Scopelogadus bispinosus has been redescribed, its species status has been restored. It has long been considered a subspecies of S. mizolepis. The species inhabits mainly the tropical and subtropical zones of the central and eastern parts of the Pacific Ocean; in the northeastern Pacific Ocean, it is found in temperate waters. A new species S. perplexus sp. n. has been described from the tropical waters of the Pacific Ocean. The new species is characterized by the presence of a large number filaments of pseudobranch (10–11), the largest of the two branchial lobes opposite the angular gill raker is relatively short; the color of the stomach surface is uniformly dark, without noticeable darkening in the back.

“Diffused neuroendocrine system (DNES)” includes a diversity of paraneuronal cells which are known to be scattered throughout the vertebrate body. They are well known to secrete a plethora of neurochemicals that act in autocrine and/or paracrine mode. Pseudobranchial neurosecretory system is one such DNES. It is found uniformly in all the catfishes and some other groups of non-catfish teleosts. However, it was not found in carps so far. The present investigation is the first time report of the presence of this system in a carp Barilius bendelisis. In catfish, the system is present in association with carotid labyrinth—a chemosensory structure known in fish and amphibians. The carotid labyrinth is a derived structure of pseudobranch. Hence, the revelation of the pseudobranchial neurosecretory system in association with pseudobranch in B. bendelisis has significant evolutionary implications.

Spiracles are a general character of gnathostomes (jawed fishes), being present in antiarch placoderms, commonly regarded as the most basal gnathostome group. The presence of spiracular tubes in acanthodians has been deduced from grooves on the neurocranium of the derived acanthodiform Acanthodes bronni from the Permian of Germany, but until now these tubes were presumed to lack an external opening, rendering them non‐functional. Here we describe the external spiracular elements in specimens of the Middle Devonian acanthodiforms Cheiracanthus murchisoni, Cheiracanthus latus and Mesacanthus pusillus from northern Scotland, and the internal structure of these elements in C. murchisoni, demonstrating that the spiracle in acanthodiforms differed from all known extant and extinct fishes in having paired cartilage‐pseudobranch structures. This arrangement represents a transitional state between the presumed basal gnathostome condition with an unconstricted first gill slit (as yet not identified in any fossil) and the derived condition with a spiracle and a single pseudobranch derived from the posterior hemibranch of the mandibular arch. We identify the main tissue forming the pseudobranch as elastic cartilage, a tissue previously unrecorded in fossils.

The pseudobranch is a gill-like epithelial elaboration that sits behind the jaw of most fishes. This structure was classically regarded as a vestige of the ancestral gill-arch like condition of the gnathostome jaw. However, more recently, hypotheses of jaw evolution by transformation of a gill arch have been challenged, and the pseudobranch has alternatively been considered a specialised derivative of the second (hyoid) pharyngeal arch. Here, we demonstrate by cell lineage tracing in a cartilaginous fish, the skate (Leucoraja erinacea), that the pseudobranch does, in fact, derive from the mandibular arch, and that it shares gene expression features and cell types with gills. We also show that the mandibular arch pseudobranch is supported by a spiracular cartilage that is patterned by ashh-expressing epithelial signalling centre. This closely parallels the condition seen in the gill arches, where cartilaginous appendages called branchial rays supporting the respiratory lamellae of the gills are patterned by ashh-expressing gill arch epithelial ridge (GAER). Taken together, these findings support serial homology of the pseudobranch and gills, and an ancestral origin of gill arch-like anatomical features from the gnathostome mandibular arch.

In hilsa (Tenualosa ilisha), pseudobranch comprises a row of parallel filaments bear numerous leaf-like lamellae arranged on both sides throughout its length. The purpose of this study was to elucidate involvement of pseudobranchial Na+, K+-ATPase (NKA) 1 α-subunit, and carbonic anhydrase (CA) in concert with H+-ATPase (HAT) compared to their branchial counterparts in freshwater acclimation of hilsa during spawning migration from off-shore of the Bay of Bengal to the Bhagirathi-Hooghly zones of the Ganga river system in India. Adult hilsa fish were collected from seawater (SW), freshwater 1 (FW1), and freshwater 2 (FW2) locations, where the salinity level was 26–28‰, 1–5‰, and 0–0.04‰, respectively. Hilsa migrating through freshwater showed a consistent decrease in the plasma osmolality, sodium (Na+) and chloride (Cl−) ion levels indicates unstable ionic homeostasis. The mRNA expression and activity of NKA 1 α-subunit in pseudobranch as well as in true gills declined with the migration to upstream locations. The pseudobranchial CA activity almost mirrors its branchial counterpart most notably while hilsa entered the freshwater zone, in the upstream river suggesting its diverse role in hypo-osmotic regulatory acclimation. Nevertheless, the H+-ATPase activity of both the tissues increased with the freshwater entry and remained similar during up-river movement into the freshwater environment. The results confirm that the pseudobranchial NKA 1 α-subunit mRNA expression and activity mimic its branchial counterpart in the process of ionoregulatory acclimation during migration through salt barriers. Also, the increase in the activities of pseudobranchial and branchial CA in concert with H+-ATPase (HAT) during freshwater acclimation of hilsa suggests their critical involvement in ion uptake.

