Magnesium chelatase (MgCh) plays a pivotal role in photosynthesis, catalyzing the insertion of magnesium into protoporphyrin IX (Proto IX), a key intermediate in chlorophyll (Chl) biosynthesis. MgCh is a heteromeric complex composed of the MgCh D subunit (CHLD), the MgCh H subunit (CHLH), and the MgCh I subunit (CHLI). The bright yellow leaves (byl) mutant was obtained through ethyl methanesulfonate (EMS) mutagenesis of the 'FT' Chinese cabbage (Brassica rapa L. ssp. pekinensis) doubled haploid line, whose Chl content, net photosynthetic rate (Pn), and non-photochemical quenching coefficient (NPQ) were decreased, and whose chloroplast development was incomplete. byl recovered to a light green phenotype under weak light conditions. Genetic analysis revealed that the bright yellow leaves phenotype of byl was caused by a single recessive nuclear gene. Using Mutmap sequencing and Kompetitive allele-specific PCR (KASP) identification, BraA01g010040.3.5C, encoding the CHLI subunit of MgCh, was identified as the candidate gene and named Brchli1. A nonsynonymous G-to-A mutation in the Brchli1 exon resulted in the substitution of aspartic acid with asparagine. Brchli1-silenced Chinese cabbage displayed bright yellow leaves with decreased Brchli1 expression. Transiently overexpressed Brchli1 in the byl mutant restored the green leaf phenotype and significantly increased relative Brchli1 expression levels. Both BrCHLI1 and its mutated variant were localized in chloroplasts. Yeast two-hybrid and luciferase complementation imaging assays demonstrated that BrCHLI1 interacted with both BrCHLD and itself. BrCHLI1 mutations did not affect its interaction with BrCHLD. Together, Brchli1 mutations impaired the function of MgCh, providing insights into the molecular mechanism of leaf coloration. Xeroderma pigmentosum is a rare autosomal recessive genodermatoses characterized by a deficiency in nucleotide excision repair. Erythropoietic protoporphyria is a rare inherited metabolic disease caused by the perturbation of heme. Xeroderma pigmentosum-erythropoietic protoporphyria is exceedingly rare. Hereby, we firstly report a young Chinese patient of xeroderma pigmentosum Group A with erythropoietic protoporphyria carrying an XPA Met214AsnfsTer7 frameshift mutation and a homozygous splicing mutation, c.315-48T>C, in the proband's intron3 of FECH. In addition, p35s‐Ami‐YGL1 experienced increased photodamage. These findings suggest that the alterations in the C:N ratio and nitrogen content in plants may be attributed to Proto IX‐mediated photodamage and chloroplast reverse transduction signaling. Besides, these results suggest that the observed increase in tissue nitrogen content in p35s‐Ami‐YGL1 does not reflect an increase in plant nitrogen absorption or use efficiency, rather it is a result of stunted carbon fixation capacity. Oral squamous-cell and pancreatic carcinomas are aggressive cancers with a poor outcome. Photodynamic therapy (PDT) consists of the use of photosensitizer-induced cell and tissue damage that is activated by exposure to visible light. PDT selectively acts on cancer cells, which have an accumulation of photosensitizer superior to that of the normal surrounding tissues. 5-aminolevulinic acid (5-ALA) induces the production of protoporphyrin IX (PpIX), an endogenous photosensitizer activated in PDT. This study aimed to test the effect of a new gel containing 5% v/v 5-ALA (ALAD-PDT) on human oral CAL-27 and pancreatic CAPAN-2 cancer cell lines. The cell lines were incubated in low concentrations of ALAD-PDT (0.05%, 0.10%, 0.20%, 0.40%, 0.75%, 1.0%) for 4 h or 8 h, and then irradiated for 7 min with 630 nm RED light. The cytotoxic effects of ALAD-PDT were measured using the MTS assay. Apoptosis, cell cycle, and ROS assays were performed using flow cytometry. PpIX accumulation was measured using a spectrofluorometer after 10 min and 24 and 48 h of treatment. The viability was extremely reduced at all concentrations, at 4 h for CAPAN-2 and at 8 h for CAL-27. ALAD-PDT induced marked apoptosis rates in both oral and pancreatic cancer cells. Elevated ROS production and appreciable levels of PpIX were detected in both cell lines. The use of ALA-PDT as a topical or intralesional therapy would permit the use of very low doses to achieve effective results and minimize side effects. Background: Candida albicans and similar species are significant pathogens in immunocompromised and hospitalized individuals, known for mucosal colonization and bloodstream/organ invasion. Many pathogenic fungi, including these species, exhibit autofluorescence (R/OF) under specific light conditions, a feature crucial for their detection. Aim: We investigated the use of a 405 nm diode laser for the direct observation of red/orange autofluorescence of Candida spp., common in the oral cavity, exploring its potential in health screenings. Methods: This study utilized cultures of Candida spp. on Sabouraud dextrose agar with Qdot 655 and 685 for fluorescence benchmarking, illuminated using a 405 nm diode laser (continuous wave, power 250 mW, 0.0425 J/cm² fluence, 0.0014 W/cm² power density). Images were captured using a yellow-filter camera at set intervals (48 to 144 h). Visual and computational analyses evaluated the R/OF in terms of presence, intensity, coloration, and intra-colony variation. Results: Most Candida strains displayed red/orange autofluorescence at all observation times, characterized by varied coloration and intra-colony distribution. Initially, there was an increase in R/OF intensity, which then stabilized in the later stages of observation. Conclusions: The majority of the Candida strains tested are capable of emitting R/OF under 405 nm laser light. According to the structural‐function hypothesis, the eggshell pigment protoporphyrin, deposited at weak spots, can strengthen the shell structure when calcium is lacking in avian species. However, this hypothesis has not been tested in species that produce pigmented eggs of uniform ground colour without spotting patterns. We tested the structural‐function hypothesis using 435 eggs produced on seven calcium diets (0.2–4.5%) given to Common Pheasants Phasianus colchicus, a species that produces a large number of eggs on a low‐calcium diet with unspotted eggshells composed of a uniform ground colour of mainly protoporphyrin. We found that pheasants on the lowest calcium diet (0.2%), which had thinner eggshells, produced eggs containing more protoporphyrin‐based coloration than four of six other diets, suggesting this species employs pigmentation as ground colour to strengthen eggshells when available calcium is low. Our results provide the first, at least partial support for a structural function for eggshell pigments producing ground colour without spotting in a species that is often calcium‐limited. This pattern may be more widespread in other ground‐nesting taxa that also produce large numbers of eggs with protoporphyrin‐based ground colour and are potentially limited by calcium during