The impacts of climate change may be particularly severe for geographically isolated populations, which must adjust through plastic responses or evolve. Here, we study an endangered Arctic plant, Primula nutans ssp. finmarchica , confined to Fennoscandian seashores and showing indications of maladaptation to warming climate. We evaluate the potential of these populations to evolve to facilitate survival in the rapidly warming Arctic (i.e. evolutionary rescue) by utilizing manual crossing experiments in a nested half-sibling breeding design. We estimate G-matrices, evolvability and genetic constraints in traits with potentially conflicting selection pressures. To explicitly evaluate the potential for climate change adaptation, we infer the expected time to evolve from a northern to a southern phenotype under different selection scenarios, using demographic and climatic data to relate expected evolutionary rates to projected rates of climate change. Our results indicate that, given the nearly 10-fold greater evolvability of vegetative than of floral traits, adaptation in these traits may take place nearly in concert with changing climate, given effective climate mitigation. However, the comparatively slow expected evolutionary modification of floral traits may hamper the evolution of floral traits to track climate-induced changes in pollination environment, compromising sexual reproduction and thus reducing the likelihood of evolutionary rescue.

Endemic plants in high mountains are projected to be at high risk because of climate change. Temporal demographic variation is a major factor affecting population viability because plants often occur in small, isolated populations. Because isolated populations tend to exhibit genetic differentiation, analyzing temporal demographic variation in multiple populations is required for the management of high mountain endemic species. We examined the population dynamics of an endemic plant species, Primula farinosa subsp. modesta, in four subalpine sites over six years. Stage-based transition matrices were constructed, and temporal variation in the projected population growth rate (λ) was analyzed using life table response experiments (LTREs). The variation in λ was primarily explained by the site × year interaction rather than the main effects of the site and year. The testing sites exhibited inconsistent patterns in the LTRE contributions of the vital rates to the temporal deviation of λ. However, within sites, growth or stasis had significant negative correlations with temporal λ deviation. Negative correlations among the contributions of vital rates were also detected within the two testing sites, and the removal of the correlations alleviated temporal fluctuations in λ. The response of vital rates to yearly environmental fluctuations reduced the temporal variation of λ. Such effects manifested especially at two sites where plants exhibited higher plasticity than plants at other sites. Site-specific temporal variation implies that populations of high mountain species likely exhibit asynchronous temporal changes, and multiple sites need to be evaluated for their conservation., (© 2022. Sugar concentration in floral nectars is an assessment required in several diverse fields of application. The widely used analysis, consisting of nectar extraction with a microcapillary and sugar concentration measurement with a light refractometer, is not reliable when the nectar is secreted in small quantities, unextractable with a microcapillary. Ancillary methods adopted in such cases are destructive, rather complicated and often provide much less precise and accurate results. The microscopic-size, low cost and biocompatibility of optical fibers were exploited to deliver light directly inside the flower with minimal invasiveness and measure instantaneously the refractometric properties of the nectar without extracting it. After comparing the new and old methods using two known nectariferous species, the new approach was validated on Primula palinuri, whose nectar is unextractable with microcapillaries. The fiber-optic probe was able to measure the nectar refractive index in P. palinuri flowers making it possible to highlight a previously undetected significant trend of the sugar concentration throughout the long anthesis of the single flowers. Changes in nectar concentrations are similar in both longistylous and brevistylous flowers. The fiber-optic refractometer is an advancement of light refractometer analysis. The trait-fitness relationship influences the strength and direction of floral evolution. To fully understand and predict the evolutionary trajectories of floral traits, it is critical to disentangle the direct and indirect effects of floral traits on plant fitness in natural populations. We experimentally quantified phenotypic selection on floral traits through female fitness and estimated the casual effects of nectar robbing with different nectar robbing intensities on trait-fitness relationships in both the L- (long-style and short-anther phenotype) and S-morph (short-style and long-anther phenotype) flowers among Primula secundiflora populations. A larger number of flowers and wider corolla tubes had both direct and indirect positive effects on female fitness in the P. secundiflora populations. The indirect effects of these two traits on female fitness were mediated by nectar robbers. The indirect effect of the number of flowers on female fitness increased with increasing nectar robbing intensity. In most populations, the direct and/or indirect effects of floral traits on female fitness were stronger in the S-morph flowers than in the L-morph flowers. In addition, nectar robbers had a direct positive effect on female fitness, but this effect varied between the L- and S-morph flowers. Background: Primula is a large genus of flowering herbs well known for their heterostyly. Currently few natural hybrids are known and reproductive barriers in this genus in the wild have received little attention. However, there is instance of hybridization between rare and widely-spread species, and conservation implications of such situation is poorly understood. In the present study, we investigated hybridization patterns and reproductive barriers between a wide spread species, Primula poissonii and a rare species P. anisodora, of which only three populations are currently known., Results: Pollinator-mediated reproductive isolation was strong between parental species but not significant between hybrids and parental species. Hand pollination experiments showed significant reduction of both fruit- and seed-set for heterospecific pollination as compared with conspecific pollination for both parental species. Furthermore, hybrids had higher fruit- and seed-set when pollinated with P. anisodora pollen as opposed to P. poissonii pollen. Microsatellites identified backcrosses to P. anisodora in two of the three populations of P. anisodora, and additionally more individuals of P. anisodora showed introgression from P. poissonii than vice versa., Conclusions: These results provide evidence for potential genetic swamping of the P. anisodora populations, which could pose a serious threat for this locally endemic species.

