Endogenous inhibitors of angiogenesis, such as thrombospondin-1 (TSP-1), are promising sources of therapeutic agents to treat angiogenesis-driven diseases, including cancer. TSP-1 regulates angiogenesis through different mechanisms, including binding and sequestration of the angiogenic factor fibroblast growth factor-2 (FGF-2), through a site located in the calcium binding type III repeats. We hypothesized that the FGF-2 binding sequence of TSP-1 might serve as a template for the development of inhibitors of angiogenesis. Using a peptide array approach followed by binding assays with synthetic peptides and recombinant proteins, we identified a FGF-2 binding sequence of TSP-1 in the 15-mer sequence DDDDDNDKIPD-DRDN. Molecular dynamics simulations, taking the full flexibility of the ligand and receptor into account, and nuclear magnetic resonance identified the relevant residues and conformational determinants for the peptide-FGF interaction. This information was translated into a pharmacophore model used to screen the NCI2003 small molecule databases, leading to the identification of three small molecules that bound FGF-2 with affinity in the submicromolar range. The lead compounds inhibited FGF-2-induced endothelial cell proliferation in vitro and affected angiogenesis induced by FGF-2 in the chicken chorioallantoic membrane assay. These small molecules, therefore, represent promising leads for the development of antiangiogenic agents. Altogether, this study demonstrates that new biological insights obtained by integrated multidisciplinary approaches can be used to develop small molecule mimics of endogenous proteins as therapeutic agents.

Hematopoietic and endothelial cell lineages share common progenitors. Accordingly, cytokines formerly thought to be specific for the hematopoietic system have been shown to affect several functions in endothelial cells, including angiogenesis. In this study, we investigated the angiogenic potential of erythropoietin (Epo), the main hormone regulating proliferation, differentiation, and survival of erythroid cells. Epo receptors (EpoRs) have been identified in the human EA.hy926 endothelial cell line by Western blot analysis. Also, recombinant human Epo (rHuEpo) stimulates Janus Kinase-2 (JAK-2) phosphorylation, cell proliferation, and matrix metalloproteinase-2 (MMP-2) production in EA.hy926 cells and significantly enhances their differentiation into vascular structures when seeded on Matrigel. In vivo, rHuEpo induces a potent angiogenic response in the chick embryo chorioallantoic membrane (CAM). Accordingly, endothelial cells of the CAM vasculature express EpoRs, as shown by immunostaining with an anti-EpoR antibody. The angiogenic response of CAM blood vessels to rHuEpo was comparable to that elicited by the prototypic angiogenic cytokine basic fibroblast growth factor (FGF2), it occurred in the absence of a significant mononuclear cell infiltrate, and it was not mimicked by endothelin-1 (ET-1) treatment. Taken together, these data demonstrate the ability of Epo to interact directly with endothelial cells and to elicit an angiogenic response in vitro and in vivo and thus act as a bona fide direct angiogenic factor.

The 45th Annual Meeting of the Italian Cancer Society (SIC), held at the Centro Congressi Giovanni XXIII in Bergamo, Italy on 9-12 November, 2003, attracted almost 400 participants. The Scientific Committee chaired by R Giavazzi (Mario Negri Institute, Bergamo) and the board of the Italian Cancer Society produced a packed and varied program. Plenary sessions with keynote speakers (24 lectures) were combined with oral proffered papers (20 selected presentations), providing a platform from laboratory science to clinical interventions. Two hundred and sixty-one abstracts were submitted to the conference and presented in poster and poster discussion sessions, providing a stimulating ground for discussion and a network of interactive collaboration among young investigators. The venue of Bergamo offered exceptional hospitality with social events in the beautiful old town and at the Modern Art Museum. We hope that the following report gives an idea of the event and will encourage the reader to attend the 46th meeting of the SIC, 24-26 October, 2004 in Pisa.

An analytical method using high performance liquid chromatography with fluorescence detection for the simultaneous determination of abamectin and ivermectin in bovine milk was developed and validated. The best recovery results were achieved by using acetonitrile for extraction of the compounds followed by solid phase extraction in cartridges containing C18 for the purification of the extract. Pre-column derivatization was accomplished with N-methylimidazole and trifluoroacetic anhydride. The method limit of detection (LOD) values for abamectin and ivermectin were 0.10 and 0.14 µg L-1 and the limit of quantification (LOQ) values were 0.18 and 0.36 µg L-1, respectively. The recoveries were from 75 to 101%, with RSD values lower than 10%. The LOD and LOQ values are lower than the maximum residue limits (MRLs) in milk established by Codex Alimentarius, European Union and the Brazilian legislation. Um método analítico empregando cromatografia líquida de alta eficiência com detecção por fluorescência para a determinação simultânea de abamectin e ivermectin em leite bovino foi desenvolvido e validado. Os melhores resultados de recuperação foram obtidos empregando acetonitrila na extração dos compostos, com purificação do extrato por extração em fase sólida em cartuchos contendo C18. A derivação pré-coluna foi realizada com N-metilimidazol e anidrido trifluoracético. Os limites de detecção (LOD) do método para abamectin e ivermectin foram 0,10 e 0,14 µg L-1 e os limites de quantificação (LOQ) foram 0,18 e 0,36 µg L-1, respectivamente. As recuperações foram de 75 a 101%, com valores de RSD abaixo de 10%. Os valores de LOD e LOQ obtidos são inferiores aos limites máximos de resíduos em leite estabelecidos pelo Codex Alimentarius, União Européia e legislação brasileira.