breeding. Decreasing the scavenging capacity of reactive oxygen species (ROS) and enhancing ROS production are the two principal objectives in the development of novel sonosensitizers for sonodynamic therapy (SDT). Herein, we designed a protoporphyrin-sensitized bismuth-based semiconductor (P-NBOF) as a sonosensitizer to generate ROS and synergistically depleted glutathione for enhanced SDT against tumors. The bismuth-based nanomaterial (NBOF) is a wide-bandgap semiconductor. Sensitization by protoporphyrin made it easier to excite electrons under ultrasonic stimulation, and the energy of the lowest unoccupied electron orbital in protoporphyrin was higher than the conduction-band energy of NBOF. Under ultrasound excitation, the excited electrons in the protoporphyrin were injected into the conduction band of the NBOF, increasing its reducing ability leading to the production of more superoxide anion radicals and also helping to increase the charge separation of protoporphyrin leading to the production of more singlet oxygen. Meanwhile, P-NBOF continuously depleted glutathione, which was not only conducive to breaking the redox balance of the tumor microenvironment to enhance the therapeutic efficacy of SDT, but also promoted its degradation and metabolism. The construction of this P-NBOF sonosensitizer thus provided an effective strategy to enhance SDT for tumors. To enhance the efficacy of sonodynamic tumor therapy, we developed a degradable protoporphyrin-sensitized bismuth-based nano-semiconductor (P-NBOF) by optimizing the band structure and glutathione-depletion ability. Protoporphyrin in P-NBOF under excitation preferentially generates free electrons, which are then injected into the conduction band of NBOF, improving the reducing ability of NBOF and promoting the separation of electron-hole pairs, thereby enhancing the production capacity of reactive oxygen species. Furthermore, P-NBOF can deplete glutathione, reduce the scavenging of reactive oxygen species, and reactivate and amplify the effect of sonodynamic therapy. Anemia is the most prevalent nutrition-related disorder worldwide. Zinc is an essential trace element for various biological processes in the body, and zinc deficiency has been associated with anemia in humans. However, the molecular mechanisms by which zinc availability alters red blood cell development remain uncertain. The present study identifies the essentiality of zinc during erythroid development, particularly for normal heme biosynthesis. G1E-ER4 mouse cells were used as an in vitro model of terminal erythroid differentiation, which featured elevated cellular zinc content by development. Restriction of zinc import compromised the rate of heme and α-globin production and, thus, the hemoglobinization of the erythroid progenitors. Heme is synthesized by the incorporation of iron into protoporphyrin. The lower heme production under zinc restriction was not due to changes in iron but was attributable to less porphyrin synthesis. The requirement of adequate zinc for erythroid heme metabolism was confirmed in another erythropoietic cell model, MEL-DS19. Additionally, we found that a conventional marker of iron deficiency anemia, the ZnPP-to-heme ratio, responded to zinc restriction differently from iron deficiency. Collectively, our findings define zinc as an essential nutrient integral to erythroid heme biosynthesis and, thus, a potential therapeutic target for treating anemia and other erythrocyte-related disorders. Simple Summary: In meningiomas, 5-aminolevulinc acid (5-ALA)-mediated fluorescence-guided resection (FGR) has been shown to improve intraoperative tumor bone and soft tissue invasion. However, several studies reported distinct limitations of FGR, e.g., for the visualization of the dura tail or CNS invasion. Notably, correlations between fluorescence and histological findings, as well as the expression of key heme synthesis pathway molecules, have been sparsely investigated. In this study, we examined 111 samples from 44 patients after an FGR of an intracranial meningioma for the presence of histopathological evidence of tumor tissue and intraoperative fluorescence, and analyzed the expression of key transporters/enzymes involved in PpIX metabolism using immunohistochemistry and qPCR. High sensitivity and specificity for the identification of tumor tissue and correlation of fluorescence and tumor tissue with expression of the enzymes/transporters were demonstrated. However, a deviating fluorescence and expression could be observed in non-neoplastic brain tissue, whereas this was lacking in the dura. Background: The usefulness of 5-ALA-mediated fluorescence-guided resection (FGR) in meningiomas is controversial, and information on the molecular background of fluorescence is sparse. Methods: Specimens obtained during 44 FGRs of intracranial meningiomas were analyzed for the presence of tumor tissue and fluorescence. Protein/mRNA expression of key transmembrane transporters/enzymes involved in PpIX metabolism (ABCB6, ABCG2, FECH, CPOX) were investigated using immunohistochemistry/qPCR. Results: Intraoperative fluorescence was observed in 70 of 111 specimens (63%). No correlation was found between fluorescence and the WHO grade (p = 0.403). FGR enabled the identification of neoplastic tissue (sensitivity 84%, specificity 67%, positive and negative predictive value of 86% and 63%, respectively, AUC: 0.75, p < 0.001), and was improved in subgroup analyses excluding dura specimens (86%, 88%, 96%, 63% and 0.87, respectively; p < 0.001). No correlation was found between cortical fluorescence and tumor invasion (p = 0.351). Protein expression of ABCB6, ABCG2, FECH and CPOX was found in meningioma tissue and was correlated with fluorescence (p < 0.05, each), whereas this was not confirmed for mRNA expression. Aberrant expression was observed in the CNS. Conclusion: FGR enables the intraoperative identification of meningioma tissue with limitations concerning dura invasion and due to ectopic expression in the CNS. This study aimed to evaluate the effects of a new photodynamic protocol (ALAD-PDT) on primary human osteoblasts (hOBs). The ALAD-PDT protocol consists of a heat-sensitive gel with 5% 5-delta aminolevulinic acid commercialized as Aladent (ALAD), combined with 630 nm LED. For this purpose, the hOBs, explanted from human mandible bone fragments, were used and treated with different ALAD concentrations (10%, 50%, 100% v/v) incubated for 45 min and immediately afterwards irradiated with a 630 nm LED device for 7 min. The untreated and unirradiated cells were considered control (CTRL). The cellular accumulation of the photosensitizer protoporphyrin IX (PpIX), the proliferation, the alkaline phosphatase (ALP) activity, and the calcium deposition were assessed. All concentrations (10, 50, 100%) determined a significant increment of PpIX immediately after 45 min of incubation (0 h) with the highest peak by ALAD (100%). The consequent 7 min of light irradiation caused a slight decrease