Abstract The present work elicits a novel approach to combating COVID-19 by synthesizing a series of azo-anchored 3,4-dihydroimidazo[4,5-b]indole derivatives. The envisaged methodology involves the l-proline-catalyzed condensation of para-amino-functionalized azo benzene, indoline-2,3-dione, and ammonium acetate precursors with pertinent aryl aldehyde derivatives under ultrasonic conditions. The structures of synthesized compounds were corroborated through FT-IR, 1H NMR, 13C NMR, and mass analysis data. Molecular docking studies assessed the inhibitory potential of these compounds against the main protease (Mpro) of SARS-CoV-2. Remarkably, in silico investigations revealed significant inhibitory action surpassing standard drugs such as Remdesivir, Paxlovid, Molnupiravir, Chloroquine, Hydroxychloroquine (HCQ), and (N3), an irreversible Michael acceptor inhibitor. Furthermore, the highly active compound was also screened for cytotoxicity activity against HEK-293 cells and exhibited minimal toxicity across a range of concentrations, affirming its favorable safety profile and potential suitability. The pharmacokinetic properties (ADME) of the synthesized compounds have also been deliberated. This study paves the way for in vitro and in vivo testing of these scaffolds in the ongoing battle against SARS-CoV-2.

Summary: Glioma cells switch between energetic pathways to adapt and resist therapies. We present a protocol for measuring mitochondrial and glycolytic ATP rates in patient-derived glioma stem-like cells using a Seahorse XF ATP rate assay. We describe steps for growing 3D glioma stem-like cells, attaching cells to the assay plate, preparing drugs, and running the ATP rate assay. We also detail procedures for imaging viable cell numbers and normalization, with tips to overcome pitfalls in Agilent Seahorse assays. : Publisher’s note: Undertaking any experimental protocol requires adherence to local institutional guidelines for laboratory safety and ethics.

Green synthesised nanoparticles (NPs) have garnered significant attention as a rapidly expanding category of materials with diverse applications. In this context, plants are widely acknowledged as a highly favourable option for green synthesis due to their adherence to the eco-friendly pathway of NPs biosynthesis. The objective of green synthesis is to reduce the utilisation of hazardous chemicals. The utilisation of silica (SiO2) NPs, synthesised from biomass as adsorbents, has shown great potential particularly in removing toxic materials from environment. When it comes to adsorbing water pollutants, functionalized SiO2 NPs are seen as potential option. However, certain functional group onto the surface of SiO2 NPs have higher affinity towards a particular pollutant. In this work, a simple and eco-friendly synthesis method (using waste bamboo leaves) have been utilised for production of SiO2 NPs. Further, a silylating procedure was used to functionalize SiO2 NPs with amine (A-SiO2) and carboxyl groups (C–SiO2) for determining adsorption capabilities towards anionic and cationic dyes i.e., Naphthol green-B (NGB) and brilliant green (BG). The adsorption mechanism has been discussed in view of different adsorption isotherm models. According to batch investigations, the functionalized SiO2 NPs showed different behaviour for dyes depending upon nature of dyes and the functional groups. The results of the study revealed that A-SiO2 NPs were having adsorption capacity of 122.10 mg/g for anionic dye (NGB) while C–SiO2 NPs showed adsorption capacity of 114.41 mg/g for cationic dye (BG). Additionally, these NPs exhibit good recyclability up to 5 cycles of reuse. Novelty of this study is the reduced energy needs for the green synthesis of the SiO2 NPs using bamboo leaves and the effect of different functional groups on the adsorption capacity of the NPs. Overall, this study provides a sustainable, and reusable approach for effective dye removal from wastewater, addressing both environmental and economic problems.

Abstract There are fewer studies on Trichoderma diversity in agricultural fields. The rhizosphere of 16 crops was analyzed for Trichoderma species in 7 districts of Rajasthan state of India. Based on DNA sequence of translation elongation factor 1α (tef-1α), and morphological characteristics, 60 isolates were identified as 11 species: Trichoderma brevicompactum, species in Harzianum clade identified as T. afroharzianum, T. inhamatum, T. lentiforme, T. camerunense, T. asperellum, T. asperelloides, T. erinaceum, T. atroviride, T. ghanense, and T. longibrachiatum. T. brevicompactum is the most commonly occurring strain followed by T. afroharzianum. No new species were described in this study. T. lentiforme, showed its first occurrence outside the South American continent. The morphological and cultural characteristics of the major species were observed, described, and illustrated in detail. The isolates were tested for their antagonistic effect against three soilborne plant pathogens fungi: Sclerotium rolfsii, Rhizoctonia solani, and Fusarium verticillioides in plate culture assays. One of the most potent strains was T. afroharzianum BThr29 having a maximum in vitro inhibition of S. rolfsii (76.6%), R. solani (84.8%), and F. verticillioides (85.7%). The potential strain T. afroharzianum BThr29 was also found to be efficient antagonists against soil borne pathogens in in vivo experiment. Such information on crop selectivity, antagonistic properties, and geographic distribution of Trichoderma species will be beneficial for developing efficient Trichoderma-based biocontrol agents.

Abstract Background Lipoarabinomannan (LAM) antigen serves as an attractive biomarker to diagnose Tuberculosis (TB). Given the limitations of current diagnostic modalities for Pleural TB, current study evaluated LAM’s potential to serve as a point-of-care test to diagnose pleural TB. Methods A cross sectional, diagnostic accuracy study was conducted during February to November 2021 in a tertiary care hospital in India. LAM antigen detection was performed on pleural fluid as well as early morning urine specimen of suspected pleural TB patients by “Alere/ Abott Determine TB LAM” lateral flow assay (LAM-LFA). The results were compared to microbiological reference standards/MRS (Mycobacterial culture or NAAT) and Composite reference standards/CRS (MRS plus Clinico-radiological diagnosis). Results A total of 170 subjects were included in the analysis, including 26 with Definite TB, 22 with Probable TB, and 122 with No TB. Compared to MRS and CRS, the sensitivity (61.54% & 45.83%) and positive predictive value (PPV) (57.14 & 78.57%) of Pleural LAM-LFA testing were found to be suboptimal, whereas the specificity (91.67% & 95.08%) and negative predictive value (NPV) (92.96% & 81.69%) of the assay were found to be good. Urinary LAM-LFA performed even worse than pleural LAM-LFA, except for its higher specificity against MRS and CRS (97.2% and 98.3%, respectively). Specificity and PPV of pleural LAM detection increased to 100% when analysed in a subgroup of patients with elevated ADA levels (receiver operating curve analysis-derived cut off value > 40 IU/ml). Conclusion Detection of LAM antigen by LFA directly from pleural fluid was found to be a useful test to predict absence of the disease if the test is negative rather than using as a POCT for diagnosis.

