1. Assessment of Google Glass for Photographic Documentation in Veterinary Forensic Pathology: Usability Study

Abstract

BackgroundGoogle Glass is a head-mounted device designed in the shape of a pair of eyeglasses equipped with a 5.0-megapixel integrated camera and capable of taking pictures with simple voice commands. ObjectiveThe objective of our study was to determine whether Google Glass is fit for veterinary forensic pathology purposes. MethodsA total of 44 forensic necropsies of 2 different species (22 dogs and 22 cats) were performed by 2 pathologists; each pathologist conducted 11 necropsies of each species and, for each photographic acquisition, the images were taken with a Google Glass device and a Nikon D3200 digital single-lens reflex (DSLR) camera. The pictures were collected, divided into 3 groups (based on the external appearance of the animal, organs, and anatomical details), and evaluated by 5 forensic pathologists using a 5-point score system. The parameters assessed were overall color settings, region of interest, sharpness, and brightness. To evaluate the difference in mean duration between necropsies conduced with Google Glass and DSLR camera and to assess the battery consumption of the devices, an additional number of 16 necropsies were performed by the 2 pathologists. In these cases, Google Glass was used for photographic reports in 8 cases (4 dogs and 4 cats) and a Nikon D3200 reflex camera in the other 8 cases. Statistical evaluations were performed to assess the differences in ratings between the quality of the images taken with both devices. Abstract

An 8-month-old female Lagotto Romagnolo dog was presented for a 1-month history of an initial severe reluctance to move, rapidly progressing to a marked stiff gait and progressive muscular weakness and evolving to tetraparesis, which persuaded the owner to request euthanasia. A primary muscle pathology was supported by necropsy and histopathological findings. Macroscopically, the muscles were moderately atrophic, except for the diaphragm and the neck muscles, which were markedly thickened. Histologically, all the skeletal muscles examined showed atrophy, hypertrophy, necrosis with calcification of the fibers, and mild fibrosis and inflammation. On immunohistochemistry, all three dystrophin domains and sarcoglycan proteins were absent. On Western blot analysis, no band was present for delta sarcoglycan. We sequenced the genome of the affected dog and compared the data to more than 900 control genomes of different dog breeds. Genetic analysis revealed a homozygous private protein-changing variant in the SGCD gene encoding delta- sarcoglycan in the affected dog. The variant was predicted to induce a SGCD:p.(Leu242Pro) change in the protein. In silico tools predicted the change to be deleterious. Other 770 Lagotto Romagnolo dogs were genotyped for the variant and all found to be homozygous wild type. Based on current knowledge of gene function in other mammalian species, including humans, hamsters, and dogs, we propose the SGCD missense variant as the causative variant of the observed form of muscular dystrophy in the index case. The absence of the variant allele in the Lagotto Romagnolo breeding population indicates a rare allele that has appeared recently.

Abstract

Idiopathic inflammatory myopathies (IIMs) represent a heterogeneous group of disorders in which skeletal muscle is inappropriately targeted by the immune system. IIMs are characterized by inflammation of muscle and varying degrees of muscle dysfunction. Extra-muscular manifestations may involve heart, skin, joints, lungs, and gastrointestinal tract. Cardiovascular involvement is a feared event because is one of the leading causes of mortality in IIM patients. As the myocardium shares many features with the skeletal muscle, it is supposed that it can be affected by the same inflammatory processes, which take place during the different forms of IIMs. However, the full extent of this link and the mechanisms behind it are still not fully understood. Animal models have greatly improved our understanding of IIM pathomechanisms and have proven to be a useful tool for discovering therapeutic drug targets. Here we report the evidence of heart muscle involvement in different animal models of spontaneous IIMs, assuming a common autoimmune mechanism and presenting them as study models for human pathology.

17. New insights into the Pathogenesis of Inflammatory Myopathies in Animals

Abstract

Inflammatory Myopathies (IMs) are a large and heterogeneous group of acquired disorders of the skeletal muscle characterized by the presence of inflammatory cells directly responsible for initiating and maintaining myofiber injury. IMs are poorly characterized in veterinary medicine. A relatively large body of descriptive knowledge is available, however, many of the factors responsible for disease initiation and perpetuation remain less well-defined and the understanding of these mechanisms is mainly based on biological plausibility from animal models and human medicine. The majority of these disorders are considered to be autoimmune disorders in which skeletal muscle is inappropriately targeted by the immune system. Autoimmune diseases develop as a result of chronic inflammation owing to interactions between genes and the environment. However, the mechanisms by which autoimmune diseases evolve remain poorly understood. Numerous evidences support that the innate immune system (including cytokines and chemokines) and adaptive immune system (including autoantibodies and antigen-specific T cells) are involved in IM pathogenesis. Furthermore, several non-immune-mediated mechanisms contribute to the pathogenesis of these disorders, including cell-stress pathways, free radicals, altered energy metabolism, protein homeostasis and mitochondrial damages. Autoantibodies are present in approximately 60-70% of human patients with IM and often specific autoantibodies are strongly associated with specific forms of IMs. The role of autoantibodies in causing muscle damage and dysfunction is still debated, however, they have proven to be immensely useful biomarkers for the diagnosis, monitoring and prognosis of IMs. Therefore, there is great interest in the identification of myositis-specific autoantibodies. In the present Ph.D. thesis: 1. we have identified novel pathogenetic mechanisms underlying leishmania-associated inflammatory myopathy in dogs. In particular, we identified circulating autoantibodies that recognize the muscle protein sarcoplasmic/endoplasmic reticulum calcium ATPase 1 (SERCA1) as the main antigen, supporting the autoimmune mechanism underlying this myopathy and the antigen mimicry pathogenesis; 2. we morphologically and molecularly characterize the white striping myopathy of the broiler chicken and, based on our results, we hypothesize an associated immune-mediated mechanism; 3. we describe the morphological and molecular findings of the inflammatory myopathy associated with natural Feline Immunodeficiency Virus infection in cats and identify circulating anti-skeletal muscle autoantibodies.