In the model species Arabidopsis thaliana phytochromes mediate dormancy and germination responses to seasonal cues experienced during seed maturation on the maternal plants. However, the effect of the maternal light environment on seed germination in native wild species has not been well studied. This is particularly important given its practical application in the context of environmental restoration, when there can be marked changes in the canopy. Plants of Primula vulgaris were grown in the field over two vegetative seasons under four shading treatments from low to high ratio of red to far-red light (R:FR). Leaf and seed traits were assessed in response to the light treatments. The germination of seeds from these four maternal environments (pre-dispersal) was investigated at seven light and five temperature treatments (post-dispersal). Thinner leaves, larger leaf area and greater chlorophyll content were found in plants growing in reduced R:FR. Shading in the maternal environment led to increased seed size and yield, although the conditions experienced by the maternal plants had no effect on seed germination. Seeds responded strongly to the cues experienced in their immediate germination environment. Germination was always enhanced under higher R:FR conditions. The observed phenotypic trait variation plays a major role in the ability of P. vulgaris to grow in a wide range of light conditions. Distyly is a mechanism promoting cross-pollination within a balanced polymorphism. Numerous studies show that the degree of inter-morph sexual organ reciprocity (SOR) within species relates to its pollen-mediated gene flow. Similarly, a lower interspecific SOR should promote interspecific isolation when congeners are sympatric, co-blooming and share pollinators. In this comparative study, we address the significance of SOR at both intra- and interspecific levels. Seventeen allopatric and eight sympatric populations representing four Primula species (P. anisodora, P. beesiana, P. bulleyana and P. poissonii) native to the Himalaya-Hengduan Mountains were measured for eight floral traits in both long- and short-styled morphs. GLMM and spatial overlap methods were used to compare intra- and interspecific SOR. While floral morphology differed among four Primula species, SOR within species was generally higher than between species, but in species pairs P. poissonii/P. anisodora and P. beesiana/P. bulleyana, the SOR was high at both intra- and interspecific levels. We did not detect a significant variation in intraspecific SOR or interspecific SOR when comparing allopatric versus sympatric populations for all species studied. As intraspecific SOR increased, disassortative mating may be promoted. As interspecific SOR decreased, interspecific isolation between co-flowering species pairs also may increase. Premise of the Study: Much research has focused on plant responses to ongoing climate change, but there is relatively little information about how climate change will affect the early plant life history stages. Understanding how global warming and changes in winter snow pattern will affect seed germination and seedling establishment is crucial for predicting future alpine population and vegetation dynamics., Methods: In a 2-year study, we tested how warming and alteration in the snowmelt regime, both in isolation and combination, influence seedling emergence phenology, first-year growth, biomass allocation, and survival of four native alpine perennial herbs on the southeastern Tibetan Plateau., Key Results: Warming promoted seedling emergence phenology of all four species and biomass per plant of two species but reduced seedling survival of three species. Prolonged snow cover partly mediated the affects of warming on Primula alpicola (survival and biomass), Pedicularis fletcheri (phenology, biomass, and root:shoot ratio) and Meconopsis integrifolia (survival). For the narrowly distributed species M. racemosa, seedling growth was additively decreased by warming and prolonged snow cover., Conclusions: Both warming and alteration of the snow cover regime can influence plant recruitment by affecting seedling phenology, growth, and survival, and the effects are largely species-specific. Thus, climate change is likely to affect population dynamics and community structure of the alpine ecosystem. Background and Aims: The outbreeding floral polymorphism heterostyly frequently breaks down, resulting in the evolution of self-fertilization as a result of homostyle formation. Here, the loss of floral polymorphism in distylous Primula oreodoxa, a sub-alpine species restricted to western Sichuan, China, was examined by investigating the ecological correlates and genetic consequences of mating system transitions. Several key questions were addressed. (1) What are the frequencies, geographical distribution and reproductive characteristics of floral morphs in distylous and homostylous populations? (2) Does increased elevation influence pollinator service and the likelihood of inbreeding in populations? (3) How often has homostyly originated and what are the consequences of the breakdown of distyly for the amounts and distribution of genetic diversity in populations?, Methods: Fourteen populations throughout the range of P. oreodoxa were sampled, and morph frequencies and floral characteristics were recorded. Polymorphism at microsatellite loci and chloroplast DNA (cpDNA) variation were used to quantify population genetic structure and genetic relationships among populations. Controlled pollinations and studies of pollen tube growth and fertility were conducted to determine the compatibility status of populations and their facility for autonomous self-pollination. Finally, visitation rates of long- and short-tongued pollinators to distylous and homostylous populations at different elevations were compared to determine if increased elevation was associated with deterioration in pollinator service., Key Results: In contrast to most heterostylous species, both distylous and homostylous morphs of P. oreodoxa are highly self-compatible, but only homostyles have the facility for autonomous self-pollination. Homostyles set significantly more fruit and seeds following open pollination than the distylous morphs. Visitation by long-tongued pollinators was significantly lower in homostylous populations, and overall rates of insect visitation decreased with elevation. Genetic diversity was significantly lower in homostylous populations, with evidence of increased inbreeding at higher elevation. Patterns of cpDNA variation were consistent with multiple transitions from distyly to homostyly and limited gene flow among populations., Conclusions: The results of this study support the hypothesis that the multiple loss of floral polymorphism in distylous P. oreodoxa is associated with unsatisfactory pollinator service, with homostyles benefiting from reproductive assurance as a result of autonomous self-pollination., (© The Author 2017. The targets and causes of phenotypic selection are crucial to understanding evolutionary ecology. However, few studies have examined selection quantitatively from multiple sources on the same trait identified the agent of natural selection experimentally. Here we quantified phenotypic selection on traits, including flowering phenology and aspects of floral display via female fitness, in the distylous perennial herb Primula alpicola. To determine the role of pollinators in generating selection effects on floral traits, we compared the phenotypic selection gradients in open-pollinated and hand-pollinated plants. Our results show that pollinator-mediated linear selection on flowering start and correlational selection on the number of flowers and scape height explains most of the net phenotypic selection on these traits suggesting pollinators played an important role in shaping floral diversity. We used path analysis and structural equation modeling (SEM) to examine how herbivores affected the relationship between floral traits and female fitness, but no significant selection was caused by seed predators. These results suggest pollinators, not herbivores maybe the significant agent of selection on flora traits.