The present work elicits the synthesis of 2,2-dimethyl-2H-[1,3]dioxino[4,5-b]pyrrole-4(7H)-ones derivatives catalyzed by heterogeneous Fe3O4 magnetic nanoparticles supported N-acetyl-PABA@Cu(II), which address the limitations of traditional organic synthesis approaches in terms of sustainable and efficient, resulting in improved yields, higher selectivity with no waste. The decorum embraces four-component reactions (4CRs) of amines, aldehydes, meldrum’s acid, and nitromethane facilitated by Fe3O4-N-acetyl-PABA-Cu(II) in the presence of ethanol (CH3CH2OH) as an ecologically benign solvent under ultrasonic irradiation at room temperature. Outstanding conversion rates and selectivity were attained by the heterogeneous catalyst through great catalytic performance. The simple work-up, easy catalyst recovery, and no base or additional activators were required. Characterization of the structures of synthesized compounds and catalysts was corroborated through different spectroanalytical techniques viz; FTIR, 1H & 13C NMR, Mass elemental analyses, Energy-Dispersive X-ray Spectroscopy (EDX), Field-Emission Scanning Electron Microscope (FE-SEM), Powder X-ray Diffraction (XRD), Thermal Gravimetric Analysis (TGA), Differential Thermal Analysis (DTA), Particle size analysis and Zeta potential.

Abstract Retractions of peer-reviewed biomedical journal articles with Indian authorship have been on the rise for many years. Our study aimed to investigate the reason behind these retractions, namely plagiarism, falsification, fabrication, duplicate publication, author conflicts, ethical issues, fake peer-reviews, and data-related issues, besides providing year-wise trends regarding retraction, authorship, impact factor, and citations. We retrieved retracted publications with Indian affiliations indexed in MEDLINE between 1 January 1990 to 31 December 2021. During this period a total of 619 papers from 372 different journals with median values (interquartile range) pertaining to impact factor [3.2 (1.5, 5.2)], retraction time [24 (10, 51)] months, pre-retraction citations [4 (1, 12)], and post-retraction citations [4 (2, 12)] were retracted. While retractions still account for a small fraction of all publications (0.1%), the overall rate of retractions, that is, the number of retractions relative to the number of newly published journal articles in a given year, has been increasing. The reasons for retractions included plagiarism (27%), falsification and fabrication (26%), duplicate publication (21%), erroneous data (12%), authorship issues (4%), fake-peer reviews (3%), and ethical and funding issues (2%). We have analysed these reasons separately and compared them with each other. Besides a spurt in retraction due to plagiarism, instances of falsification have been escalating over the past decade. Half of the papers retracted on grounds of falsification were published by repeat offender authors in high-impact journals. Furthermore, 82% of retracted papers continued to accumulate citations even after the release of the journal retraction notices. The increase in retractions raises concerns over research quality as well as the wastage of scientific resources, which is especially pressing considering the present environment of scarce funding. The problem of retractions due to reasons such as plagiarism, duplicate publication, authorship issues, and, ethical issues as well as post-retraction citations can be mitigated by educating and raising awareness on publication ethics and responsible research conduct of researchers and journal publishers. Retractions due to fabrication, falsification, and fake peer reviews are more challenging to difficult to address and require further research for the identification of effective solutions.

Present work elicits synthesis of novel azepane-acridines, assisted by titania-encapsulated γFe2O3 nanoparticles supported on Vitamin B1 (γFe2O3@TiO2-Vitamin B1) as a new magnetically recyclable heterogeneous nano-catalytic system. The reaction was accomplished using biocompatible PEG-200 (Polyethylene glycol) as the solvent under ultrasonic irradiations. The structure and morphology of synthesized magnetic nano-catalyst was ascertained based on data obtained from powder X-ray diffraction (PXRD), field emission scanning electron microscopy (FE-SEM), energy-dispersive X-ray spectroscopy (EDAX), Fourier transform infrared (FT-IR), and VSM (Vibrating-sample magnetometer) TGA, DTA and DTG techniques. The characteristic size of the prepared heterogenous nano-catalyst is 34.62 nm. Also, the structures of all the synthesized azepane-acridine heterocyclic scaffolds were corroborated through spectro-analytical data. These new compounds were found to have remarkable biological activity profile against the bacterium S.aureus with IC50 values ranging from 1.41 to 2.75 µg/mL. Further, these in-vitro studies were correlated with statistically significant 2D/3D- QSAR models. The best 2D-QSAR models reveal the positive influence of slogp and Vander Waal Surface area descriptors while the negative influence of the polar surface area and T_2_N_6 descriptors on the bioactivity with ideal values of squared correlation coefficient (r2), cross-validated correlation coefficient (q2) with high values of Fisher ratio (F-test) indicating the model to be statistically relevant. The finest 3D-QSAR models MLR (Multiple Linear Regression method) provide practical information in the characterization and differentiation of compound binding sites. Moreover, molecular docking studies were performed to evaluate the antifungal activity of synthesized scaffolds against the black fungi crystal protein, indicating remarkable bioassay scores. Atom economy, practical feasibility, cost-effectiveness, minimum reaction time, High yields, and an additive-free process are the foremost advantages of the developed methodology.

SARS-CoV, an RNA virus, is contagious and displays a remarkable degree of adaptability, resulting in intricate disease presentations marked by frequent genetic mutations that can ultimately give rise to drug resistance. Targeting its viral replication cycle could be a potential therapeutic option to counter its viral growth in the human body leading to the severe infectious stage. The Mpro of SARS-CoV-2 is a promising target for therapeutic development as it is crucial for viral transcription and replication. The derivatives of β-diketone and coumarin have already been reported for their antiviral potential and, thus, are considered as a potential scaffold in the current study for the computational design of potential analogs for targeting the viral replication of SARS-CoV-2. In our study, we used novel diketone-hinged coumarin derivatives against the SARS-CoV-2 MPro to develop a broad-spectrum antiviral agent targeting SARS-CoV-2. Through an analysis of pharmacokinetics and docking studies, we identified a list of the top 10 compounds that demonstrated effectiveness in inhibiting the SARS-CoV-2 MPro virus. On the basis of the pharmacokinetics and docking analyses, the top 5 novel coumarin analogs were synthesized and characterized. The thermodynamic stability of compounds KS82 and KS94 was confirmed by their molecular dynamics, and the stability of the simulated system indicated their inhibitory nature. Molecules KS82 and KS94 were further evaluated for their anti-viral potential using Vero E6 cells followed by RT-PCR assay against SARS-CoV-2. The test compound KS82 was the most active with the potential to inhibit SARS-CoV-2 replication in Vero E6 cells. These data indicate that KS82 prevents the attack of the virus and emerges as the primary candidate with promising antiviral properties.

