Using desi-cow waste products like Jeevamrit under natural farming is widespread among farmers for improving soil biology and productivity. Jeevamrit enhances soil chemical and microbiological properties without needing a large quantity of farmyard manure (FYM) as a sustainable farming practice with a reduced carbon footprint. Despite its traditional use, Jeevamrit faces criticism due to a lack of scientific evidence. This study investigated the comparative effect of Jeevamrit and chemical fertilizers on the growth and yield of marigold cv. Siracole. The experiment employed a randomized block design (RBD) with three replications. The mother block of marigolds was raised for both the summer and winter seasons. From this mother block, three harvesting flushes were taken and propagated from cuttings. The rooted cuttings were planted at monthly intervals and evaluated for flowering parameters and compared to those treated with RDF (30:20:20 N, P, and K g/m2). Soil supplied with Jeevamrit showed enhanced bacteria (26.33%), fungi (18.92%), and actinomycetes (31.21%) populations compared to the recommended dose of fertilizers (RDF) (i.e., N–P–K @ 30:20:20 g m−2). Jeevamrit-treated plants have a more marketable flower yield per square meter (3.98%) and a longer shelf life (9.93%) compared to RDF. The study concludes that Jeevamrit @ 2 liters m−2 is a sustainable and effective alternative to traditional fertilizers for enhancing marigold production in the mid-hills of Himachal Pradesh, where natural farming is already accepted.

Abstract Objectives High‐altitude natives have a high incidence of parangangliomas (PGL) of the head and neck, especially the carotid body tumor. The aim of this study is to describe the clinical presentation, pattern, altitude of residence, distribution, management, and follow‐up of head and neck paragangliomas (HNPGL) in our sub‐Himalayan population. Study Design Retrospective cohort study. Setting Academic tertiary care hospital. Methods Hospital records of 20 patients of HNPGL diagnosed from December 2017 to December 2021 were retrieved for analysis. Results Twenty patients with 23 HNPGL, with a mean age of 41.74 years were managed in our institute. The female‐to‐male ratio was 2.3: 1 and the mean follow‐up was 29.95 months. Nine had carotid body (CBPGL), 7 had tympanic (TPGL), 2 had jugular (JPGL), and 2 had vagal paragangliomas (VPGL). Multiple PGL were seen in 4 patients (20%). Majority of cases (all CBPGL and 57.14% of TPGL) were residents of the high altitude, and the rest were from the low altitude. Fifteen patients (8 CBPGL, 7 TPGL) were operated. There were no major complications except in a patient with large carotid body tumor required anastomosis of carotid artery. Five patients received stereotactic radiotherapy, and 1 malignant PGL received chemoradiotherapy. Conclusion In this study, JPGL and VPGL are common at low altitudes, whereas carotid body and tympanic PGL were the most common tumor at high altitudes. Being a retrospective and study small sample size, a definite conclusion is not established, however, a genetic analysis and inclusion of a wider population in a future prospective study may establish the hypothesis.

Gastrointestinal stromal tumors (GIST) are rare tumors accounting for less than 1% of primary neoplasia of the digestive tract. They can also occur outside the GIT, where they are called extra gastrointestinal stromal tumors (EGIST), which are extremely rare tumors. GISTS and EGISTS have similar histo-pathological and molecular profiles. Though these are known to be FDG avid, some of the atypical GISTS can be FDG negative or only minimally FDG avid.Here, an unusual case of a 57-year-old female with FDG avid solitary chest wall mass is presented, which after biopsy and immune-histochemistry was proven as EGIST. She underwent en block resection of the tumor and is presently on adjuvant treatment with Imatinib mesylate. This case report highlights the importance of considering EGIST as a differential diagnosis of a solitary chest wall mass and the utility of 18F- FDG PET/CT in its management.