Understanding the effects of past climatic fluctuations on the distribution and population-size dynamics of cold-adapted species is essential for predicting their responses to ongoing global climate change. In spite of the heterogeneity of cold-adapted species, two main contrasting hypotheses have been proposed to explain their responses to Late Quaternary glacial cycles, namely, the interglacial contraction versus the interglacial expansion hypotheses. Here, we use the cold-adapted plant Primula farinosa to test two demographic models under each of the two alternative hypotheses and a fifth, null model. We first approximate the time and extent of demographic contractions and expansions during the Late Quaternary by projecting species distribution models across the last 72 ka. We also generate genome-wide sequence data using a Reduced Representation Library approach to reconstruct the spatial structure, genetic diversity, and phylogenetic relationships of lineages within P. farinosa. Finally, by integrating the results of climatic and genomic analyses in an Approximate Bayesian Computation framework, we propose the most likely model for the extent and direction of population-size changes in $P$. farinosa through the Late Quaternary. Our results support the interglacial expansion of $P$. farinosa, differing from the prevailing paradigm that the observed distribution of cold-adapted species currently fragmented in high altitude and latitude regions reflects the consequences of postglacial contraction processes., (© The Author(s) 2017. The goal of biological measurement is to capture underlying biological phenomena in numerical form. The reciprocity index applied to heterostylous flowers is meant to measure the degree of correspondence between fertile parts of opposite sex on complementary (inter-compatible) morphs, reflecting the correspondence of locations of pollen placement on, and stigma contact with, pollinators. Pollen of typical heterostylous flowers can achieve unimpeded fertilization only on opposite-morph flowers. Thus, the implicit goal of this measurement is to assess the likelihood of 'legitimate' pollinations between compatible morphs, and hence reproductive fitness. Previous reciprocity metrics fall short of this goal on both empirical and theoretical grounds. We propose a new measure of reciprocity based on theory that relates floral morphology to reproductive fitness. This method establishes a scale based on adaptive inaccuracy, a measure of the fitness cost of the deviation of phenotypes in a population from the optimal phenotype. Inaccuracy allows the estimation of independent contributions of maladaptive bias (mean departure from optimum) and imprecision (within-population variance) to the phenotypic mismatch (inaccuracy) of heterostylous morphs on a common scale. Background and Aims: Polyploidization plays a key role in plant evolution. Despite the generally accepted 'minority-cytotype exclusion' theory, the specific mechanisms leading to successful establishment and persistence of new polyploids remain controversial. The majority of newly formed polyploids do not become established, because they are less common, have fewer potential mates or may not be able to compete successfully with co-occurring progenitors at lower ploidy levels. Changes in floral traits and ecological niches have been proposed as important mechanisms to overcome this initial frequency-dependent disadvantage. The aim of this study was to determine whether dodecaploids of the heterostylous P. marginata differ from their hexaploid progenitors in P. marginata and P. allionii for selected floral traits and ecological preferences that might be involved in establishment and persistence, providing a possible explanation for the origin of polyploidized populations., Methods: Floral morphological traits and ecological niche preferences among dodecaploids and their hexaploid progenitors in P. marginata and P. allionii ,: all restricted to the south-western Alps, were quantified and compared KEY RESULTS: Differences in floral traits were detected between dodecaploids and their closest relatives, but such differences might be too weak to counter the strength of minority cytotype disadvantage and are unlikely to enable the coexistence of different cytotypes. Furthermore, the results suggest the preservation of full distyly and no transition to selfing in dodecaploids. Finally, dodecaploids occur almost exclusively in environments that are predicted to be suitable also for their closest hexaploid relatives., Conclusions: In light of the results, P. marginata dodecaploids have probably been able to establish and persist by occupying geographical areas not yet filled by their closest relatives without significant evolution in their climatic and pollination niches. Species distribution models (SDMs) have been criticized for involving assumptions that ignore or categorize many ecologically relevant factors such as dispersal ability and biotic interactions. Another potential source of model error is the assumption that species are ecologically uniform in their climatic tolerances across their range. Typically, SDMs treat a species as a single entity, although populations of many species differ due to local adaptation or other genetic differentiation. Not taking local adaptation into account may lead to incorrect range prediction and therefore misplaced conservation efforts. A constraint is that we often do not know the degree to which populations are locally adapted. Lacking experimental evidence, we still can evaluate niche differentiation within a species' range to promote better conservation decisions. We explore possible conservation implications of making type I or type II errors in this context. For each of two species, we construct three separate Max-Ent models, one considering the species as a single population and two of disjunct populations. Principal component analyses and response curves indicate different climate characteristics in the current environments of the populations. Model projections into future climates indicate minimal overlap between areas predicted to be climatically suitable by the whole species vs. population-based models. We present a workflow for addressing uncertainty surrounding local adaptation in SDM application and illustrate the value of conducting population-based models to compare with whole-species models. These comparisons might result in more cautious management actions when alternative range outcomes are considered.

Time lags in responses of organisms to deteriorating environmental conditions delay population declines and extinctions. We examined how local processes at the population level contribute to extinction debt, and how cycles of habitat deterioration and recovery may delay extinction. We carried out a demographic analysis of the fate of the grassland perennial Primula veris after the cessation of grassland management, where we used either a unidirectional succession model for forest habitat or a rotation model with a period of forest growth followed by a clear-cut and a new successional cycle. The simulations indicated that P. veris populations may have an extinction time of decades to centuries after a detrimental management change. A survey of the current incidence and abundance of P. veris in sites with different histories of afforestation confirmed the simulation results of low extinction rates. P. veris had reduced incidence and abundance only at sites with at least 100 years of forest cover. Time to extinction in simulations was dependent on the duration of the periods with favourable and unfavourable conditions after management cessation, and the population sizes and growth rates in these periods. Our results thus suggest that the ability of a species to survive is a complex function of disturbance regimes, rates of successional change, and the demographic response to environmental changes. Detailed demographic studies over entire successional cycles are therefore essential to identify the environmental conditions that enable long-term persistence and to design management for species experiencing extinction debts.

Under the pressure of global warming, general expectations of species migration and evolution of adaptive traits should always be confirmed with species-specific studies. Within this framework, some species can be used as study systems to predict possible consequences of global warming also on other relatives. Unlike its mountain congeneric, Primula palinuri Petagn. has endured all the climatic fluctuations since the Pleistocene, while surviving on Mediterranean coastal cliffs. The aim of this work was to investigate the possible evolution of reproductive biological and ecological traits in P. palinuri adaptation to a warmer environment. Data showed that flowering starts in mid-winter; single flowers remain open for over a month, changing from pendulous to erect. The number of insects visiting flowers of P. palinuri increases during the flowering season, and pollination reduces flower longevity. Overall, the best pollen performances, in terms of viability and germinability, occur at winter temperatures, while pollinator activity prolongs flowering until spring. Moreover, extended longevity of single flowers optimises reproductive success. Both phenotypic plasticity and selective processes might have occurred in P. palinuri. However, we found that reproductive traits of the only Mediterranean Primula remain more associated with cold mountain habitats than warm coastal cliffs. Given the rapid trend of climate warming, migration and new adaptive processes in P. palinuri are unlikely. Heterostyly is a floral polymorphism characterized by reciprocal herkogamy maintained through high levels of mating between morphs, serviced by appropriate pollinators. We studied how differential efficiency and abundance of distinct pollinators affect plant female reproduction in self- and intra-morph incompatible distylous Primula secundiflora. Bumblebees and syrphid flies were found to be the most abundant floral visitors. Bumblebees frequently exhibited nectar-robbing behavior. Because the robbing holes were always situated between the high- and low-level organs on both morphs, nectar-robbing bumblebees only pollinated S-styled flowers. L-styled flowers set four times as many seeds as did S-styled flowers after being visited by pollen-collecting syrphid flies. The natural female fecundity and the magnitude of pollen limitation varied between the morphs within populations because of the mosaic distribution of nectar-robbing bumblebees and syrphid flies. L-styled flowers and S-styled flowers set the same number of seeds after supplemental hand pollination, indicating equivalent female reproductive potential. We suggest that bumblebee nectar robbers and syrphid flies play an important role in sustaining the floral dimorphism of heterostyly in P. secundiflora because of their complementary roles in the pollination system.

