The article informs that the protozoans of the genus Kudoa, which parasitize estuarine and marine fishes, are most commonly seen in the skeletal musculature and can create lesions of economic importance. They have also been identified in other fish tissues including the kidney, gills, fins, nervous tissue, cardiac muscle, external gut wall, and mesentery and peritoneum. The present report describes a histozoic infection of a Kudoa species in juvenile white perch, Morone americana (Gmelin), from the Chesapeake Bay, U.S., which were collected as part of a larger study of hepatic copper storage in this species.

Segments of the kidney tubules of about 16% of cod caught in the approaches to Halifax Harbour were found to contain a coccidian parasite. Macrogametes developed in an epicellular position, and microgamonts, unsporulated and sporulating oocysts, and free sporocysts, were found in the lumen of the kidney tubule. The epithelial cells of the kidney tubules were damaged and granulomas often formed around infected tubules. The presence of a suture dividing the sporocyst wall into two valves could only be determined using electron microscopy. Other coccidians which have been placed in the genus Eimeria because sutures could not be seen using light microscopy, although no Stieda body was present, may also belong to the genus Goussia. No other coccidians have been reported in the kidney tubules of marine fish, and the name Goussia spragiiei sp. nov., is proposed. [ABSTRACT FROM AUTHOR]

Myxosporeans are common parasites of fish, and uncommon parasites of amphibians, reptiles and invertebrates, that can cause significant morbidity and mortality. The common genus Myxidium infects the excretory system of turtles, yet knowledge of its pathogenicity in these hosts is limited. We offer new knowledge of morphological and ultrastructural aspects of host-parasite interactions in Myxidium infections from our recent diagnostic investigations on captive freshwater turtles listed in CITES (Appendix II). We investigated the cause of death of 2 adult Indo-Gangetic flap-shelled turtles Lissemys punctata andersonii from a zoo collection. After post-mortem examination, tissues were processed for histopathology, and special stains were used to demonstrate morphology of myxosporean spores. Additional kidney tissue, immersion-fixed in formalin, was processed for transmission electron microscopy. Both turtles were infected with a myxosporidian, Myxidium mackiei, in the kidney, which occluded 5 to 10% of the renal proximal convoluted tubules. The polysporic plasmodia contained pairs of developing and mature spores. Each mature, spindle-shaped spore had 2 asymmetric valves (1 overlapping, 1 overlapped), with 10 to 13 and 10 to 14 longitudinal ridges per valve, and 2 polar capsules each containing a polar filament with 4 to 5 turns. A pair of spores, each surrounded by a membrane-bound electron-lucent matrix, lay in an enclosing cell within the plasmodium. Regions of the parasite-host interface consisted of undulations of the parasite surface, with intense pinocytotic activity beneath, intermingled with the hosts' microvilli, and endocytotic channels at the apex of renal epithelial cells. The microvilli of the renal epithelial cells of infected tubules were frequently sheared or compressed, or occasionally missing; we did not detect other pathology induced by the parasite. Our report of M. mackiei in L. punctata is a new host record. Both individuals also had disseminated pale yellow nodules (bacterial granulomas) present in lung, heart, kidney, and skeletal muscle, and both were infected with coccidia (tentatively identified as Eimeria sp.) in multiple organs. The cause of death for one turtle was septicemia, but remained unknown for the other individual.