Abstract Current study signifies the use of nanoparticles as alternative in plant disease management to avoid harmful effect of pesticide and fungicide residue. Synthesis of nanoparticles (Ni0.5Al0.5Fe2O4) by hydrothermal method and studied their X-ray diffraction analysis (XRD), Raman spectra, and UV spectra and further successfully evaluated for antifungal activity against a soil and seed borne pathogenic fungus (Fusarium oxysporum).Among various pests, fungal pathogens are the main cause of crop destruction and we developed nanoparticles (Ni0.5Al0.5Fe2O4) which is successfully evaluated for antimycotic activity against dry rot (F. oxysporum) of ginger which causes 50–70% losses in the ginger plant. In vitro and in vivo analysis designated that the nanoparticles (Ni0.5Al0.5Fe2O4) has shown an excellent antifungal activity against F. oxysporum at 0.5 mg/ml concentration. Similarly, no disease incidence was recorded when Ni0.5Al0.5Fe2O4 nanoparticles used at 0.5 mg/ml concentration under in vivo conditions. In plants various environmental stresses (biotic and abiotic) leads to excessive production of reactive oxygen species (ROS) causing progressive oxidative damage and ultimately leads to cell death. The role of ROS in nanoparticles (Ni0.5Al0.5Fe2O4) represents by reduction in the growth inhibition of F. oxysporum. We speculated in light of these results that the cytotoxic effect of Ni0.5Al0.5Fe2O4 nanoparticles on F. oxysporum may be mediated through ROS. We can suggest the role of nanoparticles (Ni0.5Al0.5Fe2O4) gives a promising result as a fungicidal activity and could be a novel family of future new generation fungicide.

The continuum of antioxidant response dysregulation in aging/oxidative stress-driven Nlrp3 inflammasome activation-mediated inflammatory response is associated with age-related diseases. Peroxiredoxin (Prdx) 6 is a key antioxidant that provides cytoprotection by regulating redox homeostasis. Herein, using lens epithelial cells (LECs) derived from the targeted inactivation of Prdx6 gene and aging lenses, we present molecular evidence that Prdx6-deficiency causes oxidative-driven Nlrp3 inflammasome activation, resulting in pyroptosis in aging/redox active cells wherein Prdx6 availability offsets the inflammatory process. We observed that Prdx6−/− and aging LECs harboring accumulated reactive oxygen species (ROS) showed augmented activation of Nlrp3 and bioactive inflammatory components, like Caspase-1, IL-1β, ASC and Gasdermin-D. Similar to lipopolysaccharide treatment, oxidative exposure led to further ROS amplification with increased activation of the Nlrp3 inflammasome pathway. Mechanistically, we found that oxidative stress enhanced Kruppel-like factor 9 (Klf9) expression in aging/Prdx6−/− mLECs, leading to a Klf9-dependent increase in Nlrp3 transcription, while the elimination of ROS by the delivery of Prdx6 or by silencing Klf9 prevented the inflammatory response. Altogether, our data identify the biological significance of Prdx6 as an intrinsic checkpoint for regulating the cellular health of aging or redox active LECs and provide opportunities to develop antioxidant-based therapeutic(s) to prevent oxidative/aging-related diseases linked to aberrant Nlrp3 inflammasome activation.

This paper describes an unreported case of a rifaximin-induced phototoxic reaction in an otherwise healthy 24-year-old female (skin type: V). The patient developed malaise, chills, and facial swelling with accompanying redness and itching that began within a day of initiating treatment with rifaximin (200 mg twice daily), and progressively increased over the next 3–4 days. The patient revealed that they had been lying in the sun for hours due to the chills they were experiencing. Over the next 10 days they developed an exaggerated, acute, sunburn-like phototoxic reaction, with blistering over the exposed skin. A skin biopsy showed no evidence of vasculopathy, endothelial damage, or extravasation of red blood cells. The patient was treated successfully with oral prednisolone (30 mg per day for a week), topical mometasone furoate (0.1%) cream applied twice daily, levocetirizine (5 mg per day taken orally), zinc oxide (20.0%) cream applied every 3 hours during daytime, and strict sun avoidance. The possible pathomechanism of rifaximin-induced sunburn is also discussed here.

