^{2}2\theta_{14}$., Comment: 6 pages, 4 figures. Abstract

A search for exotic dark matter (DM) in the sub-GeV mass range has been conducted using 205 kg$\cdot$day data taken from a p-type point contact germanium detector of CDEX-10 experiment at China Jinping underground laboratory. New low-mass dark matter searching channels, neutral current fermionic DM absorption ($\chi+A\rightarrow \nu+A$) and DM-nucleus 3$\rightarrow$2 scattering ($\chi+\chi+A\rightarrow \phi+A$), have been analyzed with an energy threshold of 160 eVee. No significant signal was found. Thus new limits on the DM-nucleon interaction cross section are set for both models at sub-GeV DM mass region. A cross section limit for the fermionic DM absorption is set to be $\rm 2.5\times 10^{-46} cm^2$(90\% C.L.) at DM mass of 10 MeV/c$^2$. For the DM-nucleus 3$\rightarrow$2 scattering scenario, limits are extended to DM mass of 5 MeV/c$^2$ and 14 MeV/c$^2$ for the massless dark photon and bound DM final state, respectively., Comment: 5 pages, 7 figures

The LHRH receptor is specifically overexpressed in most ovarian cancers, and the integrin αvβ 3 receptor is also overexpressed on the surface of ovarian cancer cells. In this study, we designed LHRH analogues (LHRHa)/RGD co-modified paclitaxel liposomes (LHRHa-RGD-LP-PTX) to target LHRH receptor-positive ovarian cancers more effectively and enhance the anti-ovarian cancer effects.Methods: LHRHa-RGD-LP-PTX liposomes were prepared using the thin film hydration method. The morphology, physicochemical properties, cellular uptake, and cell viability were assessed. Additionally, the cellular uptake mechanism of the modified liposomes was investigated using various endocytic inhibitors. The inhibitory effect of the formulations on tumor spheroids was observed under a microscope. The co-localization with lysosomes was visualized using confocal laser scanning microscopy (CLSM), and the in vivo tumor-targeting ability of the formulations was assessed using the IVIS fluorescent imaging system. Finally, the in vivo anti-tumor efficacy of the formulations was evaluated in the armpits of BALB/c nude mice.Results: The results indicated that LHRHa-RGD-LP-PTX significantly enhanced cellular uptake in A2780 cells, increased cytotoxicity, and hand a more potent inhibitory effect on tumor spheroids of A2780 cells. It also showed enhanced co-localization with endosomes or lysosome in A2780 cells, improved tumor-targeting capability, and demonstrated an enhanced anti-tumor effect in LHRHR-positive ovarian cancers.Conclusion: The designed LHRHa-RGD-LP-PTX liposomes significantly enhanced the tumor-targeting ability and therapeutic efficacy for LHRH receptor-positive ovarian cancers. Abstract

We present improved germanium-based constraints on sub-GeV dark matter via dark matter--electron ($\chi$-$e$) scattering using the 205.4 kg$\cdot$day dataset from the CDEX-10 experiment. Using a novel calculation technique, we attain predicted $\chi$-$e$ scattering spectra observable in high-purity germanium detectors. In the heavy mediator scenario, our results achieve 3 orders of magnitude of improvement for $m_{\chi}$ larger than 80 MeV/c$^2$ compared to previous germanium-based $\chi$-$e$ results. We also present the most stringent $\chi$-$e$ cross-section limit to date among experiments using solid-state detectors for $m_{\chi}$ larger than 90 MeV/c$^2$ with heavy mediators and $m_{\chi}$ larger than 100 MeV/c$^2$ with electric dipole coupling. The result proves the feasibility and demonstrates the vast potential of a new $\chi$-$e$ detection method with high-purity germanium detectors in ultralow radioactive background., Comment: 6 pages, 3 figures. Abstract

A natural broad energy germanium (BEGe) detector is operated in the China Jinping Underground Laboratory (CJPL) for a feasibility study of building the next generation experiment of the neutrinoless double-beta (0{$\nu\beta\beta$}) decay of $^{76}$Ge. The setup of the prototype facility, characteristics of the BEGe detector, background reduction methods, and data analysis are described in this paper. A background index of 6.4$\times$10$^{-3}$ counts/(keV$\cdot$kg$\cdot$day) is achieved and 1.86 times lower than our previous result of the CDEX-1 detector. No signal is observed with an exposure of 186.4 kg$\cdot$day, thus a limit on the half life of $^{76}$Ge 0$\nu\beta\beta$ decay is set at T$_{1/2}^{0\nu}$ $>$ 5.62$\times$10$^{22}$ yr at 90% C.L.. The limit corresponds to an effective Majorana neutrino mass in the range of 4.6 $\sim$ 10.3 eV, dependent on the nuclear matrix elements., Comment: 10 pages, 15 figures