Ulcers in Atlantic menhaden Brevoortia tyrannus (Latrobe) (Clupeidae), observed along the USA east coast, have been attributed to diverse etiologies including bacterial, fungal and, recently, harmful algal blooms. To understand the early pathogenesis of these lesions, we examined juvenile Atlantic menhaden collected during their seasonal presence in Chesapeake Bay tributaries from April to October 1999 and from March to August 2000. We conducted histopathological examinations of young-of-the-year fish from the Pocomoke River tributary, which has a history of fish mortalities and high lesion prevalence. Kudoa clupeidae (Myxozoa: Myxosporea) spores were present in the muscles of fish collected in both years. Of the fish assessed by histology in April, 5 to 14% were infected, while in May 90 to 96% were infected. Infection rates remained high during the summer. Mature spores were primarily located within myomeres and caused little or no observable pathological changes. Ultrastructure showed spores with capsulogenic cells bearing filamentous projections, and a basal crescentic nucleus with mottled nucleoplasm containing cleaved, condensed chromatin. Also, a highly invasive plasmodial stage of a myxozoan was found in the lesions of juvenile Atlantic menhaden. The plasmodia were observed in fish collected between May and July, with the maximum occurrence in late June 1999 and late May 2000. Plasmodia penetrated and surrounded muscle bundles, causing grossly observable raised lesions in 73% of all fish infected with this invasive stage. Plasmodia were also detected in the visceral organs, branchial arches, and interocular muscles of some fish. Some of the invasive extrasporogonic plasmodial lesions were associated with ulcers and chronic inflammatory infiltrates. The plasmodial stage appeared to slough out of the tissue with subsequent evidence of wound healing. Ultrastructure showed plasmodia with an elaborate irregular surface, divided into distinct ectoplasm and endoplasm; the latter contained numerous spherical vegetative nuclei, secondary generative cells, and occasional cell doublets. Our ultrastructural studies indicate that the plasmodial organisms, which are important in the etiology of the skin lesions, are myxozoans, and they may represent early stages of K. clupeidae.

The successful 6 mo rehabilitation of a stranded juvenile pygmy sperm whale Kogia breviceps afforded the opportunity to study the poorly known protozoan fauna of the upper respiratory tract of cetaceans. Mucus samples were collected by holding either a petri dish or glass slides over the blowhole for 3 to 5 exhalations; preparations were examined as wet mounts, and then stained with Wrights-Giemsa or Gram stain. Blood smears were stained with Wrights-Giemsa. Unidentified spindle-shaped and unidentified broad ciliates, reported from the blowhole of the pygmy sperm whale for the first time, were seen only initially, while yeast-like organisms and bacteria were seen intermittently. Epithelial cells and white blood cells were often present in the blowhole mucus, but red blood cells were never seen. A novel trypanoplasm-like bodonid kinetoplastid biflagellate (Order Kinetoplastida) was commonly encountered in the blowhole mucus, but never in the blood. Both mature flagellates and those undergoing longitudinal binary fission were present. The elongate flagellate had a long whiplash anterior flagellum; the recurrent flagellum was attached along at least two-thirds of the body length, forming a prominent undulating membrane, and the trailing portion was short. The kinetoplast was irregularly fragmented. The flagellates were either free-swimming, or attached to host material via the free portion of the posterior flagellum. The prominent undulating membrane was characteristic of Trypanoplasma, while the fragmented kinetoplast was characteristic of some species of Cryptobia. For the novel bodonid kinetoplastid, with its unique combination of morphological features (prominent undulating membrane and fragmented kinetoplast), we propose the creation of a new genus Jarrellia. We believe this to be the first published description of a flagellate from a marine mammal, and among the first reports of a trypanoplasm-like flagellate from a warm-blooded host. We expect that a diversity of flagellates and ciliates are commonly present in the blowhole of cetaceans. Future studies on the identity of the protozoans and the health of their cetacean hosts, which are readily studied in captivity, are necessary to establish their status as commensals or parasites.

An adult female lesser flamingo (Phoeniconaias minor), caught in the African Rift Valley in 1991 and subsequently housed at the Baltimore Zoo, died of severe visceral gout in 1996. Necropsy revealed a white, moderately firm, nodular lesion, 1 cm in diameter, in the serosal wall of the small intestine. Although it was initially thought to be a tumor or focal granuloma, histologic examination revealed multiple cestodes deeply embedded at the base of the crypts between the intestinal villi, with their massive scolices (up to 3.4 mm in diameter) distending these spaces into multiple diverticulae. The mucosal epithelium surrounding the scolices was severely attenuated. Around the diverticulae, in the submucosa and muscularis, was a mild to moderate lymphocytic reaction and mild fibrosis. The proximity of multiple scolices and extensive invasion of host tissue suggested that the infection occupied a preexisting lesion. The cestodes were cyclophyllids but were distinct from any species previously reported from flamingos. Helminths should be included in differential diagnoses for gastrointestinal nodules in flamingos.