Abstract Rhamnolipids (RLs) are surface-active compounds and belong to the class of glycolipid biosurfactants, mainly produced from Pseudomonas aeruginosa. Due to their non-toxicity, high biodegradability, low surface tension and minimum inhibitory concentration values, they have gained attention in various sectors like food, healthcare, pharmaceutical and petrochemicals. The ecofriendly biological properties of rhamnolipids make them potent materials to be used in therapeutic applications. RLs are also known to induce apoptosis and thus, able to inhibit proliferation of cancer cells. RLs can also act as immunomodulators to regulate the humoral and cellular immune systems. Regarding their antimicrobial property, they lower the surface hydrophobicity, destruct the cytoplasmic membrane and lower the critical micelle concentration to kill the bacterial cells either alone or in combination with nisin possibly due to their role in modulating outer membrane protein. RLs are also involved in the synthesis of nanoparticles for in vivo drug delivery. In relation to economic benefits, the post-harvest decay of food can be decreased by RLs because they prevent the mycelium growth, spore germination of fungi and inhibit the emergence of biofilm formation on food. The present review focuses on the potential uses of RLs in cosmetic, pharmaceutical, food and health-care industries as the potent therapeutic agents.

Pseudocereals have attracted the attention of nutritionists and food technologists due to their high nutritional value. In addition to their richness in nutritional and bioactive components, these are deficient in gluten and can serve as valuable food for persons suffering from gluten allergies. Processing treatments are considered an effective way to enhance the quality of food grains. Soaking and germination are traditional and most effective treatments for enhancing the nutritional and bioactive potential as well as reducing the anti-nutritional components in food grains. This study reflects the effect of soaking and germination treatments on nutritional, bioactive, and anti-nutritional characteristics of pseudocereals. There was a significant (p ≤ 0.05) increase in nutritional and bioactive components such as crude fiber, crude protein, phenolic components, antioxidant activity, and mineral content but reduced the anti-nutrients such as tannin and phytic acid. In amaranth, there was a significant increase (p ≤ 0.05) of 7.01, 74.67, 126.62, and 87.47% in crude protein, crude fiber, phenolic content, and antioxidant activity but significant (p ≤ 0.05) reduction of 32.30% and 29.57% in tannin and phytic acid contents, respectively. Similar changes in values of crude proteins, crude fiber, phenolic content, and antioxidant activity were observed in buckwheat and quinoa. While the anti-nutritional components such as tannin and phytic acid decreased by 59.91 and 17.42%, in buckwheat and 27.08% and 47.57%, in quinoa, respectively. Therefore, soaking and germination proved to be excellent techniques to minimize the anti-nutritional component and enhance the nutritional, bioactive, and antioxidant potential of these underutilized grains.

Multiple sclerosis (MS) is an autoimmune, demyelinating, inflammatory disease of central nervous system (CNS) which is characterized by spasticity, fatigue, depression, anxiety, bowel and bladder dysfunction, impaired mobility, cognitive impairment etc. and affects approximately 2.5 million people worldwide. Disease modifying therapies for MS which help in preventing accumulation of lesions in white matter of CNS are costly and have significant adverse effects. Therefore, patients with MS are using complementary and alternative medicine (CAM) and Yoga is one of the most popular form of CAM which is being used immensely to reduce or overcome the symptoms of MS. In the current review attempted to present the potential impact of yoga practices on reducing MS related symptoms.

Introduction: Esophageal varices develop as a consequence of portal hypertension (PHT) in patients with chronic liver disease. Hence, screening of all cirrhotic patients with upper gastrointestinal endoscopy to detect the presence of significant esophageal varices implies a number of unnecessary endoscopies and has its limitation where such facilities are not available, especially in the rural part of country. Method: Patients with either sex, aged between 18 and 60 years with diagnosis of cirrhosis were studied. Detailed history, physical examination along with relevant investigations were recorded and upper gastrointestinal endoscopy was done within 2–3 days of investigation. Esophageal varices were graded as I-IV, using the Paquet grading system and patients were classified dichotomously either as having large esophageal varices (LEV) group A (Grade III-IV) and no varices group B (grade I-II). Result: A total of 50 patients with cirrhosis of liver were recruited in the study. Peroxiredoxins (Prxs) are antioxidant proteins that are involved in cellular defence against reactive oxygen species and reactive nitrogen species. Humans have six peroxiredoxins, hPrxI-VI, out of which hPrxI and hPrxII belongs to the typical 2-Cys class sharing 90% conservation in their amino acid sequence including catalytic residues required to carry out their peroxidase and chaperone activities. Despite the high conservation between hPrxI and hPrxII, hPrxI behaves differently from hPrxII in its peroxidase and chaperone activity. We recently showed in yeast that in the absence of Tsa1 and Tsa2 (orthologs of hPrx) hPrxI protects the cells against different stressors whereas hPrxII does not. To understand this difference, we expressed catalytic mutants of hPrxI in yeast cells lacking the orthologs of hPrxI/II. We found that the catalytic mutants lacking peroxidase function including hPrxIC52S, hPrxIC173S, hPrxIT49A, hPrxIP45A and hPrxIR128A were not able to grow on media with nitrosative stressor (sodium nitroprusside) and unable to withstand heat stress, but surprisingly they were able to grow on an oxidative stressor (H2O2). Interestingly, we found that hPrxI increases the expression of antioxidant genes, GPX1 and SOD1, and this is also seen in the case of a catalytic mutant, indicating hPrxI can indirectly reduce oxidative stress independently of its own peroxidase function and thus suggesting a novel role of hPrxI in altering the expression of other antioxidant genes. Furthermore, hPrxIC83T was resistant to hyperoxidation and formation of stable high molecular weight oligomers, which is suggestive of impaired chaperone activity. Our results suggest that the catalytic residues of hPrxI are essential to counter the nitrosative stress whereas Cys83 in hPrxI plays a critical role in hyperoxidation of hPrxI.

