Abstract

An increasing number of elderly individuals are experiencing postoperative cognitive dysfunction (POCD) problems after undergoing hip replacement surgery, with gut microbiota metabolites playing a role in its pathogenesis. Among these, the specific effects of trimethylamine N-oxide (TMAO) on POCD are still unclear. This study aimed to explore the role of TMAO on cognitive dysfunction and underlying mechanisms in mice. The POCD model was created through femoral fracture surgery in elderly mice, followed by cognitive function assessments using the Morris Water Maze and Novel Object Recognition tests. The gut microbiota depletion and fecal microbiota transplantation were performed to examine the relationship between TMAO levels and cognitive outcomes. The effects of TMAO treatment on cognitive dysfunction, microglial activation, and inflammatory cytokine levels in the brain were also evaluated, with additional assessment of the role of microglial ablation in reducing TMAO-induced cognitive impairment. Elevated TMAO levels were found to be associated with cognitive decline in mice following femoral fracture surgery, with gut microbiota depletion mitigating both TMAO elevation and cognitive dysfunction. In contrast, fecal microbiota transplantation from postoperative mice resulted in accelerated cognitive dysfunction and TMAO accumulation in germ-free mice. Furthermore, TMAO treatment worsened cognitive deficits, neuroinflammation, and promoted microglial activation, which were reversed through the ablation of microglia. TMAO exacerbates cognitive dysfunction and neuroinflammation in POCD mice, with microglial activation playing a crucial role in this process. 3. [Evaluation of gap-filling methods for CH 4 flux data based on eddy covariance method in the Lake Taihu, China].

Abstract

Eddy covariance method has become a key technique to measure CH 4 flux continuously in lakes. A large number of CH 4 flux data was missing due to variable reasons. In order to reconstruct a complete time series of CH 4 flux, it is necessary to find an appropriate gap-filling method to insert the CH 4 flux data gap. Based on the routine meteorological data and CH 4 flux data measured at Bifenggang site in the eastern part of the Taihu eddy flux network during 2014 to 2017, we analyzed the control factors of CH 4 flux at the half-hour scale and daily scale. With those data, we tested that whether nonlinear regression method and two machine learning methods, random forest algorithm and error back propagation algorithm, could fill the CH 4 flux gap at the half-hour scale and daily scale. The results showed that CH 4 flux at the half-hour scale was mainly influenced by sediment temperature, friction velocity, air temperature, relative humidity, latent heat flux and water temperature at 20 cm in the growing season, and was mainly affected by relative humidity, latent heat flux, wind speed, sensible heat flux and sediment temperature in non-growing season. The CH 4 flux at the daily scale was mainly affected by latent heat flux and relative humidity. Random forest model was the best in CH 4 flux data gap filling at both time scales. The random forest model with the input variables of day of year, solar elevation angle, sediment temperature, friction velocity, air temperature, water temperature at 20 cm, relative humidity, air pressure, and wind speed was more suitable for filling the CH 4 flux data gap at the half-hour scale. The random forest model with the input variables of day of year, sediment temperature, friction velocity, air temperature, water temperature at 20 cm, relative humidity, air pressure, wind speed, and downward shortwave radiation was more suitable for filling CH 4 flux data gap at the day scale. The interpolation models could fill the data gap better at daily scale than that at the half-hour scale.

4. [Effects of chamber characteristics on CO 2 and CH 4 flux at the water-air interface measured by the chamber method].

Abstract

The chamber method is widely used to measure CO 2 and CH 4 flux in inland water. However, the designs of chamber used in various studies are different and lack unified standards, which would affect the observation results. To clarify the impacts of chamber characteristics, including light transmittance, air pressure difference inside and outside the chamber, and gas mixing degree in the chamber, on CO 2 and CH 4 flux measurements at the water-air interface, we compared the effects of transparent/opaque chamber, the chamber with/without air pressure equalizing device and fan on CO 2 and CH 4 flux measurements in the aquaculture pond, based on the multi-channel closed dynamic chamber system. The results showed that, during the daytime in summer, compared with the transparent chamber which could measure the actual CO 2 flux, when CO 2 was emitted from the pond, the opaque chamber overestimated the CO 2 flux by 90%; when CO 2 was absorbed by the pond, the opaque chamber underestimated the CO 2 flux by 50%. The CH 4 diffusion flux measured by the opaque chamber was 40% lower than that measured by the transparent chamber. There was no significant difference between CO 2 and CH 4 flux measured by the chamber with and without air pressure equalizing device. CO 2 flux observed by the chamber without fan had poor representativeness, being 20% higher than that observed by the chamber with fan. Moreover, CH 4 flux emitted through different pathways could not be distinguished using the chamber without fan. Therefore, when the chamber method was used to observe the CO 2 and CH 4 flux at the water-air interface, the chamber shall be transparent and be installed with fan.

5. Amelioration of ligamentum flavum hypertrophy using umbilical cord mesenchymal stromal cell-derived extracellular vesicles.