Betta sp. is a freshwater ornamental fish which also known as a fighting fish. One of the fundamental organs to support fishes life is respiratory organ. Fighting fish is belongs to the suborder Anabantoidei which means labyrinth fishes group. The aim of the study was to know histology of the respiratory organs of Betta sp. Histological preparations were done using paraffin method, stained with Hematoxylin-Eosin (HE). The result showed that Betta sp. has a respiratory organ common fish i.e gills and additional respiratory organ structure namely labyrinth and pseudobranch that makes Betta sp can survive in a low volume of water. The gill is consists of gill arch, gill raker, gill fillament and gill lamellae. The labyrinth is consist of connective tissue and folded ephitelium. Pseudobranch according to some literature function as an additional respiratory. Functions attributed to the pseudobranch include; regulation of oxygen to the eyes, enzyme production for use in the gas bladder, osmoregulation, and many others.

The organization of the roots, ganglia and the peripheral distribution of the cranial nerves of the fully formed embryos of Oreochromis niloticus are examined in the transverse serial sections. These nerves carry fibers, which were also analyzed. The results of this study demonstrated that the glossopharyngeal nerve originates by means of only one root, which leaves the cranium through the glossopharyngeal foramen. This nerve gives fibers (visceromotor) to the first internal and external levator arcus branchialis muscles. There is a single epibranchial (petrosal) ganglion located extracranially. Nervus glossopharyngeus has three rami; pharyngeus, pretramticus and posttrematicus. The ramus pharyngeus carries only viscerosensory fibers; general for the pharyngeal epithelium and special ones for the pseudobranch. General viscerosensory fibers are also carried by rami pretrematicus and posttrematicus for the pharyngeal epithelial lining. The special sensory fibers are carried by the ramus pretrematicus for the taste buds and by ramus posttrematicus for the gill filaments. The ramus pretrematicus also carries visceromotor fibers for the first adductor arcus branchialis and to the first obliquus ventralis muscles. Resumo A organização das raízes, gânglios e a distribuição periférica dos nervos cranianos dos embriões totalmente formados de Oreochromis niloticus são examinados nas seções transversais seriais. Esses nervos carregam fibras, que também foram analisadas. Os resultados deste estudo demonstraram que o nervo glossofaríngeo se origina por meio de apenas uma raiz, que sai do crânio pelo forame glossofaríngeo. Este nervo fornece fibras (visceromotoras) para os primeiros músculos levantadores do arco branquial interno e externo. Existe um único gânglio epibranquial (petroso) localizado extracranialmente. Nervus glossopharyngeus tem três ramos; faríngeo, pretramticus e póstrematicus. O ramo faríngeo contém apenas fibras viscerossensoriais — gerais para o epitélio faríngeo e especiais para o pseudobrânquio. Fibras viscerossensoriais gerais também são transportadas por ramos pretrematicus e posttrematicus para o revestimento epitelial da faringe. As fibras sensoriais especiais são transportadas pelo ramus pretrematicus para as papilas gustativas e pelo ramus posttrematicus para os filamentos branquiais. O ramo pretrematicus também carrega fibras visceromotoras para o primeiro adutor arcus branchialis e para o primeiro músculo oblíquo ventral.

Warm water treatment, i.e. exposure to sea water at a temperature of 28–34 °C for 20–30 s, has in recent years been widely used for delousing of Atlantic salmon (Salmo salar) and rainbow trout (Oncorhynchus mykiss) in Norwegian aquaculture. High mortality and various lesions (e.g. injuries and/or bleedings in skin, fins, eyes, brain, and gills) have, however, been reported after industrial warm water treatments. The objective of this study was to reveal whether the thermal component of warm water treatment inflicts acute lesions on Atlantic salmon. The study was conducted by exposing individual, sedated Atlantic salmon post-smolts ( w ¯ = 1117 ± 250 g) to sea water at a temperature of 34 °C (warm water treatment, n = 40) or 9 °C (control treatment, n = 20) for 30 s, and subsequently conducting welfare indicator scoring and histopathological examination of their skin, fins, eyes, snout, nasal pits/mucosa, palate, gills, thymus, pseudobranch, brain, heart, liver, kidney, pyloric caeca, pancreas, and spleen. The results showed that the prevalence and severity of acute lesions were not significantly different between the two treatment groups, except for higher prevalence of injuries on the caudal (p = 0.002), dorsal (p = 0.002), and right pelvic fins (p = 0.014) in the warm water treatment group. The main cause of these fin injuries may have been a strong behavioural reaction displayed by the fish when exposed to warm water. Possible consequences of fin injuries, the use of anaesthetic, and statistical limitations were discussed. It was concluded that exposure of Atlantic salmon to sea water at a temperature of 34 °C for 30 s did not lead to any statistically significant change in the prevalence of acute lesions except an increase in minor, possibly behaviour-related, fin injuries. Detection of a lower lesion prevalence than was possible in this study, but which may concern many individuals in an industrial setting, requires examination of a larger number of fish.