Abstract

We present new constraints on light dark matter boosted by cosmic rays (CRDM) using the 205.4 kg day data of the CDEX-10 experiment conducted at the China Jinping Underground Laboratory. Abstract

Dark matter direct detection experiments mostly operate at deep underground laboratories. It is necessary to consider shielding effect of the Earth, especially for dark matter particles interacting with a large cross section. We analyzed and simulated the Earth shielding effect for dark matter at the China Jinping Underground Laboratory (CJPL) with a simulation package, CJPL Earth Shielding Simulation code (CJPL\_ESS), which is applicable to other underground locations. AimTo evaluate aspects of cognition impacted by individuals with and without normal tension glaucoma.MethodsFifty normal tension glaucoma (NTG) and 50 control patients ≥50y of age were recruited from the UCSF Department of Ophthalmology. Demographic data and glaucoma parameters were extracted from electronic medical records for both groups. Tests of executive function [Executive Abilities: Measures and Instruments for Neurobehavioral Evaluation and Research (EXAMINER)] and learning and memory [California Verbal Learning Test-Second Edition (CVLT-II)] were administered to both NTG and controls. Race, handedness, best-corrected visual acuity, maximum intraocular pressure, optic nerve cup-to-disc ratio, visual field and optic nerve optical coherence tomography parameters, and a measure of general health (Charlson Comorbidity Index) were compared between NTG and controls as well as within NTG subgroups. Multivariate linear regression was used to compare group performances on the EXAMINER battery and CVLT-II while controlling for age, sex, and years of education.ResultsNTG and controls were comparable with respect to age, sex, race, education, handedness, and the Charlson Comorbidity Index (P>0.05 for all). Performance on the EXAMINER composite score and the CVLT-II did not differ between NTG and controls (P>0.05 for both).ConclusionThis is the first prospective study in which the cognitive function of subject with NTG were evaluated using a comprehensive, computerized neurocognitive battery. Subjects with NTG do not perform worse than unaffected controls on tests of executive function, learning, and memory. Results do not support the hypothesis that individuals with NTG are at higher risk for cognitive dysfunction and/or dementia.

Abstract

PurposeTo develop a vision-targeted health-related quality-of-life instrument for patients with glaucoma who are candidates for minimally invasive glaucoma surgery (MIGS).DesignDevelopment of a health-related quality-of-life instrument.ParticipantsTwelve practicing ophthalmologists and 41 glaucoma patients.MethodsA questionnaire was constructed to assess functional limitations, vision-related symptoms, aesthetics, psychosocial issues, and surgical satisfaction for MIGS candidates. Questions were drafted after a review of the literature and subsequently refined based upon input from 1 physician and 4 patient focus groups. Nineteen cognitive interviews were used to ensure that questions were understandable to respondents.ResultsThe focus group identified the following key issues and concerns as being important to glaucoma patients: functional limitations (eg, driving), bodily discomfort (eg, stinging from drops), changes in appearance (eg, drooping eyelid), and psychosocial concerns (eg, mental burden associated with a diagnosis of glaucoma, financial burden of treatment). Cognitive interviews resulted in the following improvements to the questionnaire: changes in wording to clarify lighting conditions, and additional questions addressing psychosocial issues, such as job loss, severity of disease, and perception of MIGS.ConclusionsA patient-reported outcomes instrument, the Glaucoma Outcomes Survey, was developed to evaluate MIGS for patients with mild to moderate glaucoma. Next steps include electronic administration to patients selected from the American Academy of Ophthalmology Intelligent Research in Sight (IRIS) registry. An electronic patient-reported outcomes platform will be used to administer the questionnaire before and after MIGS. The questionnaire will improve understanding of how surgical interventions such as MIGS impact vision-targeted health-related quality-of-life in glaucoma patients.

