Foodborne parasites are a source of human parasitic infection. Zoonotic infections of humans arise from a variety of domestic and wild animals, including sheep, goats, cattle, camels, horses, pigs, boars, bears, felines, canids, amphibians, reptiles, poultry, and aquatic animals such as fishes and shrimp. Therefore, the implementation of efficient, accessible, and controllable inspection policies for livestock, fisheries, slaughterhouses, and meat processing and packaging companies is highly recommended. In addition, more attention should be paid to the education of auditors from the quality control (QC) and assurance sectors, livestock breeders, the fishery sector, and meat inspection veterinarians in developing countries with high incidence of zoonotic parasitic infections. Furthermore, both the diagnosis of zoonotic parasitic infections by inexpensive, accessible, and reliable identification methods and the organization of effective control systems with sufficient supervision of product quality are other areas to which more attention should be paid. In this review, we present some examples of successful inspection policies and recent updates on present conventional, serologic, and molecular diagnostic methods for zoonotic foodborne parasites from both human infection and animal-derived foods.

We explored the usage of Monte Carlo tree search (MCTS) and deep learning in order to predict game level difficulty in Candy Crush Saga (Candy) measured as number of attempts per success. A deep neural network (DNN) was trained to predict moves from game states from large amounts of game play data. The DNN played a diverse set of levels in Candy and a regression model was fitted to predict human difficulty from bot difficulty. We compared our results to an MCTS bot. Our results show that the DNN can make estimations of game level difficulty comparable to MCTS in substantially shorter time. Stock forecasting is a problem that is important in finance because it aids investors in financial decision making. According to the efficient market hypothesis stock markets are efficient in such a way that it's impossible to gain excess returns over the market by making decisions based on current available information. This paper evaluates the usage of machine learning algorithms and historical trading data for stock price prediction combined with investment strategies in order to test the efficient market hypothesis. The results show that none of the tested machine learning algorithms managed to gain excess returns over the market which confirms the efficient market hypothesis.

Progesteronin kohdegeenit rintasyöpäsoluissa Progesteroni l. keltarauhashormoni on pääasiassa munasarjoista ja istukasta erittyvä steroidihormoni, joka yhdessä estrogeenin kanssa säätelee merkittävästi lisääntymisbiologisia tapahtumia. Progesteronin synteettisiä muotoja progestiineja käytetään laajalti osana vaihdevuosien jälkeisessä hormonikorvaushoidossa (HRT), hormonaalisessa ehkäisyssä, sekä endometrioosin ja rintasyövän hoidossa. Laajat tutkimukset ovat kuitenkin osoittaneet, että estrogeenia ja progesteronia sisältävä yhdistelmähoito lisää riskiä sairastua rintasyöpään. Myös hormonaalisia ehkäisyvalmisteita käyttävillä on kohonnut rintasyöpäriski. Kahdessa klassisessa kohde-elimessään, kohdussa ja rintarauhasessa, progesteronilla on merkittävä vaikutus solujen kasvuun ja erilaistumiseen. Näissä elimissä progestiinit voivat joko lisätä tai estää solujen jakautumista solu – ja kudosspesifisellä tavalla. Esimerkiksi kohdun limakalvon pintasolukossa progesteroni estää estrogeenin aikaansaamaa solukasvua ja lisää solujen erilaistumista. Sen vuoksi progestiinia (mm. medroksiprogesteroniasetaatti, MPA) annetaan hormonikorvaushoidossa yhdistelmähoitona estrogeenin kanssa kontrolloimaan estrogeenin aikaansaamaa solujen kasvua ja pienentämään kohdun limakalvon syövän esiintyvyyttä. Rintarauhasessa progestiinin rooli solukasvun kontrolloijana on epäselvä. Rintarauhasessa progestiineilla on kaksitahoinen vaikutus, sillä ne sekä lisäävät että estävät solujen kasvua. Normaalissa rintarauhasessa progestiini lisää solukasvua ja pitkäaikaisen altistus progesteronille onkin rintasyövän riskitekijä. Toisaalta