Background: Green tea (Camellia sinensis) mouth rinse is found effective in reducing periodontitis. However, studies evaluating the effectiveness of green tea extracts in reducing oral halitosis and tongue coating on Indian population were scanty. Objective: The objective of this study was to evaluate the effectiveness of green tea-based mouth rinse in comparison with 0.2% chlorhexidine gluconate mouth rinse in reducing dental plaque, tongue coating, and halitosis among human volunteers. Materials and Methods: This was a parallel-arm double-blind randomized controlled trial conducted in two residential hostels in Mysuru city over 21 days. 90 adult participants were recruited and randomized into three groups: Group A: mouth rinse containing saline, Group B: 5% C. sinensis mouth rinse, and Group C: 0.2% chlorhexidine diluted to with equal quantity of water. Preintervention prophylaxis was done; tongue coating and oral halitosis scores were recorded and compared between the groups at baseline and after 21 days. Results: The mean plaque buildup at postintervention was highest in Group 1 (2.45 ± 0.38) followed by Group 3 (1.18 ± 0.12) and Group 2 (1.08 ± 0.11) in the descending order. The mean oral halitosis score was highest in Group 1 (3.00 ± 0.79) followed by Group 3 (1.53 ± 0.50) and Group 2 (1.50 ± 0.50) in the descending order. The mean tongue coating score was highest in Group 1 (1.17 ± 0.47) followed by Group 2 (0.75 ± 0.36) and Group 3 (0.69 ± 0.34) in the descending order. Conclusion: Five percent C. sinensis mouth rinse is as effective as commercially available 0.2% chlorhexidine mouthwash in reducing plaque deposition, tongue coating, and oral halitosis.

The present paper elicits the zinc oxide nanoparticles-assisted synthesis of a new series of seven-membered 1,4-diazepine heterocyclic compounds as potent lead scaffolds. Structures of synthesized compounds were corroborated using spectroanalytical techniques viz, FT-IR, 1H, 13C NMR, Mass, and elemental analysis. Also, Field emission scanning electron microscopy (FE-SEM), Transmission electron microscopy (TEM), Energy dispersive x-ray analysis (EDAX), powder X-ray diffraction (PXRD), and Fourier transform infrared spectroscopy (FT-IR) were used to establish the structure and morphology of the synthesized nanocatalyst. The clean workup procedure, high to excellent yields, relatively short reaction times, and high atom economy are the incredible advantages associated with the protocol.

Abstract The emergence of multidrug resistance coupled with shrinking antibiotic pipelines has increased the demand of antimicrobials with novel mechanisms of action. Therefore, researchers across the globe are striving to develop new antimicrobial substances to alleviate the pressure on conventional antibiotic therapies. Host-Defence Peptides (HDPs) and their derivatives are emerging as effective therapeutic agents against microbial resistance. In this study, five analogs (DP1-5) of the N-terminal (N-15) fragment of CATH-2 were designed based on the delicate balance between various physicochemical properties such as charge, aliphatic character, amphipathicity and hydrophobicity. By means of in-silico and in-vitro studies a novel peptide (DP1) with the sequence “RFGRFLRKILRFLKK” was found to be more effective and less toxic than the N-terminal CATH-2 peptide. Circular dichroism spectroscopy and differential scanning calorimetry were applied for structural insights. Antimicrobial, haemolytic, and cytotoxic activities were also assessed. The resulting peptide was characterized by low cytotoxicity, low haemolytic activity, and efficient anti-microbial activity. Structurally, it displayed strong helical properties irrespective of the solvent environment and was stable in membrane-mimicking environments. Taken together, the data suggests that DP1 can be explored as a promising therapeutic agent with possible clinical applications.

Alzheimer’s disease (AD) is caused by plaque agglomeration and entanglement in several areas of the neural cells, which leads to apoptosis. The main etiology of AD is senile dementia, which is linked to amyloid-beta (Aβ) deregulation and tau perivascular pathogeny. Hyperphosphorylated tau has a propensity for microtubules, which elevate the instability and tau-protein congregates, leading to accumulation of neurofibrillary tangles (NFTs). Tau hyperphosphorylation is susceptible to GSK-3, which has led to an emerging hypothesis regarding the pathogenesis of AD. Accordingly, attempts have been made to conduct investigations and achieve further advancements on new analogues capable of inhibiting the GSK-3 protein, which are currently in the clinical trials. In this analysis, we have evaluated certain GSK-3 inhibitor variants utilising scaffolding and framework devised techniques with pharmacological characteristics, accompanied by computational screenings (pharmacokinetics and docking). The structure-based designed analogues interacted effectively with the active amino acids of GSK-3β target protein. The in silico pharmacokinetic studies revealed their drug-like properties. The analogues with best interactions and binding scores will be considered in the future to completely demonstrate their potential relevance as viable GSK-3 inhibitors.

No drug on the market, as a single entity, participates in different pathways involved in the pathology of Alzheimer’s disease. The current study is aimed at the exploration of multifunctional chalcone derivatives which can act on multiple targets involved in Alzheimer’s disease. A series of novel aminoethyl-substituted chalcones have been developed using in silico approaches (scaffold morphing, molecular docking, and ADME) and reported synthetic methods. The synthesized analogs were characterized and evaluated biologically using different in vitro assays against AChE, AGEs, and radical formation. Among all compounds, compound PS-10 was found to have potent AChE inhibitory activity (IC50 = 15.3 nM), even more than the standard drug (IC50 = 15.68 nM). Further, the in vivo evaluation of PS-10 against STZ-induced dementia in rats showed memory improvement (Morris Water Maze test) in rats. Also, PS-10 inhibited STZ-induced brain AChE activity and oxidative stress, further strengthening the observed in vitro effects. Further, the molecular dynamic simulation studies displayed the stability of the PS-10 and AChE complex. The novel aminoethyl-substituted chalcones might be considered potential multifunctional anti-Alzheimer’s molecules.

Alzheimer’s disease (AD) is the prime cause of 65–80% of dementia cases and is caused by plaque and tangle deposition in the brain neurons leading to brain cell degeneration. β-secretase (BACE-1) is a key enzyme responsible for depositing extracellular plaques made of β-amyloid protein. Therefore, efforts are being applied to develop novel BACE-1 enzyme inhibitors to halt plaque build-up. In our study, we analyzed some Elenbecestat analogues (a BACE-1 inhibitor currently in clinical trials) using a structure-based drug design and scaffold morphing approach to achieve a superior therapeutic profile, followed by in silico studies, including molecular docking and pharmacokinetics methodologies. Among all the designed compounds, SB306 and SB12 showed good interactions with the catalytic dyad motifs (Asp228 and Asp32) of the BACE-1 enzyme with drug-likeliness properties and a high degree of thermodynamic stability confirmed by the molecular dynamic and stability of the simulated system indicating the inhibitory nature of the SB306 and SB12 on BACE 1.

A 57-year-old farmer presented with chronic cough and recurrent hemoptysis, previously treated for sputum positive pulmonary tuberculosis. Referred to us for evaluation of drug resistant tuberculosis as his sputum was persistently positive for acid fast bacilli along with radiological worsening even after 6 months of antitubercular treatment. Bronchoalveolar lavage was done and he was diagnosed with a rare mixed non-tuberculous mycobacyteria (NTM) pulmonary infection despite no immune dysfunction. He was successfully treated with multidrug regimen of rifampicin, isoniazid, ethambutol and clarithromycin.