The spiracle of elasmobranchs (sharks, skates, and rays) is a gill-slit-derived tube located behind the eye. Its inner structure was well studied in the late nineteenth to early twentieth century, but its entire morphology has rarely been characterized and is poorly understood. The present study shows the three-dimensional morphology of the spiracular tube for the first time, using resin injection and CT scanning, in the Japanese bullhead shark. The spiracular tube is characterized by the presence of two caeca (dorsal and ventral spiracular caeca) on the medial wall of the spiracular tube and the presence of a pseudobranch on the anterior wall. This study provides a basis for further studies on the morphological diversity, function, and evolution of spiracles in elasmobranch fishes. Anat Rec, 2018. © 2018 Wiley Periodicals, Inc.

Pseudobranchial neurosecretory system (PSNS) is the third Neuroendocrine (NE) system found in the gill region of fishes in close association with pseudobranch/carotid labyrinth/carotid gland and can suitably be placed under the category of "Diffused NE system (DNES)." The cells belonging to this system fall under the category of "Paraneurons," a concept proposed by Fujita and coworkers. It is found uniformly in all the catfish species and some other noncatfish group of teleosts as Atheriniformes, Channiformes, Perciformes, and Clupeiformes. The fishes, in which the PSNS is present, belong to different breathing habits. Most of these have the capacity to tolerate low O2 conditions. Leiodon cutcutia although not an air-breathing fish, is known to retain air in its stomach for varied periods when threatened. In an attempt to verify the veracity of this system in a fish of another peculiar breathing habit, ocellated puffer fish L. cutcutia (order Tetradontiformes) was investigated. The histological observations undertaken on L. cutcutia revealed the presence of a well-developed extrabranchial NE system. The findings are discussed in the light of the association of PSNS with chemosensory system and its evolution in fishes, especially in the view of the transition from aquatic to terrestrial life.

Various putative oxygen chemosensory cells are reported to be present throughout the vertebrate body performing pivotal roles in respiration by initiating responses during acute hypoxia. Since air-breathing fishes often are exposed to the oxygen-deficient milieu, in such conditions various chemosensory cells operate in an orchestrated fashion. The Pseudobranchial neurosecretory system (PSNS) a newly discovered system, is one of these. It has been placed in the category of "Diffuse NE systems (DNES)". It is found in all the catfish species and in some other non-catfish group of teleosts. In catfishes, it is present in close association with the carotid labyrinth- a chemosensory structure, known in fish and amphibians. The presence of this system in Glossogobius giuris, in association with the pseudobranch, a structure considered to be precursor of carotid labyrinth, is a significant finding. In an attempt to study the structure and organization of the pseudobranchial neurosecretory system in a non-catfish species of teleost, the present investigation was undertaken on a goby G. giuris. The histological observations, using a neurosecretion-specific stain, revealed the presence of this system in G. giuris. The findings are discussed in the light of the association of PSNS with pseudobranch and the type of "neurohaemal contact complex" formed between this neurosecretory system and the elements of the circulatory system.

The characteristics of the fish defense system, its relationship with environmental conditions and other factors are presented. In contrast to higher vertebrates the antibody response of fish is weaker and arises with a certain delay. The reaction can vary depending on the place in the body of a fish. There is a great diversity of lymphoid and lymphomyeloid organs in fish. In addition to the thymus, kidney and spleen, they include the Leydig organ, a tissue associated with the esophagus, the epigonal organ, linked to the gonads, the pericardial tissue, the orbital and the preorbital tissues, that are the tissues located in the region of the fish eyes. The mucosa-associated lymphoid tissue of fish is characterized by the lack of organized structures, dominance of specialized mucosa-IgT immunoglobulins and IgT + B cells, a variety of symbiotic bacteria that are overlaid by mucosal Igs of fish. Attention is paid to the interbranchial lymphoid tissue which was found only in salmonids fish and cutaneous immune system observed only in fish and amphibians. The role of various organs (pseudobranch, nasopharynx) and cells involved in immune reaction in fish is discussed.