in PpIX. At 48 h and 72 h, any increment of PpIX was observed. The concentration 100% associated with LED significantly increased hOB proliferation at 48 h (+ 46.83%) and 72 h (+ 127.75%). The 50% and 100% concentrations in combination to the red light also stimulated the ALP activity, + 12.910% and + 14.014% respectively. The concentration 100% with and without LED was selected for the assessment of calcium deposition. After LED irradiation, a significant increase in calcium deposition was observed and quantified (+ 72.33%). In conclusion, the ALAD-PDT enhanced proliferation, the ALP activity, and mineralized deposition of human oral osteoblasts, highlighting a promising potential for bone tissue regeneration. Protoporphyrin IX (PPIX) is a photosensitizer in the photodynamic therapy. However, its aqueous insolubility is always a challenge for extensive applications. In this work, a water-soluble derivative of PPIX were synthesized by esterification of PPIX and polyethylene glycol (PEG400), and the PPIX derivative was polymerized with hexamethylene diisocyanate to form a water-soluble polyurethane. The PPIX derivatives were characterized and the results showed that the polyurethane had the highest singlet oxygen generation efficiency among PPIX and its derivatives. The photodynamic antibacterial activity of PPIX derivatives were investigated on escherichia coli, staphylococcus aureus and methicillin-resistant staphylococcus aureus respectively. The results indicated that the polyurethane reached about 100% antibacterial rate of 40 μmol/L based on PPIX unit after 405 nm laser irradiation for 5 min. The PPIX derivatives showed high biocompatibility when co-cultured with human adipose-derived stem cells and liver cancer cells (HePG2) in the dark. With exposed to 405 nm or 635 nm laser for 5 min, about 90% and 50% HePG2 cells were killed by polyurethane, which was the best among PPIX and its derivatives. One of the main obstacles of Photodynamic Therapy (PDT) to damage and destroy abnormal cells is that most photosensitizers (Ps) have a highly hydrophobic nature with a tendency to aggregate in aqueous solutions and the non-specificity towards target cells. Nanotechnology proposes new tactics for the development of monomeric Ps nanotransporters and active targeting mole-cules with the use of biodegradable polymeric nanoparticles to improve the specificity towards target cells. The goal of this work was to optimize the synthesis of chitosan polymeric nanoparticles conjugated with protoporphyrin IX and vitamin B9 (CNPs-PpIX-B9) by the ionic gelation method from the established protocol previously carried out by our laboratory with 1.74 times fold of efficiency. They were characterized by ultraviolet-visible and infrared spectroscopy and transmission electron microscopy. The optimal conditions for CNPs synthesis was found at pH 5.11. The nanoconjugate shapes were more homogeneous and the average size resulted in 19.92 nm ± 7.52 nm. CNPs-PpIX-B9 were stable after the filter sterilization method and highly thermostable. This study aimed to evaluate the effects of a new photodynamic protocol (ALAD-PDT), consisting of 5% 5-aminolevulinic acid-gel and 630 nm-LED, already used for antibacterial effects in the treatment of periodontitis, on human gingival fibroblasts (HGF) and primary human osteoblasts (HOB). HGF and HOB were incubated with different ALAD concentrations for 45 min, and subsequently irradiated with 630 nm-LED for 7 min. Firstly, the cytotoxicity at 24 h and proliferation at 48 and 72 h were assessed. Then the intracellular content of the protoporphyrin IX (PpIX) of the ROS and the superoxide dismutase (SOD) activity were investigated at different times. Each result was compared with untreated and unirradiated cells as the control. Viable and metabolic active cells were revealed at any concentrations of ALAD-PDT, but only 100-ALAD-PDT significantly enhanced the proliferation rate. The PpIX fluorescence significantly increased after the addition of 100-ALAD, and decreased after the irradiation. Higher ROS generation was detected at 10 min in HGF, and at 30 min in HOB. The activity of the SOD enzyme augmented at 30 min in both cell types. Both BrCHLI1 and its mutated variant were localized in chloroplasts. Yeast two-hybrid and luciferase complementation imaging assays demonstrated that BrCHLI1 interacted with both BrCHLD and itself. ABSTRACT Campylobacter jejuni is a microaerophilic foodborne zoonotic pathogen of worldwide concern as the leading cause of bacterial gastroenteritis. Many strains are increasingly antibiotic resistant and new methods of control are required to reduce food-chain contamination. One possibility is photodynamic inactivation (PDI) using violet-blue (VB) light, to which C. jejuni is highly susceptible. Here, we show that flavin and protoporphyrin IX are major endogenous photosensitizers and that exposure of cells to VB light increases intracellular reactive oxygen species (ROS) to high levels, as indicated by a dichlorodihydrofluorescein reporter. Unusually for an oxygen-respiring bacterium, C. jejuni employs several ROS-sensitive iron-sulfur cluster enzymes in central metabolic pathways; we show that VB light causes rapid inactivation of both pyruvate and 2-oxoglutarate oxidoreductases, thus interrupting the citric acid cycle. Cells exposed to VB light also lose heme from c-type cytochromes, restricting electron transport, likely due to irreversible oxidation of heme-ligating cysteine residues. Evaluation of global gene expression changes by RNAseq and probabilistic modeling showed a two-stage protein damage/oxidative stress response to VB light, driven by specific regulators, including HspR, PerR, Fur, and RacR. Deletion mutant analysis showed that superoxide dismutase and the cytochrome CccA were particularly important for VB light survival and that abolishing repression of chaperones and oxidative stress resistance genes by HcrA, HspR, or PerR increased tolerance to VB light. Our results explain the high innate sensitivity of C. jejuni to VB light and provide new insights that may be helpful in exploiting PDI for novel food-chain interventions to control this pathogen. IMPORTANCE Campylobacteriosis caused by C. jejuni is one of the most widespread zoonotic enteric diseases worldwide and represents an enormous human health and economic burden, compounded by the emergence of antibiotic-resistant strains. New interventions are urgently needed to reduce food-chain contamination. Although UV light is well known to be bactericidal, it is highly mutagenic and problematic for continuous exposure in food production facilities; in contrast, narrow spectrum violet-blue (VB) light is much safer. We confirmed that C. jejuni is highly susceptible to VB light and then identified some of the global regulatory networks involved in responding to photo-oxidative damage. The identification of damaged cellular components underpins efforts to develop commercial applications of VB light-based technologies.