Abstract

Ligamentum flavum (LF) hypertrophy (LFH) has been recognised as one of the key contributors to lumbar spinal stenosis. Currently, no effective methods are available to ameliorate this hypertrophy. In this study, human umbilical cord mesenchymal stromal cell-derived extracellular vesicles (hUCMSC-EVs) were introduced for the first time as promising vehicles for drug delivery to treat LFH. The downregulation of miR-146a-5p and miR-221-3p expressions in human LF tissues negatively correlated with increased LF thickness. The hUCMSC-EVs enriched with these two miRNAs significantly suppressed LFH in vivo and notably ameliorated the progression of transforming growth factor β1(TGF-β1)-induced fibrosis in vitro after delivering these two miRNAs to mouse LF cells. The results further demonstrated that miR-146a-5p and miR-221-3p directly bonded to the 3'-UTR regions of SMAD4 mRNA, thereby inhibiting the TGF-β/SMAD4 signalling pathway. Therefore, this translational study determined the effectiveness of a hUCMSC-EVs-based approach for the treatment of LFH and revealed the critical target of miR-146a-5p and miR-221-3p. 6. [Analysis of Greenhouse Gas Emission Characteristics and Their Influencing Factors in the Algae Zone of Lake Taihu].

Abstract

In order to identify CH 4 and CO 2 emission flux characteristics and their impact factors in the algal lake zone of Lake Taihu, CH 4 and CO 2 fluxes were observed by the improved closed chamber method in Meiliang Bay in Lake Taihu. The relationships between CH 4 and CO 2 flux and meteorological factors were analyzed. The results showed that CH 4 and CO 2 fluxes had obvious diurnal variations. The CH 4 flux in the daytime was higher than that in the nighttime in spring; however, the CH 4 flux in the nighttime was higher than that in the daytime in summer. The CO 2 uptake flux in the daytime was higher than that in the nighttime in spring and summer. The algae zone of Lake Taihu was a CH 4 source for the atmosphere. The average CH 4 flux was 4.047 nmol ·(m 2 ·s) -1 and 40.779 nmol ·(m 2 ·s) -1 in spring and summer, respectively. The zone was the CO 2 sink for the atmosphere in spring and summer. The average CO 2 flux was -0.160 μmol ·(m 2 ·s) -1 and -0.033 μmol ·(m 2 ·s) -1 in spring and summer, respectively. On an hourly scale, the CH 4 emission flux was positively correlated with air temperature and water temperature ( r =0.20, P <0.01 and r =0.34, P <0.01, respectively). When wind speed was lower than 6 m ·s -1 , the CH 4 flux was positively correlated with wind speed ( r =0.71, P <0.01). The CO 2 uptake flux had a significant positive correlation with air temperature and wind speed ( r =0.14, P <0.01 and r =0.33, P <0.05, respectively). However, the CO 2 uptake flux was negatively correlated with air pressure and solar radiation ( r =-0.41, P <0.01 and r =-0.35, P <0.01, respectively). The CO 2 efflux had a significant positive correlation with wind speed ( r =0.40, P <0.05). The CO 2 efflux was negatively correlated with solar radiation ( r =-0.35, P <0.01). On a daily scale, the CH 4 emission flux had a significant positive correlation with air temperature and water temperature ( r =0.83, P <0.01 and r =0.78, P <0.01, respectively).

Abstract

We report a study of the process e^{+}e^{-}→(D^{*}D[over ¯]^{*})^{0}π^{0} using e^{+}e^{-} collision data samples with integrated luminosities of 1092 pb^{-1} at sqrt[s]=4.23 GeV and 826 pb^{-1} at sqrt[s]=4.26 GeV collected with the BESIII detector at the BEPCII storage ring. We observe a new neutral structure near the (D^{*}D[over ¯]^{*})^{0} mass threshold in the π^{0} recoil mass spectrum, which we denote as Z&#95;{c}(4025)^{0}. Assuming a Breit-Wigner line shape, its pole mass and pole width are determined to be (4025.5&#95;{-4.7}^{+2.0}±3.1) MeV/c^{2} and (23.0±6.0±1.0) MeV, respectively. The Born cross sections of e^{+}e^{-}→Z&#95;{c}(4025)^{0}π^{0}→(D^{*}D[over ¯]^{*})^{0}π^{0} are measured to be (61.6±8.2±9.0) pb at sqrt[s]=4.23 GeV and (43.4±8.0±5.4) pb at sqrt[s]=4.26 GeV. The first uncertainties are statistical and the second are systematic.

Abstract

Using a data sample collected with the BESIII detector operating at the BEPCII storage ring, we observe a new neutral state Z&#95;{c}(3900)^{0} with a significance of 10.4σ. The mass and width are measured to be 3894.8±2.3±3.2  MeV/c^{2} and 29.6±8.2±8.2  MeV, respectively, where the first error is statistical and the second systematic. The Born cross section for e^{+}e^{-}→π^{0}π^{0}J/ψ and the fraction of it attributable to π^{0}Z&#95;{c}(3900)^{0}→π^{0}π^{0}J/ψ in the range E&#95;{c.m.}=4.19-4.42  GeV are also determined. We interpret this state as the neutral partner of the four-quark candidate Z&#95;{c}(3900)^{±}.

