Abstract

We present a summary of two round-table discussions held during two subsequent workshops in Montreal (Canada) on 16 April 2014 and Ostend (Belgium) on 8 July 2015. Five species of the genus Achomosphaera Evitt 1963 and 33 of the genus Spiniferites Mantell 1850 emend. Sarjeant 1970 occuring in Pliocene to modern sediments are listed and briefly described along with remarks made by workshop participants. In addition, several holotypes and topotypes are reillustrated. Three species previously assigned to Spiniferites are here considered/accepted as belonging to other genera: Impagidinium inaequalis (Wall and Dale in Wall et al. 1973) Londeix et al. 2009, Spiniferites? rubinus (Rossignol 1962 ex Rossignol 1964) Sarjeant 1970, and Thalassiphora balcanica Baltes¸ 1971. This summary forms the basis for a set of papers that follows, where points raised during the workshops are explored in greater detail.

10. Morphology, molecular phylogeny and azaspiracid profile of Azadinium poporum (Dinophyceae) from the Gulf of Mexico

Abstract

Azadinium poporum produces a variety of azaspiracids and consists of several ribotypes, but information on its biogeography is limited. A strain of A. poporum (GM29) was incubated from a Gulf of Mexico sediment sample. Strain GM29 was characterized by a plate pattern of po, cp, x, 4′, 3a, 6″, 6C, 5S, 6‴, 2⁗, a distinct ventral pore at the junction of po and the first two apical plates, and a lack of an antapical spine, thus fitting the original description of A. poporum. The genus Azadinium has not been reported in waters of the United States of America before this study. Molecular phylogeny, based on large subunit ribosomal DNA (LSU rDNA) and internal transcribed spacer (ITS) sequences, reveals that strain GM29 is nested within the well-resolved A. poporum complex, but forms a sister clade either to ribotype B (ITS) or ribotype C (LSU). It is, therefore, designated as a new ribotype, termed as ribotype D. LSU and ITS sequences similarity among different ribotypes of A. poporum ranges from 95.4% to 98.2%, and from 97.1% to 99.2% respectively, suggesting that the LSU fragment is a better candidate for molecular discrimination. Azaspiracid profiles were analyzed using LC–MS/MS and demonstrate that strain GM29 produces predominantly AZA-2 with an amount of 45 fg/cell. The results suggest that A. poporum has a wide distribution and highlights the risk potential of azaspiracid intoxication in the United States.

Abstract

In this paper we describe a new species, Archaeperidinium saanichi sp. nov. within the Archaeperidinium minutum Jörgensen 1912 species complex. We examined the morphological variation of the cyst and motile stage by incubation experiments from sediment samples collected in coastal British Columbia (Canada), and compared it to closely related species. The theca of A. saanichi is differentiated from related species by overall size, the asymmetry of the intercalary plates and the right-sulcal plate (S.d.) not touching the cingulum. We provide a key to differentiate all closely related species. A. saanichi can be readily distinguished from A. minutum by a distinctively large cyst with a broad 2a type archeopyle and regularly spaced processes with relatively broad bases and aculeate process tips. Molecular phylogenetic analyses of large and small subunit (LSU and SSU) rDNA sequences demonstrated a close affinity of this species to A. minutum; however, the relatively high level of sequence conservation in dinoflagellate rDNA sequences made these particular markers inadequate for distinguishing one species from the other. Sediment-trap data suggest that A. saanichi has a preference for cooler temperatures and lowered salinities.