In 1995 at a sea-farm in Ireland, Atlantic salmon Salmo salar smolts were introduced in the spring and autumn into 2 marine rearing sites, one with and one without recent histories of neurologic disease and mass mortality believed to be due to parasitic encephalitis. In order to monitor disease, determine the onset and anatomic distribution of parasites and encephalitis, and determine the ultrastructure and identity of the parasite, randomly selected smolts were necropsied from both sites at intervals of 1 to 3 d for periods of 1 to 2 mo, and clinical signs and cumulative mortalities were monitored. The prevalences of parasites and encephalitis were assessed by light microscopy, and sections of brain were examined by transmission electron microscopy. No clinical signs of neurologic disease were observed, but parasites and encephalitis were detected in smolts from both sites, with prevalences of each that were significantly greater in smolts from the site with recent disease. Based on light microscopy, the parasite was first detected in smolts sampled at 26 d post-introduction. Parasites were detected with significantly greater frequency in the optic tectum of the mesencephalon than in the diencephalon, metencephalon, and myelencephalon, and were not detected in the telencephalon. Foci of non-suppurative encephalitis were detected with significantly greater frequency in the myelencephalon than in each of the other 4 anatomic subdivisions. In each anatomic subdivision the prevalence of encephalitis was significantly greater than that of parasitic infection. The ultrastructure of the parasite was consistent with that of a histozoic presporogonic multicellular developmental stage of a myxosporean, characterized by intercellular branching tubular structures containing generative cells arranged individually or in cell-in-cell doublets. Parasitic stages were located between bundles of axons, with compression of axons along segments of their plasmalemma. No mature spores were detected.

Light microscopy studies have previously shown that Bombycirhynchus sphyraenaicum is an exceptional trypanorhynch cestode, characterised by a poeciloacanthous armature and two enormous bothridia, which overlap parts of the pars bulbosa, a character combination unique within the trypanorhynchs. Plerocercoids of B. sphyraenaicum from the fish Lates calcarifer (Centropomidae) were investigated by scanning and transmission electron microscopy. Results revealed that the tegument of the anterior margin of the distal bothridial surface bears three kinds of microtriche; palmate microtriches, 8 microns high, with seven to eight digitiform processes; equalized filamentous microtriches, 5-6 microns long, with cap and base each forming 50% of the length, borne on undulations beneath the palmate microtriches, and cap-dominated filamentous microtriches, 5-6 microns long, with the cap forming 75% of the length, borne on the apex of putative sensory papillae. Integumental connections link the bases of the palmate microtriches, forming transverse girdles around the worm, which may serve to coordinate traction. At regular intervals between the palmate microtriches are papillae, covered with filamentous microtriches extending above the palmate microtriches. A cilium emerges from a bulb at the apex of each papilla; other structures in the bulb include an electron-dense cuff, and two electron dense collars. The posterior part of the bulb tapers and passes into the tegumental cytoplasm. These adorned papillae, observed for the first time in trypanorhynch cestodes, are identified as putative mechanoreceptors.

A diplomonad flagellate, Spironucleus torosa n. sp. is described from Atlantic Cod Gadus morhua and haddock Melanogrammus aeglefinus. This is believed to be the 1st confirmed report of Spironucleus from a marine fish. Organisms swimming in the rectal lumen were broadly pyriform to elongate, and measured 10.5-18.6 microns long and 3.2-13.3 microns wide; other elongate organisms were attached to the rectal epithelium, via apical extensions appearing continuous with the microvilli. The posterior end of the body was extended into a caudal projection, on either side of which was a posteriolateral ring-shaped protrusion or torus, with a recurrent flagellum emerging from its centre. A symmetrical system of microtubules and lamellae, forming a "V" in protargol impregnated specimens, supported the flanges of the body surrounding the tori, the tori themselves and the caudal projection. Supranuclear microtubules were an inverted V to U shape in transverse section, and an electron dense band accompanied the cytostomes. Lightly staining homogenous cytoplasm was usually present in the anterior part of the body, the remainder being highly vacuolated with numerous dark granules. In swimming organisms, rough endoplasmic reticulum (RER) was present around the nuclei and cytostomes, and bacteria were occasionally seen in the cytoplasm. In "attached" organisms, RER was reduced, and bacteria were absent. Hexamita salmonis Moore from Salvelinus fontinalis was studied by light and scanning electron microscopy for comparison; its cytoplasm was not highly vacuolated. The two recurrent flagella emerged close together from the blunt posterior end of the body.