Nitrogen-containing scaffolds are ubiquitous in nature and constitute an important class of building blocks in organic synthesis. The asymmetric aza-Michael reaction (aza-MR) alone or in tandem with other organic reaction(s) is an important synthetic tool to form new C–N bond(s) leading to developing new libraries of diverse types of bioactive nitrogen compounds. The synthesis and application of a variety of organocatalysts for accomplishing highly useful organic syntheses without causing environmental pollution in compliance with ‘Green Chemistry” has been a landmark development in the recent past. Application of many of these organocatalysts has been extended to asymmetric aza-MR during the last two decades. The present article overviews the literature published during the last 10 years concerning the asymmetric aza-MR of amines and amides catalysed by organocatalysts. Both types of the organocatalysts, i.e., those acting through non-covalent interactions and those working through covalent bond formation have been applied for the asymmetric aza-MR. Thus, the review includes the examples wherein cinchona alkaloids, squaramides, chiral amines, phase-transfer catalysts and chiral bifunctional thioureas have been used, which activate the substrates through hydrogen bond formation. Most of these reactions are accompanied by high yields and enantiomeric excesses. On the other hand, N-heterocyclic carbenes and chiral pyrrolidine derivatives acting through covalent bond formation such as the iminium ions with the substrates have also been included. Wherever possible, a comparison has been made between the efficacies of various organocatalysts in asymmetric aza-MR.

In the treatment of influenza, Neuraminidase inhibitors (NAIs) (Oseltamivir and Zanamivir) play a major role. The emergence of variants of influenza A (H1N1) pdm09 virus resistant to Oseltamivir is a matter of great concern as it limits its usage. Therefore, vigilant monitoring for Oseltamivir-resistant viruses has been recommended by the World Health Organization (WHO). Our study aimed to screen the influenza A (H1N1) pdm09 virus for NAI drug resistance during the outbreak of 2015-16 in North-Western India. A total of 640 H1N1pdm09 virus-positive samples were screened for drug resistance to Oseltamivir by WHO allelic discrimination real-time RT-PCR protocol. The allelic discrimination PCR protocol can detect the presence of single nucleotide polymorphisms (SNPs), the H275Y mutation is detected by this method which causes resistance to Oseltamivir. Sanger sequencing of partial fragment of NA gene (fragment IV), of 90 samples were performed to confirm the presence of NA-H275Y mutation. Neuraminidase susceptibility of 20 randomly selected isolates to Oseltamivir was tested using NA inhibition chemiluminiscence based assay. Among 640 H1N1pdm09 positive samples tested, H275Y mutation was detected in one sample (0.15%) by PCR and confirmed by Sanger sequencing also. All the 20 isolates tested for NAI susceptibility by NA star assay were found to be sensitive to Oseltamivir. WHO allelic discrimination PCR is an easy, rapid and sensitive method for high-throughput detection of resistance to Oseltamivir. Systematic regular drug resistance surveillance of Influenza A is essential to monitor the emergence and spread of drug-resistant strains.

BackgroundOmicron, a new variant of Severe Acute Respiratory Syndrome-Coronavirus 2 (SARS-CoV-2), was first detected in November 2021. This was believed to be highly transmissible and was reported to evade immunity. As a result, an urgent need was felt to screen all positive samples so as to rapidly identify Omicron cases and isolate them to prevent the spread of infection. Genomic surveillance of SARS-CoV-2 was planned to correlate disease severity with the genomic profile.MethodsAll the SARS-CoV-2 positive cases detected in the state of Rajasthan were sent to our Lab. Samples received from 24 November 2021 to 4 January 2022 were selected for Next-Generation Sequencing (NGS). Processing was done as per protocol on the Ion Torrent S5 System for 1,210 samples and bioinformatics analysis was done.ResultsAmong the 1,210 samples tested, 762 (62.9%) were Delta/Delta-like and other lineages, 291 (24%) were Omicron, and 157 (12.9%) were invalid or repeat samples. Within a month, the proportion of Delta and other variants was reversed, 6% Omicron became 81%, and Delta and other variants became 19%, initially all Omicron cases were seen in international travelers and their contacts but soon community transmission was seen. The majority of patients with Omicron were asymptomatic (56.7%) or had mild disease (33%), 9.2% had moderate symptoms, and two (0.7%) had severe disease requiring hospitalization, of which one (0.3%) died and the rest were (99.7%) recovered. History of vaccination was seen in 81.1%, of the previous infection in 43.2% of cases. Among the Omicron cases, BA.1 (62.8%) was the predominant lineage followed by BA.2 (23.7%) and B.1.529 (13.4%), rising trends were seen initially for BA.1 and later for BA.2 also. Although 8.9% of patients with Delta lineage during that period were hospitalized, 7.2% required oxygen, and 0.9% died. To conclude, the community spread of Omicron occurred in a short time and became the predominant circulating variant; BA.1 was the predominant lineage detected. Most of the cases with Omicron were asymptomatic or had mild disease, and the mortality rate was very low as compared to Delta and other lineages.

The pandemic caused by SARS-CoV-2 (SCoV-2) has impacted the world in many ways and the virus continues to evolve and produce novel variants with the ability to cause frequent global outbreaks. Although the advent of the vaccines abated the global burden, they were not effective against all the variants of SCoV-2. This trend warrants shifting the focus on the development of small molecules targeting the crucial proteins of the viral replication machinery as effective therapeutic solutions. The PLpro is a crucial enzyme having multiple roles during the viral life cycle and is a well-established drug target. In this study, we identified 12 potential inhibitors of PLpro through virtual screening of the FDA-approved drug library. Docking and molecular dynamics simulation studies suggested that these molecules bind to the PLpro through multiple interactions. Further, IC50 values obtained from enzyme-inhibition assays affirm the stronger affinities of the identified molecules for the PLpro. Also, we demonstrated high structural conservation in the catalytic site of PLpro between SCoV-2 and Human Coronavirus 229E (HCoV-229E) through molecular modelling studies. Based on these similarities in PLpro structures and the resemblance in various signalling pathways for the two viruses, we propose that HCoV-229E is a suitable surrogate for SCoV-2 in drug-discovery studies. Validating our hypothesis, Mefloquine, which was effective against HCoV-229E, was found to be effective against SCoV-2 as well in cell-based assays. Overall, the present study demonstrated Mefloquine as a potential inhibitor of SCoV-2 PLpro and its antiviral activity against SCoV-2. Corroborating our findings, based on the in vitro virus inhibition assays, a recent study reported a prophylactic role for Mefloquine against SCoV-2. Accordingly, Mefloquine may further be investigated for its potential as a drug candidate for the treatment of COVID.

