The introduced brushtail possum (Trichosurus vulpecula) is a major threat to native biodiversity and agricultural production in New Zealand. Research on non-lethal management methods is focussed on fertility control, and aims to develop zona pellucida (ZP) vaccines suitable for bait delivery to free-living possums. Vaccine delivery remains a challenge. One highly successful oral wildlife vaccine which has been widely used to control rabies in wildlife in the US and Europe, is based on a replication-limited recombinant vaccinia virus (rVV). The potential of rVV as a vaccine delivery system has yet to be tested in any Australian marsupial species. In the present study we evaluated the infectivity of rVV, as well as cell-mediated and antibody immune responses, in the marsupial brushtail possum. Possums were exposed to a model recombinant vaccinia construct (rVV399, which expresses the Eg95 antigen of the hydatid disease parasite Echinococcus granulosus) or the parent vaccinia virus strain (Lister) by applying 108 pfu of virus, drop wise onto the external surface of the nose and into the oral cavity. Both the recombinant and parent viruses persisted in the mucosal epithelium around the palatine tonsils for up to 2 weeks post-exposure. The parent vaccinia and the rVV399 construct induced peripheral blood lymphocyte reactivity against viral antigens in possums by 4 weeks post-exposure, and were still detectable at 4 months post-exposure. Serum antibody reactivity to the antigen Eg95 was recorded in 7/8 possums which received a single dose of the rVV399 construct and in 7/7 animals which received tripledose delivery, with titre end-points in the latter case exceeding 1/4000 dilution. This study demonstrates that vaccinia virus will readily infect possums via an oronasal route and is capable of generating immune reactivity against both viral and heterologous antigens. This highlights the potential of recombinant vaccinia as a wildlife delivery system for fertility control vaccines for the brushtail possum and potentially other marsupial species.

Polytopal Element Methods (PEM) allow to solve differential equations on general polygonal and polyhedral grids, potentially offering great flexibility to mesh generation algorithms. Differently from classical finite element methods, where the relation between the geometric properties of the mesh and the performances of the solver are well known, the characterization of a good polytopal element is still subject to ongoing research. Current shape regularity criteria are quite restrictive, and greatly limit the set of valid meshes. Nevertheless, numerical experiments revealed that PEM solvers can perform well on meshes that are far outside the strict boundaries imposed by the current theory, suggesting that the real capabilities of these methods are much higher. In this work, we propose a benchmark to study the correlation between general 2D polygonal meshes and PEM solvers. The benchmark aims to explore the space of 2D polygonal meshes and polygonal quality metrics, in order to identify weaker shape-regularity criteria under which the considered methods can reliably work. The proposed tool is quite general, and can be potentially used to study any PEM solver. Besides discussing the basics of the benchmark, in the second part of the paper we demonstrate its application on a representative member of the PEM family, namely the Virtual Element Method, also discussing our findings.

- Abstract
This report presents the analysis, study, and simulation of an ultrafast charging station (UFCS) for electric vehicles (EVs) operating in steady state. The electrical architecture of the charging station uses an ac bus plus two dc buses and it is supported by a storage system based on batteries and super-capacitors. The power demand of the EVs is established taking into account the electric characteristics of their batteries and the availability of the station charging points. The analysis introduces a supervisory control based on a state machine description for different operating modes, which eventually facilitates fault detection in the electrical architecture. In addition, the study proposes different methods to handle the required energy for the charging demand and a procedure for the correct sizing of both the energy storage system and the input transformer. In laboratory experiments in a reduced-scale storage system, a SCADA supervision with CAN communication has proved successful in gathering data corresponding to modes of charge and discharge in batteries and super-capacitors, and subsequently displaying them on a computer screen.

- Abstract
Bats have been implicated as the reservoir hosts of filoviruses in Africa, with serological evidence of filoviruses in various bat species identified in other countries. Here, serum samples from 190 bats, comprising 12 different species, collected in Australia were evaluated for filovirus antibodies. An in-house indirect microsphere assay to detect antibodies that cross-react with Ebola virus (Zaire ebolavirus; EBOV) nucleoprotein (NP) followed by an immunofluorescence assay (IFA) were used to confirm immunoreactivity to EBOV and Reston virus (Reston ebolavirus; RESTV). We found 27 of 102 Yinpterochiroptera and 19 of 88 Yangochiroptera samples were positive to EBOV NP in the microsphere assay. Further testing of these NP positive samples by IFA revealed nine bat sera that showed binding to ebolavirus-infected cells. This is the first report of filovirus-reactive antibodies detected in Australian bat species and suggests that novel filoviruses may be circulating in Australian bats.

Amazon population genetics: the evolutionary history of the jaguar, ocelot, pink river dolphin, woolly monkey and wattled curassow reconstructed through their genes The Amazon has more than the half of the world’s biodiversity. Nevertheless, the major fraction of the Amazon species has unknown evolutionary histories. This is also certain for mammals and birds. Population genetics, employing molecular markers and theoretical mathematics models, can reconstruct these evolutionary histories and offer very powerful tools for the application of correct conservation politics. Herein, we show a comparative view of population genetics results obtained for Amazon populations of jaguar, ocelot, pink river dolphin, woolly monkey and wattled curassow and provide recommendation for their biological conservation. Each species showed its own specific evolutionary particularities, characteristics that were not shared by the other species. This finding should be taken into consideration for any effective biological conservation program.

