Abstract Background Essential amino acid (EAA) and omega-3 fatty acid ingestion independently attenuate leg skeletal muscle disuse atrophy in uninjured persons. However, no data exist regarding the effectiveness of combined EAA and omega-3 fatty acid ingestion to mitigate skeletal muscle disuse atrophy in response to anterior cruciate ligament reconstruction (ACLR) surgery. This pilot trial will explore the feasibility of recruitment and retention of ACLR outpatients from a single center across 18 months to consume either a combination of omega-3 fatty acids and EAAs, or a placebo control, for 4 weeks before and 2 weeks after surgery. Methods Thirty adult (≥ 18 years old) ACLR outpatients will be recruited for this single center, double-blind, two-arm randomized controlled feasibility pilot trial. Participants will consume either 5 g⋅day−1 of omega-3 fatty acids (fish oil) and 40 g⋅day−1 of EAAs or 5 g⋅day−1 of a control fatty acid mixture (safflower oil) and 40 g⋅day−1 of non-essential amino acids (NEAAs). Fatty acid supplements will be consumed 4 weeks before and for 2 weeks after ACLR surgery, whereas the EAAs and NEAAs will be consumed 1 week before and for 2 weeks after ACLR surgery. The primary outcomes are feasibility of recruitment and retention, with the goal to recruit 30 outpatients across 18 months and retain 22 participants upon completion of the study protocol following 12 weeks of data collection. These results will be reported using descriptive statistics, along with reasons and timepoints for study dropout. Secondary exploratory outcomes will be reported using inferential statistics for purposes of hypothesis generation and elucidation of mechanistic targets for future work; no inferences to clinical efficacy will be made. These outcomes include integrated rates of skeletal muscle protein synthesis, skeletal muscle protein content and expression of translation factors, skeletal muscle and erythrocyte phospholipid composition, and measures of skeletal muscle mass, strength, and power. Impact This work will set the foundation for a future randomized controlled trial powered to detect an effect of EAA + omega-3 fatty acid intake on skeletal muscle size or function in response to ACLR surgery. In advance of the practicum and internship experiences, counselor educators are challenged to provide realistic simulations of the counseling relationship for students. We describe a pilot collaboration between an introductory counseling and an applied theatre course to elucidate the mutual benefits of incorporating theatrical improvisation into the counseling curriculum.

Background: Edutainment has long been used as a strategy in health promotion and public health wellness interventions. In March 2019, a large US southeastern university hosted a multimodal theatrical production entitled From Colored to Black (FCTB). The play used a historical lens to broadly address social determinants of health such as disparities in education, access to health services, fair housing and health outcomes. This creative intervention was intended to encourage progressive, justice-oriented attitudes about historically disenfranchised Black communities in North Central Florida. Methods: Driven by an arts-based education approach and constructs from narrative transportation theory, the study employed directed qualitative content analysis to evaluate audience commentary and examine how a play on historic health inequities in Black communities could activate cognition, emotion and imagination for critical thinking about present-day wellbeing. Results: Qualitative survey data in response to the artistic presentation revealed the following subthemes: fascination, assumed truthfulness, satisfaction, feelings of inspiration, enthusiasm, negative attitudes, hopelessness and emotional disconnect. Conclusions: Edutainment continues to be a strong intervention tool. Feedback indicates audience members experienced both highly immersive moments and low transportation-inducing points that ultimately influenced critical reflection on some of the themes addressed in the play.