Background: First Zika virus (ZIKV) positive case from North India was detected on routine surveillance of Dengue-Like Illness in an 85-year old female. Objective of the study was to conduct an investigation for epidemiological, clinical and genomic analysis of first ZIKV outbreak in Rajasthan, North India and enhance routine ZIKV surveillance. Method: Outbreak investigation was performed in 3 Km radius of the index case among patient contacts, febrile cases, and pregnant women. Routine surveillance was enhanced to include samples from various districts of Rajasthan. Presence of ZIKV in serum and urine samples was detected by real time PCR test and CDC trioplex kit. Few ZIKV positive samples were sequenced using the next-generation sequencing method for genomic analysis. Result: On outbreak investigation 153/2043 (7.48%) cases were found positive: 1/153 (0.65%) among contacts, 90/153 (58.8%) in fever cases, 62/153(40.5%) in pregnant females. In routine surveillance, 6/4722 (0.12%) serum samples were ZIKV positive.Majority of patients had mild signs and symptoms, no case of microcephaly and Guillain- Barre Syndrome was seen, 25 (40.3%) pregnant females delivered healthy babies, four (6.4%) reported abortion and three (4.8%) had intrauterine death, one (1.6%) child had colorectal malformation and died after few days of birth. ZIKV was found to belong to Asian lineage, mutation related to enhanced neuro-virulence and transmission in animal models was not found. Conclusion: ZIKV was endogenous to India belonging to Asian Lineage. Disease profile of the ZIKV was asymptomatic to mild. No major anomaly was observed in infants born to ZIKV positive mothers; however, long term follow up of these children is required. There is need to scale up surveillance in the virology lab network of India for early detection and control. Summary line: Zika virus infection was endogenous due to Asian Lineage with mild disease, no case of microcephaly or Guillain- Barre Syndrome was seen but children need to be followed for anomalies and surveillance of ZIKV needs to be enhanced in the country.

Patients with chronic kidney disease stage 5 are usually excluded from renal transplantation if they do not have a compatible blood group donor. Paired kidney donation and ABO-incompatible kidney transplantation are the only options for such patients. We describe the case of a 49-year-old male patient who had blood Group B positive and potential donor; his wife had blood Group nagative. The patients had an initial anti-A antibody titer of 1:32 and underwent antibody depletion with plasmapheresis (two sessions). He also received rituximab 200 mg on minus 14th days and was induced with basiliximab. At the time of transplant, his anti-A titer was 1:8. Postoperatively, the patient had delayed graft function and received one session of hemodialysis but no plasmapheresis. Postoperatively, anti-A titer was 1:8. He was discharged on the 14th postoperative day with normal renal function.

Mycetoma is disorder of subcutaneous tissue, skin, and bones, mainly the feet. Etiologically divided in Eumycetoma and actinomycetoma, since the treatment of both is different, the diagnosis is mandatory. This is the case of 35-year-old lady with swelling in left foot with multiple discharging sinuses for 5 years that was none responding to antifungal treatment. Change of treatment after the culture confirmation of Actinomadura species improves patient condition drastically.

The addition of 4 wt% Zr to Ti52V12Cr36 alloy was carried out in two different ways: arc-melting or ball-milling. The cast alloy showed rapid hydrogen absorption up to 3.6 wt% of hydrogen capacity within 15 min. Ball milling this sample worsened the kinetics, and no hydrogen absorption was registered when milling was carried out for 30 or 60 min. When zirconium is added by ball-milling, the kinetic is slower than that when addition is by arc-melting. This is due to the fact that when added by milling, zirconium does not form a ternary phase with Ti, V, and Cr but instead is just dispersed on the particles’ surface.

BACKGROUND: India lacks organized school oral health programs, resulting in limited access to oral health care among children. The peer role models, or teachers, may help in bridging the gap to enhance knowledge on self-care preventive practices. The aim of the study was to evaluate and compare the effectiveness of dental health education (DHE) offered by qualified dental professional, trained teachers, and peer role models in promoting oral hygiene status and behavior among school-going children in Mysuru, Karnataka. MATERIALS AND METHODS: This was an interventional study conducted over a period of 3 months in an academic year in three selected schools in Mysuru City, India. A total of 120 students were divided into three groups – group 1 were given DHE (dental health education) by a dental professional, group 2 were given DHE by a trained teacher, and group 3 were given DHE by peer role models. Oral health knowledge was assessed using a close-ended questionnaire, plaque levels were assessed using Turesky Gilmore Glickman modification of Quigley Hein plaque index, and gingival status was assessed using Loe and Sillness gingival index. After 3 months, the same index and questionnaire were used post intervention. RESULTS: The mean scores for knowledge on dental caries at baseline in groups 1, 2, and 3 were 3.75 ± 1.25, 3.65 ± 1.07, and 3.40 ± 1.17, respectively, with no significant difference between the groups, which changed to 4.43 ± 1.27, 3.37 ± 1.14, and 4.93 ± 0.99, respectively, following intervention. Similar results were observed with regard to knowledge on gingival and periodontal diseases. The mean plaque scores at baseline for groups 1, 2, and 3 were 4.17 ± 0.30, 3.24 ± 0.70, and 4.10 ± 0.31, respectively, which changed to 3.85 ± 0.32, 3.90 ± 0.39, and 3.69 ± 0.34, respectively, in three groups following intervention. Post intervention, plaque scores and gingival scores significantly improved in groups 1 and 3 but worsened in group 2. Overall, knowledge scores improved in groups 1 and 3 for some questions, but improvement was not noted in some questions. CONCLUSION: Under the limitations of the study, it was found that peer role models were as effective as dental professionals in providing DHE in schools.

In this paper, we report the effect of cooling rate on the microstructure and hydrogenation behaviour of TiFe alloy with 4 wt% Zr as an additive. An ingot of TiFe +4 wt% Zr was synthesized by induction melting, using industrial grade Fe and Ti. Step mold with a thickness of 25, 13, 6 and 3 mm were used to obtain different cooling rates. It was found that higher cooling rate leads to a rapid solidification and finer distribution of the secondary phase. There was no effect of cooling rate on the chemical composition of the different phases present in all thicknesses. However, faster cooling rate leads to faster first hydrogenation kinetics. The reason is that the scale of the secondary phase decreases with increasing cooling rates. The decrease in the scale of the secondary phase was evidenced by the measure of its perimeter. During activation, the hydrogenation rate limiting step for all alloys was found to be 3D growth, diffusion controlled with decreasing interface velocity. Wound healing is an intricate process of tissue repair or remodeling that occurs in response to injury. Plants and plant-derived bioactive constituents are well explored in the treatment of various types of wounds. Curcumin is a natural polyphenolic substance that has been used since ancient times in Ayurveda for its healing properties, as it reduces inflammation and acts on several healing stages. Several research studies for curcumin delivery at the wound site reported the effectiveness of curcumin in eradicating reactive oxygen species and its ability to enhance the deposition of collagen, granulation tissue formation, and finally, expedite wound contraction. Curcumin has been widely investigated for its wound healing potential but its lower solubility and rapid metabolism, in addition to its shorter plasma half-life, have limited its applications in wound healing. As nanotechnology has proven to be an effective technique to accelerate wound healing by stimulating appropriate mobility through various healing phases, curcumin-loaded nanocarriers are used for targeted delivery at the wound sites. This review highlights the potential of curcumin and its nanoformulations, such as liposomes, nanoparticles, and nano-emulsions, etc. in wound healing. This paper emphasizes the numerous biomedical applications of curcumin which collectively prepare a base for its antibiofilm and wound-healing action.