Background and Aims: Distyly is a floral polymorphism characterized by the presence of two discrete morphs with reciprocal positioning of anthers and stigmas in flowers on different plants within the same population. Although reciprocal herkogamy and associated floral traits are generally thought to be discrete and strict polymorphisms, little is known about variation in floral traits related to the distylous syndrome within and among populations of a single species. In this study, variation in floral morphology and reciprocal positioning of the sexual organs in the distylous Primula veris (cowslip) is quantified., Methods: Data were collected in ten populations occurring in two contrasting habitat types (grasslands and forests), and for each population the average level of reciprocity was assessed, the strength of the self-incompatibility system was determined, and seed production under natural conditions was quantified., Results: In grassland populations, flowers showed clear distyly with low and symmetric reciprocity indices at both the lower and upper level. In forests, P. veris produced larger flowers that showed strong deviations in stigma-anther separation, especially in the L-morph. This deviation was mainly driven by variation in stigma height, resulting in high and asymmetric reciprocity indices and the occurrence of several short-styled homostylous plants. Self-incompatibility was, however, strict in both habitats, and morph ratios did not differ significantly from isoplethy. The observed shift in reciprocity in forest populations was associated with a significant reduction in seed production in the L-morph., Conclusions: The results indicate that populations of P. veris show habitat-specific variation in flower morphology. Deviations from perfect reciprocal positioning of stigmas and anthers also translate into reduced seed production, suggesting that small changes in sexual organ reciprocity can have far-reaching ecological and evolutionary implications., (© The Author 2014. Life cycles can limit the abilities of species to track changing climatic conditions. We combined age or stage structure and a moving-habitat model to explore the effects of life history on the persistence of populations in the presence of climate change. We studied four dissimilar plant species in moving patches and found that (1) population growth rates, (2) elasticities with respect to the survival (stasis and shrinkage) components of the projection matrix, and (3) the evenness of the elasticities with respect to the components of the projection matrix all decreased as we increased the translational speeds of the patches. In addition, the value of long-distance dispersal increased with patch speed for three of the four species. Our analyses confirm that rapid growth, high fecundity, and long-distance dispersal can benefit species in moving patches. Thus, species with long generation times and limited dispersal ability are especially vulnerable to habitat movement. Stage-structured moving-habitat models can easily incorporate spatial complexity and can help us predict the effects of shifting climatic conditions.

Background and Aims: Heteromorphy in flowers has a profound effect on breeding patterns within a species, but little is known about how it affects reproductive barriers between species. The heterostylous genus Primula is very diverse in the Himalaya region, but hybrids there have been little researched. This study examines in detail a natural hybrid zone between P. beesiana and P. bulleyana., Methods: Chloroplast sequencing, AFLP (amplified fragment length polymorphism) markers and morphological comparisons were employed to characterize putative hybrids in the field, using synthetic F1s from hand pollination as controls. Pollinator visits to parent species and hybrids were observed in the field. Hand pollinations were conducted to compare pollen tube growth, seed production and seed viability for crosses involving different morphs, species and directions of crossing., Key Results: Molecular data revealed all hybrid derivatives examined to be backcrosses of first or later generations towards P. bulleyana: all had the chloroplast DNA (cpDNA) of this species. Some individuals had morphological traits suggesting they were hybrids, but they were genetically similar to P. bulleyana; they might have been advanced generation backcrosses. Viable F1s could not be produced with P. bulleyana pollen on P. beesiana females, irrespective of the flower morphs used. Within-morph crosses for each species had very low (<10 %) seed viability, whereas crosses between pin P. bulleyana (female) and pin P. beesiana had a higher seed viability of 30 %. Thus genetic incompatibility mechanisms back up mechanical barriers to within-morph crosses in each species, but are not the same between the two species. The two species share their main pollinators, and pollinators were observed to fly between P. bulleyana and hybrids, suggesting that pollinator behaviour may not be an important isolating factor., Conclusions: Hybridization is strongly asymmetric, with P. bulleyana the only possible mother and all detected hybrids being backcrosses in this direction. Partial ecological isolation and inhibition of heterospecific pollen, and possibly complete barriers to F1 formation on P. beesiana, may be enough to make F1 formation very rare in these species. Therefore, with no F1 detected, this hybrid zone may have a finite life span as successive generations become more similar to P. bulleyana.

Variation in mutualistic and antagonistic interactions are important sources of variation in population dynamics and natural selection. Environmental heterogeneity can influence the outcome of interactions by affecting the intensity of interactions, but also by affecting the demography of the populations involved. However, little is known about the relative importance of environmental effects on interaction intensities and demographic sensitivity for variation in population growth rates. We investigated how soil depth, soil moisture, soil nutrient composition, and vegetation height influenced the intensity of seed predation as well as host plant demography and sensitivity to seed predation in the perennial herb Primula farinosa. Intensity of seed predation ranged from 0% to 80% of seeds damaged among the 24 study populations and was related to soil moisture in two of four years. The effect of seed predation on plant population growth rate (lambda) ranged from negligible to a reduction in lambda by 0.70. Sensitivity of population growth rate to predation explained as much of the variation in the reductions in population growth rate due to seed predation as did predation intensity. Plant population growth rate in the absence of seed predation and sensitivity to predation were negatively related to soil depth and soil moisture. Both intensity of predation and sensitivity to predation were positively correlated with potential population growth rate and, as a result, there was no significant relationship between predation intensity and realized population growth rate. We conclude that in our study system environmental context influences the effects of seed predation on plant fitness and population dynamics in two important ways: through variation in interaction intensity and through sensitivity to the effects of this interaction. Moreover, our results show that a given abiotic factor can influence population growth rate in different directions through effects on potential growth rate, intensity of biotic interactions, and the sensitivity of population growth rate to interactions.

The deposition of surface (farinose) flavonoids on aerial parts of some Primula species is a well-documented but poorly understood phenomenon. Here, we show that flavonoid deposition on the leaves and winter buds may contribute strongly to preventing freezing damage in these plants. The ice nucleation temperature of fairy primrose (Primula malacoides) leaves covered with natural flavone was approximately 6 °C lower compared to those that had their flavone artificially removed. Additionally, farinose flavonoids on the leaves reduced subsequent electrolyte leakage (EL) from the cells exposed to freezing temperatures. Interestingly, exogenous application of flavone at 4 mg/g fresh weight to P. malacoides leaves, which had the original flavone mechanically removed, restored freezing tolerance, and diminished EL from the cells to pretreatment values. Our results suggest that farinose flavonoids may function as mediators of freezing tolerance in P. malacoides, and exogenous application of flavone could be used to reduce freezing damage during sudden but predictable frost events in other plant species., (© 2013 Institute of Botany, Chinese Academy of Sciences.)