Shore‐oblique bathymetric features occur around the world and have been statistically correlated with enhanced shoreline retreat on sandy beaches. However, the physical mechanisms that explain a causal relationship are not well understood. In this study, radar remote sensing observations and results from a phase‐resolved numerical model explore how complex morphology alters nearshore hydrodynamics. Observations at selected times during high‐energy storm events as well as a suite of idealized simulations indicate that shore‐oblique features induce strong spatial variations in the water surface elevation, wave breaking patterns, and mean current pathways. Re‐emergent offshore flows and longshore current accelerations occur near the shoreward apex of the oblique nearshore features. The results suggest that complex bathymetric morphology exerts a powerful control on nearshore hydrodynamics and increases the potential for enhanced cross‐shore and alongshore sediment transport. Plain Language Summary: Near the shoreline, underwater topography is affected by sea level, waves, currents, tides, and geological characteristics. Typically, sandbars are oriented parallel to the coastline, but shore‐oblique sandbars have also been identified around the world. In many instances, these oblique features have been correlated with zones of enhanced erosion, however the explanation for this statistical relationship is not fully understood. In this study, remote sensing observations and modeling results explore how complex underwater topographies alter coastal wave energy and flow patterns. Selected stormy periods were observed in addition to a set of idealized simulations. Results indicate that shore‐oblique features cause localized changes to wave heights, wave breaking, and current speeds. These alterations contribute to the presence of rapid offshore‐directed rip currents and accelerations in shore‐parallel currents, which could enhance the potential for sand to be transported away from these zones and contribute to erosion. Abstract Middle to Upper Ordovician ironstone and associated sedimentary rocks of the West Asturian‐Leonese and Cantabrian tectonostratigraphic zones, Spain, provide new information regarding the Palaeozoic Fe cycle and the palaeoceanography of the Rheic Ocean. Examination of drill cores and outcrops indicates the southeastern margin of this narrow seaway was a dynamic continental shelf where upwelling of ferruginous seawater and storm currents controlled lithofacies character. Parasequence composition and stacking relationships suggest ironstone accumulated during marine transgression as accommodation increased from lowstand conditions. Proximal parasequences record aggradation from deep subtidal to shoreface environments. Hummocky cross‐stratified sandstone and organic‐rich siltstone grade upwards into swaley cross‐stratified sandstone and granular Fe‐silicate‐rich ironstone capped by a flooding surface. Distal parasequences were deposited below storm wave base on the distal shelf and are composed of variably bioturbated organic‐rich siltstone with thin Fe‐chlorite and phosphorite layers. These differences in parasequence character define two different ironstone factories where Fe was concentrated and precipitated in sediment. Lithofacies associations support an emerging model for ironstone deposition where coastal upwelling delivered and stimulated the precipitation of Fe within shelf sediment. This notion provides further evidence for the development of intermittent anoxic water masses in an Ordovician ocean that was near the threshold of becoming fully ventilated. This style of Fe delivery probably represents a tipping point in the oxygenation history of the Phanerozoic oceans and is a throwback to the Precambrian when widespread anoxia allowed hydrothermal Fe to concentrate in the global ocean. New data suggest that minor extinction events punctuating the Great Ordovician Biodiversification Event may be traced to these anoxic waters, which in addition to Fe, were also enriched in biologically toxic, redox sensitive trace elements. Conversely, precipitation of upwelling‐related ironstone may have helped sequester these trace elements, providing a negative feedback response that would aid post‐extinction recovery.

Middle Ordovician phosphatic ironstone of the Welsh Basin provides new insight into the paleoenvironmental significance of ironstone and Ordovician ocean chemistry. Deposition occurred in a back-arc basin along the southern margin of Avalonia as the Rheic Ocean opened to the south. Ironstone is interpreted to have accumulated as part of an aggradational parasequence on a storm-dominated shelf with coastal upwelling. This parasequence has a laminated pyritic mudstone base that grades upward into variably bioturbated mudstone and coated grain-rich, intraclastic ironstone, which is overlain in turn by cross-stratified grainstone composed entirely of coated Fe grains. A coarser clastic parasequence composed of more proximal lithofacies rests conformably above and suggests the contact between the two parasequences is a maximum flooding surface marking the onset of highstand conditions. Lithofacies associations suggest that sustained coastal upwelling created a wedge of nutrient-rich, ferruginous seawater on the middle shelf that stimulated high surface ocean productivities. Large, coated Fe grains (granule size) composed of discontinuous and concentric carbonate fluorapatite, hematite, and chamosite cortical layers record fluctuations in pore water Eh that are interpreted to have been related to changes in upwelling intensity and intermittent storm reworking of the seafloor. Results support an emerging model for Ordovician ironstone underpinned by the development of ferruginous bottom water that was periodically tapped by coastal upwelling. Expanding, semi-restricted seaways such as the Rheic Ocean were ideal locations for the ponding of this anoxic, hydrothermally enriched seawater, especially during the early Paleozoic when the deep ocean was variably and inconsistently oxygenated. The coincidence of ironstone depositional episodes with graptolite diversification events suggests that, in addition to Fe, the sustained supply of upwelling-related P may have driven the radiation of some planktonic ecosystems during the Great Ordovician Biodiversification Event. Concomitant minor extinctions of benthic trilobites occurred as these ferruginous waters impinged on the shelf.