Language plays a vital role in the communication of ideas, thoughts, and information to others. Hearing-impaired people also understand our thoughts using a language known as sign language. Every country has a different sign language which is based on their native language. In our research paper, our major focus is on Indian Sign Language, which is mostly used by hearing- and speaking-impaired communities in India. While communicating our thoughts and views with others, one of the most essential factors is listening. What if the other party is not able to hear or grasp what you are talking about? This situation is faced by nearly every hearing-impaired person in our society. This led to the idea of introducing an audio to Indian Sign Language translation system which can erase this gap in communication between hearing-impaired people and society. The system accepts audio and text as input and matches it with the videos present in the database created by the authors. If matched, it shows corresponding sign movements based on the grammar rules of Indian Sign Language as output; if not, it then goes through the processes of tokenization and lemmatization. The heart of the system is natural language processing which equips the system with tokenization, parsing, lemmatization, and part-of-speech tagging.

A comprehensive account of recent advances in the synthesis of imidazopyridines, assisted through transition-metal-catalyzed multicomponent reactions, C–H activation/functionalization and coupling reactions are highlighted in this review article. The basic illustration of this review comprises of schemes with concise account of explanatory text. The schemes depict the reaction conditions along with a quick look into the mechanism involved to render a deep understanding of the catalytic role. At some instances optimizations of certain features have been illustrated through tables, i.e., selectivity of catalyst, loading of the catalyst and percentage yield with different substrates. Each of the reported examples has been rigorously analyzed for reacting substrates, reaction conditions and transition metals used as the catalyst. This review will be helpful to the chemists in understanding the challenges associated with the reported methods as well as the future possibilities, both in the choice of substrates and catalysts. This review would be quite appealing to a wider range of organic chemists in academia and industrial R&D sectors working in the field of heterocyclic syntheses. In a nutshell, this review will be a guiding torch to envisage: (i) the role of various transition metals in the domain dedicated towards method development and (ii) for the modifications needed thereof in the R&D sector.

Keratitis is a very serious clinical condition caused by bacteria, fungi or parasites. It is associated with serious complications like blindness or endophthalmitis if remained undiagnosed. A 42-year-old male patient with the history of foreign body inoculation presented with mild pain and redness. After clinical examination, fungal keratitis was diagnosed. Corneal scrapping was sent for microbiological investigation. A very rare fungi Colletotrichum dematium was isolated and identified. Patient was managed with Natamycin tab fluconazole and atropin eye drops. To know the geographical distribution of the rare fungi, more and more cases should be studied and reported, which will help in management of rare fungi and standardisation of therapy

Wet ball milling was used for the synthesis of Cu2ZnSnSe4 (CZTSe) nanoparticles with a kesterite structure. The prepared nanoparticles were used for ink formulation. Surfactants and binders were added to improve the ink stability, prevent agglomeration, and enhance ink adhesion. The films deposited via spin coating were annealed at different temperatures using a rapid thermal processing system in the presence of selenium powder in an inert environment. Analytical techniques, such as X-ray diffraction, Raman spectroscopy, and Fourier-transform infrared spectroscopy, were used to confirm the formation of CZTSe nanoparticles with a single-phase, crystalline kesterite structure. Field-emission scanning electron microscopy and energy-dispersive X-ray spectroscopy were used to study the surface morphology and chemical composition of the thin films before and after annealing, with and without the sodium solution. The optoelectrical properties were investigated using ultraviolet-visible spectroscopy and Hall measurements. All the prepared CZTSe thin films exhibited a p-type nature with an optical bandgap in the range of 0.82–1.02 eV. The open-circuit voltage and fill factor of the CZTSe-based devices increased from 266 to 335 mV and from 37.79% to 44.19%, respectively, indicating a decrease in the number of recombination centers after Na incorporation.

In the present study, we adopt an easy and cost-effective route for preparing Cu2ZnSnS4 (CZTS)-absorber nanoparticles by a mechanochemical method using non-toxic and environmentally benign solvents (butanol, methyl ethyl ketone, and ethanol). The gram-scale synthesis of absorber nanoparticles was achieved in a non-hazardous, zero-waste process without using high-vacuum equipment. The effects of annealing and Na incorporation on the properties of spin-coated CZTS thin films were scrutinized. The deposited samples showed kesterite crystal structure and single phase. The morphological results revealed an improvement in the surface morphology after annealing. The optical bandgaps of the thin films lied in the range of 1.50–1.57 eV with p-type nature. Finally, photovoltaic devices were fabricated, and their cell performance parameters were studied. An efficiency of 0.16% was observed. The present study provides a potential route for the cost-effective fabrication of CZTS-based photovoltaic devices.

Fusarium wilt caused by Fusarium oxysporum f. sp. lentis (Fol) has been considered as destructive seed and soil-borne disease of lentil which causes severe yield loss every year. For its biological control, twenty isolates of Trichoderma spp. were tested against thirty Fol isolates using dual culture technique. Out of twenty Trichoderma isolates, seven isolates (5593, ThL-1, ThL-4, TaL- 2, TaL- 4, ThaL-3 and TvL-6) showed maximum percent inhibition of radial growth against pathogens between 82.4% to 78.0% as compared to the rest of Trichoderma isolates. As far as molecular identification is concerned the effective six isolates after dual culture assay and highly virulent Fol isolate were molecularly identified by using ITS primers (ITS 1 and ITS 4) and remaining one isolate was taken from ITCC which was already identified as T. harzianum (5593). Seven efficient isolates of Trichoderma were used to evaluate volatile and non- volatile assay against Fol (NFLRJ-3). Isolates, 5593 and ThL-4 showed significant growth inhibition against Fol at 48 hours of incubation in both the assays as compared to the rest Trichoderma isolates. In volatile organic compound (VOC) assay ThL-4 showed highest number of seed germination and showed good seed health condition. Out of seven selected isolates of Trichoderma, all isolates were found effective to control disease and stimulated plant growth promotion activity of lentil (2017-2019). Therefore, these isolates can be utilized to formulate bio-fungicides which is the need of the hour as far as disease management is concerned.

Abdominal tuberculosis (ATB) continues to pose a major diagnostic challenge for clinicians due to its nonspecific clinical presentation, variable anatomical location and lack of sensitive diagnostic tools. In spite of the development of several assays till date; no single test has proved to be adequate for ATB diagnosis. In this study, we for the first time report the detection of circulating cell-free Mycobacterium tuberculosis (M. tuberculosis) DNA (cfMTB-DNA) in ascitic fluid (AF) samples and its utility in ATB diagnosis. Sixty-five AF samples were included in the study and processed for liquid culture, cytological, biochemical and molecular assays. A composite reference standard (CRS) was formulated to categorize the patients into 'Definite ATB' (M. tuberculosis culture positive, n = 2), 'Probable ATB' (n = 16), 'Possible ATB' (n = 13) and 'Non-TB' category (n = 34). Two molecular assays were performed, namely, the novel cfMTB-DNA qPCR assay targeting M. tuberculosis devR gene and Xpert MTB/RIF assay (Xpert), and their diagnostic accuracy was assessed using CRS as reference standard. Hydrogels, an advanced interactive system, is finding use as wound dressings, however, they exhibit restricted mechanical properties, macroscopic nature, and may not manage high exudate wounds or incorporate lipophilic actives. In this study, we developed a self-gelling solid lipid nanoparticle (SLNs) dressing to incorporate simvastatin (SIM), a lipophilic, potential wound-healing agent, clinically limited due to poor solubility (0.03 mg/mL) and absorption. The study explores unconventional and novel application of SIM. The idea was to incorporate a significant amount of SIM in a soluble form and release it slowly over a prolonged time. Further, a suitable polymeric surfactant was selected that assigned a self-gelling property to SLNs (SLN-hydrogel) so as to be used as a novel wound dressing. SLNs assign porosity, elasticity, and occlusivity to the dressing to keep the wound area moist. It will also provide better tolerance and sensory properties to the hydrogel. SIM loaded SLN-hydrogel was prepared employing an industry amenable high-pressure homogenization technique. The unique hydrogel dressing was characterized for particle size, zeta potential, Fourier transform infra-red spectroscopy, powder X-ray diffraction, differential scanning calorimetry, rheology, and texture. Significant loading of SIM (10% w/w) was achieved in spherical nanoparticule hydrogel (0.3 nm (nanoparticles) to 2 µm (gelled-matrix)) that exhibited good spreadability and mechanical properties and slow release up to 72 h. SLN-hydrogel was safe as per the organization for economic co-operation and development (OECD-404) guidelines, with no signs of irritation. Complete healing of excision wound observed in rats within 11 days was 10 times better than marketed povidone-iodine product. The presented work is novel both in terms of classifying a per se SLN-hydrogel and employing SIM. Further, it was established to be a safe, effective, and industry amenable invention.