Abstract

We report an observation of the process J/ψ→γX(1835)→γK&#95;{S}^{0}K&#95;{S}^{0}η at low K&#95;{S}^{0}K&#95;{S}^{0} mass with a statistical significance larger than 12.9σ using a data sample of 1.31×10^{9} J/ψ events collected with the BESIII detector. In this region of phase space the K&#95;{S}^{0}K&#95;{S}^{0} system is dominantly produced through the f&#95;{0}(980). By performing a partial wave analysis, we determine the spin parity of the X(1835) to be J^{PC}=0^{-+}. The mass and width of the observed X(1835) are 1844±9(stat)&#95;{-25}^{+16}(syst)  MeV/c^{2} and 192&#95;{-17}^{+20}(stat)&#95;{-43}^{+62}(syst)  MeV, respectively, which are consistent with the results obtained by BESIII in the channel J/ψ→γπ^{+}π^{-}η^{'}.

Abstract

We report the observation of the X(3823) state in the process e+e-→π+π-X(3823)→π+π-γχc1 with a statistical significance of 6.2σ, in data samples at center-of-mass energies sqrt[s]=4.230, 4.260, 4.360, 4.420, and 4.600 GeV collected with the BESIII detector at the BEPCII electron positron collider. The measured mass of the X(3823) state is (3821.7±1.3±0.7)  MeV/c2, where the first error is statistical and the second systematic, and the width is less than 16 MeV at the 90% confidence level. The products of the Born cross sections for e+e-→π+π-X(3823) and the branching ratio B[X(3823)→γχc1,c2] are also measured. These measurements are in good agreement with the assignment of the X(3823) state as the ψ(1 3D2) charmonium state.

Abstract

Based on data samples collected with the BESIII detector at the BEPCII collider at nine center of mass energies from 4.21 to 4.42 GeV, we search for the production of e^{+}e^{-}→ωχ&#95;{cJ} (J=0, 1, 2). The process e^{+}e^{-}→ωχ&#95;{c0} is observed for the first time, and the Born cross sections at sqrt[s]=4.23 and 4.26 GeV are measured to be (55.4±6.0±5.9) and (23.7±5.3±3.5)  pb, respectively, where the first uncertainties are statistical and the second are systematic. The ωχ&#95;{c0} signals at the other seven energies and the e^{+}e^{-}→ωχ&#95;{c1} and ωχ&#95;{c2} signals are not significant, and the upper limits on the cross sections are determined. By examining the ωχ&#95;{c0} cross section as a function of center of mass energy, we find that it is inconsistent with the line shape of the Y(4260) observed in e^{+}e^{-}→π^{+}π^{-}J/ψ. Assuming the ωχ&#95;{c0} signals come from a single resonance, we extract the mass and width of the resonance to be (4230±8±6)  MeV/c^{2} and (38±12±2)  MeV, respectively, and the statistical significance is more than 9σ.

Abstract

Using data collected with the BESIII detector operating at the Beijing Electron Positron Collider at center-of-mass energies of sqrt[s]=4.23, 4.26, and 4.36 GeV, we observe e(+)e(-)→π(0)π(0)hc for the first time. The Born cross sections are measured and found to be about half of those of e(+)e(-)→π(+)π(-)hc within less than 2σ. In the π(0)hc mass spectrum, a structure at 4.02  GeV/c(2) is found. It is most likely to be the neutral isospin partner of the Zc(4020)(±) observed in the process of e(+)e(-)→π(+)π(-)hc being found. A fit to the π(0)hc invariant mass spectrum, with the width of the Zc(4020)(0) fixed to that of its charged isospin partner and possible interferences with non-Zc(4020)(0) amplitudes neglected, gives a mass of (4023.9±2.2±3.8)  MeV/c(2) for the Zc(4020)(0), where the first error is statistical and the second systematic.

13. [A microfabricated ICP source base on planar spiral-shaped coil].

Abstract

Along with the development of micro-electromechanical system technology, the reports on the optical splitting systems and the photoelectric detection systems of spectrometer based on micro-electromechanical system have become increasingly popular, whereas the reports on micro excitation source in the development of micro spectrometer are few. In other words, the development of micro excitation source is the most important part in the development of the micro spectrometer. A novel low-pressure microfabricated inductively coupled plasma source is introduced, which is an emission spectrum excitation source based on surface-micromaching technology. Its radio frequency power consumption is much lower than the general inductively coupled plasma source. The source can work at an argon gas pressure of 100 Pa, and thus consumes less argon. The principle of operation for a microfabricated inductively coupled plasma source is illustrated. The layout of the planar spiral-shaped coil, the matched capacitor and the resonant capacitor is given. The fabrication process and properties of the plasma source are described. Meanwhile a novel inductively coupled plasma based on the technology of printed circuit board is introduced. In the experiment, the planar spiral-shaped coil and interdigatial capacitor were used. When the pressure of argon was 100 Pa and the radio frequency power was 3. 5 W at 13. 56 MHz the novel inductively coupled plasma was ignited. The photo of the argon ignition is given. Finally the potential application of the micro inductively coupled plasma in spectrometers is presented.