To address the problem of drug-resistant bacterial infections, a photodynamic broad-spectrum antibacterial cellulose acetate/protoporphyrin (CA/PpIX) porous microfibre membrane was prepared by electrospinning. The surface morphology of the fibrous membrane was observed by scanning electron microscope (SEM) and the distribution of PpIX on CA membrane was observed by laser confocal scanning microscope (CLSM). The change of the basic chemical structure of membrane before and after loading PpIX was analyzed by Fourier infrared spectroscopy (FT-IR) and Raman spectrometer. Through the DPBF substrate oxidation experiment and the antibacterial ability against S. aureus and E. coli, the photodynamic performance of the material was explored, and the gradient experiment of inorganic salt solution (KI, NaNO2, MgCl2) was designed to explore the effect of its interaction with 1O2 on the antibacterial performance. The results show that the loading of PpIX does not change the basic morphology of the fibrous membrane, but it gives the membrane good substrate oxidation and antibacterial properties (illumination 30 min, the antibacterial efficiency against S. aureus achieves 99.537%). KI, MgCl2and NaNO2solutions all have certain enhancement effects on photodynamic antibacterial mediated by CA/PpIX, among which KI solution has the most obvious enhancement effect. The interaction between the inorganic salt KI and 1O2 produces more free radicals, which can significantly potentiate the photodynamic antibacterial effect. The 100 mmol/L KI solution can potentiate the antibacterial efficiency of the composite fibrous membrane against S. aureus and E. coli to 99.9999%.

Protein arginine methyltransferase (PRMT) enzymes catalyze posttranslational modifications of target proteins and are often upregulated in human cancers. In this study, we purified two chemical compounds from seeds of Foeniculum vulgare based on their ability to inhibit the enzymatic activity of PRMT5. These two compounds were identified as Pheophorbide a (PPBa) and Pheophorbide b (PPBb), two breakdown products of chlorophyll. PPBa and PPBb inhibited the enzymatic activity of both Type I and Type II PRMTs with IC50 values at sub micromole concentrations, inhibited the arginine methylation of histones in cells, and suppressed proliferation of prostate cancer cells. Molecular docking results predicted that PPBa binds to an allosteric site in the PRMT5 structure with a high affinity (ΔG = -9.0 kcal/mol) via hydrogen bond, ionic, and π-π stacking interactions with amino acid residues in PRMT5. Another group of natural compounds referred to as protoporphyrins and sharing structural similarity with pheophorbide also inhibited the PRMT enzymatic activity. This study is the first report on the PRMT-inhibitory activity of the tetrapyrrole macrocycles and provides useful information regarding the application of these compounds as natural therapeutic reagents for cancer prevention and treatment. Carbon steel is one of the most employed materials in many industrial sectors due to its unique physical and mechanical properties. However, within a certain period of time, carbon steel-based materials are susceptible to corrosion under operating conditions and corrosion inhibitors are important to extending the limit of use of carbon steel. In this study, the influence of protoporphyrin from animal blood hemin as an eco-friendly corrosion inhibitor for T22 carbon steel in an acidic environment (0.5 M HCl) was conducted. The hemin isolated from animal blood extracts was modified to obtain the protoporphyrin. The dosage of protoporphyrin was varied between 40 and 200 ppm and the temperature influence were studied in the range of 298–318 K. The inhibition efficiency of protoporphyrin in 0.5 M hydrochloric acid reached up to 46.2% at a dose of 160 ppm at a temperature of 298 K. The inhibition efficiency (IE) value further decreases with increasing temperature, thereby showing the process exothermic in nature and the weakening of the inhibitor molecules to adsorb on the surface of the T22 carbon steel. The potentiodynamic polarization measurements indicate that protoporphyrin acts as a mixed-type inhibitor. The adsorption of protoporphyrin on the surface of T22 carbon steel obeys the Langmuir adsorption isotherm. The thermodynamic parameter of adsorption allows us to suggest the adsorption process was dominated by physical adsorption. Thus, these current results present a case study using protoporphyrin as a promising green inhibitor for carbon steel T22 in hydrochloric acid prepared from livestock waste. [ABSTRACT FROM AUTHOR]

Background: Acne is one of the most commonly faced dermatological complaints in our day-to-day practice. Among the various treatment modalities for acne and postacne pigmentation, chemical peeling is a promising and emerging treatment modality. Although various peeling agents are available, there is a lack of research comparing their efficacy. In addition, only a few studies have compared the efficacy of combined peels in the South Indian population. The present study aimed to compare the efficacy of alpha and beta hydroxy acid peels in the treatment of postacne pigmentation.Methods: Forty patients with postacne pigmentation were divided into two groups of 20 patients each. Patients were randomized into one of the two treatment groups consecutively based on block randomization technique with block size 4. Patients of GA peel group was treated with 50% glycolic acid (GA) and the salicylic acid (SA) peel group was treated with 30% salicylic acid. The procedure was repeated every two weeks up to six weeks and the primary outcome was assessed every two weeks. The assessment with photographic documentation was done by a blinded evaluator from our department of dermatology.Results: The mean age of the study population was 21±2.29 years. Out of 40 patients, 24 were females and 16 were males. GA and SA peel groups showed significant improvement from first follow-up onwards in the study. However, GA peel showed higher percentage of postacne pigmentation reduction than SA peel in all the four follow-ups. In GA peel group, >75% reduction in postacne pigmentation was observed in 45 percent of the patients. Whereas in the SA peel group, none of the patients achieved >75% reduction of postacne pigmentation. Pairwise comparison between both groups were statistically significant (P<0.0001).Conclusion: Our results