Reduction of pollen flow can affect plant abundance and population viability and cause selection on plant mating system and floral traits. Little is known on the effect of this phenomenon in species naturally restricted to small and isolated habitats, that may have developed strategies to cope with long-term isolation and small population size. We investigated the pollination ecology of the endemic distylous winter-flowering P. allionii to verify the possible limitation of female fitness due to reduced pollinator visits. We recorded a higher production of pollen grains in long-styled morph, and a higher seed set in short-styled morph. The high intra-morph variability of sexual organ position may explain the hybridization phenomena allowing and easier intra-morph pollination. The fruit set is constant, although its winter-flowering period might decrease pollen transfer. Nevertheless, the lower competition for pollinators with neighbouring plants and the long-lasting anthesis may offset its reproductive success. Even if our results show no evidence of imminent threats, changes in plant-pollinator interactions might increase inbreeding, resulting in an increased extinction risk.

Spatial variation in the direction of selection drives the evolution of adaptive differentiation. However, few experimental studies have examined the relative importance of different environmental factors for variation in selection and evolutionary trajectories in natural populations. Here, we combine 8 y of observational data and field experiments to assess the relative importance of mutualistic and antagonistic interactions for spatial variation in selection and short-term evolution of a genetically based floral display dimorphism in the short-lived perennial herb Primula farinosa. Natural populations of this species include two floral morphs: long-scaped plants that present their flowers well above the ground and short-scaped plants with flowers positioned close to the ground. The direction and magnitude of selection on scape morph varied among populations, and so did the frequency of the short morph (median 19%, range 0-100%; n = 69 populations). A field experiment replicated at four sites demonstrated that variation in the strength of interactions with grazers and pollinators were responsible for among-population differences in relative fitness of the two morphs. Selection exerted by grazers favored the short-scaped morph, whereas pollinator-mediated selection favored the long-scaped morph. Moreover, variation in selection among natural populations was associated with differences in morph frequency change, and the experimental removal of grazers at nine sites significantly reduced the frequency of the short-scaped morph over 8 y. The results demonstrate that spatial variation in intensity of grazing and pollination produces a selection mosaic, and that changes in biotic interactions can trigger rapid genetic changes in natural plant populations.

Background: Primula species are important early spring garden plants with a centre of diversity and speciation in the East Himalaya-Hengduan Mountains in Western China. Studies on population genetics, speciation and phylogeny of Primula have been impeded by a lack of genomic resources. In the present study, we sequenced the transcriptomes of two closely related primrose species, Primula poissonii and Primula wilsonii, using short reads on the Illumina Genome Analyzer platform., Results: We obtained 55,284 and 55,011 contigs with N50 values of 938 and 1,085 for P. poissonii and P. wilsonii, respectively, and 6,654 pairs of putative orthologs were identified between the two species. Estimations of non-synonymous/synonymous substitution rate ratios for these orthologs indicated that 877 of the pairs may be under positive selection (Ka/Ks > 0.5), and functional enrichment analysis revealed that significant proportions of the orthologs were in the categories DNA repair, stress resistance, which may provide some hints as to how the two closely related Primula species adapted differentially to extreme environments, such as habitats characterized by aridity, high altitude and high levels of ionizing radiation. It was possible for the first time to estimate the divergence time between the radiated species pair, P. poissonii and P. wilsonii; this was found to be approximately 0.90 ± 0.57 Mya, which falls between the Donau and Gunz glaciation in the Middle Pleistocene. Primers based on 54 pairs of orthologous SSR-containing sequences between the two Primula species were designed and verified. About half of these pairs successfully amplified for both species. Of the 959 single copy nuclear genes shared by four model plants (known as APVO genes), 111 single copy nuclear genes were verified as being present in both Primula species and exon-anchored and intron-spanned primers were designed for use., Conclusion: We characterized the transcriptomes for the two Primula species, and produced an unprecedented amount of genomic resources for these important garden plants. Evolutionary analysis of these two Primula species not only revealed a more precise divergence time, but also provided some novel insights into how differential adaptations occurred in extreme habitats. Furthermore, we developed two sets of genetic markers, single copy nuclear genes and nuclear microsatellites (EST-SSR). Both these sets of markers will facilitate studies on the genetic improvement, population genetics and phylogenetics of this rapidly adapting taxon.

The shape of the relationship between intensity of biotic interactions and strength of selection is important for spatial variation in selection, but is little explored. We quantified interactions and selection in 69 populations of the short-lived herb Primula farinosa. As predicted because of saturation and depletion effects, the strength of selection on a discrete and on a continuously varying floral display trait were in several cases significantly non-linearly related to the mean intensity of mutualistic and antagonistic interactions. Strength of selection was strongest at low levels of fruit initiation and at high intensities of seed predation. Seed predation varied more among populations than did fruit initiation and could explain a larger proportion of the among-population variation in strength of selection. Our results support the contention that interaction intensity affects selection strength, and suggests that for mutualistic and antagonistic interactions that can be saturated or depleted, this relationship is sometimes non-linear., (© 2012 Blackwell Publishing Ltd/CNRS.)

A combined study was carried out on the micromorphology and chemistry of glandular trichomes with focus on Primula vialii and P. vulgaris, respectively. Epifluorescence microscopy was applied to study the auto-fluorescent properties of flavonoids and their localization in glandular trichomes. Both species differed in the morphology of the glandular trichomes and in the exudate flavonoid composition. Leaf glands from P. vialii and from some species of subgen. Primula exhibited uniform glandular fluorescence, but notable differentiation was observed within a single leaf of P. vulgaris. Our observations indicate that exudate flavonoids are not transported from the tissue to the glandular hairs. Conversely, only the newly isolated glycoside 1 (kaempferol 3-O-(2"'-rhamnosyl)-robinobioside) was obtained from leaf tissue of P. vialii after removal of the exudate. Its structure was confirmed by NMR and mass spectrometry. This glycoside was not detected in tissue extracts of P. vulgaris after similar treatment. The observed chemical diversity is discussed, with focus on possible correlation with glandular structures and tissue differentiation in Primula, and also against other studied species. Aspects of biosynthesis in relation to tissue-specific flavonoid diversification are shortly addressed.

Background and Aims: Heterostyly is a floral polymorphism that has fascinated evolutionary biologists since Darwin's seminal studies on primroses. The main morphological characteristic of heterostyly is the reciprocal placement of anthers and stigmas in two distinct (distyly) floral morphs. Variation in the degree of intermorph sexual reciprocity is relatively common and known to affect patterns of pollen transfer within species. However, the partitioning of sexual organ reciprocity within and between closely related species remains unknown. This study aimed at testing whether intermorph sexual reciprocity differs within vs. between primrose species that hybridize in nature and whether the positions of sexual organs are correlated with other floral traits., Methods: Six floral traits were measured in both floral morphs of 15 allopatric populations of Primula elatior, P. veris and P. vulgaris, and anther-stigma reciprocity was estimated within and between species. A combination of univariate and multivariate approaches was used to test whether positions of reproductive organs were less reciprocal between than within species, to assess correlations between sexual organ positions and other corolla traits, and to quantify differences between morphs and species., Key Results: The three species were morphologically well differentiated in most floral traits, except that P. veris and P. vulgaris did not differ significantly in sexual organ positions. Overall, lower interspecific than intraspecific sexual organ reciprocity was detected. This decrease was marked between P. elatior and P. vulgaris, intermediate and variable between P. elatior and P. veris, but negligible between P. veris and P. vulgaris., Conclusions: Differences in anther and stigma heights between the analysed primrose species were of the same magnitude or larger than intraspecific differences that altered pollen flow within other heterostylous systems. Therefore, it is possible to suggest that considerable reductions of sexual organ reciprocity between species may lower interspecific pollen flow, with potential effects on reproductive isolation.