The development of osmoregulatory and gas exchange organs was studied in larval yellowfin tuna (Thunnus albacares) from 2 to 25 days post-hatching (2.9-24.5 mm standard length, SL). Cutaneous and branchial ionocytes were identified using Na+/K+-ATPase immunostaining and scanning electron microscopy. Cutaneous ionocyte abundance significantly increased with SL, but a reduction in ionocyte size and density resulted in a significant decrease in relative ionocyte area. Cutaneous ionocytes in preflexion larvae had a wide apical opening with extended microvilli; however, microvilli retracted into an apical pit from flexion onward. Lamellae in the gill and pseudobranch were first detected ~ 3.3 mm SL. Ionocytes were always present on the gill arch, first appeared in the filaments and lamellae of the pseudobranch at 3.4 mm SL, and later in gill filaments at 4.2 mm SL, but were never observed in the gill lamellae. Unlike the cutaneous ionocytes, gill and pseudobranch ionocytes had a wide apical opening with extended microvilli throughout larval development. The interlamellar fusion, a specialized gill structure binding the lamellae of ram-ventilating fish, began forming by ~ 24.5 mm SL and contained ionocytes, a localization never before reported. Ionocytes were retained on the lamellar fusions and also found on the filament fusions of larger sub-adult yellowfin tuna; however, sub-adult gill ionocytes had apical pits. These results indicate a shift in gas exchange and NaCl secretion from the skin to branchial organs around the flexion stage, and reveal novel aspects of ionocyte localization and morphology in ram-ventilating fishes.

The jellyfishes of the genus Chrysaora are present in all of the world's oceans, but the toxicity of their venoms has not yet been thoroughly characterized. The zebrafish as a toxicology model can be used for general toxicity testing of drugs and the investigation of toxicological mechanisms. The aim of this study was to evaluate the effect of crude venom from jellyfish Chrysaora sp., a species of jellyfish observed in the tropical lagoons of the Gulf of Mexico, on the zebrafish Danio rerio. Juvenile zebrafish were injected with different concentrations of venom from Chrysaora sp. via intraperitoneal and subcutaneous injections. The effects of the venom were determined by histopathological analysis and through the measurement of hemolytic and phospholipase A2 activities. The crude venom was examined by SDS-PAGE. The effect of sublethal concentrations of crude venom from Chrysaora sp. on D. rerio was hemorrhaging in the eyes, while the histopathological analysis demonstrated that the primary organs targeted were the pseudobranch, which displayed hyperemia, and the gill, which displayed hyperplasia and hypertrophy. The blood analysis exhibited hemolysis, nuclear abnormalities, and echinocytes by the action of phospholipase A2, which was determined to have 596 units of activity/mg of protein in the venom. The crude venom has proteins with molecular weights ranging from 250 to 6 kDa, with more density in the bands corresponding to 70, 20 and 15 kDa. The venom of Chysaora sp. caused disturbances in circulation associated with vascular dilation due to the localized release of inflammatory mediators. The hemolysis of erythrocytes was caused by the action of phospholipase A2. These findings not only provide an excellent study model but also have a great pharmacological potential for designing new drugs and for the elucidation of the mechanisms of action of and treatment against stings.

The ultrastructure of the pseudobranch of the euryhaline, self-fertilizing fish, Rivulus marmoratus (Cyprinodontidae) was studied with thin sectio and freeze-fracture transmission electron microscopy. In specimens raised from birth in 1% or 200% seawater, the pseudobranch contains two mitochondria-rich cell types, pseudobranchial cells and chloride cells, each of which has an extensive tubular system. Chloride cells only occur on the lateral aspects of the pseudobranch, with their apical crypts open to the environment. Apical crypt invagination to a “pit” structure and multicellular complexes occur in both salinity extremes. The ultrastructure of chloride cells in 1% and 200% SW is consistent with that described previously in the opercular epithelium, opercular skin, and gill of this species; elaboration of mitochondria and basolateral membrane accompanies increased environmental salinity (King et al.: Cell and Tissue Research 257:367–377, 1989). Pseudobranchial cells constitute the majority of the cells; they do not extend to the surface but have a more organized tubular system that is continuous with the basal membrane. These cells do not exhibit ultrastructural changes in response to increased salinity. © 1993 Wiley-Liss, Inc.

