22. Combined Pretreatment as an Effective Technology in Breaking of Phenolic Polymer Lignin from Sustainable Biomass: Bambusa balcooa

Abstract

Bamboo biomass is a potential source for the production of monomeric sugars containing high cellulose content with low amount of lignin. However, for efficient hydrolysis, the biomass treatment by effective pretreatment technique is required to minimize lignin content and other barrier components. During present study, the bamboo biomass was treated with different physical, chemical, biological and combined treatments to reduce the lignin content. Among all the pretreatments, the maximum lignin removal (14.5%) was obtained with the combined chemical and biological treatment under 2% NaOH+1% H2O2 +WDP2 fungal culture (5 plugs) conditions. In addition, lignolytic fungus and NaOH pretreatment was mainly effective in removing lignin, whereas the H2O2 pretreatment efficiently minimize cellulose crystallinity. To analyze structural changes of raw and treated biomass, we used scanning electron microscopy and fourier transform infrared spectroscopy. The structural analysis indicated that all treatments causes disruption in the biomass structure and loses the compactness of the biomass which facilitates the biomass conversion during hydrolysis process. The findings of the present study indicate effective pretreatment methods in breaching the recalcitrancy of the potential lignocellulosic biomass for maximum hydrolysis.

26. Phenotypic and genotypic characterization of antimicrobial resistance in Enterococcusspecies from dairy niches

Abstract

The present work was carried out to investigate the prevalence of antimicrobial resistance (AMR) in dairy enterococci. Dairy samples comprising of raw milk and traditionally fermented milks were collected from local surrounding areas and a total of 140 enterococci strains were isolated from the dairy samples and further, 104 isolates were tested for antimicrobial susceptibility. The most prevalent resistance was observed against cephalosporins followed by fosfomycin, rifampicin, erythromycin etc. Multi-antibiotic resistance index of the tested isolates ranged from 0.09 to 0.36. Minimum inhibitory concentration testing showed very high cut-offs against erythromycin, tetracycline, cefepime and cefotaxime in certain isolates. Further, Extended Spectrum β-lactamase (ESBL) production tested by double disc synergy method confirmed 9 isolates as ESBL positive along with the presence of blaTEMgene in certain isolates. In addition, AMR genes against aminoglycoside, tetracyclines,macrolides, chloramphenicol along with multi-drug transporter gene (emeA) was also detected in many isolates. The resistant enterococci isolates also showed a high frequency of the presence of mobile genetic elements viz.Transposons (Tn-5397 and Tn-916/Tn-1545 family) and Integrons (intI1, intI3). Finally, virulence traits such as gelatinase and hemolysin production along with gelEand cylAgenes was also detected in many of the resistant enterococci isolates. Although, the virulence traits could not be directly correlated with the AMR resistance of the isolates, the results from the study demonstrated the wide prevalence of AMR in dairy enterococci that could greatly contribute to the problem of antimicrobial resistance in the food chain.

29. Efficacy of Reuterin and Bacteriocins Nisin and Pediocin in the Preservation of Raw Milk from Dairy Farms.

Abstract

Research background. In the current scenario of milk production in developing and developed countries, several factors influence the shelf-life of raw milk and add significant numbers of microbial contaminants that drastically lower the initial microbial quality leading to milk spoilage by the time it reaches the processing units. Experimental approach. The present study was undertaken to investigate the biopreservative efficacy of reuterin system along with different combinations of bacteriocins in controlling the initial microflora of raw milk at farm level. Lactobacillus reuteri strain LR47, having effective antimicrobial activity, was shortlisted from our previous study and further characterized for reuterin production and tested in raw milk system. Results and conclusions. Preliminary testing of the cell-free supernatant from L. reuteri LR47 demonstrated significant growth inhibition of the majority of the tested bacterial indicators of milk spoilage. Further genetic analysis of the L. reuteri LR47 revealed the presence of two genes (pduC and dhaB) involved in the utilization of glycerol to produce reuterin via two different pathways. The strain LR47 was also found to possess comparatively higher capacity to convert glycerol into reuterin when checked through colourimetric assay. In the raw milk biopreservation experiment with reuterin alone or in combination with bacteriocins, the highest level of growth suppression in the total bacterial load and coliform counts was observed in the sample that was treated with a combination of reuterin, nisin and pediocin. The treatment combining these three natural biopreservatives at specific concentrations was able to maintain the initial microbial quality and extend the shelf-life of raw milk by 6 h at 37 °C based on the microbial counts and physicochemical properties, viz. pH and titratable acidity. In conclusion, the results confirm that the use of reuterin in combination with bacteriocins is a promising approach for temporary control of the raw milk microflora and extension of its shelf-life until further processing. Novelty and scientific contribution. This study demonstrates for the first time the use of reuterin for the extension of shelf-life of raw milk as an alternative treatment method.