The participatory rural appraisals were exercised during 2014 in temperate Himalayas in eight gram panchayats of Sangrah block of district Sirmour of Himachal Pradesh. The purpose was to unravel the problems being faced by the farmers, prioritise them and devise apriori based solutions for the upliftment of the peasants. The project area had 22 villages spread over 5611.09 ha. Out of which, 701.02 ha was the cultivable area in the possession of 12906 cultivators. The area was rugged, had steep topography and was stratified into ladder type fields. The matrix analysis extrapolated the prime problem of monkey and wild animals menace. Other problems ranking in the descending order were: erratic irrigation and drinking water supply, lack of awareness about new techniques, ginger rot, poor connectivity by road and communication, migration of youth from villages to cities, unavailability of agricultural inputs like seeds, fertilizers etc., soil erosion, hindrance due to local politics, erratic electricity supply, abandoned agriculture due to public distribution system, lacking minimum support price and crop insurance of each crop, unconsolidated land, lacking higher education facility, labour scarcity due to MGNREGA and migration, defunct apple cultivars, frequent blockage and breakage of irrigation kuhals (channels) and invasion of obnoxious weeds. The insight solutions and the unleashed problems lies in farming bi-pronged strategy which must concentrate on i) convergence of schemes, and ii) apriori based approaches for the upliftment of rural community of the temperate Himalayas. The outcome of the present study suggested seven proposals for improving income resources of hilly peasants.

The present study was undertaken to evaluate anxiolytic effect of 70% hydroethanolic extract of leaves of Cissampelos pareira in murine models. C. pareira (Menispermaceae) is rich in alkaloids, and phytochemical results showed that it contains alkaloids, flavanoids, terpenoids, steroids, etc., Anxiolytic activity was evaluated by using elevated plus maze test (EPM), light dark (LandD) model, and forced swim test (FS) models in rats. The efficacy of extract (100, 200, 400 mg/kg) was compared with control as well as standard diazepam (DZ; 2 mg/kg, p.o.) in EPM, LandD model, and imipramine (IM; 2.5 mg/kg, p.o.) in FS model. The results showed that DZ and extract significantly increased the number of entries, time spent in open arm, head dip counts, and rearing time, while they decreased fecal count in EPM. DZ and extract also significantly increased the number of crossings and time spent in light compartment, while they decreased duration of immobility in LandD model. In case of FS model, IM and extract significantly increased mobility and swimming time. Thus, the results confirm that hydroethanolic extract of C. pareira has the potential to be used in the management of anxiety-like behavior in a dose of 200 and 400 mg/kg. Further study is required to explore the plant and its parts for anxiolytic potential.