Background The myxosporean parasite Parvicapsula pseudobranchicola commonly infects farmed Atlantic salmon in northern Norway. Heavy infections are associated with pseudobranch lesions, runting and mortality in the salmon populations. The life-cycle of the parasite is unknown, preventing controlled challenge experiments. The infection dynamics, duration of sporogony, tissue tropism and ability to develop immunity to the parasite in farmed Atlantic salmon is poorly known. We conducted a field experiment, aiming at examining these aspects. Methods Infections in a group of Atlantic salmon were followed from before sea-transfer to the end of the production (604 days). Samples from a range of tissues/sites were analysed using real-time RT-PCR and histology, including in situ hybridization. Results All salmon in the studied population rapidly became infected with P. pseudobranchicola after sea-transfer medio August. Parasite densities in the pseudobranchs peaked in winter (November-January), and decreased markedly to March. Densities thereafter decreased further. Parasite densities in other tissues were low. Parasite stages were initially found to be intravascular in the pseudobranch, but occurred extravascular in the pseudobranch tissue at 3 months post-sea-transfer. Mature spores appeared in the pseudobranchs in the period with high parasite densities in the winter (late November-January), and were released (i.e. disappeared from the fish) in the period January-March. Clinical signs of parvicapsulosis (December-early February) were associated with high parasite densities and inflammation in the pseudobranchs. No evidence for reinfection was seen the second autumn in sea. Conclusions The main site of the parasite in Atlantic salmon is the pseudobranchs. Blood stages occur, but parasite proliferation is primarily associated with extravascular stages in the pseudobranchs. Disease and mortality (parvicapsulosis) coincide with the completion of sporogony. Atlantic salmon appears to develop immunity to P. pseudobranchicola. Further studies should focus on the unknown life-cycle of the parasite, and the pathophysiological effects of the pseudobranch infection that also could affect the eyes and vision. International audience; Gene expression of key ion transporters (the Na+/K+-ATPase NKA, the Na+, K+-2Cl− cotransporter NKCC1, and CFTR) in the gills, opercular inner epithelium, and pseudobranch of European seabass juveniles (Dicentrarchus labrax) were studied after acute transfer up to 4 days from seawater (SW) to freshwater (FW). The functional remodeling of these organs was also studied. Handling stress (SW to SW transfer) rapidly induced a transcript level decrease for the three ion transporters in the gills and operculum. NKA and CFTR relative expression level were stable, but in the pseudobranch, NKCC1 transcript levels increased (up to 2.4-fold). Transfer to FW induced even more organ-specific responses. In the gills, a 1.8-fold increase for NKA transcript levels occurs within 4 days post transfer with also a general decrease for CFTR and NKCC1. In the operculum, transcript levels are only slightly modified. In the pseudobranch, there is a transient NKCC1 increase followed by 0.6-fold decrease and 0.8-fold CFTR decrease. FW transfer also induced a density decrease for the opercular ionocytes and goblet cells. Therefore, gills and operculum display similar trends in SW-fish but have different responses in FW-transferred fish. Also, the pseudobranch presents contrasting response both in SW and in FW, most probably due to the high density of a cell type that is morphologically and functionally different compared to the typical gill-type ionocyte. This pseudobranch-type ionocyte could be involved in blood acid-base regulation masking a minor osmotic regulatory capacity of this organ compared to the gills.

We describe organogenesis at a histological level in American shad (Alosa sapidissima) larvae from 0 until 45 days after hatching (DAH). Larval development was divided into four stages based on the feeding mode, external morphological features, and structural changes in the organs: stage 1 (0–2 DAH), stage 2 (3–5 DAH), stage 3 (6–26 DAH) and stage 4 (27–45 DAH). At early stage 2 (3 DAH), American shad larvae developed the initial digestive and absorptive tissues, including the mouth and anal opening, buccopharyngeal cavity, oesophagus, incipient stomach, anterior and posterior intestine, differentiated hepatocytes, and exocrine pancreas. The digestive and absorptive capacity developed further in stages 2 to 3, at which time the pharyngeal teeth, taste buds, gut mucosa folds, differentiated stomach, and gastric glands could be observed. Four defined compartments were discernible in the heart at 4 DAH. From 3 to 13 DAH, the excretory systems started to develop, accompanied by urinary bladder opening, the appearance and development of primordial pronephros, and the proliferation and convolution of renal tubules. Primordial gills were detected at 2 DAH, the pseudobranch was visible at 6 DAH, and the filaments and lamellae proliferated rapidly during stage 3. The primordial swim bladder was first observed at 2 DAH and started to inflate at 9 DAH; from then on, it expanded constantly. The spleen was first observed at 8 DAH and the thymus was evident at 12 DAH. From stage 4 onwards, most organs essentially manifested an increase in size, number, and complexity of tissue structure.

Living and fixed samples of Schistosoma mansoni -infected Biomphalaria glabrata snails were used to determine the relative contributions of different snail tissues to cercarial emergence (shedding). Three methods of observations were employed: (1) direct microscopical observations of shedding snails; (2) microscopic analysis of 5 μm serial sections (HE stained) of actively shedding snails; and (3) scanning electron microscopic (SEM) observations of snails that were fixed while actively shedding. For this investigation, there were advantages and disadvantages to using each method. We confirmed the results of others that there were 3 tissues of the snail that contributed most prominently to cercarial release (mantle collar, pseudobranch, and headfoot). Based on histological analysis of cercarial accumulations in presumed shedding sites in these 3 tissues, 57% of the cercariae could be seen in the mantle collar, 30.6% in the pseudobranch, and 12.5% in the headfoot. Other anterior structures were involved to a much lesser extent. SEM observations clearly showed cercariae emerging either body first, tail first, or likely emerging en masse from blebs, especially from the mantle collar. These studies provide a more quantitative appraisal of the role the different anterior snail tissues play in cercarial emergence.