Abstract

Abstract Glaucoma remains a leading cause of blindness globally. Minimally invasive treatment techniques are rapidly expanding the availability of therapeutic options for glaucoma. These include devices aimed at enhancing outflow through the subconjunctival space, Schlemm’s canal, and suprachoroidal space, sustained-release drug delivery devices, and extraocular devices aiming to reduce glaucomatous progression through other novel means. In this review, we provide an overview of several novel devices either newly available or in development for the medical and surgical management of glaucoma. Further studies are required to determine the long-term efficacy of these devices and how they will integrate into the current landscape of glaucoma management.

Abstract

We present weakly interacting massive particles (WIMPs) search results performed using two approaches of effective field theory from the China Dark Matter Experiment (CDEX), based on the data from both CDEX-1B and CDEX-10 stages. In the nonrelativistic effective field theory approach, both time-integrated and annual modulation analyses were used to set new limits for the coupling of WIMP-nucleon effective operators at 90% confidence level (C.L.) and improve over the current bounds in the low $m_{\chi}$ region. In the chiral effective field theory approach, data from CDEX-10 were used to set an upper limit on WIMP-pion coupling at 90% C.L. Abstract

We present the improved constraints on couplings of solar axions and more generic bosonic dark matter particles using 737.1 kg-days of data from the CDEX-1B experiment. The CDEX-1B experiment, located at the China Jinping Underground Laboratory, primarily aims at the direct detection of weakly interacting massive particles using a p-type point-contact germanium detector. We adopt the profile likelihood ratio method for analysis of data in the presence of backgrounds. An energy threshold of 160 eV was achieved, much better than the 475 eV of CDEX-1A with an exposure of 335.6 kg-days. This significantly improves the sensitivity for the bosonic dark matter below 0.8 keV among germanium detectors. Limits are also placed on the coupling $g_{Ae} < 2.48 \times 10^{-11}$ from Compton, bremsstrahlung, atomic-recombination and de-excitation channels and $g^{eff}_{AN} \times g_{Ae} < 4.14 \times 10^{-17}$ from a $^{57}$Fe M1 transition at 90\% confidence level., Comment: version accepted by PRD, 9 pages, 11 figures

Abstract

We report constraints on the dark photon effective kinetic mixing parameter (${\kappa}$) with data taken from two ${p}$-type point-contact germanium detectors of the CDEX-10 experiment at the China Jinping Underground Laboratory. The 90\% confidence level upper limits on ${\kappa}$ of solar dark photon from 205.4 kg-day exposure are derived, probing new parameter space with masses (${m_V}$) from 10 to 300 eV/${c^2}$ in direct detection experiments. Considering dark photon as the cosmological dark matter, limits at 90\% confidence level with ${m_V}$ from 0.1 to 4.0 keV/${c^2}$ are set from 449.6 kg-day data, with a minimum of ${\rm{\kappa=1.3 \times 10^{-15}}}$ at ${\rm{m_V=200\ eV/c^2}}$., Comment: 5 pages, 6 figures

Abstract

We report results on the searches of weakly interacting massive particles (WIMPs) with sub-GeV masses ($m_{\chi}$) via WIMP-nucleus spin-independent scattering with Migdal effect incorporated. Analysis on time-integrated (TI) and annual modulation (AM) effects on CDEX-1B data are performed, with 737.1 kg$\cdot$day exposure and 160 eVee threshold for TI analysis, and 1107.5 kg$\cdot$day exposure and 250 eVee threshold for AM analysis. The sensitive windows in $m_{\chi}$ are expanded by an order of magnitude to lower DM masses with Migdal effect incorporated. New limits on $\sigma_{\chi N}^{\rm SI}$ at 90\% confidence level are derived as $2\times$10$^{-32}\sim7\times$10$^{-35}$ $\rm cm^2$ for TI analysis at $m_{\chi}\sim$ 50$-$180 MeV/$c^2$, and $3\times$10$^{-32}\sim9\times$10$^{-38}$ $\rm cm^2$ for AM analysis at $m_{\chi}\sim$75 MeV/$c^2-$3.0 GeV/$c^2$., Comment: 5 pages, 4 figures