suggest that the GA peel was more effective than SA peel in improving postacne pigmentation. Currently, several classes of oral therapies for psoriasis are in use, in development, or in investigative stages. Standard non-biologic treatments for psoriasis, such as methotrexate, cyclosporine, and acitretin, have generally unfavorable safety profiles and are not ideal for long-term use. This review will address the safety and efficacy of existing and novel oral therapies for psoriasis that target inflammatory pathways via modulation of phosphodiesterase 4 (PDE4), Janus kinases (JAKs), sphingosine 1-phosphate (S1P), A3 adenosine receptors, and rho-associated kinase 2 (ROCK2), with an emphasis on JAK inhibitors. Anemia is the most prevalent nutrition-related disorder worldwide. Zinc is an essential trace element for various biological processes in the body, and zinc deficiency has been associated with anemia in humans. However, the molecular mechanisms by which zinc availability alters red blood cell development remain uncertain. The present study identifies the essentiality of zinc during erythroid development, particularly for normal heme biosynthesis. G1E-ER4 mouse cells were used as an in vitro model of terminal erythroid differentiation, which featured elevated cellular zinc content by development. Restriction of zinc import compromised the rate of heme and α-globin production and, thus, the hemoglobinization of the erythroid progenitors. Heme is synthesized by the incorporation of iron into protoporphyrin. The lower heme production under zinc restriction was not due to changes in iron but was attributable to less porphyrin synthesis. The requirement of adequate zinc for erythroid heme metabolism was confirmed in another erythropoietic cell model, MEL-DS19. Additionally, we found that a conventional marker of iron deficiency anemia, the ZnPP-to-heme ratio, responded to zinc restriction differently from iron deficiency. Collectively, our findings define zinc as an essential nutrient integral to erythroid heme biosynthesis and, thus, a potential therapeutic target for treating anemia and other erythrocyte-related disorders.

Cerebral small vascular disease (CSVD) has a high incidence worldwide, but its pathological mechanisms remain poorly understood due to the lack of proper animal models. The current animal models of CSVD have several limitations such as high mortality rates and large-sized lesions, and thus it is urgent to develop new animal models of CSVD. Ultrasound can activate protoporphyrin to produce reactive oxygen species in a liquid environment. Here we delivered protoporphyrin into cerebral small vessels of rat brain through polystyrene microspheres with a diameter of 15 μm, and then performed transcranial ultrasound stimulation (TUS) on the model rats. We found that TUS did not affect the large vessels or cause large infarctions in the brain of model rats. The mortality rates were also comparable between the sham and model rats. Strikingly, TUS induced several CSVD-like phenotypes such as cerebral microinfarction, white matter injuries and impaired integrity of endothelial cells in the model rats. Additionally, these effects could be alleviated by antioxidant treatment with N-acetylcysteine (NAC). As control experiments, TUS did not lead to cerebral microinfarction in the rat brain when injected with the polystyrene microspheres not conjugated with protoporphyrin. Background: Actinic keratoses (AKs) are sun-induced cutaneous lesions that may progress to squamous cell carcinoma (SCC). Photodynamic therapy (PDT) is an ideal treatment option for AKs because it allows for treatment of field cancerization, selective destruction of diseased tissue, good cosmetic outcomes, and limited downtime.Objectives: This study sought to determine the efficacy and safety of pretreating AKs of the dorsal hands and forearms with adapalene gel, an inexpensive and over-the-counter retinoid, prior to debridement of the target area and PDT with aminolevulinic hydrochloride acid (ALA, 10%) gel and narrowband red light.Methods: Fifteen patients with AK lesions of the right or left dorsal hands or forearms were pretreated with adapalene gel (0.1%) twice daily for one week prior to ALA-PDT. The other hand or forearm was treated with ALA-PDT (standard therapy), but not pretreated. For PDT, all treated areas were debrided with sandpaper, degreased with acetone, incubated for one hour with 10% ALA gel under occlusion, and illuminated with narrowband red light (~635 nm). All patients experienced one PDT treatment session.Results: Eight weeks after treatment, 12 subjects in the adapalene-pretreated group achieved 50% to 100% clearance compared to 10 subjects in the standard therapy group. The median lesion count reduction in the adapalene-pretreated group was -79% compared to -57% in the standard therapy group, and this difference was significant (P=0.0164). The treatment was well-tolerated and the level of patient satisfaction was high.Conclusions: Pretreatment with adapalene gel twice daily for one week may enhance efficacy in a single ALA-PDT treatment of AK lesions of the dorsal hand or forearm. This study aimed to evaluate the effects of a new photodynamic protocol (ALAD-PDT), consisting of 5% 5-aminolevulinic acid-gel and 630 nm-LED, already used for antibacterial effects in the treatment of periodontitis, on human gingival fibroblasts (HGF) and primary human osteoblasts (HOB). HGF and HOB were incubated with different ALAD concentrations for 45 min, and subsequently irradiated with 630 nm-LED for 7 min. Firstly, the cytotoxicity at 24 h and proliferation at 48 and 72 h were assessed. Then the intracellular content of the protoporphyrin IX (PpIX) of the ROS and the superoxide dismutase (SOD) activity were investigated at different times. Each result was compared with untreated and unirradiated cells as the control. Viable and metabolic active cells were revealed at any concentrations of ALAD-PDT, but only 100-ALAD-PDT significantly enhanced the proliferation rate. The PpIX fluorescence significantly increased after the addition of 100-ALAD, and decreased after the irradiation. Higher ROS generation was detected at 10 min in HGF, and at 30 min in HOB. The activity of the SOD enzyme augmented at 30 min in both cell types. In conclusion, ALAD-PDT not only showed no cytotoxic effects, but had pro-proliferative effects on HGF and HOB, probably via ROS generation.

