5. B cells mediate lung ischemia/reperfusion injury by recruiting classical monocytes via synergistic B cell receptor/TLR4 signaling

Abstract

Ischemia/reperfusion injury-mediated (IRI-mediated) primary graft dysfunction (PGD) adversely affects both short- and long-term outcomes after lung transplantation, a procedure that remains the only treatment option for patients suffering from end-stage respiratory failure. While B cells are known to regulate adaptive immune responses, their role in lung IRI is not well understood. Here, we demonstrated by intravital imaging that B cells are rapidly recruited to injured lungs, where they extravasate into the parenchyma. Using hilar clamping and transplant models, we observed that lung-infiltrating B cells produce the monocyte chemokine CCL7 in a TLR4-TRIF-dependent fashion, a critical step contributing to classical monocyte (CM) recruitment and subsequent neutrophil extravasation, resulting in worse lung function. We found that synergistic BCR-TLR4 activation on B cells is required for the recruitment of CMs to the injured lung. Finally, we corroborated our findings in reperfused human lungs, in which we observed a correlation between B cell infiltration and CM recruitment after transplantation. This study describes a role for B cells as critical orchestrators of lung IRI. 16. Public Pedagogy and Representations of Higher Education in Popular Film: New Ground for the Scholarship of Teaching and Learning

Abstract

Constructions of teaching, learning, and the university within popular culture can exert an important influence on public understandings of higher education, including those held by faculty and students. As such, they constitute a rich site of inquiry for the scholarship of teaching and learning. Drawing on the notion of film as 'public pedagogy,' this article analyses representations of higher education within 11 top grossing and/or critically acclaimed films released in 2014. We identify three broad themes across these texts--the purpose of higher education, relationships between students and professors, and the creation of academic identities--and consider the implications and functions of these representational patterns for teaching, learning, and SoTL. Particular attention is given to the difference between the framing of science and arts and humanities disciplines, and to how this might resonate with the contemporary 'crisis of the humanities.'

38. Just Entertainment? Student and Faculty Responses to the Pedagogy of Media Representations of Higher Education

Abstract

Hollywood film has been positioned as a form of public pedagogy -- a site of efficacious informal learning. Acknowledging this educative potential, this article examines the ways in which faculty and students perceive and respond to representations of higher education offered by popular film. Drawing on focus groups and interviews with 22 participants, we discuss the key themes about teaching and learning that faculty and students observed in filmic representations -- including an emphasis on genius, the presence or absence of effort, and a prevalence of extreme faculty-student relationships -- and consider the extent to which these lessons shape viewers' understanding and experiences of teaching, learning, and universities. While many participants suggested these representations are 'just entertainment' and thus have limited impact, their comments nonetheless indicate ways in which films shape their own and others' experiences and perceptions of the university, underscoring the need for further research in this area.

Abstract

Tertiary lymphoid organs are aggregates of immune and stromal cells including high endothelial venules and lymphatic vessels that resemble secondary lymphoid organs and can be induced at nonlymphoid sites during inflammation. The function of lymphatic vessels within tertiary lymphoid organs remains poorly understood. During lung transplant tolerance, [Foxp3.sup.+] cells accumulate in tertiary lymphoid organs that are induced within the pulmonary grafts and are critical for the local downregulation of alloimmune responses. Here, we showed that tolerant lung allografts could induce and maintain tolerance of heterotopic donor-matched hearts through pathways that were dependent on the continued presence of the transplanted lung. Using lung retransplantation, we showed that [Foxp3.sup.+] cells egressed from tolerant lung allografts via lymphatics and were recruited into donor-matched heart allografts. Indeed, survival of the heart allografts was dependent on lymphatic drainage from the tolerant lung allograft to the periphery. Thus, our work indicates that cellular trafficking from tertiary lymphoid organs regulates immune responses in the periphery.