Cliffs worldwide are known to be reservoirs of relict biodiversity. Despite the presence of harsh abiotic conditions, large endemic floras live in such environments. Primula palinuri Petagna is a rare endemic plant species, surviving on cliff sites along a few kilometres of the Tyrrhenian coast in southern Italy. This species is declared at risk of extinction due to human impact on the coastal areas in question. Population surveys have shown that most of the plants are old individuals, while seedlings and plants at early stages of development are rare. We followed the growth of P. palinuri plants from seed germination to the adult phase and analysed the morphoanatomical traits of plants at all stages of development. Our results showed that the pressure of cliff environmental factors has been selected for seasonal habitus and structural adaptive traits in this species. The main morphoanatomical modifications are suberized cell layers and accumulation of phenolic compounds in cell structures. These features are strictly related to regulation of water uptake and storage as well as defence from predation. However, we found them well established only in adult plants and not in juvenile individuals. These findings contribute to explain the rare recruitment of the present relict populations, identifying some of the biological traits which result in species vulnerability.

Agricultural land use results in direct biodiversity decline through loss of natural habitat, but may also cause indirect cross-habitat effects on conservation areas. We conducted three landscape-scale field studies on 67 sites to test the hypothesis that mass flowering of oilseed rape (Brassica napus) results in a transient dilution of bees in crop fields, and in increased competition between crop plants and grassland plants for pollinators. Abundances of bumble-bees, which are the main pollinators of the grassland plant Primula veris, but also pollinate oilseed rape (OSR), decreased with increasing amount of OSR. This landscape-scale dilution affected bumble-bee abundances strongly in OSR fields and marginally in grasslands, where bumble-bee abundances were generally low at the time of Primula flowering. Seed set of Primula veris, which flowers during OSR bloom, was reduced by 20 per cent when the amount of OSR within 1 km radius increased from 0 to 15 per cent. Hence, the current expansion of bee-attractive biofuel crops results in transient dilution of crop pollinators, which means an increased competition for pollinators between crops and wild plants. In conclusion, mass-flowering crops potentially threaten fitness of concurrently flowering wild plants in conservation areas, despite the fact that, in the long run, mass-flowering crops can enhance abundances of generalist pollinators and their pollination service.

Herbivores can have strong deleterious effects on plant growth, reproduction, and even survival. Because these effects might be strongly interrelated, the direct consumptive effects of herbivores and a variety of indirect effects are difficult to untangle. Reductions in growth, for example, may strongly impact the flowering behaviour of plant species in the current season, but at the same time incur costs to survival, growth and reproduction in the next growing season(s). To get better insights in the effects of herbivory on the flowering behaviour of the long-lived polycarpic grassland herb Primula veris L., flowering patterns were monitored over ten consecutive years under two treatments (grazing and control mowing regimes). We tested the hypothesis that the size at flowering was affected by the presence of herbivores, and whether this translated into costs to future reproduction and survival. Overall, grazed plants were significantly smaller than control plants, and the size at which plants flowered was also significantly smaller when herbivores were present. The transition probability of flowering and of surviving into the next year was significantly smaller for all plants in the current year if they had been grazed than if they had been mown, indicating that herbivory incurred costs to both flowering and survival. Grazed plants also needed longer to start flowering, had fewer flowers and flowered less frequently, causing a significantly lower proportion of flowering adults in the population. These results suggest that the observed regression in plant size due to herbivory does not allow plants to capture enough resources to guarantee regular flowering in the longer run.

In flowering plants, pollen dispersal is often the major contributing component to gene flow, hence a key parameter in conservation genetics and population biology. A cost-effective method to assess pollen dispersal consists of monitoring the dispersal of fluorescent dyes used as pollen analogues. However, few comparisons between dye dispersal and realized pollen dispersal have been performed to validate the method. We investigated pollen dispersal in two small populations of the insect-pollinated herb Primula elatior from urban forest fragments using direct (paternity analyses based on microsatellite DNA markers) and indirect (fluorescent dyes) methods. We compared these methods using two approaches, testing for the difference between the distance distributions of observed dispersal events and estimating parameters of a dispersal model, and related these results to dye dispersal patterns in three large populations. Dye and realized (based on paternity inference) pollen dispersal showed exponential decay distributions, with 74.2-94.8% of the depositions occurring at <50 m and a few longer distance dispersal events (up to 151 m). No significant difference in curve shape was found between dye and realized pollen dispersal distributions. The best-fitting parameters characterizing the dye dispersal model were consistent with those obtained for realized pollen dispersal. Hence, the fluorescent dye method may be considered as reliable to infer realized pollen dispersal for forest herbs such as P. elatior. However, our simulations reveal that large sample sizes are needed to detect moderate differences between dye and realized pollen dispersal patterns because the estimation of dispersal parameters suffers low precision.

Nectar robbers are thought rarely to pollinate flowers, especially those with sexual organs hidden within corollas. In this study, we examined whether robbers pollinate flowers of distylous Primula secundiflora. Distylous plants have two floral morphs. Pin flowers have long styles and short stamens, and thrum flowers have short styles and long stamens. Flowers of P. secundiflora were commonly robbed by bumble-bees, and robbing holes were always situated between high and low sexual organs for both floral morphs. We observed that pollen grains of pin flowers were removed while thrum flowers received fresh pollen grains immediately after flowers were robbed. We manipulated flowers so that only nectar robbers could visit them. This resulted in 98 per cent of thrum flowers and 6 per cent of pin flowers setting fruit, and seed number per thrum fruit was also significantly higher than per pin fruit. Our findings suggest that nectar robbers transfer pollen from pin flowers to thrum flowers effectively, and consequently increase male fitness of the pin morph and female fitness of the thrum morph. Such asymmetrical pollen flow caused by nectar robbers may act as an important selective agent in floral fitness and evolution of distyly.