Industri tekstil banyak menggunakan pewarna sintetik karena mudah dalam pewarnaan dan murah, tetapi memiliki dampak yang buruk bagi lingkungan sekitar. Kesadaran akan pentingnya menjaga lingkungan membuat peusahaan tekstil mencoba kembali menggunakan pewarna alami. Kerabang telur puyuh memiliki protoporphyrin IX yang dapat digunakan sebagai pewarna. Kondisi optimum ekstraksi protoporphyrin IX dari kerabang telur puyuh perlu diteliti lebih lagi ditinjau dari komposisi pelarut, metode, dan siklus ekstraksi. Kerabang telur puyuh dikeringkan kemudian diekstraksi menggunakan pelarut metanol 96% dan HCl (v/v) (HCl(p) dan HCl 5%, v/v) dengan metode maserasi dan ultrasonik. Siklus ekstraksi dioptimalkan dengan variasi hingga 5 siklus. Penetapan kadar protoporphyrin dilakukan dengan menggunakan spektroskopi UV-VIS berdasarkan persamaan Lambert-Beer. Ekstrak difraksinasi menggunakan klorofom dengan rasio 1 : 2 v/v (klorofom : ekstrak). Fase organik dipindai dengan menggunakan spektrofotometer UV-Vis. Hasil penelitian menunjukkan bahwa ekstrak kerabang telur puyuh mengandung protoporphyrin IX, terlihat dari hasil spektra yang cocok dengan spektra protoporphyrin standard. Kondisi ekstraksi protoporphyrin yang optimum didapatkan dengan menggunakan pelarut metanol 96% dan HCl(p) 5% (v/v) secara maserasi. Siklus ekstraksi protoporphyrin yang optimal adalah 3 siklus dengan kadar protoporphyrin dalam ekstrak adalah 1,92 ×10-2 % b/b.

This study hypothesizes that bacteria inhabiting shale rock affect the content of the sedimentary cobalt protoporphyrin present in it and can use it as a precursor for heme synthesis. To verify this hypothesis, we conducted qualitative and quantitative comparative analyses of cobalt protoporphyrin as well as heme, and heme iron in shale rock that were (i) inhabited by bacteria in the field, (ii) treated with bacteria in the laboratory, and with (iii) bacterial culture on synthetic cobalt protoporphyrin. Additionally, we examined the above-mentioned samples for the presence of enzymes involved in the heme biosynthesis and uptake as well as hemoproteins. We found depletion of cobalt protoporphyrin and a much higher heme concentration in the shale rock inhabited by bacteria in the field as well as the shale rock treated with bacteria in the laboratory. Similarly, we observed the accumulation of protoporphyrin in bacterial cells grown on synthetic cobalt protoporphyrin. We detected numerous hemoproteins in metaproteome of bacteria inhabited shale rock in the field and in proteomes of bacteria inhabited shale rock and synthetic cobalt protoporhyrin in the laboratory, but none of them had all the enzymes involved in the heme biosynthesis. However, proteins responsible for heme uptake, ferrochelatase and sirohydrochlorin cobaltochelatase/sirohydrochlorin cobalt-lyase were detected in all studied samples.

Biosynthesis of chlorophyll involves several enzymatic reactions of which many are shared with the heme biosynthesis pathway. Magnesium chelatase is the first specific enzyme in the chlorophyll pathway. It catalyzes the formation of Mg-protoporphyrin IX from the insertion of Mg2+ into protoporphyrin IX. The enzyme consists of three subunits encoded by three genes. The three genes are named Xantha-h, Xantha-g and Xantha-f in barley (Hordeum vulgare L.). The products of the genes have a molecular weight of 38, 78 and 148 kDa, respectively, as mature proteins in the chloroplast. Most studies on magnesium chelatase enzymes have been performed using recombinant proteins of Rhodobacter capsulatus, Synechocystis sp. PCC6803 and Thermosynechococcus elongatus, which are photosynthetic bacteria. In the present study we established a recombinant expression system for barley magnesium chelatase with the long-term goal to obtain structural information of this enigmatic enzyme complex from a higher plant. The genes Xantha-h, -g and -f were cloned in plasmid pET15b, which allowed the production of the three subunits as His-tagged proteins in Escherichia coli BL21(DE3)pLysS. The purified subunits stimulated magnesium chelatase activity of barley plastid extracts and produced activity in assays with only recombinant proteins. In preparation for future structural analyses of the barley magnesium chelatase, stability tests were performed on the subunits and activity assays were screened to find an optimal buffer system and pH. In plants, tetrapyrrole biosynthesis occurs in chloroplasts, the reactions being catalysed by stromal and membrane-bound enzymes. The tetrapyrrole moiety is a backbone for chlorophylls and cofactors such as sirohaems, haems and phytochromobilins. Owing to this diversity, the potential cytotoxicity of some precursors and the associated synthesis costs, a tight control exists to adjust the demand and the fluxes for each molecule. After synthesis, haems and phytochromobilins are incorporated into proteins found in other subcellular compartments. However, there is only very limited information about the chaperones and membrane transporters involved in the trafficking of these molecules. After summarizing evidence indicating that glutathione transferases (GST) may be part of the transport and/or degradation processes of porphyrin derivatives, we provide experimental data indicating that tau glutathione transferases (GSTU) bind protoporphyrin IX and haem moieties and use structural modelling to identify possible residues responsible for their binding in the active site hydrophobic pocket. Finally, we discuss the possible roles associated with the binding, catalytic transformation (i.e. glutathione conjugation) and/or transport of tetrapyrroles by GSTUs, considering their subcellular localization and capacity to interact with ABC transporters. Differences in avian eggshell pigmentation could be an honest signal of female quality that males use to inform their nestling provisioning effort. We investigated whether among-individual variation in protoporphyrin-based eggshell pigmentation in house wrens (Troglodytes aedon) reflects female fitness-associated traits and whether males use that information. Females laying lighter clutches were older and larger than females laying darker clutches. Nestlings hatching from lighter clutches had greater size-corrected mass on post-hatch day 11, a measure that strongly predicts survival and recruitment to the breeding population. To test whether male provisioning effort responds to clutch pigmentation, we used a reciprocal clutch cross-fostering design, swapping dark