Abstract

BackgroundMany therapeutic options are available to glaucoma patients. One recent therapeutic option is minimally invasive glaucoma surgical (MIGS) devices. It is unclear how patients view different treatments and which patient-reported outcomes would be most relevant in patients with mild to moderate glaucoma. We developed a questionnaire for patients eligible for MIGS devices and a patient preference study to examine the value patients place on certain outcomes associated with glaucoma and its therapies.ObjectivesTo summarize the progress to date.MethodsQuestionnaire development: We drafted the questionnaire items based on input from one physician and four patient focus groups, and a review of the literature. We tested item clarity with six cognitive interviews. These items were further refined. Patient preference study: We identified important benefit and risk outcomes qualitatively using semi-structured, one-on-one interviews with patients who were eligible for MIGS devices. We then prioritized these outcomes quantitatively using best-worst scaling methods.ResultsQuestionnaire testing: Three concepts were deemed relevant for the questionnaire: functional limitations, symptoms, and psychosocial factors. We will evaluate the reliability and validity of the 52-item draft questionnaire in an upcoming field test. Patient preference study: We identified 13 outcomes that participants perceived as important. Outcomes with the largest relative importance weights were "adequate IOP control" and "drive a car during the day."ConclusionsPatients have the potential to steer clinical research towards outcomes that are important to them. Incorporating patients' perspectives into the MIGS device development and evaluation process may expedite innovation and availability of these devices.

Abstract

Glaucomatous neurodegeneration, a blinding disease affecting millions worldwide, has a need for the exploration of new and effective therapies. Previously, the glucagon-like peptide-1 receptor (GLP-1R) agonist NLY01 was shown to reduce microglia/macrophage activation, rescuing retinal ganglion cells after IOP elevation in an animal model of glaucoma. GLP-1R agonist use is also associated with a reduced risk for glaucoma in patients with diabetes. In this study, we demonstrate that several commercially available GLP-1R agonists, administered either systemically or topically, hold protective potential in a mouse model of hypertensive glaucoma. Further, the resulting neuroprotection likely occurs through the same pathways previously shown for NLY01. This work contributes to a growing body of evidence suggesting that GLP-1R agonists represent a viable therapeutic option for glaucoma.

Background/aims To investigate the rates of structural and functional progression of primary open-angle glaucoma in an African ancestry cohort and identify risk factors for progression.Methods This retrospective study included 1424 eyes from glaucoma cases in the Primary Open-Angle African American Glaucoma Genetics cohort, with ≥2 visits for retinal nerve fibre layer (RNFL) thickness and mean deviation (MD) measurements over ≥6-month follow-up. The rates of structural progression (change in RNFL thickness/year) and functional progression (change in MD/year) were calculated from linear mixed effects models, accounting for intereye correlation and longitudinal correlation. Eyes were categorised as slow, moderate or fast progressors. Risk factors for progression rates were assessed using univariable and multivariable regression models.Results The median (interquartile) rates of progression were −1.60 (−2.05 to –1.15) µm/year for RNFL thickness and −0.40 (−0.44 to –0.34) decibels/year for MD. Eyes were categorised as slow (structural: 19%, functional: 88%), moderate (structural: 54%, functional: 11%) and fast (structural: 27%, functional: 1%) progressors. Abstract

Purpose: We describe a patient with elevated intraocular pressure (IOP) secondary to an oral water bolus and examine the utility of the water-drinking test. Observations: A 66-year-old male with a history of hypertension presented with headache, bilateral retro-orbital ache, and blurry vision. Symptoms began shortly after his radiation treatment for prostate cancer, for which he consumed a water bolus to fill his bladder 30 minutes prior to treatment initiation. On exam, he had bilateral elevated IOP that responded to topical IOP-lowering medications. Gonioscopy demonstrated open angles and fundus exam showed non-glaucomatous optic nerves with pronounced retinal venous tortuosity. The water-drinking test showed a peak intraocular pressure of 20 mmHg in the right eye (5 mmHg increase from baseline) and 23 mmHg in the left eye (8 mmHg increase from baseline), suggesting impairment of the outflow system in the left compared to the right eye. He was started on topical IOP-lowering therapy and followed in our clinic as a glaucoma suspect. Conclusions: Consumption of a water bolus can be associated with IOP elevation and may be a risk factor in patients with otherwise normal IOPs at risk for glaucoma. The water-drinking test was historically used as provocative testing for open-angle glaucoma and may have an updated role in evaluating at-risk patients without ocular hypertension.