Rattan palms are one of the important nontimber forest produce which forms an integral part of the tribal population and contribute significantly in the rural economy. The demand of the rattan resources is substantial but most of the raw material is being collected from natural stands; and therefore, there is tremendous pressure on natural populations. Calamus guruba is one of the important rattan species of India and extensively harvested for a wide range of end products. In the present study, three populations of C. guruba naturally growing in Mizoram were characterized for their genetic diversity and population structure using Inter-Simple Sequence Repeat (ISSR) markers. A total of 197 bands generated in PCR amplification of 67 individuals with 10 ISSR primers. Among all the primers UBC 808 displayed maximum 25, while UBC 807 and UBC 827 showed the lowest 13 numbers of bands. Relatively high genetic diversity (PPB = 98.98 %, h = 0.271, I = 0.341) was recorded for the species with a moderate level of genetic differentiation (PhiPT = 0.282). Analysis of molecular variance revealed that the genetic differentiation among the populations is the significant and large proportion of the genetic variation (72%) resides among the individuals within a population, whereas only 28 % were found among the populations. UPGMA tree, PCoA plot and STRUCTURE analysis revealed that two ancestral groups captured the entire genetic variability. The baseline study on C. guruba is of paramount importance in devising programs for conservation and improvement.

Fungi are reported to cause a range of superficial to invasive human infections. These often result in high morbidity and at times mortality. Conventional antifungal agents though effective invariably exhibit drug interactions, treatment-related toxicity, and fail to elicit significant effect, thus indicating a need to look for suitable alternatives. Fungi thrive in humid, nutrient-enriched areas. Such an environment is well-supported by the oral cavity. Despite this, there is a relatively low incidence of severe oral and periodontal fungal infections, attributed to the presence of antimicrobial peptides hosted by saliva, viz. histatin 5 (Hstn 5). It displays fungicidal activity against a variety of fungi including Candida albicans, Candida glabrata, Candida krusei, Cryptococcus neoformans, and unicellular yeast-like Saccharomyces cerevisiae. Candida albicans alone accounts for about 70% of all global fungal infections including periodontal disease. This review intends to discuss the scope of Hstn 5 as a novel recourse for the control of fungal infections.

India is at risk of Zika virus transmission due to high prevalence of its vector Aedes aegypti. Rajasthan, a state in the north-west region of India, has also high prevalence of Aedes mosquito. First laboratory confirmed case of Zika virus disease in Rajasthan was reported on 21 September 2018 in Jaipur. The Government of Rajasthan quickly implemented a containment strategy to contain the outbreak and prevent further spread of this disease. Strategy included active human and mosquito surveillance, laboratory testing and sequencing of the virus, integrated vector control measures, intersectoral coordination, risk communication and social mobilisation, all in a predefined geographic area around the epicentre. Timely action with appropriate coordination at all levels with multiple stakeholders contained the outbreak successfully. In all, 159 confirmed cases were reported from in and around the 3 km containment zone in Shastri Nagar area of Jaipur City and routine surveillance. Following this, a specially developed laboratory-based surveillance strategy was put in place to ensure that the disease does not spread beyond the containment zone. No fresh case was reported subsequently within or beyond the containment zone.

Vancomycin causes different types of hypersensitivity reactions, ranging from localized skin reactions to generalized cardiovascular collapse. However, cases of vancomycin-induced leukocytoclastic vasculitis are rare. In this article, we present a case where the patient developed palpable purpura on his bilateral lower limbs following treatment with vancomycin. He was diagnosed with vancomycin-induced leukocytoclastic vasculitis that resolved without sequelae after withdrawal of vancomycin.

Due to their outstanding and versatile properties, polybenzoxazines have quickly occupied a great niche of applications. Developing the ability to polymerize benzoxazine resin at lower temperatures than the current capability is essential in taking advantage of these exceptional properties and remains to be most challenging subject in the field. The current review is classified into several parts to achieve this goal. In this review, fundamentals on the synthesis and evolution of structure, which led to classification of PBz in different generations, are discussed. Classifications of PBzs are defined depending on building block as well as how structure is evolved and property obtained. Progress on the utility of biobased feedstocks from various bio-/waste-mass is also discussed and compared, wherever possible. The second part of review discusses the probable polymerization mechanism proposed for the ring-opening reactions. This is complementary to the third section, where the effect of catalysts/initiators has on triggering polymerization at low temperature is discussed extensively. The role of additional functionalities in influencing the temperature of polymerization is also discussed. There has been a shift in paradigm beyond the lowering of ring-opening polymerization (ROP) temperature and other areas of interest, such as adaptation of molecular functionality with simultaneous improvement of properties.

Recent advances in woundcare is targeted towards developing active-dressings, where multiple components are combined to provide a suitable environment for rapid healing. The aim of the present research is to study the preparation of biomimic composite wound dressings by the grafting of hydrogel on silk fibroin fabric. The swelling ability of hydrogel grafted silk fibroin fabric was optimized by changing the initiator concentration. In order to impart antimicrobial properties, these dressing are further coated sono-chemically with zinc oxide nanoparticles. The water vapor transmission rate of the prepared samples was measured. The conformation of silk fibroin proteins after grafting with hydrogel was also confirmed using Fourier Transform Infrared Spectroscopy (FTIR). The morphology of the zinc oxide-coated silk fibroin fabric and hydrogel-coated silk fibroin was studied using Scanning Electron Microscope (SEM). The antimicrobial activity of the zinc oxide-coated samples was studied against E coli. The cytocompatibility of the prepared dressing materials were evaluated using L929 fibroblast cells. MTT assay and phase contrast microscopic studies showed that the adherence, growth, and proliferation of the L929 fibroblast cells that were seeded on zinc oxide nanoparticles on the functionalized hydrogel-coated silk fibroin dressing was significantly higher than that of pure silk fibroin due to the highly porous, bio-mimic structure that allowed ease of passage of nutrients, growth factors, metabolites, and the exchange of gases which is beneficial for successful regeneration of damaged tissues. The expression of TNF-α and IL-2 were not significantly higher than that of control. The proposed composite dressing would be a promising material for wound dressing and regenerative medicine but in order to prove the efficacy of these materials, more in vivo experiments and clinical tests are required to be conducted in future.