rintarauhasen kasvaimissa ja viljellyissä rintasyöpäsoluissa, progestiini estää estrogeenin aikaansaamaa kasvua, johon liittyy myös solujen erilaistumisen lisääntyminen. Progesteronin vaikutus elimistössä välittyy spesifisen vastaanottajamolekyylin, progesteronireseptorin (PR) kautta. Hormonin aktivoima PR säätelee geenien ilmentymistä sitoutumalla niiden säätelyalueille. Aktivoimalla tai estämällä geenien ilmentymistä progesteroni vaikuttaa solukasvuun ja erilaistumiseen. Mitä ovat nämä geenit? Tässä väitöskirjatyössä etsittiin progesteronin ja laajalti käytetyn synteettisen progestiinin MPA:n kohdegeenejä viljellyistä rintasyöpäsoluista. Tunnistimme useita uusia progesteronin ja MPA:n kohdegeenejä. Tuloksissa on erityisen merkittävää se, että löytämämme geenit voivat osaltaan selittää progesteronin paradoksaaliset vaikutukset rintarauhasen soluissa. Löysimme progestiinien aktivoimia geenejä, joilla tiedetään olevan kasvua estäviä vaikutuksia, mutta toisaalta myös geenejä joiden tiedetään edistävän solujen kasvua. Tutkimus antaa tukea aiemmin esitetylle hypoteesille, että progestiinit säätelisivät sekä kasvua edistäviä että kasvua estäviä geenejä ja solujen ympäristö yms. ratkaisee tapahtuuko soluissa kasvuinhibitio vai kasvustimulus. Kolmen löydetyn proteiinin (DLG5, GPR30 ja S100P) rakennetta ja roolia kasvunsäätelyssä ja rintasyövässä tutkittiin tarkemmin. Ottaen huomioon, että maailmanlaajuisesti miljoonat naiset käyttävät progestiineja ja että näihin liittyy mm. kohonnut rintasyövän riski, on tärkeää tietää progestiinien solukasvuun vaikuttavia mekanismeja molekyylitasolla. Väitöskirjatyössä tehdyt havainnot auttavat osaltaan ymmärtämään progestiinien kasvunsäätelyn mekanismeja. Progesterone is a steroid hormone that plays a key role in the development, differentiation and maintenance of female reproductive tissues. Synthetic compounds with progesterone-like activity, progestins, are widely used in oral contraception, hormone replacement therapy (HRT), the treatment of some gynecological disorders and the treatment of endometrial and advanced breast cancer. Medroxyprogesterone acetate (MPA) is a major synthetic progestin used in oral contraceptives, the treatment of endometriosis, and hormonal replacement therapy and occasionally for metastatic breast cancer treatment. In the mammary gland, progesterone is a paradoxical hormone: on one hand, it has a proliferative and a differentiative role in normal mammary gland development and in preparing the breast for lactation during pregnancy; on the other hand, progesterone/progestins inhibit the growth of breast cancer cells in vivo and in vitro. Further, progestins inhibit the estrogen-mediated growth of the endometrial epithelium and are therefore combined with estrogen in postmenopausal HRT to eliminate the increased risk of developing endometrial cancer resulting from long-term estrogen-only therapy. However, the risk of developing breast cancer is substantially increased in women using combined HRT. The effects of progestins on cell growth are primarily exerted through the progesterone receptor (PR), which functions as a ligand-inducible transcription factor. Upon activation by a ligand, PR affects gene transcription and triggers the expression of its primary target genes, which in turn leads to the activation of further downstream targets. What are these primary and downstream target genes? In this work, to better understand the molecular mechanism of progestin action, a set of various molecular biology techniques was utilized to identify genes whose expression is altered in response to progestins in cultured breast cancer cells. Dozens of novel progesterone/progestin target genes in breast cancer cells were identified. Of particular interest in this work, several genes were identified that point to a dual role of progestins in controlling the growth of the mammary breast epithelium. In summary, the genes identified in this study point to the capacity of progestins either to stimulate or inhibit cell growth on a transcriptional level. Attention was further focused on three of the primary target genes: DLG5 (a putative tumor suppressor gene), S100P (a potential tumor biomarker), and GPR30 (an orphan receptor).