The ontogenetic development of the immune system was studied during the egg phase and the early post-hatch period of rainbow trout. Quantitative real-time PCR (qPCR) was used to assess the timing and degree of expression of 9 important immune relevant genes and EF1-α. Further, immunohistochemical staining using monoclonal antibodies was applied on rainbow trout embryos and larvae in order to localize five different protein molecules (MHCII, CD8, IgM, IgT and SAA) in the developing tissue and immune organs. Maternally transferred transcripts of EF1-α mRNA were detected in the unfertilized egg. Early onset of expression was seen for all immune genes at very low levels. The amount of mRNA slowly increased and peaked around and after hatching. The highest increases were seen for MHCII, C3, C5 and SAA. Immunohistochemistry using five monoclonal antibodies showed positive staining from day 84 post fertilization. Skin, gills, intestine, pseudobranch and thymus showed reactivity for MHCII, thymus for CD8, gill mucus for IgT and pseudobranch and cartilage associated tissue for SAA. The importance of detected factors for early protection of eggs and larvae is discussed.

A new species of Neobathyclupea is described from two specimens collected off Myanmar and off Socotra Island. The new species is most similar to N. malayana, but differs from it and other congeners in having jet-black coloured fins, larger pseudobranch, dorsal-pterygiophore pattern and some body proportions. Neobathyclupea malayana is re-diagnosed. Individual variations in armament of the preopercle and in the dorsal-pterygiophore patterns within Bathyclupeidae are discussed.

The pseudobranchs of two euryhaline teleost species, the milkfish (Chanos chanos) and the Mozambique tilapia (Oreochromis mossambicus), were studied after acclimization to different salinities using optical and electron microscopy. The milkfish pseudobranch was the lamellae-free type, with separate lamellae along the filaments containing two groups of mitochondria (Mt)-rich cells: chloride cells (CCs) and pseudobranch type cells (PSCs). Conversely, the tilapia pseudobranch was the embedded type, covered with connective tissues and with only one group of Mt-rich PSCs. Chloride cells were identified according to the apical openings and branched tubular networks around randomly distributed and diversely shaped Mt. Pseudobranchs type cells, however, were characterized according to the orderly arrangement of parallel tubules around closely packed Mt; both the tubules and the Mt were distributed in the vascular side of the cell, but were absent from the apical region. Compared with those of seawater (SW)-acclimated milkfish, the pseudobranchial lamellae of freshwater (FW) specimens were longer on average, and the Mt of the CCs had fewer cristae, were less electron-dense, and were often vacuolated. The Mt in the PSCs of FW-acclimated milkfish and tilapia were larger and more electron-dense than those of their SW-acclimated counterparts; in addition, more tubules were found to aggregately surround the Mt and basolateral membranes in the PSCs of fish from the hypo-osmotic environment. Conversely, the PSCs of tilapia were periodic acid-Schiff (PAS)-positive, and Mt in PSCs were concentrated with more parallel arrays of the tubule system than those of milkfish. Therefore, salinity-dependent changes in the ultrastructures of PSCs suggest their potential role in energy metabolism of both lamellae-free and embedded pseudobranchs, whereas the PAS-positive staining characteristics suggest a role in releasing or storaging polysaccharides in the embedded pseudobranch. The genus Sebastes (rockfishes) is an ecologically diverse group of fish, more than 60 species occurring off the Oregon, Washington, and California coasts. As of 2004, seven species of rockfish were classified as overfished by the Pacific Fishery Management Council. Because rockfishes often experience barotrauma to varying degrees when forced up from depth, the management of discard mortality has been difficult. In this study, the macroscopic, morphological, and physiological responses to decompression of black rockfish S. melanops, blue rockfish S. mystinus, and yellowtail rockfish S. flavidus, all nearshore species, were investigated. The rockfish were adjusted to 4.5 atmospheres absolute (ATA; 35 m) over a period of 7–10 d in hyperbaric pressure chambers and when neutrally buoyant were rapidly brought to surface pressure in a simulated ascent. SUMMARYThe fish gill is a highly specialized and complex organ that performs a variety of important physiological functions. In this article, we briefly review the innervation of important structures of the branchial region, such as the gill filaments, respiratory lamellae and pseudobranch, and discuss the physiological significance of this innervation within the context of homeostatic functions of the gill, such as oxygen sensing and ion regulation. Studies in zebrafish utilizing techniques of confocal microscopy and immunolabelling, with specific antibodies against neuronal markers, have recently led to the characterization of innervation patterns in the gills not attained with traditional techniques of histochemistry and electron microscopy. We will discuss the association of putative sensory nerve fibres with O2-chemoreceptive neuroepithelial cells and the implications of dual sensory pathways for cardiorespiratory and vascular control. In addition, the idea of the neural control of ion regulation in the gill based on the apparent innervation of mitochondria-rich cells, and the role of innervation in the pseudobranch, will be presented.