Tuberculosis is one of the top 10 causes of death worldwide according to the World Health Organization. Central nervous system involvement is usually the least common presentation of tuberculosis occurring in about 1% of all cases but yet can have very devastating outcomes. Lupus nephritis is one of the most common complications of systemic lupus erythematosus with up to two thirds of patients presenting with some degree of renal dysfunction. The mainstay of treatment is glucocorticoids; however, to sustain remission, steroid sparing agents such as cyclophosphamide, azathioprine and mycophenolate mofetil are used. Such patients, in addition to their baseline dysfunctional immune system, have a heightened risk of infections due to these drugs. In this article, we present a young woman who had recently been started on mycophenolate mofetil for control of class V lupus nephritis who presented with headaches, sinus pressure, and fevers. She had a protracted course of hospitalization as she failed to improve clinically and to respond to conventional therapy for acute bacterial sinusitis and meningitis. She was empirically started on antitubercular therapy 9 days after hospitalization. The diagnosis was not confirmed until day 18, the day results of cerebrospinal fluid acid-fast bacillus culture was reported. This case is reported to highlight the challenges in diagnosing Mycobacterium tuberculosis infection in an immunocompromised state and to demonstrate that its presentation can mimic numerous other conditions. Clinicians must maintain a high index of suspicion of Mycobacterium tuberculosis infection in such patients who present with nonspecific or unexplainable symptoms.

Introduction. Hyperkalemia is a commonly encountered clinical problem. Pseudohyperkalemia is believed to be an in vitro phenomenon that does not reflect in vivo serum potassium and therefore should not be treated. Here, we present a case who unfortunately underwent unnecessary treatment because of failure to detect the common lab abnormality of pseudohyperkalemia. Case Presentation. A 91-year-old female with a history of chronic lymphocytic leukemia presented to the emergency with nausea and vomiting 24 hours after her first chemotherapy with chlorambucil. Physical examination was overall unremarkable. She had a leukocytosis of 210 × 103/µL with 96% lymphocytes along with chronic anemia with hemoglobin of 8.1 g/dL. Her initial sodium and potassium levels were normal. During the clinical course, her potassium progressively worsened and failed to improve despite standard medical treatment. Patient ultimately underwent dialysis. Conclusions. Differentiating true hyperkalemia from pseudohyperkalemia is very important in selected group of patients to avoid unnecessary medications, higher level of care, and unnecessary procedure including dialysis. We want to emphasize the importance of simple yet profound knowledge of technique of blood draws and basic metabolic panel processing for every clinician in day-to-day practice.

Survival of Ehrlichia chaffeensis depends on obligatory intracellular infection. One of the barriers to E. chaffeensis research progress has been the inability, using conventional techniques, to generate knock-out mutants for genes essential for intracellular infection. This study examined the use of Peptide Nucleic Acids (PNAs) technology to interrupt type IV secretion system (T4SS) effector protein expression in E. chaffeensis followed by intracellular complementation of the effector to determine its requirement for infection. Successful E. chaffeensis infection depends on the E. chaffeensis-specific T4SS protein effector, ehrlichial translocated factor-1 (Etf-1), which induces Rab5-regulated autophagy to provide host cytosolic nutrients required for E. chaffeensis proliferation. Etf-1 is also imported by host cell mitochondria where it inhibits host cell apoptosis to prolong its infection. We designed a PNA specific to Etf-1 and showed that the PNA bound to the target region of single-stranded Etf-1 RNA using a competitive binding assay. Electroporation of E. chaffeensis with this PNA significantly reduced Etf-1 mRNA and protein, and the bacteria's ability to induce host cell autophagy and infect host cells. Etf-1 PNA-mediated inhibition of ehrlichial Etf-1 expression and E. chaffeensis infection could be intracellularly trans-complemented by ectopic expression of Etf-1-GFP in host cells. These data affirmed the critical role of bacterial T4SS effector in host cell autophagy and E. chaffeensis infection, and demonstrated the use of PNA to analyze the gene functions of obligate intracellular bacteria.

Trichoderma spp. are most popular biocontrol agents (BCA) known worldwide for their great ability to combat different soil and foliar diseases of agricultural crops. The use of biocontrol agents in IPM strategies requires the effective strains compatible with pesticides which can be well utilized in an integrated pest management system and also suitable to reduce the residual effect of pesticides. Keeping this in view, the study was designed to evaluate to lerance levels and biodegradation of benzimidazole fungicide (Carbendazim) by Trichoderma harzianum, Trichoderma viride and Trichoderma atroviride at three concentrations 100, 150 and 200 ppm were determined. The maximum per cent inhibition of T.harzianum, T.viride and T.atroviride in carbendazim amended media were 60, 83 and 93 at with spore production of 2.0×106, 1.4×106 and 0.8×106spore/µl at 200 ppm. Biodegradation per cent of carbendazim after 5 days of incubation determined through HPLC analysis were 85% in T.harzianum, 47% in T.viride and 21% in T.atroviride, thus clearly indicating the biodegradation property of Trichoderma spp.

Transition metal complexes of Co(II), Ni(II) and Cu(II) metal ions with general stoichiometry [M(L)X]X and [M(L)SO4], where M = Co(II), Ni(II) and Cu(II), L = 3,3’-thiodipropionic acid bis(4-amino-5-ethylimino-2,3-dimethyl-1-phenyl-3-pyrazoline) and X = NO3-, Cl- and OAc-, have been synthesized and structurally characterized by elemental analyses, molar conductance measurements, magnetic susceptibility measurements and spectral techniques like IR, UV and EPR. The nickel(II) complexes were found to have octahedral geometry, whereas cobalt(II) and copper(II) complexes were of tetragonal geometry. The covalency factor (β) and orbital reduction factor (k) suggest the covalent nature of the complexes. The ligand and its complexes have been screened for their antifungal and antibacterial activities against three fungi, i.e. Alternaria brassicae, Aspergillus niger and Fusarium oxysporum and two bacteria, i.e. Xanthomonas compestris and Pseudomonas aeruginosa.

Penile cysts are rare benign lesions. Their clinical or pathological diagnosis is extremely difficult. Less than 200 cases have been reported in literature. Most of them are asymptomatic and present since birth, but usually they are only detectable in adolescence or adulthood. These lesions rarely interfere with sexual function. These cysts most likely arise from ectopic urethral mucosa sequestered in the penile skin during embryologic development. We report a rare case of a 55-year-old man with a 4.5 × 4 cm nodule on the prepuce (ventral surface), which appeared two years ago and interfered with his sexual function. The nodule was excised by circumcision and a histopathological examination revealed a mucoid cyst of penis with squamous metaplasia. No recurrence was observed at a one year followup and the patient’s sexual function returned to normal.