with light clutches and light with dark; in controls, we swapped light with light clutches and dark with dark. Shortly before hatching, clutches were returned to their original nest to avoid confounding effects of nestling quality on male provisioning. Contrary to the sexual selection hypothesis, clutch pigmentation had no effect on male provisioning. Males were probably able to observe eggshell pigmentation and thus had information about female quality, but they did not use this information to modulate their nestling provisioning. Purpose: Heterogeneity within GBMs and variability of visualized fluorescence combine to confer practical limitations to the technique of optical imaging. A biometric analysis was planned to objectively ascertain and analyse this phenomenon Methods: 25 adult glioblastoma subjects undergoing resection were prospectively accrued. Biopsies were taken from various parts of the tumor and safe peritumoral zones. White light (WL) and visualized fluorescence was subjectively recorded. Corresponding histopathology [coalescent (C) or infiltrating (I) tumor] and protoporphyrin-IX (PPIX) levels were assayed. Results: WL was very sensitive for detecting tumor. SF was more specific and had high positive predictive value for detecting tumor. WF on the other hand had a poor discriminatory efficacy. Mean PPIX levels were 3.0, 2.01 and 0.16 for SF, WF, and NF respectively. WF had a wide variable range of PPIX levels. Within the coalescent tumor areas, there was a variable distribution of fluorescence (both subjective as well as objective PPIX levels) with only 54% samples showing SF and high PPIX. In seven cases this discordance was noted within the same tumor (biological heterogeneity). Conclusions: Fluorescence may miss important tumor areas even if objective assessment is used. Histologically similar tumor areas may exhibit contrasting fluorescence properties, a phenomenon which needs further investigation and elucidation of underlying mechanisms which could potentially be manipulated to optimize the utility of fluorescence guidance. Background and aim: This study was carried out to investigate antiplasmodial activities of Diospyros mespiliformis (DM) and Mondia whitei (MW) in Plasmodium berghei-infected mice. Materials and methods: Air-dried stem of DM and root of MW were soaked in methanol, decanted and concentrated to give extracts. Parts of these extracts were partitioned successively to give dichloromethane, ethylacetate and methanol fractions. Mice (18 ± 3 g) were infected with Plasmodium berghei-infected erythrocytes from a donor mouse and were treated with the extracts, while the drug control group received 10 mg/kg body weight of artesunate and the parasitized control received the vehicle (5% v/v DMSO). Percentage parasitemia and clearance were estimated from thin films of blood smear. Hematological parameters were determined using standard methods. Both extracts were also tested on in vitro inhibition of b-hematin formation. Results, discussion and conclusion: Results showed that DM had the least percentage parasitemia (0.67%) and highest percentage parasite clearance (84.7%) while the MW had 0.89% percentage parasitemia and 79.7% clearance at the highest dose used after the seventh day relative to untreated control. The cell free antiplasmodial activity of the fractions and extracts of both DM and MW revealed that DM significantly inhibited b-hematin formation than MW. The packed cell volume, white blood cell count, Lymphocyte, Eosinophil, Monocyte and Neutrophil significantly increased in the treated groups compared with the control. In this research we are reporting for the first time the elaboration y characterization of titanium dioxide sensitized solar cells based on nanocomposites of protoporphyrin IX and microwave reduced graphene oxide quantum dots. Reduced graphene oxide quantum dots were synthesized by microwave assisted cutting of reduced graphene oxide and mixed with protoporphyrin IX previous to be drop-casted on titanium dioxide. Characterization was done by dynamic light scattering, ultraviolet visible spectroscopy, Raman spectroscopy, atomic force microscopy, field electron scanning electron microscopy, impedance spectroscopy and current voltage measurements. There were blue and red shifts of Raman and absorption bands of protoporphyrin+reduced graphene oxide quantum dots nanocomposites, evidencing strong interactions because of π − π electrons stacking, this fact was confirmed by microscopy images showing full coverage of protoporphyrin surface by reduced graphene oxide quantum dots. Electrical characterization of titanium dioxide+protoporphyrin+reduced graphene oxide quantum dots showed one of the highest photo-responses and lowest resistivities, in line with impedance measurements in which these composites presented up to ∼ 200% increase in capacitance, versus TiO 2 alone. Efficiency of the optimized cell was 6.13 %, this was a value 9.4 % higher versus titanium dioxide+protoporphyrin cell. The reasons for the behavior of optimized cells are discussed based on a bands diagram showing the possibility of a rectifier effect and because of the strong affinity between protoporphyrin and reduced graphene oxide quantum dots. 5-Aminolevulinic acid (5-ALA) is a naturally occurring non-proteinogenic amino acid, which contributes to the diagnosis and therapeutic approaches of various cancers, including glioblastoma (GBM). In the present study, we aimed to investigate whether 5-ALA exerted cytotoxic effects on GBM cells. We assessed cell viability, apoptosis rate, mRNA expressions of various apoptosis-related genes, generation of reactive oxygen species (ROS), and migration ability of the human U-87 malignant GBM cell line (U87MG) treated with 5-ALA at different doses. The half-maximal inhibitory concentration of 5-ALA on U87MG cells was 500 μg/mL after 7 days; 5-ALA was not toxic for human optic cells and NIH-3T3 cells at this concentration. The application of 5-ALA led to a significant increase in apoptotic cells, enhancement of Bax and p53 expressions, reduction in Bcl-2 expression, and an increase in ROS generation. Furthermore, the application of 5-ALA increased the accumulation of U87MG cells in the SUB-G1 population, decreased the expression of cyclin D1, and reduced the migration ability of U87MG cells. Our data indicate the potential cytotoxic effects of 5-ALA on U87MG cells. Further studies are required to determine the spectrum of the antitumor activity of 5-ALA on GBM.