The physiological relevance of the teleost pseudobranch as a remnant of a reduced gill arch is still unclear. Numerous hypotheses have been proposed regarding its physiological role, but direct confirmatory evidence is lacking. The close relationship by serial blood flow arrangement with the fish eye's choroid rete has sparked the idea that pseudobranchial preconditioning of blood pH may facilitate initiation of the Root effect and thus support the establishment of high oxygen tensions for retinal diffusive supply. This idea was critically tested by studies on isolated pseudobranchs in situ (Oncorhynchus mykiss), perfused with RBC/Ringer or RBC/plasma suspensions of widely varied composition (pH 7.4-8.2). Detailed analysis of inflowing as compared to effluent perfusates indicated normal aerobic metabolism expressed by a rise in Pco2 (+0.39 +/- 0.13 mmHg x +/- SD), an oxygen utilization of 25% and a high oxygen consumption of approximately 400 nmol g(-1 ) min(-1). Upon passage of the pseudobranch, pH (corrected for Haldane effect) was only slightly acidified (-0.03 to -0.10), [HCO3(-)] and [lactate] were slightly enhanced (+0.51 mmol l(-1) or 0.13 mmol l(-1), respectively). In order to test for yet unknown plasma components involved in pseudobranch function, a second series of experiments was conducted using RBC-suspensions in fresh plasma instead of Ringer, with results closely resembling those of the RBC/Ringer series. Lacking any physiologically significant correlation with the level of perfusate pH, the obtained data indicate pseudobranchial basic metabolic activity rather than pH regulatory characteristics. Also the observed absolute changes in pH are negligible in terms of pH regulation towards the Root-threshold. Accordingly, the present experiments as well as plausibility evaluation of mechanisms do not support the idea of blood pH pre-adjustment prior to entry into the choroid rete structure of the teleost eye to facilitate the Root-mediated oxygen release.

Diagnosis of SAV infections has traditionally been based upon clinical observations together with a set of histopathological findings in exocrine pancreas, heart and skeletal muscle, but recently, real-time RT-PCR assays have been developed as a supplement for the detection of SAV. The aim of this study was to determine tissue tropism of SAV1 and SAV3 in Atlantic salmon Salmo salar L. in order to identify the most suitable tissues for real-time RT-PCR diagnostic assays. The results indicated that the pseudobranch and the heart (ventricle) are the most useful tissues for such assays, regardless of disease status. The pyloric caecae with associated pancreatic tissue is unsuitable for diagnosis using this method. The use of real-time RT-PCR enabled viral RNA detection at all stages of the disease, including in surviving fish six months after infection. Considering the short production cycle of farmed salmonids, this suggests that surviving Atlantic salmon may become life-long asymptomatic carriers of SAV after an infection.

We previously reported that long-term (54 days), repeated intraperitoneal exposure to low doses of tributyltin (TBT; 0.3 mg/kg) inhibited the metabolic activation of co-administered benzo[a]pyrene (BaP; 3 mg/kg) in the Arctic charr (Salvelinus alpinus); BaP, in turn, stimulated the metabolism and/or excretion of TBT. Here, we report the results of histopathological examinations of liver, kidney and pseudobranch tissue samples originating from these same fish. The results revealed higher lesion incidences at all sampling time points (Days 8, 32 and 56) among BaP-exposed fish compared with fish exposed to either TBT alone or combined with BaP. The severity of lesions like necrosis was also higher in BaP-exposed fish. Moreover, hepatic basophilic foci were observed exclusively in fish exposed to BaP alone. Together, these results provide new evidences that TBT can antagonize BaP toxicity in fish exposed to both pollutants under controlled laboratory conditions. In contrast, BaP does not appear to provide protection against TBT toxicity.

Background: Parvicapsula pseudobranchicola (Myxozoa) causes widespread infections in farmed Atlantic salmon in northern Norway. Heavily infected salmon become runts, probably due to vision impairment or blindness. The salmon are likely infected by waterborne actinospores, released by an alternating annelid host, but the life cycle of P. pseudobranchicola is unknown. Seatrout and Arctic charr have been considered possible hosts for the parasite, but firm evidence has been lacking. Findings: We show for the first time the presence of mature spores of P. pseudobranchicola in seatrout. The seatrout were infected with high intensities of P. pseudobranchicola in the pseudobranchs in early April. The presence of mature spores in early spring suggests that the fish had been infected late the previous year, a pattern of infection similar to that observed for farmed salmon stocked in autumn. Although heavily infected, the fish did not display any symptoms consistent with parvicapsulosis. The results suggest that the life cycle of P. pseudobranchicola is more adapted to seatrout, rather than to Atlantic salmon. Conclusions: The presence of mature spores of P. pseudobranchicola in seatrout confirms that seatrout is a natural host for this myxosporean and this is also the first record of these spores in the pseudobranch of a wild salmonid. Furthermore, wild trout from non-farming areas may become heavily infected with P. pseudobranchicola, developing pseudobranch pathology resembling that of farmed Atlantic salmon suffering from parvicapsulosis