Recent advances in stochastic demography provide tools to examine the importance of random and periodic variation in vital rates for population dynamics. In this study, we explore with simulations the effect of disturbance regime on population dynamics and viability. We collected 7 years of demographic data in three populations of the perennial herb Primula farinosa, and used these data to examine how variation in vital rates affected population viability parameters (stochastic growth rate, lambda(S)), and how vital rates were related to weather conditions. Elasticity analysis indicated that the stochastic growth rate was very sensitive to changes in regeneration, quantified as the production, survival, and germination of seeds. In one of the study years, all seedlings and mature plants in the demography plots died. This extinction coincided with the driest summer during the study period. Simulations suggested that a future increase in the frequency of high-mortality years due to climate change would result in reduced population growth rate, and an increased importance of survival in the seed bank for population viability. The results illustrate how the limited demographic data typically available for many natural systems can be used in simulation models to assess how environmental change will affect population viability.

The reproductive ability related to the population size of the endangered and endemic species Primula merrilliana Schltr. was investigated. In 26 populations observed, only four contain more than 500 flowering individuals, whereas most of them (53.8%) consist of less than 100 flowering individuals. Though the ratio of pin and thrum plants keeps its balance at 1:1 for all populations, the frequency of pin and thrum flowers was significantly biased in most small populations. Population size strongly affected reproductive success; plants in small populations produced significantly fewer flowers and fruits per plant and fewer seeds per fruit, and therefore fewer seeds per plant. The floral density was another important factor that influenced the reproductive success of P. merrilliana, because four main reproductive success parameters (fruits per plant, seeds per fruit, seeds per plant, and the proportion of flowers setting fruit) were all positively correlated with floral density. The size of plants and the number of leaves per plant (measure of habitat quality) were not influenced by the variation of population size, suggesting that the reduced fecundity in small populations may not be a consequence of lower habitat quality. Inbreeding depression and pollen limitation as a result of less attractiveness in small populations are therefore likely explanations for these patterns.

Theory predicts that, with conflicting selection pressures mediated by mutualists and antagonists, alternative reproductive strategies can be maintained by negative frequency-dependent selection if it results in rare-morph advantage. We combined field experiments and surveys of natural populations to determine whether selection on floral display is frequency dependent in the self-incompatible herb Primula farinosa, which is polymorphic for inflorescence height and occurs in a short-scaped and a long-scaped morph. Among short-scaped plants, both pollination success, quantified as initiation of fruits and seeds, and seed predation were positively correlated with the relative frequency of the long-scaped morph. The relative strength of these effects and the direction of the resulting frequency-dependent selection on scape morph varied among years and populations. The results suggest that both mutualists and antagonists may mediate frequency-dependent selection and that frequency dependence may vary from positive to negative with rare-morph advantage, depending on the relative strength of these interactions.

The flowering phenology of alpine-snowbed plants varies widely depending on the time of snowmelt. This variation may cause spatial and temporal heterogeneity in pollen dispersal, which in turn may influence genetic structure. We used spatial autocorrelation analyses to evaluate relative effect of segregation in flowering time and physical distance on fine-scale spatial genetic structure (SGS) of a snowbed herb Primula cuneifolia sampled in 10-m grids within a continuous snow patch (110 x 250 m) using nine allozyme loci. Although the individual flower lasts for

Primula sieboldii E. Morren is a clonal herbaceous species with a short foliar period from spring to early summer. We have studied the temperature-dependence of the rate of respiration at the whole-ramet level throughout the phenological stages of P. sieboldii to reveal its photosynthate-utilization strategy. P. sieboldii ramets were grown in a chamber enabling simulation of seasonal changes in temperature. Rates of respiration were measured at three phenological stages--the foliar period, the before-chilling defoliated (BCD) period, and the after--chilling defoliated (ACD) period. In the foliar period the rate of respiration, on a biomass basis at 20 degrees C (R (20)), of the above-ground plant parts was 2.5 times that of the below-ground parts. The R (20) of the below-ground parts in the foliar period was 6.5 times that in the BCD period and 1.6 times that in the ACD period. Estimation of the ramet carbon balance under these growth conditions showed that ramets respired 87% of total photosynthate production during the experimental period (8.5 months). Respiratory consumption in the foliar period accounted for 70% of the yearly total, whereas 24 and 6% were consumed in the BCD and ACD periods, respectively. An extremely low rate of respiration during the long defoliated period led to a positive net annual carbon balance for P. sieboldii ramets.

Grazing reduces litter thickness and vegetation height and may thereby indirectly affect reproductive success and selection on floral characters in plants with prostrate growth. Reductions in litter thickness and vegetation height should influence both the resource status of plants with leaves positioned close to the ground and the significance of inflorescence height for interactions with pollinators and seed predators. We experimentally examined how simulated grazing of surrounding vegetation affected pollen limitation, fruit predation and fecundity of short-scaped and long-scaped Primula farinosa, which differ markedly in floral display and therefore in expected attractiveness to pollinators. Litter removal and pruning of surrounding vegetation increased fruit and seed production per plant in the year of the treatment and the probability of flowering in the following year. Pollen limitation of fruit and seed production was stronger in the short-scaped morph than in the long-scaped morph, but was not significantly affected by litter removal and simulated grazing of surrounding vegetation. Supplemental hand-pollination reduced seed size in the year of the treatment and flowering probability in the second year, and these effects did not differ among scape morphs or grazing treatments. The results suggest that grazing indirectly favours seed production in P. farinosa, mainly because it increases the resource status of plants that escape damage. Contrary to expectation, there was no strong evidence that litter accumulation and tall vegetation increase the severity of pollen limitation or reduce the relative performance of the short-scaped morph.

Population context should influence pollination success and selection on floral display in animal-pollinated plants because attraction of pollinators depends not only on the characteristics of individual plants, but also on the attractiveness of co-occurring conspecifics. The insect-pollinated herb Primula farinosa is polymorphic for inflorescence height. Natural populations may include both long-scaped plants, which present their flowers well above the soil surface, and short-scaped plants, with their flowers positioned close to the ground. We experimentally tested whether seed production in short-scaped P. farinosa varied with local morph frequency and surrounding vegetation height. In tall vegetation, short-scaped plants in polymorphic populations produced more fruit and tended to produce more seeds than short-scaped plants did in monomorphic populations. In low vegetation, population composition did not significantly affect fruit and seed output of short-scaped plants. The results suggest that long-scaped plants facilitate short-scaped plants in terms of pollinator attraction and that the facilitation effect is contingent on the height of the surrounding vegetation. The documented facilitation should contribute to the maintenance of the scape length polymorphism in ungrazed areas where litter accumulates and vegetation grows tall.