An assembly of hybrid hydrogel derived from Carbon Dot (CD), Protoporphyrin IX (PpIX) and DNA is reported here. PpIX and CD were covalently conjugated to 5′-phosphate termini of Cytosine (C) rich single strand DNA (ssDNA) separately. The CD-DNA-PpIX hybrid hydrogel was assembled through the formation of intercalated motif (i-motif) DNA network structure from CD-DNA and PpIX-DNA conjugates where CD act as crosslinkers in the hydrogel as well as energy donor to excite the photosensitizer (PS), PpIX. While hydrogel derived from only PpIX-DNA conjugate showed an inadequate sol-gel transition, the same could be precisely achieved from the CD-DNA-PpIX hybrid hydrogel by controlled pH adjustment. Distinct photophysical properties of CD and PpIX including fluorescence emission potentially enable tracking of the PS loading and hydrogel dissolution that was visually detectable under UV illumination. Reactive Oxygen Species (ROS) generation and subsequent killing of gram-positive bacteria (Staphylococcus aureus (S. aureus)) were observed following excitation of PpIX (acceptor) in the hybrid hydrogel either through energy transfer from CD or by direct irradiation of PpIX with visible light. Complete dissolution of hydrogel and sustained release of PpIX and subsequent ROS generation was achieved over 10–11 days that could kill S. aureus systematically. This study provides a promising strategy to address self-quenching and solubility of PS for its sustained release in Antimicrobial Photodynamic Therapy (A-PDT) applications through smart hydrogel formulation. Birds' eggs often exhibit considerable variation in the presence and pattern of superficial pigmentation, or maculation. However, despite long appreciation of the nature of this variation both between and within species, there is still poor understanding of why such a diversity of patterns has evolved. Accurately quantifying eggshell maculation is therefore key to elucidating its functional significance. Maculation is typically quantified from photographs, and so measurements may be biased by the perceived distortion introduced by the curvature of the eggshell. Here, I consider egg geometry in order to derive measures of curved distance and surface area that can be used to characterize eggshell maculation patterns more accurately, and demonstrate the utility of this approach for quantifying and visualizing eggshell maculation from standard photographs. Highlights • Polyaniline/protoporphyrin composites were prepared in acidic suspension of PpIX. • The PANI/PpIX composites exhibited nanosheet structure with nanoparticles. • The composites exhibited outstanding sensing performance toward VOCs. Abstract Polyaniline/protoporphyrin nanocomposites were prepared via a simple chemical method in the acidic suspension of protoporphyrin. The scanning electron microscope images revealed that the polyaniline/protoporphyrin composites exhibited an interesting nanosheet structure decorated with nanoparticles, which is rather different with the usual nanofiber morphology of polyaniline. The formation of the nanosheet structure is because protoporphyrin molecules may exist as a bilayer form at low pH, which is similar with the phospholipid bilayer in membranes of cells. To demonstrate the application potential of the composites, the sensing performance of the composites was tested when exposed to four volatile organic compounds, including trimethylamine, triethylamine, ethanol, and ethyl acetate. The composites exhibited highest response value (S) of 39.482 toward trimethylamine, and fast response time of 2–4 s toward trimethylamine and triethylamine. Introduction: Fluorescence guided surgery has developed over the last 2 decades as a formidable augmentation strategy to promote maximal safe resection and diagnostic accuracy within gliomas. The majority of the literature evidence supporting this modality utilizes 5-aminolevulinic acid in the setting of high-grade gliomas. The role for fluorescence guided surgery in low-grade gliomas is less well defined.Results: This review describes the existing literature discussing the utilization of 5-aminolevulinic acid for fluorescence guided surgery in low-grade gliomas, including its pertinence in identification of anaplastic foci and potential role in guiding resection following combination with augmentation strategies for detection.Conclusion: The advance in operative technology and growth of research analyzing 5-aminolevulinic acid will continue to enhance the role of fluorescence guided surgery within the standard of surgical management for low-grade gliomas. Porphyrias are disorders caused by defects in the biosynthetic pathway of heme. Their manifestations can be divided into three distinct syndromes, each attributable to the accumulation of three distinct classes of molecules. The acute neurovisceral syndrome is caused by the accumulation of the neurotoxic porphyrin precursors, delta aminolevulinic acid, and porphobilinogen; the syndrome of immediate painful photosensitivity is caused by the lipid-soluble protoporphyrin IX and, the syndrome of delayed blistering photosensitivity, caused by the water-soluble porphyrins, uroporphyrin, and coproporphyrin. Porphyrias can manifest with one, or with a combination, of these syndromes, depending on whether one or more types of molecules are being accumulated. Iron plays a significant role in some of these conditions, as evidenced by improvements in both clinical manifestations and laboratory parameters, following iron depletion in porphyria cutanea tarda, or iron administration in some cases of X-linked erythropoietic protoporphyria. While the pathophysiology of a specific type of porphyrias, the protoporphyrias, appears to favor the administration of zinc, results so far have been conflicting, necessitating further studies in order to assess its potential benefit. The pathways involved in each disease, as well as insights into their pathobiological processes are presented, with an emphasis on the development of photosensitivity reactions. [ABSTRACT FROM AUTHOR]