Studies of ion regulation by mitochondria-rich cells (MRCs) of transport epithelia in fish have revealed many processes by which ion homeostasis is achieved. However, the control of these mechanisms and, particularly, the extent of nervous system involvement are not completely understood. We characterized the potential innervation of MRCs in various gill and extrabranchial tissues involved in ion transport in the model vertebrate the zebrafish. Confocal and conventional microscopy of whole-mount preparations were combined with immunofluorescence techniques to label MRCs with antibodies against a subunit of the enzyme Na+/K+-ATPase and nerve fibers with a zebrafish neuronal marker, zn-12. MRCs of the gill filaments were identified by their morphology and migration out to the lamellae in response to ion-poor water acclimation. Gill MRCs were intimately associated with nerve fibers originating from outside the filaments. MRCs of the opercular epithelium resembled those of the gill and were also located adjacent to nerve fibers. Mitochondria-rich “pseudobranch cells” were identified in the pseudobranch by immunofluorescence and labeling of dissociated cells with the mitochondrial marker DASPEI. Pseudobranch MRCs resembled gill MRCs and received innervation from a dense network of nerve fibers. In larvae, MRCs were distributed across the surface of the skin. These cells were situated among a dense network of varicose nerve fibers, and some MRCs of the skin displayed extensive cytoplasmic processes. Evidence is presented suggestive of widespread association of MRCs with the nervous system in transport epithelia and the neural control of MRC-mediated ion regulation in teleost fish. J. Comp. SUMMARYAlthough the retina has one of the highest metabolic rates among tissues,certain teleost fishes lack any vascular supply to this organ which, in combination with the overall thickness of the organ, results in extremely long diffusion distances. As the only way to compensate for these obstacles, oxygen partial pressure (PO2) in the eyes of such fish is elevated far above atmospheric values. Although not supported by any direct evidence, the enhancement of PO2 is considered to be related to the Root effect, the release upon acidification of Hb-bound O2 into physical dissolution, possibly supported by counter-current multiplication similar to the loop of Henle.The present study evaluates the magnitude of intraocular PO2 enhancement under tightly controlled physiological conditions, to directly confirm the involvement of the Root effect on intraocular PO2 in the retina of rainbow trout Oncorhynchus mykiss. Intraocular PO2 was determined with special polarographic microelectrodes inserted into the eye. PO2profiles established in vivo by driving electrodes through the entire retina yielded average PO2 values between 10 mmHg (1.3 kPa) at the inner retinal surface and 382 mmHg (50.9 kPa) close to the outer retinal limit (Bruch's membrane). According to estimates on the basis of the diffusion distances determined from sections of the retina(∼436 μm at the site of PO2 measurement)and literature data on specific oxygen consumption, the in vivodetermined values would be sufficient to cover the oxygen demand of the retina with some safety margin.For a clear and direct in-tissue-test as to the involvement of the Root effect, an isolated in vitro eye preparation was established in order to avoid the problem of indirect blood supply to the eye from the dorsal aorta only via the pseudobranch, a hemibranch thought to modulate blood composition before entry of the eye. Any humoral effects (e.g. catecholamines)were eliminated by perfusing isolated eyes successively with standardized red blood cell (RBC) suspensions in Ringer, using trout (with Root) and human(lacking any Root effect) RBC suspension. To optimize perfusate conditions for maximal Root effect, the Root effect of trout RBCs was determined in vitro via graded acidification of individual samples equilibrated with standardized gas mixtures. During perfusion with trout RBC, PO2 at the outer retinal limit was 99 mmHg(13.2 kPa), but fell by a factor of 3.3 upon perfusion with human RBC in spite of higher total oxygen content (TO2 2.8 for trout vs 3.9 mmol l-1 for human RBC). Upon reperfusion with trout RBC, PO2 was restored immediately to the original value. This regularly observed pattern indicated a highly significant difference (P=0.003) between perfusion with trout (with Root effect;high retinal PO2) and perfusion with human (no Root effect; low retinal PO2) RBC suspension,thus clearly demonstrating that the Root effect is directly involved and a crucial prerequisite for the enhancement of PO2in the retina of the teleost eye.

Similar to gas secretion into the swimbladder, some teleosts also exhibit ocular gas secretion to supply their thick and avascular retina with oxygen. The process in the eye, however, is complicated by the fact that the retina is supplied with arterial blood only indirectly through the pseudobranch, a reduced gill arch of still unknown function. Approaches for closer analysis of the physiological implications of this serial blood supply arrangement and the associated process of O2 release include complete unilateral vascular isolation of eye and/or pseudobranch while maintaining normal blood flow characteristics for undisturbed operation of the counter-current rete mirabile.