Although the effects of deterministic factors on population viability often are more important than stochasticity, few researchers have dealt with the effect of deterministic habitat changes on plant population demography. We assessed population viability for the perennial herb Primula veris L. and identified targets for management based on demographic data from five different habitat types representing different degrees of canopy closure. We conducted replicate studies at the border of the distribution area and in more central parts. Demographic patterns were similar between the two regions. Most study populations had a positive population growth, and only populations in late phases of forest succession showed consistently negative trends. The populations of open habitats had high seedling recruitment, and the populations of early and middle forest succession had high seed production. The importance of survival for population growth rate increased with increasing habitat closure, whereas the importance of growth and reproduction decreased. Results of the elasticity analysis suggested that the best method to manage decreasing late-successional populations is to increase survival of the largest individuals. The life-table response experiment (LTRE) analysis, however showed that survival of the largest individuals contributed little to differences in population growth rates of different habitats, whereas seed production and growth of small individuals were more important. Moreover direct perturbation of the performance of the largest stages showed that late-successional populations would not attain positive population growth even if the largest stages had no mortality at all. We conclude that restoration of recruitment is the only possibility for positive population growth in late-successional populations of P. veris, although the elasticities of recruitment transitions are low. Our results also suggest that retrospective demographic methods such as LTRE constitute an important and necessary complement to prospective methods such as elasticities in identifying management targets.

Hummock-and-hollow microtopography is common in wetlands of the Qinghai-Tibetan Plateau. The physical environment of hummocks contrasts strongly with that of hollows. To address how Primula nutans Georgi, a herbaceous species broadly distributed on the plateau, can inhabit both hummocks and hollows, we investigated the plasticity of its morphology and biomass growth in relation to the environmental variables during the growing season. The results are as follows: (1) total daily photosynthetic photon flux density, maximum soil temperature, and diurnal soil temperature range were significantly larger on the hummocks than in the hollows; (2) individual ramets had smaller leaves, higher leaf mass per unit area, and shorter petiole and peduncle length on the hummocks, but leaf and root dry weights per ramet differed little between microsites; (3) P. nutans allocated most of its dry mass to peduncles and flowers and the proportion of reproductive mass was significantly higher for plants in the hollows than on the hummocks; (4) the coefficients of variation of most morphological and biometric parameters was higher on hummocks than in hollows. The results suggest that P. nutans favors hummocks but can tolerate hollows through morphological adjustments in that the mass investment to peduncles and flowers is much larger in plants in the hollows than hummocks.

Primula (c. 430 species) and relatives (Primulaceae) are paradigmatic to our understanding of distyly. However, the common co-occurrence of distyly and monomorphy in closely related groups within the family has made the interpretation of its evolution difficult.Here, we infer a chloroplast DNA (cpDNA) phylogeny for 207 accessions, including 51% of the species and 95% of the sections of Primula with monomorphic populations, using Bayesian methods. With this tree, we infer the distribution of ancestral states on critical nodes using parsimony and likelihood methods. The inferred cpDNA phylogeny is consistent with prior estimates. The most recent common ancestor (MRCA) of Primula is resolved as distylous using both methods of inference. However, whether the distyly in Primula, Hottonia, and Vitaliana arose once or three independent times is not clear. We conclude that monomorphism in descendants of the MRCA of Primula is derived from distyly in all cases. Thus, scenarios for the evolution of distyly that rely on the persistence of primitive monomorphy (such as in Primula section Sphondylia) require re-evaluation.

The methods of production of inter-specific hybrids in Primula are categorized into four steps: (1) emasculation, (2) pollination, (3) rescue culture of immature embryo, and (4) confiration of hybridity and ploidy level of the regenerated plants. Although most of the Primula pecies have a heteromorphic self-incompatibility system, an emasculation step is usually needed to avoid self-pollination since self-incompatibility is not always complete. At the resue culture step, addition of plant hormones (e.g., auxin, cytokinin, and gibberellin) to the culture medium is proved to be effective. The hybridity of the plants is efficiently confirmed at seedling stage by DNA analysis in addition to the comparison of morphological characters. The analysis of relative DNA contents by flow cytometry is easy and rapid technique to confirm hybridity and to estimate ploidy level and genomic combination.

In plant populations where reproductive output is limited by pollinator visitation, plants with attractive floral displays should have a selective advantage. We examined the effect of inflorescence height on pollination success in Primula farinosa, which is dimorphic for scape length. To test the hypothesis that fruit and seed initiation are more strongly pollen-limited in the short-scaped than in the long-scaped morph, and that this difference is affected by spatio-temporal variation in pollen limitation, we conducted a hand-pollination experiment in four populations over 2 yr. Pollen limitation of fruit initiation varied among populations and years, and was stronger in the short-scaped than in the long-scaped morph. The results suggest that interactions with pollinators will need to be considered for a full understanding of the maintenance of this striking polymorphism. The study also shows that, although pollen limitation is likely to vary in space and time in many plant species, such variation is not necessarily associated with variation in selection on floral characters.

Background and Aims: Petals are important for Primula sieboldii because of the commercial value of its flowers, and their form is a target characteristic for breeding. An appropriate understanding of petal form in terms of genetic mechanisms and environmental effects is necessary for improvement of this species. The aim of this study was to establish a quantitative evaluation method of petal shape by elliptic Fourier descriptors and principal component analysis (EF-PCA), and thus to investigate genotypic and environmental effects on petal morphology., Methods: EF-PCA describes an overall shape mathematically by transforming coordinate information concerning its contours into elliptic Fourier descriptors (EFDs) and summarizing the EFDs by principal component analysis. To examine varietal effects on principal component (PC) scores and petal area among commercial varieties, nested ANOVAs were performed (since the samples had a hierarchical structure with four sources, i.e. variety, plant, flower and petal)., Key Results: Petal shape variation could be evaluated successfully and the symmetrical and asymmetrical elements of the overall shape variation could be detected. The proportions of the variance component due to varietal differences were more than 70 % in the first five PCs of the symmetrical elements and petal area. By contrast, the proportions due to varietal effects of all PCs of the asymmetrical elements were less than 20 %, and the proportions of the variation within a flower were more than 75 %. It was also demonstrated that the yearly variance of petal shape was small, and that of petal area was large., Conclusions: Within a flower the major source of the symmetrical elements is genotypic and the asymmetrical elements are strongly affected by the environment. With respect to petal area, the contribution of genotypes is also large; it is, however, affected by the macro-environment more notably than is petal shape.

Current issues in population dynamics are discussed in the context of The Royal Society Discussion Meeting 'Population growth rate: determining factors and role in population regulation'. In particular, different views on the centrality of population growth rates to the study of population dynamics and the role of experiments and theory are explored. Major themes emerging include the role of modern statistical techniques in bringing together experimental and theoretical studies, the importance of long-term experimentation and the need for ecology to have model systems, and the value of population growth rate as a means of understanding and predicting population change. The last point is illustrated by the application of a recently introduced technique, integral projection modelling, to study the population growth rate of a monocarpic perennial plant, its elasticities to different life-history components and the evolution of an evolutionarily stable strategy size at flowering.