All patients were classified as FH(+) and ROS(+), FH(+) and ROS(-), or FH(-) and ROS(-), based on the FH and ROS combined test of voided urine. Demographic information, pathological results, and status of the FH and ROS combined test were reviewed retrospectively. The relationship between FH and ROS combined test status and tumor stage and grade was evaluated using logistic regression.Results: This study included 120 UC patients with a median age of 69 years (interquartile range [IQR] 62– 77 years). Eighteen patients (15%) were diagnosed with upper tract urothelial carcinoma, and the others (85%) were diagnosed with bladder cancer. The pathological stages for those with FH(+) and ROS(+) at diagnosis were 25.0% Ta, 45.8% T1, and 29.2% ≥T2. The pathological stages for those with FH(+) and ROS(-) at diagnosis were 23.5% Ta, 35.3% T1, and 41.2% ≥T2. The pathological stages for those with FH(-) and ROS(-) at diagnosis were 52.6% Ta, 26.3% T1, and 21.1% ≥T2. After adjusting for clinical factors, including age, sex, and smoking history, FH(+) and ROS(-) were independent risk factors for muscle-invasive UC (≥T2 stage) at diagnosis (odds ratio [OR] 3.379; 95% confidence interval [CI] 1.103– 10.355; P=0.033) in the univariate and multivariate logistic regression analyses.Conclusion: Among patients with newly diagnosed UC, FH(+) and ROS(-) might have an association with a more advanced pathological stage. Abstract

AIM: To evaluate aspects of cognition impacted by individuals with and without normal tension glaucoma. METHODS: Fifty normal tension glaucoma (NTG) and 50 control patients ≥50y of age were recruited from the UCSF Department of Ophthalmology. Demographic data and glaucoma parameters were extracted from electronic medical records for both groups. Tests of executive function [Executive Abilities: Measures and Instruments for Neurobehavioral Evaluation and Research (EXAMINER)] and learning and memory [California Verbal Learning Test–Second Edition (CVLT-II)] were administered to both NTG and controls. Race, handedness, best-corrected visual acuity, maximum intraocular pressure, optic nerve cup-to-disc ratio, visual field and optic nerve optical coherence tomography parameters, and a measure of general health (Charlson Comorbidity Index) were compared between NTG and controls as well as within NTG subgroups. Multivariate linear regression was used to compare group performances on the EXAMINER battery and CVLT-II while controlling for age, sex, and years of education. RESULTS: NTG and controls were comparable with respect to age, sex, race, education, handedness, and the Charlson Comorbidity Index (P>0.05 for all). Performance on the EXAMINER composite score and the CVLT-II did not differ between NTG and controls (P>0.05 for both). CONCLUSION: This is the first prospective study in which the cognitive function of subject with NTG were evaluated using a comprehensive, computerized neurocognitive battery. Subjects with NTG do not perform worse than unaffected controls on tests of executive function, learning, and memory. Results do not support the hypothesis that individuals with NTG are at higher risk for cognitive dysfunction and/or dementia.

The hyperplasia of airway smooth muscle cells (ASMCs) promotes the development of airway remodeling in asthma. Therefore, we attempted to evaluate the regulatory function of H19 in the proliferation and migration of ASMCs.Methods: The expressions of H19 and miR-21 were detected using qRT-PCR. PDGF-BB-induced abnormal proliferation and migration of ASMCs was used as the airway remodeling model in vitro. The expressions of H19 and miR-21 were modified by transfection with pcDNA3.1-H19 and miR-21 mimic, respectively. CCK-8 assay, flow cytometry-based cell cycle analysis was conducted to examine the proliferation ability of ASMCs. The migration ability was measured by transwell assay. Dual-luciferase reporter system was carried out to find the potential relationship between miR-21 and H19 or PTEN. Western blot was conducted to detect the expressions of PCNA, MMP-9, α-SMA, PTEN, and the phosphorylation level of Akt.Results: LncRNA-H19 expression was decreased and microRNA-21 expression was increased in serum samples of children with asthma and PDGF-BB-stimulated ASMCs. Overexpression of H19 reduced the proliferation and migration ability of ASMCs with PDGF-BB treatment and these changes were reversed by miR-21 mimic. H19 promoted the protein level of PTEN via sponging miR-21. Overexpression of H19 suppressed miR-21-induced phosphorylation of Akt, and the suppression effect of H19 on phosphorylation of Akt was significantly reduced after transfecting shPTEN in ASMCs.Conclusion: In this study, overexpression of H19 suppressed the proliferation and migration of ASMCs induced by PDGF-BB via miR-21/PTEN/Akt axis, which could be a potential biomarker and target for treating hyperplasia of airway smooth muscle cells.Keywords: lncRNA H19, asthmatic children, airway smooth muscle cell, miR-21, airway remodeling