Many affected counties have had experienced a shortage of personal protective equipment (PPE) during the coronavirus disease (COVID-19) pandemic. We aimed to investigate the needs of healthcare professionals and the technical difficulties faced by them during the initial outbreak. A cross-sectional web-based survey was conducted among the healthcare workforce in the most populous cities from three Latin American countries in April 2020. In total, 1,082 participants were included. Of these, 534 (49.4%), 263 (24.3%), and 114 (10.5%) were physicians, nurses, and other professionals, respectively. At least 70% of participants reported a lack of PPE. The most common shortages were shortages in gown coverall suits (643, 59.4%), N95 masks (600, 55.5%), and face shields (569, 52.6%). This paper provides a starting point for researchers and practitioners from biology, medicine, physics and engineering who can benefit from an up-to-date literature survey on patient-specific bone fracture modelling, simulation and risk analysis. This survey hints at a framework for devising realistic patient-specific bone fracture simulations. This paper has 18 sections: Section 1 presents the main interested parties; Section 2 explains the organzation of the text; Section 3 motivates further work on patient-specific bone fracture simulation; Section 4 motivates this survey; Section 5 concerns the collection of bibliographical references; Section 6 motivates the physico-mathematical approach to bone fracture; Section 7 presents the modelling of bone as a continuum; Section 8 categorizes the surveyed literature into a continuum mechanics framework; Section 9 concerns the computational modelling of bone geometry; Section 10 concerns the estimation of bone mechanical properties; Section 11 concerns the selection of boundary conditions representative of bone trauma; Section 12 concerns bone fracture simulation; Section 13 presents the multiscale structure of bone; Section 14 concerns the multiscale mathematical modelling of bone; Section 15 concerns the experimental validation of bone fracture simulations; Section 16 concerns bone fracture risk assessment. 33. First report of Trypanosoma dionisii (Trypanosomatidae) identified in Australia

Trypanosomes are blood-borne parasites that can infect a variety of different vertebrates, including animals and humans. This study aims to broaden scientific knowledge about the presence and biodiversity of trypanosomes in Australian bats. Molecular and morphological analysis was performed on 86 blood samples collected from seven different species of microbats in Western Australia. Phylogenetic analysis on 18S rDNA and glycosomal glyceraldehyde phosphate dehydrogenase (gGAPDH) sequences identified Trypanosoma dionisii in five different Australian native species of microbats; Chalinolobus gouldii, Chalinolobus morio, Nyctophilus geoffroyi, Nyctophilus major and Scotorepens balstoni. In addition, two novel, genetically distinct T. dionisii genotypes were detected and named T. dionisii genotype Aus 1 and T. dionisii genotype Aus 2. Genotype Aus 2 was the most prevalent and infected 20.9% (18/86) of bats in the present study, while genotype Aus 1 was less prevalent and was identified in 5.8% (5/86) of Australian bats. Morphological analysis was conducted on trypomastigotes identified in blood films, with morphological parameters consistent with trypanosome species in the subgenus Schizotrypanum. This is the first report of T. dionisii in Australia and in Australian native bats, which further contributes to the global distribution of this cosmopolitan bat trypanosome.

34. Coronaviruses and Australian bats: a review in the midst of a pandemic

Australia’s 81 bat species play vital ecological and economic roles via suppression of insect pests and maintenance of native forests through pollination and seed dispersal. Bats also host a wide diversity of coronaviruses globally, including several viral species that are closely related to SARS-CoV-2 and other emergent human respiratory coronaviruses. Although there are hundreds of studies of bat coronaviruses globally, there are only three studies of bat coronaviruses in Australian bat species, and no systematic studies of drivers of shedding. These limited studies have identified two betacoronaviruses and seven alphacoronaviruses, but less than half of Australian species are included in these studies and further research is therefore needed. There is no current evidence of spillover of coronaviruses from bats to humans in Australia, either directly or indirectly via intermediate hosts. The limited available data are inadequate to determine whether this lack of evidence indicates that spillover does not occur or occurs but is undetected. Conversely, multiple international agencies have flagged the potential transmission of human coronaviruses (including SARS CoV-2) from humans to bats, and the consequent threat to bat conservation and human health. Australia has a long history of bat research across a broad range of ecological and associated disciplines, as well as expertise in viral spillover from bats. This strong foundation is an ideal platform for developing integrative approaches to understanding bat health and sustainable protection of human health.

Cognitive functions are important correlates of health outcomes across the life-course. Individual differences in cognitive functions are partly heritable. Epigenetic modifications, such as DNA methylation, are susceptible to both genetic and environmental factors and may provide insights into individual differences in cognitive functions. Epigenome-wide meta-analyses for blood-based DNA methylation levels at ~420,000 CpG sites were performed for seven measures of cognitive functioning using data from 11 cohorts. CpGs that passed a Bonferroni correction, adjusting for the number of CpGs and cognitive tests, were assessed for: longitudinal change; being under genetic control (methylation QTLs); and associations with brain health (structural MRI), brain methylation and Alzheimer's disease pathology. Current chromium concentrations in natural waters amount to only a few micrograms per liter. Electrothermal atomic absorption spectrometry (ETAAS) is one of the most suitable techniques for its determination. However, it was found that high iron concentrations (3-4 orders of magnitude) may cause serious atomic signal enhancement in Cr analysis. It is discussed that Fe may cause either spectral or chemical (or both) interferences on Cr determination. The goal of this paper is to develop multivariate calibration models not affected by such interferences. Three multivariate regression methods were applied. One of them was linear in its nature (partial least squares, PLS), and two were nonlinear (polynomial PLS and locally weighted regression). PLS was revealed to be a suitable and convenient alternative to simplify current laboratory workload in order to cope with chemical and spectral interferences caused by a major metal (Fe) when determining another trace metal (Cr) by ETAAS. Three effects, which modify the atomic peak shape, due to the aging of the atomizers were simulated to evaluate to what extent they affect the predictions, namely, peak-shift, peak enhancement (depletion), and increased random noise. The ETAAS-PLS methodology was satisfactorily tested with water samples and two CRMs.