Using Gondwana as an example, we show how the geological record can be interrogated to detect significant changes in mantle convection patterns at critical junctures in Earth's evolution. Evidence of major changes in mantle circulation in the aftermath of late Neoproterozoic-early Paleozoic Gondwana assembly is provided by widespread (i) plume-related magmatism around Gondwana's periphery, (ii) ironstone deposits related to mantle plume-ocean ridge interaction and enhanced hydrothermal activity, and (iii) super-mature clastic deposits that reflect epeirogenic uplift triggered by mantle upwelling beneath Gondwana combined with deep tropical weathering. In our model, Gondwana assembled above a region of mantle downwelling in which subducted slabs between the converging Gondwanan continents descended to the core-mantle boundary. Renewed subduction along Gondwana's periphery yielded early arc magmas. But as downwelling beneath Gondwana evolved into upwelling as a result of the ponding of subducted slabs at the base of the mantle, mantle plumes rose from the margins of the nascent upwelling to interact with the edges of Gondwana, where they penetrated the peripheral subduction zones via slab windows, tears and transform faults to generate voluminous calc-alkalic crustal melts in hydrated arc regions and A-type magmas in dry back-arc regions. The plumes also underplated oceanic lithosphere and interacted with adjacent ocean ridges, thereby enhancing hydrothermal activity and the flux of bioessential nutrients, leading to the recurrence of marine iron-rich sedimentary rocks in the geological record. At the same time, upwelling beneath a tropical to equatorial Gondwana led to epeirogenic uplift, deep weathering and erosion, resulting in the production of widespread super-mature clastic deposits. We contend that major changes in mantle convection patterns were encoded into the geological record of Gondwana assembly, influenced global-scale mantle convection patterns, and should be incorporated into geodynamic models for the assembly of Pangea. How well are our schools preparing students to become global citizens who can communicate in languages other than English? To answer this question, we surveyed a nationally representative sample of more than 5,000 U.S. public and private elementary and secondary schools. Results identify current patterns and shifts over time in five key areas of K-12 foreign language education: amount of language instruction; languages and program types offered; curriculum and instruction; teacher certification, professional development, and teacher shortages; and the effects of education reform. Results reveal that despite some positive developments, overall foreign language instruction has decreased over the past decade and the achievement gap has widened. The article concludes with recommendations on how to progress toward achieving foreign language proficiency for all students. (Contains 7 notes, 7 figures, and 5 tables.)

Nonprofit family-run organizations, such as Tennessee Voices for Children (TVC), are providing leadership in advocating for and delivering services to children and families in need. Utilizing a family-driven approach and a staff partially comprised of parent-professionals, TVC's Nashville Connection and Family Connection programs have strengthened families by providing alternatives to state custody for children and families living with serious emotional or behavioral problems. TVC's Nashville Connection and Family Connection programs did this by coordinating support services, building community bridges, and providing comprehensive in-home services. (Contains 1 table.)