Abstract

PurposeTo investigate the association between ultrasound energy, expressed as cumulative dissipated energy (CDE), and changes in intraocular pressure (IOP) after uncomplicated cataract surgery by phacoemulsification.MethodsIn this prospective study, nonglaucomatous subjects underwent cataract surgery by phacoemulsification. IOP was compared by clustered linear regression at 4 separate time-points: preoperative, 1 day, 1 month, and 3 months after cataract surgery. Changes in the IOP were evaluated as a function of CDE using univariate and multivariate clustered linear regression models, which adjusted for sex, ethnicity, age, axial length, spherical equivalent, mean preoperative Shaffer gonioscopy grade of all 4 quadrants, cataract grade, preoperative IOP, central corneal thickness, and use of both eyes in the same subject.ResultsOne hundred sixty-one eyes (89 Asian, 49 white, 12 African, and 11 Hispanic) from 116 nonglaucomatous subjects were analyzed. The 161 eyes included 81 right and 80 left eyes. The 89 Asian eyes included 46 Chinese, 35 Filipino, and 8 Vietnamese. Preoperative IOP was 14.9±3.2 mm Hg. Postoperative IOP significantly increased to 16.0±4.9 mm Hg at 1 day (P=0.037) and decreased to 12.4±3.1 and 12.3±3.0 mm Hg at 1 and 3 months, respectively (both P0.05).ConclusionsThe amount of ultrasound energy delivered to the eye during phacoemulsification, expressed as CDE, was not associated with postoperative changes in IOP.

This work is on the Physics of the B Factories. Part A of this book contains a brief description of the SLAC and KEK B Factories as well as their detectors, BaBar and Belle, and data taking related issues. Part B discusses tools and methods used by the experiments in order to obtain results. The results themselves can be found in Part C. Please note that version 3 on the archive is the auxiliary version of the Physics of the B Factories book. This uses the notation alpha, beta, gamma for the angles of the Unitarity Triangle. Our study uses cationic/anionic polyelectrolyte coated vesicular phospholipid gels (VPGs) loaded with cytarabine (Ara-C) that enhance in vitro and in vivo anti-glioma effects.Methods: Sodium cholesteryl sulfate (SCS) or octadecylamine (ODA) incorporated in a phospholipids phase were used to prepare charged VPGs, and cationic ϵ-polylysine (PLL) coated VPGs (PLL-SCS VPGs) and anionic γ-polyglutamic acid (PGA) coated VPGs (PGA-ODA VPGs) were prepared via electrostatic interactions, respectively. The morphology, particle size, zeta potential, rheology properties, and in vitro release were then characterized. The in vitro cytotoxicity and cellular uptake were evaluated on U87-MG glioma cells. The in vivo antitumor effects were studied on BALB/c nude mice bearing a right flank U87-MG glioma model.Results: The TEM images and physicochemical properties of cationic/anionic polyelectrolyte coated VPGs exhibited that polymers covered on the vesicular surface. The results of rheologic property analysis showed that cationic/anionic polyelectrolyte coated VPGs enhanced the viscosity of uncoated VPGs. The in vitro release experiments revealed that cationic/anionic polyelectrolyte coated VPGs kept Ara-C sustained release up to 18 days. Specially, compared with PLL-SCS VPGs, PGA-ODA VPGs demonstrated higher in vitro cytotoxicity and cellular uptake efficiency in U87-MG glioma cells, and enhanced in vivo antitumor effects when subcutaneously injected around the tumor. No severe toxicity appeared in the right flank U87-MG glioma model of BALB/c nude mice.Conclusion: Anionic γ-PGA coated VPGs were superior to cationic PLL coated VPGs in terms of improving the anti-glioma effect for local delivery.Keywords: biodegradable depots, polyelectrolyte coating, sustained release, glioma, local therapy