Studies on the effect of covering material on off-season cut flower production of chrysanthemum (Dendranthema grandiflora Tzvelev) were carried out at the experimental farm of the Department of Floriculture and Landscape Architecture, Dr Yashwant Singh Parmar University of Horticulture and Forestry, Nauni, Solan, Himachal Pradesh, during 2011 in naturally ventilated polyhouse. The experiment was carried out using three types of covering materials(viz. tarpaulin, high density polyethylene (HDPE) and black sateen cloth) and four cultivars each of standard (Purnima, Yellow Star, Tata Century and White Star) and spray (Ajay, Birbal, Nanako and White Bouquet) varieties. Findings revealed that HDPE was the best alternative covering material to tarpaulin as optimum plant height (83.33 cm), plant spread (40.54 cm), maximum number of cut stems (4.21), duration of flowering (33.54 days) and earliest flower bud formation (91.07 days) and flowering were obtained in plants under HDPE. Flowering was earlier under controlled photoperiod than natural photoperiod. Peak flowering was earliest (140.36 days) in plants under HDPE* cover as compared to plants under natural photoperiodic conditions which took maximum number of days (176.19), to come to peak flowering. The varieties were assigned to different response group under controlled photoperiodic conditions with HDPE. All the varieties tested were found suitable for off-season flower production.

A study was conducted to see the effect of three growing media (soil from natural habitat, soil from natural habitat and FYM (2:1), v/v and soil, sand and FYM (2:1:1), three pinching treatments (No pinch, single pinch and double pinch) and paclobutrazol (0, 75, 150 ppm) on growth and flowering of seedlings as well as vegetatively propagated (rooted stem cuttings) of Phillipines violet or blue bell barleria (Barleria cristata L.). Plants raised through seeds and rooted cuttings differed significantly among themselves for various growth and flowering parameters. Plants raised under both methods of propagation showed optimum plant height (1.5 to 2.5 times to the height of the pot) when grown in soil from natural habitat and FYM with single and double pinching in absence of application of paclobutrazol for growing as pot plant. Seedlings recorded maximum number of secondary side shoots (20.58), leaves (177.42), flower clusters (16.17), flowers (23.58)/plant when grown in soil from natural habitat and FYM with double pinching and with the application of 75 ppm paclobutrazol and seed raised plants showed early visible bud formation and flowering when grown in all growing media without pinching and application of 75 ppm paclobutrazol. However, the cutting raised plants recorded maximum number, of secondary side shoots (16.00), leaves (171.67), flower clusters (17.83), flowers (24.42)/plant when grown in soil from natural habitat and FYM with double pinching and with the application of 75 ppm paclobutrazol. Seed raised plants showed better results in case of plant height, length of primary side shoots and plant spread than cutting raised plants while cutting raised plants recorded early visible bud formation and flowering and maximum number of flower clusters/plant than seed raised plants. Based on the findings of present investigations, seed raised Barleria cristata. plants grown in pots containing soil from natural habitat added with FYM (2:1, v/v), double pinching and application of 75 ppm paclobutrazol is recommended for better growth and flowering.

Radiation Therapy (RT) plays a pivotal role in the curative approach for carcinoma of the cervix. Inspite of the emergence of various new conformal techniques in RT, conventional techniques still hold vital importance. Majority of the patients worldwide are treated with 2D-RT techniques. 2D-RT techniques have been proven to be non-inferior and simpler in comparison to 3D-RT in the context of carcinoma of the cervix. However, inadequate target volume coverage with improper portal design can preclude the chances of cure. We demonstrate the need for abolishing guesswork in terms of target volume determination through the example of a patient′s sagittal magnetic resonance image showing a case of the retroverted uterus which would have been likely to be missed from the treatment portals if they were designed using definitions based on bony landmarks.

Viral hepatitis resulting in chronic liver disease is an important clinical challenge and insight into the cellular processes that drive pathogenesis will be critical in order to develop new diagnostic and therapeutic options. Nuclear inclusions in viral and non-viral hepatitis are well documented and have diagnostic significance in some disease contexts. However, the origins and functional consequences of these nuclear inclusions remain elusive. To date the clinical observation of nuclear inclusions in viral and non-viral hepatitis has not been explored at depth in murine models of liver disease. Herein, we report that in a transgenic model of hepatitis B surface antigen mediated hepatitis, murine hepatocytes exhibit nuclear inclusions. Cells bearing nuclear inclusions were more likely to express markers of cell proliferation. We also established a correlation between these inclusions and oxidative stress. N-acetyl cysteine treatment effectively reduced oxidative stress levels, relieved endoplasmic reticulum (ER) stress, and the number of nuclear inclusions we observed in the transgenic mice. Our results suggest that the presence of nuclear inclusions in hepatocytes correlates with oxidative stress and cellular proliferation in a model of antigen mediated hepatitis.