Nandu, a listserv for elementary school foreign language teachers, was designed to address the need for peer discussion and up-to-date resources. In this article, the authors report on the findings of a small-scale study of what Nandu listserv participants discuss. Analyses of postings during a six-month period showed that teachers use the listserv to resolve problems that they face in their daily practice, and when conflict arises, they resolve it amicably and (as they explicitly claim) learn from it. The findings of this study will be of interest to practitioners and teacher educators who want to learn more about the day-to-day concerns of foreign language teachers in order to provide relevant resources and staff development. Current and prospective teachers will find that listserv participation offers not only timely information, but also the opportunity to re-examine their assumptions through collegial discussion. Numerous reports and articles have decried the mediocrity of U.S. students' foreign language skills and have called for improved language education. One study that looked at foreign language education in 19 countries suggests several characteristics of successful foreign language programs: starting early, providing an articulated framework, treating language as a core subject, having strong supportive leaders, focusing on professional development, actively promoting proficiency, and nourishing heritage languages. Both Europe and Canada take language learning seriously: The European Union's 2004-2006 action plan asserts that multilingualism is a basic skill for all European citizens. Canada's action plan for foreign language education promotes the use of English and French, the two official languages. (Contains 1 figure.)

Patients with pancreatic cancer have dismal prognoses, and novel therapies are urgently needed. Mutations of the KRAS oncogene occur frequently in pancreatic cancer and represent an attractive target. Direct targeting of the predominant KRAS pathways have been challenging and research into therapeutic strategies have been now refocused on pathways downstream of KRAS, phosphoinositide 3-kinase (PI3K) and mitogen-activated protein kinase (MAPK [MEK]). We hypothesized that concurrent inhibition of the PI3K and MEK pathways would result in synergistic antitumor activity, as it would circumvent the compensatory feedback loop between the two pathways. We investigated the combined effect of the PI3K inhibitor, GDC0941, and the MEK inhibitor, AZD6244, on cell viability, apoptosis and cell signaling in a panel of pancreatic cancer cell lines. An in vivo analysis was conducted on pancreatic cancer xenografts. While BxPC-3 (KRAS wild type) and MIA PaCa-2 (KRAS mutated) cell lines were sensitive to GDC0941 and AZD6244 as single agents, synergistic inhibition of tumor cell growth and induction of apoptosis were observed in both cell lines when the two drugs were combined. Interestingly, phosphorylation of the cap-dependent translational components, 4E-binding protein (p-4E-BP1) and S6 was found to be closely associated with sensitivity to GDC0941 and AZD6244. In BxPC-3 cell xenografts, survival differences were observed between the control and the AZD6244, GDC0941, and combination groups. Our study provides the rationale for concurrent targeting of the PI3K and MEK pathways, regardless of KRAS status, and suggests that phosphorylation of 4E-BP1and S6 can serve as a predictive biomarker for response to treatment.

To gain a better idea of what other countries were doing in foreign language education, the Center for Applied Linguistics (CAL) collected information about policies and practices in 19 countries in diverse regions of the world. The exploratory study found that successful language programs shared common features that the U.S. could learn from: they start language instruction early, maintain a well-articulated framework, provide strong leadership, offer language as a core subject, provide quality teacher education, teach language through content, use technology creatively, and support heritage language instruction.

5-Lipoxygenase (5-LO) catalyzes the two initial steps in the biosynthesis of leukotrienes (LT), a group of inflammatory lipid mediators derived from arachidonic acid. Here, we investigated the regulation of 5-LO mRNA expression by alternative splicing and nonsense-mediated mRNA decay (NMD). In the present study, we report the identification of 2 truncated transcripts and 4 novel 5-LO splice variants containing premature termination codons (PTC). The characterization of one of the splice variants, 5-LOΔ3, revealed that it is a target for NMD since knockdown of the NMD factors UPF1, UPF2 and UPF3b in the human monocytic cell line Mono Mac 6 (MM6) altered the expression of 5-LOΔ3 mRNA up to 2-fold in a cell differentiation-dependent manner suggesting that cell differentiation alters the composition or function of the NMD complex. In contrast, the mature 5-LO mRNA transcript was not affected by UPF knockdown. Thus, the data suggest that the coupling of alternative splicing and NMD is involved in the regulation of 5-LO gene expression.

Phosphorus is the central ingredient in fertilizer that allows modern agriculture to feed the world's population. This element, also critical in a host of industrial applications, is a nonrenewable resource that is sourced primarily from the phosphatic mineral apatite, hosted in sedimentary and igneous ores. World phosphate resources are estimated by the U.S. Geological Survey at ca. 300,000 Mt, of which 95% are sedimentary and 5% are igneous. Current known USGS reserve estimates are sufficient for a maximum of 200 to 300 years; the exploration and discovery of new resources, enhanced mining technologies, and new technologies aimed at the recovery and recycling of P from sewage and agricultural runoff will all contribute to extending P production. Igneous ores are generally associated with Phanerozoic carbonatites and silica-deficient alkalic intrusions that typically average 5 to 15 wt % P2O5, which can be beneficiated to high-grade concentrates of at least 30 wt % P2O5 with few contaminants. Carbonatites are typically the smallest and youngest parts of a carbonatite-alkaline rock complex that formed during fractional crystallization of a calcic parental alkaline silicate melt, or from liquid immiscibility of a carbonate-rich nephelinite that underwent magmatic fractionation and differentiation during ascent from the mantle source. Fluorapatite generally crystallizes early, near the liquidus, and over a small temperature interval below the apatite saturation temperature that varies strongly with temperature, SiCL and CaO concentrations, and the aluminosity of the melt. Carbonatite-alkaline rock complexes commonly possess a concentric, zonal structure thought to reflect caldera volcanism. Pathfinder elements in soils, sediments, tills, and vegetation include Nb, rare earth elements (REEs), P, Ba, Sr, F, U, and Th, and in water, F, Th, and U are indicators. Remote sensing techniques with the ability to identify minerals rich in CO3, REEs, and Fe2+ that are characteristic of carbonatites are also important exploration tools that may provide vectors to ore. Sedimentary phosphorite is a marine bioelemental sedimentary rock that contains >18 wt % P2O5. While small peritidal phosphorites formed in Precambrian coastal environments, economically significant upwelling- related phosphorite did not accumulate until the late Neoproterozoic and continued through the Phanerozoic. Coastal upwelling delivered deep, P-rich waters to continental shelves and in epeiric seas to drive phospho-genesis and form the largest phosphorites on Earth. High-grade deposits formed as a result of hydraulic concentration of phosphate grains to form granular beds with minimal gangue. The amalgamation of these beds into decameter-thick, stratiform ore zones is generally focused along the maximum flooding surface, which is a primary exploration target in upwelling-related phosphorite. In addition to P, other elements concentrated in igneous and sedimentary phosphorites are Se, Mo, Zn, Cu, and Cr, which are important agricultural micronutrients. Other saleable by-products include U and REEs. The U concentration in sedimentary phosphorite is generally between 50 and 200 ppm, but can be as high as 3,000 ppm, making it an increasingly important source of U for the nuclear industry. The concentration of REEs in some sedimentary phosphorites is comparable to the world's richest igneous and Chinese clay-type REE deposits. The source of the dissolved P in upwelling ocean water is ultimately derived from the chemical weathering of continental rocks, the process that links igneous and sedimentary phosphorites through time and space. The covarying temporal relationship of igneous and sedimentary deposits suggests that plate tectonics and the concentration of apatite in a progressively more felsic crust underpins the feedback processes regulating the biogeochemical cycling of P Critical to the generation of greenfield exploration targets is the recognition that large P deposits emerged in the late Neoproterozoic. The geological environments conducive for exploration can be constrained from an understanding of ore-forming processes by the use of complementary petrological techniques, including fieldwork, petrography, sedimentology, sequence stratigraphy, and geochemistry.
